Die vorliegende Erfindung bezieht sich auf ein Verfahren zur Herstellung neuer Chinazolinthione der Formel:

der Buchstabe n die Zahl 0 oder eine ganze Zahl von 1 bis 3, jedes der Symbole R1, R2 und R3 Wasserstoff, ein Halogenatom, eine Nitrogruppe, einen Alkyl-, Alkoxy-, Alkylthio-, Alkylsulfonyl- oder Alkylsulfinylrest mit jeweils 1 bis 4 Kohlenstoffatomen oder einen Trifluormethylrest und R4 einen Cycloalkylrest mit 3 bis 6 Kohlenstoffatomen bedeuten.
In den Verbindungen der obigen Formel I kann man als Halogenatome Chlor-, Brom-, Jod- und Fluoratome einsetzen.
Beispiele von Alkylgruppen mit 1 bis 4 Kohlenstoffatomen sind Methyl-, Äthyl-, n-Propyl-, Isopropyl-, n-Butyl-, Isobutylund tert.-Butylgruppen. Beispiele von Alkoxygruppen mit 1 bis 4 Kohlenstoffatomen sind Methoxy-, Äthoxy-, n-Propoxy-, Isopropoxy-, n-Butoxy-, Isobutoxy- und tert.-Butoxyreste.
Beispiele von Cycloalkylgruppen mit 3 bis 6 Kohlenstoffatomen sind Cyclopropyl-, Cyclobutyl-, Cyclopentyl-, Cyclohexyl-, Methylcyclopropyl-, Dimethylcyclopropylreste usw. Stellt die -CnHn-Gruppe eine Alkylengruppe mit 1 bis 3 Kohlenstoffatomen dar, so kommen beispielsweise Methylen-, Äthylen-, l-Methyläthylen-, 2-Methyläthylen- und Trimethylenreste in Frage.
Verbindungen der Formel I und deren pharmazeutisch annehmbare Säureadditionssalze (z. B. die Salzsäure-, Bromwasserstoffsäure-, Schwefelsäure-, Phorphorsäure-, Salpetersäure-, Essigsäure-, Maleinsäure-, Fumarsäure-, Weinsäure-, Bernsteinsäure- oder Citronensäure-Additionssalze von solchen Verbindungen) besitzen hervorragende pharmakologische Eigenschaften und sind insbesondere ausgezeichnete entzündungshemmende und analgetische Mittel. Sie eignen sich auch als Zwischenprodukte für die Herstellung von anderen Arzneimitteln.
Das erfindungsgemässe Verfahren besteht darin, dass man eine Verbindung der Formel:

Die Umsetzung der Verbindungen der Formel IV-b mit äquimolaren Mengen oder grösseren Mengen an Phosgen wird vorzugsweise in Gegenwart eines inerten Lösungsmittels, wie z. B. Äther, Benzol, Chloroform, Toluol, Dioxan usw., durchgeführt. Die Umsetzung erfolgt vorzugsweise in Gegenwart eines säurebindenden Mittels. Beispiele geeigneter säurebindender Mittel sind tertiäre organische Basen, wie z. B. Tri äthylamin, Tributylamin, Pyridin oder N-Methylpiperidin, Alkalimetallhydroxyde, wie z. B. Natriumhydroxyd oder Kaliumhydroxyd, sowie ,Alkalimetallcarbonate, wie z. B. Natriumcarbonat oder Kaliumcarbonat.
Die als Ausgangsprodukte eingesetzten Verbindungen der Formel IV-b können dadurch erhalten werden, dass man ein Indolderivat der Formel:

Beispiele von für die Oxydation der Indolderivate der Formel VII verwendeten Oxydationsmitteln sind Ozon, Wasserstoffperoxyd, Persäuren, wie z. B. Perameisensäure, Peressigsäure und Perbenzoesäure, Chromsäure, Kaliumpermanganat und dgl. Das bevorzugte Oxydationsmittel ist Chromsäure oder Ozon. Die Oxydationsreaktion erfolgt vorzugsweise in Gegenwart eines Lösungsmittels oder eines Lösungsmittelgemisches. Die Wahl des Lösungsmittels hängt vom verwendeten Oxydationsmittel ab. Als solche kommen Wasser, Aceton, Tetrachlorkohlenstoff, Essigsäure, Schwefelsäure und dgl. in Frage. Das Oxydationsmittel wird in stöchiometrischer Menge oder in einer grösseren Menge verwendet. Die Reaktionstemperatur schwankt je nach dem verwendeten Oxydationsmittel.
Die Hydrolyse der Verbindungen der Formel VIII verläuft in Gegenwart eines Hydrolysiermittels. Beispiele von Hydrolysiermitteln sind Mineralsäuren, wie z. B. Salzsäure und Schwefelsäure, Alkalimetallhydroxyde, wie z. B. Natriumhydroxyd und Kaliumhydroxyd, Erdalkalimetallhydroxyde, wie z. B.
Calciumhydroxyd und Bariumhydroxyd, Alkalimetallcarbonate, wie z. B. Natriumcarbonat und Kaliumcarbonat, sowie Ammoniakverbindungen, wie z. B. Ammoniumhydroxyd. Die Hydrolyse erfolgt vorzugsweise in einem Lösungsmittel oder in einem Lösungsmittelgemisch. Einige Beispiele geeigneter Lösungsmittel sind Wasser, Methanol, Äthanol, Aceton, Dimethylsulfoxyd und dgl., sowie Mischungen davon.
Die Verbindungen der Formel IV-b können auch dadurch erhalten werden, dass man Benzonitrilderivate der Formel:

Die Erfindung sei nun anhand der folgenden Beispiele erläutert.
Herstellung der Ausgangsprodukte a) Eine Lösung von 50 g Chromianhydrid in 50 cm3 Wasser wird tropfenweise einer Suspension von 60,2 g l-Cyclopropylmethyl-3-phenyl-5-chlorindol-2-carbonsäureäthylester in 340 cm3 Eisessig bei 20 bis 25eC zugegeben. Das Gemisch wird dann während 6 Stunden auf 50 bis 55oC erwärmt. Das Reaktionsgemisch wird hierauf in Wasser gegossen und mit Toluol extrahiert. Die Toluolextrakte werden vereinigt, mit
Wasser gewaschen und über Natriumsulfat getrocknet, worauf man das Lösungsmittel unter vermindertem Druck entfernt.
Auf diese Weise erhält man 61,8 g rohes 2-(N-Cyclopropylme- thyl-äthoxalylamino)-5 -chlorbenzophenon in Form eines Ols.
Durch Auskristallisierenlassen aus einer Mischung von Ätha nol und Petroläther gelangt man zu farblosen Kristallen vom Smp. 67 bis 68.
Das so erhaltene rohe 2-(N-Cyclopropylmethyläthoxalylamino)-5-chlorbenzophenon wird in 260 cm3 Dimethylsulfoxyd gelöst. Die Lösung wird hierauf mit 57 g 40 %oder Kaliumhydroxydlösung versetzt und die Mischung während 2 Stunden auf 50 bis 55oC erhitzt. Das Reaktionsgemisch wird mit Wasser verdünnt und die ausgefällten Niederschläge werden durch Filtrieren gesammelt, mit Wasser gewaschen und getrocknet, wobei man 43,4 g rohes 2-Cyclopropylmethylamino-5-chlorbenzophenon vom Smp. 77 bis 78oC erhält.
Durch Umkrisfallisieren aus Äthanol erhält man das reine Produkt mit einem Schmelzpunkt von 86 bis 87.
b) Eine Mischung von 25 cm3 konzentrierter Salzsäure und 25 cm3 Wasser wird einer Lösung von 2,5 g 2-(N-Cyclopropylmethyläthoxalyl-amino)-5-chlorbenzophenon in 62,5 cm3 Äthanol hinzugegeben. Das Gemisch wird während 6 Stunden unter Rückfluss erhitzt. Nach dem Kühlen wird das Reaktionsgemisch unter vermindertem Druck eingeengt, mit
100 cm3 Wasser verdünnt und mit Chloroform extrahiert. Die Chloroformextrakte werden vereinigt, nacheinander mit Wasser und mit 20%iger Kaliumhydroxydlösung gewaschen und über Natriumsulfat getrocknet. Das Chloroform wird unter vermindertem Druck entfernt, wobei man 2-Cyclopropylmethylamino-5-chlorbenzophenon in quantitativer Menge erhält.
c) Unter Anwendung einer ähnlichen Methode wie jener von Abschnitt a), jedoch durch Ersatz des
1 -Cyclopropylmethyl-3 -phenyl-5-chlorindol2-carbonsäureäthylesters durch
1-Cyclopropylmethyl-3-phenyl-5-chlor-indol-2-carbonsäure, erhält man 2-(N-Cyclopropylmethylhydroxyoxalyl-amino)-5- chlorbenzophenon in Form eines Öls.
Eine Mischung von 8 g 2-(N-Cyclopropylmethyl-hydroxyoxalyl-amino)-5-chlorbenzophenon, 9,2 g Natriumhydroxyd und 100 cm3 Wasser wird während 2 Stunden unter Rückfluss erhitzt. Das Reaktionsgemisch wird gekühlt und die Niederschläge werden durch Filtrieren gesammelt, mit Wasser gewaschen und getrocknet, wobei man 2-Cyclopropylmethylamino5-chlorbenzophenon mit einem Schmelzpunkt von 82 bis 830C erhält. Dieses Produkt ist identisch mit der Verbindung gemäss Beispiel 1, wie dies durch das Infrarotabsorptionsspektrum nachgewiesen wird.
d) Eine Lösung von 70 g Chromianhydrid in 70 cm3 Wasser wird tropfenweise einer Suspension von 73,6 g l-Cyclopropylmethyl-2-cyano-3-phenyl-5-chlorindol in 500 cm3 Eisessig bei Zimmertemperatur hinzugegeben. Das Gemisch wird über Nacht bei Zimmertemperatur gerührt. Hierauf wird das Reaktionsgemisch filtriert und das Filtrat in Wasser gegossen und mit Chloroform extrahiert. Die Chloroformextrakte werden vereinigt, mit Wasser gewaschen und über Natriumsulfat getrocknet, worauf man das Lösungsmittel unter vermindertem Druck entfernt. Auf diese Weise erhält man 55 g 2-(N-Cyclo propylmethyl-cyanocarbonylamino)-5 -chlorbenzophenon in Form eines Ols.
Zu einer Lösung von 34 g 2-(N-Cyclopropylmethyl-cyanocarbonylamino)-5-chlor-benzophenon in 200 cm3 Äthanol gibt man eine Lösung von 120 g Natriumhydroxyd in 300 cm3 Wasser hinzu, worauf man die Mischung während 1 Stunde unter Rückfluss erhitzt. Das Reaktionsgemisch wird mit Chloroform extrahiert. Die Chloroformextrakte werden vereinigt, mit Wasser gewaschen und über Natriumsulfat getrocknet und das Lösungsmittel unter vermindertem Druck entfernt. Der Rückstand wird auf Siliciumdioxydgel chromatographiert und mit Chloroform eluiert, wobei man 2-Cyclopropylmethylamino-5-chlorbenzophenon, Smp. 82 bis 830C, erhält. Dieses Produkt ist identisch mit der Verbindung gemäss Abschnitt a), wie dies durch das Infrarotabsorptionsspektrum nachweisbar ist.
e) Zu einer Lösung von 17,5 g 1-ss-Cyclohexyläthyl)-3- phenyl-5 -chlorindol-carbonsäureäthylester in 95 cm3 Eisessig gibt man tropfenweise eine Lösung von 11,5 g Chromianhydrid in 11,5 cm3 Wasser bei 20 bis 25cC hinzu. Das Gemisch wird während 30 Minuten bei Zimmertemperatur gerührt und während 5 Stunden auf 50 bis 55 C erhitzt. Nach dem Kühlen wird das Reaktionsgemisch in 500 cm3 Wasser gegossen und mit Chloroform extrahiert. Die Extrakte werden vereinigt, mit Wasser gewaschen und über Natriumsulfat getrocknet, worauf man das Lösungsmittel unter vermindertem Druck entfernt.
Auf diese Weise erhält man 17,5 g 2-[N-(ss-Cyclohexyläthyl)- äthoxalylamino]-5-chlorbenzophenon in Form eines Öls.
Zu einer Lösung von 17,5 g des so erhaltenen 2-[N- (f3-Cyclohexyläthyl) -äthoxalylamino] - 5 -chlorbenzophenon in 400 cm3 Äthanol gibt man tropfenweise 150 cm3 konzentrierte Salzsäure hinzu, worauf man das Gemisch während 7 Stunden unter Rückfluss erhitzt. Hierauf wird das Lösungsmittel unter vermindertem Druck entfernt. Der Rückstand wird mit 300 cm3 kaltem Wasser versetzt und das Gemisch mit konzentriertem Ammoniumhydroxyd neutralisiert und hierauf mit Äther extrahiert. Die Extrakte werden vereinigt, mit Wasser gewaschen und über Natriumsulfat getrocknet, worauf man das Lösungsmittel unter vermindertem Druck entfernt. Auf diese Weise erhält man 10,7 g 2-(P-Cyclohexyläthylamino)-5chlorbenzophenon in Form eines braunen Öls.
f) Eine Lösung von 13,5 g Chromianhydrid in 13,5 cm3 wird zu einer Lösung von 17,4 g l-Cyclopropylmethyl-3-phenyl-5trifluormethylindol-2-carbonsäureäthylester in 100 cm3 Eisessig bei 20 bis 25 C hinzugegeben. Das Gemisch wird während 30 Minuten bei Zimmertemperatur gerührt und hierauf während 7 Stunden auf 50 bis 55 C erhitzt. Nach dem Kühlern wird das Reaktionsgemisch in 500 cm3 Wasser gegossen und 2-mal mit jeweils 150 cm3 Chloroform extrahiert. Die kombinierten Extrakte werden mit Wasser gewaschen, über Natriumsulfat getrocknet und im Vakuum zur Trockne eingeengt, wobei man 16,4 g 2-(N-Cyclopropylmethyläthoxalyl- amino)-5 -trifluormethylbenzophenon in Form eines Öls erhält.
Das so erhaltene 2-(N-Cyclopropylmethyläthoxalylamino)-5 -trifluormethylben- zophenon wird in 200 cm3 einer 20%gen wässrigen Kaliumhydroxydlösung gelöst. Das Gemisch wird gerührt und während Stunden auf 70 bis 800C erhitzt, worauf man in einem Eisbad kühlt.
Der gelbe Niederschlag wird durch Filtrieren gesammelt, mit Wasser gewaschen und getrocknet, wobei man 7,47 g 2-Cyclo propylmethylamino-5-tritluormethylbenzophenon, Smp. 102,0 bis 103,50C erhält.
g) Aus 11,8 g Brombenzol und 1,0 g Lithiummetall in 40 ml wasserfreiem Äther wird eine Phenyllithiumlösung hergestellt und dieser Lösung unter Eiskühlung eine Lösung von 5,2 g 2 Cyclopropylmethylamino-5-chlorbenzonitril in 50 ml wasserfreiem Äther tropfenweise zugegeben. Die entstandene Reaktionsmischung wird über Nacht bei Raumtemperatur gerührt und hierauf unter Rühren in 200 ml Eiswasser gegeben. Die Ätherschicht wird abgetrennt, mit Wasser gewaschen und über wasserfreiem Natriumsulfat getrocknet. Das Lösungsmittel wird unter vermindertem Druck entfernt, wobei 2-Cyclopropyl-methylamino-5-chlorbenzophenonimin in Form eines dunkelbraunen Öls erhalten wird.
h) Ein Gemisch von 2,0 g 2-Cyclopropylmethylamino-5nitrobenzophenon, 20 ml flüssigem Ammoniak und 0,1 g Zinkchlorid wird während 3 Tagen auf 1200C in einem Druckgefäss erhitzt. Der überschüssige Ammoniak wird abgedampft und der Rückstand aus Diisopropyläther umkristallisiert; dabei wird 2-Cyclopropylmethylamino-5-nitIobenzoph vom PSm . 100-101oC erhalten.
i) Zu einer Lösung von 3,85 g 2-Cyclopropylmethylamino- 5-chlorbenzophenonimin und 12 cm3 Triäthylamin in 70 cm3 Benzol gibt man tropfenweise unter Kühlen 70 cm3 einer 10 %eigen Phosgenlösung in Benzol hinzu. Das Gemisch wird während 30 Minuten bei Zimmertemperatur gerührt und hierauf im Vakuum zur Trockne eingeengt.
Der Rückstand wird in 100 cm3 verdünnter wässriger Natriumcarbonatlösung und mit 100 cm3 Chloroform versetzt und das Gemisch gerührt. Die wässrige Schicht wird mit Chloroform extrahiert. Die organischen Schichten werden vereinigt, mit Wasser gewaschen und über Natriumsulfat getrocknet. Das Lösungsmittel wird unter vermindertem Druck entfernt und der Rückstand aus Äthanol umkristallisiert, wobei man 1 Cyclopropylmethyl-4-phenyl-6-chlor-2(1H)-chinazolin, Smp.
171 bis 1720C, erhält.
Nach derselben Arbeitsweise kann man die folgenden Verbindungen der Formel II (Ausgangsprodukte) herstellen: 1 -Cyclopropylmethyl-4-phenyl-6-brom-2(1H) chinazolinon, Smp. 163-1640C, l-Cyclopropylmethyl-4-phenyl-6-fluor-2(1 chinazolinon, Smp. 168,5-169,50C, l-Cyclopropylmethyl-4-phenyl-2(1H)-chinazo- linon.

Beispiel 1
Ein Gemisch von 2,0 g 1-Cyclopropylmethyl-4-phenyl-6- nitro-2(1H)-chinazolinon, 6 g Phosphorpentasulfid und 40 ml Pyridin wird während 20 Stunden unter Rückfluss zum Sieden erhitzt.
Nach Abkühlen wird das Reaktionsgemisch in 200 ml Wasser gegossen und das erhaltene Gemisch mit Chloroform extrahiert. Die Chloroformschicht wird nacheinander mit verdünnter Salzsäure und Wasser gewaschen und hierauf über wasserfreiem Natriumsulfat getrocknet. Das Chloroform wird unter vermindertem Druck verdampft und der Rückstand an einer Kieselgelsäule chromatographiert; durch Eluieren mit Chloroform erhält man 0,1 g 1-Cyclopropylmethyl-4-phenyl- 6-nitro-2(1H)-chinazolmthion. Durch Umkristallisieren aus Äthanol und Chloroform erhält man Plättchen von dunkeloranger Farbe und Smp. 205-2060C.
Beispiel 2
Ein Gemisch von 3,4 g 1-Cyclopropylmethyl-4-phenyl-6- chlor-2(1H)-chinazolinon, 11 g Phosphorpentasulfid und 40 ml Pyridin wird während 17 Stunden am Rückfluss zum Sieden erhitzt. Die weitere Ausarbeitung wird analog Beispiel 1 durchgeführt. Man erhält dabei 3,3 g l-Cyclopropylmethyl- 4-phenyl-6-chlor-2(1H)-chinazolinthion in Form oranger Nadeln vom Smp. 232-2340C.

In the compounds of the above formula I, the halogen atoms used can be chlorine, bromine, iodine and fluorine atoms.
Examples of alkyl groups having 1 to 4 carbon atoms are methyl, ethyl, n-propyl, isopropyl, n-butyl, isobutyl and tert-butyl groups. Examples of alkoxy groups with 1 to 4 carbon atoms are methoxy, ethoxy, n-propoxy, isopropoxy, n-butoxy, isobutoxy and tert-butoxy radicals.

The invention will now be explained using the following examples.
Preparation of the starting products a) A solution of 50 g of chromium anhydride in 50 cm3 of water is added dropwise to a suspension of 60.2 g of 1-cyclopropylmethyl-3-phenyl-5-chloroindole-2-carboxylic acid ethyl ester in 340 cm3 of glacial acetic acid at 20 to 25 ° C. The mixture is then heated to 50 to 55 ° C for 6 hours. The reaction mixture is then poured into water and extracted with toluene. The toluene extracts are combined with
Washed with water and dried over sodium sulfate, whereupon the solvent is removed under reduced pressure.
In this way, 61.8 g of crude 2- (N-cyclopropylmethyl-ethoxalylamino) -5-chlorobenzophenone are obtained in the form of an oil.
Colorless crystals with a melting point of 67 to 68 are obtained by letting them crystallize out of a mixture of ethane nol and petroleum ether.
The crude 2- (N-cyclopropylmethylethoxalylamino) -5-chlorobenzophenone thus obtained is dissolved in 260 cm3 of dimethyl sulfoxide. 57 g of 40% or potassium hydroxide solution are then added to the solution and the mixture is heated to 50 to 55 ° C. for 2 hours. The reaction mixture is diluted with water and the precipitated precipitates are collected by filtration, washed with water and dried to give 43.4 g of crude 2-cyclopropylmethylamino-5-chlorobenzophenone, mp 77 to 78 ° C.
Recrystallization from ethanol gives the pure product with a melting point of 86 to 87.
b) A mixture of 25 cm3 of concentrated hydrochloric acid and 25 cm3 of water is added to a solution of 2.5 g of 2- (N-cyclopropylmethylethoxalyl-amino) -5-chlorobenzophenone in 62.5 cm3 of ethanol. The mixture is refluxed for 6 hours. After cooling, the reaction mixture is concentrated under reduced pressure with
Diluted 100 cm3 of water and extracted with chloroform. The chloroform extracts are combined, washed successively with water and with 20% strength potassium hydroxide solution and dried over sodium sulfate. The chloroform is removed under reduced pressure, 2-cyclopropylmethylamino-5-chlorobenzophenone being obtained in a quantitative amount.
c) Using a method similar to that in section a), but replacing the

1-Cyclopropylmethyl-3-phenyl-5-chloro-indole-2-carboxylic acid, 2- (N-cyclopropylmethylhydroxyoxalyl-amino) -5-chlorobenzophenone is obtained in the form of an oil.
A mixture of 8 g of 2- (N-cyclopropylmethyl-hydroxyoxalyl-amino) -5-chlorobenzophenone, 9.2 g of sodium hydroxide and 100 cm3 of water is refluxed for 2 hours. The reaction mixture is cooled and the precipitates are collected by filtration, washed with water and dried to give 2-cyclopropylmethylamino5-chlorobenzophenone melting at 82-830C. This product is identical to the compound according to Example 1 as evidenced by the infrared absorption spectrum.
d) A solution of 70 g of chromium anhydride in 70 cm3 of water is added dropwise to a suspension of 73.6 g of l-cyclopropylmethyl-2-cyano-3-phenyl-5-chloroindole in 500 cm3 of glacial acetic acid at room temperature. The mixture is stirred at room temperature overnight. The reaction mixture is then filtered and the filtrate is poured into water and extracted with chloroform. The chloroform extracts are combined, washed with water and dried over sodium sulfate, whereupon the solvent is removed under reduced pressure. In this way, 55 g of 2- (N-cyclo propylmethyl-cyanocarbonylamino) -5-chlorobenzophenone are obtained in the form of an oil.
A solution of 120 g of sodium hydroxide in 300 cm3 of water is added to a solution of 34 g of 2- (N-cyclopropylmethyl-cyanocarbonylamino) -5-chlorobenzophenone in 200 cm3 of ethanol, whereupon the mixture is refluxed for 1 hour. The reaction mixture is extracted with chloroform. The chloroform extracts are combined, washed with water and dried over sodium sulfate and the solvent is removed under reduced pressure. The residue is chromatographed on silica gel and eluted with chloroform, 2-cyclopropylmethylamino-5-chlorobenzophenone, mp 82 to 830 ° C., being obtained. This product is identical to the compound according to section a), as can be demonstrated by the infrared absorption spectrum.
e) To a solution of 17.5 g of 1-ss-cyclohexylethyl) -3-phenyl-5-chloroindole-carboxylic acid ethyl ester in 95 cm3 of glacial acetic acid, a solution of 11.5 g of chromium anhydride in 11.5 cm3 of water is added dropwise at 20 to 25cC added. The mixture is stirred for 30 minutes at room temperature and heated to 50 to 55 ° C. for 5 hours. After cooling, the reaction mixture is poured into 500 cm3 of water and extracted with chloroform. The extracts are combined, washed with water and dried over sodium sulfate, whereupon the solvent is removed under reduced pressure.
In this way, 17.5 g of 2- [N- (ss-cyclohexylethyl) ethoxalylamino] -5-chlorobenzophenone are obtained in the form of an oil.
150 cm3 of concentrated hydrochloric acid are added dropwise to a solution of 17.5 g of the 2- [N- (f3-cyclohexylethyl) ethoxalylamino] -5-chlorobenzophenone in 400 cm3 of ethanol, whereupon the mixture is refluxed for 7 hours heated. The solvent is then removed under reduced pressure. 300 cm3 of cold water are added to the residue and the mixture is neutralized with concentrated ammonium hydroxide and then extracted with ether. The extracts are combined, washed with water and dried over sodium sulfate, whereupon the solvent is removed under reduced pressure. In this way, 10.7 g of 2- (P-cyclohexylethylamino) -5chlorobenzophenone are obtained in the form of a brown oil.
f) A solution of 13.5 g of chromium anhydride in 13.5 cm3 is added to a solution of 17.4 g of l-cyclopropylmethyl-3-phenyl-5-trifluoromethylindole-2-carboxylic acid ethyl ester in 100 cm3 of glacial acetic acid at 20 to 25 C. The mixture is stirred for 30 minutes at room temperature and then heated to 50 to 55 ° C. for 7 hours. After cooling, the reaction mixture is poured into 500 cm3 of water and extracted twice with 150 cm3 of chloroform each time. The combined extracts are washed with water, dried over sodium sulfate and evaporated to dryness in vacuo, 16.4 g of 2- (N-cyclopropylmethylethoxalyl-amino) -5-trifluoromethylbenzophenone being obtained in the form of an oil.
The 2- (N-cyclopropylmethylethoxalylamino) -5 -trifluoromethylbenzophenone obtained in this way is dissolved in 200 cm3 of a 20% aqueous potassium hydroxide solution. The mixture is stirred and heated to 70-80 ° C. for hours, after which it is cooled in an ice bath.
The yellow precipitate is collected by filtration, washed with water and dried to give 7.47 g of 2-cyclopropylmethylamino-5-tritluoromethylbenzophenone, m.p. 102.0-103.50C.
g) A phenyllithium solution is prepared from 11.8 g of bromobenzene and 1.0 g of lithium metal in 40 ml of anhydrous ether and a solution of 5.2 g of 2-cyclopropylmethylamino-5-chlorobenzonitrile in 50 ml of anhydrous ether is added dropwise to this solution while cooling with ice. The resulting reaction mixture is stirred overnight at room temperature and then poured into 200 ml of ice water with stirring. The ether layer is separated off, washed with water and dried over anhydrous sodium sulfate. The solvent is removed under reduced pressure to give 2-cyclopropyl-methylamino-5-chlorobenzophenone imine as a dark brown oil.
h) A mixture of 2.0 g of 2-cyclopropylmethylamino-5-nitrobenzophenone, 20 ml of liquid ammonia and 0.1 g of zinc chloride is heated in a pressure vessel to 1200C for 3 days. The excess ammonia is evaporated and the residue is recrystallized from diisopropyl ether; 2-Cyclopropylmethylamino-5-nitIobenzoph is from PSm. 100-101oC received.
i) To a solution of 3.85 g of 2-cyclopropylmethylamino-5-chlorobenzophenone imine and 12 cm3 of triethylamine in 70 cm3 of benzene, 70 cm3 of a 10% phosgene solution in benzene are added dropwise with cooling. The mixture is stirred for 30 minutes at room temperature and then concentrated to dryness in vacuo.
100 cm3 of dilute aqueous sodium carbonate solution and 100 cm3 of chloroform are added to the residue, and the mixture is stirred. The aqueous layer is extracted with chloroform. The organic layers are combined, washed with water and dried over sodium sulfate. The solvent is removed under reduced pressure and the residue is recrystallized from ethanol, 1 cyclopropylmethyl-4-phenyl-6-chloro-2 (1H) -quinazoline, mp.

A mixture of 2.0 g of 1-cyclopropylmethyl-4-phenyl-6-nitro-2 (1H) -quinazolinone, 6 g of phosphorus pentasulfide and 40 ml of pyridine is refluxed for 20 hours.
After cooling, the reaction mixture is poured into 200 ml of water and the mixture obtained is extracted with chloroform. The chloroform layer is washed successively with dilute hydrochloric acid and water, and then dried over anhydrous sodium sulfate. The chloroform is evaporated under reduced pressure and the residue is chromatographed on a silica gel column; eluting with chloroform gives 0.1 g of 1-cyclopropylmethyl-4-phenyl-6-nitro-2 (1H) -quinazolmthione. Recrystallization from ethanol and chloroform gives platelets of dark orange color and melting point 205-2060C.
Example 2
A mixture of 3.4 g of 1-cyclopropylmethyl-4-phenyl-6-chloro-2 (1H) -quinazolinone, 11 g of phosphorus pentasulfide and 40 ml of pyridine is refluxed for 17 hours. The further elaboration is carried out analogously to Example 1. 3.3 g of 1-cyclopropylmethyl-4-phenyl-6-chloro-2 (1H) -quinazolinethione are obtained in the form of orange needles with a melting point of 232-2340C.
