Erfindungsgemäss gelangt man zu den neuen Verbindungen der Formel I, indem man ein reaktionsfähiges, funktionelles Derivat einer Säure der Formel II,

Die erfindungsgemässe Umsetzung gemäss Verfahren stellt eine Kondensationsreaktion für Amide dar.
Aus der Ergotchemie ist bekannt, welche reaktionsfähigen funktionellen Derivate der 9,10-Lyserg- bzw. 9,10-Dihydroly
sergsäure bei solchen Umsetzungen verwendet und unter welchen Bedingungen diese Umsetzungen durchgeführt werden können.
Als reaktionsfähige funktionelle Derivate einer Säure der Formel II kann beispielsweise das Säurechloridhydrochlorid, das Säureazid oder ein gemischtes Anhydrid einer Säure der Formel II mit Schwefelsäure oder Trifluoressigsäure eingesetzt werden.
Vorzugsweise verwendet man die reaktionsfähigen funktionellen Derivate, die erhältlich sind durch Umsetzung einer Säure der Formel II mit einem aus einem Chlorierungsmittel und einem N-di(nieder)alkylsubstituierten Säureamid einer aliphatischen Carbonsäure mit 1 bis 3 Kohlenstoffatomen gebildeten Amidchlorid. Als Chlorierungsmittel können beispielsweise Thionylchlorid, Phosgen oder Oxalylchlorid eingesetzt werden.
Für die Bildung des benötigten Amidchlorids geeignete Säureamide sind beispielsweise Dimethylformamid oder Dimethylacetamid.
Das Verfahren wird zweckmässig in einem organischen Lösungsmittelgemisch durchgeführt. Geeignete Lösungsmittel sind z.B. Chloroform, Methylenchlorid, Dimethylformamid oder Acetonitril.
Man arbeitet vorzugsweise in Gegenwart eines Kondensationsmittels. Zweckmässig werden tertiäre organische Basen, beispielsweise Triäthylamin, vorzugsweise aber Pyridin und Homologe eingesetzt.
Die Umsetzung kann bei Temperaturen zwischen - 30 bis + 20 durchgeführt werden. Man kann unter Normaldruck arbeiten.
Die Verbindungen der Formel I können in freier Form als Base oder in Form von Additionssalzen mit Säuren vorliegen.
Aus den freien Basen lassen sich in bekannter Weise Säureadditionssalze herstellen und umgekehrt.
Die Verbindungen der Formel III sind neu.
In einer ersten Stufe werden hierbei Verbindungen der Formel IV,

Die Reduk tion erfolgt vorzugsweise mit Lithiumaluminiumhydrid in einem unter den Reaktionsbedingungen inerten Lösungsmittel, vor zugsweise cyclische oder offenkettige Äther, wie z.B. Tetrahy drofuran, Dioxan, Diäthyläther, Diglyme oder Dibutyläther.
Die Reduktion erfolgt vorzugsweise bei Temperaturen zwischen etwa -20 und +20" .

worin R, R1 und R2 obige Bedeutung haben, kann analog zu bekannten Methoden unter Verwendung einer Verbindung der Formel V und S( + )-R-Benzyloxy-malonsäureäthylesterchlorid, worin R obige Bedeutung hat, erfolgen.
Die weiteren Synthesestufen sind im Prinzip Teilstufen der Curtius Methode bzw. Modifikation davon und erfolgen unter Verwendung von Verbindungen der Formel VI. Die Ester der Formel VI werden analog zu bekannten Methoden in der ihnen entsprechenden freien Säure über das Carbonsäurechlorid in das Säureazid überführt, das analog zu bekannten Methoden zu einer Verbindung der Formel III umgesetzt wird.
Die erfindungsgemäss hergestellten Verbindungen der Formel I und ihre Salze zeigen in der pharmakologischen Prüfung interessante Eigenschaften, aufgrund derer sie als Heilmittel verwendet werden können. So weisen sie insbesondere hypotensive Wirkung auf, wie in Tierversuchen festgestellt werden konnte. Weiterhin zeigen sie serotoninantagonistische und gefässtonisierende Aktivität, zudem zentrale Effekte, die sowohl dämpfender als auch erregender Natur sein können, und beeinflussen periphere wie auch zentrale Kreislaufparameter. Sie können daher bei der Behandlung zahlreicher Kreislauferkrankungen, vaskulär bedingter Schmerzzustände und Geriatrie Verwendung finden.
Die neuen Verbindungen können als Arzneimittel allein oder in entsprechenden Arzneiformen für orale, enterale oder parenterale Verabreichung verwendet werden. Zwecks Herstellung geeigneter Arzneiformen werden diese mit pharmakologisch indifferenten Hilfsstoffen verarbeitet.
In den nachfolgenden Beispielen, die die Erfindung näher erläutern, ihren Umfang aber in keiner Weise einschränken sollen, erfolgen alle Temperaturangaben in Celsiusgraden.
Beispiel I 1 -Methyl-6' -desoxo-9, 1 O-dihydroergocristin
150 ml abs. Dimethylformamid (DMF) werden unter Rühren auf - 15" gekühlt und tropfenweise mit einer Lösung von 1,14 ml (17,14 m Mol) Oxalylchlorid in 15 ml abs. Acetonitril versetzt. Anschliessend trägt man 4,87 g (17,14 m Mol) trockene 1-Methyl-9,10-dihydro-lysergsäure ein, wobei unter vor übergehender Lösung ein grauer Niederschlag ausfällt. Nach 30 Minuten Rühren bei 0 verdünnt man unter starkem Kühlen mit 37,5 ml abs.
Pyridin und trägt 5,14 g (2R,SS,lOaS,lObS) 2-Amino-2-isopropyl-3-oxo-5 -benzyl- 10b-hydroxy-perhy- drooxazolo[3 ,2-a]pyrrolo[2, 1-c]pyrazin.2,5 -Naphtalindisulfon- säure (ca. 8,5 m Mol) ein, rührt kräftig während 2 Stunden und lässt die Temperatur langsam von -10 bis 0 steigen. Zur Aufarbeitung wird unter starker Kühlung mit 40 ml Citrat Puffer pH=4 verdünnt und mit 2 N-Sodalösung alkalisch gestellt. Nach Extraktion mit Chloroform, das einige % Äthanol enthält, Trocknen und Eindampfen des Extrakts, wird das Rohprodukt an 500 g Kieselgel chromatographiert, wobei die Titelverbindung mit 4% Methanol in Methylenchlorid eluiert wird und aus Aceton kristallisiert.
Smp. 183-185" [a] D20= - 770 (c=l, Pyridin).
Das als Ausgangsmaterial benötigte 2-Amino-2-isopropyl3-oxo-5-benzyl-10b-hydroxy-perhydrooxazolo[3,2-a]pyrro- 10[2,1-c]pyrazin.2,5-Naphtalindisulfonsäure wird auf folgendem Wege erhalten: a) (2S, 7aS) -2 -Benzyl-perhydro- 8-oxo-pyrroloj2, 1 -cjpyrazin
Man suspendiert unter Stickstoff 36,4 g (960 m Mol) Lithi umaluminiumhydrid in 1,3 Liter abs. Tetrahydrofuran und tropft während 45 Minuten eine Lösung von 117,2 g (480 m Mol) L-Phe-L-Pro-Lactam in 1,4 Liter abs. Tetrahydrofuran zu.
Nach 18 Stunden Rühren bei - 10" wird bei dieser Temperatur der Reihe nach vorsichtig mit 350 ml Essigester und 350 ml Wasser verdünnt. Der Reaktionsbrei wird filtriert, wobei anschliessend noch haftendes Reaktionsprodukt durch wiederholte Extraktion des Filterkuchens mit siedendem Methylenchlorid extrahiert wird. Die vereinigten organischen Phasen werden mit Natriumsulfat getrocknet und eingedampft. Die reine Titelverbindung kristallisiert direkt aus Methylenchlorid/Äther in hellbeigen Kristallen vom Smp.135-140 ; [a] D20= - 3,50 (c= 1 in Äthanol).
b) (2R, 5S, 1 OaS, I ObS)2- Carbäthoxy-2-isopropyl-3-oxo-5- benzyl-lOb-hydroxy-perhydro-oxazolo[3,2-alpyrrolol2,1-c]- pyrazin
47,8 g (207 m Mol) 2S,7aS)-2-Benzyl-perhydro-8-oxo-pyr rolo[2,1-cjpyrazin in 160 ml abs. Dioxan werden mit 74,4 g (249 m Mol) S(+)-Isopropyl-benzyloxy-malonsäure-äthyl- esterchlorid und 34,9 g (270 m Mol) N-Äthyldiisopropylamin vermengt und 1 Stunde bei +70 gerührt. Nach Verdünnen mit 1 Liter Äther wird zweimal mit gesättigter Natriumbicarbonatlösung geschüttelt und über Natriumsulfat getrocknet.
Nach Eindampfen des Lösungsmittels werden 138 g eines bräunlichen Öls erhalten, das in 1,5 Liter Äthanol gelöst und bei 60 nach Versetzen mit 60 g Palladium-Aktivkohle (10% Pd) hydriert wird, wobei nach 14 Stunden etwa 6,5 Liter Wasserstoff absorbieren. Nach Filtration und Eindampfen des Filtrates kristallisiert die Titelverbindung nach Aufnehmen in Isopropyläther in gelblichen Prismen vom Smp.99-100 ; [aj D20= - 57,70 (c=0,65 in CHCl3).
c) (2R,5S, 1OaS,1ObS)2-Carboxy-2-isopropyl-3-oxo-5-ben- zyl-l Ob-hydroxy-perhydro-oxazolo[3,2-aapyrrolof 1- cjpyra - zin
39,3 g (97,8 m Mol) 2R,5S,10aS, IObS)2-Carbäthoxy-2- isopropyl-3-oxo-5-benzyl- 10b-hydroxy-perhydro-oxazolo [3,2-a]pyrrolo-[2,1-c]pyryzin werden in 245 ml Methanol und 245 ml 2 N-Natriumhydroxrad gelöst und 3 Stunden bei Raumtemperatur gerührt. Zur Aufarbeitung wird bei 0 mit 2 N-Salzsäure auf pH=4,5 gestellt und wiederholt mit Essigester extrahiert. Nach Trocknen des organischen Extraktes und Eindampfen des Lösungsmittels unterhalb +30 kristallisiert die Titelverbindung nach Verdünnen mit Äther. Smp. 126-128 (Zers.).
Sie enthält nach 5 Stunden Trocknen bei 30 im Hochvakuum 1/2 Mol Kristallwasser.
d) (2R, 5S, iOaS IObS)2-Amino-2-isopropyl-3-oxo-5-ben- zyl-10b-hydroxy-perhydro-oxazolo[3,2-a]pyrrolo[2,1-c]pydra- zin. 2, 5-Naphtalindisulfonsäure
In ein auf - 15" gekühltes, gerührtes Gemisch von 4,33 ml (56,3 m Mol) abs. Dimethylformamid und 30 ml abs. Acetonitril tropft man innert 10 Minuten eine Lösung von 3,84 ml (45 m Mol) Oxalylchlorid in 15 ml abs. Acetonitril zu und rührt noch weitere 10 Minuten. Nach Verdünnen mit 45 ml abs.
Äther werden 11,25 g (30 m Mol) (2R,5S,10aS,10bS)2-Carb oxy-2-isopropyl-3-oxo-5-benzyl-lOb-hydroxy-perhydro- oxazolo[3,2b]pyrrolo[2,1-c]pyrazin.l/2 H20 rasch eingetragen, wobei eine klare Lösung entsteht, die noch 10 Minuten bei - 10" gerührt wird. Dann überschichtet man mit einer Lösung von 20 g Natriumazid in 100 ml Wasser, verdünnt mit 300 ml Methylenchlorid und schüttelt das 2-Phasengemisch 4 Minuten kräftig. Dann fügt man 150 ml eiskalte, gesättigte Natriumbicarbonatlösung hinzu, trennt die organische Phase ab und trocknet gut über Natriumsulfat. Nach Einengen des Lösungsmittels unterhalb 30 wird mit einer Lösung von 7,78 g (27 m Mol) 2,5 Naphtalindisulfonsäure in 200 ml Dimethoxyäthan und 20 ml Acetonitril versetzt und im Vakuum langsam eingeengt.
Nach Verdünnen mit abs. Äther wird das Säureazid als Salz in Form eines beigen Pulvers isoliert. 6,35 g ( 9 m Mol) des getrockneten Pulvers werden mit 120 ml Dimethoxyäthan, das 0,166 ml ( 11 m Mol) enthält, 1 Stunde auf 80" erhitzt, wobei unter ständigem Kratzen ein bräunliches, halbkristallines Pulver anfällt, das nach Kühlen, Filtrieren und Trocknen bei Raumtemperatur im Hochvakuum direkt zur Kondensation mit einer geeignet substituierten Lysergsäure verwendet werden kann.
Beispiel 2 6' -Desoxo-9, 1 O-dihydro-ergotamin
Man verfährt in allen Stufen analog zu Beispiel 1, wobei (2S,7aS)-2-Benzyl-perhydro-8-oxo-pyrrolo[2,1-c]pyrazin mit 67,5 g (249 m Mol) S(+)-Methyl-benzyloxymalonsäure-äthylester-chlorid acyliert wird. Der analoge Cyclolester (2R,5S, 10aS,lObS)-2-Carbäthoxy-2-methyl-3-oxo-5-benzyl-lOb- hydroxy-perhydro-oxazolo[3,2-a]pyrrolo[2,1-c]pyrazin kristallisiert aus abs. Äther.
Smp. 106-110 ; [α]D20= - 53" (c=1,Äthanol).
Die analoge Cyclolcarbonsäure (2R,5S,10aS,10bS)-2- Carboxy-2-methyl-3-oxo-5 -benzyl-lOb-hydroxy-perhydro oxazolo[3,2-a]pyrrolo[2,1-c]pyrazin kristallisiert aus Essigester/ Äther.
Smp. 122" (Zers.); [a] D20= - 700 (c=1, Pyridin).
Der analoge Curtiusabbau führt über ein halbkristallines Aminocyclol (2R,SS,lOaS,lObS)2-Amino-2-methyl-3-oxo-5- benzyl-10b-hydroxy-perhydro-oxazolo[3,2-a]pyrrolo[2,1-c]- pyrazin.2,5-Naphtalindisulfonsäure, welches wiederum als 2,5 Naphtalindisulfonat isoliert und mit 9,10-Dihydrolysergsäure analog Beispiel 1 gekoppelt wird. Die so erhaltene Titelverbindung kristallisiert aus 90%igem Aceton.
Smp. 165-168 (Zers.); [α]D20= 87 (c= 1 (c=l in Pyridin).

For example, thionyl chloride, phosgene or oxalyl chloride can be used as the chlorinating agent.
Acid amides suitable for the formation of the required amide chloride are, for example, dimethylformamide or dimethylacetamide.
The process is advantageously carried out in an organic solvent mixture. Suitable solvents are e.g. Chloroform, methylene chloride, dimethylformamide or acetonitrile.
It is preferably carried out in the presence of a condensing agent. Tertiary organic bases, for example triethylamine, but preferably pyridine and homologs are expediently used.
The reaction can be carried out at temperatures between - 30 to + 20. You can work under normal pressure.

The other stages of the synthesis are in principle sub-stages of the Curtius method or modification thereof and are carried out using compounds of the formula VI. The esters of the formula VI are converted into the corresponding acid in the corresponding free acid via the carboxylic acid chloride into the acid azide, which is converted into a compound of the formula III analogously to known methods.
The compounds of the formula I prepared according to the invention and their salts show interesting properties in pharmacological testing, on the basis of which they can be used as medicaments. In particular, they have a hypotensive effect, as could be determined in animal experiments. Furthermore, they show serotonin-antagonistic and vasonizing activity, as well as central effects, which can be both damping and arousing, and influence peripheral and central circulatory parameters. They can therefore be used in the treatment of numerous circulatory diseases, vascular pain and geriatrics.
The new compounds can be used as pharmaceuticals alone or in appropriate pharmaceutical forms for oral, enteral or parenteral administration. In order to produce suitable pharmaceutical forms, these are processed with pharmacologically indifferent auxiliaries.
In the following examples, which explain the invention in more detail but are not intended to restrict its scope in any way, all the temperatures are given in degrees Celsius.
