Die vorliegende Erfindung betrifft ein neues Verfahren zum Herstellen von [l-Oxo-(2-phenyl-, -halogenphenyl- oder -thienyl)-2-methyl-6,7-dichlor- 5-indanyloxy]-D erivaten. Pharmazeutische Untersuchungen zeigten, dass die oben erwähnten Produkte wirksame diuretische und saluretische Mittel sind, die bei der Behandlung von Zuständen herangezogen werden können, welche bei Elektrolyt- und Flüssigkeitsretention auftreten.
Die oben erwähnten Verbindungen werden insbesondere auch bei der Behandlung von Hypertonie verwendet. Ausserdem vermögen diese Verbindungen die Harnsäurekonzentration in dem Körper bei durch Vorbehandlung erreichten Niveaus zu halten und sogar eine Abnahme der Harnsäurekonzentration zu bewirken.
Wenn die Verbindungen in therapeutischen Dosierungen in herkömmlichen Trägern verabreicht werden, setzen sie die Natrium- und Chioridionenmenge im Körper wirksam herab, senken gefährliche und übermässig hohe Flüssigkeitsniveaus auf annehmbare Niveaus und lindern allgemein solche Zustände, die üblicherweise mit Ödemen verbunden sind. Ausserdem wird durch die Verbindungen ein Hauptproblem überwunden, das mit vielen der gegenwärtig verfügbaren Diuretika und Saluretika verbunden ist. So haben viele der gegenwärtig verfügbaren Diuretika und Saluretika beispielsweise die Neigung, bei der Verabreichung Hyperurikämie herbeizuführen, die bewirken kann, dass Harnsäure oder Natriumurat oder beide im Körper ausfallen, was leichte bis schwere Fälle von Gicht verursachen kann.
Die erfindungsgemäss erhältlichen Verbindungen stellen nun ein wirksames Mittel dar, um solche Patienten, und zwar sowohl Menschen als auch Tiere, die eine diuretische und saluretische Behandlung verlangen, zu behan dem, ohne dass man das Risiko eingeht, dass das Auftreten von Gicht gefördert wird.
Es ist daher ein Ziel der vorliegenden Erfindung, ein spezifisches, sehr wirksames, vereinheitlichtes Verfahren zur Herstellung der genannten Verbindungen bereitzustellen. Diese können in ihre nichttoxischen, pharmazeutisch annehmbaren Salze überführt werden, falls sie als freie Säure vorliegen. Ein Vorteil des erfindungsgemässen Verfahrens ist der, dass es die Verwendung von verhältnismässig billigeren Ausgangsstoffen als früher durchgeführte Verfahren gestattet.

In den erfindungsgemäss erhaltenen Verbindungen ist R vorzugsweise der Methylrest. Der Halogenphenylrest R' ist bevorzugt p-Chlor- oder p-Fluorphenyl oder 2-Thienyl.
Wie schon erwähnt, wird das erfindungsgemässe Verfahren in Gegenwart einer Lewis-Säure, die als Katalysator dient, durchgeführt. Bevorzugt arbeitet man mit Aluminiumchlorid oder Zinnchlorid und mit einem Lösungsmittel, insbesondere einem inerten, organischen Lösungsmittel, das sich nicht merklich mit den Reaktanten umsetzt. Typisch für solche inerte Lösungsmittel sind Methylenchlorid, Tetrachloräthan, Hexan, Kohlenstoffdisulfid oder Nitrobenzol. Das Verfahren kann bei einer Temperatur von etwa 0 bis zur Rückflusstemperatur des Lösungsmittels, wobei 20 bis 50 bevorzugt werden, so lange durchgeführt werden, bis die Umsetzung praktisch vollständig zu Ende ist; im allgemeinen wird die Umsetzung jedoch bei etwa 40 C während etwa 4 bis 6 Stunden durchgeführt.
Das a,a -disubstituierte ss-Halogenpropionylhalogenid der Formel (III) kann nach der allgemeinen Methode von Nicolaus, Mariani und Teste ( A New General Synthesis for a-Substituted Propionic Acid and its Derivatives , Bruno J.R. Nicolaus, Luigi Mariani and Emilio Testa (Lepetit Spa, Milan) Chem. Abst. 55 27271d (1961) hergestellt werden.
Das Reaktionsprodukt kann aus dem Reaktionsgemisch nach aus dem Stand der Technik bekannten Methoden, wie durch Extrahieren der gewünschten Verbindung aus ihrer Reaktionsmischung mit einem Lösungsmittel, Abdampfen des genannten Lösungsmittels und nachfolgendes Umkristallisieren des gewünschten Indanons aus Aceton oder einem anderen geeigneten Lösungsmittel, isoliert werden.
Wenn in den erfindungsgemäss erhaltenen Verbindungen der Formel (I) R für Alkyl mit 1-5 C-Atomen steht, so kann an dieser Verbindung eine Ätherspaltung durchgeführt werden, gemäss der die Verbindung der Formel I z. B. mit einem Ätherspaltungsreaktanten umgesetzt wird. Dann kann das gebildete 5-Hydroxyprodukt mit einer Halogenessigsäure oder Pseudohalogenessigsäure oder einem Ester davon der Formel

Wenn R" in dem Halogenessigsäureester für Niedrigalkyl steht, kann das gebildete Produkt der Formel I hydrolysiert werden, um die freie Säure herzustellen.
Die erwähnte Ätherspaltung wird gewöhnlich mit ätherspaltenden Reaktanten, wie Aluminiumchlorid, Pyridinhydrochlorid, Bromwasserstoff, Natrium in flüssigem Ammoniak und dergleichen, durchgeführt. Bei Verwendung von Aluminiumchlorid kann das Lösungsmittel Heptan, Kohlenstoffdisulfid, Methylenchlorid und dergleichen sein, und wenn z. B. Pyridinhydrochlorid verwendet wird, ist es nicht notwendig, ein Lösungsmittel zu verwenden.
Im allgemeinen wird diese Ätherspaltung bei Temperaturen von Raumtemperatur bis zur Rückflusstemperatur irgendeines der verwendeten Lösungsmittel. für genügend lange Zeit durchgeführt, um die Bildung der Hydroxyverbindung praktisch zu vervollständigen.
Diejenige Reaktion, bei der das nach der Ätherspaltung erhaltene Produkt mit einer Halogenessigsäure oder einem Ester umgesetzt wird, wird im allgemeinen in Gegenwart einer Base, wie Alkalicarbonat, -hydroxid oder -alkoxid, wie Kaliumcarbonat, Natriumcarbonat, Kaliumhydroxid, Natriumhydroxid, Natriumäthoxid und dergleichen, ausgeführt. Verwendbar ist jedes beliebige Lösungsmittel, das gegenüber den Reaktanten inert oder praktisch inert ist und in dem die Reaktanten angemessen löslich sind. Äthanol, Dimethylformamid, Benzol und Toluol beispielsweise haben sich als besonders vorteilhafte Lösungsmittel erwiesen. Die Umsetzung kann bei einer Temperatur im Bereich von etwa 25 C bis zur Rückflusstemperatur des speziell verwendeten Lösungsmittels ausgeführt werden.
Die Umsetzung mit der Halogenessigsäure oder dem Ester ist im allgemeinen in etwa 10 bis 60 Minuten beendet. Wenn z. B.
der Halogenessigsäureester verwendet wird, kann der erhaltene Ester nach Methoden, die dem Fachmann geläufig sind, zu der freien Säure hydrolysiert werden.
Die vorliegende Erfindung wird durch die folgenden Beispiele veranschaulicht.
Beispiel 1
2-Methyl-2-phenyl-5-methoxy-6,7-dichlor-1 -indanon
177 g (1,0 Mol) 2,3-Dichloranisol, 280 g (2,10 Mol) wasserfreies Aluminiumchlorid und 545 ml Kohlenstoffdisulfid werden unter einer Stickstoffatmosphäre vermischt. Das Gemisch wird bei 25 bis 30 C gerührt, während 261,5 g (1 Mol) a -Methyl -phenyl-ss -brompropionylchlorid im Verlauf von 11/2 Stunden zugesetzt werden. Das Reaktionsgemisch wird bei 25 bis 30 1 Stunde lang und dann bei 43 bis 45" 4 Stunden lang gealtert. Die Charge, die schliesslich über Nacht bei 25" gerührt worden ist, wird sorgfältig in einer Mischung aus 2,73 Liter Eis und Wasser abgeschreckt.
Die Temperatur steigt während des Abschreckens auf 28" an, und die Charge wird 10 Minuten lang gealtert. Die Schichten werden getrennt, und die wässrige Phase wird mit 2 x 550 ml Toluol extrahiert.
Die vereinigte organische Phase wird mit 275 ml 10%der Natriumbicarbonatlösung gewaschen, über Magnesiumsulfat getrocknet und im Vakuum zur Trockne eingeengt. Man erhält 207 g (93 %) 2-Methyl-2-phenyl-5-methoxy-6,7-dichlor-1-indanon.
Nach der oben beschriebenen Arbeitsweise werden unter Verwendung von a-Methyl-p-chlorphenyl- oder -2-thienyl-ss - brompropionylchlorid anstelle von a -Methyl-a -phenylp-brom- propionylchlorid äquivalente Mengen an 2-Methyl-2-p-chlor phenyl-5-methoxy-6,7-dichlor-1 -indanon bzw. 2-Methyl-2-(2 thienyl)-5-methoxy-6,7-dichlor-1-indanon hergestellt.
Beispiel 2 (1-Oxo-2-phenyl-2-methyl-6,7-dichlor-5-indanyloxy)- essigsäure
Diese Verbindung wird nach derselben Arbeitsweise, wie sie in Beispiel 1 angewandt wurde, jedoch mit der Abänderung hergestellt, dass 2,3-Dichlorphenoxyessigsäure anstelle von 2,3-Dichloranisol verwendet wird.
Nach der oben beschriebenen Arbeitsweise werden unter Verwendung von a -Methyl-a -p-chlorphenyl-oder-2-thienyl-ss - brompropionylchlorid anstelle von a -Methyle -phenyl-ss -brom- propionylchlorid äquivalente Mengen an [1 -Oxo-2 -methyl-2 - (p-chlorphenyl-oder-2-thienyl)-2-methyl-6,7 -dichlor-5 -indanyl.

Beispiel 3
Herstellung von (1 -Oxo-2-phenyl-2 -methyl-6,7-dichlor-
5 -indanyloxy) -essigsäure
Schritt A 2-Phenyl-2-methyl-5 -hydroxy-6, 7-dichlor-1 -indanon
3,5 g (0,0109 Mol) 2-Phenyl-2-methyl-5-methoxy-6,7-di- chlor-1-indanon werden zu 180 ml trockenem Hexan gegeben, und es werden 4,36 g (0,0327 Mol) Aluminiumchlorid hinzugefügt. Das Gemisch wird 1 Stunde unter Rückfluss gekocht, und das Hexan wird von dem gummiartigen Rückstand abdekantiert, der dann zu 200 ml Eiswasser, die 15 ml 12n Chlorwasserstoffsäure enthalten, gegeben wird. 2,84 g (0,00926 Mol) 2-Phenyl-2-methyl-5-hydroxy-6,7dichlor-1-indanon werden zu 40 ml Dimethylformamid (DMF) gegeben. 3,21 g (0,0232 Mol) Kaliumcarbonat und 3,34 g (0,0232 Mol) Äthylbromacetat werden hinzugegeben. Das Gemisch wird 21/2 Stunden lang bei 55 bis 60 C gerührt, dann werden 40 ml 10%ges Natriumhydroxid hinzugesetzt, und das Gemisch wird 11/2 Stunden lang bei 80 bis 85" C gerührt. Das Gemisch wird dann zu 500 ml 2%iger Chiorwasser- stoffsäure gegeben. Der sich abscheidende Feststoff wird in Äther aufgenommen. Der Ätherauszug wird mit Wasser gewaschen, über Magnesiumsulfat getrocknet und eingedampft.
Es bleibt ein gummiartiger Feststoff zurück, der aus Essigsäure auskristallisiert wird. Man erhält 183 g (1-Oxo-2-phenyl-2methyl-6,7-dichlor-5 -indanyloxy) -essigsäure.
Nach der oben beschriebenen Arbeitsweise werden unter Verwendung von 2-p-Chlorphenyl-oder-2-(2-thienyl)-2-methyl-5-methoxy-6,7-dichlor-1-indanon anstelle von 2-Phenyl-2 methyl-5-methoxy-6,7-dichlor-1 -indanon äquivalente Mengen an [1-Oxo-2-(p-chlorphenyl-bzw.-2-thienyl)-2-methyl-6,7-di- chlor-5 -indanyloxy] -essigsäure hergestellt.
