Die Erfindung betrifft ein Verfahren zur Herstellung neuer Riechstoffe. Bei diesen Riechstoffen handelt es sich um die Verbindungen der Formel

Die Oxydation des Alkohols II kann nach an sich bekannten Methoden durchgeführt werden. Geeignete Oxydationsmittel sind beispielsweise Silbercarbonat in Gegenwart von Diatomeenerde, sauerstoffhaltige Derivate, wie Oxyde von Übergangsmetallen, z. B. von Chrom, Mangan oder Nickel. Ein bevorzugtes Oxydationsmittel ist insbesondere Mangandioxyd Pyridin (J. R. Holum, J. Org. Chem. 26, 4814 [1961]). Bei dieser Methode wird vorzugsweise bei Raumtemperatur in einem inerten Lösungsmittel, z. B. Pentan oder Hexan, gearbeitet. Aber auch Chromtrioxyd ist bevorzugt.
Wenn Mangandioxyd als Oxydationsmittel verwendet wird, bleibt die geometrische Konfiguration des Ausgangsmaterials (cis- oder trans-Alkohol II oder Gemische derselben) praktisch vollständig erhalten. Wenn Chromtrioxyd verwendet wird, vorzugsweise in Gegenwart einer organischen Base wie Pyridin, weist das erhaltene Keton trans-Konfiguration auf, und zwar unabhängig davon, ob der Ausgangsalkohol II cisoder trans-Konfiguration aufweist.
Die trans-Isomeren können gewünschtenfalls durch Isomerisierung der cis-Isomeren mittels einer Säure in einem inerten Lösungsmittel erhalten werden. Die Isomerisierung kann mittels Protonensäuren, insbesondere jenen Säuren, die in bekannter Weise Ketone enolisieren, z. B. p-Toluolsulfonsäure, Trifluoressigsäure, und Salzsäure, durchgeführt werden.
Man kann aber auch Lewis-Säuren, z. B. Bortrifluorid oder Jod, verwenden. Die Isomerisierung wird zweckmässigerweise in einem inerten Lösungsmittel, z. B. einem aromatischen Kohlenwasserstoff wie Benzol oder Toluol, einem aliphatischen oder cycloaliphatischen Kohlenwasserstoff wie Heptan oder Cyclohexan oder einem Äther wie Monoglym, Diglym oder Dioxan, ausgeführt. Die Temperatur, bei welcher die Isomerisierung durchgeführt wird, ist nicht kritisch. Man kann beispielsweise die Isomerisierung so durchführen, dass man die zu isomerisierende Substanz mit dem Lösungsmittel und einer katalytischen Menge des sauren Isomerisierungsmittels mischt und das Gemisch bei Raumtemperatur während mehrerer Stunden, z. B. 12 Stunden, stehenlässt.
Die erfindungsgemäss erhältlichen Verbindungen I bzw.
deren Isomerengemische besitzen besondere Riechstoffeigenschaften. So ist beispielsweise der Geruchstyp eines Gemisches der Isomeren Ia, Ib und Ic typisch agreste (ländlich), die Geruchsnoten können wie folgt charakterisiert werden: nach Tabak und Eucalyptussamen, rauchig (Isoeugenol), fruchtig.
Die Geruchsnoten sind überraschenderweise völlig verschieden von denjenigen des strukturell nahe verwandten 2,3,6,6-Tetramethyl-1-crotonyl-cyclohexens - (1 und 2), welches aus der DOS 2 022 216 bekanntgeworden ist. Jenes Isomerengemisch verfügt insbesondere über eine holzige Note, etwas an Chinolin und Nüsse erinnernd, schwach gewürzartig.
Die Verbindungen I bzw. deren Isomerengemisch können demgemäss auf Grund ihrer interessanten olfaktorischen Eigenschaften als Riechstoffe Verwendung finden, z. B. in der Parfümerie zur Herstellung von Riechstoffkompositionen wie Parfums bzw. zur Parfümierung von Produkten aller Art, wie z. B. Seifen, Waschmitteln, festen und flüssigen Detergentien Aerosolen oder anderen kosmetischen Produkten, wie Salben.
Gesichtsmilch, Schminke, Lippenstiften, Badesalzen, Badeölen, usw., dienen.
Das Isomerengemisch I eignet sich aufgrund seiner sehr natürlichen Noten insbesondere zur Modifizierung bekannter Kompositionen, z. B. solchen vom Typ Chypre. Es eignet sich sehr gut in Kombination mit Holznoten, wie z. B. von p-tert.
Butylcyclohexylacetat, Sandelholzöl, Patchouliöl, Cedrylacetat, Methyljonon.
Die Konzentration des erfindungsgemässen Gemisches I kann je nach dem Verwendungszweck innerhalb weiter Grenzen variieren, beispielsweise zwischen etwa 1 (Detergentien) und etwa 15 Gew. % (alkoholische Lösungen). In Parfumbasen bzw. Konzentraten können die Konzentrationen selbstverständlich auch höher liegen.
In den folgenden Beispielen sind die Temperaturen in Celsiusgraden angegeben.
Beispiel 1
In einen Rundkolben, der mit Rührer und Thermometer versehen ist, gibt man 2,7 kg trockenes Hexan, 1 kg Mangandioxyd (J. Chem. Soc. [1952], 1104) und 100 g 2-Äthyl3,6,6-trimethyl- 1 -(1 -hydroxy-2-butenyl)-2-cyclohexen (enthaltend ca. 85 % a-Isomeres). Man rührt das Gemisch 48 Stunden bei 20 . Man filtriert anschliessend über Cellit und wäscht mit Äther nach. Man entfernt die Lösungsmittel des Filtrats durch Destillation und gewinnt so 95 g rohes 2-Äthyl-3,6,6 trimethyl- 1-crotonoyl-2-cyclohexen. Der Anteil der Isomeren entspricht dem oben für das Ausgangsmaterial angegebenen.
Durch Destillation werden 58,8 g (58,9% Ausbeute) des reinen Materials erhalten: Siedepunkt 63-66 /0,07-1 mm Hg, nu20= 1,4945-1,4959.
Das als Ausgangsmaterial verwendete 2-Äthyl-3,6,6-tri methyl- 1 - (1 -hydroxy-2-butenyl)-2-cyclohexen kann wie folgt erhalten werden:
In einen Rundkolben, der mit Thermometer und Rührer ausgestattet ist, gibt man 162 g (0,9 Mol) 3-Äthyl-4,7-dimethyl-octa-2,6-dienal und 220 ml Diäthyläther. Unter Rühren werden tropfenweise 102 g (1,1 Mol) frisch destilliertes Anilin zugegeben, und alsdann wird noch 11/2 Stunden weitergerührt. Das gebildete Wasser wird abdekantiert und die ätherische Lösung der gebildeten Schiffschen Base über Natriumsulfat getrocknet.
Unter Stickstoffatmosphäre werden in einem Rundkolben 880 g konzentrierte Schwefelsäure auf 200 abgekühlt. Innerhalb einer Stunde gibt man nun bei dieser Temperatur die ätherische Lösung der Schiffschen Base zu. Man rührt das Reaktionsgemisch 30 Minuten bei 200 weiter und giesst auf 1,5 kg zerstossenes Eis. Man extrahiert mit Äther, wäscht dreimal mit Wasser, hierauf mit einer 10%gen Natriumcarbonatlösung und schliesslich noch einmal mit Wasser. Nach Abdampfen des Äthers bleiben 128 g rohes 2-Äthyl-3,6,6-trimethyl-1-formyl-2-cyclohexen zurück, die nach fraktionierter Destillation 62,2 g (38,4%) des reinen Materials (enthaltend ca. 85 % a-Isomeres) ergeben; Siedepunkt 83-88 , nD20 = 1,4773.
In einen Rundkolben, der mit Rührer, Kühler, Thermometer und Tropftrichter versehen ist, gibt man 25,2 g (1,05 g-Atome) Magnesiumspäne in 80 ml getrocknetem Tetrahydrofuran. Unter Rühren gibt man nun langsam 133,1 g (1,1 g-Atome) 1-Propenbromid in 150 ml getrocknetem Tetrahydrofuran zu. Man bringt das Gemisch 30 Minuten auf Rückflusstemperatur und lässt nachher abkühlen (12 Stunden).
Nun gibt man bei 10 eine Lösung von 180,3 g (1 Mol) des oben erhaltenen Aldehyds in 250 ml getrocknetem Tetrahydrofuran zu. Man hält das Reaktionsgemisch 6 Stunden bei 40 , lässt wieder abkühlen, gibt feuchten Äther zu und zersetzt mittels einer gesättigten Ammonchloridlösung. Man wäscht die organische Schicht mit Wasser, mit Weinsäure und hierauf nochmals mit Wasser. Nach dem Abdampfen des Lösungsmittels gewinnt man 239 g rohes 2-Äthyl-3,6,6-trimethyl- 1 -(1 -hydroxy-2-butenyl)-2-cyclohexen (enthaltend ca. 85 % a-Isomeres), deren Destillation 117 g (53 %) Reinprodukt vom Siedepunkt 88-95 /0,2 mm Hg ergeben; nun20 = 1,4923-1,4942.
Beispiel 2
Zu einer auf ca. 5-10 gekühlten und unter Stickstoff gehaltenen Suspension von 2,4 g (0,35 g-Atom) feingeschnittenem Lithiumband in 40 ml abs. Tetrahydrofuran werden einige Kristalle Diphenyl zugefügt. Dann werden 2 ml einer Lösung von 9,4 g (70 mMol) Allylphenyläther in 10 ml abs.
Äther zugegeben. Nach Auftreten eines grünlichen Farbtons wird der Rest der Lösung tropfenweise bei -15" zugefügt.
Nach der Zugabe wird 30 Minuten bei 0 weitergerührt.
Die resultierende Allyllithiumlösung wird vom überschüssigen Lithium abdekantiert und in einen Tropftrichter transferiert.
Man gibt nun diese Lösung tropfenweise zu einer auf 600 gekühlten und unter Stickstoff gehaltenen Lösung von 4,48 g (20 mMol) 2-Äthyl-3 ,6,6-trimethyl-2-cyclohexen- 1-carbon- säureäthylester in 40 ml abs. Äther. Nach der Zugabe lässt man das Gemisch auf Zimmertemperatur erwärmen, giesst die Reaktionsmischung auf Eiswasser und extrahiert mit 3 Portionen Hexan. Die vereinigten Hexanlösungen werden mit Wasser (3 x ) neutralgewaschen, über Natriumsulfat getrocknet und eingedampft. Durch fraktionierte Destillation des Rückstandes erhält man 3,75 g (85 %) 2-Äthyl-3,6,6-trimethyl-1-[but-3-enoyl]-2-cyclohexen. Sdp. 57-58 /0,04 mm Hg, nD20 = 1,4865.
Eine Suspension von 3,5 g (15,9 mMol) 2-Äthyl-3,6,6 trimethyl- 1 -[but-3 -enoyl]-2-cyclohexen und 2,0 g Pottasche in 40 ml Aceton werden 24 Stunden bei Raumtemperatur unter Stickstoff gerührt. Vom Salz wird abfiltriert und das Filtrat eingedampft. Der Rückstand ergibt nach der fraktionierten Destillation 3,2 g (91,5%) 2-Äthyl-3,6,6-trimethyl-1crotonoyl-2-cyclohexen. Sdp. 59-60 /0,04 mm Hg, n,ZO = 1,4952.
= 1,4952.

The oxidation of the alcohol II can be carried out by methods known per se. Suitable oxidizing agents are, for example, silver carbonate in the presence of diatomaceous earth, oxygen-containing derivatives such as oxides of transition metals, e.g. B. of chromium, manganese or nickel. A preferred oxidizing agent is especially manganese dioxide pyridine (J. R. Holum, J. Org. Chem. 26, 4814 [1961]). This method is preferably carried out at room temperature in an inert solvent, e.g. B. pentane or hexane worked. Chromium trioxide is also preferred.
If manganese dioxide is used as the oxidizing agent, the geometric configuration of the starting material (cis- or trans-alcohol II or mixtures thereof) is practically completely retained. If chromium trioxide is used, preferably in the presence of an organic base such as pyridine, the ketone obtained has the trans configuration, regardless of whether the starting alcohol II has the cis or trans configuration.
The trans isomers can, if desired, be obtained by isomerizing the cis isomers using an acid in an inert solvent. The isomerization can be carried out by means of protic acids, especially those acids which enolize ketones in a known manner, e.g. B. p-toluenesulfonic acid, trifluoroacetic acid, and hydrochloric acid.
But you can also Lewis acids such. B. boron trifluoride or iodine, use. The isomerization is conveniently carried out in an inert solvent, e.g. B. an aromatic hydrocarbon such as benzene or toluene, an aliphatic or cycloaliphatic hydrocarbon such as heptane or cyclohexane or an ether such as monoglyme, diglyme or dioxane. The temperature at which the isomerization is carried out is not critical. For example, the isomerization can be carried out by mixing the substance to be isomerized with the solvent and a catalytic amount of the acidic isomerizing agent and the mixture at room temperature for several hours, e.g. B. 12 hours.
The compounds I or
their isomer mixtures have special fragrance properties. For example, the odor type of a mixture of isomers Ia, Ib and Ic is typically agreste (rural), the odor notes can be characterized as follows: after tobacco and eucalyptus seeds, smoky (isoeugenol), fruity.
The odor notes are surprisingly completely different from those of the structurally closely related 2,3,6,6-tetramethyl-1-crotonyl-cyclohexene - (1 and 2), which has become known from DOS 2,022,216. This mixture of isomers has a woody note, somewhat reminiscent of quinoline and nuts, slightly spicy.
The compounds I or their mixture of isomers can accordingly be used as odorants because of their interesting olfactory properties, eg. B. in perfumery for the production of fragrance compositions such as perfumes or for perfuming products of all kinds, such. B. soaps, detergents, solid and liquid detergents, aerosols or other cosmetic products such as ointments.
Facial milk, make-up, lipsticks, bath salts, bath oils, etc., are used.
Because of its very natural notes, the isomer mixture I is particularly suitable for modifying known compositions, e.g. B. those of the chypre type. It works very well in combination with wood notes, such as B. from p-tert.
Butylcyclohexyl acetate, sandalwood oil, patchouli oil, cedrylacetate, methylionon.
The concentration of the mixture I according to the invention can vary within wide limits depending on the intended use, for example between about 1 (detergents) and about 15% by weight (alcoholic solutions). In perfume bases or concentrates, the concentrations can of course also be higher.
In the following examples the temperatures are given in degrees Celsius.

2.7 kg of dry hexane, 1 kg of manganese dioxide (J. Chem. Soc. [1952], 1104) and 100 g of 2-ethyl3,6,6-trimethyl- are placed in a round bottom flask equipped with a stirrer and thermometer. 1 - (1-hydroxy-2-butenyl) -2-cyclohexene (containing approx. 85% a-isomer). The mixture is stirred for 48 hours at 20. It is then filtered through cellite and washed with ether. The solvent from the filtrate is removed by distillation and 95 g of crude 2-ethyl-3,6,6 trimethyl-1-crotonoyl-2-cyclohexene are obtained. The proportion of isomers corresponds to that given above for the starting material.
Distillation gives 58.8 g (58.9% yield) of the pure material: boiling point 63-66 / 0.07-1 mm Hg, nu20 = 1.4945-1.4959.
The 2-ethyl-3,6,6-tri methyl- 1 - (1-hydroxy-2-butenyl) -2-cyclohexene used as starting material can be obtained as follows:
162 g (0.9 mol) of 3-ethyl-4,7-dimethyl-octa-2,6-dienal and 220 ml of diethyl ether are placed in a round-bottom flask equipped with a thermometer and stirrer. 102 g (1.1 mol) of freshly distilled aniline are added dropwise with stirring, and stirring is then continued for a further 11/2 hours. The water formed is decanted off and the ethereal solution of the Schiff base formed is dried over sodium sulfate.
880 g of concentrated sulfuric acid are cooled to 200 in a round bottom flask under a nitrogen atmosphere. The ethereal solution of the Schiff base is then added at this temperature within an hour. The reaction mixture is stirred for a further 30 minutes at 200 and poured onto 1.5 kg of crushed ice. It is extracted with ether, washed three times with water, then with a 10% sodium carbonate solution and finally again with water. After evaporation of the ether, 128 g of crude 2-ethyl-3,6,6-trimethyl-1-formyl-2-cyclohexene remain, which after fractional distillation 62.2 g (38.4%) of the pure material (containing approx. 85% a-isomer) yield; Boiling point 83-88, nD20 = 1.4773.
25.2 g (1.05 g-atoms) of magnesium shavings in 80 ml of dried tetrahydrofuran are placed in a round-bottomed flask equipped with a stirrer, condenser, thermometer and dropping funnel. 133.1 g (1.1 g atoms) of 1-propene bromide in 150 ml of dried tetrahydrofuran are then slowly added with stirring. The mixture is brought to reflux temperature for 30 minutes and then left to cool (12 hours).
A solution of 180.3 g (1 mol) of the aldehyde obtained above in 250 ml of dried tetrahydrofuran is then added at 10. The reaction mixture is kept at 40 for 6 hours, allowed to cool again, moist ether is added and decomposed using a saturated ammonium chloride solution. The organic layer is washed with water, with tartaric acid and then again with water. After evaporation of the solvent, 239 g of crude 2-ethyl-3,6,6-trimethyl-1 - (1-hydroxy-2-butenyl) -2-cyclohexene (containing about 85% a-isomer) are obtained, their distillation 117 g (53%) of pure product with a boiling point of 88-95 / 0.2 mm Hg result; nun20 = 1.4923-1.4942.
Example 2
To a suspension, cooled to approx. 5-10 and kept under nitrogen, of 2.4 g (0.35 g atom) of finely cut lithium tape in 40 ml of abs. A few crystals of diphenyl are added to tetrahydrofuran. Then 2 ml of a solution of 9.4 g (70 mmol) of allyl phenyl ether in 10 ml of abs.
Ether added. When a greenish hue appears, the rest of the solution is added dropwise at -15 ".
After the addition, stirring is continued at 0 for 30 minutes.
The resulting allyl lithium solution is decanted from the excess lithium and transferred to a dropping funnel.
This solution is then added dropwise to a solution, cooled to 600 and kept under nitrogen, of 4.48 g (20 mmol) of 2-ethyl-3,6,6-trimethyl-2-cyclohexene-1-carboxylic acid ethyl ester in 40 ml of abs . Ether. After the addition, the mixture is allowed to warm to room temperature, the reaction mixture is poured into ice water and extracted with 3 portions of hexane. The combined hexane solutions are washed neutral with water (3 ×), dried over sodium sulfate and evaporated. Fractional distillation of the residue gives 3.75 g (85%) of 2-ethyl-3,6,6-trimethyl-1- [but-3-enoyl] -2-cyclohexene. Bp 57-58 / 0.04 mm Hg, nD20 = 1.4865.
A suspension of 3.5 g (15.9 mmol) of 2-ethyl-3,6,6 trimethyl-1 - [but-3-enoyl] -2-cyclohexene and 2.0 g of potash in 40 ml of acetone are stored for 24 hours stirred at room temperature under nitrogen. The salt is filtered off and the filtrate is evaporated. After fractional distillation, the residue gives 3.2 g (91.5%) of 2-ethyl-3,6,6-trimethyl-1crotonoyl-2-cyclohexene. Bp 59-60 / 0.04 mm Hg, n, ZO = 1.4952.
= 1.4952.
