La présente invention a pour objet un procédé de préparation d'un nouveau dérivé de pyridine présentant en particulier une activité protectrice, surtout au niveau du foie, vis-à-vis de lésions provoquées par des agents toxiques ou infectieux et, en général, vis-à-vis d'agents qui provoquent des troubles hépatiques et circulatoires, ainsi que vis-à-vis de lésions athérosclérotiques ou inflammatoires.
Le composé préparé suivant l'invention répond à la formule:

L'invention comprend aussi la préparation des sels d'addition du composé (I) avec des acides minéraux (comme le chlorhydrate, le sulfate, etc.) ou organiques (comme le fumarate, le citrate, le tartrate, le palmoate, etc.).
Le procédé de préparation du composé de formule (I) suivant l'invention est caractérisé en ce qu'on fait réagir un halogénure de N-(x-benzoylthiopropionyl)glycine (II) sur la 2,6-dihydroxyméthylpyridine (III).
Le composé (III) est un composé connu, et le composé (II) peut être préparé par des procédés classiques bien connus.
L'exemple suivant est donné à titre d'illustration de l'invention.
Exemple:
a) Préparation du chlorure de N-(x-benzoylthiopropionyl)- glycine:
On ajoute à 5,4 g de N-(-benzoylthiopropionyl)glycine en suspension dans 25 cm3 de benzène, sous agitation, 10 g de chlorure de thionyle. On maintient le produit réactionnel à la température ambiante jusqu'à ce que tout le solide passe en solution. La solution jaune ainsi formée est évaporée sous vide, reprise avec du benzène et évaporée à nouveau jusqu'à ce que tout le chlorure de thionyle restant soit éliminé. On dissout l'huile résiduelle dans
15 cm3 de chloroforme anhydre.
b) Préparation de la bis-[N-{x-henzoylthiopropionyl)glycl lo.ry- méthyl]-2 6-pyridine:
A la solution obtenue en a) et maintenue à 10 C, on ajoute sous agitation et sous azote 1,3 g de 2,6-dihydroxyméthylpyridine dissous en 10 cm3 de pyridine anhydre. L'addition se fait sans dépasser 0 C. L'addition terminée, on laisse reposer à la température ambiante pendant 48 h, puis on évapore le solvant sous atmosphère d'azote. Le résidu est traité par une solution saturée de bicarbonate de sodium et extrait plusieurs fois avec du chloroforme. Les extraits sont séchés sur Na2SO4 et ensuite concentrés jusqu'à 25 cm3. Cette solution est chromatographiée sur une colonne de silice (150 g) en éluant avec du chloroforme.
La fraction principale, consistant en le diester désiré de 2,6-dihydroxyméthylpyridine contenant encore quelques impuretés, est chromatographiée à nouveau sur une courte colonne de silice, puis éluée avec du chloroforme. Le produit, qui fond à 86 C, présente une analyse spectroscopique (IR et RMN) et une analyse élémentaire en accord avec la structure de la formule (I) précitée.
Le composé de formule (I) présente une activité antitoxique, hépatoprotectrice et antiathérosclérotique qui le rend particulièrement intéressant en thérapeutique.
En complément d'information, on donne ci-après les résultats d'études toxicologique et pharmacologique effectuées sur le composé de formule (I).
1. Etude toxicologique
Le produit révèle une faible toxicité. Par les essais, la DLs() du produit a été évaluée à 1,45 g et 1,75 g par voie orale, chez le rat et la souris respectivement. Chez le cobaye, la DLso par voie orale est de 1,05 g.
Par voie intrapéritonéale, la DLso chez la souris est de 625 mg/kg.
La toxicité chronique du produit est aussi très favorable. Une administration quotidienne chez le rat, pendant 2 mois, de 200 mg/kg de produit par voie orale a été bien tolérée.
L'examen du poids des animaux, de la formule sanguine, de la glycémie, de l'azotémie et de l'excrétion urinaire n'a montré aucune altération.
On n'a également observé aucune altération à l'examen histologique au niveau des principaux organes.
2. Etude pharmacologique
a) Action protectrice antitoxique dans l'itztoxication par les métaux lourds
On a évalué la protection développée par le composé de formule (I) dans les intoxications expérimentales provoquées par l'administration chez la souris de chlorure de mercure ou d'éthylmercurethiosalicylate de sodium.
L'administration préalable de 200 mg/kg du composé par voie intrapéritonéale protège presque à 100% des animaux recevant par voie intrapéritonéale 20 mg/kg de chlorure de mercure ou
100 mg/kg d'éthylmercurethiosalicylate de sodium.
Le même effet de protection du produit a été observé à la dose de 150 mg/kg vis-à-vis de la mortalité provoquée chez la souris par 50 mg/kg, par voie intrapéritonéale, de sulfate de cuivre ou 200 mg/kg de phosphate de cuivre.
b) Action protectrice dans I 'i'itoxicatùm erpérimentale par le tétraclziorure de carbone
L'administration chez le rat de tétrachlorure de carbone (cul4) provoque une altération précoce du métabolisme du foie qui se manifeste en principe par une forte infiltration lipidique de cet organe.
Dans les expériences, I'administration préalable du composé de formule (I) à une dose de 100 ou 200 mg/kg par voie intrapéritonéale ou orale empêche l'infiltration lipidique au niveau du foie provoquée par l'administration orale de 10 cm3/kg d'une solution de CCl4 à 10% dans l'huile d'olive.
c) Action protectrice dans I 'i'1toxîcatù ' erpérimentale par l'éthionine
L'infiltration lipidique hépatique chez le rat, induite par l'administration par voie intrapéritonéale de I g/kg d'éthionine fractionnée en 3 doses dans un seul jour, est inhibée par l'administration dans les 8 jours précédents de 200 mg/kg par voie orale du composé de l'invention.
d) Action protectrice dans l intorication par I orygl)ne h! perha- risque
Le composé de formule (I), à une dose de 300 mg/kg par voie orale, empêche chez la souris l'apparition de l'état convulsif et la mort de la souris enfermée dans une chambre hyperbarique sous 4 atmosphères d'oxygène.
e) Action protectrice vis-à-vis de lésions vasculaires provoquées par le p-aminopropionitrile
L'angiolathyrisme induit par l'administration chez le rat mis au régime de 0,4% de a-aminopropionitrile pendant 30 j est inhibé par l'administration pendant la même période de temps de 4% du composé de formule (I). Chez les animaux traités par le produit, les lésions vasculaires sont beaucoup moins évidentes, même à l'examen microscopique.
f) Action antiathérosclérotique
L'administration chez le lapin d'un régime hypercholestérolé miant (1% de cholestérol dans le régime alimentaire) pendant
10 semaines et, en même temps, de 20 mg/kg/j du composé de formule (I) empêche l'hypercholestérolémie et la formation de lésions athérosclérotiques au niveau de l'aorte et des artères.
g) Action antibradykinine
L'action phlogogène de la bradykinine se trouve réduite par l'administration du composé de formule (I). 200 à 400 mg/kg du composé par voie orale empêchent, en effet, I'oedéme de la patte du rat produit par l'injection dans celle-ci de 0,5 cm3 d'une solution de bradykinine.
h) Action sur l'agrégation des plaquettes
La présence du composé de formule (I) inhibe in vitre et in vho l'agrégation des plaquettes induites par l'A DP (adénosine diphosphate).
Le composé de formule (I) est utilisable en thérapeutique humaine pour ses activités antitoxiques, antihypercholes térolémiante et anti-inflammatoire, dans la thérapie et la prévention des lésions hépatiques d'origine toxique, réactive ou métabolique, ainsi que dans la thérapie et la prévention de l'athérosclèrose et dans toutes les maladies nécessitant un apport soufré.
Il peut être administré par voie orale, parentérale, rectale ou topique. Pour ces divers modes d'administration, il est présenté sous forme de comprimés, de capsules, de gélules, de suppositoires ou de pommade, dans lesquels le principe actif est associé aux véhicules ou excipients appropriés à ces présentations pharmaceutiques.
Chaque dose unitaire peut contenir de 0,1 à I g de principe actif.
On donne ci-après quelques formulations pharmaceutiques:
a) comprimés, capsules ou gélules contenant 0,25 g de principe actif dans un excipient approprié;
b) solution injectable, contenant 0,1 g de principe actif/cm3, dans un solvant approprié:
c) suppositoires, contenant 0,5 g de principe actif dans une base pour suppositoires:
d) pommade, contenant 10% de principe actif dans une base appropriée à ce type de préparation pharmaceutique.
Les doses susceptibles d'être administrées en 24 h varient selon l'application envisagée.
La dose moyenne est de 1-2 comprimés, capsules ou gélules, 2-3 fois par jour par administration orale; I injection intramusculaire ou intraveineuse 1 ou 2 fois par jour; I ou 2 suppositoires par jour par voie rectale: une application topique de la pommade 3-4 fois par jour.

In addition to information, the results of toxicological and pharmacological studies carried out on the compound of formula (I) are given below.
1. Toxicological study

The same protective effect of the product was observed at a dose of 150 mg / kg with respect to the mortality caused in mice by 50 mg / kg, intraperitoneally, of copper sulphate or 200 mg / kg of copper phosphate.
b) Protective action in the experimental i'itoxicom by carbon tetraclzioride
The administration in rats of carbon tetrachloride (cul4) causes an early deterioration of the metabolism of the liver which is manifested in principle by a strong lipid infiltration of this organ.

f) Anti-atherosclerotic action

g) Antibradykinin action
The phlogogenic action of bradykinin is reduced by the administration of the compound of formula (I). 200 to 400 mg / kg of the compound orally prevent the edema of the rat paw produced by the injection therein of 0.5 cm 3 of a bradykinin solution.
h) Action on platelet aggregation
The presence of the compound of formula (I) inhibits in vitre and in vho the aggregation of platelets induced by DPP (adenosine diphosphate).
The compound of formula (I) can be used in human therapy for its antitoxic, antihyperchola terolemic and anti-inflammatory activities, in the therapy and prevention of hepatic lesions of toxic, reactive or metabolic origin, as well as in therapy and prevention of atherosclerosis and in all diseases requiring a sulfur intake.
It can be administered orally, parenterally, rectally or topically. For these various modes of administration, it is presented in the form of tablets, capsules, hard capsules, suppositories or ointment, in which the active principle is combined with the vehicles or excipients suitable for these pharmaceutical presentations.
Each unit dose can contain from 0.1 to 1 g of active principle.
Some pharmaceutical formulations are given below:
a) tablets, capsules or hard capsules containing 0.25 g of active principle in an appropriate excipient;
b) solution for injection, containing 0.1 g of active ingredient / cm3, in an appropriate solvent:

The doses likely to be administered in 24 hours vary according to the envisaged application.
The average dose is 1-2 tablets, capsules or capsules, 2-3 times a day by oral administration; I intramuscular or intravenous injection once or twice a day; I or 2 suppositories per day rectally: topical application of the ointment 3-4 times per day.
