PATENTANSPRÜCHE
1. Verfahren zur Herstellung von Dicyclohexylaminsalzen von Cephalosporinderivaten der Formel

mit einer A-2 oder A-3 Doppelbindung und worin R1 für Phenoxy, Phenyl oder Thienyl und R2 für Wasserstoff oder die Acetoxygruppe stehen, dadurch gekennzeichnet, dass die Cephalosporinderivate der Formel I in gelöstem Zustand mit Dicyclohexylamin umgesetzt und die gebildeten Salze in kristalliner Form isoliert werden.
2. Verfahren gemäss Anspruch 1, dadurch gekennzeichnet, dass die Cephalosporinderivate der Formel I als Gemische von A-2 und A-3-Isomeren vorliegen, die durch Ringerweiterung aus Estern von entsprechenden Penicillinsulfoxid-Derivaten erhältlich sind, und die gebildeten Salze mittels einer fraktionierten Kristallisation isoliert werden.
3. Verwendung der nach dem Verfahren gemäss Anspruch 1 erhaltenen Salze zur Desacylierung der Seitenkette Ri - CH2 - CO-NH-.

Verseifung Verbindungen der Formel I als Isomerengemisch erhalten werden, die Zwischenprodukte darstellen und vor ihrer Weiterverarbeitung gereinigt werden müssen. Für diese
Reinigung sind Verfahren bekannt, z.B. aus der
DOS 2 240 224. die jedoch aufwendig, zeitraubend und meist mit groben \'verlusten durchführbar sind. Bei dem nach der Ringerweiterung zur Reinigung anfallenden Produkt handelt es sich um ein Gemisch von A2- und A3-Isomeren, die womöglich gleichzeitig getrennt isoliert werden sollen.
Es wurde nun gefunden, dass diese Reinigung und gleichzeitige Trennung von A2- und A3-Isomeren durch Salzbildung mit Dicyclohexylamin durchgeführt werden kann.
Das erfindungsgemässe Verfahren wird beispielsweise so ausgeführt, dass man das bei der Ringerweiterung anfallende Reaktionsgemisch in einem Lösungsmittel, z.B. in Butylacetat oder Essigester, löst und mit Dicyclohexylamin versetzt. Das Salz des A3-Isomeren kristallisiert meist rasch aus und kann nach bekannten Methoden isoliert und gegebenenfalls gereinigt werden.
Durch diese Verfahren wird die bisher in zwei Stufen mit relativ geringen Ausbeuten durchgeführte Reinigung und Trennung des A3- vom A2-Isomeren in einer Stufe und mit hohen Ausbeuten vorgenommen.
Die so erhaltenen gereinigten Zwischenprodukte können als erfindungsgemässe Verwendungsmöglichkeit anschliessend einer Deacylierung unterworfen werden. Bisher musste man dafür die gereinigten Verbindungen in ein Salz überführen, z.B. durch Behandeln mit N-Äthylpiperidin. Bei dem erfindungsgemässen Verfahren fallen jedoch nach der Reinigung und Trennung die zur Weiterverarbeitung vorgesehenen Zwischenprodukte bereits als Dicyclohexylaminsalze an, die direkt der Deacylierung unterworfen werden können.
Obwohl die Umsetzung mit Dicyclohexylamin bevorzugt dazu dient, Gemische von A2- und A3-Cephemcarbonsäuren zu reinigen und zu trennen, ist die Erfindung nicht darauf beschränkt und bezieht sich ebenfalls auf die Reinigung und Isolierung von gesonderten A2- und A3-Cephemcarbonsäuren.
In den nachfolgenden Beispielen, die die Erfindung näher erläutern, ihren Umfang aber in keiner Weise einschränken sollen, erfolgen alle Temperaturangaben in Celsiusgraden.
Beispiel 1 Dicyclohexylaminsalz der 7-Phenoxyacetamido-3 -methyl-3 cephem-4-carbonsäure
Eine Lösung von 3,5 g 7-Phenoxyacetamido-3-methyl-3 cephem-4-carbonsäure in 50 ml Essigester wird mit 1,8 g Dicyclohexylamin unter Rühren versetzt. Dabei kristallisiert das Salz aus, es wird abgesaugt, mit Essigester gewaschen und getrocknet. Es werden 4,8 g (91%) erhalten, Fp. 182-1840 (Zers.), [a]2D0 = +63,50 (c = 2 in Methylenchlorid).
Beispiel 2 Fraktionierung der 7-Phenoxyacetamido-3-methyl-3- und 2 cephem4-carbonsäure
Eine Lösung von 3 g 7-Phenoxyacetamido-3-methyl-3 cephem-4-carbonsäure und 0,5 g 7-Phenoxyacetamido-3methyl-2-cephem-4-carbonsäure in 100 ml Butylacetat wird mit 1,8 g Dicyclohexylamin versetzt. Das dabei ausgefallene Salz wird abgesaugt, mit Äther gewaschen und getrocknet: 4,3 g, Fp. 171-174" (Zers.). [alD20 = 62,70 (c = 2 in Methylenchlorid).
Beim Aufkonzentrieren der Mutterlauge auf ca. 20 ml kristallisiert das Salz der A2-Säure aus: 0,45 g, Fp. 170-172" (Zers.), [oÜD0 = +332 (c = 2 in Methylenchlorid).
Die Dünnschichtchromatographie (Kieselgelplatten, Nussbaumer-System als Laufmittel) zeigt eine Verunreinigung durch die A3-Säure an. Nussbaumersystem: 80 Teile Butylacetat, 15 Teile n-Butanol, 40 Teile Eisessig, 24 Teile Wasser und 5 Teile Methanol.
Beispiel 3
Dicyclohexylaminsalz der 7-Phenoxyacet amido-3-methyl-2-cephem-4-carbonsäure
Eine Lösung von 0,7 g 7-Phenoxyacetamido-3-methyl-2
cephem-4-carbonsäure in 10 ml Aceton wird mit 0,4 g Dicyclohexylamin versetzt. Beim Stehen bei Raumtemperatur kristallisiert das Salz langsam aus. Absaugen, Waschen mit Aceton und Trocknen ergibt 0,9 g Dicyclohexylaminsalz, Fp.
173-175 , [a]2D0 = +359 (c = Methylenchlorid). Es ist dünnschichtchromatographisch rein.
Beispiel 4
Dicyclohexylaminsalz der 7-Phenoxyacet amido-3-methyl-3 -cephem-4-carbonsäure
Eine Lösung von 3,3 g 7-Phenoxyacetamido-3-methyl-3cephem-4-carbonsäure in 150 ml Essigester wird unter Rühren tropfenweise mit 1,8 g Dicyclohexylamin versetzt, wobei sofort ein feiner Niederschlag ausfällt. Absaugen, Waschen mit Essigester und Trocknen ergibt 4,6 g Salz (Fp. 167-171", Zersetzung). [aj200 = +62,3 (c = 2inMethylenchlorid).
Beispiel 5
Dicyclohexylaminsalz der 7-Thienyl acetamido-cephalosporansäure
2 g 7-Thienylacetoamido-cephalosporansäure werden in 200 ml Essigsäureäthylester gelöst und langsam unter Umschwenken mit einer Lösung von 910 mg Dicyclohexylamin in 50 ml Essigsäureäthylester versetzt. Dabei fällt das Salz als voluminöser Niederschlag aus. Nach kurzem Stehen bei Raumtemperatur wird abgesaugt, mit Äther gewaschen und getrocknet.
Ausbeute: 2,5 g, ca. 86% d. Th. Fp. = 185-189 (Zers.) .
[a]2D0 = +33 (c = 2%inCHCl).
Beispiel 6
Dicyclohexylaminsalz der 7-Phenoxy acetamido-cephalosporansäure
200 mg 7-Phenoxyacetamido-cephalosporansäure werden in 30 ml Essigsäureäthylester gelöst, mit 3 ml einer Lösung von 90 mg Dicyclohexylamin in Essigester langsam versetzt und mit einem Glasstab angerieben. Kurz darauf setzt die Kristallisation ein. Nach einer Stunde Stehen im Eisschrank wird der Niederschlag abgesaugt, mit Äther gewaschen und getrocknet.
Ausbeute: 221 mg, ca. 76% d. Th. Fp. = 158-1630, [a]2D0 = +42 (c = 2% in CHCl3).
Beispiel 7
Dicyclohexylaminsalz der 7-Phenyl acetamido-cephalosporansäure
200 mg 7-Phenylacetamido-cephalosporansäure werden in einer Mischung aus 3 ml Äthylalkohol und 27 ml Essigsäure äthylester gelöst und unter ständigem Umschwenken mit einer Lösung von 95 mg Dicyclohexylamin in 3 ml Essigsäureäthylester langsam versetzt. Nach wenigen Minuten fällt das Salz langsam aus und die Fällung wird durch Stehenlassen im Eisschrank während 30 Minuten vervollständigt. Es wird abgesaugt, mit Äther gewaschen und getrocknet.
Ausbeute: 243 mg, ca. 82% d. Th. Fp. 179-1850, [a]2D0 +340 (c = 2 in CHCI3) .

It has now been found that this purification and simultaneous separation of A2 and A3 isomers can be carried out by salt formation with dicyclohexylamine.
The process according to the invention is carried out, for example, in such a way that the reaction mixture obtained during ring expansion is dissolved in a solvent, e.g. in butyl acetate or ethyl acetate, and mixed with dicyclohexylamine. The salt of the A3 isomer usually crystallizes out quickly and can be isolated and, if necessary, purified by known methods.
By means of this process, the purification and separation of the A3- from the A2-isomer, previously carried out in two stages with relatively low yields, is carried out in one stage and with high yields.
The purified intermediate products obtained in this way can then be subjected to deacylation as a possible use according to the invention. Up to now you had to convert the purified compounds into a salt, e.g. by treating with N-ethylpiperidine. In the process according to the invention, however, after the purification and separation, the intermediate products intended for further processing are obtained as dicyclohexylamine salts which can be subjected directly to deacylation.
Although the reaction with dicyclohexylamine is preferably used to purify and separate mixtures of A2- and A3-cephemcarboxylic acids, the invention is not limited to this and also relates to the purification and isolation of separate A2- and A3-cephemcarboxylic acids.
In the following examples, which explain the invention in greater detail but are not intended to limit its scope in any way, all temperatures are given in degrees Celsius.
Example 1 Dicyclohexylamine salt of 7-phenoxyacetamido-3-methyl-3-cephem-4-carboxylic acid
A solution of 3.5 g of 7-phenoxyacetamido-3-methyl-3-cephem-4-carboxylic acid in 50 ml of ethyl acetate is mixed with 1.8 g of dicyclohexylamine while stirring. The salt crystallizes out, it is filtered off with suction, washed with ethyl acetate and dried. 4.8 g (91%) are obtained, melting point 182-1840 (decomp.), [A] 2D0 = +63.50 (c = 2 in methylene chloride).
Example 2 Fractionation of 7-phenoxyacetamido-3-methyl-3- and 2-cephem4-carboxylic acids
1.8 g of dicyclohexylamine are added to a solution of 3 g of 7-phenoxyacetamido-3-methyl-3-cephem-4-carboxylic acid and 0.5 g of 7-phenoxyacetamido-3methyl-2-cephem-4-carboxylic acid in 100 ml of butyl acetate. The salt which has precipitated out is filtered off with suction, washed with ether and dried: 4.3 g, melting point 171-174 "(decomp.). [AlD20 = 62.70 (c = 2 in methylene chloride).
When the mother liquor is concentrated to approx. 20 ml, the salt of the A2 acid crystallizes out: 0.45 g, melting point 170-172 "(decomp.), [OUD0 = +332 (c = 2 in methylene chloride).
Thin-layer chromatography (silica gel plates, Nussbaumer system as the mobile phase) shows contamination by the A3 acid. Nussbaumer system: 80 parts of butyl acetate, 15 parts of n-butanol, 40 parts of glacial acetic acid, 24 parts of water and 5 parts of methanol.
Example 3
Dicyclohexylamine salt of 7-phenoxyacetamido-3-methyl-2-cephem-4-carboxylic acid
A solution of 0.7 g of 7-phenoxyacetamido-3-methyl-2
cephem-4-carboxylic acid in 10 ml of acetone is mixed with 0.4 g of dicyclohexylamine. The salt slowly crystallizes out on standing at room temperature. Sucking off, washing with acetone and drying gives 0.9 g of dicyclohexylamine salt, m.p.
173-175, [a] 2D0 = +359 (c = methylene chloride). It is pure by thin-layer chromatography.
Example 4
Dicyclohexylamine salt of 7-phenoxyacetamido-3-methyl-3-cephem-4-carboxylic acid
A solution of 3.3 g of 7-phenoxyacetamido-3-methyl-3cephem-4-carboxylic acid in 150 ml of ethyl acetate is admixed dropwise with 1.8 g of dicyclohexylamine while stirring, a fine precipitate immediately separating out. Sucking off, washing with ethyl acetate and drying gives 4.6 g of salt (melting point 167-171 ", decomposition). [Aj200 = +62.3 (c = 2 in methylene chloride).
Example 5
Dicyclohexylamine salt of 7-thienyl acetamido-cephalosporanic acid
2 g of 7-thienylacetoamido-cephalosporanic acid are dissolved in 200 ml of ethyl acetate and a solution of 910 mg of dicyclohexylamine in 50 ml of ethyl acetate is slowly added while swirling. The salt precipitates out as a voluminous precipitate. After standing briefly at room temperature, it is filtered off with suction, washed with ether and dried.
Yield: 2.5 g, approx. 86% of theory. Th. Mp = 185-189 (dec.).

Example 6
Dicyclohexylamine salt of 7-phenoxy acetamido-cephalosporanic acid
200 mg of 7-phenoxyacetamido-cephalosporanic acid are dissolved in 30 ml of ethyl acetate, 3 ml of a solution of 90 mg of dicyclohexylamine in ethyl acetate are slowly added and the mixture is rubbed with a glass rod. Shortly afterwards, crystallization begins. After standing in the refrigerator for an hour, the precipitate is filtered off with suction, washed with ether and dried.

Example 7
Dicyclohexylamine salt of 7-phenyl acetamido-cephalosporanic acid
200 mg of 7-phenylacetamido-cephalosporanic acid are dissolved in a mixture of 3 ml of ethyl alcohol and 27 ml of ethyl acetate and a solution of 95 mg of dicyclohexylamine in 3 ml of ethyl acetate is slowly added while swirling continuously. After a few minutes, the salt slowly precipitates out and the precipitation is completed by standing in the refrigerator for 30 minutes. It is suctioned off, washed with ether and dried.
