Die Erfindung ist ein neues Verfahren zur Herstellung von in 7-Stellung durch die Nitrogruppe substituierten 1,4 -Benzodiazepin-2-on-Derivaten.
Es ist bekannt, dass die in 7-Stellung durch Nitrosubstituierten 1 ,4-Benzodiazepin-2-on-Derivate vorzügliche, einen physiologischen Schlaf herbeiführende hypnotische Wirkungen haben. Zur Herstellung dieser Verbindungen sind aus der Literatur mehrere Methoden bekannt. So wird in der britischen Patentschrift 972967 die Kondensation von 2-Amino-5-nitrobenzophenon mit einem Glycinester beschrieben, während in der deutschen Patentschrift 1136 709 der Ringschluss von 2-Bromacetamido-5-nitro-benzophenon mit Ammoniak beschrieben wurde.
Es ist ferner zur Herstellung von in 7-Stellung durch Nitro substituierten 1 ,4Benzodiazepin-2-on-Denvaten auch eine Methode bekannt, nach welcher die Nitrogruppe durch das nachträgliche Nitrieren des Benzodiazepinrings eingeführt wird (britische Patentschrift 972965 bzw. deutsche Patentschrift 1 288 609); in der ungarischen Patentschrift 155 373 wurde die Oxidation der entsprechenden l,3,4,5-Tetrahydro-1,4- -benzodiazepin-2-on-Derivaten beschrieben.
Die Herstellung von in 7-Stellung durch Nitrosubstituierten 1 ,4-Benzodiazepinderivaten bereitet mehr Schwierigkeiten, als die Synthese von in der 7-Stellung andere Substituenten enthaltenden Verbindungen: so kann z.B. nach der in der britischen Patentschrift 972967 beschriebenen Methode die 7-Nitro-Verbindung nur mit sehr schwachen Ausbeuten hergestellt werden. Zur Kondensation von 2-Glycylamido-5 -nitro-benzophenon muss hier der Ausgangstoff auf Temperaturen von 165 bis 187"C erwärmt werden (vgl. deutsche Patentschrift 1 136 709), was ernsthafte technologische Schwierigkeiten bereitet, und die Ausbeuten sind wegen der Wärmeempfindlichkeit des Produkts nicht befriedigend.
Bei dem nachträglichen Nitrieren des Benzodiazepinrings (britische Patentschrift 972965 bzw. deutsche Patentschrift 1 288 609) treten unerwünschte Nebenreaktionen ein, da die Nitrierung nicht eindeutig in der 7-Stellung erfolgt.
Es wurde nun gefunden, dass die in 7-Stellung durch Nitrosubstituierten 1,SBenzodiazepin-2-on-Derivate der Formel I

Die Überführung der 1,3A,5-Tetra4ydrobenzodiazepin- derivate der Formel IV in die entsprechenden 1,3-Dihydro -benzodiazepinderivate der Formel V erfolgt auf die in der ungarischen Patentschrift 155 373 beschriebene Weise, und zwar derart, dass man das Tetrahydrobenzodiazepinderivat mit Brom behandelt und dann aus dem erhaltenen bromierten Zwischenprodukt durch alkalische Behandlung Bromwasserstoff abspaltet.
In den erfindungsgemäss herstellbaren Verbindungen der Formel I bedeutet R vorzugsweise ein Wasserstoffatom, kann aber auch eine Methylgruppe oder eine andere Alkylgruppe bis zu 5C-Atomen bedeuten.
Das Entfernen der Carbobenzoxygruppe Z kann mit Hilfe von in Eisessig gelöster Bromwasserstoffsäure in sehr einfacher Weise, mit praktisch quantitativer Ausbeute durchgeführt werden. Als letzter Schritt wird das Tetrahydrobenzodiazepinderivat mit Brom behandelt und aus dem Produkt wird in alkalischem Medium Bromwasserstoff abgespalten.
Sämtliche Schritte des erfindungsgemässen Verfahrens sind technologisch leicht durchführbar; weitere Vorteile des Verfahrens sind, dass keine erhöhten Temperaturen erforderlich sind und dass die Produkte mit ausgezeichneten Ausbeuten erhalten werden.
Das erfindungsgemässe Verfahren wird durch das nachstehende Beispiel näher veranschaulicht. Die im Beispiel angegebenen Schmelzpunkte wurden mit einem Dr. Tottoli'schen Apparat ermittelt. Die dünnschichtchromatograph i- schen Untersuchungen wurden auf Kieselgel nach Stahl durchgeführt, die Entwicklung erfolgte mittels Chlor und Tolidin. Die Struktur der Verbindungen wurde auch durch infrarot-spektroskopischen Untersuchungen kontrolliert.
Beispiel a) 1,3 ,4,5-Tetrahydro-2H-5-phenyl-4-benzyloxycarbo- nyl-7-amino- 1 ,4-benzodiazepin-2-on.
Die Lösung von 22,0 g (0,087 Mol) 1,3,4,5-Tetrahydro -5-phenyl-7-amino-2H- 1 ,4-benzodiazepin-2-on in 750 ml Chloroform wird mit der Lösung von 3,6 g Natriumshy- droxyd in 90 ml Wasser versetzt, dann wird unter lebhaftem Rühren und Kühlen die Lösung von 16,5 g Benzyloxycarbonylchlorid in 90 ml Chloroform tropfenweise zugesetzt.
Das Reaktionsgemisch wird 5 Stunden zwischen -2"C und -4"C, dann 2 Stunden bei 0 C gerührt. Die Chloroform-Phase wird dann abgetrennt, zuerst zweimal mit je 30 ml einer 0,4%igen Salzsäurelösung, dann zweimal mit je 180 ml 2N Salzsäure ausgeschüttelt. Die mit 2N Salzsäure gewonnenen Auszüge werden vereinigt, mit verdünnter Ammoniumhydroxydlösung neutralisiert und das zuerst in öliger Form ausscheidende, dann sich unter Kühlung verfestigende Produkt wird abfiltriert und mit Wasser gewaschen. Es werden auf diese Weise 30,5 g rohes 1,3.4,5- -Tetrahydro-4- carbobenzoxy-5-phenyl-7-amino-2H- l ,4- -benzodiazepin-2-on (90% d.Th.) erhalten.
Für analytische Zwecke wird das Produkt aus Äthanol umkristallisiert: F. 165-1680C; R=0,65 (Chloroform-Methanol 9:1).
Analyse: berechnet: C 71,4% H 5,5% N 10,9% gefunden: C 71,6% H 5,5% N 10,8% b) 1,3 ,4,5-Tetrahydro-5-phenyl-7-nitro-2H- 1 ,4-benzodiazepin-2-on.
2,85 g (0,00735 Mol) 1,3,4,5-Tetrahydro-4-carbobenzoxy-5-phenyl- 7-amino-2H- 1 ,4benzodiazepin-2- on werden im Gemisch von 15 ml 2N Salzsäure und 30 ml Wasser gelöst; das Lösen des Ausgangsstoffs kann durch Zusatz eines Dispergiermittels erleichtert werden. Die Lösung wird auf 0 C abgekühlt und dann mit der Lösung von 0,72 g Natriumnitrit in 3,0 ml Wasser tropfenweise versetzt. Nach Beendigung der Diazotierung wird das Reaktionsgemisch durch die vorsichtige Zugabe von festem Natriumhydrogencarbonat neutralisiert, dann mit 6,0 g festem Natriumhexanitrokobaltat (III) versetzt. Die ausgeschiedene orangefarbige Komplexverbindung wird kalt abfiltriert und dann sofort einer vorher zubereiteten wässrigen Lösung von Natriumnitrit, Kupfer(II)-sulfat und Kupfer(I)-oxyd zugesetzt.
Das Gemisch wird 5 Stunden bei Zimmertemperatur gerührt, dann wird der Niederschlag abfiltriert, mit Wasser gewaschen und mit Dichlormethan extrahiert. Die Dichlormethanlösung wird nach dem Trocknen unter vermindertem Druck zur Trockne verdampft. Es werden 3,0 g rohes 1,3 ,4,5-Tetrahydro-2H-4-carbobenzoxy-5-phenyl-7-nitro- 1,4 -benzodiazepin-2-on erhalten. Das Produkt kann für analytische Zwecke aus Isopropanol umkristallisiert werden; F. 179-181"C.
Das auf obige Weise erhaltene Produkt wird ohne weitere Reinigung decarbobenzoxyliert; zu diesem Zweck wird das rohe Produkt mit 15 ml 5,5 N Bromwasserstofflösung in Eisessig versetzt und unter Luftausschluss bei Zimmertemperatur 30 Minuten gerührt. Dann wird das Reaktionsgemisch mit 150 ml trockenem Äther versetzt, das ausgeschiedene 1,3 ,4,5-Tetrahydro-phenyl-7-nitro-2H- 1 ,4-benzo- diazepin-2-on-hydrobromid wird abfiltriert und mit Äther gewaschen. Es werden 2,5 g rohes Hydrobromid erhalten.
Dieses Produkt kann unmittelbar, ohne weitere Reinigung weiterverarbeitet werden; für analytische Zwecke kann durch wiederholte Verteilung zwischen Dichlormethan und verdünnter Ammoniaklösung, dann zwischen Dichlormethan und verdünnter Salzsäure ein analytisch reines Produkt erhalten werden; F. 232-234"C; R,=0.35 (Hexan-Essigsäure-Chloroform 1:1:8).
c) 1 ,3-Dihydro-5-phenyl-7-nitro-2H- 1 .4-benzodiazepin -2-on.
1,4 g (0,005 Mol) l,3,4,5-Tetrahydro-5-phenyl-7-nitro- -2H-1 ,4-benzodiazepin-2-on werden in 60 ml Chloroform gelöst und die Lösung bei 0 C mit 15 ml molarer Bromlösung in Chloroform tropfenweise versetzt. Das Gemisch wird 10 Minuten gerührt und dann, noch immer bei 0 C.
mit 35 ml 2N Natriumhydroxydlösung tropfenweise versetzt. Das Reaktionsgemisch wird in einen Scheidetrichter übergetragen und mit 80 ml Wasser nachgewaschen. Die wäss rige Phase wird abgetrennt und durch Zugabe von Salzsäure auf pH = 6 gestellt. Der erhaltene Niederschlag wird durch Ausschütteln mit Essigester gelöst. die abgetrennte organische Lösung wird getrocknet und unter vermindertem Druck verdampft. Als Rückstand wird 1,1 g 1,3-Dihydro-5 -phenyl-7-nitro-2H-l ,4-benzodiazepin-2-on (78,5% d.Th.) erhalten. Durch Umkristallisieren aus Äthanol kann ein analytisch reines Produkt erhalten werden: F. 218-220"C; Rf=0,3 (Hexan-Essigester-Chloroform 1:1:8).
The invention is a new process for the preparation of 1,4-benzodiazepin-2-one derivatives substituted in the 7-position by the nitro group.
It is known that the 1,4-benzodiazepin-2-one derivatives substituted by nitro in the 7-position have excellent hypnotic effects that induce physiological sleep. Several methods are known from the literature for the preparation of these compounds. A method is also known for the preparation of 1,4-benzodiazepin-2-one derivatives substituted in the 7-position by nitro, according to which the nitro group is introduced by subsequent nitration of the benzodiazepine ring (British patent 972965 or German patent 1 288 609 ); in the Hungarian patent specification 155 373 the oxidation of the corresponding 1,3,4,5-tetrahydro-1,4- benzodiazepin-2-one derivatives was described.
The preparation of 1,4-benzodiazepine derivatives which are substituted in the 7-position by nitro poses more difficulties than the synthesis of compounds which contain other substituents in the 7-position: e.g. according to the method described in British patent specification 972967, the 7-nitro compound can only be prepared with very poor yields. For the condensation of 2-glycylamido-5-nitro-benzophenone, the starting material must be heated to temperatures of 165 to 187 "C (cf. German Patent 1,136,709), which causes serious technological difficulties, and the yields are because of the heat sensitivity of the Product not satisfactory.
In the subsequent nitration of the benzodiazepine ring (British patent specification 972965 or German patent specification 1,288,609), undesirable side reactions occur, since the nitration does not clearly take place in the 7-position.

The conversion of the 1,3A, 5-tetra4ydrobenzodiazepine derivatives of the formula IV into the corresponding 1,3-dihydrobenzodiazepine derivatives of the formula V is carried out in the manner described in the Hungarian patent specification 155,373, in such a way that the tetrahydrobenzodiazepine derivative is mixed with bromine treated and then split off from the brominated intermediate obtained by alkaline treatment hydrogen bromide.
In the compounds of the formula I which can be prepared according to the invention, R preferably denotes a hydrogen atom, but can also denote a methyl group or another alkyl group with up to 5 carbon atoms.
The removal of the carbobenzoxy group Z can be carried out very easily with the aid of hydrobromic acid dissolved in glacial acetic acid, with practically quantitative yield. As a final step, the tetrahydrobenzodiazepine derivative is treated with bromine and hydrogen bromide is split off from the product in an alkaline medium.
All steps of the process according to the invention can be carried out easily from a technological point of view; further advantages of the process are that no elevated temperatures are required and that the products are obtained in excellent yields.
The method according to the invention is illustrated in more detail by the example below. The melting points given in the example were determined using a Dr. Tottoli's apparatus determined. The thin-layer chromatographic investigations were carried out on silica gel according to Stahl; the development was carried out using chlorine and tolidine. The structure of the compounds was also checked by infrared spectroscopic studies.
Example a) 1,3,4,5-Tetrahydro-2H-5-phenyl-4-benzyloxycarbonyl-7-amino-1,4-benzodiazepin-2-one.

The mixture is stirred for 5 hours at room temperature, then the precipitate is filtered off, washed with water and extracted with dichloromethane. The product obtained in the above manner is decarbobenzoxylated without further purification; for this purpose, the crude product is mixed with 15 ml of 5.5 N hydrogen bromide solution in glacial acetic acid and stirred with exclusion of air at room temperature for 30 minutes. 150 ml of dry ether are then added to the reaction mixture, and the 1,3,4,5-tetrahydro-phenyl-7-nitro-2H-1,4-benzodiazepin-2-one hydrobromide which has separated out is filtered off and washed with ether . 2.5 g of crude hydrobromide are obtained.
This product can be used immediately without further purification; for analytical purposes, an analytically pure product can be obtained by repeated partitioning between dichloromethane and dilute ammonia solution, then between dichloromethane and dilute hydrochloric acid; F. 232-234 "C; R, = 0.35 (hexane-acetic acid-chloroform 1: 1: 8).
c) 1,3-Dihydro-5-phenyl-7-nitro-2H-1 .4-benzodiazepin -2-one.
