Gegenstand der vorliegenden Erfindung ist die Herstellung neuer Derivate des Rifamycins S- und -SV mit hoher antibiotischer Wirksamkeit. Es handelt sich um in 3-Stellung durch eine aliphatisch substituierte Aminogruppe substituierte Rifamycin S-Verbindungen (I) der folgenden Formel

und ihre entsprechenden Hydrochinone, die Rifamycin SV-Derivate (Verbindungen II), worin A einen gegebenenfalls an den C-Atomen durch Niederalkylgruppen substituierten 4R-1-Piperazinylrest bedeutet, wobei R ein gesättigter oder ungesättigter cycloaliphatischer oder cycloaliphatischaliphatischer Kohlenwasserstoffrest darstellt. In den ungesättigten cycloaliphatisch-aliphatischen Verbindungen können Doppelbindungen sowohl im cycloaliphatischen wie im aliphatischen Teil vorhanden sein. Der genannte Rest R ist somit z.
B. eine im cycloaliphatischen Teil durch Alkylgruppen, insbesondere Niederalkylgruppen, in erster Linie Methylgruppen substituierte Cycloalkyl-, Cycloalkenyl-, Cycloalkyl Alkyl-, Cycloalkenyl-alkyl- oder Cycloalkenyl-alkenyl-Gruppe, wobei die Cycloalkylreste vorzugsweise 3 bis 8 Ring C-Atome und vor allem 5 bis 6 Ring C-Atome und die Cycloalkenylgruppen vorzugsweise 5 bis 8 Ring C-Atome aufweisen. In den Cycloalkyl- bzw. Cycloalkenyl-alkyl- und -alkenyl Resten ist die Alkylen bzw. Alkenylengruppe vorzugsweise eine niedere Alkylen bzw. Alkenylgruppe mit 1 bis 7 C-Atomen.
Beispiele für die genannten niederen Alkylgruppen sind: Methyl, Äthyl, Propyl, Isopropyl, gerade oder verzweigte, in beliebiger Stellung verbundene Butyl-, Amyl-, Hexyl- oder Heptylgruppen. Cycloalkylgruppen mit 3 bis 8 C-Atomen im Ring sind z. B. Cyclopropyl-, Cyclobutyl-, Cyclopentyl, Cyclohexyle Cycloheptyl-, 4-Methyl-cyclohexyl-, 2,6-Dimethyl-cyclohexyl-, 3,5-Dimethyl-cyclohexyl-Gruppen. Unter den Cycloalkenyl-Resten sind z. B. zu nennen:
1-Cyclopentenyl, 2-Cyclopentenyl, 3-Cyclopentenyl, 2,4-Cyclopentadienyl, 1 -Cyclohexenyl, 2-Cyclohexenyl, 3-Cyclohexenyl- und Iplyclohexadienyl. In den oben genannten Cycloalkyl- alkyl- oder Cycloalkenyl-alkyl-Resten sind z. B. die soeben genannten Reste jeweils in 1-Stellung z.
B. an einen Me thylen-, Athylen-, Trimethylen-, Tetramethylen-, Pentamethy len-, Hexamethylen-, Heptamethylen- oder an einen Propylen-, 1,2-Dimethyläthylen-, 1,1,2,2-Tetramethyläthylen-Rest und in den Cycloalkyl- bzw. Cycloalkenylalkenyl-Resten z. B.
an 1- oder 2-Propenylen oder 1,3-Butadienylen gebunden.
Gegebenenfalls vorhandene niedere Alkylgruppen an den C-Atomen des Piperazinrests besitzen 1 bis 7 C-Atome; es sind vor allem Methylgruppen, wobei in erster Linie 1 oder 2 solcher Gruppen vorhanden sind.
Unter den Verbindungen (I) und (II) sind besonders solche hervorzuheben, in welchen die Gruppe A einen an den C-Atomen unsubstituierten 4R-1 -Piperazinylrest bedeutet, worin R einen cycloaliphatisch-aliphatischen Rest darstellt, insbesondere einen Cycloalkylniederalkyl und einen Cycloalkenylniederalkyl-Rest wobei der Cycloalkylrest vorzugsweise 3 bis 8 Ring C-Atome, der Cycloalkenyl-Rest 5 bis 8 C-Atome aufweist, und der Niederalkyl-Teil 1 bis 4, insbesondere 1 oder 2, C-Atome besitzt, sowie entsprechende Verbindungen, die im Niederalkyl-Teil von R eine oder eventuell zwei Doppelbindungen aufweisen. Im Niederalkyl-Teil mit 1 bis 4 C-Atomen kann die Kohlenstoffkette gerade oder verzweigt sein; Doppelbindungen gehen vorzugsweise nicht von dem am Stickstoff des Piperazinrestes benachbarten Kohlenstoffatom aus.
Unter diesen Verbindungen seien jene genannt, in denen der Niederalkylteil eine Methylengruppe darstellt und der cycloaliphatische Teil einen Cyclopentyl-, Cyclohexyl- oder entsprechende mono-ungesättigte oder durch Methylgruppen, insbesondere 1 bis 2 Methylgruppen, substituierte Reste bedeuten.
Unter den neuen Verbindungen sind insbesondere zu nennen: das 3-(4-CyclohexylmethyI-l-piperazinylt rifamycin S und -SV das 3i4-(4-Methylcyclohexyl-methyl)- l-piperazinyl)rifamycin S und -SV das 3i4(4-Tert butyl-cyclohexyl-methyl > 1-piperazinyl}rifamycin S und -SV das 3{444-lsobutylcyclohexyl-methylF 1-piperazinyl}rifamycin S und -SV das 3+4(4-Isopropylcyclohexyl-methyl > 1-piperazinylirifamycin S und -SV das 3-C4-(3-Methylcyclohexyl-methylt 1-piperazinylirifamycin S und
-SV das 3+43,4Dimethylcyclohexyl-methylY 1-piperazinylirifamycin S und -SV das 3{443,5-Dimethylcyclohexyl-methylS 1-piperazinyl}rifamycin S und -SV das 3i4-(3-Cyclohexenyl-methyl)-l-piperaziny rifamycin S und -SV das 3 < 4-Cyclopropylmethyl-1-piperazinylY rifamycin S und -SV das 3*-Cycloheptylmethyl-1-piperazinylS rifamycin S und -SV das 3*42-Cyclohexyl-2-PropylS 1-piperazinylirifamycin S und -SV oder das 3-[4-(2-Cyclohexyl-2-butyl)-piperazinyl]- rifamycin S und -SV.
3-Aminorifamycin S- und -SV-Verbindungen mit antibioti scher Wirkung sind bereits bekannt. So werden im Patent
Nr. 512 510 Rifamycin S- und -SV-Verbindungen, die in 3-Stel lung durch eine Aminogruppe aliphatischen Charakters sub stituiert sind, beschrieben. Jene Verbindungen besitzen eine hohe antibiotische Wirkung und weisen auch einen antituber kulösen Effekt auf. Die Verbindungen der vorliegenden An meldung zeichnen sich nun gegenüber den genannten Verbin dungen des französischen Patentes 1490183 und auch gegen über anderen bekannten Rifamycin-Derivaten mit antituber kulöser Wirkung durch einen erhöhten antituberkulösen Ef fekt und durch geringere Toxizität aus, wie sich in vivo nach weisen lässt.
Die hohe antituberkulöse Wirkung der neuen Verbindun gen kann z. B. an der Maus nachgewiesen werden. So zeigen die Verbindungen bei peroraler Verabreichung an mit Mycobacterium bovis infizierten Mäusen eine ausgesprochene tuberkulostatische Wirkung (ED 50) in Dosen zwischen 0,5 mg/kg. und 40 mg/kg. Zum Beispiel weist das 344-Cyclohe- xylmethyl-1-piperazinyl > nfamycin SV in diesem Test eine EDso von 1 mg/kg. Die Verbindungen zeichnen sich zudem durch eine sehr grosse therapeutische Breite aus, indem eine nennenswerte Toxizität erst bei sehr hoher Dosis auftritt. So beträgt z; B. die LDso bei der oben genannten spezifischen Verbindung einen Wert von über 1000 mg/kg.
Die neuen Verbindungen können daher als Heilmittel in erster Linie für tuberkulöse Infektionen, aber auch für andere Infektionen, wie z. B. für Lepra, verwendet werden. Die neuen Verbindungen sind aber auch wertvolle Zwischenprodukte für die Herstellung anderer nützlicher Stoffe, insbesondere von pharmakologisch wirksamen Verbindungen. Sie können ferner als Futtermittelzusätze und zur Konservierung von Nahrungsmitteln verwendet werden.
Gemäss der vorliegenden Erfindung werden die neuen Verbindungen I und II nach dem Verfahren des Patentes 512 510 erhalten, d. h. durch Umsetzung des Rifamycin S mit einem Amin der Formel HA, worin A die gleiche Bedeutung wie oben hat.
Das erhaltene 3-Amino-Substitutionsprodukt des Rifamycins S oder Rifamycins SV kann direkt aus dem Reaktionsgemisch isoliert werden. Wenn erwünscht, kann vor oder nach der Isolierung ein erhaltenes Hydrochinon zum Chinon oxydiert oder ein erhaltenes Chinon zum Hydrochinon reduziert, und/oder eine erhaltene Verbindung in ein Salz übergeführt werden.
Die Umsetzung des Rifamycin S mit dem Amin HA wird zweckmässig in einem Hydroxygruppen-freien organischen
Lösungsmittel, vorzugsweise von geringer Polarität, wie halogenierten aliphatischen Kohlenwasserstoffen, wie Methylenchlorid, Chloroform, Estern oder Äthern, wie z. B. Essig ester, Butylacetat, Amylacetat oder Cellosolve oder in Tetrahs drofuran und in erster Linie in Dioxan ausgeführt. Wenn das umzusetzende Amin flüssig ist, kann auch auf die Zugabe eines Lösungsmittels verzichtet werden. Man verwendet zweckmässig einen Überschuss (5 bis 10 Mol) an Amin. Die Reaktion wird vorteilhaft bei Zimmertemperatur oder, z. B.
bei langsamem Verlauf, bei erhöhter Temperatur, z. B. zwischen Zimmertemperatur und 100 , durchgeführt. Der Verlauf der Reaktion kann dünnschichtchromatographisch verfolgt werden.
Das Reaktionsprodukt liegt in der Reaktion teilweise in
Form des Chinons und teilweise in Form des Hydrochinons vor. Es ist vorteilhaft, das Reaktionsprodukt vor der Aufarbeitung in dieser Hinsicht zu vereinheitlichen, d. h., das vorhandene Hydrochinon zum Chinon zu oxydieren bzw. das vor handene Chinon zum Hydrochinon zu reduzieren, und dann das 3-Amino-Derivat in der einen bzw. der anderen Form zu isolieren. Die Oxydation wird vorteilhaft mit anorganischen
Oxydationsmitteln, vorzugsweise Ferricyankalium vorgenom men, die Reduktion vorzugsweise mit Ascorbinsäure oder Na-dithionit.
Die Isolierung des Reaktionsproduktes aus den so erhaltenen Reaktionsgemischen oder auch aus den unmittelbar er haltenen Reaktionsgemischen erfolgt in an sich bekannter
Weise, z. B. durch Verdünnen mit Wasser, und/oder, gegebe nenfalls durch Neutralisieren mit einer wässerigen Säure, z. B. Mineralsäure oder vorteilhaft Zitronensäure, und Zugabe eines eines mit Wasser nicht mischbaren Lösungsmittels, wie z. B. eines chlorierten Kohlenwasserstoffs, z. B. Chloroform oder Methylenchlorid, wobei das Reaktionsprodukt in die organische Phase übergeht, aus welcher es durch die üblichen Methoden d. h. Trocknen, Abdampfen und Kristallisation und/oder Chromatographie oder anderen üblichen Reinigungsmethoden in reiner Form erhalten werden kann.
Die solchermassen gewonnenen Chinone bzw. Hydrochinone können leicht ineinander übergeführt werden, z. B.
durch Behandlung mit den oben genannten Reduktions- bzw.
Oxydationsmitteln.
Die Chinone sind meist violettrot gefärbte Verbindungen. Die Hydrochinone sind meist gelb gefärbt und kristallisieren gut. Die Hydrochinone bilden Metallsalze, von denen besonders die Alkalisalze wegen deren Wasserlöslichkeit Bedeutung haben. Mit Säuren bilden die Chinone und die Hydrochinone Säureadditionssalze und gegebenenfalls auch quaternäre Ammoniumsalze, insbesondere mit Estern niederer Alkanole mit Halogenwasserstoffsäuren, Schwefelsäuren oder Sulfonsäuren. Zur Herstellung der Säureadditionssalze verwendet man vor allem Säuren, die zur Bildung therapeutisch verwendbarer Salze geeignet sind.
Als solche seien beispielsweise genannt: Halogenwasserstoffsäuren, Schwefelsäuren, Phosphorsäuren, Salpetersäure, Perchlorsäure; aliphatische, alicyclische, aromatische oder heterocyclische Carbonoder Sulfonsäuren, wie Ameisen-, Essig-, Propion-, Bernstein-, Glykol; Milch-, Äpfel-, Wein-, Zitronen-, Ascorbin-, Malein-, Hydroxymalein- oder Brenztraubensäure; Phenylessig, Benzoe-, p-Aminobenzoe-, Anthranil-, p-Hydroxybenzoe-, Salicyloder p-Aminosalicylsäure, Embonsäure, Methansulfon-, Äthansulfon-, Hydroxyäthansulfon-, Äthylensulfonsäure; Halogenbenzolsulfon-, Toluolsulfon-, Naphthalinsulfonsäuren oder Sulfanilsäure; Methionin, Tryptophan, Lysin oder Arginin.
Diese oder andere Salze der neuen Verbindungen, wie z. B. die Pikrate, können auch zur Reinigung der erhaltenen Basen dienen, indem man die Basen in Salze überführt, diese abtrennt und aus den Salzen wiederum die Basen freimacht.
Infolge der engen Beziehung zwischen den Basen in freier Form und in Form ihrer Salze sind im vorausgegangenen und nachfolgend unter den freien Basen sinn- und zweckgemäss gegebenenfalls auch die entsprechenden Salze zu verstehen.
Die neuen Verbindungen können z. B. in Form pharmazeutischer Präparate Verwendung finden. Diese enthalten die Verbindungen in Mischung mit einem für die enterale, topische oder parenterale Applikation geeigneten pharmazeutischen organischen oder anorganischen, festen oder flüssigen Trägermaterial. Für die Bildung desselben kommen solche Stoffe in Frage, die mit den neuen Verbindungen nicht reagieren, wie z. B. Wasser, Gelatine, Laktose, Stärke, Stearylalkohol, Magnesiumstearat, Talk, pflanzliche Öle, Benzylalkohole, Gummi, Propylenglykol, Polyalkylenglykole, Vaseline, Cholesterin oder andere bekannte Arzneimittelträger. Die pharmazeutischen Präparate können z. B. als Tabletten, Dragees, Salben, Cremen, Kapseln oder in flüssiger Form als Lösungen, Suspensionen oder Emulsionen vorliegen.
Gegebenenfalls sind sie sterilisiert und/oder enthalten Hilfsstoffe, wie Konservierungs-, Stabilisierungs-, Netz- oder Emulgiermittel, Lösungsvermittler oder Salze zur Veränderung des osmotischen Drucks oder Puffer. Sie können auch noch andere therapeutisch wertvolle Substanzen enthalten. Die Präparate werden nach üblichen Methoden erhalten.
Die Erfindung betrifft auch jene Ausführungen des oben beschriebenen Verfahrens zur Herstellung der neuen 3-Aminorifamycin-Verbindungen, bei denen man von einer auf irgendeiner Stufe als Zwischenprodukt erhältlichen Verbindung ausgeht und die fehlenden Verfahrenschritte durchführt oder die Ausgangsstoffe unter den Reaktionsbedingungen bildet.
Die neuen Verbindungen können auch in der Tiermedizin, z. B. in einer der oben genannten Formen, verwendet werden.
Die Erfindung wird in den nachfolgenden Beispielen be schrieben. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel 1
Man versetzt eine Lösung von 30 g Rifamycin S in 100 ml Dioxan mit 30 g 1-Cyclohexylmethylpiperazin und lässt bei Raumtemperatur solange stehen, bis die anfangs violettblaue Farbe des Reaktionsgemisches nach orangebraun umgeschlagen ist. Hierauf versetzt man mit Wasser und säuert mit Zitronensäure an. Das gewünschte Material wird mit Chloroform aufgenommen. Nach dem Trocknen und Eindampfen der Chloroformlösung löst man den verbleibenden Rückstand in wenig Methanol und setzt tropfenweise konzentrierte wässrige Ascorbinsäurelösung zu, bis die anfangs dunkle Färbung der Methanollösung goldgelb geworden ist.
Nach kurzem Stehen kristallisiert 3(4-Cyclohexylmethyl-pipe- razin-1-ylnfamycin-SV, das nach zweimaligem Umkristallisieren aus Chlorofrom-Methanol-Wasser völlig rein erhalten wird. Smp. i76-178". In der Mutterlauge der ersten Kristallisation befindet sich neben dem gewünschten Reaktionsprodukt fast ausschliesslich Rifamycin-SV.
UV-Spektrum in 0,01 n alkohol HCI, Maxima in nm log e): 228(4,53), 298(4,28), 433(3,90).
Beispiel 2
In völlig analoger Arbeitsweise wie im vorigen Beispiel beschrieben entsteht aus 25 g Rifamycin S in 40 ml Dioxan mit 30 g 1-Cyclopropylmethylpiperazin das 344-Cyclopropyl- methyl-piperazin-i-ylfrifamycin-SV, das nach dreimaliger Kristallisation aus Chloroform-Methanol-Wasser in gelben Kristallen vom Smp. 217 erhalten wird. UV-Spektrum in 0,01 n alkohol, HCI, Maxima in nm (log s): 225(4,54); 272(4,30); 430(3,81).
Beispiel 3
Pharmazeutische Präparate enthaltend 344-Cyclohexyl- methyl-1-piperazinylfrifamycin-SV als Antibiotikum in Form von Steckkapseln.
Zusammensetzung 3 -(4-Cyclohexylmethyl 1-piperazinyl)-riSamycin SV 100,00 mg Lactose 50.00 mg Aethylcellulose 1,50 mg Stearinsäure 1,50 mg
153,00 mg Herstellung
1. Die aktive Substanz wird mit der Lactose gemischt.
2. Man löst die Äthylcellulose in der 10fachen Menge Methylenchlorid.
3. Die Mischung 1) wird mit der Lösung 2) befeuchtet und durch ein Sieb mit 3 bis 5 mm Maschenweite geschlagen, und bei einer 40 nicht übersteigenden Temperatur getrocknet.
4. Das trockene Granulat wird durch ein Sieb mit 0,5 mm Maschenweite geschlagen und mit der pulverförmigen Stearinsäure vermischt. Die Mischung wird sodann in Steckkapseln der Grösse 2 abgefüllt.
Beispiel 4
In analoger Weise wie im Beispiel 1 beschrieben setzt man 10 g Rifamycin S mit 10 g N-Cyclobutylmethyl-piperazin um und erhält so das 344-Cyclobutylmethyl-1-piperazinyl)- rifamycin-SV in gelben Kristallen ab, die nach zweimaligem Umkristallisieren aus Chloroform-Methanol-Wasser sich oberhalb 195 "C langsam zersetzen. Weitere Mengen befinden sich in den beiden letzten Mutterlaugen.
UV-Spektrum in 0,01 n alkohol. HCI, Maxima in nm (log s): 230 (4,58), 299 (4,30), 435 (3,90).
Beispiel 5
In analoger Weise wie im Beispiel 1 beschrieben setzt man 10 g Rifamycin S mit 12 g N-Cyclopentylmethyl-piperazin um und erhält so das 3(4-Cyclopentylmethyl-1-piperazi- nylfrifamycin-SV von Smp. 180-185 "C (Zersetzung).

Setzt man Rifamycin S in der unten angegebenen Weise mit den in der ersten Spalte der untenstehenden Tabelle angeführten N-substituierten Piperazinen um, so erhält man die durch den entsprechend N'-substituierten 1-Piperazinylrest in 3-Stellung substituierten Rifamycin-SV-Derivate, die die in der Tabelle angeführten physikalischen Daten besitzen.

The subject of the present invention is the production of new derivatives of rifamycins S and -SV with high antibiotic effectiveness. They are rifamycin S compounds (I) of the following formula which are substituted in the 3-position by an aliphatically substituted amino group

and their corresponding hydroquinones, the rifamycin SV derivatives (compounds II), in which A is a 4R-1-piperazinyl radical which is optionally substituted on the carbon atoms by lower alkyl groups, where R is a saturated or unsaturated cycloaliphatic or cycloaliphatic-aliphatic hydrocarbon radical. In the unsaturated cycloaliphatic-aliphatic compounds, double bonds can be present both in the cycloaliphatic and in the aliphatic part. The said radical R is thus z.

Any lower alkyl groups present on the carbon atoms of the piperazine radical have 1 to 7 carbon atoms; they are mainly methyl groups, with 1 or 2 such groups being present in the first place.
