Die Erfindung betrifft ein Verfahren zur Herstellung von heterocyciischen Verbindungen der Formel

worin Het einen Pyridazinyl-, Pyrimidyl-, Pyrazinyl- oder Pyridylrest darstellt, der mindestens einen Substituenten ausgewählt aus Niederalkylamino und Diniederalkylamino mit bis zu 7 C-Atomen in jedem der Niederalkylteile, Niederalkylenimino und Hydroxyniederalkylenimino mit 3-7 C-Atomen, Oxaniederalkylenimino oder Thianiederalkylenimino mit jeweils 4 oder 5 C-Atomen und Azaniederalkylenimino mit 2-6 C-Atomen, und der daneben noch weitere Substituenten enthalten kann, R1 Wasserstoff oder Methyl und R2 Niederalkyl, gegebenenfalls substituiertes Phenylniederalkyl, Carboxyniederalkyl oder funktionell abgewandeltes Carboxyniederalkyl bedeuten, ihren N-Oxiden und Salzen dieser Verbindungen.
Vor und nachstehend wird unter einem niederen Rest insbesondere ein solcher Rest mit bis zu 7 C-Atomen, vor allem bis zu 4 C-Atomen verstanden.
Niederalkylreste sind Reste mit bevorzugt bis zu 7 C-Atomen, vor allem bis zu 4 C-Atomen und sind z.B. Methyl, Äthyl, n-Propyl, i-Propyl, i-Propyl oder unverzweigtes oder an beliebiger Stelle gebundenes oder verzweigtes Butyl, Pentyl, Hexyl oder Heptyl.
Gegebenenfalls substituierte Phenylniederaikyfreste sind Mederalkylreste mit bis zu 7 C-Atomen, vor allem bis zu 4 C-Atomen, die in beliebiger Stelle durch gegebenenfalls substituierte Phenylgruppen substituiert sind. Als Phenylsubstituenten kommen vor allem Niederalkyl-, Niederalkoxygruppen, der Trifluormethylrest und Halogenatome in Frage. Als Beispiele sind 3-Phenyk-n-propyl oder vor allem Benzyl und 2-Phenyläthyl zu nennen.
Carboxyniederalkylreste sind Niederalkyfreste mit bevorzugt bis zu 7 C-Atome, vor allem bis zu 4 C-Atome, die in beliebiger Stelle durch Carboxy substituiert sind, wie z.B.
3-Carboxy-n-propyl, 4-Carboxy-n-butyl und vor allem Carboxymethyl und 2-Carboxyäthyl.
Funktionell abgewandeltes Carboxyniederalkyl ist z.B.
verestertes Carboxyniederalkyl, amidiertes Carboxyniederalkyl oder Cyanoniederaikyl.
Verestertes Carboxyniederalkyl ist z.B. mit einem aliphatischen Alkohol verestertes Carboxyniederalkyl. Der Niederalkylteil des veresterten Carboxyniederalkyls hat bevorzugt bis zu 7 C-Atome, vor allem bis zu 4 C-Atome. Aliphatische Alkohole sind solche, in denen die Hydroxylgruppe an ein C-Atom gebunden ist, welches nicht Glied eines aromatischen Systems ist. Geeignete aliphatische Alkohole sind z.B.
Cycloalkanole, wie solche mit 3-7, insbesondere 5-7 Ringgliedern, z.B. Cyclopropanoi, Cyclopentanol, Cyclohexanol und Cycloheptanol, Cycioalkyiniederaikanole, die z.B. obige Cycloalkylteile enthalten, wie Cyclopentyl-methanol, Cyciohexyl-methanol, 2-Gyclohexyi-äthanol und Cycloheptyl-methanol, Phenylniederalkanole, wie 2-Phenyläthanol und Benzylalkohol, wobei Phenylreste auch durch Halogen, Niederalkyl und oder Niederalkoxy, wie die oben genannten substituiert sein können, und insbesondere Niederalkanole, wie n-Propanol, iso-Propanol, geradkettiges oder verzweigtes Butanol, Pentanol, Hexanol oder Heptanol, und insbesondere Methanol oder Äthanol.
Verestertes Carboxyniederalkyl ist vor allem Methoxycarbonylmethyl, 2-Meth oxycarbonyl-äthyl, Äthoxycarbonylmethyl und 2-Äthoxy- carbonyläthyl.
Amidiertes Carboxyniederalkyl ist substituiertes oder unsubstituiertes Carbamoylniederalkyl. Der Niederalkylteil des amidierten Carboxyniederalkyls hat bevorzugt bis zu 7 C-Atome, vor allem bis zu 4 C-Atome. Substituiertes Carbamoyl hat z.B. die Formel -CONR5R6,worinR5Was- serstoff oder Niederalkyl ist, R6 Niederalkyl ist oder R5 und R6 zusammen Niederalkylen, Oxaniederalkylen, Thianiederalkylen oder Azaniederalkylen ist. Niederalkylen ist verzweigtes oder insbesondere geradkettiges Niederalkylen mit insbesondere 3-7, vor allem 4-6 C-Atomen in der Alkylenkette. Oxaniederalkylen ist verzweigtes oder insbesondere geradkettiges Oxaniederalkylen mit insbesondere 4 oder 5 C-Atomen in der Oxaalkylenkette.
Thianiederalkylen ist verzweigtes oder insbesondere geradkettiges Thianiederalkylen mit insbesondere 4 oder 5 C-Atomen in der Thiaalkylenkette. Azaniederalkylen ist verzweigtes oder geradkettiges Azaniederalkylen mit insbesondere 2-6, vor allem 4-6 C-Atomen in der Azaalkylenkette.
Amidiertes Garboxyniederalkyl ist demnach vor allem Carbamoylmethyl, 2-Carbamoyläthyl, N,N-Dimethylcarbamoyhnethyl, 2-(N,N-Dimethylcarba moyl)-äthyl, N,N-Diäthylcarbamoylmethyl, 2-(N,N-Diäthylcarbamoyl)-äthyl, Pyrrolidinocarbonyltnethyl, 2-Pyrrolidino- carbonyl-äthyl, Piperidinocarbonylmethyi, 2-Piperidinocarbonyläthyl, Morpholinocarbonylmethyl, 2-Morpholinocarbonyläthyl, Thiomorpholinocarbonylmethyi, 2-Thiomor pholinocarbonyläthyl, 2,6-Dimethylthiomorpholinocarbonylmethyl, 2- (2',6'-Dimethylthiomorpholinocarbonyl)-äthyl, Piperazinocarbonylmethyl,

Cyanoniederalkyi hat im Niederalkylteil vorzugsweise bis zu 7 C-Atome und vor allem bis zu 4 C-Atome und ist z.B. 3-Cyano-n-propyl, 4-Cyano-n-butyl, 5-Cyano-n-pentyl und vor allem 2-Cyanoäthyl und Cyanomethyl.

Als zusätzliche Eigenschaft besitzt ein Teil dieser Verbindungen eine sogenannte intrinsic sympathominetic activity (ISA) , das heisst, diese Verbindungen bewirken neben der p-Blockade (= Hauptwirkung) eine partielle p-stimula- tion.
Die neuen Verbindungen können daher zur Behandlung von Erkrankungen des Herz- und Kreislaufsystems verwendet werden. Die 3-Rezeptorenblocker können z.B. zur Therapie von Angina pectoris, Hypertonie und Herzrhytmusstörungen Anwendung finden. Die cardioselektiven Präparate bringen gegenüber den nicht cardioselektiven Vorteil, dass in den Dosen, welche für eine Blockade der p-Rezepto- ren des Herzens benötigt werden, noch keine Blockade von p-Rezeptoren in anderen Organen zu erwarten ist. Das Risiko, unerwünschte Nebenwirkungen, wie z.B. einen Bronchospasmus auszulösen, ist demnach sehr gering.
Im Gegensatz zu den cardioselektiven blockieren die nicht cardioselektiven Präparate entweder die -Rezeptoren in allen Organen etwa gleich stark oder in bestimmten Organen (wie z.B. in den Gefässen) bevorzugt.
Sie können aber auch als wertvolle Zwischenprodukte für die Herstellung anderer nützlicher Stoffe, insbesondere pharmazeutisch wirksamer Verbindungen verwendet werden.
Für Verbindungen der Formel I, worin Het substituiertes Pyrazinyl ist, stellt Formel I Verbindungen der allgemeinen Formel Ia

amino oder Diniederalkylamino mit bis zu 7 C-Atomen in jedem der Niederalkylteile, Acylamino, Acylaminoniederalkyl, Acylaminoniederalkenyl oder gegebenenfalls substituiertes Carbamoyl ist und n gleich 1, 2 oder 3 ist, wobei mindestens einer der Reste R8 Niederalkyienimino, Hydroxyniederalkylenimino, Oxaniederalkylenimino, Thianiederalkylenimino, Azaniederalkylenimino, Niederalkylamino oder Diniederalkylamino, der angegebenen Art ist.
Die neuen Verbindungen besitzen wertvolle pharmakologische Eigenschaften. Die Hauptwirkung der substituierten Pyridine, Pyrazine und Pyrimidine besteht in einer Blockade adrenergischer ,8-Rezeptoren, die sich z.B. als Hemmwirkung gegenüber Effekten von bekannten p-Rezeptoren-Stimula- toren in verschiedenen Organen nachweisen lässt: Hemmung der Isoproterenol-Tachykardie am isolierten Meerschweinchenherzen und der Isoproterenoi-Reiaxation an der isolierten Meerschweinchentrachea bei Konzentrationen von 0,001 bis 3 ,pg/ml, Hemmung der IsoproterenoloTachykardie und -Vasodilatation an der narkotisierten Katze bei intravenöser Gabe von 0,01 bis 30 mg/kg i.v.
Die genannten Verbindungen gehören entweder der Klasse der nicht cardioselektiven p-Rezeptorenblocker an, d.h. sie blockieren die ,8-Rezepto- ren an den Gefässsen bzw. in der Trachea in ähnlichen oder sogar kleineren Dosen bzw. Konzentrationen, als die p-Rezeptoren im Herzen, oder gehören der Klasse der sogenannten cardioselektiven 48-Rezeptorenblocker an, d.h. sie blockieren die p-Rezeptoren des Herzens schon in einem Dosen- bzw.
und die entsprechenden N-Oxide dar, worin
R1 und R2 die gleichen Bedeutungen haben wie oben,
R7 Wasserstoff, Cyano, Niederalkyl, Niederalkoxy-niederalkyl, Niederalkoxyniederalkenyl, Acylamino, Acylaminoniederalkyl oder Acylaminoniederalkenyl ist und
R4 obige Bedeutung hat.

Geeignete Substituenten für gegebenenfalls substituiertes Phenyl sind Halogen, Trifluormethyl, Niederalkyl, Nie deraltkenyl, Niederaikoxymethyl, Niederalkoxy und Niederalkenyloxy. Gegebenenfalls substituiertes Phenyl ist somit vor allem Phenyl, Chlorphenyl, Bromphenyl, Trifluormethylphenyl, Tolyl, Methoxymethylphenyl, Methoxyphenyl, Äthoxyphenyl und Allyloxyphenyl.
Niederalkoxyreste haben bevorzugt bis zu 7 C-Atome vor allem bis zu 4 C-Atome, wie z.B. Äthoxy, Propoxy, i-Propoxy, gerades oder verzweigtes, an beliebiger Stelle ge bundenes Butyl-, Pentyl-, Hexyl- oder Heptyloxy oder vor allem Methoxy.
Niederalkoxyniederalkyl hat im Niederalkylteil des Niederalkoxyteils bevorzugt bis zu 7 C-Atome, vor allem bis zu 4 C-Atome, wie iso- oder n-Propyl, gerades oder verzweigtes, in beliebiger Stelle gebundenes Butyl, Pentyl, Hexyl oder Heptyl, besonders Äthyl und vor allem Methyl. Der den Niederaikoxyteil tragende Niederalkylteil hat bevorzugt bis zu 7 C-Atome, vor allem bis zu 4 C-Atome. Beispielsweise ist Niederalkoxyniederalkyl Methoxymethyl, 2-Äthoxy äthyl, 3-Methoxy-n-propyl, 3-Äthoxy-n-propyl, 4-Methoxy -n-butyl oder vor allem 2-Methoxyäthyl und Äthoxymethyl.
Niederalkoxyniederalkenyl sind Niederalkenylreste mit bevorzugt bis zu 7 C-Atomen und vor allem bis zu 4 C-Atomen, die an beliebiger Stelle durch Niederalkoxy substituiert sind. Niederalkoxy hat im Niederaikylteil bevorzugt bis zu 7 C-Atome und vor allem bis zu 4 C-Atome. Niederalkoxyniederalkenyl ist somit bevorzugt 2-Methoxyvinyl, 2-Äthoxyvinyl und vor allem 3-Methoxyallyl und 3-Äthoxyallyl.
Niederalkenyloxyreste sind Reste mit bevorzugt bis zu 7 C-Atome, insbesondere mit 3 oder 4 C-Atome, wie der Methallyioxy- oder vor allem Allyloxyrest.
Niederalkoxyniederalkoxy hat in den Niederalkylteilen je bis zu 7 C-Atome, vor allem bis zu 4 C-Atome und ist z.B. Methoxymethoxy, Äthoxymethoxy, 2-Methoxy-äthoxy, 4-Methoxy-n-butoxy und insbesondere 3-Methoxy-n-propoxy.
Niederalkylthioreste haben bevorzugt bis zu 7 C-Atome, vor allem bis zu 4 C-Atome, wie z.B. Äthylthio, n-Propylthio, n-Butylthio, i-Propylthio oder insbesondere Methylthio.
Niederalkylaminoreste sind Reste mit bevorzugt bis zu 7 C-Atomen, insbesondere bis zu 4 C-Atomen, wie z.B.
Äthyl-, Propyl-, i-Propyl-, gerades oder verzweigtes, an beliebiger Stelle gebundenes Butyl-, Pentyl, Hexyl- oder Heptylamino oder vor allem Methylamino.
Diniederalkylaminoreste sind Reste mit bevorzugt bis zu 7 C-Atomen. insbesondere bis zu 4 C-Atomen in jedem der Niederalkylteile. Die beiden Niederalkylreste sind voneinander unabhängig und bilden zusammen mit dem Stickstoffatom Reste wie z.B. Diäthyl-, Methyläthyl-, Diäthyl-, Dipropyl-, Dibutylamino oder insbesondere Dimethylamino.
Niederalkyleniminoreste sind Reste mit z.B. 4 bis 8 Ringgliedern, wobei der Niederalkylenteil verzweigtes oder insbesondere geradkettiges Niederaikylen ist mit insbesondere 3-7, vor allem 4-6 C-Atomen in der Alkylenkette. Beispiele sind Pyrrolidino oder Piperidino.
Hydroxyniederalkyleniminoreste sind Reste mit z.B. 4-8 Ringgliedern, wobei der hydroxysubstituierte Niederalkylenteil verzweigtes oder insbesondere geradkettiges Niederalkylen ist, mit insbesondere 3-7, vor allem 4-6 C-Atomen in der Alkylenkette. Beispiele sind 4-Hydroxypiperidino oder 3-Hydroxypyrrolidino.
Oxaniederalkylenimino ist verzweigtes oder insbesondere geradkettiges Oxaniederalkylenimino mit insbesondere 4 oder 5 C-Atomen in der Oxaalkylenkette. Als Beispiel ist insbesondere Morpholino zu nennen.
Thianiederalkylenimino ist verzweigtes oder insbesondere geradkettiges Thianiederalkylenimino mit insbesondere 4 oder 5 C-Atomen in der Thiaalkylenkette. Als Beispiele sind insbesondere Thiomorpholino und 2,6-Dimethylthiomorpholino zu nennen.
Azaniederalkylenimino ist verzweigtes oder insbesondere geradkettiges Azaniederalkylenimino mit insbesondere 2-6, vor allem 4-6 C-Atomen in der Azaalkylenkette. Beispiele sind Piperazino, N'-Methylpiperazino oder N'-(P-Hydroxy- äthyl)-piperazino.
Acylamino ist z.B. niederes Alkanoylamino mit bis zu 7 C-Atomen und vor allem bis zu 4 C-Atomen im Niederalkylteil oder gegebenenfalls substituiertes Aroylamino, wie gegebenenfalls substituiertes Benzoylamino oder gegebenenfalls substituiertes Arylniederalkanoylamino oder Niederalkoxycarbonylamino.
Niedere Alkanoylaminoreste sind z.B. n-Propionylamino, n-Butyrylamino, n-Valerylamino, n-Hexanoylamino, n-Heptanoylamino oder vor allem Acetylamino. Gegebenenfalls substituierte Benzoylaminoreste sind z.B. Benzoylamino oder Niederalkoxybenzoylamino, wie n-Propoxybenzoylamino und vor allem Methoxy- und Äthoxybenzoylamino, oder Niederalkylbenzoylamino, wie n-Propyl-, gerades oder verzweigtes, in beliebiger Stelle gebundenes Butyl-, Pentyl-, Hexyl- oder Heptylbenzoylamino und vor allem Methylund Äthylbenzoylamino, oder Trifluormethylbenzoylamino oder Halobenzoylamino, wie Fluor-, Brom- und ganz besonders Chlorbenzoylamino.
Gegebenenfalls substituierte Arylniederalkanoylaminoreste sind Niederalkanoylaminoreste, die an beliebiger Stelle des Niederalkylteils z.B. gegebenenfalls substituierte Phenylgruppen tragen. Die Phenylgruppen können die gleichen Substituenten tragen, wie oben für gegebenenfalls substituierte Benzoylaminoreste erwähnt. Als Beispiele für gegebenenfalls substituiertes Arylniederalkanoylamino sind vor allem Phenylacetyiamino, 3-Phenyl-n -propionyramino, 4-Phenyl-n-butyryiamino, Chlorphenylacetylamino und Bromphenylacetylamino zu nennen.
Nie deralkoxycarb onylamino hat im Niederalkylteil von Niederalkoxy vorzugsweise bis zu 7 C-Atome und vor allem bis zu 4 C-Atome und ist z.B. n-Propoxycarbonylamino, n-Butoxycarbonylamino, i-Propoxycarbonylamino, tert.-Butoxycarbonylamino und vor allem Methoxycarbonylamino und Äthoxycarbonylamino.
Acylaminoniedernlkylreste sind Niederalkylreste mit vorzugsweise bis zu 7 C-Atomen und vor allem bis zu 4 C-Atomen, die in beliebiger Stelle durch Acylaminoreste substituiert sind. Acylamino ist z.B. niederes Alkanoylamino mit bis zu 7 C-Atomen und vor allem bis zu 4 C-Atomen im Niederalkylteil oder gegebenenfalls substituiertes Aroylamino, wie gegebenenfalls substituiertes Benzoylamino oder gegebenenfalls substituiertes Arylniederalkanoylamino oder Niederalkoxycarbonylamino.
Niedere Alkanoylaminoreste sind z.B. n-Propionylamino, n-Butyrylamino, n-Valerylamino, n-Hexanoylamino, n-Heptanoylamino oder vor allem Acetylamino. Gegebenenfalls substituierte Benzoylaminoreste sind z.B. Benzoylamino oder Niederalkoxybenzoylamino, wie n-Propoxybenzoylamino und vor allem Methoxy- und Äthoxybenzoylamino, oder Niederalkylbenzoylamino, wie n-Propyl-. gerades oder verzweigtes, in beliebiger Stelle gebundenes Butyl-, Pentyl-, Hexyl- oder Heptylbenzoylamino und vor allem Methylund Äthylbenzoyiamino, oder Trifluormethylb enzoylamino oder Halobenzoylamino, wie Fluor-, Brom- und ganz besonders Chlorbenzoylamino. Gegebenenfalls substituierte Arylniederalkanoylaminoreste sind Niederalkanoylaminoreste, die an beliebiger Stelle des Niederalkylteils z.B. gegebenenfalls substituierte Phenylgruppen tragen.
Die Phenylgruppen können die gleichen Substituenten tragen, wie oben für gegebenenfalls substituierte Benzoylaminoreste erwähnt.
Als Beispiele für gegebenenfalls substituiertes Arylniederalkanoylamino sind vor allem Phenylacetylamino, 3-Phenyl -n-propionylamino, 4-Phenyl-n-butyryiamino, Chlorphenylacetylamino und Bromphenylacetylamino zu nennen. Niederalkoxycarbonylamino hat im Niederalkylteil von Niederalkoxy vorzugsweise bis zu 7 C-Atome und vor allem bis zu 4 C-Atome und ist z.B.
n-Propoxycarbonylamino, n-Butoxycarbonylamino, i-Propoxycarbonylamino, tert. Butoxycarbonylamino und vor allem Methoxycarbonylami na und Äthoxycarbonyiamino. Als Beispiele für Acylaminoniederalkylreste sind demnach vor allem Acetylaminomethyl, 2-Acetylamino-äthyl, Benzoylaminomethyl, 2-Benzoylaminoäthyl, Methoxybenzoylaminomethyl, 2-Methoxybenzoylaminoäthyl, Äthoxybenzoylaminomethyl, 2-Äthoxybenzoylaminoäthyl, Methylbenzoylaminomethyl, 2- Methyl- benzoylaminoäthyl, Äthylbenzoylaminomethyl, 2-Äthylbenzoylaminoäthyl, Chlorbenzoylaminomethyl, 2-Chlorbenzoylaminoäthyl, Phenylacetylaminomethyl, 2-Phenylacetylamino äthyl, Chlorphenylacetylaminomethyl, 2-Chlorphenylacetylaminoäthyl, Methoxycarbonylaminomethyl, Äthoxycarbonylaminomethyl, 2-Methoxycarbonylaminoäthyl, 2-Äthoxycarbonylaminoäthyl,
3-Acetylaminopropyl und 3-Methoxycarbonylaminopropyl zu nennen.
Acylaminoniederalkenyl trägt im Niederalkenylteil bevorzugt bis zu 7 C-Atome und vor allem bis zu 4 C-Atome.

Niederalkylreste sind Reste mit bevorzugt bis zu 7 C-Atomen, vor allem bis zu 4 C-Atomen und sind z.B. Methyl, Äthyl, n-Propyl, i-Propyl oder unverzweigtes oder an beliebiger Stelle gebundenes oder verzweigtes Butyl, Pentyl, Hexyl oder Heptyl.
Hydroxyniederalkyl hat im Niederalkylteil vorzugsweise bis zu 7 C-Atome, vor allem bis zu 4 C-Atome und ist z.B.
3-Hydroxy-propyl, 2-Hydroxy-propyl, 1 -Methyl-2-hydroxy- äthyl oder verzweigtes oder an beliebiger Stelle gebundenes oder verzweigtes Hydroxybutyl, Hydroxypentyl, Hydroxyhexyl, Hydroxyheptyl und vor allem Hydroxymethyl und 2-Hydroxyäthyl.
Niederalkenyl hat bevorzugt bis zu 7 C-Atome und vor allem bis zu 4 C-Atome, wie Vinyl, 2-Methylvinyl, Methallyl und besonders Allyl.
Geeignete Substituenten für gegebenenfalls substituiertes Phenyl sind Halogen, Trifluormethyl, Niederalkyl, Niederalkenyl, Niederalkoxymethyl, Niederalkoxy und Nieder alkenyioxy. Gegebenenfalls substituiertes Phenyl ist somit vor allem Phenyl, Chlorphenyl, Bromphenyl, Trifluormethylphenyl, Tolyl, Methoxymethylphenyl, Methoxyphenyl, Äthoxyphenyl und Allyloxyphenyl.
Niederalkoxyreste haben bevorzugt bis zu 7 C-Atome, vor allem bis zu 4 C-Atome, wie z.B. Äthoxy, Propoxy, i-Propoxy, gerades oder verzweigtes, an beliebiger Stelle gebundenes Butyl-,. Pentyl-, Hexyl- oder Heptyloxy oder vor allem Methoxy.
Niederalkoxyniederalkyl hat im Niederalkylteil des Nie deralkoxyteils bevorzugt bis zu 7 C-Atome, vor allem bis zu 4 C-Atome, wie iso- oder n-Propyl, gerades oder ver zweigtes, in beliebiger Stelle gebundenes Butyl, Pentyl, Hexyl oder Heptyl, besonders Äthyl und vor allem Methyl. Der den Niederalkoxyteil tragende Niederalkylteil hat bevorzugt bis zu 7 C-Atome, vor allem bis zu 4 C-Atome. Beispiels weise ist Niederalkoxyniederalkyl, Methoxymethyl, Äthoxy methyl, 3-Methoxy-n-propyl, 3-Äthoxy-n-propyl, 4-Methoxy-n-butyl oder vor allem 2-Methoxyäthyl und 2-Äthoxy äthyl.
Niederalkoxyniederalkenyl sind Niederalkenylreste mit bevorzugt bis zu 7 C-Atomen und vor allem bis zu 4 C-Atomen, die an beliebiger Stelle durch Niederalkoxy substituiert sind. Niederalkoxy hat im Niederalkylteil bevorzugt bis zu 7 C-Atome und vor allem bis zu 4 C-Atome. Niederalkoxyniederalkenyl ist somit bevorzugt 2- Methoxyvinyl, 2-Äthoxyvinyl und vor allem 3-Methoxyallyl und 3-Äthoxyallyl.
Niederalkenyloxyreste sind Reste mit bevorzugt bis zu 7 C-Atome, insbesondere mit 3 oder 4 C-Atome, wie der Methallyloxy- oder vor allem Allyloxyrest.
Niederaikoxyniederalkoxy hat in den Niederalkylteilen je bis zu 7 C-Atome, vor allem bis zu 4 C-Atome und ist z.B. Methoxymethoxy, Äthoxymethoxy, 2-Methoxy-äthoxy, 4-Methoxy-n-butoxy und insbesondere 3-Methoxy-n-propoxy.
Niederalkylthioniederalkoxy hat in den Niederalkylteilen je bis zu 7 C-Atome, vor allem bis zu 4 C-Atome und ist z.B. Methylthiomethoxy, Äthylthiomethoxy, 3-Methylthiopropoxy, 4-Methylthiobutoxy und insbesondere 2-Methylthioäthoxy, 2-Athylthioäthoxy oder 2-(n-Propylthio) -äthoxy.
Niederalkylthioreste haben bevorzugt bis zu 7 C-Atome, vor allem bis zu 4 C-Atome, wie z.B. Äthylthio, n-Propylthio, n-Butylthio, i-Propylthio oder insbesondere Methylthio.
Niederalkylthioniederalkyl hat in den Niederalkylteilen je bis zu 7 C-Atome, vor allem bis zu 4 C-Atome und ist z.B. Methylthiomethyl, Äthylthiomethyl, 3 -Methylthiopro- pyl, 4-Methylthiobutyl und insbesondere 2-Methylthioäthyl, 2-Äthylthioäthyl oder 2-(n-Propylthio)-äthyl.
Niederalkylaminoreste sind Reste mit bevorzugt bis zu 7 C-Atomen, insbesondere bis zu 4 C-Atomen, wie z.B.
Äthyl-, Propyl-, i-Propyl-, gerades oder verzweigtes, an beliebiger Stelle gebundenes Butyl-, Pentyl-, Hexyl- oder Heptylamino oder vor allem Methylamino.
Diniederalkylaminoreste sind Reste mit bevorzugt bis zu 7 C-Atomen, insbesondere bis zu 4 C-Atomen in jedem der Niederalkylteile. Die beiden Niederalkylreste sind voneinander unabhängig und bilden zusammen mit dem Stickstoffatom Reste wie z.B. Diäthyl-, Methyläthyl-, Dipropyl-, Dibutylamino oder insbesondere Dimethylamino.
Niederalkylenaminoreste sind Reste mit z.B. 4 bis 8 Ringgliedern, wobei der Niederalkylenteil verzweigtes oder insbesondere geradkettiges Niederalkylen ist mit insbesondere 3-7, vor allem 4-6 C-Atomen in der Alkylenkette. Beispiele sind Pyrrolidino oder Piperidino.
Hydroxyniederalkyleniminoreste sind Reste mit z.B. 4-8 Ringgliedern, wobei der hydroxysubstituierte Niederalkylenteil verzweigtes oder insbesondere geradkettiges Niederalkylen, ist mit insbesondere 3-7, vor allem 4-6 C-Atomen in der Alkylenkette. Beispiele sind 4-Hydroxypiperidino oder 3-Hy droxypyrrolidino.
Oxaniederalkylenimino ist verzweigtes oder insbesondere geradkettiges Oxaniederalkylenimino mit insbesondere 4 oder 5 C-Atomen in der Oxaalkylenkette. Als Beispiel ist insbesondere Morpholino zu nennen.
Thianiederalkylenimino ist verzweigtes oder insbesondere geradkettiges Thianiederalkylenimino mit insbesondere 4 oder 5 C-Atomen in der Thiaalkylenkette. Als Beispiele sind insbesondere Thiomorpholino und 2,6-Dimethylthiomorpholino zu nennen.
Azaniederalkylenimino ist verzweigtes oder insbesondere geradkettiges Azaniederalkylenimino mit insbesondere 2-6, vor allem 4-6 C-Atomen in der Azaalkylenkette. Beispiele sind Piperazino-N'-Methylpiperazino oder N'-(P-Hydroxy- äthyl)-piperazino.
Acylamino ist z.B. niederes Alkanoylamino mit bis zu 7 C-Atomen und vor allem bis zu 4 C-Atomen im Niederalkylteil oder gegebenenfalls substituiertes Aroylamino, wie gegebenenfalls substituiertes Benzoylamino oder gegebenenfalls substituiertes Arylniederalkanoylamino oder Niederalkoxycarbonylamino.
Niedere Alkanoylaminoreste sind z.B. n-Propionylamino, n-Butyrylamino, n-Valerylamino, n-Hexanoylamino, n-Heptanoylamino oder vor allem Acetylamino. Gegebenenfalls substituierte Benzoylaminoreste sind z.B. Benzoylamino oder Niederalkoxybenzoylamino, wie n-Propoxybenzoylamino und vor allem Methoxy- und Äthoxybenzoylamino, oder Niederalkylbenzoylamino, wie n-Propyl-, gerades oder verzweigtes, in beliebiger Steile gebundenes Butyl-, Pentyl-, Hexyl- oder Heptylbenzoylamino und vor allem Methylund Äthylbenzoylamino, oder Trifluormethylbenzoylamino oder Halobenzoylamino, wie Fluor-, Brom- und ganz besonders Chlorbenzoylamino.
Gegebenenfalls substituierte Aryl niederalkanoyiaminoreste sind Niederalkanoylaminoreste, die an beliebiger Stelle des Niederalkylteils z.B. gegebenenfalls substituierte Phenylgruppen tragen. Die Phenylgruppen können die gleichen Substituenten tragen, wie oben für ge gebenenfalls substituierte Benzoylaminoreste erwähnt. Als Beispiele für gegebenenfalls substituiertes Arylniederalkanoylamino sind vor allem Phenyiacetylamino, 3-Phenyl-n -propionylamino, 4-Phenyi-n-butyrylamino, Chlorphenylacetylamino und Bromphenylacetylamino zu nennen.
Niederalkoxycarbonylamino hat im Niederalkylteil von Niederalkoxy vorzugsweise bis zu 7 C-Atome und vor allem bis zu 4 C-Atome und ist z.B. n-Propoxycarbonylamino, n-Butoxycarbonylamino, i-Propoxycarbonylamino, tert.-Butoxycarbonylamino und vor allem Methoxycarbonylamino und Äthoxycarbonylamino.
Acylaminoniederalkylreste sind Niederalkylreste mit vorzugsweise bis zu 7 C-Atomen und vor allem bis zu 4 C Atomen, die in beliebiger Steile durch Acylaminoreste substituiert sind. Acylamino ist z.B. niederes Alkanoylamino mit bis zu 7 C-Atomen und vor allem bis zu 4 C-Atomen im Niederalkylteil oder gegebenenfalls substituiertes Aroyl- amino, wie gegebenenfalls substituiertes Benzoylamino oder gegebenenfalls substituiertes Arylniederalkanoylamino oder Niederalkoxycarbonylamino.

Gegebenenfalls substituierte Arylniederalkanoylaminoreste sind Niederalkanoylamino- reste, die an beliebiger Stelle des Niederalkylteils z.B. gegebenenfalls substituierte Phenylgruppen tragen. Die Phenylgruppen können die gleichen Substituenten tragen, wie oben für gegebenenfalls substituierte Benzoylaminoreste erwähnt.
Als Beispiele für gegebenenfalls substituiertes Arylniederalkanoylamino sind vor aMem Phenylacetylamino, 3-Phenyl -n-propionylamino, 4-Phenyl-n-butyrylamino, Chlorphenylacetylamino und Bromphenylacetylamino zu nennen. Niederalkoxycarbonylamino hat im Niederalkylteil von Niederalkoxy vorzugsweise bis zu 7 C-Atome und vor allem bis zu 4 C-Atome und ist z.B.
n-Propoxycarbonylamino, n-But oxycarbonylamino, i-Propoxycarbonylamino, tert.Butoxycarbonylamino und vor allem Methoxycarbonylamino und Athoxycarbonylamino. Als Beispiele für Acylaminonieder alkylreste sind demnach vor allem Acetylaminomethyl, 2-Acetylamino-äthyl, Benzoylaminomethyl,
2-Benzoylamino- äthyl, Methoxybenzoylaminomethyl, 2-Methoxybenzoyl- aminoäthyl, Athoxybenzoylaminomethyl, 2-Athoxybenzoyl- aminoäthyl, Methylbenzoylaminomethyl, 2-Methylbenzoylaminoäthyl, Äthylbenzoylaminomethyl, 2-Äthylbenzoylami- noäthyl, Chlorbenzoylaminomethyl, 2-Chlorbenzoylamino- äthyl, Phenylacetylaminomethyl, 2-Phenylacetylaminoäthyl, 3-Acetylaminopropyl, 3-Methoxycarbonylaminopropyl, Methoxycarbonylaminomethyl, Äthoxycarbonylaminome- thyl, 2-Methoxycarbonylaminoäthyl und 2-Äthoxycarbonylaminoäthyl zu nennen.
Acylaminoniederalkenyl trägt im Mederalkenylteil bevorzugt bis zu 7 C-Atome und vor allem bis zu 4 C-Atome, während Acylamino z.B. Mederalkanoylamino, gogebenen- falls substituiertes Benzoylamino oder gegebenenfalls substituiertes Phenylaiederalkanoylamino oder Mederalkoxy- carbonylamino ist. Pyridin, Triäthylamin, Natriumhydroxyd, Kaliumhydroxyd, Natriummethylat, Kaliummethylat, Natrium äthylat oder Kaliumäthylat.
Die genannten Reaktionen werden in üblicher Weise in An- oder Abwesenheit von Verdünnungs-, Kondensationsund/oder katalytischen Mitteln, bei erniedrigter, gewöhnlicher oder erhöhter Temperatur, gegebenenfalls im geschlossenen Gefäss durchgeführt.
Je nach den Verfahrensbedingungen und Ausgangsstoffen erhält man die Endstoffe in freier Form oder in der ebenfalls in der Erfindung inbegriffenen Form ihrer Säureadditionssalze. So können beipieisweise basische, neutrale oder gemischte Salze, gegebenenfalls auch Hemi-, Mono-, Sesqui- oder Polyhydrate davon, erhalten werden. Die Säureadditionssalze der neuen Verbindungen können in an sich bekannter Weise in die freie Verbindung übergeführt werden, z.B. mit basischen Mitteln, wie Alkalien oder Ionenaustauschern. Andererseits können die erhaltenen freien Basen mit organischen oder anorganischen Säuren Salze bilden. Zur Herstellung von Säureadditionssalzen werden inbesondere solche Säuren verwendet, die zur Bildung von therapeutisch verwendbaren Salzen geeignet sind.
Als solche Säuren seien beispielsweise genannt: Halogenwasserstoffsäuren, Schwefelsäuren, Phosphorsäuren, Salpetersäure, Perchlorsäure, aliphatische, alicyclische, aromatische oder heterocyclische Carbon- oder Sulfonsäuren, wie Ameisen-, Essig-, Propion-, Bernstein-, Glykol-, Milch-, Äpfel-, Wein-, Zitronen-, Ascorbin-, Malein-, Fumar- oder Brenztraubensäure; Phenylessig-, Benzoe-, Anthranil-, p-Hydroxybenzoe-, Salicylsäure, Embonsäure, Methansulfon-, Äthansulfon-, Hydroxyäthansulfon-, Äthylensulfonsäure; Halogenbenzolsulfon-, Toluolsulfon-, Naphthalinsulfonsäure, Sulfanilsäure oder Cyclohexylaminsulfonsäure.
Diese oder andere Salze der neuen Verbindungen, wie z.B. die Pikrate oder Perchlorate, können auch zur Reinigung der erhaltenen freien Basen dienen, indem man die freien Basen in Salze überführt, diese abtrennt und aus den Salzen wiederum die Basen freisetzt. Infolge der engen Beziehungen zwischen den neuen Verbindungen in freier Form und in Form ihrer Salze sind im Vorausgegangenen und nachfolgend unter den freien Verbindungen sinn- und zweckmässig, gegebenenfalls auch die entsprechenden Salze zu verstehen.
Die Erfindung betrifft auch diejenigen Ausführungsformen des Verfahrens bei denen der Ausgangsstoff in Form eines Salzes vorliegt.
Die neuen Verbindungen können je nach der Wahl der Ausgangsstoffe und Arbeitsweisen, als optische Antipoden oder Racemate oder, sofern sie mindestens zwei asymmetische Kohlenstoffatome enthalten auch als Isomerengemische (Racematgemische) vorliegen.
Erhaltene Isomerengemische (Racematgemische) können aufgrund der physikalisch-chemischen Unterschiede der Bestandteile in bekannter Weise in die beiden stereoisomeren (diastereomeren) reinen Racemate aufgetrennt werden, beispielsweise durch Chromatographie und/oder fraktionierte Kristallisation.
Erhaltene Racemate lassen sich nach bekannten Methoden, beispielsweise durch Umkristallisation aus einem optisch aktiven Lösungsmittel, mit Hilfe von Mikroorganismen oder durch Umsetzen mit einer, mit der racemischen Verbindung Salze bildenden optisch aktiven Säure und Trennung der auf diese Weise erhaltenen Salze, z.B. aufgrund ihrer verschiedenen Löslichkeiten, in die Diastereomeren, aus denen die Antipoden durch Einwirkung geeigneter Mittel freigesetzt werden können, zerlegen. Besonders gebräuchliche, optisch aktive Säuren sind z.B. die D- und L-Formen von Weinsäure, Di-o-Toluylweinsäure, Äpfelsäure, MandeL- säure, Camphersulfonsäure oder Chinasäure. Vorteilhaft isoliert man den wirksameren der beiden Antipoden.
Zweckmässig verwendet man für die Durchführung der erfindungsgemässen Reaktionen solche Ausgangsstoffe, die zu den eingangs besonders erwähnten Gruppen von Endstoffen und besonders zu den speziell beschriebenen oder hervorgehobenen Endstoffen führen.
Die Ausgangsstoffe können nach an sich bekannten Methoden erhalten werden.
Verbindungen der Formeln IIa, IIb, IIc oder IId kön nen z.B. erhalten werden, wenn man (R7) (R4)-Pyrazinole mit Epichlorhydrin umsetzt, und anschliessend das erhaltene Reaktionsprodukt der Formel

Die neuen Verbindungen können als Heilmittel, z.B. in Form pharmazeutischer Präparate, Verwendung finden, welche sie oder ihre Salze in Mischung mit einem z.B. für die enterale oder parenterale Applikation geeigneten pharmazeutischen, organischen oder anorganischen, festen oder flüssigen Trägermaterial enthalten. Fir die Bildung desselben kommen solche Stoffe in Frage, die mit den neuen Verbindungen nicht reagieren, wie z.B. Wasser, Gelatine, Milchzucker, Stärke, Magnesiumstearat, Talk, pflanzliche Öle, Benzylalkohole, Gummi, Polyalkylenglykole, Vaseline, Cholesterin oder andere bekannte Arzneimittelträger. Die pharmazeutischen Präparate können z.B. als Tabletten, Dragees, Kapseln, Suppositorien, Salben, Crem oder in flüssiger Form als Lösungen (z.B. als Elixier oder Sirup), Suspensionen oder Emulsionen vorliegen.
Gegebenenfalls sind sie sterilisiert und bzw. oder enthalten Hilfsstoffe, wie Konservierungs-, Stabilisierungs-, Netz- oder Emulgiermittel, Salze zur Veränderung des osmotischen Druckes oder Puffer. Sie können auch noch andere therapeutisch wertvolle Stoffe enthalten. Die Präparate welche auch in der Veterinärmedizin Verwendung finden können, werden nach üblichen Methoden gewonnen.
Die tägliche Dosis beträgt etwa 40-150 mg im Falle eines Warmblüters vo netwa 75 kg Körpergewicht.
Die folgenden Beispiele erläutern die Erfindung ohne sie jedoch einzuschränken. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel I
9,8 g (0,04 Mol) 3-Chlor-2-(3'-isopropylamino-2'-hydroxy -propyloxy)-pyrazin werden mit 12,1 g (0,12 Mol) 4-Hydroxypiperidin 1 Stunde auf 130"C erwärmt. Nach dem Abkühlen wird das Reaktionsgemisch in Chloroform-Äther 1: 3 aufgenommen und mit 2-n Natronlauge und Wasser gewaschen.
Die organische Phase wird über Natriumsulfat getrocknet und im Wasserstrahlvakuum eingedampft. Man erhält 3-(4' -Hydroxy- 1 '-piperidyl)-2-(3'-isopropylamino-2'-hydroxy- propyloxy)-pyrazin, Smp. 88-91" aus Äther-Pentan. Das daraus mit Cyclohexylaminosulfonsäure bereitete Cyclamat kristallisiert aus Aceton; Smp. 90-920.
Beispiel 2
In analoger Weise wie im Beispiel 1 beschrieben, erhält man aus 9,8 g (0,04 Mol) 3-Chlor-2-(3'-isopropylamino-2' -hydroxy-propyloxy)-pyrazin und 12,0 g (0,12 Mol) N-Methylpiperazin 3-(4-Methyi-l -piperazinyl)-2-(3'-isopropyl- amino-2'-hydroxy-propyloxy)-pyrazin, Smp. 62-64" aus Petroläther. Das daraus mit Fumarsäure hergestellte Fumarat kristallisiert aus Methanol, Smp. 200-201".
Beispiel 3
In analoger Weise wie in den Beispielen 1-2 beschrieben lassen sich auch folgende Verbindungen herstellen:

The acid addition salts of the new compounds can be converted into the free compound in a manner known per se, e.g. with basic agents such as alkalis or ion exchangers. On the other hand, the free bases obtained can form salts with organic or inorganic acids. For the preparation of acid addition salts, those acids are used in particular which are suitable for the formation of therapeutically useful salts.

These or other salts of the new compounds, e.g. the picrates or perchlorates can also be used to purify the free bases obtained by converting the free bases into salts, separating these and in turn liberating the bases from the salts. As a result of the close relationships between the new compounds in free form and in the form of their salts, the free compounds in the preceding and in the following are meaningful and expedient, if appropriate also to mean the corresponding salts.
The invention also relates to those embodiments of the method in which the starting material is in the form of a salt.
Depending on the choice of starting materials and working methods, the new compounds can be present as optical antipodes or racemates or, provided they contain at least two asymmetric carbon atoms, also as isomer mixtures (racemic mixtures).
Mixtures of isomers (mixtures of racemates) obtained can be separated into the two stereoisomeric (diastereomeric) pure racemates in a known manner on the basis of the physico-chemical differences between the constituents, for example by chromatography and / or fractional crystallization.
Racemates obtained can be purified by known methods, for example by recrystallization from an optically active solvent, with the aid of microorganisms or by reaction with an optically active acid which forms salts with the racemic compound and separation of the salts obtained in this way, e.g. due to their different solubilities, decompose into the diastereomers, from which the antipodes can be released by the action of suitable agents. Optically active acids which are particularly common are e.g. the D- and L-forms of tartaric acid, di-o-toluyltartaric acid, malic acid, mandelic acid, camphorsulphonic acid or quinic acid. It is advantageous to isolate the more effective of the two antipodes.
For carrying out the reactions according to the invention, it is expedient to use those starting materials which lead to the groups of end products particularly mentioned at the beginning and especially to the end products specifically described or emphasized.
The starting materials can be obtained by methods known per se.

The new compounds can be used as medicaments, e.g. in the form of pharmaceutical preparations which use them or their salts in admixture with a e.g. contain pharmaceutical, organic or inorganic, solid or liquid carrier material suitable for enteral or parenteral administration. They can also contain other therapeutically valuable substances. The preparations, which can also be used in veterinary medicine, are obtained using conventional methods.
The daily dose is about 40-150 mg in the case of warm-blooded animals weighing about 75 kg.
