Die vorliegende Erfindung bezieht sich auf ein Verfahren zur Herstellung von neuen, wirksamen Acylderivaten des Antibiotikums WS-4545, die gegen eine Anzahl Mikroorganismen wirksam sind und zu pharmazeutischen Präparaten verarbeitet und therapeutisch bei der Behandlung von Infektionen verwendet werden können.
Mit den neuen Verbindungen lassen sich pharmazeutische Präparate herstellen, die als wirksame antibakterielle Mittel Acylderivate des genannten Antibiotikums WS-4545 enthalten, wobei diese Präparate zur Behandlung von durch Bakterien verursachten Infektionskrankheiten bei Mellschen und Tieren dienen können.
Das Antibiotikum WS-4545 entspricht der folgenden Formel

und wird dadurch erhalten, dass man Streptomyces sapporonensis ATCC-21532 oder Mutanten desselben unter aeroben Bedingungen in einem Nährboden fermentiert.
Der für die Herstellung des neuen Antibiotikums WS4545 brauchbare Mikroorganismus ist eine neu entdeckte Streptomycesart, die aus einer in der Stadt Sapporo in Japan gefundenen Erdprobe isoliert wurde. Eine Kultur des lebenden Organismus wurde bei der American Type Culture Collection, unter der Nummer ATCC 21532 hinterlegt und der dauernden Vorratskultursammlung derselben einverleibt; der Mikroorganismus wird im folgenden als Streptomyces sapporonensis bezeichnet.
Selbstverständlich ist die Erfindung nicht auf die Verwendung des hier beschriebenen speziellen Organismus oder von Organismen, die den hier zur Erläuterung beschriebenen Wachstums- und Mikroskopischen Eigenschaften vollständig entsprechen, für die Herstellung des neuen Antibiotikums beschränkt. Es können auch aus dem beschriebenen Mikroorganismus in üblicher Weise, wie mit Röntgenstrahlen, ultravioletter Strahlung und Stickstofflost, erzeugte Mutanten für die Zwecke der Erfindung verwendet werden.

Serumspiegel und Ausscheidung im Urin bei gesunden
Freiwilligen nach intramuskulärer Verabreichung
Die Serum- und Urinspiegel wurden bei 5 gesunden Freiwilligen bestimmt. 5 erwachsene männliche Freiwillige mit Gewichten im Bereich von 60,0 bis 75,0 kg wurde WS-4545 in einer einzigen Dosis von 1000 mg bzw. 5 mg intramuskulär verabreicht, 0,5, 1, 2, 3 und 5 Stunden nach der Verabreichung wurden Blutproben entnommen. Urinproben wurden in den Zeiträumen 0-1, 1-3, 3-6, 6-8, 8-10 und 10-24 Stunden nach der Verabreichung gesammelt. Die Konzentrationen von WS-4545 in dem Serum und dem Urin wurden nach dem Schalenverfahren bestimmt. Es wurden folgende Resultate erhalten:

Es wurde gefunden, dass das Antibiotikum WS-4545, welches Hydroxylgruppen hat, mit einem Acylierungsmittel leicht zu den entsprechenden Acylderivaten acyliert werden kann. Mit anderen Worten: das Antibiotikum WS-4545 wird mit einer Carbonsäure oder einem reaktionsfähigen Derivat davon zu einem WS-4545-Ester dieser Carbonsäure umgesetzt.
Als Acylierungsmittel können für die Zwecke der Erfindung Carbonsäuren der Formel
R-OH [II] worin R eine von einer Carbonsäure abgeleitete Acylgruppe darstellt, und deren reaktionsfähige, von der Carboxylgruppe abgeleitete Derivate verwendet werden; als Acylierungsmittel kommen beispielsweise aliphatische Carbonsäuren, aromatische Carbonsäuren, heterocyclische Carbonsäuren und deren reaktionsfähige Derivate in Betracht.
In der Formel II kann R die folgenden Bedeutungen haben: eine gesättigte oder ungesättigte aliphatische Kohlenwasserstoffcarbonylgruppe, wie beispielsweise Alkanoyl, z. B.
Acetyl-, Propionyl-, Butyryl-, Isobutyryl-, Valeryl-, Isovaleryl-, Pivaloyl-, 2-Äthylbutyryl-, Caproyl-, Palmitoyl-Stearoylreste, usw., mit 2 bis 20 Kohlenstoffatomen und Alkenoyl, zum Beispiel Acryloyl-, Methacryloyl-, Crotonoyl-, Oleoyl-, Linoleoyl-, Linolenoylreste, usw., mit 2 bis 20 Kohlenstoffatomen, oder gesättigte oder ungesättigte cycloaliphatische Kohlenwasserstoffcarbonylreste, wie beispielsweise Cycloalkancarbonyl-, z. B. Cyclohexalcarbonyl- und Cycloalkencarbonyl, z. B. Cyclohexenylcarbonyl, oder durch cycloaliphatische Kohlenwasserstoffreste substituierte aliphatische Kohlenwasserstoffcarbonylgruppen, wie beispielsweise Cycloalkanalkanoyl, z. B. Cyclohexylacetyl, Cyclohexylpropionyl usw., mit 7 bis 13 Kohlenstoffatomen.
Ferner kann die aliphatische Kohlenwasserstoffgruppe der aliphatischen Carbonsäure unverzweigt oder verzweigt oder durch ein Heteroatom, wie Sauerstoff oder Schwefel, unterbrochen sein. Beispiele solcher Reste sind Methoxyacetyl-, Methylthioacetyl-, Butylthioacetyl-, Methoxycarbonyl-, Äthoxycarbonyl-, Cyclohexylthioacetylreste usw.
In den aromatischen Carbonsäuren der Formel II kann R einen Aroylrest, z. B. Benzoyl, Toluoyl, Xyloyl, Naphthoyl usw., oder eine arylsubstituierte aliphatische Kohlenwasserstoffcarbonylgruppe, wie Aralkanoyl, z. B. Phenylacetyl-, Phenylpropionyl-, a-Methyl-a-toluoylreste usw., und Aralkenoyl, z. B. Cinnamoyl usw., darstellen. Die aliphatische Kohlenwasserstoffgruppe, d. h. die Alkan- oder Alkengruppe, mit 1 bis 6 Kohlenstoffatomen, die Bestandteil dieser arylsubstituierten aliphatischen Kohlenwasserstoffcarbonylgruppen, d. h. Aralkanoyl- bzw.
Aralkenoylgruppen ist, kann durch ein Heteroatom, wie Sauerstoff oder Schwefel, unterbrochen sein; Beispiele solcher Reste sind Phenoxyacetyl, Phenylthioacetyl usw;
In den heterocyclischen Carbonsäuren der Formel II kann R eine durch einen heterocyclischen Rest substituierte Carbonylgruppe sein, wobei dieser heterocyclische Reste monocyclisch oder bicyclisch oder mit Benzol anelliert sein kann, 5 oder 6 Ringglieder enthält und mindestens eines der Heteroatome Stickstoff, Sauerstoff und Schwefel aufweist; Beispiele solcher Reste sind Nicotinoyl, Isonicotinoyl, 2-Furoyl, 2 Thenoyl- Benzofuroyl, Benzothenoyl, usw. R kann auch einen derartigen Rest darstellen, der durch eine aliphatische Koh lenwasserstoffcarbonylgruppe, z.
B. eine Alkanoyl- oder Alkenoylgruppe, substituiert ist, wie beispielsweise 1H- oder 2H Tetrazolylacetyl-, Thienylacetyl-, Furylacetyl-, 3-Benzothiazolylacetyl-, 2-Oxo-3-benzothiazolylacetyl-, 3-Indolylacetyl-, Morpholinoacetylreste, usw.; die aliphatische Kohlenwasserstoffgruppe, d. h. die Alkan- oder Alkengruppe, kann durch ein Heteroatom, wie Sauerstoff und Schwefel, unterbrochen sein und 1 bis 6 Kohlenstoffatome enthalten.
Bei den oben erwähnten aliphatischen, aromatischen und heterocyclischen Carbonsäuren kann die aliphatische Kohlenwasserstoffgruppe, d. h. die Alken-, Alkan-, Cycloalkan- bzw.
Cycloalkengruppe, der aromatische Ring, d. h. der Arylrest, und der heterocyclische Ring durch einen oder mehrere Substituenten substituiert sein, wie beispielsweise Halogen, z. B.
Chlor, Brom, Fluor usw., Alkoxy, z. B. Methoxy, Äthoxy usw., Amino, Acylamino, z. B. Acetylamino, Acyloxy, z. B.
Acetoxy, Nitro, Hydroxyl usw. Beispiele solcher substituierter Carbonsäuren sind: Trichlorpropionsäure, Benzolylglycin, Brombenzoesäure, Chlorbenzoesäure, Nitrobenzoesäure, Methoxybenzoesäure, 3,4-Dimethoxybenzoesäure, 3,4-Dimethylbenzoesäure, 3,4,5-Trimethoxybenzoesäure, 3,4-Dimethoxyzimtsäure, 4-Chlorzimtsäure, Chlorphenylpropionsäure, Chlorphenoxyessigsäure, 5-Nitrofuran-2-carbonsäure, 5 Chlorbenzofuran-2-carbonsäure, 5-Chlor-2-oxo-3-benzothiazolylessigsäure usw.
Beispiele von reaktionsfähigen Derivaten der Carbonsäuren der Formel II sind Säurehalogenide, z. B. Säurechloride, Säurebromide usw., Säureanhydride, z. B. gemischte Säureanhydride mit Alkylschwefelsäuren, Alkylphosphorsäuren, aliphatischen oder aromatischen Carbonsäuren usw., Säureamide, z. B. von Imidazol, in 4-Stellung substituiertem Imidazol usw. abgeleitete Säureamide, Säureester, z. B. Methylester, Äthylester, Cyanomethylester, p-Nitrophenylester usw., und Säureazide. Wenn man die Carbonsäure als solche oder in Form eines Salzes, z. B. des Natriumsalzes, Kaliumsalzes usw., verwendet, ist es manchmal empfehlenswert, für die Reaktion ein Kondensationsmittel zu verwenden. Beispiele solcher Kondensationsmittel sind N,N-Dicylohexylcarbodümid, Polyphosphorsäure, Äthylpolyphosphat, Isopropylphosphat usw.
Die Reaktion wird gewöhnlich in einem inerten Lösungsmittel, wie Wasser, Aceton, Dioxan, Acetonitril, Pyridin, Chloroform, Äthylacetat, Tetrahydrofuran, Äthylendichlorid, Dimethylsulfoxyd, Dimethylformamid, Dimethylacetamid usw., ausgeführt. Erforderlichenfalls kann im Reaktionsgemisch eine basische Substanz, z. B. ein Alkalimetallcarbonat, Alkalimetallhydrogencarbonat, Trialkylamin, Pyridin usw., vorhanden sein. Die Reaktionstemperatur ist nicht speziell beschränkt, aber die Reaktion wird gewöhnlich unter Kühlen oder bei Raumtemperatur ausgeführt. Die acylierte Verbindung kann in üblicher Weise isoliert und gereinigt werden.
Bei der erfindungsgemässen Acylierungsreaktion können selbstverständlich Monoacylderivate, Diacylderivate oder Triacylderivate des Antibiotikums WS-4545 oder Gemische davon erzeugt werden, was von der verwendeten Menge an Acylierungsmittel abhängt.
Das WS-4545 wird also mit einer molaräquivalenten Menge eines Acylierungsmittels acyliert, um überwiegend das Monoacylderivat herzustellen. Wenn man mit annähernd zwei molaren Äquivalenten eines Acylierungsmittels acyliert, so erhält man hauptsächlich das Diacylderivat. Wird das WS4545 mit mehr als drei molaren Äquivalenten eines Acylierungsmittels behandelt, so wird überwiegend das Triacylderivat gebildet. Wenn bei der Reaktion ein Gemisch aus Monoacylderivat, Diacylderivat und Triacylderivat gebildet wird, kann jedes der Acylderivate in üblicher Weise aus dem Reaktionsgemisch isoliert werden, beispielsweise durch Chromatographie oder Kristallisation. Ferner können Monoacylderivate mit mindestens einem molaren Äquivalent eines Acylierungsmittels mit einem anderen Acylrest als das Monoacylderivat acyliert werden, um gemischte Diacylderivate bzw. Triacylderivate herzustellen.
Die Acylierungsreaktion gemäss der Erfindung kann durch das folgende Reaktionsschema wiedergegeben werden:

Im obigen Formelschema entspricht das Monoacylderivat des Antibiotikums WS-4545 der Formel III, worin RI eine Acylgruppe und RII sowie RIII Wasserstoff bedeuten, während das Diacylderivat der Formel III entspricht, worin RI und Rll Acylgruppen und R111 Wasserstoff darstellen, und das Triacylderivat entspricht der Formel III, worin R1, RII und RIII Acylgruppen bedeuten.

Die so hergestellten Acylderivate von WS-4545 werden bei oraler Verabreichung in viel höherer Konzentration vom Körper resorbiert und können im Körper in dieses Antibiotikum, das gegen pathogene Bakterien wirksam ist, umgewandelt werden. Daher können die Acylderivate als Antibiotika, die bei oraler Verabreichung zur Behandlung von Infektionskrankheiten bei Menschen und Tieren wirksam sind, verwendet werden. Dies zeigen die folgenden pharmakologischen Tests.

Verabreichung der Acylderivate von WS-4545 bei gesunden
Freiwilligen
Die Serum- und Urinspiegel wurden bei drei gesunden Freiwilligen bestimmt. Drei erwachsene männliche Freiwillige mit Gewichten im Bereich von 60,0 bis 75,0 kg wurde je eine Probe in einer einzigen Dosis von 1000 mg bzw. 500 mg oral verabreicht, 1, 2, 3, 5 und 8 Stunden nach der Verabreichung wurden Blutproben entnommen. Urinproben wurden in den Zeiträumen von 0-1, 1-3, 3-6, 6-9 und 9-24 Stunden nach der Verabreichung gesammelt. Die Konzentrationen des WS-4545 im Serum und im Urin wurden mittels des Schalenverfahrens bestimmt. Ein entsprechender Test auf die Ausscheidung im Urin wurde mit dem Antibiotikum WS-4545 ausgeführt. Die erfindungsgemäss erhältlichen Verbindungen können für die Verabreichung analog wie andere antibiotische Substanzen auf jede herkömmliche Weise formuliert werden.
So können die erfindungsgemäss erhältlichen Verbindungen in Form von pharmazeutischen Präparaten, beispielsweise in fester, halbfester oder flüssiger Form, verwendet werden, die ein Acylderivat im Gemisch mit einem pharmazeutischen organischen oder anorganischen Träger oder Exzipiens, das für die äusserliche oder parenterale Anwendung geeignet ist, enthalten. Die Wirkstoffe können beispielsweise mit den üblichen Trägern für Tabletten, Pillen, Kapseln, Suppositorien, Lösungen, Emulsionen, wässrigen Suspension und anderen, für die Anwendung geeigneten Formen gemischt werden.
Verwendbare Träger sind Glucose, Lactose, Akaziengummi, Gelatine, Mannit, Stärkepaste, Magnesiumtrisilikat, Talkum, Maisstärke, Keratin, kolloidales Siliciumdioxyd, Kartoffelstärke, Harnstoff und andere Träger, die für die Verwendung bei der Herstellung von Präparaten in fester, halbfester oder flüssiger Form geeignet sind, und ausserdem können Hilfsstoffe, Stabilisatoren, Verdickungsmittel und Färbemittel sowie Geruchsstoffe verwendet werden. Die Präparate können auch Konservierungsmittel oder bakteriostatische Mittel enthalten, um die Aktivität des Wirkstoffes in den gewünschten Präparaten stabil zu halten. Der Wirkstoff WS-4545 und/oder seine Acylderivate werden den Präparaten in einer solchen Menge einverleibt, dass die gewünschte therapeutische Wirkung gegen den bakteriellen Infektionsprozess oder den Infektionszustand erzielt wird.
Die Dosierung oder die therapeutisch wirksame Menge der Verbindungen hängen zwar von dem Alter und dem Zustand der einzelnen Patienten ab und müssen entsprechend variiert werden, aber eine tägliche Dosis von ca. 0,5 bis 5 g, vorzugsweise 1 bis 2 g, des Wirkstoffes pro Tag wird im allgemeinen zur Behandlung von Krankheiten verabreicht, gegen die das Antibiotikum oder seine Derivate verwendet werden können.
Die folgenden Beispiele erläutern die Erfindung.
Herstellung von Acylderivaten des Antibiotikums WS-4545
Das Antibiotikum WS-4545 wurde als Ausgangsmaterial für die Herstellung von Acylderivaten verwendet und wird im folgenden als WS-4545-Substanz bezeichnet.
Beispiel 1
Eine Lösung von 100 mg WS-4545-Substanz in 0,5 cm3 Pyridin wurde unter Kühlen auf - 100 C tropfenweise mit 0,5 cm3 Essigsäureanhydrid versetzt und das Gemisch 3 Stunden lang gerührt. Das Reaktionsgemisch wurde mit Eiswasser versetzt und die Lösung der Gefriertrocknung unterworfen. Die gefriergetrocknete Substanz wurde aus einem Gemisch aus Aceton und Äther kristallisiert, wobei man Kristalle erhielt, die aus Aceton umkristallisiert wurden und 50 mg des Essigsäureesters von WS-4545-Substanz in Form von Prismen vom Schmelzpunkt 213 bis 2150 C ergaben.
Analyse für Cl4H2008N2:
Berechnet: C 48,83; H 5,86; N 8,14;
Gefunden: C 48,91; H 5,81; N 8,08.
Beispiel 2
Eine Lösung von 12,08 g WS-4545-Substanz in 48 cm3 Pyridin wurde unter Kühlen auf C bis - 100 C unter Rühren im Verlauf von 1,5 Stunden tropfenweise mit 7,8 g Propionsäureanhydrid versetzt. Das Reaktionsgemisch liess man über Nacht stehen und versetzte es mit 100 cm3 Wasser, worauf die Lösung unter vermindertem Druck eingeengt wurde. Der Rückstand wurde in 100 cm3 Wasser gelöst und die Lösung mit Äthylacetat gewaschen und dann unter vermindertem Druck eingeengt. Beispiel 3
Eine Lösung von 6,04 g WS-4545-Substanz in 30 cm3 Pyridin wurde unter Kühlen in einem Eiswasserbad und unter Rühren im Verlauf einer Stunde tropfenweise mit 5,81 g Palmitoylchlorid versetzt. Das Gemisch wurde bei der gleichen Temperatur eine Stunde lang und bei Raumtemperatur ebenfalls eine Stunde lang weiter gerührt. Das Reaktionsgemisch wurde mit 60 cm3 kaltem Wasser versetzt und die Lösung unter vermindertem Druck eingeengt. Der Rückstand wurde in Äthylacetat gelöst und die Lösung nacheinander mit Wasser, 2,30/obiger Salzsäure, wässriger Natriumbicarbonatlösung und Wasser gewaschen und dann über Magnesiumsulfat getrocknet.
Das Lösungsmittel wurde abdestilliert und der Rückstand zweimal aus einem Gemisch von Acetonitril und Isopropyläther umkristallisiert, wobei man 5,0 g des Palmitinsäureesters von WS-4545-Substanz in Form farbloser Kristalle vom Schmelzpunkt 156 bis 1570 C erhielt.

Beispiel 6
Eine Lösung von 500 mg WS-4545-Substanz in 2 cm3 Pyridin wurde bei Raumtemperatur tropfenweise mit 2 cm9 Essigsäureanhydrid versetzt und das Gemisch 5 Stunden lang gerührt. Das Reaktionsgemisch wurde in Eiswasser gestellt, wobei sich Kristalle abschieden. Diese wurden abfiltriert und mit Wasser gewaschen und darauf aus Methanol umkristallisiert, wobei man 600 mg des Triessigsäureesters von WS4545-Substanz in Form farbloser Prismen vom Schmelzpunkt 247 bis 2500 C (Zersetzung) erhielt.

Beispiel 7 (A) Eine Lösung von 30,2 g WS-4545-Substanz in 90 cm3 Pyridin wurde bei Raumtemperatur unter Rühren im Verlauf einer Stunde tropfenweise mit 17,4 g n-Buttersäureanhydrid versetzt, worauf das Gemisch eine Stunde lang weiter gerührt wurde. Das Reaktionsgemisch wurde mit Wasser versetzt und die Lösung unter vermindertem Druck eingeengt. Der Rückstand wurde in Äthylacetat gelöst und die Lösung mit wässriger Natriumbicarbonatlösung und dann mit Wasser gewaschen und über Magnesiumsulfat getrocknet. Das Lösungsmittel wurde unter vermindertem Druck abdestilliert und der Rückstand mit Isopropyläther gewaschen, wobei rohe Kristalle erhalten wurden.
Diese wurden aus einem Gemisch aus Äthylacetat und Benzol umkristallisiert, wobei Kristalle gebildet wurden, die mit Isopropyläther gewaschen und getrocknet wurden; man erhielt 24,3 g n-Buttersäureester von WS4545-Substanz in Form farbloser Kristalle vom Schmelzpunkt 139 bis 1400 C.

(B) Die bei der obigen Umkristallisationsoperation erhaltene Mutterlauge wurde unter vermindertem Druck eingeengt. Der Rückstand wurde auf einer mit Silikagel gefüllten Säule chromatographiert, die mit einem Gemisch aus Chloroform und Methanol im Verhältnis 15:1 eluiert wurde. Das Eluat wurde unter vermindertem Druck eingeengt und der Rückstand aus einem Gemisch aus Äthylacetat und Benzol umkristallisiert, wobei man 2,0 g des Di-n-buttersäureesters von WS-4545-Substanz in Form farbloser Kristalle vom Schmelzpunkt 160,5 bis 162,50 C erhielt.

Beispiel 8
Eine Lösung von 12,08 g WS-4545-Substanz in 36 cm Pyridin wurde bei Raumtemperatur unter Rühren im Verlauf von einer Stunde tropfenweise mit 6,7 g Benzoylchlorid versetzt. Das Gemisch wurde eine weitere Stunde lang gerührt und dann mit Wasser versetzt. Die Lösung wurde unter vermindertem Druck eingeengt und der Rückstand in Sithyl- acetat gelöst. Die Lösung wurde nacheinander mit Wasser, wässriger Natriumbicarbonatlösung und Wasser gewaschen und über Magnesiumsulfat getrocknet. Das Lösungsmittel wurde unter vermindertem Druck abdestilliert und der Rückstand aus einem Gemisch aus Äthylacetat und Äther umkristallisiert, wobei man 12,0 g des Benzoesäureesters von WS-4545-Substanz in Form farbloser Kristalle vom Schmelzpunkt 1350 C erhielt.

Beispiel 9
Eine Lösung von 2,0 g WS-4545-Substanz in 6 cm Pyridin wurde unter Kühlen in Eiswasser im Verlauf von ca. 30 Minuten tropfenweise mit 1,4 g p-Chlorbenzoylchlorid versetzt.
Man liess das Gemisch bei der gleichen Temperatur eine Stunde lang stehen, worauf das Kühlbad weggenommen und die Reaktion 3 Stunden lang fortgesetzt wurde. Das Reaktionsgemisch wurde mit 50 cms Eiswasser versetzt und die Lösung mit Äthylacetat extrahiert. Die Äthylacetatschicht wurde nacheinander mit Wasser, 20/obiger wässriger Natrium bicarbonatlösung und Wasser gewaschen und über Magnesiumsulfat getrocknet. Das Äthylacetat wurde abdestilliert und der Rückstand aus Äthylacetat umkristallisiert; diese Umkristallisationsoperation wurde zweimal ausgeführt, wobei man 0,9 g des p-Chlorbenzoesäureesters von WS-4545-Substanz in Form farbloser Kristalle vom Schmelzpunkt 135 bis 1370 C erhielt.

Man rührte das Gemisch eine weitere Stunde lang und liess es über Nacht stehen. Das Reaktionsgemisch wurde mit Wasser versetzt und das Lösungsmittel unter vermin dertem Druck abdestilliert. Der Rückstand wurde in Äthylacetat gelöst. Die Lösung wurde nacheinander mit Wasser, wässriger Natriumbicarbonatlösung und Wasser gewaschen und über Magnesiumsulfat getrocknet. Das Lösungsmittel wurde unter vermindertem Druck abdestilliert und der Rückstand auf eine Säule mit Silikagel gegeben. Die gewünschte Verbindung wurde auf dem Silikagel in der Säule adsorbiert und mit einem Gemisch aus Chloroform und Äthylacetat im Verhältnis 9:1 eluiert.
Das Eluat wurde unter vermindertem Druck eingeengt und der Rückstand aus Benzol umkristallisiert, wobei man 1,02 g des Tribenzoesäureesters von WS-4545-Substanz in Form farbloser Kristalle vom Schmelzpunkt 1650 C erhielt.

Beispiel 18
Eine Lösung von 2,0 g WS-4545-Substanz in 12 cms Pyridin wurde unter Kühlen in einem Eiswasserbad mit 0,8 g Nicotinsäureanhydrid versetzt, worauf das Gemisch 1,5 Stunden lang gerührt wurde. Danach wurde das Kühlbad weggenommen und das Reaktionsgemisch weitere 5 Stunden lang gerührt. Das Reaktionsgemisch wurde mit Wasser versetzt und die Lösung unter vermindertem Druck eingeengt. Der Rückstand wurde mit Äthylacetat extrahiert und die Äthylacetatschicht mit wässriger Natriumbicarbonatlösung gewaschen und über Magnesiumsulfat getrocknet. Das Äthylacetat wurde abdestilliert und der Rückstand aus Äthylacetat umkristallisiert, wobei man 0,8 g des Nicotinsäureesters von WS4545-Substanz in Form blassgelber Kristalle vom Schmelz punkt 1430 C erhielt.

Das Gemisch wurde bei der gleichen Temperatur 2 Stunden lang weiter gerührt und mit 40 cm3 Wasser versetzt. Das Lösungsmittel wurde unter vermindertem Druck abdestilliert und der Rückstand in Wasser gelöst. Die wässrige Lösung wurde mit Äthylacetat und dann mit gesättigter Natriumchloridlösung gewaschen und mit Äthylacetat extrahiert. Die Äthylacetatschicht wurde mit wässriger Natriumbicarbonatlösung gewaschen und über Magnesiumsulfat getrocknet.
Das Lösungsmittel wurde unter vermindertem Druck abdestilliert und der Rückstand aus einem Gemisch aus Äthylacetat und Benzol umkristallisiert, wobei man 0,93 g des 2-Furancarbonsäureesters von WS-4545-Substanz in Form farbloser Kristalle vom Schmelzpunkt 1250 C erhielt.

Beispiel 22
Eine Lösung von 1,0 g des in Beispiel 11 erhaltenen p Brombenzoesäureesters von WS-4545-Substanz in 3 cm3 Pyridin wurde bei Raumtemperatur unter Rühren im Verlauf von 20 Minuten tropfenweise mit 3,0 cm2 Essigsäureanhydrid versetzt. Das Gemisch wurde bei Raumtemperatur weitere 4 Stunden lang gerührt und mit Wasser versetzt. Die Lösung wurde unter vermindertem Druck eingeengt und der Rückstand in Wasser gelöst. Die Lösung wurde nacheinander mit Wasser, wässriger Natriumbicarbonatlösung und Wasser gewaschen und über Magnesiumsulfat getrocknet.
Das Lö sungsmittel wurde unter vermindertem Druck abdestilliert und der Rückstand auf eine Säule mit Silikagel gegeben, worauf der adsorbierte Rückstand mit einem Gemisch aus Chloroform und Athylacetat im Verhältnis 8:2 eluiert wurde.
Das Eluat wurde unter vermindertem Druck eingeengt, wobei rohe Kristalle erhalten wurden. Diese wurden aus einem Gemisch aus Äthylacetat und Benzol umkristallisiert und ergaben 0,5 g des Diacetyl-p-brombenzoylderivates von WS-4545 Substanz in Form farbloser Kristalle vom Schmelzpunkt 2100 C.

Beispiel 23
Eine Lösung von 2,0 g des p-Brombenzoesäureesters von WS-4545-Substanz in 6 cm3 Pyridin wurde bei Raumtemperatur unter Rühren im Verlauf von 30 Minuten tropfenweise mit 1,45 g Benzoylchlorid versetzt. Das Gemisch wurde weitere 5 Stunden lang bei Raumtemperatur gerührt und über Nacht stehen gelassen. Das Reaktionsgemisch wurde mit Wasser versetzt und unter vermindertem Druck eingeengt.
Der Rückstand wurde in Äthylacetat gelöst und nacheinander mit Wasser, wässriger Natriumbicarbonatlösung und Wasser gewaschen. Nach Trocknen der Lösung wurde das Lösungsmittel unter vermindertem Druck abdestilliert. Der Rückstand wurde auf eine Säule mit Silikagel gegeben und der adsorbierte Rückstand mit Chloroform eluiert. Das Eluat wurde unter vermindertem Druck eingeengt, wobei rohe Kristalle gebildet wurden. Diese wurden aus Äthylacetat umkristallisiert und ergaben 1,51 g des Dibenzoyl-p-brombenzoylderivates von WS-4545-Substanz in Form von Nadeln vom Schmelzpunkt 177 bis 1780 C.
Beispiel 24
Eine Lösung von 2,0 g des in Beispiel 10 erhaltenen m Brombenzoesäureesters von WS-4545-Substanz in 6 cm3 Pyridin wurde unter Kühlen und Rühren im Verlauf von 30 Minuten tropfenweise mit 40 cm3 Essigsäureanhydrid versetzt. Das Gemisch wurde weitere 3 Stunden lang gerührt und über Nacht in einem Kühlschrank stehen gelassen. Das Reaktionsgemisch wurde mit Wasser versetzt und die Lösung unter vermindertem Druck eingeengt. Der Rückstand wurde mit Wasser gewaschen und getrocknet und ergab 2,2 g rohe Kristalle. Diese wurden aus Aceton umkristallisiert, wobei man das Diacetyl-m-brombenzoylderivat von WS-4545-Substanz in Form von Kristallen vom Schmelzpunkt 217 bis 2180 C erhielt.
Beispiel 25
Zu einer Lösung von 3,02 g der WS-4545-Substanz in 12 ml einer Mischung von 9 ml Pyridin und 3 ml Aceton gibt man tropfenweise unter Rühren und unter Kühlen auf 0 bis 50 C mit Hilfe eines Eiswasserbades innerhalb von 1 Stunde eine Lösung von 3,02 g Hexanoylchlorid in 3 ml Aceton hinzu. Dann wird das Reaktionsgemisch während 1 Stunde bei der gleichen Temperatur gerührt. Hierauf versetzt man das Reaktionsgemisch mit dem 3fachen Volumen an kaltem Wasser und konzentriert dann diese Lösung unter vermindertem Druck. Der Rückstand wird in Äthylacetat gelöst und die Lösung nacheinander mit Wasser, wässriger Natriumbicarbonatlösung und Wasser gewaschen und hierauf über Magnesiumsulfat getrocknet. Dann wird die Lösung unter vermindertem Druck eingeengt und der Rückstand aus einer Mischung von Benzol und n-Hexan zum Auskristallisieren gebracht, wobei man 2,3 g rohe Kristalle erhält.
Die rohen Kristalle werden aus Acetonitril umkristallisiert, wobei man 1,8 g des Hexansäureesters der WS-4545-Substanz in Form von farblosen Kristallen vom Smp. 144 bis 1450 C erhält.

Zu einer Lösung von 10,9 g der WS-4545-Substanz in 60 ml Pyridin gibt man tropfenweise unter Rühren bei 30 C innerhalb von 25 Minuten 6,6 g 3-Vinylpropionsäureanhydrid hinzu. Das Reaktionsgemisch wird hierauf während 30 Minuten bei 30 C und hierauf während 14 Stunden bei 3 bis 170 C gerührt. Nach beendeter Umsetzung wird das Pyridin aus dem Reaktionsgemisch unter vermindertem Druck entfernt. Hierauf wird der Rückstand mit Eiswasser versetzt und die Lösung hierauf mit Äther gewaschen. Die Lösung wird mit Natriumchlorid gesättigt und hierauf mit Äthylacetat extrahiert. Die Äthylacetatschicht wird mit einer mit Natriumchlorid gesättigten, wässrigen Lösung gewaschen und hierauf über Magnesiumsulfat getrocknet, wobei man 9,0 g eines amorphen Pulvers erhält. Dieses amorphe Pulver wird in Benzol gelöst.
Die Benzollösung wird über Nacht stehen gelassen, wobei sich 7,1 g rohe Kristalle ausscheiden. Diese rohen Kristalle werden aus einer Mischung von Äthylacetat und Diisopropyläther umkristallisiert, wobei man 6,1 g 3-Vinylpropionsäureester der WS-4545-Substanz in Form von farblosen Kristallen vom Smp. 170 bis 1720 C erhält.

In eine Lösung von 9,7 g WS-4545-Substanz in 30 ml Pyridin werden tropfenweise innerhalb von 30 Minuten bei 50 C 16,76 g 2-(3,4,5-Trimethoxy)-phenylessigsäureanhydrid eingerührt. Dann wird das Reaktionsgemisch über Nacht weitergerührt. Hierauf wird das Reaktionsgemisch mit der dreifachen Volumenmenge Wasser versetzt und anschliessend eingeengt. Der Rückstand wird in Äthylacetat gelöst. Diese Äthylacetatlösung wird mit einer mit Natriumbicarbonat gesättigten wässrigen Lösung bei einer unter 50 C liegenden Temperatur versetzt und hierauf kräftig gerührt. Diese mit Natriumbicarbonat gesättigte wässrige Lösung wird mit einer kleinen Menge Kochsalz versetzt und hierauf gerührt. Die Äthylacetatschicht wird mit Wasser gewaschen und hierauf über Magnesiumsulfat getrocknet, wobei man ein gelbes, rohes Pulver erhält.
Dieses Pulver wird mit Äthylacetat trituriert, wobei man 4,83 g 2-(3,4,5-Trimethoxy)-phenylessigsäureester der WS4545-Substanz in Form von farblosen Kristallen vom Smp.
147 bis 1500 C erhält.

Zu einer Lösung von 20 g der WS-4545-Substanz in 150 ml Pyridin gibt man tropfenweise unter Kühlen 19,43 g Thienylessigsäureanhydrid hinzu, worauf man das Gemisch während 5 Stunden rührt. Dann wird das Reaktionsgemisch mit Wasser versetzt und die Lösung unter vermindertem Druck eingeengt. Der Rückstand wird in Äthylacetat gelöst und die Lösung nacheinander mit Wasser, mit einer wässrigen Natriumbicarbonatlösung und mit Wasser gewaschen und hierauf über Magnesiumsulfat getrocknet. Dann wird das Lösungsmittel abdestilliert und der Rückstand mit Aktivkohle behandelt, worauf man den Rückstand aus einer Mischung von Äthylacetat und Isopropyläther umkristallisiert.
Auf diese Weise erhält man Kristalle, welche nach dem Umkristallisieren aus einer Mischung von Äthylacetat und Isopropyläther 13,5 g des Thienylessigsäureesters der WS-4545-Substanz in Form von farblosen Prismen ergeben. Smp.: 171-1720 C.

Zu einer Lösung von 24,2 g WS-4545-Substanz in 70 ml Pyridin gibt man tropfenweise 19,7 g Äthylthioessigsäureanhydrid unter Rühren und Kühlen auf 5 C bis 0 C innerhalb von I Stunde hinzu.
Dann wird das Reaktionsgemisch während einer weiteren Stunde bei dieser gleichen Temperatur gerührt und hierauf über Nacht bei Zimmertemperatur stehen gelassen. Dann wird das Reaktionsgemisch mit 3 Volumen Wasser versetzt und die Lösung hierauf unter vermindertem Druck bei 500 C eingeengt, wobei man einen gelben, öligen Rückstand erhält. Dieser Rückstand wird in 200 ml Äthylacetat gelöst und die Lösung mehrere Male mit kalter, 10%iger wässriger Salzsäurelösung so lange gewaschen, bis die wässrige Schicht sauer ist, wodurch das Pyridin entfernt wird. Die gewünschte Verbindung wird zweimal mit jeweils 50 ml Äthylacetat aus der ausgesalzenen wässrigen Schicht extrahiert. Die vereinigten Extrakte werden zweimal mit einer gesättigten wässrigen Natriumbicarbonatlösung gewaschen und die wässrige Schicht wird durch Aussalzen und durch Extraktion mit Äthylacetat behandelt.
Die Äthylacetatschicht wird dreimal mit gesättigter Natriumchloridlösung gewaschen, über Magnesiumsulfat getrocknet und unter vermindertem Druck eingeengt, wobei man 26 g eines rohen Pulvers erhält. Das Pulver aus 200 ml einer Mischung von Benzol und Äthylacetat (Mischungsverhältnis 1:1) wird zum Auskristallisieren gebracht, wobei man 24,76 g farblose Nadeln erhält. Diese Nadeln werden aus 200 ml Äthylacetat umkristallisiert, wobei man 9,95 g des Äthylthioessigsäureesters der WS-4545-Substanz in Form von farblosen Nadeln, die sich bei 142 bis 1450 C zersetzen, erhält.

The present invention relates to a process for the preparation of new, effective acyl derivatives of the antibiotic WS-4545 which are effective against a number of microorganisms and which can be made into pharmaceutical preparations and used therapeutically in the treatment of infections.
The new compounds can be used to produce pharmaceutical preparations which contain acyl derivatives of the antibiotic WS-4545 mentioned as effective antibacterial agents, these preparations being able to serve for the treatment of infectious diseases caused by bacteria in Mellschen and animals.
The WS-4545 antibiotic corresponds to the following formula

and is obtained by fermenting Streptomyces sapporonensis ATCC-21532 or mutants thereof under aerobic conditions in a nutrient medium.
The microorganism that can be used to produce the new antibiotic WS4545 is a newly discovered species of Streptomyces that was isolated from a soil sample found in the city of Sapporo, Japan. A living organism culture has been deposited with the American Type Culture Collection under number ATCC 21532 and has been incorporated into the permanent culture inventory collection; the microorganism is referred to below as Streptomyces sapporonensis.
Of course, the invention is not restricted to the use of the specific organism described here or of organisms which completely correspond to the growth and microscopic properties described here for the purpose of illustration for the production of the new antibiotic. It has been found that the antibiotic WS-4545, which has hydroxyl groups, can easily be acylated to the corresponding acyl derivatives with an acylating agent. The compounds obtainable according to the invention can be formulated for administration in any conventional manner analogous to other antibiotic substances.

Example 21 (A) A solution of 3.02 g of WS-4545 substance in 18 cms of pyridine was added dropwise with 1.43 g of 2 furoyl chloride over 30 minutes while cooling to -5 to -100 ° C. while stirring. The mixture was further stirred at the same temperature for 2 hours, and 40 cm 3 of water was added. The solvent was distilled off under reduced pressure and the residue was dissolved in water. The aqueous solution was washed with ethyl acetate and then with saturated sodium chloride solution and extracted with ethyl acetate. The ethyl acetate layer was washed with an aqueous sodium bicarbonate solution and dried over magnesium sulfate.
The solvent was distilled off under reduced pressure and the residue was recrystallized from a mixture of ethyl acetate and benzene, 0.93 g of the 2-furancarboxylic acid ester of WS-4545 substance being obtained in the form of colorless crystals with a melting point of 1250.degree.

(B) The mother liquor obtained in the above recrystallization operation was concentrated under reduced pressure and the residue was applied to a column of silica gel; the adsorbed residue was eluted with a mixture of chloroform and methanol in a ratio of 15: 1. The eluate was concentrated under reduced pressure and the residue was recrystallized from a mixture of chloroform and methanol, whereby 0.6 g of the di-2-furancarboxylic acid ester of WS-4545 substance was obtained in the form of colorless crystals with a melting point of 183 to 1840.degree.

Example 22
A solution of 1.0 g of the bromobenzoic acid ester of WS-4545 substance obtained in Example 11 in 3 cm 3 of pyridine was admixed dropwise with 3.0 cm 2 of acetic anhydride at room temperature with stirring over the course of 20 minutes. The mixture was stirred at room temperature for an additional 4 hours and water was added. The solution was concentrated under reduced pressure and the residue was dissolved in water. The solution was washed successively with water, aqueous sodium bicarbonate solution and water and dried over magnesium sulfate.
The solvent was distilled off under reduced pressure and the residue was applied to a column of silica gel, whereupon the adsorbed residue was eluted with a mixture of chloroform and ethyl acetate in the ratio 8: 2.
The eluate was concentrated under reduced pressure to obtain crude crystals. These were recrystallized from a mixture of ethyl acetate and benzene and gave 0.5 g of the diacetyl-p-bromobenzoyl derivative of WS-4545 substance in the form of colorless crystals with a melting point of 2100 C.

Example 23
A solution of 2.0 g of the p-bromobenzoic acid ester of WS-4545 substance in 6 cm 3 of pyridine was added dropwise with 1.45 g of benzoyl chloride over 30 minutes at room temperature while stirring. The mixture was stirred for an additional 5 hours at room temperature and left to stand overnight. The reaction mixture was added with water and concentrated under reduced pressure.
The residue was dissolved in ethyl acetate and washed successively with water, aqueous sodium bicarbonate solution and water. After the solution was dried, the solvent was distilled off under reduced pressure. The residue was applied to a column of silica gel, and the adsorbed residue was eluted with chloroform. The eluate was concentrated under reduced pressure, whereby crude crystals were formed. These were recrystallized from ethyl acetate and gave 1.51 g of the dibenzoyl-p-bromobenzoyl derivative of WS-4545 substance in the form of needles with a melting point of 177 to 1780 C.
Example 24
A solution of 2.0 g of the m bromobenzoic acid ester of WS-4545 substance obtained in Example 10 in 6 cm 3 of pyridine was added dropwise with 40 cm 3 of acetic anhydride over a period of 30 minutes while cooling and stirring. The mixture was stirred for an additional 3 hours and left in a refrigerator overnight. Water was added to the reaction mixture and the solution was concentrated under reduced pressure. The residue was washed with water and dried to give 2.2 g of crude crystals. These were recrystallized from acetone, whereby the diacetyl-m-bromobenzoyl derivative of WS-4545 substance was obtained in the form of crystals with a melting point of 217 to 2180.degree.
Example 25
To a solution of 3.02 g of the WS-4545 substance in 12 ml of a mixture of 9 ml of pyridine and 3 ml of acetone, a solution is added dropwise over the course of 1 hour while stirring and cooling to 0 to 50 ° C. with the aid of an ice water bath of 3.02 g of hexanoyl chloride in 3 ml of acetone are added. Then the reaction mixture is stirred for 1 hour at the same temperature. Three times the volume of cold water is then added to the reaction mixture, and this solution is then concentrated under reduced pressure. The residue is dissolved in ethyl acetate and the solution washed successively with water, aqueous sodium bicarbonate solution and water and then dried over magnesium sulfate. The solution is then concentrated under reduced pressure and the residue is crystallized from a mixture of benzene and n-hexane, giving 2.3 g of crude crystals.
The crude crystals are recrystallized from acetonitrile, 1.8 g of the hexanoic acid ester of the WS-4545 substance being obtained in the form of colorless crystals with a melting point of 144 to 1450.degree.

To a solution of 10.9 g of the WS-4545 substance in 60 ml of pyridine, 6.6 g of 3-vinylpropionic anhydride are added dropwise with stirring at 30 ° C. over the course of 25 minutes. The reaction mixture is then stirred at 30 ° C. for 30 minutes and then at 3 to 170 ° C. for 14 hours. When the reaction has ended, the pyridine is removed from the reaction mixture under reduced pressure. Ice water is then added to the residue and the solution is then washed with ether. The solution is saturated with sodium chloride and then extracted with ethyl acetate. The ethyl acetate layer is washed with an aqueous solution saturated with sodium chloride, and then dried over magnesium sulfate to obtain 9.0 g of an amorphous powder. This amorphous powder is dissolved in benzene.
The benzene solution is left to stand overnight, 7.1 g of crude crystals separating out. These crude crystals are recrystallized from a mixture of ethyl acetate and diisopropyl ether, 6.1 g of 3-vinylpropionic acid ester of the WS-4545 substance being obtained in the form of colorless crystals with a melting point of 170 to 1720.degree.

16.76 g of 2- (3,4,5-trimethoxy) phenylacetic anhydride are stirred into a solution of 9.7 g of WS-4545 substance in 30 ml of pyridine over a period of 30 minutes at 50.degree. Then the reaction mixture is stirred further overnight. Three times the volume of water is then added to the reaction mixture and the mixture is then concentrated. The residue is dissolved in ethyl acetate. This ethyl acetate solution is mixed with an aqueous solution saturated with sodium bicarbonate at a temperature below 50 C and then stirred vigorously. This aqueous solution saturated with sodium bicarbonate is mixed with a small amount of common salt and then stirred. The ethyl acetate layer is washed with water and then dried over magnesium sulfate to give a crude yellow powder.
This powder is triturated with ethyl acetate, whereby 4.83 g of 2- (3,4,5-trimethoxy) phenylacetic acid ester of the WS4545 substance are obtained in the form of colorless crystals of mp.

The residue is dissolved in ethyl acetate and the solution washed successively with water, with an aqueous sodium bicarbonate solution and with water and then dried over magnesium sulfate. The solvent is then distilled off and the residue is treated with activated charcoal, whereupon the residue is recrystallized from a mixture of ethyl acetate and isopropyl ether.
In this way, crystals are obtained which, after recrystallization from a mixture of ethyl acetate and isopropyl ether, give 13.5 g of the thienylacetic acid ester of the WS-4545 substance in the form of colorless prisms. M.p .: 171-1720 C.

Example 43
19.7 g of ethylthioacetic anhydride are added dropwise to a solution of 24.2 g of WS-4545 substance in 70 ml of pyridine over the course of 1 hour, while stirring and cooling to 5 ° C. to 0 ° C.
The reaction mixture is then stirred for a further hour at this same temperature and then left to stand overnight at room temperature. 3 volumes of water are then added to the reaction mixture, and the solution is then concentrated under reduced pressure at 500 ° C., a yellow, oily residue being obtained. This residue is dissolved in 200 ml of ethyl acetate and the solution is washed several times with cold, 10% strength aqueous hydrochloric acid solution until the aqueous layer is acidic, whereby the pyridine is removed. The desired compound is extracted twice from the salted-out aqueous layer, each time with 50 ml of ethyl acetate. The combined extracts are washed twice with a saturated aqueous sodium bicarbonate solution and the aqueous layer is treated by salting out and by extraction with ethyl acetate.
The ethyl acetate layer is washed three times with a saturated sodium chloride solution, dried over magnesium sulfate and concentrated under reduced pressure to obtain 26 g of a crude powder. The powder of 200 ml of a mixture of benzene and ethyl acetate (mixing ratio 1: 1) is caused to crystallize out, giving 24.76 g of colorless needles. These needles are recrystallized from 200 ml of ethyl acetate, 9.95 g of the ethylthioacetic acid ester of the WS-4545 substance being obtained in the form of colorless needles which decompose at 142 to 1450.degree.
