Gegenstand der Erfindung ist ein Verfahren zur Herstellung neuer 4- oder 5-Nitro-imidazole der Formel I

worin einer der Reste R1 und R2 Wasserstoff oder Niederalkyl und der andere die Nitrogruppe ist, Rb Wasserstoff, oder gegebenenfalls substituiertes Niederalkyl ist, R Oxo oder Thioxo ist, R5 Wasserstoff, gegebenenfalls substituiertes Niederalkyl oder Phenyl, Acyl oder ein Heteroarylrest ist und R6 und R7 unabhängig voneinander Wasserstoff oder Niederalkyl oder gemeinsam den fehlenden Teil eines gegebenenfalls substituierten ankondensierten Benzolkerns bedeuten, und ihrer Salze.
Vorstehend und nachstehend ist unter einem niederen Rest, wie z.B. einem Niederalkyfrest, ein Rest, z.B. ein Al kylrest zu verstehen, welcher nicht mehr als 8 und insbesondere nicht mehr als 4 Kohlenstoffatome enthält.
Niederalkyl ist somit geradkettiges oder verzweigtes, in beliebiger Stellung gebundenes Octyl, Heptyl, Hexyl, Pentyl oder insbesondere Butyl, Propyl oder speziell Äthyl oder Methyl. Substituiertes Niederalkyl ist insbesondere Niederalkyl, welches einen oder mehrere Substituenten, die gleich oder verschieden sein können, trägt.
Als solche Substituenten sind insbesondere zu nennen: Aryl, vor allem gegebenenfalls substituiertes Phenyl, die Hydroxyl- und Mercaptogruppe, Halogen, vor allem Fluor, Chlor oder Brom, Niederalkoxy, vor allem Butoxy, Propoxy, Isopropoxy oder speziell Äthoxy oder Methoxy, Alkylmercapto, wie z.B. die durch Niederalkyl substituierte Mercaptogruppe, vor allem Butylmercapto, Isopropylmercapto oder speziell Äthyl- oder Methylmercapto, die freie Aminogruppe, sekundäre Aminogruppen, vor allem gegebenenfalls substituierte Anilinogruppen oder Niederalkylaminogruppen, wie Butyl-, Propyl-, Isopropyl- oder speziell Äthyl- oder Methylamino, oder tertiäre Aminogruppen, wie gegebenenfalls substituierte N-Niederalkylanilinogruppen, zB. die N-Butyl-, -Propyl-,
Isopropyl- oder speziell -Äthyl- oder -Methyl-anilinogruppe oder vor allem Diniederalkylaminogruppen, z.B. Dibutyl-, Diisobutyl-, Dipropyl-, Diisopropyl-, Äthylmethyl- oder speziell Diäthyl- oder Dimethylamino, sowie insbesondere Alkylenamino- und Oxa-, Azaoder Tniaalkylenaminogruppen, wie die gegebenenfalls C-me thylierte Pyrrolidino-, Piperidino-, Morpholino-, Thiomorpholino-, 2,6-Dimethylthiomorpholino-, Piperazino-, N'-Methylpiperazino- oder N'-68-Hydroxyäthyl)-piperazinogruppe, oder Sulfonylgruppen, wie Arylsulfonylgruppen, z.B. gegebe- nenfalls substituierte Benzolsulfonylgruppen, oder Alkan-, vor allem Niederalkansulfonylgruppen, z.B. die Methanoder Äthansulfonylgruppe.
Acyl ist insbesondere der Rest einer von einem aromatischen oder aliphatischen Kohlenwasserstoffrest abgeleiteten Carbonsäure, vor allem eine Aroylgruppe, wie z.B. eine gegebenenfalls substituierte Benzoylgruppe, oder eine Alkanoylgruppe, speziell eine Niederalkanoylgruppe, wie zB, Valeroyl Isovaleroyl, Pivaloyl, Butyryl, Isobutyryl, Propionyl, Formyl und speziell Acetyl, oder der Rest einer organischen Sulfon säure, vor allem einer aromatischen Sulfonsäure, wie z. B.
der Benzol-, Toluol- oder Brombenzolsulfonsäure, oder einer aliphatischen Sulfonsäure, z.B. der Methan- oder Äthansul- fonsäure.
Unter gegebenenfalls substituiertem Phenyl oder Benzoyl oder einem gegebenenfalls substituierten ankondensierten Benzolring sind sowohl diese Gruppen selbst als auch durch einen, zwei oder mehr als zwei gleiche oder verschiedene Substituenten, wie Niederalkyl, Niederalkoxy, Halogen, vor allem Chlor oder Brom, Nitro und/oder Trifluormethyl substituierte Phenylgruppen, Benzoylgruppen und ankondensierte Benzolringe zu verstehen.
Als Halogenatome kommen insbesondere Fluor- oder Bromatome und vor allem Chloratome in Betracht.
Ein Heteroarylrest R5 ist beispielsweise ein in 2-Stellung gebundener Imidazolyfrest, vor allem 2-Imidazolyfrest der auch in l-Stellung insbesondere durch Acyl, Niederalkyl oder substituiertes Niederalkyl, in einer der Stellungen 4 oder 5 durch Niederalkyl und in der anderen der genannten Stellungen durch Niederalkyl oder die Nitrogruppe substituiert sein kann.
Die neuen Imidazole besitzen wertvolle pharmakologische Eigenschaften. So zeigen sie insbesondere Wirkungen gegen Bakterien, speziell grammnegative Keime, Protozoen und Würmer, wie Trichomonaden, Schistosomen, Coccidien, Filarien und vor allem Amoeben, wie sich im Tierversuch zeigen lässt, z.B. an der Leber von gesunden Hamstern, die künstlich mit Entamoeba histolytica infiziert ist, bei Gabe von Dosen von etwa 30 bis etwa 300 mg/kg/p.o. Die neuen Imidazole können daher als Mittel gegen Schistosomen, Filarien, Trichomonaden, Bakterien und insbesondere gegen Amoeben verwendet werden. Ferner können die neuen Imidazole als Ausgangs- oder Zwischenprodukte für die Herstellung anderer, insbesondere therapeutisch wirksamer Verbindungen dienen.
Bevorzugt ist die Gruppe Ia derjenigen Verbindungen der Formel I, worin der Rest R1 Wasserstoff oder Niederalkyl ist, R2 die Nitrogruppe ist, R3 Wasserstoff, Niederalkyl, Hydroxyniederalkyl, Aminoniederalkyl, Mono- oder Di-niederalkylamino-niederalkyl, Alkylenaminoniederalkyl, gegebenenfalls C-methyliertes Morpholino-, Thiomorpholino- oder N'-Methylpiperazino-niederalkyl, Niederalkoxyniederalkyl, Halogenniederalkyl, Niederalkylmercaptoniederalkyl ist, R" Oxo oder Thioxo ist, R5 eine der für R3, angegebenen Bedeutungen hat Niederalkanoyl oder gegebenenfalls durch Niederalkyl, Niederalkoxy, Halogen, Trifluormethyl und/oder Nitro substituiertes Benzoyl bedeutet und R,
und RT unabhängig voneinander Wasserstoff oder Niederalkyl bedeuten oder gemeinsam den fehlenden Teil eines gegebenenfalls durch Niederalkyl, Niederalkoxy, Halogen, Trifluormethyl und/ oder Nitro substituierten ankondensierten Benzolkerne bilden.
Insbesondere geeignet ist die Gruppe Ib derjenigen Verbindungen der Formel I, worin R1 Wasserstoff oder Niederalkyl und R2 die Nitrogruppe ist, R > Wasserstoff, Niederalkyl, Hydroxyniederalkyl, vor allem 2-Hydroxypropyl, 1oder 2-Hydroxyäthyl odder Hydroxymethyl, Amine, Monooder Diniederalkylamino-, Pyrrolidino, Piperidino-, Morpholino-, Thiomorpholino- odedr N'-Methylpiperazinoniederalkyl, vor allem -methyll oder -äthyl, Chlor- oder Bromniederalkyl, vor allem -äthyl, Niederalkoxy-, vor allem Methoxyoder Athoxyniederalkyl ist, R4 Oxo oder Thioxo ist, R5 eine der für R8 angegebenen Bedeutungen hat oder ein Rest der

Vor allem geeignet ist jedoch die Gruppe Id derjenigen Verbindungen der Formel 1, worin R1 Wasserstoff oder Niederalkyl und R2 die Nitrogruppe ist, R4 Oxo oder Thioxo ist, R3 und R5 unabhängig voneinander Wasserstoff, Niederalkyl, 2-Hydroxyäthyl oder -propyl, Hydroxymethyl, 2-Diniederalkylaminoäthyl oder -propyl oder Dirsederalkylaminomethyl ist und R5 auch Niederalkanoyl sein karin, und R5 und R7 unabhängig voneinander Wasserstoff oder Niederalkyl bedeuten oder gemeinsam den fehlenden Teil eines ankondensierten Benzolkernes bilden.
Von den vorstehend genannten Verbindungen der Gruppe Id ist wiederum die Gruppe Ic hervorzuheben, worin R1 Wasserstoff oder Methyl und R2 die Nitrogruppe ist, R4 Oxo oder Thioxo ist, R, und R5 unabhängig voneinander Niederalkyl, Hydroxymethyl oder Hydroxyäthyl bedeuten und R5 auch Niederalkanoyl sein kann und R6 und R7 Wasserstoff sind.
Ganz besondere Erwähnung verdient jedoch die Gruppe If derjenigen Verbindungen der Formel I, worin R1 Wasserstoff und R2 die Nitrogruppe ist, R3 Wasserstoff oder vor allem Niederalkyl mit 1-4 Kohlenstoffatomen, R4 Oxo oder vor allem Thioxo ist, R5 Niederalkyl mit 1-4 Kohlenstoffatomen, vor allem Methyl oder gegebenenfalls durch Methyl, Methoxy, Chlor, Brom, Trifluormethyl substituiertes vor allem aber unsubstituiertes Phenyl ist und R6 und R7 Wasserstoff bedeuten, namentlich das l-Methyl-3-(1-methyl-5-nitro-2-imidazolyl)-benzimidazol- -2-(3H)-on,
das 1-Methyl-3-(1-methyl-5-nitro-2-imidazolylfibenzimidazol- -2-(3H)-thion, das 1 -Methyl-3-(l -methyl-5-nitro-2-imidazolyl)-4-imidazolin- -2-on und ganz besonders das l-Methyl-3-(1-methyl-5-nitro-2-imidazolyl)-4-imidazolin- -2-thion, welches an der Leber von gesunden Hamstern, die künstlich mit Entamoeba histolytica infiziert wurde, bei Gaben von Dosen zwischen 30 und 100 mg/kg/p.o. eine deutliche Abzessfreiheit bewirkt.
Das erfindungsgemässe Verfahren zur Herstellung der neuen 4- oder 5-Nitroimidazole ist dadurch gekennzeichnet, dass man eine Verbindung der Formel II

wobei R1, R2, R3, R4, R5, R6 und R7 die angegebenen Bedeutungen haben, R'4 ein Sauerstoff- oder vor allem ein Schwefelatom ist und X und Y unter Hinterlassung einer direkten Bindung abspaltbare Reste sind, kondensiert, d.h. X und Y unter Hinterlassung einer direkten Bindung abspaltet.
So kann einer der Reste X und Y, vor allem Y ein kationisch abspaltbarer Rest, z.B. ein Metallatom, z.B. ein Alkalimetall- oder Erdalkalimetallatom, oder insbesondere ein Wasserstoffatom und der andere, insbesondere X, ein austauschbarer Rest sein.
Ein austauschbarer Rest ist z.B. eine reaktionsfähige ver ätherte Hydroxylgruppe, eine freie oder verätherte Mercaptogruppe, eine Ammoniumgruppe oder insbesondere eine reak tionsfähige veresterte Hydroxylgruppe oder eine Sulfonylgruppe.
Eine reaktionsfähige verätherte Hydroxylgruppe ist z.B.
eine mit einem aromatischen oder aliphatischen, vor allem einem niederen aliphatischen Alkohol verätherte Hydroxylgruppe, wie eine gegebenenfalls substituierte Phenoxygruppe oder eine Alkoxygruppe, vor allem eine Niederalkoxygruppe, speziell Methoxy oder Äthoxy.
Eine verätherte Mercaptogruppe ist z.B. eine gegebenenfalls substituierte Phenylmercapto- oder Benzylmercaptogruppe oder insbesondere eine Niederalkylmercaptogruppe, wie die Äthyl- oder Methylmercaptogruppe.
Eine Ammoniumgruppe ist insbesondere eine quaternäre Ammoniumgruppe, vor allem eine Tri-niederalkylammonium- gruppe, z.B. die Trimethyl- oder Triäthyl-ammoniumgruppe oder das Kation einer aromatischen Stickstoffbase, z.B. die Pyridinium- oder Chinoliniumgruppe.
Eine reaktionsfähige veresterte Hydroxylgruppe ist insbesondere eine durch eine starke anorganische oder organische Säure, vor allem eine Halogenwasserstoffsäure, wie Chlorwasserstoffsäure, Bromwasserstoffsäure oder Jodwasserstoffsäure, oder eine organische Sulfonsäure, insbesondere eine aromatische Sulfonsäure, z.B. Benzolsulfonsäure, p-Brombenzolsulfonsäure oder p-Toluolsulfonsäure, oder eine Alkan-, vor allem Niederalkansulfonsäure, z.B. Methansulfonsaure, Äthansulfonsäure oder eine olefinische Sulfonsäure, z.B. Äthansulfonsäure, veresterte Hydroxylgruppe.
Eine Sulfonylgruppe ist insbesondere eine von einer organischen Sulfonsäure, insbesondere von einer aromatischen Sulfonsäure, z.B. Benzolsulfonsäure, p-Brombenzolsulfonsäure oder p-Toluolsulfonsäure, einer Alkan-, vor allem Niederalkansulfonsäure, z.B. Methansulfonsäure oder Äthansulfonsäure, oder einer olefinischen Sulfonsäure, z.B. Äthan- sulfonsäure abgeleitete Sulfonylgruppe.
Die Umsetzung kann in üblicher Weise durchgeführt werden. So arbeitet man bei der Umsetzung einer Verbindung der Formel II, worin X ein austauschbarer Rest, z.B. einer der genannten austauschbaren Reste ist, mit einer Verbindung der Formel III, bzw. IIIa, worin Y Waserstoff ist, vorzugsweise in Gegenwart eines basischen Kondensationsmittels oder man setzt die Verbindung der Formel III bzw. IIIa in Form eines Salzes, z.B. eines Metallsalzes um, wie eines Alkalioder Erdalkalimetallsalzes, welches z.B. aus einer Verbindung der Formel III bzw. IIIa und einer starken Base, z.B.
einem Amid, einer Kohlenwasserstoffverbindung, einem Alkoholat, dem Hydroxid oder insbesondere dem Hydrid eines Metalles, z.B. eines Alkalimetalles, wie des Lithiums, des Kaliums oder vor allem des Natriums, oder eines Erdalkalimetalles, wie des Magnesiums oder Calciums, oder diesem selbst erhältlich ist und ohne Isolierung verwendet werden kann. Basische Kondensationsmittel sind z.B. Alkali- oder Erdalkalihydroxide, wie Natrium-, Kalium- und Calciumhydroxid oder organische tertiäre Stickstoffbasen, wie Trialkylamine, z.B. Triäthyl- oder Trimethylamin, oder aromatische Stickstoffbasen, z.B. Prridin oder Chinolin. Man kann ferner auch einen Überschuss der Verbindung der Formel III bzw.
Illa verwenden, insbesondere, wenn R5 nicht Wasserstoff ist. Vorteilhaft arbeitet man bei erhöhter Temperatur und/ oder in Gegenwart eines inerten Lösungsmittels, insbesondere eines inerten polaren Lösunwnittels, z*B. von Acetonitril, Dimethylsulfoxid, Tetramethylharnstoff, eines höhersiedenden Äthers, z.B. des Dioxans, Diphenyläthers, Diisopropyl äthers, eines Äthers des Äthylenglykols oder von Tetrahydrofuran, von Wasser, eines höheren Alkohols, einer der genannten Stickstoffbasen oder insbesondere von Dimethylformamid.
Geht man von Verbindungen der Formel III bzw. lila aus, worin Y und R5 Wasserstoff bedeuten, können bei geeigneter Wahl der Reaktionsbedingungen und Verwendung doppelt molarer Mengen einer Verbindung der Formel II sowohl l-Mono-(4-oder 5-nitro-2-imidazolyl)-4-imidazolin-2- -one oder -thione als auch 1 ,3-Bis-( oder 5-nitro-2-imidazolyl)-4-imidazolin-2-one oder -thione der Formel I erhalten werden.
In erhaltene Verbindungen kann man im Rahmen der Definition der Endstoffe Substituenten einführen, abwandeln oder abspalten.
So kann man erhaltene Verbindungen, die mindestens ein substituierbares an ein aromatisches Kohlenstoffatom gebundenes Wasserstoffatom enthalten, in an sich bekannter Weise substituieren, beispielsweise in der beschriebenen Weise nitrieren.
Bei der Nitrierung von Imidazolen der Formel IV

worin R3, R4, R5, Rss und R7 die angegebenen Bedeutungen haben und mindestens einer der Reste R'1 und R'2 Wasserstoff ist und der andere auch Niederalkyl sein kann, kann je nach den Verfahrensbedingungen und Ausgangsstoffen neben der Nitrogruppe in 4- oder 5-Stellung des Imidazolringes auch eine oder mehr als eine Nitrogruppe in den 2ständigen Rest eingeführt werden. So kann z.B. die Nitrierung eines 1 ,3-Bis-(2-imidazolyl)-4-imidazolin-2-ons oder -thions selektiv erfolgen. Je nach den Reaktionsbedingungen und der Stärke des Nitrierungsmittels können beide Imid azolringe oder nur ein Imidazolring in 4- und/oder 5-Stellung nitriert werden.
Weiterhin kann man erhaltene Verbindungen, die mindestens ein substituierbares, an ein aromatisches Kohlenstoffatom gebundenes Wasserstoffatom besitzen, in an sich bekannter Weise durch Umsetzung mit einem elementaren Halogen, z.B. mit Chlor oder Brom, halogenieren. Dabei arbeitet man unter den üblichen Bedingungen, vorzugsweise in einem inerten Lösungsmittel, wie einem halogenierten Kohlenwasserstoff, z.B. Tetrachlormethan, oder eines Äthers vorteilhaft bei Anwesenheit eines Katalysators, wie einer Lewis-Säure, z.B. eines Halogenides des Bors oder Aluminiums oder eines übergangsmetalles, z.B. des Eisens, Kupfers, Zinks oder bei Anwesenheit von fein verteilten katalytisch wirkenden Metallen, z.B. eines der genannten Metalle, speziell von Eisen.
Weiterhin kann man die genannten Verbindungen in an sich bekannter Weise alkylieren zum Beispiel indem man sie mit einem reaktionsfähigen Derivat eines Kohlenwasserstoffes, z.B. eines in genannter Weise reaktionsfähig veresterten Alkohols, vor allem eines Alkanols, umsetzt. Dabei arbeitet man vorteilhaft in einem inerten Lösungsmittel, wie einem halogenierten Kohlenwasserstoff, zum Beispiel Tetrachlormethan oder Trichloräthan, einem geeigneten Kohlenwasserstoff, wie einem Alkan, oder einem geeigneten Äther, wie Diisopropyläther, bei erhöhter Temperatur und vorteilhaft bei Anwesenheit einer hierfür üblichen Lewissäure, wie einer der genannten Lewissäuren, eines Halogenides, insbesondere des Chlorides des Aluminiums, des Zinns oder des Zinks.
Erhaltene Verbindungen, worin mindestens einer der Substituenten R3 und R5 Wasserstoff ist, kann man in an sich bekannter Weise durch einen Substituenten R3, und/oder R5 substituieren, beispielsweise zur Herstellung von Verbindungen in denen der Substituent R3 und/oder R5 eine Alkyl-, Aralkyl- oder Arylgruppe ist, worin gegebenenfalls vorhandene Heteroatome durch mindestens 2 Kohlenstoffatome vom Ringstickstoffatom getrennt sind, durch Reaktion mit reaktionsfähigen Estern von Alkoholen der Formeln R3-OH und/ oder Rb-OH alkylieren. Reaktionsfähige Ester sind dabei solche mit starken anorganischen Säuren oder organischen Sulfonsäuren, vor allem mit Halogenwasserstoffsäuren, z.B.
Chlor-, Brom- oder Jodwasserstoffsäure, oder Schwefelsäure, oder Aryl- oder Alkansulfonsäuren, vor allem Phenyl, wie Toluolsulfonsäuren. Dabei arbeitet man in an sich bekannter Weise, zB. indem man die zu alkylierende Verbindung in Form eines Metall-, wie AUralimetallsalzes umsetzt, oder in Gegenwart eines basischen Kondensationsmittels, besonders eines Metallsalze bildenden Kondensationsmittels, wie eines Amides, des Hydrides, einer Kohlenwasserstoffverbindung, des Hydroxides, eines Alkoholats oder Carbonates eines Alkalimetalles.
Die Einführung eines Restes R3 und/oder R5 der genannten Art kann aber auch gegebenenfalls durch Behandlung mit einer Diazoverbindung der Formel R',tN3N erfolgen, worin RXo bis auf die Doppelbindung zum Stickstoff hin dem Alkoholrest R3 oder R4 entspricht.
Verbindungen, in denen der Rest R3 und/oder R5 ein eine Hydroxylgruppe oder eine freie oder substituierte Aminogruppe tragender Methylrest, besonders ein Hydroxymethyloder sek.- oder tert.Aminomethylrest ist, werden durch Reaktion mit Formaldehyd gegebenenfalls in Gegenwart von Ammoniak oder Aminen erhalten.
Die Einführung der Hydroxymethylgruppe geschieht durch einfache Reaktion mit Formaldehyd, gegebenenfalls in Form eines Formaldehyd-Donators, wie 1,3,5-Trioxan oder Paraformaldehyd, vorteilhaft in Gegenwart eines basischen Kondensationsmittels, wie eines Alkalihydroxyds oder -carbonats, oder tertiärer Amine oder quaternärer Ammoniumhydroxyde, wie Triäthylamin oder Benzyltrimethylammoniumhydroxyd.
Die Einführung der Aminomethylgruppe erfolgt zweckmässig nach Art einer Mannich-Reaktion, z.B. mit Formaldehyd, unter Verwendung eines Salzes des Ammoniaks oder Amins. Der Formaldehyd kann auch hier in Form eines Donators, wie 1,3,5-Trioxan oder Paraformaldehyd, gegebenenfalls in Gegenwart einer Säure, verwendet werden.
Soll ein 2-Hydroxyniederalkylrest, z.B. der 2-Hydroxy äthyl oder 2-Hydroxypropylrest, eingeführt werden, so kann man die erhaltene Verbindung mit einem 1,2-Epoxyalkan, z.B. mit Äthylenoxid oder Propylenoxid, umsetzen.
Bei der N-Substitution von Verbindungen der Formel I, worin R5, Wasserstoff ist, ist indes zu beachten, dass aufgrund der bekannten Tautomerie von 4(5)-substituierten Imidazolen je nach den angewandten Mitteln und Reaktionsbedingungen wahlweise 4-Nitro- oder 5-Nitro-2-imidazolyl-verbindungen der Formel I oder Gemische beider Isomeren erhalten werden können. So kann man beispielsweise bei der Umsetzung eines 1- unsubstituierten 4(5)-Nitroimidazols der Formel I mit einem reaktionsfähigen Ester eines Alkohols R3-OH, z.B. mit einem Alkylhalogenid, Alkylsulfat oder Sulfonsäurealkylester, in Gegenwart eines basischen Kondensationsmittels, wie eines Amides, des Hydrides, des Hydroxides, eines Alkoholates oder einer Kohlenwasserstoffverbindung eines Alkalimetallels, überwiegend das 4-Nitro-isomere erhalten.
Andererseits erhält man beispielsweise bei Durchführung der Alkylierung unter neutralen oder sauren Bedingungen, wie bei der Umsetzung eines in 1-Stellung unsubstituierten 4(5)-Nitro-imidazols mit einem 1,2-Epoxyalkan bei Anwesenheit einer starken Säure, z.B. einer niederen aliphatischen Carbonsäure, überwiegend das 5-Nitro-isomere.
Die genannten Verbindungen, die mindestens ein substituierbares Stickstoffatom enthalten, kann man weiterhin in üblicher Weise acylieren, z.B. durch Umsetzung mit einem üblichen Acylierungsmittel, wie einem reaktionsfähigen Derivat einer Carbonsäure, insbesondere einer gegebenenfalls substituierten Benzoesäure oder einer Niederalkancarbonsäure, z.B. der Essigsäure, oder einer organischen Sulfonsäure, insbesondere einer der genannten Sulfonsäuren. Zur Acylierung geeignet ist insbesondere das Anhydrid oder das Keton einer der genannten Säuren oder das gemischte Anhydrid derselben mit einer starken anorganischen Säure, wie einer Halogen-, insbesondere Chlor- oder Bromwasserstoffsäure, oder einer organischen Säure, oder ein aktiviertes Amid oder ein aktivierter Ester einer der genannten Säuren.
Aktivierte Ester sind z;B. Ester mit elektronenanziehenden Strukturen, wie Ester von Phenol, Thiophenol, p-Nitrophenol, Cyanmethylalkohol und ähnlichen. Aktivierte Amide sind z.B. die N-Acylderivate von Pyrazolen, wie 3,5-Dimethylpyrazol oder Imidazolen, wie Imidazol selbst. Je nach der Natur der Acylierungskomponente kann die Verwendung eines Kondensationsmittels zweckmässig sein. So begünstigen disubstituierte Carbodiimide die Reaktion der Säuren, Basen, wie tertiäre Amine, z.B. Triniederalkylamino, N,N-Diniederalkylaniline oder aromatische tertiäre Stickstoffbasen wie Pyridin oder Chinolin, oder anorganische Basen, wie Alkalioder Erdalkalihydroxide, -carbonate oder -bicarbonate, z.B.
Natrium-, Kalium- oder Calciumhydroxid oder Natrium-, Kalium- oder Calcium-(bi)carbonat, oder Acylationen die Reaktion von Säureanhydriden und Säurehalogeniden.
Weiterhin kann man eine Verbindung der Formel I, worin R5 Wasserstoff ist, durch Umsetzung mit einer Verbindung der Formel II in ein symmetrisches oder, sofern erwünscht, unsymmetrisches 1,3-Bis-(4- oder 5-nitro-2-imidazolyl)-4-imidazolin-2-on oder -thion überführen.
Weiterhin kann man in erhaltenen Verbindungen Substituenten abwandeln. So kann man erhaltene Hydroxylverbindungen durch Umsetzung mit einem reaktionsfähigen Derivat eines Kohlenwasserstoffes, wie einem in genannter Weise reaktionsfähig veresterten Alkohol, vor allem Alkanol, in Alkoxyverbindungen überführen. Dabei arbeitet man in üblicher Weise, z.B. wie für die Alkylierung von Verbindungen mit substituierbaren Stickstoffatomen angegeben. Für die O-Alkylierung von erhaltenen Verbindungen mit enolisierbaren Oxogruppen sind insbesondere auch Ketale, wie 2,2 Dimethoxypropan oder Orthoester, z.B. Orthoameisensäureester geeignet.
Ferner kann man die genannten Verbindungen auch durch Umsetzung mit einem üblichen Halogenierungsmittel, wie einem der genannten, z.B. einer Halogenwasserstoffsäure, vor allem Chlor- oder Bromwasserstoffsäure, oder einem Halogenid, vor allem einem Chlorid oder Bromid der Schwefelsäure, schwefligen Säure, der Phosphorsäure oder der phosphorigen Säure, in Halogen-, insbesondere in Chloroder Bromverbindungen umwandeln.
In erhaltenen Verbindungen, die mindestens eine Halogenalkyl-, insbesondere Halogenniederalkylgruppe enthalten, kann man diese ferner in üblicher Weise, z.B. durch Umsetzung mit einem Amin, vorzugsweise in einem inerten Lösungsmittel, wie einem Lösungsmittel mit polaren funktionellen Gruppen, z.B. einem Äther, vor allem einem Diniederalkyläther oder einem cyclischen Äther, einem Alkohol, vor allem einem Niederalkanol, einem N,N-Dialkyl- vor allem N,N-Dimethyl-carbonsäureamid, vor allem Dimethylformamid oder Dimethylacetamid, einem Tetraalkylharnstoff, vor allem Tetramethylharnstoff, Hexamethylphosphosäuretriamid oder Dimethylsulfoxid, vorteilhaft bei Anwesenheit eines basischen Kondensationsmittels, wie eines der genannten, oder mit einem Metallsalz, insbesondere einem Alkalimetallsalz, wie dem Natriumsalz oder Kaliumsalz, eines Amins in eine Aminoalkylgruppe umwandeln.
Ferner kann man Halogenalkylgruppen in üblicher Weise in eine Mercaptoalkylgruppe überführen, z.B. durch Umsetzung mit Schwefelwasserstoff oder insbesondere mit einem geeigneten Derivat davon, wie Thioharnstoff, oder speziell einem Salz einer Thiocarbonsäure, wie einem Alkali-Thiolacetat, und gegebenenfalls Freisetzung der Mercaptogruppe nach üblichen Methoden, z. B.
durch Hydrolyse eines primär erhaltenen Isothiuroniumsalzes oder durch Umesterung eines primär erhaltenen Thiolesters, insbesondere mit einem Niederalkanol bei Anwesenheit eines Alkalimetall-niederalkanolates. In ähnlicher, an sich bekannter Weise kann man eine Halogenalkylverbindung, z.B.
durch Umsetzung mit einem Metallmercaptid, vor allem einem Alkalimercaptid, das auch unter den Reaktionsbedin gungen aus einem Mercaptan, insbesondere einem Niederalkylmercaptan und einer starken Base, wie einem Alkalimetallhydroxid oder- alkoholat in situ erhalten und ohne Isolierung eingesetzt werden kann, in eine Alkylmercaptoalkylverbindungen umwandeln.
In erhaltenen Verbindungen, in denen R4 Thioxo ist, kann man R4 in üblicher Weise in die Oxogruppe umwandeln, insbesondere nach an sich bekannten Hydrolysemethoden, z. B.
durch Behandeln der erhaltenen Thioxo-Verbindung mit einem alkalischen Mittel, wie einem Alkalihydroxyd, in Gegenwart eines Oxydationsmittels, wie Wasserstoffperoxyd.
In erhaltenen Verbindungen, in denen R4 Oxo ist, kann man R4 in üblicher Weise in die Thioxogruppe umwandeln, insbesondere durch Behandeln der erhaltenen Oxo-Verbindung mit geeigneten Sulfiden, wie Phosphorpentasulfid, Alu miniumsulfid, Siliciumdisulfid oder Borsulfid.
Ferner kann man erhaltene Endstoffe durch Abspaltung von Substituenten in andere Endstoffe überführen. So kann man in erhaltenen Verbindunge, die mindestens eine an ein Stickstoffatom gebundene Acylgruppe tragen, diese in üblicher Weise, z.B. durch Hydrolyse, vorzugsweise in einem wasserhaltigen Lösungsmittel, wie einem der genannten, bei Anwesenheit einer starken Säure, insbesondere einer starken anorganischen Säure, wie einer der genannten, und zweckmässig bei erhöter Temperatur, durch Wasserstoff ersetzen.
Die nachträglichen Umwandlungen können einzeln oder in Kombination und in beliebiger Reihenfolge vorgenommen werden. Bei den einzelnen Operationen ist darauf zu achten, dass andere funktionelle Gruppen nicht angegriffen werden.
Je nach den Verfahrensbedingungen und Ausgangsstoffen erhält man die Endstoffe in freier Form oder in der ebenfalls in der Erfindung inbegriffenen Form ihrer Säureadditionssalze. So können beispielsweise basische, neutrale oder gemischte Salze, gegebenenfalls auch Hemi-, Mono-, Sesquioder Polyhydrate davon erhalten werden. Die Säureadditionssalze der neuen Verbindungen können in an sich bekannter Weise in die freie Verbindung übergeführt werden, z.B. mit basischen Mitteln, wie Alkalien oder Ionenaustauschern. Anderseits können die erhaltenen freien Basen mit organischen oder anorganischen Säuren Salze bilden. Zur Herstellung von Säureadditionssalzen werden insbesondere solche Säuren verwendet, die zur Bildung von therapeutisch verwendbaren Salzen geeignet sind.
Als solche Säuren seien beispielsweise genannt: Halogenwasserstoffsäuren, Schwefelsäuren, Phosphorsäuren, Salpetersäure, Perchlorsäure, aliphatische, alicyclische, aromatische oder heterocyclische Carbonoder Sulfonsäuren, wie Ameisen-, Essig-, Propion-, Bernstein-, Glykol-, Milch-, Äpfel-, Wein-, Zitronen-, Ascorbin-, Malein-, Hydroxymalein- oder Brenztraubensäure; Phenylessig-, Benzoe-, p-Aminobenzoe-, Anthranil-, p-Hydroxybenzoe-, Salicyl- oder p-Aminosalicylsäure, Embonsäure, Methansulfon-, Äthansulfon-, Hydroxyäthansulfon-, Äthylensulfonsäure; Halogenbenzolsulfon-, Toluolsulfon-, Naphthalinsulfonsäure oder Sulfanilsäure; Methionin, Trypthophan, Lysin oder Arginin.
Diese oder andere Salze der neuen Verbindungen, wie z.B.
die Pikrate, können auch zur Reinigung der erhaltenen freien Basen dienen, indem man die freien Basen in Salze überführt, diese abtrennt und aus den Salzen wiederum die Basen freimacht. Infolge der engen Beziehungen zwischen den neuen Verbindungen in freier Form und in Form ihrer Salze sind im Vorausgegangenen und nachfolgend unter den freien Verbindungen sinn- und zweckmässig, gegebenenfalls auch die entsprechenden Salze zu verstehen.
Die Erfindung betrifft auch diejenigen Ausführungsformen eines Verfahrens, bei denen man einen Ausgangsstoff in Form eines Salzes und/oder Racemates bzw. Antipoden oder insbesondere eines unter den Reaktionsbedingungen erhältlichen rohen Reaktionsgemisches einsetzt.
Je nach der Zahl der asymmetrischen C-Atome und der Wahl der Ausgangsstoffe und Arbeitsweisen können die neuen Verbindungen als Racematgemische, als Racemate oder als optische Antipoden vorliegen.
Racematgemische können aufgrund der physikalischchemischen Unterschiede der Bestandteile in bekannter Weise in die reinen Racemate aufgetrennt werden, z.B. durch Chromatographie und/oder fraktionierte Kristallisation.
Die Auftrennung von erhaltenen Racematen in die optischen Antipoden kann in an sich bekannter Weise durchgeführt werden. Erhaltene Racemate können z.B. in Ester von optisch aktiven Säuren oder vorzugsweise in Salze mit optisch aktiven Säuren überführt werden. Besonders gebräuchliche optisch aktive Säuren sind z.B. die D- und L-Formen von Weinsäure, Di-o-Toluylweinsäure, Diacetyl-Weinsäure, Äpfelsäure, Mandelsäure, Camphersäure, Camphersulfonsäure, Bromcamphersulfonsäure und Chinasäure.
Die erhaltenen Gemische von diastereoisomeren Salzen werden aufgrund von physikalisch-chemischen Unterschieden, z.B. der Löslichkeit, der Kristallisationsfähigkeit usw., in die einzelnen Salze aufgetrennt und die optisch aktiven Antipoden aus den Salzen freigesetzt. Ferner kann man ein erhaltenes Racemat in Salzform mit einem optisch aktiven Metall Komplexsalz oder ein erhaltenes Racemat in freier Form mit einem optisch aktiven Metall-Komplex-Hydroxyd umsetzten und das geringer lösliche Produkt abtrennen und die optisch reine Verbindung freisetzen. Geeignete optisch aktive Metallkomplexe sind z.B. optisch aktive Kobaltnitrat-Komplexverbindungen.
Man kann ferner erhaltene Racemate auch durch fraktioniertes Kristallisieren, gegebenenfalls aus einem optisch aktiven Lösungsmittel, oder durch Chromatographie, insbesondere Dünnschichtchromatographie, an einem optisch aktiven Trägermaterial oder mit Hilfe von Mikroorganismen in die optisch aktiven Antipoden auftrennen. Gemische von diastereoisomeren Verbindungen werden in üblicher Weise aufgrund ihrer physikalisch-chemischen Unterschiede, wie solchen der Löslichkeit, der Siedepunkte usw., z.B. durch fraktioniertes Kristallisieren oder Destillieren, in die reinen isomeren Verbindungen aufgetrennt. Dabei isoliert man vorteilhafterweise das pharmakologisch wirksamere reine Isomere, insbesondere den wirksameren bzw. weniger toxischen Antipoden.
Erfindungsgemäss kann man aber auch die Endprodukte in Form der reinen Racemate bzw. optischen Antipoden erhalten, indem man ein oder mehrere asymmetrische C-Atome enthaltende Ausgangsstoffe in Form der reinen Racemate bzw. optischen Antipoden einsetzt.
Die Ausgangsstoffe sind bekannt oder können, sofern sie neu sind, nach an sich bekannten Methoden erhalten werden.
Zweckmässig verwendet man für die Durchführung der erfindungsgemässen Reaktionen solche Ausgangsstoffe, die zu den eingangs besonders erwähnten Gruppen von Endstoffen und besonders zu den speziell beschriebenen und hervorgehobenen Endstoffen führen.
Die neuen Verbindungen können z.B. in Form pharmazeutischer Präparate Verwendung finden, welche sie in freier Form oder gegebenenfalls in Form ihrer Salze, besonders der therapeutisch verwendbaren Salze, in Mischung mit einem z.B. für die enterale oder parenterale Applikation geeigneten pharmazeutischen organischen oder anorganischen, festen oder flüssigen Trägermaterial enthalten. Für die Bildung desselben kommen solche Stoffe in Frage, die mit den neuen Verbindungen nicht reagieren, wie z.B. Wasser, Gelatine, Lactose, Stärke, Stearylalkohol, Magnesiumstearat, Talk, pflanzliche öle, Benzylalkohole, Gummi, Propylenglykole, Vaseline oder andere bekannte Arzneimittelträger. Die pharmazeutischen Präparate können z.B. als Tabletten, Dragees, Kapseln, Suppositorien oder in flüssiger Form als Lösungen (z.B. als Elixier oder Sirup), Suspensionen oder Emulsionen vorliegen.
Gegebenenfalls sind sie sterilisiert und/oder enthalten Hilfsstoffe, wie Konservierungs-, Stabilisierungs-, Netzoder Emulgiermittel, Lösungsvermittler oder Salze zur Ver änderung des osmotischen Druckes oder Puffer. Sie können auch andere therapeutisch wertvolle Substanzen enthalten. Die pharmazeutischen Präparate werden nach üblichen Methoden gewonnen. Die Dosierung der neuen Verbindungen kann je nach der Verbindung und den individuellen Bedürfnissen des Patienten variieren. Die übliche Tagesdosis für einen ca. 75 kg schweren Warmblüter liegt zwischen etwa 0,25 und 1,0 g.
Die neuen Verbindungen können auch in der Tiermedizin, z.B. in einer der oben genannten Formen oder in Form von Futtermitteln oder von Zusatzmitteln für Tierfutter verwendet werden. Dabei werden z.B. die üblichen Streck- und Verdünnungsmittel bzw. Futtermittel angewendet.
Die Erfindung wird in dem folgenden Beispiel näher beschrieben. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel
In eine Suspension von 4,8 g 50%igem Natriumhydrid in 100 ml Dimethylformamid tropft man innerhalb von 15 Minuten unter Rühren bei 20 bis 30 eine Lösung von 11,4 g 2-Mercapto-l-methyl-imidazol in 60 ml Dimethylformamid.
Die erhaltene Lösung wird dann innerhalb von 30 Minuten unter Rühren bei 20 bis 300 in eine Lösung von 20,5 g l-Me- thyl-2-methylsulfonyl-5-nitro-imidazol in 80 ml Dimethylformamid eingetropft. Anschliessend lässt man noch eine Stunde bei 1000 rühren und dampft das Reaktionsgemisch ein.
Der Eindampfrückstand wird mit 200 ml Methylenchlorid und 200 ml Wasser ausgeschüttelt, der Methylenchloridextrakt abgetrennt, mit 50 ml Wasser gewaschen, über wasserfreiem Magnesiumsulfat getrocknet und eingedampft. Der Eindampfrückstand ergibt nach zweimaligem Umkristallisieren aus Essigester reines l-Methyl-2[(1 -methyl-5-nitro-2-imidazolyl)- -mercapto]-imidazol vom Smp. 123-124".
Die vereinigten Mutterlaugen der Kristallisation werden eingedampft und an einer mit 550 g Kieselgel gefüllten Chromatographiesäule von 50 mm Durchmesser getrennt. Dabei eluiert man mit Methylenchlorid und fängt Fraktionen von je 600 ml auf. Die 17. bis 24. Fraktion werden vereinigt, ein gedampft und aus 85 ml abs. Äthanol umkristallisiert. Man erhält so reines l-Methyl-3-(1-methyl-5-nitro-2-imidazolyl)- -4-imidazolin-2-thion der Formel
