Bei der therapeutischen Anwendung physiologisch wirksamer Peptide, z. B. des Corticotropins, MSH, Calcitonins, des Hypertensins, Insulins, Wachstumshormons, Glucagons, Thyrotropins, LHRH, TRH, Vasopressins, Bradykinins u. a., besteht ein bisher noch nicht befriedigend gelöstes Problem darin, dass die Peptide im Körper zu schnell inaktiviert werden. Man hat auf verschiedene Weise versucht, die Wirksamkeit zu verlängenn, z. B. beim ACTH indem man Komplexe der Peptide mit anorganischen Stoffen wie schwerlöslichen Zinkverbindungen, beispielsweise Zinkhydroxyd oder Zinkphosphat, oder mit organischen Stoffen wie Gelatine, Polyvinylpyrrolidon, Phosphorsäureestern von Polyphenolen oder Polyalkoholen, beispielsweise Polyphloretinphosphat oder alkalischen oder sauren Polyaminosäuren beispielsweise Protamin oder Polyglutaminsäuren bildete.
Abgesehen davon dass die erzielte Wirkungsverlängerung nicht voll befriedigt, weisen gewisse der hergestellten Präparate auch den Nachteil auf, dass sie Allergien hervorrufen können.
Es wurde nun gefunden, dass man eine wesentlich stärkere Wirkungsverlängerung als bisher erzielen kann, wenn man als Trägermaterial für das Peptid ein Gel aus einem Fettsäure-Aluminiumsalz und einem bei Zimmertemperatur flüssigen Fettsäureester, insbesondere einem pflanzlichen Ö1, verwendet. Gegenstand der vorliegenden Erfindung ist daher ein speziell zur Herstellung injizierbarer öliger Peptid Präparate geeigneter Träger, der aus einem Gel enthaltend eine Mischung von 0,1 bis 10% (G/V) Fettsäure-Aluminiumsalz und einem bei Zimmertemperatur flüssigen Fettsäureester besteht.
Das als Träger verwendete Gel besteht aus einer Mischung von ca. 0,1 bis 10%, vorzugsweise 1-3 (Gewicht:Volumen) fettsaurem Aluminium und einem flüssigen langkettigen Fettsäureester. Das fettsaure Aluminiumsalz enthält als Fettsäure vor allem Stearinsäure, doch können auch andere langkettige gesättigte oder ungesättigte Fettsäuren mit 12 bis 22 Kohlenstoffatomen, z. B. die oben genannten, verwendet werden. Das fettsaure Aluminiumsalz ist vorzugsweise ein Monofettsäuresalz, z. B. Monostearat, enthält als pro Grammatom Aluminium ein Mol Fettsäure. Man kann aber auch Dioder Trifettsäuresalze des Aluminiums venvenden, insbesondere auch die im Handel erhältlichen Produkte, die nicht immer der chemischen Bezeichnung genau entsprechen, sondern nach ihrem Aluminiumhydroxidgehalt, z.
B. entsprechend der Formel Al(OH)n (Cl7H3sCOoH)3-ns worin n für 0,1 oder 2 steht, benannt werden. Aluminiummonostearat enthält beispielsweise gemäss U. S. Pharmacopeia XVIII 14,5 bis 16,0% Al2O3 und entsprechend variierende Mengen von Stearinsäure oder anderen Fettsäuren, besonders Palmitinsäure.
Der flüssige Fettsäureester enthält als Säurekomponente eine langkettige Fettsäure mit 8-22, besonders 12-22 Kohlenstoffatomen, wie z. B. die oben erwähnten Fettsäuren. Die Alkoholkomponente hat maximal 6 Kohlenstoffatome und ist ein ein- oder mehrwertiger, z. B. ein-, zwei- oder dreiwertiger Alkohol, z. B. Methanol, Äthanol, Propanol, Butanol oder Pentanol oder deren Isomere, vor allem aber Glycol oder Glyzerin. Als Fettsäureester sind daher beispielsweise zu nennen: Äthyloleat, Isopropylmyristat, Isopropylpalmitat, Labrafil M 2735 (Polyoxyäthylenglyzerintrioleat der Firma Gattefosse, Paris), Miglyol 812 (Triglyzerid gesättigter Fettsäuren oder Kettenlänge C8 bis C12 der Firma Chemische Werke Witten/Ruhr, Deutschland), besonders aber vegetabile Öle wie Baumwollsaatöl, Mandelöl, Olivenöl, Ricinusöl, Sesamöl, Sojabohnenöl, Rüböl, vor allem Erdnussöl.
Das Gel wird in bekannter Weise hergestellt, z. B. indem man den Ester zusammen mit dem Aluminiumsalz langsam unter Rühren auf ca. 140 C erhitzt. Bei ca. 115-125" C bildet sich ein klares Gel. Wenn die Temperatur von ca.
140 C ereicht ist, entfernt man die Heizquelle und lässt das Gel unter Rühren auf 1200 C und von dieser Temperatur an ohne Rühren auf Zimmertemperatur abkühlen. Das Gel sollte vorzugsweise eine Viskosität von 20-250 cP vorzugsweise 50-100 cP, bei 20 aufweisen.
Die mit dem erfindungsgemässen Träger hergestellten Präparate, z. B. ACTH-Präparate, weisen eine Verlängerung der Wirkung von mehreren Tagen, z. B. 7-8 Tagen auf. Dies konnte nach dem Stand der Technik keinesfalls vorhergesehen werden. Überraschend ist vor allem auch, dass diese lange Wirkung mit den üblichen ausserordentlich kleinen Dosen von Peptid, z. B. mit Bruchteilen eines Milligramms, erreicht werden kann.
Als physiologisch wirksame Peptide kommen die eingangs genannten Peptide in Betracht, vor allem Calcitonine und Peptide mit MSH- und ACTH-Wirkung. Zu den letzteren gehören die Peptide mit der Struktur der natürlichen MSHund ACTH-Peptide und synthetische Peptide oder Peptidamide mit kürzerer und/oder hinsichtlich einzelner-Aminosäuren abgeänderter Aminosäuresequenz. Als ACTH-wirksame Peptide oder Peptidamide sind z. B. diejenigen mit einer N-terminalen Sequenz bis zur 16. bis 39. Aminosäure der natürlichen Corticotropine zu nennen. In dieser Sequenz können eine oder mehrere der Aminosäuren vor allem der Aminosäuren 1 bis 4, fehlen wie z. B. in Angew.
Chemie 83, 155 (1971) gezeigt, oder es können eine oder mehrere Aminosäuren, z. B. in den Stellungen 1-5, 11, 15-18 und 25-33 durch andere Aminosäuren, Aminosäure 1 auch durch eine Desaminocarbonsäure ersetzt sein. So können beispielsweise die Serinreste Serin und/oder Serin3 durch Glycin oder Alanin, Tyrosin2 durch Phenylalanin, Methionin4 durch a-Amino-niederalkylessigsäure, worin Niederalkyl 2-4 Kohlenstoffatome aufweist, z. B. Norvalin, Valin, Norieucin, Leucin, Isoleucin, a-Aminobuttersäure, Glutaminsäures durch Glutamin, Lysin11 und/oder Lysin15,16 durch Ornithin, Arginin17,18 durch Lysin oder Ornithin, Aminosäure25 durch Valin. Serin1 kann auch z.
B. durch Prolin, a- oderss-Alanin, Threonin, Propionsäure, ss-Aminopropionsäure, ss-Hydroxy-propionsäure, Buttersäure, y Aminobuttersäure, a-Aminoisobuttersäure, Valeriansäure, Caprionsäure, E-Aminocapronsäure oder Phenylglycin ersetzt sein. Sämtliche Aminosäuren, ausser derjenigen in N-terminaler 1-Stellung haben notwendigerweise L-Konfiguration.
Vorzugsweise enthalten die neuen Präparate ACTH-wirksame Peptide, deren erste Aminosäure D-Konfiguration aufweist, vor allem D-Serin; weiter werden vorzugsweise Peptide mit einer Kettenlänge von 18-24 Aminosäuren, vor allem mit 18-19 Aminosäuren verwendet. Hervorzuheben sind auch Präparate, welche C-terminale N-unsubstituierte Amide der genannten ACTH-wirksamen Peptide enthalten, vor allem solche von Peptiden mit einer Sequenz von 18 Aminosäuren, in erster Linie D-Ser1-Lysl7 > 8-ss118-Cortico- tropin-Lysl8-amid. Peptide mit 25-39 Aminosäuren weisen im Bereich 25-33 vorzugsweise die für menschliches ACTH (vgl. Nature New Biology 235, 114 [1927]) festgestellte Sequenz auf.

Synthetische a-MSH-Peptide, welche, wie im US-Patent 3 803 309 beschrieben, eine die Ausschüttung des Wachstumshormons regulierende Wirkung haben und ebenfalls in den neuen Präparaten verwendet werden können, sind die im obigen Patent genannten Peptide, besonders solche mit der Sequenz der ersten 11-16 Aminosäuren des ACTH, worin die für ACTH oben genannten Austauschaminosäuren vorhanden sein können, und worin die erste Aminosäure vorzugsweise acyliert, besonders acetyliert ist, in erster Linie a-MSH und D-Ser1-a-MSH.
In den Präparaten zu verwendende Calcitonine sind die bekannten natürlichen Calcitonine wie Schweine-Calcitonin, menschliches Calcitonin, Salm-Calcitonin, Rinder-Calcitonin, Schaf-Calcitonin, sowie synthetische Analoge, in denen eine oder mehrere Aminosäuren durch andere ersetzt sind, z. B.
Methionin durch Norleucin, oder in denen die erste Aminosäure durch eine Desaminosäure oder eine Acylaminosäure ersetzt ist.
In den Präparaten wird das Peptid in freier Form oder in Form eines therapeutisch verwendbaren Säureadditionssalzes angewendet. Solche Salze leiten sich ab z. B. von anorganischen Säuren, wie Halogenwasserstoffsäuren, beispielsweise Salzsäure oder Bromwasserstoffsäure, Perchlorsäure, Salpetersäure oder Thiocyansäure, Schwefel- oder Phosphorsäure, oder organischen Säuren, wie Ameisensäure, Essigsäure, Propionsäure, Glykolsäure, Milchsäure, Brenztraubensäure, Oxalsäure, Malonsäure, Bernsteinsäure, Maleinsäure, Fumarsäure, Äpfelsäure, Weinsäure, Zitronensäure, Ascorbinsäure, Hydroxymaleinsäure, Dihydroxymaleinsäure, Benzoesäure, Phenylessigsäure, 4-Aminobenzoesäure, 4-Hydroxybenzoesäure, Anthranilsäure, Zimmtsäure, Mandelsäure, Salicylsäure, Pamoasäure, 4-Aminosalicylsäure,2-Phenoxybenzoesäure, 2 Acetoxybenzoesäure, Methansulfonsäure, Äthansulfonsäure, Hydroxyäthansulfonsäure,
Benzolsulfonsäure, p-Toluolsulfon säure, Naphthalinsulfonsäure oder Sulfanilsäure. Besonders vorteilhaft sind Salze mit langkettigen Fettsäuren, z. B. solchen mit 12-22 Kohlenstoffatomen wie Laurinsäure, Tridecylsäure, Myristinsäure, Pentadecylsäure, Palmitinsäure, Margarinsäure, Stearinsäure, Nonadecylsäure, Arachinsäure, Eicosacarbonsäure-(1), Behensäure, und entsprechenden ungesättigten Säuren wie z. B. Ölsäure, Elaidinsäure, Erucasäure, Brasidinsäure, Linolsäure.
Die Präparate werden hergestellt, indem man das Peptid oder Peptidsalz mit dem Gel vermischt. Dabei kann man beispielsweise so vorgehen, dass man die erforderliche Dosis des Peptids oder Peptidsalzes mikronisiert, z. B. auf eine Teilchengrösse von höchstens 50, vorzugsweise unter 20, und mit einer kleinen Menge des Gels verreibt und das erhaltene Konzentrat dann mit dem restlichen Gel verdünnt, oder man kann das Peptid bzw. Peptidsalz mit einer kleinen Menge des Esters mikronisieren und dann mit dem Gel vermischen. Ferner besteht die Möglichkeit die Peptide in einem Lösungsmittel, z. B. einem Konservierungsmittel wie Benzylalkohol, zu lösen und dann mit einer entsprechenden Menge des vorgängig hergestellten Gels zu vermischen.
So hergestellte Injektionspräparate zeichnen sich durch einen besonders feinen Verteilungsgrad der Präparate aus.
Die Herstellung der Injektionspräparate erfolgt unter antimikrobiellen Bedingungen. Dabei kann man so vorgehen, dass man die Wirkstoffe und Hilfsstoffe vorgängig der Abfüllung einer geeigneten antimikrobiellen Behandlung unterzieht. Das Abfüllen der keimfreien öligen Präparate in geeignete Behälter sowie das Verschliessen erfolgt ebenfalls unter antimikrobiellen Bedingungen.
Die in den Präparaten anzuwendende wirksame Dosis des Peptids ist diejenige Einzeldosis, die bei Anwendung des Peptids in wässeriger Suspension oder Lösungen verabreicht wird. Diese Dosis, die von der Art des Peptids abhängt, ist bekannt. Bei ACTH-Peptiden liegt sie z. B. bei 0,03 bis 3,0 mg/ml. Dem Gel können Konservierungsmittel, z. B. Benzylalkohol beigefügt werden.
Die nachstehenden Beispiele dienen zur Illustration der Erfindung. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel 1
Zu 100 ml Erdnussöl werden 2,0 g gepulvertes Aluminiummonostearat gegeben. Diese Suspension wird gut durchgemischt und unter Rühren und langsamem Erwärmen (2-3" pro Minute) auf 1.40" C erhitzt (bei ca. 115-125" bildet sich ein klares Gel). Dann wird die Heizung abgestellt und weiter gerührt, bis die Temperatur auf 1200 C gefallen ist. Von dieser Temperatur an lässt man das Gel ohne Rühren auf Zimmertemperatur abkühlen.
Dieser Träger kann wie folgt zur Herstellung injizierbarer ACTH-Peptidpräparate verwendet werden: a) Man erwärmt 2,0 ml des Gels auf ca. 30-40 und vermischt es mit 0,6 mg feinst gemahlenem (Korngrösse unter 20,u) D-SerÚ-Lys17,18-ss1-18-corticotropin-Lys18-amid Hexaacetat. Diese Mischung wird unter Rühren mit weiterem Gel von Raumtemperatur auf 20,0 ml verdünnt, so dass in 1,0 ml Gel 0,03 mg Peptidamidsalz enthalten sind.
b) Man erwärmt 2,0 ml des Gels auf ca. 30-40" und vermischt es mit einer Lösung von 0,6 mg D-Ser1-Lys17 18 ssl-18¯ Corticotropin-Lys18-amid-hexastearat in 0,4 g Benzylalkohol.
Die Mischung wird durch Zugabe von weiterem Gel auf 20,0 ml unter Rühren verdünnt, so dass 0,03 mg Peptidamidsalz pro ml Gel enthalten sind.
c) Man erwärmt 2,0 ml des Gels auf ca. 30-40 und vermischt es mit 6,0 mg feinst gemahlenem (Korngrösse unter 20 ) D-SerÚ-Lys17,18-ssÚ-18-Corticotropin-Lys18-amid. Die Mischung wird durch Zugabe von weiterem Gel auf 20,0 ml unter Rühren verdünnt, so dass 0,3 mg Peptidamid pro ml Gel erhalten sind.
d) Man erwärmt 2,0 ml des Gels auf 30-40" und vermischt es mit 20,0 mg feinst gemahlenem (Korngrösse unter 20 ist) Synathen 6 (ssl-24-Corticotropin-hexaacetat). Man verdünnt unter Rühren mit weiterem Gel auf 20,0 ml, so dass pro ml Gel 1,0 mg Peptidsalz enthalten sind.
Beispiel 2
Zu 100 ml Miglyol < @ 812 werden 3,5 g gepulvertes Aluminiummonostearat gegeben. Diese Suspension wird gut durchgemischt und unter Rühren und langsamem Erwärmen (2-3 > pro Minute) auf ca. 140 C erhitzt (bei ca. 115 bis 125 C bildet sich ein klares Gel). Dann wird die Heizung abgestellt und unter Rühren auf 1200 abgekühlt. Von dieser Temperatur an lässt man das Gel ohne Rühren auf Zimmertemperatur abkühlen.
Das Gel kann wie folgt zur Herstellung injizierbarer ACTH-Peptidpräparate verwendet werden:
Man envärmt 2,0 ml des Gels auf ca. 30-40" und vermischt es mit 10,0 mg feinst gemahlenem (Korngrösse unter 20C") D-serÚ-Lys17,18-ss1-18-corticotropin-Lys18-amid Die Mischung wird unter Rühren mit weiterem Gel auf 20,0 ml verdünnt, so dass 0,5 mg Peptidamid pro ml Gel vorhanden sind.
In the therapeutic use of physiologically active peptides, e.g. B. corticotropins, MSH, calcitonins, hypertensins, insulin, growth hormone, glucagons, thyrotropins, LHRH, TRH, vasopressins, bradykinins and the like. a., a problem that has not yet been satisfactorily resolved is that the peptides in the body are inactivated too quickly. Apart from the fact that the prolonged effect achieved is not fully satisfactory, certain of the preparations produced also have the disadvantage that they can cause allergies.
It has now been found that a much greater prolongation of activity than before can be achieved if a gel made of a fatty acid aluminum salt and a fatty acid ester, in particular a vegetable oil, which is liquid at room temperature, is used as the carrier material for the peptide. The present invention therefore relates to a carrier especially suitable for the production of injectable oily peptide preparations, which consists of a gel containing a mixture of 0.1 to 10% (w / v) fatty acid aluminum salt and a fatty acid ester which is liquid at room temperature.
