4. Verfahren nach Anspruch 1 zur Herstellung von 1 -(Thiazolin-2-yl)-2-amino-6-(a-hydroxy-a-äthylbenzyl)benzimidazol, dadurch gekennzeichnet, dass man l-(Thia- zolin-2-yl i-2-amino-6-benzoylbenzimidazol mit Äthyllithium umsetzt und die Zwischenverbindung anschliessend hydrolysiert.

Das Auftreten von Virenkrankheiten im oberen Atmungstrakt ist immens. Man hat geschätzt, dass allein in den Vereinigten Staaten von Amerika fast 1 Milliarde Fälle jährlich auftreten. In England durchgeführte Untersuchungen (Tyrell und Bynoe sowie Lancet, 16,1966) zeigen, dass 74% der Personen mit Erkältungen durch Rhinoviren infiziert sind. Es wurden bereits mehr als 80 Stämme von Rhinoviren identifiziert, und die Entwicklung einer einfachen Vakzine gegen Rhinoviren ist daher nicht möglich. Die Chemotherapie scheint somit ein besseres Mittel zu sein.
Die Fähigkeit chemischer Verbindungen zur Unterdrükkung des Virenwachstums in vitro kann leicht unter Verwendung eines Virus-Plättchensuppressionstests gezeigt werden, der dem von Siminoffin Applied Microbiology, 9(1)66 (1961) beschriebenen Test ähnlich ist.

Aus der US-PS 3 749717 sind 1 -Thiazolinyl-2-(heterocyclische)-benzimidazole bekannt, die sich als Anthelmintika und entzündungshemmende Mittel verwenden lassen.
Aus der US-PS 3 825 537 sind l-Thiazolinyl-2-amino-benzimidazole bekannt, die sich als Anthelmintilca und entzündungshemmende Mittel einsetzen lassen.
In der US-PS 3 833 574 wird ein Verfahren zur Herstellung von l-Thiazolinylbenzimidazolin-2-onen beschrieben, die als antiinflammatorische Mittel verwendbar sind.
Aus der DOS 2446266 sind 1 -Thiazolinyl-2-phenyl-benz- imidazole bekannt, die sich als Anthelmintika eignen.
Ziel der Erfindung ist es, ein Verfahren zur Herstellung von Verbindungen zu schaffen, mit denen sich das Wachstum von Viren, insbesondere Rhinoviren, Polioviren, Coxackie- Viren, Echoviren, Mengoviren und Influenzaviren unterdrücken bzw. bekämpfen lässt.

Der Ausdruck tautomeres Benzimidazol bedeutet ein
Benzimidazol-Reagens, das an jedem Stickstoffatom mit einem Wasserstoffatom substituiert sein kann. Das Benzimidazol-Reagens, das am Stickstoff unsubstituiert ist und eine Substituentengruppe in der 5-Stellung des Benzolmolekülteils enthält, besitzt eine entsprechende tautomere Form, die im Gleichgewicht mit ihrer Form ist, bei der der Substituent alternativ in der 6-Stellung steht. Das Isomerengemisch kann bezeichnet werden, indem man die möglichen wechselseitigen Stellungen als 5(6) bezeichnet.
Die folgenden Definitionen betreffen die verschiedenen Ausdrücke, die hier verwendet werden. Der Ausdruck C1-C3-Alkyl bedeutet geradkettige oder verzweigte Alkylreste mit 1 bis 3 Kohlenstoffatomen, nämlich Methyl, Äthyl, Propyl und Isopropyl. Die Angabe Cl-C3-Alkyl umfasst in ihrer Definition auch die Angabe Ci-C2-Alkyl . Die Angabe C1-C7-Alkyl bedeutet geradkettige oder verzweigtkettige Alkylreste mit 1 bis 7 Kohlenstoffatomen wie Methyl, Äthyl, Propyl, Isopropyl, Butyl, Isobutyl, sek.-Butyl, Pentyl, Isopentyl, Hexyl, Isohexyl, Heptyl, Isoheptyl, 2,4-Dimethyl-3-pentyl, tert.-Butyl und Neopentyl.
Die Angabe Thienyl bedeutet die an der 2- oder 3-Stellung gebundene Thiophengruppe.
Die Angabe C1-C4-Acyl bedeutet geradkettige oder verzweigtkettige Acylgruppen mit 1 bis 4 Kohlenstoffatomen wie Formyl, Acetyl, Propionyl, Butyryl und 2-Methylpropionyl.
Die Angabe C3-C6-Cycloalkyl bedeutet gesättigte Cycloalkylreste mit 3 bis 6 Kohlenstoffatomen wie Cyclopropyl, Methylcyclopropyl, Cyclobutyl, Cyclopentyl und Cyclohexyl. Die Angabe (C3-C6-Cycloalkyl)-methyl bedeutet eine durch Cycloalkyl, wie es oben bei der Angabe C3-C6-Cycloalkyl definiert wurde, substituierte Methylgruppe. Die Angabe (C3-C6-Cycloalkyl)-äthyl bedeutet Äthylgruppen, die durch oben definiertes Cycloalkyl substituiert sind, wobei die Substition vornehmlich am l-C-Atom vorliegt.
Das Cl-C7-Alkylmagnesiumhalogenid ist ein geeignetes Grignardreagens. Auf die Umsetzung damit erfolgt die Hydrolyse. Cl-C7-Alkyllithium ergibt ebenfalls ein Produkt ähnlich dem Grignardreagens. Die bevorzugten Lösungsmittel für das Alkylierungsverfahren sind inerte organische Lösungsmittel wie Tetrahydrofuran, aromatische Verbindungen wie Benzol oder Toluol und Äther wie Diäthyläther.
Der für die Umsetzung im allgemeinen verwendete Temperaturbereich liegt im Bereich von etwa 25"C bis zur Rückflusstemperatur des Lösungsmittels.
Die Ausgangsmaterialien der Formel (11) können wie im belgischen Patent Nr. 845 642 bzw. britischen Patent Nr.
1 562 823 beschrieben hergestellt werden.
Die Thiazolinyl-benzimidazol-Produkte können nach an sich bekannten Verfahren wie durch Filtration und Konzentration des Filtrats zur Kristallisationsinduzierung isoliert werden. Alternativ kann das Reaktionsgemisch zur Trockene eingedampft werden und der Rückstand mit einem geeigneten Lösungsmittel wie Aceton oder Methanol zur Abtren nung und Entfernung unlöslicher Materialien behandelt werden. Die das Produkt enthaltende Lösung kann zur Kristallisation des Produktes konzentriert oder unter Bildung eines zweiten Rückstandes eingedampft werden. Dieser Rückstand kann dann z.B. aus Methanol umkristallisiert werden. Die Benzimidazol-Verbindung wird gewöhnlich durch Abfiltrieren oder Zentrifugieren isoliert.
Die 5(6)-Isomeren können durch fraktionierte Kristallisation oder durch Säulenchromatographie getrennt werden.
Normalerweise kristallisiert das 6-Isomer zuerst aus der Lösung des Gemisches aus.
Die Thiazolinyl-benzimidazol-Verbindungen der Formel IA, worin R Cl-C4-Acyl bedeutet, werden durch Umsetzung von 1 -Thiazolinyl-2-amino-5(6)-subst.-benzimidazolen hergestellt, wie oben beschrieben, vorzugsweise mit den Anhydriden von Essigsäure, Propionsäure oder Buttersäure oder dem gemischten Anhydrid aus Ameisensäure und Essigsäure, Propionsäure oder Buttersäure. Falls R Formyl bedeutet, können sie durch Umsetzung des gemischten Anhydrids aus Ameisensäure und Essigsäure erhalten werden.
Die folgenden Beispiele erläutern die Herstellung der Verbindungen der Formel (I). Der Ausdruck m/e , der bei der Charakterisierung der Produkte verwendet wird, bedeutet das Masse-zu-Ladungs-Verhältnis der Ionen, die im Massenspektrum der Produkte auftreten. Im allgemeinen entsprechen die Werte den Molekulargewichten der Hauptpeaks.
Ausfüllrungsbeispiel I
4 g (12,4 mMol) 1 -(Thiazolin-2-yl)-2-amino-6-benzoylbenzimidazol werden zu 1000 ml Tetrahydrofuran unter Stickstoff bei einer Temperatur von - 10"C gegeben. Zu der Lösung gibt man tropfenweise unter Stickstoff 50 ml (1,6 Mol) n-Butyllithium in Hexan. Die Temperatur steigt auf -5"C. Die Temperatur der Lösung wird 1 h bei -5 bis - l00C gehalten. Die Lösung kann sich auf 25 C erwärmen und wird 3 h bei dieser Temperatur gehalten. Die Lösung wird in 11 Wasser gegossen, eingedampft, mit Tetrahydrofuran gewaschen und filtriert; man erhält 4,56 g l-(Thiazolin-2-yl) 2-amino-6-(a-hydroxy-a-n-butylbenzyl)-benzimidazol, Fp.
206 bis 209"C, als weissen Feststoff.
Ausführungsbeispiel 2
Man arbeitet, wie in Beispiel 1 beschrieben, verwendet jedoch 5,0 g (15,5 mMol) 1 -(Thiazolin-2-yl)-2-amino-6-benzoylbenzimidazol, 500 ml Tetrahydrofuran und 50 ml (2,1 Mol in Diäthyläther) Isopropylmagnesiumbromid; man erhält 1,24 g 1 -(Thiazolin-2-yl)-2-amino-6-(a-hydroxy-a-isopropylbenzyl)-benzimidazol, Fp. 212 bis 215"C.

Man arbeitet, wie in Beispiel 1 beschrieben, verwendet jedoch 2,0 g (6,22 mMol) l-(Thiazolin-2-yl)-2-amino-6-ben- zoyl-benzimidazol, 23 ml (1,25 Mol in Benzol) Äthyllithium und 500 ml Tetrahydrofuran; man erhält 2,5 g l-(Thia zolin-2-yl)-2-amino-6-(a-hydroxy-a-äthylbenzyl)-benzimi- dazol, Fp. 106 bis 208"C, m/e 352.
Untersuchungsmethoden
Nierenzellen (BSC-1) oder Helazellen (5-3) afrikanischer grüner Meerkatzen lässt man bei einer Temperatur von 37"C in 25 cm3-Falconkolben in Medium 199 wachsen, das 5% inaktiviertes fötales Rinderserum (FBS), Penicillin (150 Einheiten/l ml) und Streptomycin (150 Fg/ml) enthält. Sobald zusammenhängende Monoschichten gebildet sind, entfernt man das überstehende Wachstumsmedium und gibt in jeden Kolben 0,3 ml einer geeigneten Virusverdünnung (Echovirus, Mengovirus, Coxsachievirus, Poliovirus und Rhinovirus).
Nach einstündiger Absorption bei Zimmertemperatur überdeckt man die mit den Viren infizierte Zellschicht mit einem Medium, das einen Teil 1 %ges Ionagar Nr.2 und einen Teil doppelstarkes Medium 199 mit FBS, Penicillin und Streptomycin, das Arzneimittel in Konzentrationen von 100, 50,25, 12, 6, 3 und 0 ug/ml aufweist, enthält. Der keinen Wirkstoff enthaltende Kolben dient als Kontrolle für die Untersuchung. Die Herstellung entsprechender Stammlösungen der Thiazolinyl-benzimidazolverbindungen der Formel (I) erfolgt in Dimethylsulfoxid bei einer Konzentration von 104 ug/ml.
Die Kolben werden 72 h bei einer Temperatur von 37"C mit Polio-, Coxsackie-, Echo- und Mengoviren und 120h bei 32"C mit Rhinoviren inkubiert. Die Influenzaviren Ann Carbor, Maryland B, Massachusetts B, Hong Kong A, Pr-8a und Taylor C (Arten A, B) werden 72 h bei 37"C unter Verwendung von MDCK-Zellen (Madin-Darby-Hundenierenzellen) inkubiert. Plättchen sind an denjenigen Stellen zu sehen, an denen das Virus die Zellen infiziert hat und sich reproduziert. Zur Inaktivierung des Virus und zur Fixierung der Zellschicht auf der Oberfläche des Kolbens versetzt man jeden Kolben mit einer Lösung aus 10%igem Formalin und 2% Natriumacetat. Die Virusplättchen werden unabhängig von ihrer Grösse nach Anfärben der umgebenden Zellflächen mit Kristallviolett gezählt.
Die Plättchenauszählung vergleicht man bei jeder Wirkstoffkonzentration mit der Kontrollauszählung. Die Aktivität des jeweiligen Wirkstoffs wird in Form der prozentualen Plättchenreduktion oder der prozentualen Inhibierung ausgedrückt. Als Mass für die Aktivität lässt sich wahlweise auch die durch das Symbol Iso angegebene Wirkstoffkonzentration verwenden, die die Plättchenbildung um 50% inhibiert.
Die bei den obigen Versuchen erhaltenen Ergebnisse sind in Form der Inhibierung von Poliovirus Typ I ausgedrückt, da dieses Virus leicht wächst und zu übereinstimmenden Ergebnissen führt. Die Aktivität der Verbindungen der Formel (I) wird gegenüber anderen Viruskulturen wie Coxsackie (A9, A21, B5), Echovirus (Stämme 1-4), Mengo, Rhinovirus (25 Stämme), Polio (Typ I, II, III) und Influenzaviren wie Ann Arbor, Maryland B, Massachusetts B, Hong Kong A, Pr-8A und Taylor C (Typen A, B) bestimmt. Die Versuchsergebnisse für verschiedene Thiazolinyl-benzimidazol-Verbindungen sind in der folgenden Tabelle I zusammengefasst.
In der Tabelle ist die Spalte 1 die Beispielnummer der verschiedenen chemischen Beispiele aufgeführt; in der Spalte 2 ist das Isomer in der 5(6)-Stellung angegeben, und in den Spalten 3 bis 10 ist diejeweilige prozentuale Virsuplättchen-Reduktion bei Wirkstoffverdünnungen von 0,75 bis 100 Rg/ml aufgeführt.

Die Thiazolinylbenzimidazol-Verbindungen werden sowohl als reine Verbindungen als auch als Isomerengemische geprüft. Beide Isomeren inhibieren das Viruswachstum; das 6-Isomer ist im allgemeinen aktiver als das 5Isomer.
Die Verbindungen der Formel (I) unterdrücken im allgemeinen das Wachstum verschiedener Viren, wenn sie zu dem Medium zugegeben, indem das Virus gezüchtet wird. Die Verbindungen der Formel (1) können daher in wässrigen Lösungen, bevorzugt mit einem oberflächenaktiven Mittel, zur Dekontaminierung von Oberflächen verwendet werden, auf denen Polio-, Coxsackie-, Rhino- und Influenzaviren vorhanden sind, wiez.B. Krankenhaus-Glaswaren, Krankenhausarbeitsflächen und ähnliche Flächen, die zur Herstellung von Nahrungsmitteln verwendet werden.
Die erfindungsgemäss erhaltenen Verbindungen können warmblütigen Tieren und Menschen in einer Dosis von 1 bis 300 mg/kg Tierkörpergewicht oral verabreicht werden. Die Verabreichung kann bedarfsweise periodisch wiederholt werden. Im allgemeinen wird die antivirale Verbindung alle 4 bis 6 Stunden verabreicht.
Bevorzugt werden die Verbindungen der Formel (I) zusammen mit einem oder mehreren, für die besondere Art der Verabreichung geeigneten Adjuvantien verwendet. Im Falle der oralen Verabreichung wird die Verbindung mit pharmazeutischen Verdünnungsmitteln oder Trägern wie Lactose, Saccharose, Stärkepulver, Cellulose, Talk, Magnesiumstearat, Magnesiumoxid, Calciumsulfat Akazienpulver, Gelatine, Natriumalginat, Natriumbenzoat und Stearinsäure modifiziert. Zubereitungen dieser Art lassen sich für entsprechende Verabreichungszwecke als Tabletten oder eingeschlossen in Kapseln formulieren. Darüberhinaus können die erfindungsgemäss hergestellten Verbindungen auch parenteral verabreicht werden.
Die Verbindungen können ebenfalls mit einer Flüssigkeit vermischt und in Form von Nasentropfen verabreicht werden oder als Intranasalspray verwendet werden.

The occurrence of viral diseases in the upper respiratory tract is immense. It has been estimated that nearly 1 billion cases occur annually in the United States alone. Studies conducted in England (Tyrell and Bynoe and Lancet, 16.1966) show that 74% of people with colds are infected with rhinoviruses. More than 80 strains of rhinoviruses have been identified, and development of a simple rhinovirus vaccine is therefore not possible. Chemotherapy appears to be a better remedy.
The ability of chemical compounds to suppress virus growth in vitro can easily be demonstrated using a virus platelet suppression test similar to that described by Siminoffin Applied Microbiology, 9 (1) 66 (1961).

The aim of the invention is to provide a method for producing compounds with which the growth of viruses, in particular rhinoviruses, polioviruses, coxackie viruses, echoviruses, mengoviruses and influenza viruses can be suppressed or combated.

Thiazolinyl-benzimidazole compounds which come under the general formula (I) are e.g. the following connections:
I - (thiazolin-2-yl) -2-amino-5 (6) - (a-hydroxy-a-methylbenzyl) benzimidazole, I - (thiazolin-2-yl) -2-amino-5 (6) - (a-Hydroxy-an-hexyl-benzyl) -benzimidazole, and 1 - (thiazolin-2-yl) -2-amino-5 (6) - [a-hydroxy-a- (2,4-dimethyl) -3-pentyl) benzyl] benzimidazole.
The term tautomeric benzimidazole means a
Benzimidazole reagent, which may be substituted with a hydrogen atom on each nitrogen atom. The benzimidazole reagent, which is unsubstituted on nitrogen and contains a substituent group in the 5-position of the benzene moiety, has a corresponding tautomeric form which is in equilibrium with its form in which the substituent is alternatively in the 6-position. The mixture of isomers can be designated by designating the possible mutual positions as 5 (6).
The following definitions refer to the various terms used here. The term C1-C3-alkyl means straight-chain or branched alkyl radicals having 1 to 3 carbon atoms, namely methyl, ethyl, propyl and isopropyl. Cl-C3-alkyl also includes Ci-C2-alkyl in its definition. C1-C7-alkyl means straight-chain or branched-chain alkyl radicals having 1 to 7 carbon atoms, such as methyl, ethyl, propyl, isopropyl, butyl, isobutyl, sec-butyl, pentyl, isopentyl, hexyl, isohexyl, heptyl, isoheptyl, 2,4 -Dimethyl-3-pentyl, tert-butyl and neopentyl.
Thienyl means the thiophene group attached at the 2- or 3-position.
C1-C4-acyl means straight-chain or branched-chain acyl groups having 1 to 4 carbon atoms, such as formyl, acetyl, propionyl, butyryl and 2-methylpropionyl.
C3-C6-cycloalkyl means saturated cycloalkyl radicals having 3 to 6 carbon atoms, such as cyclopropyl, methylcyclopropyl, cyclobutyl, cyclopentyl and cyclohexyl. The statement (C3-C6-cycloalkyl) -methyl means a methyl group substituted by cycloalkyl as defined above for the statement C3-C6-cycloalkyl. The statement (C3-C6-cycloalkyl) -ethyl means ethyl groups which are substituted by cycloalkyl defined above, the substitution being predominantly present on the 1-C atom.
The Cl-C7 alkyl magnesium halide is a suitable Grignard reagent. The hydrolysis then takes place on the reaction. Cl-C7 alkyl lithium also gives a product similar to the Grignard reagent. The preferred solvents for the alkylation process are inert organic solvents such as tetrahydrofuran, aromatic compounds such as benzene or toluene and ethers such as diethyl ether.
The temperature range generally used for the reaction is in the range from about 25 ° C. to the reflux temperature of the solvent.
The starting materials of formula (11) can be prepared as described in Belgian Patent No. 845 642 or British Patent No.
1 562 823 can be produced.
The thiazolinyl-benzimidazole products can be isolated by methods known per se, such as by filtration and concentration of the filtrate to induce crystallization. Alternatively, the reaction mixture can be evaporated to dryness and the residue treated with a suitable solvent such as acetone or methanol to remove and remove insoluble materials. The solution containing the product can be concentrated to crystallize the product or evaporated to form a second residue. This residue can then e.g. be recrystallized from methanol. The benzimidazole compound is usually isolated by filtration or centrifugation.
The 5 (6) isomers can be separated by fractional crystallization or by column chromatography.
The 6-isomer normally crystallizes out of the solution of the mixture first.
The thiazolinyl-benzimidazole compounds of the formula IA, in which R is Cl-C4-acyl, are prepared by reacting 1-thiazolinyl-2-amino-5 (6) -subst. -Benzimidazoles, as described above, preferably with the anhydrides of acetic acid, propionic acid or butyric acid or the mixed anhydride of formic acid and acetic acid, propionic acid or butyric acid. If R is formyl, they can be obtained by reacting the mixed anhydride of formic acid and acetic acid.
The following examples illustrate the preparation of the compounds of formula (I). The term m / e used in the characterization of the products means the mass-to-charge ratio of the ions that appear in the mass spectrum of the products. In general, the values correspond to the molecular weights of the main peaks.
Completion example I
4 g (12.4 mmol) of 1 - (thiazolin-2-yl) -2-amino-6-benzoylbenzimidazole are added to 1000 ml of tetrahydrofuran under nitrogen at a temperature of -10 ° C. The procedure is as described in Example 1, but 5.0 g (15.5 mmol) of 1 - (thiazolin-2-yl) -2-amino-6-benzoylbenzimidazole, 500 ml of tetrahydrofuran and 50 ml (2.1 mol in diethyl ether) isopropyl magnesium bromide; 1.24 g of 1 - (thiazolin-2-yl) -2-amino-6- (a-hydroxy-a-isopropylbenzyl) benzimidazole are obtained, mp 212 to 215 ° C.
EXAMPLE 3
The procedure is as described in Example 1, but 2.0 g (6.22 mmol) of 1- (thiazolin-2-yl) -2-amino-6-benzoyl-benzimidazole, 23 ml (1.25 mol in benzene) ethyl lithium and 500 ml tetrahydrofuran; 2.5 g of l- (thiazolin-2-yl) -2-amino-6- (a-hydroxy-a-ethylbenzyl) -benzimizazole are obtained, mp. 106 to 208 "C, m / e 352.
Examination methods
Kidney cells (BSC-1) or hela cells (5-3) African green vervet monkeys are grown at a temperature of 37 "C in 25 cm3 falcon flasks in medium 199 containing 5% inactivated fetal bovine serum (FBS), penicillin (150 units / 1 ml) and streptomycin (150 Fg / ml) As soon as continuous monolayers are formed, the supernatant growth medium is removed and 0.3 ml of a suitable virus dilution (echovirus, mengovirus, coxsachievirus, poliovirus and rhinovirus) is added to each flask.
After absorption for one hour at room temperature, the cell layer infected with the viruses is covered with a medium comprising a part of 1% total ion agar No. 2 and a part of double-strength medium 199 with FBS, penicillin and streptomycin, the drug in concentrations of 100, 50.25 , 12, 6, 3 and 0 µg / ml. The flask containing no active ingredient serves as a control for the examination. Corresponding stock solutions of the thiazolinyl-benzimidazole compounds of the formula (I) are prepared in dimethyl sulfoxide at a concentration of 104 μg / ml.
The flasks are incubated for 72 hours at a temperature of 37 ° C. with polio, coxsackie, echo and mengoviruses and for 120 hours at 32 ° C. with rhinoviruses. Influenza viruses Ann Carbor, Maryland B, Massachusetts B, Hong Kong A, Pr-8a and Taylor C (species A, B) are incubated for 72 hours at 37 "C using MDCK cells (Madin-Darby dog kidney cells). Platelets can be seen in those places where the virus has infected and reproduced the cells. To inactivate the virus and to fix the cell layer on the surface of the flask, a solution of 10% formalin and 2% sodium acetate is added to each flask. The virus platelets are counted regardless of their size after staining the surrounding cell areas with crystal violet.
The platelet count is compared to the control count at each drug concentration. The activity of the respective active ingredient is expressed in the form of the percentage reduction in platelets or the percentage inhibition. The active substance concentration indicated by the symbol Iso, which inhibits the formation of platelets by 50%, can optionally also be used as a measure of the activity.
The results obtained in the above experiments are expressed in terms of the inhibition of type I poliovirus, since this virus grows easily and gives consistent results. The activity of the compounds of formula (I) is compared to other virus cultures such as Coxsackie (A9, A21, B5), echovirus (strains 1-4), Mengo, rhinovirus (25 strains), polio (type I, II, III) and influenza viruses as determined by Ann Arbor, Maryland B, Massachusetts B, Hong Kong A, Pr-8A and Taylor C (types A, B). The test results for various thiazolinyl-benzimidazole compounds are summarized in Table I below.
In the table, column 1 shows the sample number of the various chemical samples; column 2 shows the isomer in the 5 (6) position, and columns 3 to 10 show the percentage viral platelet reduction at drug dilutions of 0.75 to 100 Rg / ml.

The thiazolinylbenzimidazole compounds are tested both as pure compounds and as mixtures of isomers. Both isomers inhibit virus growth; the 6 isomer is generally more active than the 5 isomer.
The compounds of formula (I) generally suppress the growth of various viruses when added to the medium by growing the virus. The compounds of formula (1) can therefore be used in aqueous solutions, preferably with a surface-active agent, for the decontamination of surfaces on which polio, coxsackie, rhino and influenza viruses are present, such as e.g. Hospital glassware, hospital work surfaces and similar surfaces used in the manufacture of food.
The compounds obtained according to the invention can be administered orally to warm-blooded animals and humans in a dose of 1 to 300 mg / kg of animal body weight. The administration can be repeated periodically if necessary. Generally, the antiviral compound is administered every 4 to 6 hours.
The compounds of the formula (I) are preferably used together with one or more adjuvants which are suitable for the particular mode of administration. In the case of oral administration, the compound is modified with pharmaceutical diluents or carriers such as lactose, sucrose, starch powder, cellulose, talc, magnesium stearate, magnesium oxide, calcium sulfate, acacia powder, gelatin, sodium alginate, sodium benzoate and stearic acid. Preparations of this type can be formulated as tablets or enclosed in capsules for appropriate administration purposes. In addition, the compounds produced according to the invention can also be administered parenterally.
The compounds can also be mixed with a liquid and administered in the form of nasal drops or used as an intranasal spray.
