Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung von 18-Methylensteroiden der Formel

worin R eine veresterte Hydroxylgruppe bedeutet und in welchen in 6,7-Stellung eine weitere Doppelbindung vorhanden sein kann.
Die veresterte Hydroxygruppe R leitet sich vorzugsweise von organischen Säuren der aliphatischen, alicyclischen, aromatischen oder heterocyclischen Reihe ab, insbesondere von solchen mit 1-18 Kohlenstoffatomen, z.B. der Ameisensäure, Essigsäure, Propionsäure, der Buttersäuren, Valeriansäuren, wie n-Valeriansäure, oder Trimethylessigsäure, Trifluoressigsäure, der Capronsäuren, wie p-Trimethyl-propionsäure oder Diäthylessigsäure, der Oenanth-, Capryl-, Pelargon-, Caprin-, Undecylsäuren, der Undecylensäure, der Laurin-, Myristin-, Palmitin- oder Stearinsäuren, der Ölsäure, oder einer Cyclopropan-, -butan-, pentan- und -hexancarbonsäure, einer Cycloalkyl-alkancarbonsäure, z.B.
Cyclopropylmethancarbonsäure, Cyclobutylmethancarbonsäure, Cyclopentyläthancarbonsäure, Cyclohexyläthancarbonsäure, der Benzoesäure, von Phenoxyalkansäuren, wie Phenoxyessigsäure, von Dicarbonsäuren, wie Bernsteinsäure, Phthalsäure, Chinolinsäure, der Furan-2-carbonsäure, 5-tert.-Butyl-furan-2-carbonsäure, 5-Brom-furan-2 -carbonsäure, der Nicotinsäure oder der Isonicotinsäure, oder von Sulfonsäuren, wie Benzolsulfonsäuren, oder von anorganischen Säuren, wie z.B. Phosphor- oder Schwefelsäuren.
Insbesondere stellt R eine mit einer niederaliphatischen Carbonsäure veresterte Hydroxygruppe dar, wie Essigsäure, Propionsäure, Trimethylessigsäure, Valeriansäure oder Capronsäure.
Die neuen Verbindungen der obigen Formel (I) besitzen wertvolle pharmakologische Eigenschaften. So weisen sie neben einer gestagenen Wirkung insbesondere eine ovulationshemmende Wirkung auf, wie sich im Tierversuch z.B. an der Ratte zeigt. Die neuen Verbindungen können daher als Gestagene und Contraceptiva Verwendung finden. Die neuen Verbindungen sind aber auch wertvolle Zwischenprodukte für die Herstellung anderer nützlicher Stoffe, insbesondere von pharmakologisch wirksamen Verbindungen.
Besonders hervorzuheben ist das 17cc-Acetoxy-18-methy- len-l 9-nor-progesteron und sein 6-Dehydroderivat.
Das Verfahren zur Darstellung der Verbindungen der obigen Formel I ist dadurch gekennzeichnet, dass man in einer Verbindung der Formel

und in welcher auch eine Doppelbindung in 6,7-Stellung vorhanden sein kann, die 17-OH-Gruppe verestert.
Wenn erwünscht können erhaltene Verbindungen in 6,7 Stellung unter Einführung einer Doppelbindung dehydriert werden.
Die Ausgangsstoffe der Formel II können z.fl. nach dem Verfahren la oder lb des französischen Patentes Nr. 1 601140 hergestellt werden. Man geht z.B. von entsprechenden in 17 Stellung substituierten 19-Nor-5-pregnen-20-hydroxy-18-säure -(18-20)-Laktonen aus, die in 3-Stellung eine freie oder geschützte Hydroxygruppe oder eine ketalisierte Oxogruppe aufweisen und gelangt durch Anwendung des genannten Verfahrens lb und gegebenenfalls Ausbildung der A4-3-Oxogruppe und evtl. Dehydrierung in 6-Stellung direkt zu Verbindungen der obigen Formel II.
Beim Verfahren la des genannten französischen Patentes geht man vorteilhaft von in 17-Stellung gemäss der Formel II substituierten Verbindungen aus, in denen die 20-Oxogruppe ketalisiert ist, wobei auch eine 3-Oxogruppe mit Vorteil solchermassen geschützt ist. Man gelangt so zu 20-Monoketalen und 3,20-Diketalen der obigen Formel II, in welchen dann die ketalisierten Oxogruppen freigesetzt werden.
Die verfahrensgemässe Veresterung der 17,a-Hydroxy- gruppe kann in an sich bekannter Weise vorgenommen werden, z.B. durch Behandlung mit einem Säureanhydrid oder Säurehalogenid in Gegenwart einer tertiären Base wie Collidin, oder einer starken Säure, wie Perchlorsäure, wenn erwünscht, in Anwesenheit eines inerten Lösungsmittels, vorzugsweise bei etwas erhöhter Temperatur.
Die gegebenenfalls vorzunehmende Einführung einer zusätzlichen Doppelbindung in 6,7-Stellung kann in an sich bekannter Weise ausgeführt werden, z.B. mit einem Chinon, z.B.
Chloranil oder Dicyanodicyanbenzochinon, in einem organischen Lösungsmittel z.B. in Toluollösung.
Die gemäss der vorliegenden Erfindung herzustellenden Verbindungen der obigen Formel I können zur Herstellung von pharmazeutischen Präparaten für die Human- oder Veterinärmedizin verwendet werden. Als pharmazeutische Träger verwendet man organische oder anorganische Stoffe, die für die enterale, z.B. orale, parenterale oder topische Gabe geeignet sind. Für die Bildung derselben kommen solche Stoffe in Frage, die mit den neuen Verbindungen nicht reagieren, wie z.B. Wasser, Gelatine, Milchzucker, Stärke, Magnesium-stea- rat, Talk, pflanzliche Öle, Benzylalkohole, Gummi, Polyalkylenglykole, Vaseline, Cholesterin und andere bekannte Arzneimittelträger. Die pharmazeutischen Präparate können in fester Form, z.B. Cremen vorliegen.
Gegebenenfalls sind diese pharmazeutischen Präparate sterilisiert und bzw. oder enthalten Hilfsstoffe, wie Konservierungs-, Stabilisierungs-, Netz- oder Emulgiermittel, Salze zur Veränderung des osmotischen Drukkes oder Puffer. Sie können auch noch andere therapeutisch wertvolle Stoffe enthalten. Die neuen Verbindungen können auch als Ausgangsprodukte für die Herstellung anderen wertvoller Verbindungen dienen.
Die Verbindungen der vorliegenden Erfindung können auch als Futterzusatzmittel verwendet werden.
Die Erfindung wird in dem folgenden Beispiel näher beschrieben. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel
1,0 g 3,20-Dioxo-17,x-hydroxy-|A4st8(l8a)-l 8-Homo-l9-nor -pregnadien werden in einem Gemisch bestehend aus 70,4 ml absolutem Essigester, 0,02 ml 70%iger Perchlorsäure und 9,60 ml Essigsäureanhydrid gelöst und 5 Minuten bei Zimmertemperatur gerührt. Die Reaktionslösung wird auf Eis/ Wasser gegossen, 20 Minuten stark gerührt und zweimal mit Äther extrahiert. Die organischen Phasen werden nacheinan der mit Wasser, ln-Natronlauge und Wasser gewaschen, getrocknet und im Wasserstrahlvakuum eingedampft. Das so erhaltene amorphe 3,17a-Diacetoxy-20-oXo-L536,18(188,.18 -19-nor-pregnatrien wird in 17 ml Chloroform und 120 ml Methanol gelöst, mit 43 ml ln-Natronlauge versetzt und 1 Minute bei Zimmertemperatur gerührt.
Die Reaktionslösung wird auf Eis/Wasser ausgetragen, mit 2n-Salzsäure schwach angesäuert und zweimal mit Methylenchlorid extrahiert. Die organischen Phasen wäscht man nacheinander mit Wasser, ges.Natriumhydrogencarbonatlösung und Wasser, trocknet und dampft im Wasserstrahlvakuum ein. Nach Adsorption des anfallenden Schaumes (in Toluol-Lösung) an SOfacher Menge Silicagel und Eluieren mit Toluol-Essigester (98 : 2)-Gemisch erhält man das kristalline 3,20-Dioxo-17v,-acetoxy-A4b18(l3a)- -18-homo-19-nor-pregnadien, das nach Umlösen aus Methylenchlorid-Äther bei 200-201 C schmilzt.
