PATENTANSPRÜCHE
1. Ticrfuttermischung auf Basis von Kohlenhydraten, Eiweiss und Fetten, gekennzeichnet durch einen Gehalt an 5-20 ppm des Antibiotikums Zink-Bacitracin und einer solchen Menge an proteolytischen Enzymen, dass eine enzymatische Aktivität von 0,05-2,5 mTU/g vorliegt.
2. Tierfuttermischung nach Anspruch 1, dadurch gekennzeichnet, dass die proteolytischen Enzyme saure Proteasen mit einem Wirkungsspektrum im Bereich von pH Wert 2,5-6,5 sind.
3. Tierfuttermischung nach Anspruch 1 oder 2, dadurch gekennzeichnet, dass sie noch weitere Zusatzstoffe enthält.
Hochleistungstierfutter, wie sie heute üblicherweise in der Intensiv-Tierhaltung verwendet werden, enthalten eine Reihe von Zusatzstoffen mit prophylaktischer und/oder nutritiver Wirkung. Hierzu zählen u. a. Antibiotika und Enzyme. Während sich die Verwendung von Antibiotika bereits in Mischfuttern aller Typen durchgesetzt hat, werden Enzyme in der Praxis noch nicht in grösserem Umfange verwendet.
Die in der modernen Intensiv-Tierhaltung verwendeten Futtermischungen sind im allgemeinen in allen Nährstoffkomponenten so weit optimiert, dass eine weitere Verbesserung nicht ohne weiteres möglich erscheint. Überraschen derweise wurde jedoch gefunden, dass auch optimal zusammengesetzte Hochleistungsfutter hinsichtlich der Futterverwertung und/oder des damit erzielbaren Gewichtszuwachses noch verbessert werden können, wenn man diesen Futtern bestimmte Antibiotika zusammen mit proteolytischen Enzymen zusetzt.
Gegenstand der Erfindung ist dementsprechend eine Tierfuttermischung auf Basis von Kohlenhydraten, Eiweiss, Fetten und gegebenenfalls üblichen Zusatzstoffen, gekennzeichnet durch einen Gehalt von 5-20 ppm des Antibiotikums Zink-Bacitracin und einer solchen Menge an proteolytischen Enzymen, dass eine enzymatische Aktivität von 0,05-2,5 mTU/g vorliegt.
Das Antibiotikum Zink-Bacitracin gehört zur Gruppe der Polypetid-Antibiotika. Es handelt sich bei den Handelspräparaten um Mischungen verschiedener Polypeptide mit einem Zinkgehalt bis zu 20 Gew.-%. Die Ausgangsform Bacitracin wird in bekannter Weise von Stämmen des Bacillus licheniformis produziert.
Geeignete proteolytische Enzyme sind vor allem durch Züchten von Mikroorganismen und Abtrennen aus den Nährlösungen erhältlich. Die Verfahren hierzu sind bekannt.
Beispielsweise werden proteolytische Enzyme aus Bacillus licheniformis, Bacillus natta, Bacillus subtilis usw. verwendet.
Besonders bevorzugt werden saure Proteasen, z. B. aus Aspergillus niger oder aus Rhizopus rhizopodiformis entsprechend der DE Patentanmeldung P 2 528 490.6. Derartige Proteasen haben ein besonders breites Wirkungsspektrum im schwach sauren Bereich von pH-Wert 2,5-6,5.
Handelsübliche Futtermischungen sind für die speziellen Bedürfnisse der einzelnen Tierarten möglichst optimal zusammengestellt. Die Grundlage bilden Kohlenhydrate vor allem aus Getreidekomponenten, Mais od. dgl. Als Eiweissträger dienen in erster Linie Sojaschrot, Fischmehl, Tierkörpermehl, Kleie und dgl. Fehlende Aminosäuren, z. B. Methionin, können zusätzlich zugegeben werden. Fette werden in Form von pflanzlichen oder tierischen Fetten oder Abfallfetten beigefügt. Zum Körperaufbau werden ferner Salze, wie Dikalziumphosphat, Kalziumkarbonat, Kochsalz zugesetzt. Gegebenenfalls wird die Futtermischung durch Zugabe von Spurenelementen, Vitaminen, Ballaststoffen usw. abgerundet.
Auch fermentativ hergestellte Substanzen, wie Einzellerproteine aus Erdölfraktionen oder Alkoholen, diverse Hefen, Algeneiweiss oder sonstige, eventuell auch aus Abfallstoffen gewonnene Substanzen, können z.T. in erheblichem Masse Bestandteil der Futterrezeptur sein.
Die erfindungsgemässe Tierfuttermischung enthält neben den üblichen jeweils für eine bestimmte Tierart abgestimmten Bestandteilen zusätzlich 5-20 ppm, vorzugsweise 1015 ppm, des genannten Antibiotikums und proteolytische Enzyme in einer solchen Menge, dass die enzymatische Aktivität 0,05-2,5 mTU/g, vorzugsweise 0,2-0,5 mTU/g beträgt. Insbesondere wird eine Kombination aus Zink-Bactracin und einer sauren Pilz-Protease bzw. Proteasenmischung eingesetzt. Der damit erzielte Zuwachs bzw. die Verbesserung der Futterverwertung beträgt bis zu 4% im Vergleich zu entsprechenden, lediglich Antibiotikum enthaltenden Kontrollmischungen.
Die wirksame Kombination aus Zink-Bacitracin und Enzymen bewährt sich vor allem in Futter für Masthähnchen.
Sie kann jedoch auch für alle anderen Tierarten nützlich sein, bei denen die Verwendung von Zink-Bacitracin alleine bereits von Vorteil ist, z. B. in der Schweinemast und in Legehennenfutter.
Das Antibiotikum wird zweckmässig in Form einer Vormischung dem Futter zugesetzt, z.B. gebunden an Sojaextraktionsschrot. Bei Verwendung von mehlförmigen Futterzusammensetzungen wird auch die Enzymkomponente als Vormischung appliziert. Als Trägersubstanz fungiert eine beliebige Futterkomponente, z. B. ebenfalls Sojaschrot. Bei Verwendung dampfpelletierten Futters muss die Zumischung der Enzyme in geeignet stabilisierter Form erfolgen, um eine Desaktivierung durch Feuchtigkeit und Hitze während des Pelletierens zu verhindern. Ein derartiges Verfahren ist z. B. Gegenstand der DE Patentanmeldung P 2 602 260.6.
Zur Bestimmung der Enzymeinheit (TU) wird die proteolytische Aktivität der Protease nach dem bekannten Prinzip von Anson ermittelt: eine geeignet verdünnte Menge Enzymlösung wird bei 40"C 20 Minuten lang mit einem gleichen Volumen einer 1,2gewichtsprozentigen Caseinlösung inkubiert, wobei diese 0,6 Gew.-% Milchsäure, 6 Mol Harnstoffund 0,1 Mol Zitronen- oder Essigsäure enthält. Der pH-Wert der Caseinlösung wird durch Zusatz von 2n Natronlauge auf 4,5 eingestellt. Nach der Inkubation wird im Volumenverhältnis 1:1 mit 0,4n Trichloressigsäure versetzt, der sich bildende Niederschlag von unverdautem Casein abfiltriert, und im Filtrat die beim Abbau entstandenen Protein-Spaltstücke können nach einer beliebigen Eiweissbestimmungsmethode ermittelt werden. Geeignet hierfür ist z.
B. das von Layne in Methode of Enzymology 3 (1957) Seiten 448ff. beschriebene Verfahren.
Für jede Messprobe muss ein Blindwert angefertigt werden, bei dem zuerst Trichloressigsäure und dann Caseinlösung zugesetzt wird. Dieser Blindwert gibt neben dem Reagenzien-Leerwert den Anteil an niedermolekularen Peptiden an, der bereits vor der Verdauung in der Enzymlösung vorhanden ist. Die Differenz zwischen Haupt- und Blindwert wird bei der angegebenen Methode dann mit der Extinktion verglichen, die eine bestimmte Menge Tyrosin bei dieser Bestimmung liefert. Diese Menge Tyrosin ist dann ein Mass für die proteolytische Aktivität des vorliegenden Enzyms: eine Enzymeinheit (TU) ist diejenige Menge Enzym, die aus einer Caseinlösung in einer Minute Spaltprodukte freisetzt, die die gleiche Extinktion besitzen, wie eine 1 M Tyrosinlösung. Ublich ist die Angabe in mTU = 10-3 TU.

Beispiel
160 männliche Masteintagsküken der Herkunft Lohmann wurden in Batteriehaltung 6 Wochen lang jeweils mit gleichartig zusammengesetzten Futtermischungen, jedoch a) ohne Zusatz von Antibiotikum oder Enzym b) mit Zusatz von 10 ppm Zink-Bacitracin c mit Zusatz von 10 ppm Zink-Bacitracin und 0,3 mTU/g saure Proteasen aus Aspergilius niger gefüttert und nach Abschluss des Versuches das Endgewicht und die Futterverwertung bestimmt. Unter Futterverwertung wird das Verhältnis Futterverbrauch : Zuwachs verstanden.

The feed mixtures used in modern intensive animal husbandry are generally optimized in all nutrient components to such an extent that further improvement does not appear to be possible without further ado. Surprisingly, however, it was found that even optimally composed high-performance feed can still be improved with regard to feed conversion and / or the weight gain that can be achieved by adding certain antibiotics to these feeds together with proteolytic enzymes.
The invention accordingly relates to an animal feed mixture based on carbohydrates, protein, fats and, if appropriate, conventional additives, characterized by a content of 5-20 ppm of the antibiotic zinc bacitracin and an amount of proteolytic enzymes such that an enzymatic activity of 0.05 -2.5 mTU / g is present.
The antibiotic zinc bacitracin belongs to the group of polypeptide antibiotics. The commercial preparations are mixtures of different polypeptides with a zinc content of up to 20% by weight. The starting form bacitracin is produced in a known manner from strains of Bacillus licheniformis.
Suitable proteolytic enzymes can be obtained primarily by growing microorganisms and separating them from the nutrient solutions. The procedures for this are known.
For example, proteolytic enzymes from Bacillus licheniformis, Bacillus natta, Bacillus subtilis, etc. are used.
Acidic proteases, e.g. B. from Aspergillus niger or from Rhizopus rhizopodiformis according to DE patent application P 2 528 490.6. Such proteases have a particularly broad spectrum of activity in the weakly acidic range of pH 2.5-6.5.
Commercial feed mixtures are optimally put together for the special needs of the individual animal species. The basis is formed by carbohydrates mainly from cereal components, maize or the like. Soybean meal, fish meal, animal meal, bran and the like serve primarily as protein carriers. Missing amino acids, e.g. B. methionine, can also be added. Fats are added in the form of vegetable or animal fats or waste fats. Salts such as dicalcium phosphate, calcium carbonate and common salt are also added to the body. If necessary, the feed mixture is rounded off by adding trace elements, vitamins, fiber, etc.
Substances produced by fermentation, such as single-cell proteins from petroleum fractions or alcohols, various yeasts, algae protein or other substances that may also have been obtained from waste materials, can in part. to a large extent be part of the feed formula.
The animal feed mixture according to the invention contains, in addition to the usual constituents which are in each case matched to a specific animal species, an additional 5-20 ppm, preferably 1015 ppm, of the antibiotic mentioned and proteolytic enzymes in such an amount that the enzymatic activity 0.05-2.5 mTU / g, is preferably 0.2-0.5 mTU / g. In particular, a combination of zinc bactracin and an acidic fungal protease or protease mixture is used. The growth or improvement in feed conversion achieved in this way is up to 4% compared to corresponding control mixtures containing only antibiotics.
The effective combination of zinc bacitracin and enzymes has proven itself particularly in feed for broilers.
However, it can also be useful for all other animal species where the use of zinc bacitracin alone is already beneficial, e.g. B. in pig fattening and laying hen feed.
The antibiotic is conveniently added to the feed in the form of a premix, e.g. bound to soybean meal. When using flour-like feed compositions, the enzyme component is also applied as a premix. Any feed component, e.g. B. also soybean meal. When using steam-pelleted feed, the enzymes must be added in a suitably stabilized form to prevent deactivation by moisture and heat during pelleting. Such a method is e.g. B. Subject of DE patent application P 2 602 260.6.
To determine the enzyme unit (TU), the proteolytic activity of the protease is determined according to the known principle of Anson: a suitably diluted amount of enzyme solution is incubated at 40 ° C. for 20 minutes with an equal volume of a 1.2% by weight casein solution, this being 0.6 % By weight of lactic acid, 6 mol of urea and 0.1 mol of citric or acetic acid. The pH of the casein solution is adjusted to 4.5 by adding 2N sodium hydroxide solution. After the incubation, the volume ratio is 1: 1 with 0.4N Trichloroacetic acid is added, the precipitate formed is filtered off from undigested casein, and the protein cleavage pieces formed during the breakdown can be determined by any protein determination method.

A blank value must be prepared for each measurement sample, in which first trichloroacetic acid and then casein solution are added.
