Es wurden bereits zahlreiche Versuche unternommen, Arzneistoffe zur Behandlung von Thrombosen zu entwik keln. Aus der US-PS 3 622 615 sind N2-(p-Tolylfulfonyl)-Largininester bekannt, die zur Behandlung von thrombophilen Diathesen geeignet sind. Schliesslich sind in der DE OS 24 438 851 N2-Dansyl-L-argininester und -amide vorgeschlagen, die Thrombin-Inhibitoren darstellen und sich zur Therapie oder zur Prophylaxe von Thrombosen eignen.
Der Erfindung liegt die Aufgabe zugrunde, neue N2-substituierte L-Argininamide der nachstehenden allgemeinen Formel I zu schaffen, die hochspezifische Thrombin-Inhibitoren darstellen und sich zur Behandlung oder Prophylaxe von thrombophilen Diathesen eignen. Diese Aufgabe wird durch die Erfindung gelöst.
Die Erfindung betrifft somit den im Anspruch gekennzeichneten Gegenstand.

Zur Herstellung der Verbindungen der allgemeinen Formel I können verschiedene Verfahren angewendet werden, die von den Ausgangsverbindungen und bzw. oder Zwischenprodukten abhängen.
(a) Kondensation eines L-Argininamids mit einem Sulfonylhalogenid
Dieses Verfahren kann durch folgendes Reaktionsschema erläutert werden:

Die Kondensation wird mit praktisch äquimolaren Mengen des Sulfonylhalogenids der allgemeinen Formel IV, vorzugsweise einem Chlorid, in Gegenwart einer Base durchgeführt. Die Reaktionsbedingungen sind die gleichen wie für das Verfahren (1) (b). Die eingesetzten L-Argininamide der allgemeinen Formel V können durch Schutz der Guanidinogruppe und der a-Aminogruppe des Arginins durch Nitrierung, Acetylierung, Formylierung, Phthaloylierung, Trifluoracetylierung, p-Methoxybenzyloxycarbonylierung, Benzoylierung, Benzyloxycarbonylierung, tert.-Butoxycarbonylierung oder Tritylierung hergestellt werden. Sodann wird das erhaltene NG-substituierte N2-substituierte L-Arginin mit einem Amin in an sich bekannter Weise, beispielsweise nach der Säurechlorid-, Azid-, gemischten Anhydrid-, aktivierten Ester- oder Carbodiimid-Methode umgesetzt.
Danach wird die Schutzgruppe in an sich bekannter Weise abgespalten.
(b) Umsetzung von N2-substituierten L-Arginylhalogeniden mit Aminen

Das Arginylhalogenid der allgemeinen Formel X, vorzugsweise das Chlorid, wird mit mindestens äquimolaren Mengen eines Amins der allgemeinen Formel III umgesetzt.
Die Umsetzung wird vorzugsweise in Gegenwart eines Lösungsmittels, beispielsweise eines basischen Lösungsmittels, wie Dimethylformamid oder Dimethylacetamid, oder eines halogenierten Kohlenwasserstoffs, wie Chloroform oder Di chlormethan, durchgeführt. Das Lösungsmittel kann in einem verhältnismässig breiten Mengenbereich verwendet werden. Vorzugsweise wird das Lösungsmittel in der 5- bis 100fachen Gewichtsmenge des N2-substituierten L-Arginylhalogenids der allgemeinen Formel X eingesetzt. Die Reaktionstemperatur liegt vorzugsweise im Bereich von -10 "C bis Raumtemperatur. Die Reaktionszeit hängt unter anderem von der Art des eingesetzten Amins und der Reaktionstemperatur ab. Gewöhnlich beträgt sie 5 Minuten bis 10 Stunden.
Die erhaltenen N2-substituierten L-Argininamide können nach den vorstehend beschriebenen Methoden isoliert und gereinigt werden.
Die verfahrensgemäss eingesetzten N2-substituierten L Arginylhalogenide der allgemeinen Formel X können durch Umsetzung eines N2-substituierten L-Arginins der allgemeinen Formel II

in der R' die vorstehende Bedeutung hat, mit mindestens äquimolaren Mengen eines Halogenierungsmittels, wie Thionylchlorid, Phosphoroxychlorid, Phosphortrichlorid, Phos phorpentachlorid oder Phosphortribromid, hergestellt werden. Vorzugsweise wird die Halogenierung in einem inerten Lösungsmittel, beispielsweise in einem chlorierten Kohlenwasserstoff, wie Chloroform oder Dichlormethan, oder einem Äther, wie Tetrahydrofuran oder Dioxan, durchgeführt. Das Lösungsmittel kann in einem verhältnismässig breiten Mengenbereich verwendet werden. Vorzugsweise wird das Lösungsmittel in der 5- bis 100fachen Gewichtsmenge des N2-substituierten L-Arginins eingesetzt.
Die Reaktionstemperatur liegt vorzugsweise im Bereich von -10 "C bis Raumtemperatur. Die Reaktionszeit hängt unter anderem vom verwendeten Halogenierungsmittel und der Reaktionstemperatur ab. Gewöhnlich beträgt sie 15 Minuten bis 5 Stunden.
(c) Umsetzung von N2-substituierten L-Argininestern mit primären Aminen
Dieses Verfahren kann durch folgendes Reaktionsschema erläutert werden:

R2 und R' haben die vorstehend angegebene Bedeutung, R7 ist ein Cl¯l0-Alkylrest.
Die Umsetzung wird mit etwa 2 bis 10 Äquivalenten des primären Amins der allgemeinen Formel XIII und gegebenenfalls in einem Lösungsmittel, beispielsweise einem Alkohol, wie Methanol oder Äthanol, einem Äther, wie Di äthyläther oder Tetrahydrofuran, einem Kohlenwasserstoff, wie Benzol oder Toluol, einem halogenierten Kohlenwasserstoff, wie Chloroform oder Dichlormethan, oder Wasser, bei Raumtemperatur während eines Zeitraums von mehreren Stunden bis zu mehreren Tagen durchgeführt. Zur Beschleunigung der Umsetzung kann das Reaktionsgemisch auf Temperaturen bis zum Siedepunkt des verwendeten Amins oder Lösungsmittels erhitzt werden. Es kann auch ein basischer Katalysator, wie Natriummethoxid oder Pyridin, verwendet werden.
Nach beendeter Umsetzung wird das entstandene N2-substituierte L-Argininamid der allgemeinen Formel XIV abfiltriert, oder der Überschuss an Amin und/ oder das Lösungsmittel wird verdampft, der Rückstand mit Wasser gewaschen und das Produkt aus einem Lösungsmittel, wie wasserhaltiges Methanol, umkristallisiert.
Die Verbindungen der allgemeinen Formel I bilden mit den verschiedensten anorganischen und organischen Säuren Salze. Die Produkte der vorstehend beschriebenen Reaktionen können in Form der freien Base oder als Salz mit einer Säure isoliert werden. Zur Herstellung pharmakologisch verträglicher Salze können die freien Basen mit einer anorganischen Säure, wie Salzsäure, Bromwasserstoffsäure, Jodwasserstoffsäure, Salpetersäure, Schwefelsäure oder Phosphorsäure, oder mit einer organischen Säure, wie Essigsäure, Citronensäure, Maleinsäure, Bernsteinsäure, Milchsäure, Weinsäure, Gluconsäure, Benzoesäure, Methansulfonsäure, Äthansulfonsäure, Benzolsulfonsäure oder p-Toluolsulfonsäure, umgesetzt werden. Die Behandlung der Salze mit einer Base liefert die freien Amide oder Ester.
Die Verbindungen der allgemeinen Formel I und ihre Salze mit Säuren sind wertvolle Arzneistoffe, die spezifisch Thrombin hemmen. Die Verbindungen können daher zur Bestimmung von Thrombin im Blut und zur Behandlung oder Vorbeugung von thrombophilen Diathesen eingesetzt werden.
Die Thrombinhemmung der Verbindungen der Erfindung wird mit der Wirksamkeit des aus der US-PS 3 622 615 bekannten Thrombininhibitors N2-(p-Toluolsulfonyl)-L-ar gininmethylesters durch Ermittlung der Fibrinogen-Koagulationszeit verglichen.
Die Messung wurde folgendermassen durchgeführt: Es wird eine Fibrinogenlösung durch Auflösen von 150 mg Rinder-Fibrinogen (Cohn-Fraktion I) in 40 ml eines Borat Kochsalz-Puffers (pH 7,4) hergestellt. Zur Herstellung einer Blindprobe werden 0,8 ml dieser Fibrinogenlösung mit 0,1 ml eines Borat-Kochsalz-Puffers (pH 7,4) versetzt. Weitere 0,8 ml der ursprünglichen Fibrinogeniösung werden mit 0,1 ml einer Probelösung im selben Puffer versetzt. Schliesslich werden diese beiden Lösungen in einem Eisbad mit jeweils 0,1 ml einer Thrombinlösung (5 Einheiten pro ml) versetzt.
Nach erfolgter Zugabe werden die Reaktionsgemische sofort aus dem Eisbad genommen und in ein auf 25 "C thermostatisiertes Bad verbracht. Die Koagulationszeit ist die Zeitspanne, die zwischen dem Einbringen in das auf 25 "C erwärmte Bad und dem Zeitpunkt des ersten Auftretens von Fibrinfäden verstreicht. In den Fällen, in denen keine Probezugabe erfolgte, beträgt die Koagulationszeit 50 bis 55 Sekunden.
Die Ergebnisse sind nachstehend in Tabelle I zusammengefasst. Der Ausdruck zur Verdoppelung der Koagulationszeit erforderliche Konzentration bedeutet die Konzentration des Wirkstoffs, die erforderlich ist, um die normale Koagulationszeit von 50 bis 55 Sekunden auf 100 bis 110 Sekunden zu verdoppeln. Die zur Verdoppelung der Koagulationszeit erforderliche Konzentration beträgt bei Verwendung von N2-(p-Tolylsulfonyl)-L-argininmethylester 1100 MoL
Die in Tabelle I angegebenen Inhibitoren der allgemeinen Formel I werden durch die Reste R und R' gekennzeichnet. Ferner ist die zur Salzbildung verwendete Säure angegeben.
Bei intravenöser Verabfolgung einer Lösung der Verbindungen der allgemeinen Formel I an Tiere bleibt die Thrombininhibierung im Blutkreislauf 1 bis 3 Stunden bestehen.
Die Halbwertszeit der Verbindungen im Blutkreislauf beträgt etwa 30 Minuten. Die physiologischen Bedingungen der Versuchstiere (Ratten, Kaninchen, Hunde und Schimpansen) werden gut beibehalten. Die durch Thrombin-Infusion verursachte experimentelle Fibrinogenabnahme bei Tieren wird mit gutem Erfolg durch die gleichzeitig erfolgte Infusion der Verbindungen der Erfindung unter Kontrolle gehalten.
Die akute Toxizität (LDso) wird 24 Stunden nach oraler Verabfolgung der Verbindungen der allgemeinen Formel I an männliche Mäuse mit einem Körpergewicht von 20 g bestimmt. Sie beträgt etwa 1000 bis 10 000 mg/kg Körpergewicht.
Die Verbindungen können in Form üblicher Arzneipräparate parenteral, d. h. intramuskulär, intravenös oder subkutan, oder oral verabfolgt werden. Die Arzneipräparate enthalten neben dem Wirkstoff übliche Trägerstoffe und/ oder Verdünnungsmittel und/oder Hilfsstoffe.
Die Beispiele erläutern die Erfindung.
Beispiel 1
5,0 ml (0,069 Mol) kaltes Thionylchlorid werden unter kräftigem Rühren mit 1,0 g (0,00236 Mol) N2-(6,7-Dimethoxy-2-naphthalinsulfonyl)-L-arginin versetzt. Das Reaktionsgemisch wird 1 Stunde bei Raumtemperatur umgesetzt. Danach werden 100 ml wasserfreier Diäthyläther zugegeben, und die entstandene Fällung wird abfiltriert und gründlich mit 50 ml wasserfreiem Diäthyläther gewaschen.
Das als Pulver erhaltene N2-(6,7-Dimethoxy2-naphthalinsulfonyl)-L-arginylchlorid-dihydrochlorid wird unter Rühren in eine Lösung von 1,2 g (0,012 Mol) 4-Methypiperidin in 10 ml Chloroform eingetragen. Das Gemisch wird 3 Stunden bei Raumtemperatur stehengelassen. Danach werden das Lösungsmittel und überschüssiges 4-Methylpiperidin unter vermindertem Druck abdestilliert, und der Rückstand wird in 20 ml Chloroform gelöst. Die Chloroformlösung wird mit gesättigter wässriger Kochsalzlösung gewaschen, über Natriumsulfat getrocknet und unter vermindertem Druck eingedampft. Der Rückstand wird mit 10 ml Essigsäure und 100 ml wasserfreiem Diäthyläther versetzt. Hierbei scheidet sich ein öliges Produkt aus. Der Äther wird dekantiert und das ölige Produkt gründlich mit wasserfreiem Diäthyläther gewaschen.
Es wird das 4-Methyl 1 [N2 -(6,7-dimethoxy-2-naphthalinsulfonyl)-L-arginylj-pipe- ridin-monoacetat als Pulver erhalten. Ausbeute 1,1 g (84% d. Th.).

Danach wird das Gemisch 1 Stunde bei 0 bis 5 "C und weitere 2 Stunden bei Raumtemperatur gerührt. Hierauf wird das Reaktionsgemisch mit 100 ml wasserfreiem Diäthyläther versetzt und der Überstand dekantiert. Der ölige Rückstand wird mit 50 ml wasserfreiem Diäthyläther gewaschen. Es wird das N2-(6, 7-Dimethoxy-2-naphthalinsulfonyl)-L-arginylchloriddihydrochlorid als Pulver erhalten. Dieses Produkt wird in eine Lösung von 1,31 g (0,015 Mol) N-Methyl-N-butylamin in 10 ml Chloroform eingetragen.

Beispiel 3
Ein Gemisch von 1,00 g (0,0037 Mol) 4-Äthyl-l-(L-arginyl)-piperidin und 0,61 g (0,0044 Mol) Kaliumcarbonat in 10 ml Wasser wird auf 0 "C abgekühlt und innerhalb 30 Minuten unter kräftigem Rühren tropfenweise mit einer Lösung von 1,25 g (0,0044 Mol) 6,7-Dimethoxy-2-naphthalin sulfonylchlorid in 30 ml Dioxan versetzt. Eine Lösung von 1,00 g (0,0041 Mol) 4-(L-Arginyl)-morpholin in 50 ml Chloroform und 0,52 g (0,0052 Mol) Tri äthylamin wird bei Raumtemperatur unter Rühren mit 1,48 g (0,0052 Mol) 6,7-Dimethoxy-2-naphthalinsulfonylchlorid versetzt. Nach Sstündigem Rühren bei Raumtemperatur wird das Reaktionsgemisch mit 10 ml Wasser aufgeschlämmt. Die wässrige Phase wird abgetrennt und die Chloroformphase über Natriumsulfat getrocknet. Nach Zusatz von 2 ml Essigsäure zur Chloroformlösung wird das Gemisch eingedampft. Es hinterbleibt ein viskoses Öl, das mit einem Gemisch von Methanol und Diäthyläther umgefällt wird. Es wird das 4-fN2-(6,7-Dimethoxy- 2-naphthalinsulfonyl)-L-arginyl]-morpholin-acetat in 66prozentiger Ausbeute erhalten.

Example 2
A suspension of 1.00 g (0.00236 mol) of N2- (6,7-dimethoxy-2-naphthalenesulfonyl) -L-arginine in 20 ml of tetrahydrofuran is added in portions with 0.98 g (0.0047 mol) of phosphorus pentachloride placed under ice cooling. A solution of 1.00 g (0.0041 mol) of 4- (L-arginyl) morpholine in 50 ml of chloroform and 0.52 g (0.0052 mol) of triethylamine is stirred at room temperature with 1.48 g ( 0.0052 mol) of 6,7-dimethoxy-2-naphthalenesulfonyl chloride.
