La présente invention a pour objet un procédé de préparation de composés ayant le noyau pyrazolo [3,4-b] pyridine, n'étant pas substitués en position 1, c'est-à-dire qu'il n'y a qu'un hydrogène libre et pas d'autres substituants sar l'atome d'azote dans cette position, et ayant divers substituants en position 4 et un groupe carbonyle fixé en position 5. La position 3 peut être non substituée ou substituée. La position 6 est de préférence, mais pas nécessairement, non substituée. Le substituant en position 4 peut être un radical hydroxy, halogéno, alcoxy inférieur, amino acyclique ou hétérocyclique du type décrit ci-dessous ou un groupe hydrazino. En la position 5, le groupe carbonyle fixé à l'atome de carbone du noyau peut porter un groupe hydroxyle, alcoxy inférieur, phényle ou phényle substitué ou amino acyclique ou hétérocyclique du type précité.

Les divers produits finals obtenus par le procédé de l'invention sont utiles localement comme agents antimicrobiens, par exemple pour combattre des infections dues à des micro-organismes tels que Staphylococcus aureus, et aussi comme agents dépressifs du système nerveux central pour soulager les états d'angoisse et de tension.
Les exemples suivants illustrent l'invention et des formes d'exécution préférées. On peut obtenir d'autres produits de la même manière en modifiant les ingrédients de manière appropriée. Toutes les températures sont en degrés centigrade.
Exemple 1:
Ethylester d'acide 4butylamino-3-méthyl-lH-pyrazolo[3,4- bjpyridine-5-carboxylique.
A (a) Diéthylesterd'acide[[[1-(2-furyl)méthyl-3-méthyl- 5-pyrazolyljaminojméthylêne]malonîque.
On chauffe 177 g (1 mole) de l-(2-furyl)méthyl-3-méthyl-5aminopyrazole et 216 g (1 mole) de diéthylester d'acide éthoxyméthylènemalonique à 130"C jusqu'à ce qu'on ait chassé par distillation la quantité théorique d'alcool. On recristallise l'huile restante, c'est-à-dire le diéthylester d'acide [[[l-(2-furyl)méthyl-3 méthyl-5-pyrazolyl]amino]méthylène]malonique dans le méthanol; on obtient 305 g (rendement de 88%) qui fondent à 95" C.
(b) Ethylester d'acide 4-hydroxy 2-furyl)méthyl-3- méthylpyrazolo[3,4-bApyridine-5-carboxylique.
On dissout 347 g (1 mole) de diéthylester d'acide [[[1-(2- furyl)mèthyl-3-méthyl-5-pyrazolyl]aminojméthylène]malonique dans 1 litre de diphényléther et on chauffe à 2400 C pendant 2 heures. On chasse par distillation continue l'méthanol formé. On sépare le solvant sous vide. L'éthylester d'acide 4-hydroxy-l-(2- furyl) méthyl-3-métliylpyrazolo[3,4b]pyridine-5-carboxylique reste et on le recristallise dans le méthanol; on obtient 182 g (rendement de 60%); il fond à 82"C.
(c) Ethylester d'acide 4-éthoxy-1- (2-furyl)méthyl-3- méthylpyrazolo[3,4-bapyridine-5-carboxylique.
On met en suspension 150 g (0,5 mole) d'éthylester d'acide 4 hydroxy- 1 -(2-furyl)méthyl-3-méthylpyrazolo[3,4-b]pyridine-5- carboxylique, 140 g de carbonate de potassium et 155 g d'iodure d'éthyle dans 500 ml de diméthylformamide et on chauffe en agitant à 60"C pendant 10 heures. A ce moment, on filtre l'excès de carbonate de potassium et l'iodure de potassium précipité. On dilue le filtrat avec 500 ml d'eau. L'éthylester d'acide 4-éthoxy-1- (2-furyl)méthyl-3-méthylpyrazolo[3 ,4-b]pyridine-5-carboxylique précipite et on le recristallise dans l'hexane; on obtient 125 g (rendement de 76%); il fond à 82"C.
(d) Ethylester d'acide 4-butylamino-1-(2-furyl)méthyl-3- methyl-4-butylaminopyrazolo[3,4-bipyridine-5-carboxy- lique.
On dissout 32,8 g (0,1 mole) d'éthylester d'acide 4-éthoxy-1-(2 furyl)-methyl-3-méthylpyrazolo-[3,4b]pyridine-5-carboxylique dans 100 ml de dioxanne et on chauffe à reflux pendant 5 heures avec 11 g (0,15 mole) de n-butylamine. On évapore alors à sec le solvant et on recristallise le résidu dans l'hexane; on obtient 25,5 g (rendement de 72%) d'éthylester d'acide 4-butylamino-1-(2- furyl)méthyl-3-méthyl-4-butylaminopyrazolo[3,4-b]pyridine-5carboxylique qui fond à 77" C.
B. Ethylester d'acide 4-butylamino-3-méthyl-lH-pyrazolo
[3,4-b]pyridine-5-carboxylique.
On met en suspension 17,8 g (0,05 mole)d'éthylester d'acide 4butylamino- l-(2-furyl)méthyl-3-méthylpyrazolo[3,4bjpyridine carboxylique et 11,1 g (0,1 mole) de bioxyde de sélénium dans 50 ml de diéthylèneglycoldimèthyléther et on chauffe à 160"C. On ajoute quelques gouttes d'eau et on maintient la température pendant 1,5 heure. Après refroidissement on filtre le mélange et op le dilue avec 20 ml d'eau. Il se forme des cristaux jaunes pâles d'éthylester d'acide 4-butylamino-3-méthyl- 1 -H-pyrazolo[3 ,4- b]pyridine-5-carboxylique et on les recristallise dans l'èthanol; on obtient 10,2 g (rendement de 74%); il fond à 174-176 C.
Exemple 2: 4-Butylamino-lH-pyrazolo[3,4b]pyridine-5-diéthylaminocar- boxamide.
A (a) Diéthylester d'acide[[[l-(4-picolyl)-5- pyrazolylJaminolméthylène]malonique .
On chauffe 174 g de 1-(4-picolyl)-5-aminopyrazole et 216 g de diéthylester d'acide éthoxyméthylènemalonique, en agitant, à 1400 C, jusqu'à ce que la quantité théorique d'alcool soit chassée par distillation. Le mélange réactionnel cristallise au refroidissement. Après recristallisation dans l'acétate d'éthyle on obtient 220 g de diéthylester d'acide [[[1 -(4-picolyl)-5-pyrazolyl]amino]mé- thylene]malonique (rendement de 65%) qui fond à 95-97 C.
(b) Ethylester d'acide 4-hydroxy-1- (4-picolyl) -1H- pyrazolo[3,4-bApyridine-5-carboxylique.
On chauffe 86 g (0,25 mole) de diéthylester d'acide [[[1-(4 picolyl)-5-pyrazolyl]amino]méthylènejmalonique à 240"C pendant 15 minutes. On refroidit l'huile foncée et on ajoute 200 ml de méthanol. L'éthylester d'acide 4-hydroxy-1-(4-picolyl)-1H-pyra- zolo[3,4-b]pyridine-5-carboxylique cristallise à l'abandon; on obtient 33 g (rendement de 44%) qui fondent à 140 C.
B (c) Ethylester d'acide 4-hydroxy-lH-pyrazolo.
[3,4-bApyridine-5-carboxylique.
On dissout 3 g (0,01 mole) d'éthylester d'acide 4-hydroxy-l-(4- picolyl)-1H-pyrazolo[3,4-b]pyridine-5-carboxylique dans 20 ml d'acide acétique. On ajoute 2,2 g (0,02 mole) de bioxyde de sélénium et 2-3 gouttes d'eau. On chauffe le mélange à reflux pendant 30 minutes puis on le filtre. L'éthylester d'acide 4-hydroxy-lH- pyrazolo[3,4-b]pyridine-5-carboxylique précipite au refroidissement. Après recristallisation dans l'acide acétique on obtient 1,8 g (rendement de 87%); il fond à 275"C.
(d) Ethylester d'acide 4-éthoxy-1H-pyrazolo[3,4-b]pyridine-
5-carboxylique.
On chauffe 4,1 g (0,02 mole) d'éthylester d'acide 4-hydroxy1H-pyrazolo[3,4-b]pyridine-5-carboxylique, 5,6 g (0,04 mole) de carbonate de potassium et 3,5 g (0,022 mole) d'iodure d'éthyle dans 30 ml de diméthylformamide en agitant, pendant 10 heures à 60 C. On sépare alors par filtration l'excès de carbonate de potassium et on ajoute 30 ml d'eau. L'éthylester d'acide 4-éthoxy1H-pyrazolo[3,4-b]pyridine-5-carboxylique précipite et on le recristallise dans le méthanol; on obtient 2 g (rendement de 42,5%); il fond à 180 C.
(e) Ethylester d'acide 4-butylamino-lH-pyrazolo- [3 ,4bapyridine-5-carboxylique.
On traite 2,35 g (0,01 mole) d'éthylester d'acide 4-èthoxy-îH-
pyrazolo[3,4-b]pyridine-5-carboxylique avec 2,2 g (0,03 mole) de
butylamine à 900 C pendant une heure. On refroidit alors le mélange, on le dilue avec 20 ml d'eau et on sépare par filtration le
précipité cristallin blanc. Après recristallisation dans le diéthylé
ther, on obtient 1,7 g (rendement de 72%) d'éthylester d'acide 4
butylamino-lH-pyrazolo[3,4-b]pyridine-5-carboxylique qui fond à
181 C.
(f) Acide 4-butylamino-1H-pyrazolo[3.4.b]pyridine-5-carboxy-
lique.
On traite 2,6 g (0,01 mole) d'éthylester d'acide 4-butylamino-
1H-pyrazolo[3,4-b]pyridine-5-carboxylique avec 1,1 g d'hydroxyde de sodium dans 30 ml d'éthanol pendant 20 heures à
la température ordinaire. On sépare le solvant sous vide et on dissout le résidu dans 10 ml d'eau. Après acidification avec l'acide
acétique, l'acide 4-butylamino-1H-pyrazolo[3,4b]pyridine-5- carboxylique se solidifie et on le sépare par filtration. On purifie le produit par recristallisation dans l'acide acétique; on obtient 1,9 g (rendement de 82%); il fond à 225"C.
(g) 4-Butylamino-5-diéthylaminocarbonyl-1H-pyrazolo[3,4.b]-
pyridine.
On chauffe à reflux 2,3 g (0,01 mole) d'acide 4-butylamino-
1H-pyrazolo[3,4-b]-5-carboxylique avec 10 ml de chlorure de thionyle pendant 5 heures. On sépare alors sous vide l'excès de chlorure de thionyle, on dissout le résidu dans 20 ml de tétrahydrofuranne sec et on ajoute 2 g de diéthylamine en refroidissant.
On laisse reposer le mélange pendant 24 heures, puis on évapore à
sec le solvant et on ajoute 20 ntl d'eau au résidu. On filtre la 4butylamino-5-diéthylaminocarbonyl-1 H-pyrazolo[3,4-b]pyridine et on la recristallise dans l'acétate d'éthyle pour obtenir 2,1 g (rendement de 70%) qui fondent à 130 C.
Exemple 3:
Ethylester d'acide 4- (2-cyclohexylidène) hydrazino-l H-pyra-
zolo[3,4-b]pyridine-5-carboxylique.
(a) Ethylester d'acide 4-chloro-IH-pyrazolo[3,4-b]pyridine-5-
carboxylique.
On chauffe à reflux 20,7 g (0,1 mole) d'éthylester d'acide 4
hydroxy-lH-pyrazolo[3,4-b]pyridine-5-carboxylique pendant
5 heures avec 100 ml d'oxychlorure de phosphore. On chasse par distillation distillation l'excès d'oxychlorure de phosphore et on verse le
résidu huileux sur de la glace. Après neutralisation avec de
l'ammoniac aqueux, I'éthylester d'acide 4-chloro-1H-pyrazolo[3,4-
bgpyridine-5-carboxylique se sépare et on le recristallise dans
l'ethanol; on obtient 10,5 g (rendement de 47%) qui fondent à
169-171 C.
(b) Ethylester d'acide 4hydrazino-lH-pyrazolo[3,4-b]-
pyridine-5-carboxylique.

265 C avec décomposition.
Exemple 4: S-Benzoyl-4 (2-aminobutylJ -I H-pyrazolo[3,4-bApyridine.
A (a) Ethylester d'acide [[[1-(2-furyl)méthylpyrazolyl]-
amino]méthylène]benzoylacétique.
On chauffe 163 g (1 mole) de l-(2-furyl)méthyl-5-aminopyra- zole et 248 g (1 mole) d'éthylester d'acide éthoxyméthylèneben zoylacétique à 1300C jusqu'à ce qu'il ne distille plus d'alcool (approximativement 1 heure). On dissout 1,7 g (0,005 mole) de 5-benzoyl-4-éthoxy-l-(2furyl)méthylpyrazolo[3,4-b]pyridine dans 5 ml de diéthylèneglycoldiméthyléther; on ajoute 1,1 g de bioxyde de sélénium et on chauffe le mélange, en agitant, à 160"C. acetic acid, 4-butylamino-1H-pyrazolo [3,4b] pyridin-5-carboxylic acid solidifies and is separated by filtration. The product is purified by recrystallization from acetic acid; 1.9 g are obtained (yield 82%); it melts at 225 "C.
(g) 4-Butylamino-5-diethylaminocarbonyl-1H-pyrazolo [3,4.b] -
pyridine.
2.3 g (0.01 mol) of 4-butylamino- acid are heated to reflux.

reflux for 15 minutes with 1 ml of hydra hydrate

hydrazino-1H-pyrazolo [3,4-b] pyridin-5-carboxylic separates and
it is recrystallized from butanol; 3.5 g are obtained (yield of
64%) which melt at 350 "C.
(c) 4- (2-Cyclohexylidene) hydrazino acid ethyl ester
1H-pyrazolo [3,4-b] pyridine-5-carboxylic acid.
2.21 g (0.01 mole) of acid ethyl ester 4 are suspended
hydrazino-1H-pyrazolo [3,4-b] pyridine-5-carboxylic in 5 ml
acetic acid. Add 1 g of cyclohexanone and heat the
mixing at reflux for 10 minutes. 10 ml of water are added.
After cooling the 4- (2-cyclohexylidene) hydrazino- 1H
pyrazolo [3,4-b] pyridine precipitates and is recrystallized from acid
acetic; 2.2 g are obtained (yield of 73%) which melt at
265 C with decomposition.
Example 4: S-Benzoyl-4 (2-aminobutylJ -I H-pyrazolo [3,4-bApyridine.
A (a) Acid ethyl ester [[[1- (2-furyl) methylpyrazolyl] -
amino] methylene] benzoylacetic.
163 g (1 mole) of 1- (2-furyl) methyl-5-aminopyrazole and 248 g (1 mole) of ethoxymethylene benzoylacetic acid ethyl ester are heated to 1300C until it no longer distills of alcohol (approximately 1 hour). The oily residue crystallizes, and after cooling and recrystallization from hexane, 310 g (yield 85%) of acid ethyl ester [[[1- (2-furyl) methyl-5-pyrazolyl] amino] are obtained. methylene] benzoylacetic which melts at 75 77o C.
(b) 5-Benzoyl-4-hydroxy-1- (2-furyl) methylpyrazolo [3,4-b] -
pyridine.
36.5 g of [[[1- (2-furyl) methyl-5-pyrazolyl] amino] methylene] benzoylacetic acid ethyl ester is dissolved in 50 ml of diphenyl ether and the mixture is refluxed at 260 C for 30 minutes. After distillation of the solvent, a dark oil is obtained which crystallizes after the addition of methanol. After recrystallization, 20 g (61% yield) of 5-benzoyl-4-hydroxy-1- (2-furyl) methylpyrazolo [3,4-b] pyridine are obtained, which melts at 102 ° C.
(c) 5-Benzoyl-4-ethoxy-1- (2-furyl) methylpyrazolo
[3,4bApyridine.
3.3 g (0.01 mol) of 5-benzoyl-4-hydroxy-1- (2-furyl) methylpyrazolo [3,4-b] pyridine is dissolved in 20 ml of dimethylformamide. 2.8 g of potassium carbonate and 3.1 g of ethyl iodide are added and the mixture is heated for 12 hours at 600 ° C. The excess potassium carbonate is filtered off and water is added. 5-Benzoyl-4-ethoxy-1- (2-furyl) methylpyrazolo [3,4-b] pyridine precipitates and is recrystallized from hexane; 3 g are obtained (yield 86%) which melt at 70 ° C.
B (d) 5-Benzoyl-4-ethoxy-1H-pyrazolo [3,4-bipyridine.
1.7 g (0.005 mol) of 5-benzoyl-4-ethoxy-1- (2furyl) methylpyrazolo [3,4-b] pyridine are dissolved in 5 ml of diethylene glycol dimethyl ether; 1.1 g of selenium dioxide are added and the mixture is heated, with stirring, to 160 ° C. After adding a drop of water, the temperature is maintained for one hour. The mixture is filtered hot and the mixture is filtered. -benzoyl-4-ethoxy-1H-pyra zolo [3,4b] pyridine precipitates on cooling After recrystallization from butanol, 1 g (yield 77%) is obtained which melts at 195-197 C.

0.65 g (0.0025 mol) of 5-benzoyl-4ethoxy-1H-pyrazolo [3,4-b] pyridine is refluxed with 1 ml of butylamine for 10 minutes. The mixture is cooled and 10 ml of water are added. 5-Benzoyl-4- (2-aminobutyl) - 1 H-pyrazolo [3, 4-b] pyridine precipitates, is filtered and recrystallized from butanol; 1.1 g (76% yield) are obtained which melts at 175 ° C.
