Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung des neuen Antibiotikum-Derivates Tetrahydro-SL 7810/F.
Erfindungsgemäss gelangt man zum Tetrahydro-SL 7810/F, indem man das Antibiotikum SL 7810/F katalytisch hydriert.
Die Hydrierung des Antibiotikums SL 7810/F wird vorteilhafterweise ausgeführt, indem man das Antibiotikum SL 7810/F in einem geeigneten Lösungsmittel in Gegenwart von katalytisch erregtem Wasserstoff hydriert. Als Lösungsmittel kommen vorzugsweise niedere aliphatische Alkohole, wie z. B. Methanol oder Äthanol, in Frage. Die Hydrierung erfolgt zweckmässigerweise in schwach saurem Bereich, z. B. in Gegenwart von schwachen organischen Säuren, wie Essigsäure. Die Hydrierung erfolgt bei Temperaturen zwischen 10 und 40 , vorzugsweise zwischen 20 und 25 , unter Atmosphärendruck oder wenig erhöhtem Druck in Gegenwart eines Palladium-Katalysators, wie z. B. Palladium auf Kohle oder Bariumsulfat. Das auf diese Weise erhaltene hydrierte Derivat des Antibiotikums SL 7810/F wird hierauf in an sich bekannter Weise gereinigt, z.
B. durch Chromatographie an Kieselgel, z. B. mit Chloroform-Methanol. Das als Ausgangsprodukt benützte Antibiotikum SL 78101F kann, wie in nachstehendem Beispiel beschrieben, hergestellt werden.
Tetrahydro-SL 7810/F ist antibiotisch wirksam und zwar gegen Hefen und Pilze und kann als Heilmittel verwendet werden. Im Reihenverdünnungstest wurden die folgenden minimalen Hemmkonzentrationen gefunden (in ,ug/ml): Saccharomyces cerevisiae 0,01 Hansuela anomala 0,01 Rhodotorula rubra > 100 Kloeckera apiculata Cryptococcus neoformans > 100 Candida albicans 0,03 Candida albicans 5897 > 100 Candida albicans 439 0,03 Candida albicans Blasticidin res. > 100 Candida tropicalis > 100 Candida tropicalis CK4 > 100 Candida krusei 0,03 Candida albicans H12 0,1
Hervorzuheben ist die gute Stabilität von Tetrahydro-SL 7810/F. Ein trockenes amorphes Pulver von Tetrahydro-SL 7810/F kann bei Raumtemperatur über längere Zeit gelagert werden.
Als Heilmittel können das neue Antibiotikum-Derivat allein oder in geeigneten Arzneiformen gemeinsam mit anorganischen oder organischen, pharmakologisch indifferenten Hilfsstoffen verabreicht werden. Beispielsweise werden sie als Bestandteil einer Salbe eingesetzt, wobei die Konzentration in der Salbe ca. 5-50 mg Wirkstoff pro g Salbe betragen kann.
In dem nachfolgenden Beispiel, welches die Ausführung des Verfahrens erläutert, den Umfang der Erfindung aber in keiner Weise einschränken soll, erfolgen alle Temperaturangaben in Celsiusgraden.
Beispiel: Tetrahydro-SL 7810/F
Eine Suspension von 1 g Palladium-Kohle (10% Pd) in 250 ml Alkohol-Eisessig (1:1) wird 30 Minuten vorhydriert.
Anschliessend gibt man eine Lösung von 5 g SL 7810/F in 50 ml Alkohol-Eisessig (1:1) zu. Nach 3 Stunden Hydrierung bei 20 unter Atmosphärendruck wird der Katalysator auf einer Talknutsche abgetrennt und das klare Filtrat im Vakuum eingedampft. Der Eindampfrückstand wird in Chlors form-Methanol (1:1) gelöst und an 10 g Kieselgel 60 Merck Korngrösse 0,063-0,2 mm) imprägniert und auf eine Säule von 500 g Kieselgel gegeben. Die anschliessende Elution mit Chloroform-Methanol (4:1) liefert das Hydrierungsprodukt von SL 7810/F.
Die Laufstrecken von Tetrahydro-SL 7810/F im Dünnschichtchromatogramm gehen aus folgender Tabelle hervor:

Als Sprühreagens kann eine 0,20/obige Ce(SO4)2-Lösung in 50 /0iger Schwefelsäure verwendet werden. Bei Erwärmen der Dünnschichtplatten gibt Tetrahydro-SL 7810/F eine gelbliche Färbung.

Das Antibiotikum-Derivat Tetrahydro-SL 7810/F ist bei Raumtemperatur leicht löslich in Methanol und Äthanol, schwer löslich in Chloroform, Aceton, Äther, Petroläther, Hexan und praktisch unlöslich in Wasser.
Das als Ausgangsmaterial verwendete Antibiotikum SL 7810/F wird wie folgt hergestellt:
Eine Sporensuspension, die zur Beimpfung der Nährlösung dient, wird aus einer Kultur des ursprünglich isolierten Stammes von Aspergillus rugulosus Thom et Raper hergestellt.
Der verwendete Stamm von Aspergillus rugulosus Thom et Raper wurde aus einer auf dem Jaunpass in der Schweiz gefundenen Bodenprobe isoliert und eine Probe davon beim United States Department of Agriculture (Northern Utilization Research and Development Division), Peoria, 111., USA deponiert und steht der Öffentlichkeit unter der Kulturnummer NRRL 8039 zur Verfügung.
Man beimpft mit einer Sporensuspension des Stammes NRRL 8039 200 ml Nähragar folgender Zusammensetzung: 20 g Cerelose 2g Pepton
2 g Malzextrakt
2 g Hefeextrakt
2 g primäres Kaliumphosphat
2 g Magnesiumsulfat (MgSO4 7H2O) 15g Agar
1 Liter entionisiertes Wasser Nach 10 Tagen Inkubation bei 27 werden die gebildeten Sporen in 200 ml sterilem Wasser suspendiert und damit ein Stahlfermenter beimpft, der 50 Liter folgender Nährlösung enthält:
20 g Cerelose 10 g Stärke lOg Sojapepton
3 g Calciumcarbonat
2 g Natriumnitrat 1 g primäres Kaliumphosphat
0,5 g Kaliumchlorid
0,5 g Magnesiumsulfat (MgSO4 7H2O) 1 mol Antischaummittel 1 Liter entionisiertes Wasser Dieser Fermenter wird unter Rühren während 42 Stunden bei 27 inkubiert, wobei die Belüftung 1 Liter Luft/miniLiter Medium beträgt bei einer Rührgeschwindigkeit von 250 U/min.
Die Kultur in diesem 50 I-Fermenter dient als Vorkultur für einen 500 I-Fermenter, der die gleiche Nährlösung enthält und 66 Stunden bei 27 inkubiert wird. Die Belüftungsrate beträgt wiederum 1 Liter Luft/min/Liter Medium, die Rührgeschwindigkeit aber nur 150 U/min. Nach 66 Stunden Inkubationsdauer wird die Kulturbrühe geerntet und aufgearbeitet.
450 1 Kulturbrühe werden mit 2 N Salzsäure auf pH 7,0 gestellt, mit 500 1 Essigester versetzt und mit dem Dispase Reaktor homogenisiert. Anschliessend wird die organische Phase mit dem Separator von der Brühe getrennt und mit 50 1 Wasser gewaschen. Dieser Extraktionsschritt wird 2 2mal wiederholt. Die erhaltenen 3 Extrakte werden vereinigt und unter Vakuum (Wasserringpumpe) bei 20-40 zur Trockne eingeengt; Ausbeute 320 g Rohextrakt.
Dieses Material wird an der 4fachen Menge Kieselgel Merck 60 (Korngrösse 0,063-0,20 mm) fraktioniert Die Eluate mit Chloroform +10% Methanol und Chloroform +200in Methanol enthalten inaktive Begleitstoffe. Die Chromatographiesäule wird anschliessend mit Chloroform-Methanol (1:1) eluiert und die Fraktionen mit Aktivität gegen Candida albicans vereinigt. Durch chromatographische Reinigung von 16,4 g aktivem Material an 1 kg Kieselgel Merck 60 mit Chloroform +300/0 Methanol als Elutionsmittel und nachfolgende Gelfiltration der aktiven Fraktionen an 1,2 kg Sephadex LH20 mit Methanol wird SL 7810/F in annähernd reiner Form gewonnen. Dieses Präparat wird unter Erwärmen in Aceton gelöst und die klare Lösung auf Y4 des Volumens eingeengt.
Nach 3 Stunden Stehen bei -15 wird der entstandene Niederschlag abfiltriert und nach Waschen mit Aceton und Äther 24 Stunden bei Raumtemperatur über P2O5 im Hochvakuum getrocknet. SL 7810/F fällt als farbloses amorphes Pulver vom Smp. 160-163 an (auf dem Kofler-Block); [a]=-- 48,3 (c=0,806 in Methanol).
The present invention relates to a process for the production of the new antibiotic derivative Tetrahydro-SL 7810 / F.
According to the invention, the Tetrahydro-SL 7810 / F is obtained by catalytically hydrogenating the antibiotic SL 7810 / F.
The hydrogenation of the antibiotic SL 7810 / F is advantageously carried out by hydrogenating the antibiotic SL 7810 / F in a suitable solvent in the presence of catalytically excited hydrogen. The solvents used are preferably lower aliphatic alcohols, such as. B. methanol or ethanol in question. The hydrogenation is conveniently carried out in a weakly acidic range, e.g. B. in the presence of weak organic acids such as acetic acid. The hydrogenation is carried out at temperatures between 10 and 40, preferably between 20 and 25, under atmospheric pressure or slightly elevated pressure in the presence of a palladium catalyst, such as. B. palladium on carbon or barium sulfate. The hydrogenated derivative of the antibiotic SL 7810 / F obtained in this way is then purified in a manner known per se, e.g.
B. by chromatography on silica gel, e.g. B. with chloroform-methanol. The antibiotic SL 78101F used as the starting product can be prepared as described in the example below.
Tetrahydro-SL 7810 / F has an antibiotic effect against yeast and fungi and can be used as a remedy. The good stability of Tetrahydro-SL 7810 / F should be emphasized. A dry amorphous powder of Tetrahydro-SL 7810 / F can be stored for a long time at room temperature.
As a remedy, the new antibiotic derivative can be administered alone or in suitable drug forms together with inorganic or organic, pharmacologically indifferent auxiliary substances. For example, they are used as a component of an ointment, the concentration in the ointment being about 5-50 mg of active ingredient per g of ointment.
In the following example, which explains the implementation of the method but is not intended to restrict the scope of the invention in any way, all temperatures are given in degrees Celsius.
Example: Tetrahydro-SL 7810 / F
A suspension of 1 g of palladium-carbon (10% Pd) in 250 ml of alcohol-glacial acetic acid (1: 1) is pre-hydrogenated for 30 minutes.
A solution of 5 g SL 7810 / F in 50 ml alcohol-glacial acetic acid (1: 1) is then added. After hydrogenation for 3 hours at 20 atmospheric pressure, the catalyst is separated off on a talc filter and the clear filtrate is evaporated in vacuo. The evaporation residue is dissolved in chlorine form methanol (1: 1) and impregnated on 10 g of silica gel 60 (Merck grain size 0.063-0.2 mm) and placed on a column of 500 g of silica gel. The subsequent elution with chloroform-methanol (4: 1) yields the hydrogenation product of SL 7810 / F.
The distances traveled by Tetrahydro-SL 7810 / F in the thin-layer chromatogram are shown in the following table:

A 0.20 / above Ce (SO4) 2 solution in 50/0 sulfuric acid can be used as a spray reagent. When the thin-layer plates are heated, Tetrahydro-SL 7810 / F gives a yellowish color.

The antibiotic derivative Tetrahydro-SL 7810 / F is easily soluble in methanol and ethanol at room temperature, sparingly soluble in chloroform, acetone, ether, petroleum ether, hexane and practically insoluble in water.
The antibiotic SL 7810 / F used as the starting material is produced as follows:
A spore suspension which is used to inoculate the nutrient solution is prepared from a culture of the originally isolated strain of Aspergillus rugulosus Thom et Raper.
The strain of Aspergillus rugulosus Thom et Raper used was isolated from a soil sample found on the Jaunpass in Switzerland and a sample of it was deposited at the United States Department of Agriculture (Northern Utilization Research and Development Division), Peoria, 111., USA and is available from the Public under the culture number NRRL 8039.
200 ml of nutrient agar of the following composition are inoculated with a spore suspension of the strain NRRL 8039: 20 g cerelose 2 g peptone
2 g of malt extract
2 g yeast extract
2 grams of primary potassium phosphate
2 g magnesium sulfate (MgSO4 7H2O) 15 g agar
1 liter of deionized water After 10 days of incubation at 27, the spores formed are suspended in 200 ml of sterile water and a steel fermenter is inoculated with it that contains 50 liters of the following nutrient solution:

0.5 g magnesium sulfate (MgSO4 7H2O) 1 mol antifoam agent 1 liter deionized water This fermenter is incubated with stirring for 42 hours at 27, the aeration being 1 liter air / mini liter medium at a stirring speed of 250 rpm.
The culture in this 50 l fermenter serves as a preculture for a 500 l fermenter which contains the same nutrient solution and is incubated at 27 for 66 hours. The aeration rate is again 1 liter of air / min / liter of medium, but the stirring speed is only 150 rpm. After an incubation period of 66 hours, the culture broth is harvested and processed.
450 l of culture broth are adjusted to pH 7.0 with 2N hydrochloric acid, 500 l of ethyl acetate are added and the mixture is homogenized with the Dispase reactor. The organic phase is then separated from the broth with the separator and washed with 50 l of water. This extraction step is repeated 2 times. The 3 extracts obtained are combined and concentrated to dryness under vacuum (water ring pump) at 20-40; Yield 320 g of crude extract.
This material is fractionated on 4 times the amount of silica gel Merck 60 (particle size 0.063-0.20 mm). The eluates with chloroform + 10% methanol and chloroform + 200 in methanol contain inactive accompanying substances. The chromatography column is then eluted with chloroform-methanol (1: 1) and the fractions with activity against Candida albicans are combined. By chromatographic purification of 16.4 g of active material on 1 kg of Merck 60 silica gel with chloroform +300/0 methanol as eluent and subsequent gel filtration of the active fractions on 1.2 kg of Sephadex LH20 with methanol, SL 7810 / F is obtained in an approximately pure form . This preparation is dissolved in acetone with warming and the clear solution is concentrated to of the volume.
After standing for 3 hours at -15, the precipitate formed is filtered off and, after washing with acetone and ether, dried for 24 hours at room temperature over P2O5 in a high vacuum. SL 7810 / F is obtained as a colorless amorphous powder with a melting point of 160-163 (on the Kofler block); [a] = -48.3 (c = 0.806 in methanol).
