Die Erfindung betrifft ein Verfahren zur Herstellung von 1,4-disubstituierten 2(1H)-Chinazolinen. Die erfindungsgemäss hergestellten Verbindungen zeigen vorzügliche pharmakologische Wirkungen und dienen als entzündungshemmende Mittel, Analgetika und als Mittel zur Beeinflussung des Zentral- nervensystems.
Aufgabe der Erfindung war es, ein neues, in grossem Massstab durchführbares Verfahren zur Herstellung neuer 1,4 disubstituierter 2(1H)-Chinazolinone mit vorzüglichen pharmakologischen Wirkungen zur Verfügung zu stellen.
Diese Aufgabe wird durch die Erfindung gelöst.
Gegenstand der Erfindung ist somit ein Verfahren zur Herstellung von 2(1H)-Chinazolinonen der allgemeinen For- mel:

Die erfindungsgemäss hergestellten Verbindungen der allgemeinen Formel I können als niedere Alkylreste, z.B. die Methyl-, Äthyl-, n-Propyl-, Isopropyl-, n-Butyl-, Isobutyloder tert.-Butylgruppe, als niedere Alkoxyreste z.B. die Me- thoxy, Äthoxy-, n-Propoxy-, Isopropoxy-, n-Butoxy-, Iso- butoxy- und tert.-Butoxygruppe, als nie'dere Cycloalkylreste z.B. die Cyclopropyl-, Cyclobutyl-, Cyclopentyl, Cyclohexyl, Methylcyclopropyl- und Dimethylcyclopropylgruppe, als nie- dere Alkylthioreste z.B. die Methylthio- und Äthylthiogruppe, als Trihalogenmethyfreste z.B. die Trifluormethyl-, Trichlormethyl- und Chlordifluormethylgruppe, als Cycloalkenylreste z.B.
die Cyclopentenyl- und Cyclohexenylgruppe und als Halogenatome Fluor-, Chlor-, Brom- und Jodatome enthalten.
Die Alkylenreste der allgemeinen Formel -CnH2n- können z.B. die Methylen-, Äthylen-, l-Methyläthylen-, 2-Methyl äthyen- und Trimethylengruppe sein.
Die Umsetzung wird vorzugsweise in einem Lösungsmittel oder einem Lösungsmittelgemisch durchgeführt. Spezielle Beispiele solcher Lösungsmittel sind Methanol, Äthanol, Iso- propanol, tert.-Butanol, Mono- und Dialkyläther von Äthylenglykol und ihre Derivate, Äthylenglykol, Diäthylenglykol, Dioxan, Aceton, Pyridin, Benzol, Toluol, Xylol, Dimethylsulfoxid und Dimethylformamid. Diese Lösungsmittel können einzeln oder als Gemische verwendet werden. Der Ammoniak kann in die Umsetzung gasförmig, als Lösung in einem Alkohol, z.B. in Methanol oder Äthanol, als flüssiger Ammoniak oder als Ammoniumsalz, z.B. Ammoniumacetat, Am- moniumformiat, Ammoniumcarbamat, Ammoniumphosphat oder Ammoniumcarbonat, aus dem während der Umsetzung Ammoniak freigesetzt wird, eingebracht werden.
Wenn ein Ammoniumsalz eingesetzt wird, kann die Umsetzung in Ge- genwart oder in Abwesenheit einer Base, wie Natriumhydroxid, Kaliumhydroxid, Kaliumcarbonat, Natriummethylat, Natriumäthylat, Kaliumäthylat, Triäthylamin oder Pyridin, durchgeführt werden. Die Umsetzung kann auch durch Erhitzen einer Verbindung der allgemeinen Formel II in flüssigem Ammoniak oder durch Umsetzen einer Verbindung der allgemeinen Formel II mit einem Ammoniumsalz in der Schmelze durchgeführt werden. Nach vollständiger Umsetzung wird das Reaktionsgemisch vorzugsweise eingeengt oder mit Wasser verdünnt, und der so erhaltene Nieder- schlag wird mit Vorteil mit einem Lösungsmittel, wie einem Äther oder einem Alkohol gewaschen, wobei ein 1 ,4-disubsti- tuiertes 2(1H)-Chinazolinon der allgemeinen Formel I erhalten wird.
Die als Ausgangsverbindungen eingesetzten Verbindungen der allgemeinen Formel II, unter denen sich neue Ver- bindungen befinden, können in einfacher Weise sowohl durch Umsetzen eines entsprechenden 2-N-monosubstituierten Ami- nophenylketons mit einem niederen Halognameisensäurealkylester in Gegenwart oder Abwesenheit eines Lösungsmittels und eines basischen Kondensationsmittels als auch durch Umsetzen eines entsprechenden 2-Alkoxycarbonylaminophe- nylketons mit einem Halogenwasserstoffsäure oder einem Sulfonsäureester der allgemeinen Formel
R-CnH2n-OH in der R und n die vorstehend angegebene Bedeutung haben, in einem Lösungsmittel hergestellt werden.

Die erfindungsgemäss hergestellten Verbindungen der allgemeinen Formel 1 sind neu und weisen sowohl ausgezeichnete entzündungshemmende und analgetische Wirkungen als auch vorzügliche Wirkungen auf das Zentralnervensystem bei geringer Toxizität auf. Die erfindungsgemäss hergestellten Verbindungen sind daher wertvolle ArzneimitteL Sie können ferner als Zwischenprodukte zur Herstellung anderer Arzneimittel dienen.
So zeigt z.B. l-Cyclopropylmethyl- -4-phenyl- 6-chlor-2(1 H)-chinazolinon eine bemerkenswerte EIemnwirkung auf das durch Carragenin induzierte modem der Ratte, wobei keine toxischen Symptome beobachtet werden und nach oraler Verabreichung von 1000 mg/kg an Rat tell keine okkulte Blutung in den Faeces festgestellt wird.
Die erfindungsgemäss hergestellten Verbindungen weisen eine stärkere entzündungshemmende Wirkung als 1,2-Diphe- nyl-3,5-dioxo-4n-butylpyrazolidin auf, und die akute, subakute und chronische Toxizität der erfindungsgemäss hergestellten Verbindungen ist viel geringer als die Toxizität von 1 ,2-Diphenyl-3 ,5-dioxo-4-n-butylpyrazolidin. Die erfindungsgemäss hergestellten Verbindungen können parenteral oder oral in üblichen Dosierformen, d.h., in fester oder flüssiger Forst, wie in Form von Tabletten, Dragees, Kapseln, Suspensionen, Lösungen und Elixieren, verabreicht werden, wobei die Dosierung den jeweiligen Erfordernissen angepasst wird.
Das Beispiel erläutert die Erfindung.
Beispiel
Ein Gemisch aus 3,2 g 2.(2-Äthoxyäthylamino)-5-chlor- -2'-fluorbenzophenon und 10 ml Chlorameisensäureäthylester wird 5 Stunden auf 110 C erhitzt. Das Reaktionsgemisch wird unter vermindertem Druck eingedampft, wobei 2-[N-(2 -Äthoxyäthyl)-äthoxycarbonylamino] -5-chlor-2'-fluorbenzo- phenon als braunes ö1 erhalten wird. Diese Verbindung wird in 50 ml Dimethylsulfoxid gelöst. Die so erhaltene Lösung wird mit 8 g Ammoniumacetat und 1,2 g Kaliumhydroxid versetzt. Das erhaltene Gemisch wird 20 Stunden auf einem Ölbad bei einer Badtemperatur von 1300C erhitzt. Nach dem Abkühlen wird das Reaktionsgemisch unter Rühren in 300 ml Wasser gegossen.
Der Niederschlag wird abfiltriert, mit Di äthyläther und anschliessend mit Wasser gewaschen und ge- trocknet, wobei 1-(2-Äthoxyäthyl)-4-(o.fluorphenyl)-5-chor- -2(1H)-chinazolinon in Form blassgelber Nadeln vom Fp. 146 bis 147"C erhalten wird.
