Description
Die Erfindung betrifft ein lyophilisiertes Inokulationsmaterial für die Erzeugung eines Schimmelrasens auf Lebensmittel sowie ein Verfahren zur Herstellung von Inokulationsmaterial. The invention relates to a lyophilized inoculation material for producing a mold lawn on food and a method for producing inoculation material.
Materialien zur Übertragung von Schimmelpilzkulturen (Inokulationsmaterialien) auf natürliche oder synthetische Substrate enthalten als Bestandteile die Sporen der Pilze und/ oder Teile des Mycels (Hyphen). Im Interesse eines möglichst hochwirksamen Präparates muss die Überführung der ursprünglichen Kulturen in haltbare technisch verwertbare Formen derart geführt werden, dass die Zahl der keimfähigen Anteile besonders hoch ist. Aus Gründen der Haltbarkeit und auch aus finanziellen Überlegungen (Transportkosten) empfiehlt sich bei der Herstellung grösserer Mengen von Inokulationsmaterialien die lyophilisierte Form. Sie besitzt jedoch wegen der besonderen Struktur der Schimmelpilzsporen und des Mycels erhebliche praktische Nachteile. Bei Inokulationsmaterial für technische Zwecke, bei dem es darauf ankommt, z.B. die Oberfläche von Nahrungsmitteln (Wurst, Käse etc.) möglichst schnell mit einem dichten Rasen eines erwünschten Schimmelpilzes zu überziehen, benötigt man ein Aussaatmaterial, dessen Keimdichte gegenüber dem Material beispielsweise für Stammsammlungen um den Faktor 100 oder 1000 grösser sein müssen. Nach diesen Vorbemerkungen ist es klar, dass eine derartige Erhöhung der Keimdichte und andererseits der Dispersion nur erreicht werden kann, wenn das Trocknungsverfahren erheblich verbessert wird. Stellt man auf dem Wege der üblichen Lyophilisierung aus ganzen Pilzkulturen oder Abschwemmungen Trockenpräparate her, so gelingt es nicht, durch Verwendung von Flüssigkeiten, den ursprünglichen Dispersionsgrad der Pilzkulturen und ihrer Sporen wieder herzustellen. Verwendet man so beschriebene Inokulationsmaterialien zur Herstellung von wässerigen Aufschwemmungen, in denen z.B. Rohwürste durch Tauchen beimpft werden sollen, so stellte man fest, dass die Verteilung der Keime auf der Oberfläche der Würste sehr ungleich ausfällt. Man erhält punktförmiges Wachstum und daneben Stellen, an denen keinerlei Wachstum stattgefunden hat. Derartige Würste sind vom Aussehen nicht ansprechend und überdies können sich an den unbewachsenen Stellen Fremdschimmel ansiedeln, was dem eigentlichen Sinn einer gezielten Beimpfung naturgemäss widerspricht. Es muss also erreicht werden, dass nach der Lyophilisierung und Wiederanschwemmung in Wasser Präparate hoher Gleichmässig-keit erzielt werden können, welche beim Tauchen die Oberfläche der beimpften Materialien mit einem gleichmässigen Pilzrasen überziehen. Untersucht man mikroskopisch ein lyophilisiertes Schimmelpilz-Präparat, so stellt man fest, dass sowohl die Sporen als auch die Hyphen in verklumpter Form vorliegen. Dies ist leicht erklärlich, da die fadenförmige Struktur der Hyphen eine «Verfilzung» erleichtert, zum anderen die Oberfläche der Sporen mit feinen Protuberanzen besetzt ist. Letztere sollen in der Natur das Festhaften der Sporen auf einer geeigneten Unterlage begünstigen. Während des Lyophilisierens führen die Protuberanzen dazu, dass sich Sporen und Hyphen fest miteinander verbinden. Diese Verbindung kann durch die nachfolgende Aufschwemmung mit Wasser zur Anwendung als Tauchflüssigkeit nicht gelöst werden. Es bleiben Zusammenklumpungen, die auf der zu beimpfenden Fläche eben nur «Inseln» bilden. Wollte man mit einem derartigen Inokulationsmaterial einen gleichmässigen Pilzrasen erzielen, so müsste die Keimkonzentration (Sporen) unverhältnismässig hoch gewählt werden, was nicht allein aus preislichen Gründen unmöglich ist. Nach den oben dargelegten wissenschaftlichen Erkenntnissen lässt sich also ein hoch aktives und gleichmässigen Schimmelrasen erzeugendes Impfmaterial nur herstellen, wenn es gelingt, die Aggregation der Pilzsporen untereinander und mit dem Mycelgeflecht zu verhindern. Dies kann dadurch geschehen, dass man solche Stoffe zusetzt, die beim Trocknungsvorgang die als Kolloide zu betrachtenden Schimmelpilzsporen und Hyphen mit einem Schutzkolloid überziehen, bzw. die Dispergierung der Sporen beim Auflösevorgang vollständig bewirkt. Diese Stoffe können auch bereits Bestandteile des zur Anzüchtung benutzten Nährmediums sein, das dann mit verarbeitet werden muss.
