Die Erfindung betrifft ein Verfahren zur Herstellung von 7a-Methyl-östradiol-Derivaten der Formel

in der R1 Wasserstoff, einen niederen Alkylrest oder einen gesättigten, ausschliesslich Sauerstoff als Heteroatome enthaltenden heterocyclischen Rest oder einen physiologisch unbedenklichen Acylrest bedeutet, das dadurch gekennzeichnet ist, dass man in entsprechenden Östron-Derivaten die 17 Oxogruppe reduziert.
In den erhaltenen Verbindungen der Formel I können freie Hydroxylgruppen mit physiologisch unbedenklichen Siren verestert oder veresterte Hydroxylgruppen verseift werden. Auch kann man in erhaltenen neuen Verbindungen der Formel I freie OH-Gruppen verithern.
Als physiologisch unbedenkliche Esterreste kommen vor allem die Acylreste solcher Siren in Frage, die in der Steroidchemie üblicherweise zur Veresterung freier Hydroxylgruppen venvendet werden; beispielsweise genannt seien Essigsiure, Propionsiure, Hydroxy- und Chlorpropionsäuren, Cyclopentylpropionsäure, Capronsäure, Önanthsäure, Undecyl sure, Benzoesäure, Mono- und Dichloressigsiure, Trimethylessigsäure, Bernsteinsäure u.a.
Im Hinblick auf die pharmazeutische Anwendung der Verfahrensprodukte sind als Ätherreste vorzugsweise Methyl-, Äthyl- und Tetrahydropyranylreste geeignet.
Die Anlagerung von Wasserstoff an die 17ständige Carbonylgruppe kann durch Hydrieren in Gegenwart eines üblichen Katalysators, welcher in Gegenwart von Wasserstoff die Reduktion von Fiinfringketonen bewirkt, erfolgen. Ausserdem kann der Wasserstoff aus Metallhydriden auf das 17-Keton übertragen werden. Als Wasserstoffdonatoren haben sich insbesondere gemischte Metallhydride wie Natriumborhydrid, Lithiumaluminiumhydrid und Lithium-tritert-butoxy-aluminiumhydrid bewährt.
Geeignete Lösungsmittel sind zum Beispiel Ather, wie Diäthyläther Tetrahydrofuran und Dioxan, oder Kohlenwasserstoffe, wie Benzol und Toluol. Als tertiäre Alkohole kommen beispielsweise tert.-Butyl- und tert.-Amylalkohol in Frage.
Da das 1 -Hydroxy-7a -methyl-6stron als Resorzinderivat im alkalischen Milieu eine nicht sehr bestindige Verbindung ist, kann es zweckmässig sein, von solchen Verbindungen auszugehen, bei denen die Hydroxygruppen in 1- und 3-Stellung verestert oder veräthert sind. Wenn beispielsweise die Atherreste nur als intermediäre Schutzgruppen eingefuhrt werden sollen, ist es vorteilhaft, mit Dihydropyran zu veräthern, weil diese Reste nach erfolgter Umsetzung besonders leicht abspaltbar sind.
1,3-Diester werden bei der Umsetzung an der 17-Ketogruppe leicht angegriffen.
Die Reduktion kann auch so ausgefiihrt werden, dass im Ausgangsprodukt vorhandene Estergruppen erhalten bleiben.
Die Acylierung in 1- und 3-Stellung erfolgt vorzugsweise mit Pyridin/Siureanhydrid bei Raumtemperatur. Zur Ver ätherung in 1- und 3-Stellung dienen vor allem alkylierende Verbindungen, wie vorzugsweise Diazomethan und Dialkylsulfate.
Zur Veresterung der,17fi-Hydroxygruppe in den erhaltenen 1,3-Diestern und 1,3-Diithern lässt man auf das Steroid beispielsweise Siureanhydride in Gegenwart starker Siren, wie p-Toluolsulfonsiure, oder Pyridin/Säureanhydrid in der Wärme einwirken. Die letztgenannten Methoden kinnen auch benutzt werden, um die freie Trihydroxy-Verbindung unmittelbar in das Triacylat zu überführen. Aus den Triacylaten kdnnen durch schonende partielle Verseifung die 1- und die 3-OH-Gruppe freigesetzt werden.
1,3-Diester und 1,3-Diither können mit Dihydropyran in Gegenwart einer starken Siure, wie p-Toluolsulfonsäure, in die entsprechenden 17-Tetrahydropyranyläther iberfihrt werden. Die Veritherung der 17-OH-Gruppe in den erfin dungsgemiss herstellbaren 1,3-Diithern mit einem Alkylrest wird vorzugsweise mit Alkylhalogeniden in flüssigem Ammoniak durchgefiihrt. Die beiden zuletzt genannten Methoden ermoglichen es auch, alle drei OH-Gruppen der Trihydroxy Verbindung in einem Arbeitsgang zu veräthern.
Aus 1,3-Diacyl-17-tetrahydropyranyl-Derivaten können durch alkalische Verseifung die 1- und die 3-OH-Gruppe freigesetzt werden.
Will man zu 1,3,17ss-Trihydroxy-Verbindungen gelangen, deren 17-OH-Gruppe alkyliert oder acyliert ist, so ist es besonders vorteilhaft, das 1-Hydroxy-östron mit Dihydropyran zu veräthern, nach der Reduktion in die erhaltene 17ss-OH- Gruppe den gewünschten Alkyl- oder Acylrest einzuffihren und anschliessend die Tetrahydropyranylgruppen aus 1,3 Stellung selektiv abzuspalten. Nachtriglich können die erhaltenen 1,3-Dihydroxy-Verbindungen in gewiinschter Weise veräthert oder verestert werden.
Die Verfahrensprodukte besitzen wertvolle therapeutische Eigenschaften. Sie zeichnen sich beispielsweise durch uber- raschend starke östrogene und ovulationshemmende Wirksamkeiten aus. Die Verbindungen kdnnen peroral und auch parenteral appliziert werden.
Die folgende Tabelle zeigt die hohe flberlegenheit der neuen Verbindungen gegenüber Östradiol-3-acetat (II) bei oraler Applikation im Tierversuch. Die östrogene Wirkung wurde an kastrierten Rattenweibchen im Allen-Doisy-Test bestimmt. Die ovulationshemmende Wirkung wurde an normalen Rattenweibchen durch Tubeninspektion ermittelt, als WD50 wird diejenige Tagesdosis angegeben, die bei 50% der Tiere die Ovulation unterdriickt.

Ferner besitzen die neuen Verbindungen die Eigenschaft, die Implantation (Nidation) zu hemmen. Die stattgefundene oder unterbliebene Implantation von Blastozysten wurde bei Ratten durch Uterusinspektion ermittelt. Gravide Ratten erhielten vom 1. bis 7. Tag der Graviditit täglich die zu testende Substanz in bestimmten Dosen p. o. Am 9. Tag wurden die Tiere getötet. Es wurde die Zahl der Tiere festgestellt, bei denen keine Implantation stattgefunden hatte.
Zum therapeutischen Gebrauch können die neuen Ostratriolderivate mit den in der galenischen Pharmazie üblichen Zusätzen, Trägersubstanzen und Geschmackskorrigentien nach an sich bekannten Methoden zu den üblichen Arzneimittelformen verarbeitet werden.
Indikationsgebiete der Arzneimittel auf Basis der erfin dungsgemäss herstellbaren Verbindungen sind Krankheitserscheinungen, bei denen eine Behandlung mit Östrogen, gegebenenfalls in Kombination mit gestagen wirksamen Substanzen, angezeigt ist. Beispielsweise genannt seien das Klimakterium und lessen Folgeerkrankungen, Mammakarzi nom nach der Menopause, Prostatakarzinom, periphere Durchblutungsstörungen, Amenorrhoe; insbesondere in Kombination mit beispielsweise Norethisteronacetat sind die erfindungsgemäss erhältlichen Verbidungen auch zur Ruhigstellung des Ovars geeignet.
Das als Ausgangsmaterial des vorliegenden Verfahrens dienende und in der Literatur noch nicht beschriebene 1-Hydroxy-7a-methylöstron wird beispielsweise wie folgt hergestellt:
Zur Suspension von 42 g 7a -Methyl5stron (hergestellt durch Hydrolyse des 3-Tetrahydropyranylithers) in 600 ml Eisessig gibt man 120 g Bleitetraacetat und rührt 16 Stunden bei Raumtemperatur unter Feuchtigkeitsabschluss. Danach gibt man den Ansatz in 600 ml Eiswasser, saugt den Niederschlag ab, wäscht mit Wasser und nimmt den Filterriickstand in Methylenchlorid auf. Die Substanzlösung wird mit Natriumbicarbonatlösung und Wasser neutral gewaschen, getrocknet und eingeengt. Das Konzentrat filtriert man mit Methylenchlorid iiber 400 g Silicagel (+ 10% Wasser).
Die substanzhaltigen Fraktionen werden vereinigt und vom Lösungsmittel befreit. Man erhält 7 g 7α-Methyl-#1,4-östra- dien-10P-ol-3,17-dion-acetat, das nach Umkristallisieren aus Diäthyläther bei 198-199 C schmilzt.

Eine Suspension von 13,0 g 7a-Methyl-A 16-6stradien- 10P-ol-3,17-dion-acetat in 125 ml Essigsäureanhydrid wird tropfenweise mit 0,7 ml konzentrierter Schwefelsäure versetzt und 3 Stunden bei Raumtemperatur geriihrt, wobei die Substanz langsam in Lösung geht. Dann gibt man den Ansatz in die 10fache Menge Eiswasser, dem man 7 g Natriumcarbonat zusetzt, riihrt eine Stunde und filtriert ab. Der gewaschene und getrocknete Rtickstand wird aus Methanol umkristallisiert. Man erhält 7 g 7α-Methyl-#1,3,5(10)-östratrien-1,3- diol-17-on-1,3-diacetat, das nach Umkristallisieren aus Methylenchlorid/Hexan bei 166-166,5 C schmilzt.

Beispiel 1
Zur Lösung von 2,7 g 7α-Methyl-#1,3,5(10)-östratrien- 1,3-diol-17-on-1,3-diacetat in 60 ml absolutem Tetrahydrofuran gibt man unter Ruhren und Aussenkühlung mit Eis 5,5 g Lithiumtritert. -butoxy-aluminiumhydrid. Man lässt eine halbe Stunde reagieren bei 0 C und gibt danach vorsichtig 13 ml Eisessig hinzu. Man verdiinnt mit anther, schiittelt die organische Phase mit eiskalter, verdiinnter Salzsäure und wäscht danach mit Natriumbicarbonatlösung und Wasser neutral.
Nach Trocknen tiber Natriumsulfat und Abdampfen des Lösungsmittels erhält man 2 g 7α-Methyl-#1,3,5(10)-östra- trien-1,3,17ss-triol-1,3-diacetat, das nach Umkristallisieren aus Hexan/Diäthyläther bei 160-162 C schmilzt.

Beispiel 2
1 g 7a -Methyl-A 1,3,5(10) Bstratrien-1,3 -diol-17-on-1,3- diacetat werden in 25 ml Eisessig mit 20 mg Platinoxyd bei Normaldruck und Raumtemperatur bis zur Aufnahme von 1,1 Mol-Aquivalent Wasserstoff hydnert, danach wird vom Katalysator abfiltrielt und die Ldsung am Rotationsverdampfer zur Trockne eingedampft. Der Ruckstand wird in Methylenchlorid aufgenommen, mit Natriumbicarbonatlösung und Wasser neutral gewaschen, getrocknet und eingedampft. Man erhält 900 mg 7α-Methyl-#1,3,5(10)-östratrien-1,3,17ss-triol-1,3- diacetat, das nach Umkristallisieren aus Hexan/Diäthyläther bei 160-162 C schmilzt.

Beispiel 3
Die Ldsung von gemäss Beispiel 2 erhaltenem 1,5 g 7a Methyl-#1,3,5(10)-östratrien-1,3,17ss-triol-1,3-diacetat in 70 ml Benzol wird durch Abdestillieren von 20 ml Lösungs- mittel azeotrop getrocknet. In gleicher Weise wird die Lösungs von 15 mg p-Toluolsulfonsäure in 20 ml Benzol von Feuchtigkeit befreit. Nach Abkuhlung auf 0 ' C werden beide Lösun- gen vereinigt, mit 1 ml Dihydropyran versetzt und 1,5 Stunden geriihrt. Danach wird mit Natriumbicarbonat und Wasser gewaschen, getrocknet und eingedampft. Man erhält 1,4 g 7a -Methyl-A 1'3'5(10) -östratrien-1,3,17ss-triol-1,3 -diacetat 17ss-tetrahydropyranyläther als chromatographisch einheitliches C)l.

Beispiel 4
1,5 g 7a -Methyl-#1,3,5(10)-östratrien-1,3,17ss-triol- 1,3-diacetat werden in 5 ml Pyridin gelöst, mit 5 ml Essigsäureanhydrid versetzt und eine Stunde auf dem Dampfbad erwärmt. Dansch gibt man den Ansatz in die 10fache Menge Eiswasser, filtriert den Niederschlag ab und trocknet im Vakuum. Man erhilt 1,4 g 7a-Methyl-A 1,3,5(10)-östratrien- 1,3,17ss-triol-triacetat als chromatographisch einheitliches Öl.
