Gegenstand der vorliegenden Anmeldung ist ein Verfahren zur Auftrennung von racemischen Phenylpropanolaminen der Formel I (siehe Formelblatt), worin R1 für einen mit einem oder zwei Chloratomen substituierten Phenylrest steht, dadurch gekennzeichnet, dass man racemische Phenylpropanolamine der Formel I in an sich bekannter Weise auftrennt, z. B. indem man die Racemate mittels optisch aktiven Säuren in das Gemisch ihrer diastereomeren Salze überführt, daraus die optischen Antipoden der racemischen Phenylpropanolamine der Formel I isoliert und gewünschtenfalls in ihre Säureadditionssalze überführt.
Die optischen Antipoden der racemischen Phenylpropanolamine der Formel I zeichnen sich in der pharmakologischen Prüfung durch stark ausgeprägte Effekte aus und können daher als Heilmittel verwendet werden. Sie besitzen eine ausgeprägte anorexigene/antiobese Wirkung, wie sie bei Gewichtsveränderung und am Futterkonsum an der Ratte sowie am Futterkonsum des Totenkopfaffens festgestellt wurde. Für die obengenannte Anwendung wird die Dosierung davon abhängen, welche Verabreichungsform und Behandlung gewünscht wird. Gute Resultate werden jedoch erreicht mit einer täglichen Dosierung von 2,5 bis mehr als 20 mg pro kg Körpergewicht des Testtieres; diese Dosis kann nötigenfalls in 2 bis 3 Anteilen oder auch als Retardform verabreicht werden. Die durchschnittliche Tagesdosis für grössere Säugetiere kann 10-100 mg betragen.
Als Heilmittel können die optischen Antipoden der Verbindungen der Formel I bzw. ihre physiologisch verträglichen Säureadditionssalze allein oder in geeigneten Arzneiformen mit pharmakologisch indifferenten Hilfsstoffen verabreicht werden.
In dem nachfolgenden Beispiel, das die Erfindung näher erläutern, ihren Umfang aber in keiner Weise einschränken soll, erfolgen alle Temperaturangaben in Celsiusgraden.

Beispiel (-) 2-Amino-3-(3 ,4-dichlorophenyl) -propanol und (+) 2-Amino-3 -(3,4-dichlorophenyl)-propanol a) 33 g racemisches 2-Amino-3-(3,4-dichlorophenyl)- propanol löst man in 0,5 1 Methanol und versetzt mit einer Lösung von 57 g (-)-Di-o,o'(p-toluoyl)-L-weinsäure in 1,5 1 Methanol und gibt noch 450 ml Wasser und 250 ml Methanol hinzu. Man lässt stehen und filtriert nach 48 Stunden vom ausgeschiedenen Niederschlag ab. Der Niederschlag wird mit einem Gemisch von 150 ml Methanol und 30 ml Wasser gewaschen, hierauf erneut in 2,2 1 heissem Methanol gelöst, filtriert und 450 ml Wasser zugesetzt. Man engt die Lösung etwas ein, kühlt auf 0 ab und lässt das (-) 2-Amino-3-(3,4dichlorophenyl)-propanol -hydrogen -di-o-(p-toluoyl) -Ltartrat auskristallisieren.
b) 34,2 g des so erhaltenen Salzes der optisch aktiven Base werden hierauf in 200 ml Wasser gelöst. Die Lösung wird mit wässriger Natronlauge auf pH 11 gestellt und 2mal mit Äthylacetat extrahiert. Die Extrakte werden je 2mal mit Wasser und 30%iger Kochsalzlösung gewaschen. Anschliessend wird eingedampft und das zurückbleibende Öl aus Äther/ Petroläther umkristallisiert. Man erhält (-) 2-Amino-3-(3,4dichlorophenyl)-propanol, Smp. 77-79 ; [a]20D = -24,4 (c = 1 in Dimethylformamid).
c) Die unter a) erhaltene methanolische Mutterlauge wird eingedampft. Aus dem Rückstand erhält man das (+) 2 -Amino-3 -(3,4-dichlorophenyl) -propanol-hydrogen-(-) -di o-(p-toluoyl) -L-tartrat.
d) (+) 2-Amino-3 -(3,4-dichlorophenyl) -propanol wird nach dem unter b) beschriebenen Verfahren erhalten, Smp.
70-77 ; [a]20D = +21,4 (c = 1 in Dimethylformamid).
PATENTANSPRUCH
Verfahren zur Auftrennung von racemischen Phenylpropanolaminen der Formel I (siehe Formelblatt), worin R1 für einen mit einem oder zwei Chloratomen substituierten Phenylrest steht, dadurch gekennzeichnet, dass man racemische Phenylpropanolamine der Formel I mittels optisch aktiven Säuren in das Gemisch ihrer diastereomeren Salze überführt, daraus die optischen Antipoden der racemischen Phenylpropanolamine der Formel I isoliert und gewünschtenfalls in ihre Säureadditionssalze überführt.

The present application relates to a process for the resolution of racemic phenylpropanolamines of the formula I (see formula sheet), in which R1 stands for a phenyl radical substituted by one or two chlorine atoms, characterized in that racemic phenylpropanolamines of the formula I are separated in a manner known per se, z. B. by converting the racemates into the mixture of their diastereomeric salts by means of optically active acids, isolating the optical antipodes of the racemic phenylpropanolamines of the formula I therefrom and, if desired, converting them into their acid addition salts.
The optical antipodes of the racemic phenylpropanolamines of the formula I are distinguished in the pharmacological test by strongly pronounced effects and can therefore be used as medicaments. They have a pronounced anorexigenic / antiobese effect, as was found in the weight change and in the food consumption of rats and in the food consumption of the squirrel monkey. For the above application, the dosage will depend on the form of administration and treatment desired. However, good results are achieved with a daily dosage of 2.5 to more than 20 mg per kg of body weight of the test animal; if necessary, this dose can be administered in 2 to 3 portions or as a sustained-release form. The average daily dose for larger mammals can be 10-100 mg.
The optical antipodes of the compounds of the formula I or their physiologically tolerable acid addition salts can be administered as remedies alone or in suitable medicinal forms with pharmacologically inert auxiliaries.
In the following example, which explains the invention in more detail but is not intended to limit its scope in any way, all temperatures are given in degrees Celsius.

Example (-) 2-Amino-3- (3, 4-dichlorophenyl) -propanol and (+) 2-Amino-3 - (3,4-dichlorophenyl) -propanol a) 33 g of racemic 2-amino-3- ( 3,4-dichlorophenyl) - propanol is dissolved in 0.5 1 of methanol and treated with a solution of 57 g of (-) - di-o, o '(p-toluoyl) -L-tartaric acid in 1.5 1 of methanol and add 450 ml of water and 250 ml of methanol. The solution is adjusted to pH 11 with aqueous sodium hydroxide solution and extracted twice with ethyl acetate. The extracts are washed twice with water and 30% sodium chloride solution. It is then evaporated and the remaining oil is recrystallized from ether / petroleum ether. (-) 2-Amino-3- (3,4dichlorophenyl) propanol, mp 77-79; [a] 20D = -24.4 (c = 1 in dimethylformamide).
c) The methanolic mother liquor obtained under a) is evaporated. The (+) 2-amino-3 - (3,4-dichlorophenyl) -propanol-hydrogen - (-) -di-o- (p-toluoyl) -L-tartrate is obtained from the residue.
d) (+) 2-Amino-3 - (3,4-dichlorophenyl) propanol is obtained by the process described under b), m.p.
