Die Erfindung betrifft ein Verfahren zur Herstellung des neuen, hypotensiv wirksamen 2-(2'.6'-Dichlorphenylamino) -äthylguanidins der Formel I und seiner Säureadditionssalze.

Substituierte Phenylaminoalkylguanidine wurden bereits in dem US-Patent 3 200 151 (Brit. Prior. 15.2.1961) beschrieben. Aus dieser seit ca. 14 Jahren bekannten Verbindungsklasse konnte jedoch keine Verbindung in die Therapie der Hypertonie eingeführt werden. Im Gegensatz dazu verfügt das erfindungsgemäss hergestellte 2-(2'.6'-Dichlorphenylarnino)-äthylguanidin über solche pharmakologischen Eigenschaften, die einen vorteilhaften Einsatz dieser Verbindung zur Behandlung der Hypertonie ermöglichen.
In die vergleichenden pharmakologischen Untersuchungen, die die Überlegenheit der erfindungsgemässen Verbindung beweisen, werden neben 2-(2'.6'-Dichlorphenylamino)-äthylguanidin- -hemisulfat (1) folgende, zum Teil neue (B, D) zum Teil bekannte (E) und in der zitierten US-Patentschrift beschriebene Verbindungen (A, C) einbezogen: 2-(2'.6'-Dimethylphenylamino)-äthylguanidin-hemisulfat (A), 2-(2'.4'.6'-Trichlorphenylamino)-äthylguanidin-hemisulfat (B), 2-[N-Methyl-N- (2'.6'-dimethylphenyl)-amino]-äthylguanidin- -hemisulfat (C), 2-[N-Methyl-N-(2'.6'-dichlorphenyl)-amino]-äthylguanidin- -hemisulfat (D), 2-Guanidinomethyl-1.4-benzodioxan-hemisulfat (E).
2-Guanidinomethyl- 1 .4-benzodioxan-hemisulfat ist ein unter dem Handelsnamen Guanoxansulfat bekanntes Präparat zur Behandlung der Hypertonie und besitzt eine ähnliche Wirkungsqualität wie die vorstehend aufgeführten Verbindungen.
Für die Beurteilung der Wirkungsqualität und -quantität von Antihypertensiva, deren Wirkung auf adrenerger Neuronenblockade beruht, werden spieziell zwei Parameter herangezogen: Der Nickhautprolaps an der wachen Katze und der Grad der Veränderung des Katecholamingehaltes (karte.
cholamindepletion) im Herzen.
Die Methode Nickhautprolaps an der wachen Katze ist eine Modifikation der von Mc Lean [J. Pharmacol. exp. Ther.
129 (1960) 11] beschriebenen Verfahrensweise. Sie eignet sich zum Nachweis von sympathikolytischen Wirkungen insbesondere bei oraler Applikation am wachen Tier. Der Grad der Tonusminderung des 3. Augenlides (Palpebra tertia) wird als Wirkungsstärke in Noten von 0 bis 3 ausgedrückt.
Der Wirkungseintritt wird in Stunden und die Wirkungsdauer in Tagen angegeben. Aus der Tabelle 1 geht hervor, dass bei gleicher Dosierung die Verbindung I bezüglich Wirkungsstärke und Wirkungsdauer allen zum Vergleich herangezogenen Verbindungen wesentlich überlegen ist. Die Überlegenheit der Verbindung I gilt auch gegenüber den am aromatischen Stickstoffatom durch eine Methylgruppe substituierten Verbindungen C und D und speziell gegenüber der Verbindung C, die in der zitierten US-Patentschrift als bevorzugte Verbindung besonders hervorgehoben wird.

Aus den von uns durchgeführten Untersuchungen geht weiterhin hervor, dass auch so geringfügig erscheinende Ver änderungen im Molekül der Verbindung I, wie der Austausch der Chloratome durch Methylgruppen (Verbindung A) oder die Einführung eines zusätzlichen Chloratoms in 4-Stellung (Verbindung B), zu einer wesentlichen Abnahme der Wirkungsstärke und Wirkungsdauer bzw. zum völligen Wirkungsverlust führen (siehe Tabelle 1).
Über eine wesentlich geringere Wirkungsstärke als die Verbindung I verfügt auch das zur Behandlung der Hypertonie eingeführte Präparat Guanoxansulfat (E).
Ein weiterer Vorzug der Verbindung I gegenüber allen anderen, zum Vergleich herangezogenen Verbindungen ist der an dem Modell Nickhautprolaps gefundene verzögerte Wirkungsbeginn, der darauf hinweist, dass hier eine schonende und protrahiert einsetzende hypotensive Wirkung vorliegt.
Die Methode Katecholaminbestimmung im Herzen dient zum Nachweis einer selektiven Effektivität auf die terminale Speichergranula des sympathischen Nervensystems. Bei den Verbindungen des hier untersuchten Wirkungstyps werden die als Transmitter fungierenden Katecholamine aus ihren Speichern verdrängt, wodurch es infolge verminderter Ka techolaminaktivität zu einer Hypotension kommt. Die Organaufbereitung und die fluorimetrische Bestimmung der Katecholamine erfolgt nach einer Methode von v. Euler [Acta physiol. scand. 33 (1955) suppl. 118]. Aus dem in der Tabelle 2 aufgezeigten Grad der Katecholamindepletion im Rattenherzen ist zu entnehmen, dass die Verbindung I allen zum Vergleich herangezogenen Verbindungen überlegen ist.
Eine im Vergleich zur Verbindung I besonders geringe Wirkung zeigt auch an diesem Modell das am aromatischen Stickstoffatom durch eine Methylgruppe substituierte Derivat C.

Die akute Toxizität der untersuchten Verbindungen wurde unter intravenöser Applikation an Mäusen (männlich, 18 bis 22 g Gewicht, Stamm H) bestimmt. Die Ermittlung der LD 50 und ihrer Vertrauensgrenzen erfolgte nach der Methode von Litchfieid und Wilcoxon (Ther, Grundlagen der exp. Arzneimittelforschung, Wiss. Verl. -Ges. Stuttgart 1965). Wie aus der Tabelle 3 hervorgeht, sind die am aromatischen Stickstoffatom durch eine Methylgruppe substituierten Verbindungen zwei- bis dreimal toxischer als die übrigen Verbindungen.
Die Verbindungen A, B und C besitzen zwar die gleiche Toxizität wie die Verbindung I, unterscheiden sich aber von dieser erheblich in der Wirkungsstärke und Wirkungsdauer (siehe Tabellen 1 und 2).

Aus den vorliegenden pharmakologischen Untersuchungsbefunden ist ersichtlich, dass das erfindungsgemäss hergestellte 2-(2'.6'-Dichlorphenylamino)-äthylguanidin in nicht vorherzusehender Weise allen zum Vergleich herangezogenen Verbindungen an Wirkungsdauer und Wirkungsstärke wesentlich überlegen ist. Die starke und langanhaltende Wirkung der Verbindung I ermöglicht es, die bei der Behandlung schwerer und mittelschwerer Hypertoni efälle erforderliche Applikationsfolge pro Zeiteinheit zu verringern und bei den Patienten ein konstantes Blutdruckniveau einzustellen.
Ein besonderer Vorteil des 2-(2'.6'-Dichlorphenylamino) -äthylguanidins ist die protrahiert einsetzende Wirkung. Alle bisher bekannten Antihypertensiva, deren Wirkung auf adrenerger Neuronenblockade beruht, haben den Nachteil, dass ihre Wirkung brüsk einsetzt. Als Folge davon treten orthostatische Dysregulationen auf. Dagegen tritt diese Nebenwirkung unter der erfindungsgemäss hergestellten Verbindung nur in untergeordnetem Masse auf. Aber auch andere typische Nebenwirkungen adrenerger Neuronenblocker, wie Adynamie, Kopfschmerzen, Potenzstörungen und Diarrhoe treten vergleichsweise in geringerem Masse auf. Die therapeutische Dosis zur Behandlung hypertoner Patienten beträgt bei oraler Gabe im allgemeinen 10 bis 60 mg täglich.
Das erfindungsgemäss erhältliche 2-(2'.6'-Dichlorphenylamino)-äthylguanidin der Formel I kann in Form der freien Base oder in Form eines Säureadditionssalzes mit nicht toxischen anorganischen oder organischen Säuren hergestellt werden. Beispiele für Säureadditionssalze sind das Hydrochlorid, Hydrobromid, Hernisulfat, Phosphat, Nitrat, Acetat, Tartrat, Salicylat, Tolualsulfonaft
Gemäss der Erfindung werden das 2-(2'.6'-Dichlorphenylamino)-äthylguanidin der Formeln bzw.
dessen Salze dadurch hergestellt, dass man 2-(2'.6'-Dichlorphenylamino) -äthylamin der Formel

worin R, eine Iminogruppe und R2 eine Alkylthiogruppe mit 1 bis 3 C-Atomen, eine Altkoxygruppe mit 1 bis 3 C-Atomen oder eine gegebenenfalls substituierte Pyrazolylgruppe, oder R1 und R2 zusammen ein 3-fach an das benachbarte C-Atom gebundenes N-Atom bedeuten, umsetzt, wobei einer der Reaktionspartner der Formeln II und III als Säureadditionssalz, insbesondere mit Mineralsäuren, eingesetzt wird.
Beispiele für Verbindungen der allgemeinen Formel III sind Cyanamid, S-Methyiisothioharnstoff undl 1-Guanyl-3.5- -dimethylpyrazol und deren Salze, insbesondere Salze mit Mineralsäuren wie Salzsäure, Bromwasserstoffsäure und vorzugsweise Schwefelsäure und Salpetersäure.
Die Umsetzung von 2-(2'.6'-Dichlorphenylamino)-äthyl- amin der Formel II oder dessen Säureaddlitionssalz mit einer Verbindung der allgemeinen Formel III oder dessen Säureadditionssalz kann in Abwesenheit oder in Anwesenheit von Lösungsmitteln erfolgen. Bei Abwesenheit von Lösungsmitteln werden die Reaktionspartner der allgemeinen Formeln II und III bis zum Schmelzen erhitzt und die Reaktipn als Schmelzreaktion durchgeführt. Als Lösungsmittel eignen sich zum Beispiel Alkohole mit 1 bis 4 C-Atomen, Dimethylformamid und Gemische derselben mit Wasser. Ein bevorzugtes Lösungsmittel ist Wasser. Die Reaktion wird bei Temperaturen zwischen 20 und l500C durchgeführt.
Die Verbindungen der allgemeinen Formel III können im allgemeinen sowohl in äquimolaren Mengen als auch im Überschuss verwendet werden. Bei Einsatz von Cyanamid wird jedoch ein Überschuss an Cyanamid, vorzugsweise bis zum fünffachen (in Molen), bevorzugt.
Beispiel 1
Ein Gemisch von 10,25 g 2-(2'.6'-Dichlorphenylamino)- -äthylamin und 60 ml Wasser wird mit 2 n Salzsäure bis zum pH-Wert 7 versetzt und nach Zugabe von 25 ml 35%iger wässriger Cyanamidlösung 20 Stunden unter Rückfluss erhitzt. Nach dem Erkalten wird das ausgeschiedene Dicyandiamid abgetrennt und das Filtrat mit 20 mi 30%iger Natronlauge versetzt. Das dabei ausgefallene kristalline Produkt wird abgesaugt und im Vakuum über Phosphorpentoxid getrocknet und aus Toluol umkristallisiert.
Ausbeute 10,4 g (S4% der Theorie) 2-(2'.6'-Dichlorphenylamino)-äthylguanidin, Fp. 101-109 C.
Beispiel2
Eine Lösung von 12,1 g 2-(2'.6'-DichlorphenFylamino)- -äthylamin-hydrochiorid und 6,3 g Cyanamid in 50 ml Äthylalkohol wird 20 Stunden unter Rückfluss erhitzt. Danach entfernt man das Lösungsmittel durch Destillation und versetzt den Rückstand mit 50 ml Äther. Das dabei erhaltene kristalline Produkt wird abgesaugt und aus Äthanol/Äther umgefällt. Ausbeute 9,8 g (69% der Theorie) 2-(2'.6'-Dichlorphenylamino)-äthylguanidin-hydrochlorid, Fp. 115 bis 1200C.
Durch Wiederholung dieser Reinigungsoperation wird ein Schmelzpunkt von 127 - 1290C erreicht.
Beispiel 3
16 g 2-(2'.6'-Dichlorphenylamino)-äthylamin werden mit 2 n Sch,vefelsäure bis zum pH-Wert 7 versetzt und nach Zugabe von 130 inl 10-prozentiger wässriger Cyanamidlösung 20 Stunden unter Rückfluss erhitzt. Das nach dem Erkalten erhaltene Produkt wird abgetrennt und durch zweimaliges Umfällen aus Methanol/Wasser (10:1) gereinigt. Ausbeute 14,4 g (62,4% der Theorie) 2-(2'.6'-Dichlorphenylamino)- -äthyiguanidin-hemisulfat, Fp. 218 bis 22300.
Beispiel 4
Ein Gemisch von 214 g 2-(2'.6'-Dichlorphenylamino)- -äthylamin und 146 g S-Methylisothiuroniumsulfat in 650 ml Wasser wird 8 Stunden im siedenden Wasserbad erhitzt.
Nach dem Stehen über Nacht wird das auskristallisierte Produkt abgetrennt und aus Methanol/Wasser (10: 1) umgefällt. Ausbeute 251,4 g (83% der Theorie) 2-(2'.6'-Dichlor phenylamino)-äthylguanidin-hemisulfat, Ep 221 bis 2230C.
Beispiel 5
Zu einer Lösung von 2 g 2-(2'.6'-Dichlorphenylamino) -äthylguanidin in 3 ml Äthylalkohol werden 0,56 ml Eisessig gegeben. Die Lösung wird zur Trockne eingeengt und der Rückstand mit Äther gewaschen. Ausbeute 2,1 g 2-(2'.6' -Dichlorphenylamino > -äthylguanidin.acetat, Fp. 118 bis 1200C.

Aus einer Lösung von 2 g 2-(2'-6'-Üichlorphenylamino)- -äthylguanidin in 3 ml Äthylalkohol erhält man durch Zugabe von 1,4 g 48%ige Bromwasserstoffsäure, 2,4 g 2-(2'.6' -Dichlorphenylamino)-äthylguanidin-hydrobromid, Fp. 150 bis 15300.

Substituted phenylaminoalkylguanidines have already been described in US Pat. No. 3,200,151 (Brit. Prior. February 15, 1961). However, from this class of compounds, which has been known for about 14 years, no compound could be introduced into the therapy of hypertension. In contrast to this, the 2- (2'.6'-dichlorophenylamino) -ethylguanidine prepared according to the invention has pharmacological properties which enable this compound to be used advantageously for the treatment of hypertension.
In the comparative pharmacological investigations which prove the superiority of the compound according to the invention, in addition to 2- (2'.6'-dichlorophenylamino) ethylguanidine hemisulphate (1), the following, partly new (B, D) partly known (E ) and in the cited US patent described compounds (A, C) included: 2- (2'.6'-Dimethylphenylamino) -äthylguanidin-hemisulfat (A), 2- (2'.4'.6'-Trichlorphenylamino) ethylguanidine hemisulfate (B), 2- [N-methyl-N- (2'.6'-dimethylphenyl) amino] ethylguanidine hemisulfate (C), 2- [N-methyl-N- (2 ' .6'-dichlorophenyl) amino] ethylguanidine hemisulphate (D), 2-guanidinomethyl 1,4-benzodioxane hemisulphate (E).
2-guanidinomethyl-1,4-benzodioxane hemisulfate is a preparation known under the trade name guanoxane sulfate for the treatment of hypertension and has a similar quality of action to the compounds listed above.
To assess the quality and quantity of antihypertensive drugs, the effect of which is based on adrenergic neuron blockade, two parameters are used: the nictitating membrane prolapse in the awake cat and the degree of change in the catecholamine content (map.
cholamine depletion) in the heart.
The nictitating membrane prolapse method in the awake cat is a modification of that of Mc Lean [J. Pharmacol. exp. Ther.
129 (1960) 11] described procedure. It is suitable for the detection of sympatholytic effects, particularly when administered orally to the conscious animal. The degree of tonus reduction of the third eyelid (palpebra tertia) is expressed as potency in grades from 0 to 3.
The onset of action is given in hours and the duration of action in days. Table 1 shows that with the same dosage, compound I is significantly superior to all the compounds used for comparison in terms of potency and duration of action. The superiority of the compound I also applies to the compounds C and D which are substituted on the aromatic nitrogen atom by a methyl group and especially to the compound C, which is particularly emphasized as the preferred compound in the cited US patent.

The investigations carried out by us also show that even minor changes in the molecule of compound I, such as the replacement of the chlorine atoms with methyl groups (compound A) or the introduction of an additional chlorine atom in the 4-position (compound B), occur lead to a significant decrease in the strength and duration of action or to a complete loss of effectiveness (see Table 1).
The preparation guanoxane sulfate (E), introduced for the treatment of hypertension, is also much less potent than compound I.
A further advantage of compound I over all other compounds used for comparison is the delayed onset of action found in the model of the nictitating membrane prolapse, which indicates that there is a gentle and protracted hypotensive action here.
The method of determining catecholamines in the heart serves to demonstrate a selective effectiveness on the terminal storage granules of the sympathetic nervous system. In the case of the compounds of the type of activity examined here, the catecholamines acting as transmitters are displaced from their stores, which leads to hypotension as a result of reduced catecholamine activity. The organ preparation and the fluorometric determination of the catecholamines is carried out according to a method by v. Euler [Acta physiol. scand. 33 (1955) suppl. 118]. From the degree of catecholamine depletion in the rat heart shown in Table 2, it can be seen that compound I is superior to all compounds used for comparison.
In this model, too, the effect of the derivative C, which is substituted by a methyl group on the aromatic nitrogen atom, shows a particularly low effect compared to compound I.

The acute toxicity of the compounds investigated was determined by intravenous administration to mice (male, 18 to 22 g weight, strain H). The LD 50 and its confidence limits were determined using the method of Litchfieid and Wilcoxon (Ther, Basis der exp. Arzneimittelforschung, Wiss. Verl. -Ges. Stuttgart 1965). As can be seen from Table 3, the compounds substituted by a methyl group on the aromatic nitrogen atom are two to three times more toxic than the other compounds.
Compounds A, B and C have the same toxicity as compound I, but differ from this considerably in the strength and duration of action (see Tables 1 and 2).

From the available pharmacological test results it can be seen that the 2- (2'.6'-dichlorophenylamino) -ethylguanidine produced according to the invention is, in an unforeseeable way, significantly superior to all the compounds used for comparison in terms of duration and potency. The strong and long-lasting action of the compound I makes it possible to reduce the application sequence per unit of time required in the treatment of severe and moderate hypertension and to set a constant blood pressure level in the patient.
A particular advantage of 2- (2'.6'-dichlorophenylamino) ethylguanidine is the protracted effect. All previously known antihypertensive drugs, the effect of which is based on adrenergic neuron blockade, have the disadvantage that their effect starts abruptly. As a result, orthostatic dysregulations occur. In contrast, this side effect occurs only to a subordinate extent with the compound prepared according to the invention. But other typical side effects of adrenergic neuron blockers, such as adynamia, headache, erectile dysfunction and diarrhea, also occur to a relatively lesser extent. The therapeutic dose for the treatment of hypertensive patients is generally 10 to 60 mg daily when given orally.
The 2- (2'.6'-dichlorophenylamino) ethylguanidine of the formula I obtainable according to the invention can be prepared in the form of the free base or in the form of an acid addition salt with non-toxic inorganic or organic acids. Examples of acid addition salts are the hydrochloride, hydrobromide, hernisulfate, phosphate, nitrate, acetate, tartrate, salicylate, toluene sulfone juice

where R, an imino group and R2 an alkylthio group with 1 to 3 carbon atoms, an alkoxy group with 1 to 3 carbon atoms or an optionally substituted pyrazolyl group, or R1 and R2 together an N- bonded 3 times to the adjacent carbon atom Mean atom, converted, one of the reactants of the formulas II and III being used as an acid addition salt, in particular with mineral acids.
Examples of compounds of the general formula III are cyanamide, S-methylisothiourea and 1-guanyl-3,5- dimethylpyrazole and their salts, in particular salts with mineral acids such as hydrochloric acid, hydrobromic acid and preferably sulfuric acid and nitric acid.
The reaction of 2- (2'.6'-dichlorophenylamino) ethyl amine of the formula II or its acid addition salt with a compound of the general formula III or its acid addition salt can take place in the absence or in the presence of solvents. In the absence of solvents, the reactants of the general formulas II and III are heated until they melt and the reaction is carried out as a melting reaction. Suitable solvents are, for example, alcohols with 1 to 4 carbon atoms, dimethylformamide and mixtures thereof with water. A preferred solvent is water. The reaction is carried out at temperatures between 20 and 1500C.
The compounds of the general formula III can generally be used either in equimolar amounts or in excess. When using cyanamide, however, an excess of cyanamide, preferably up to five times (in moles), is preferred.

A mixture of 10.25 g of 2- (2'.6'-dichlorophenylamino) - ethylamine and 60 ml of water is mixed with 2N hydrochloric acid up to pH 7 and after addition of 25 ml of 35% aqueous cyanamide solution for 20 hours heated under reflux. After cooling, the precipitated dicyandiamide is separated off and the filtrate is treated with 20 ml of 30% strength sodium hydroxide solution. The crystalline product which has precipitated out is filtered off with suction and dried over phosphorus pentoxide in vacuo and recrystallized from toluene.
Yield 10.4 g (S4% of theory) 2- (2'.6'-dichlorophenylamino) ethylguanidine, melting point 101-109 C.
Example2
A solution of 12.1 g of 2- (2'.6'-DichlorphenFylamino) - ethylamine hydrochloride and 6.3 g of cyanamide in 50 ml of ethyl alcohol is heated under reflux for 20 hours. The solvent is then removed by distillation and 50 ml of ether are added to the residue. The resulting crystalline product is filtered off with suction and reprecipitated from ethanol / ether. Yield 9.8 g (69% of theory) of 2- (2'.6'-dichlorophenylamino) ethylguanidine hydrochloride, melting point 115 to 1200C.
By repeating this cleaning operation a melting point of 127-1290C is reached.

16 g of 2- (2'.6'-dichlorophenylamino) ethylamine are mixed with 2N sulfuric acid up to pH 7 and, after addition of 130 ml of 10 percent aqueous cyanamide solution, heated under reflux for 20 hours. The product obtained after cooling is separated off and purified by reprecipitating twice from methanol / water (10: 1). Yield 14.4 g (62.4% of theory) of 2- (2'.6'-dichlorophenylamino) - ethyiguanidine hemisulfate, melting point 218 to 22300.
Example 4
A mixture of 214 g of 2- (2'.6'-dichlorophenylamino) - ethylamine and 146 g of S-methylisothiuronium sulfate in 650 ml of water is heated in a boiling water bath for 8 hours.
After standing overnight, the product which has crystallized out is separated off and reprecipitated from methanol / water (10: 1). Yield 251.4 g (83% of theory) of 2- (2'.6'-dichlorophenylamino) ethylguanidine hemisulphate, Ep 221 to 2230C.
Example 5
0.56 ml of glacial acetic acid are added to a solution of 2 g of 2- (2'.6'-dichlorophenylamino) ethylguanidine in 3 ml of ethyl alcohol. The solution is concentrated to dryness and the residue is washed with ether. Yield 2.1 g of 2- (2'.6'-dichlorophenylamino> -äthylguanidine.acetate, melting point 118 to 1200C.
Examples 6
From a solution of 2 g of 2- (2'-6'-Üichlorphenylamino) - ethylguanidine in 3 ml of ethyl alcohol, by adding 1.4 g of 48% hydrobromic acid, 2.4 g of 2- (2'.6 ' Dichlorophenylamino) ethylguanidine hydrobromide, m.p. 150 to 15300.
