In dieser Beschreibung soll die Bezeichnung nieder eine geradkettige oder verzweigte Kohlenstoffkette mit 1 bis 6 Kohlenstoffatomen bedeuten, während die Bezeichnung Alkyl Methyl, Äthyl, Propyl, Isopropyl, Butyl, Isobutyl, tert.-Butyl, Pentyl, Hexyl etc. und Halogen Fluor, Chlor, Brom oder Jod bedeuten.
Die vorliegende Erfindung bezieht sich somit auf ein neues Verfahren für die Herstellung von Verbindungen der Formel I unter Verwendung eines neuen Zwischenproduktes, nämlich der Verbindung der Formel IV.
Die Verbindungen der Formel I sind bekannt und lassen sich beispielsweise gemäss jap. Auslegeschrift Nr. 17 015/ 1965 und Schweiz. Patent. Nr. 478 749 herstellen. Unter die Verbindungen der Formeln fallen die 1,2-Dialkyl-3-diphenyl- methylenpyrrolidine, welche als solche eine atropinähnliche Wirkung ausüben und ebenfalls als Zwischenprodukte für die Herstellung eines antispasmodischen Mittels, nämlich 1,1,2 -Trialkyl-3 - diphenylmethylenpyrrolidiniumhalogenid, wertvoll sind, während 1,2-Dialkyl-3-(10,- 11 -dihydro-SH-dibenzo- -[a,d]-cyclohepten- oder -octen-5-yliden)-pyrrolidine bekanntlich als Mittel gegen die Parkinson'sche Krankheit wertvoll sind.
Bisher ist bekannt geworden, dass Verbindungen der Formel I beispielsweise dadurch erhalten werden können, dass man 3-Diphenylmethylen-l-pyrrolin mit einem Alkylierungsmittel umsetzt und hierauf das so erhaltene Produkt reduziert (vergleiche japanische Patentanmeldung Nr. 17 015/ 1965) oder indem man ein Immoniumsalz von 3-(10,11-dihydro-5H-dibenzo-[a,d]-cyclohepten- oder -octen-5-yliden)- 1 - -pyrrolin reduziert (vergl. Schweiz. Patent Nr. 478-749). Im Vergleich zu diesen bekannten Methoden besitzt die neue Synthese unter Anwendung des vorliegenden Verfahrens Vorteile insofern, als der Arbeitsvorgang durch die Hydrolysenreaktion und durch die Dehydratationsreaktion, welche nacheinander durchgeführt werden, vereinfacht wird, während überdies die Gesamtausbeute höher ist.
Ferner lassen sich die eingesetzten Verbindungen der Formeln II und III leicht aus dem Reaktionsgemisch in reinem Zustande zurückgewinnen, worauf man sie für die spätere Reaktionsstufe verwenden kann.
Zuest wird also das Ausgangsprodukt IV so gewonnen, dass man ein Methanderivat der Formel II mit einem 3-Pyrrolidinon der Formel III in Gegenwart eines alkalischen Kondensationsmittels oder, sofern X in der Verbindung II ein Halogenatom darstellt, in Gegenwart von Magnesium umsetzt. Als alkalisches Kondensationsmittel kann man beispielsweise ein Alkalimetall, z.B. Lithium, Natrium, Kalium usw., oder eine Alkalimetallverbindung der Formel R4-X3, worin R4 ein Alkalimetall und X3 Wasserstoff, ein Amin, einen Alkylrest oder einen Arylrest, beispielsweise einen Phenyl-, Tolyl-, Xylyl-, Naphthylrest usw., od. einen Aralkylrest, beispielsweise einen Benzyl-, Tolylmethyl-, Xylylmethyl-, Phenyläthyl-, 3-Phenylpropylrest usw., bedeuten, verwenden.
Diese alkalischen Kondensationsmittel werden je nach der zu verwendenden Methanverbindung der Formel II ausgewählt. Bei dieser Umsetzung verwendet man ein Lösungsmittel, wie z.B. Tetrahydrofuran, Äther, n-Hexan, Dioxan, oder ein anderes beliebiges Lösungsmittel, welches während der Reaktion inert ist. Die Umsetzung erfolgt im allgemeinen bei Temperaturen, welche zwischen einer niedrigen Temperatur und ungefähr 1000C liegen, doch wird man bei möglichst niedriger Temperatur arbeiten.
Bei dieser Umsetzung wird man das Alkalimetall vorzugsweise im Überschuss verwenden, wenn die Verbindung der Formel II, worin X ein Halogenatom darstellt, mit dem Alkalimetall umgesetzt wird, wobei man die Umsetzung ebenfalls bei einer niedrigen Temperatur durchführen wird, um Nebenreaktionen zu vermeiden, wie z.B. ein Ersatz des Halogenatoms durch den Rest X3, welcher ein Alkyl- oder Aralkykest wäre. Dies geschieht auf Wegen einer eventuellen Selbstkondensation.
Das so erhaltene Reaktionsprodukt wird dann hydrolysiert. Die Hydrolyse erfolgt leicht durch Zugabe von Wasser und kann durch die Anwesenheit einer sauren oder basischen Substanz beschleunigt werden. Die auf diese Weise erhaltenen Verbindungen der Formel IV, nämlich 1,2-Di-nieder -alkyl-3-pyrrolidinole, sind neue Verbindungen und können gewünschtenfalls in Kristallform aus dem Reaktionsgemisch isoliert werden. Man kann sie aber auch ohne Isolierung für das anschliessende Verfahren, nämlich der Dehydratationsstufe, verwenden.
Die Dehydratationsreaktion erfolgt vorzugsweise durch Erhitzen und/oder durch Behandeln von Verbindungen der Formel IV mit einem Katalysator, wie z.B. Aluminiumoxyd, Aluminiumsalzen, wie z.B. Aluminiumchlorid usw., oder mit einem Dehydratationsmittel, wie z.B. Salzsäure, Schwefelsäure, einer aromatischen Sulfonsäure, Phosphorpentoxyd, Phosphorsäure, Metaphosphorsäure, Oxalsäure, Ameisensäure, einem Anhydrid einer organischen Säure, einem anorganischen oder organischen Säurechlorid, Phosphoroxychlorid, einem wasserfreien anorganischen Salz usw. Die Reaktions- temperatur hängt vom zu verwendenden Mittel ab.
Der Reaktionsmechanismus lässt sich wie folgt beschreiben. Es wird vorausgesetzt, dass die Gruppe X Wasserstoff oder Halogen im Methanderivat der Formel II darstellt. In diesem Falle wird diese Gruppe durch das Alkalimetall im alkalischen Kondensationsmittel ersetzt, wobei das erhaltene Produkt mit den 3-Pyrrolidinonverbindungen der Formel III reagiert, wobei das in der Verbindung II eingeführte Metall eine Kombination mit dem Sauerstoffatom in den Verbindungen der Formel III eingeht, wodurch eine Metall-O-Bindung einerseits entsteht und andererseits das Kohlenstoffatom im Methananteil der Verbindungen der Formel II sich mit dem Kohlenstoffatom, das sich in der 3-Stellung des Pyrrolidinringes der Verbindungen III befindet, vereinigt, um eine C-C -Bindung einzugehen,
worauf das Metall im erhaltenen Produkt in Freiheit gesetzt und durch Hydrolyse durch ein Wasserstoffatom ersetzt wird, wobei man Verbindungen der Formel IV erhält. Hierauf werden die Verbindungen der Formel IV dehydratisiert, worauf man die gewünschten Verbindungen der Formel I erhält. Im Falle einer Umsetzung einer Verbindung der Formel II, bei welcher X ein Halogenatom darstellt, mit Magnesium, erfolgt die Umsetzung nach einem ähnlichen Reaktionsmechanismus, wie jener der oben beschrieben worden ist.
In diesem Falle wird zuerst eine halogenierte Magnesiumgruppe erzeugt, indem Magnesium as das Methanderivat der Formel II gebunden wird, worauf diese Gruppe unter Verbindung mit dem Sauerstoffatom in den 3-Pyrrolidinonverbindungen der Formel III in eine halogenierte Magnesium-O-Gruppe übergeht, während eine C-C-Bindung zwischen dem Kohlenstoffatom im Methananteil der Verbindung II und dem Kohlenstoffatom in der 3-Stellung des Pyrrolidinringes der Verbindungen der Formel III gebildet wird, worauf der halogenierte Magnesiumteil in der halogenierten Magnesium-O-Gruppe in Freiheit gesetzt wird und durch Hydrolyse ein Wasserstoffatom aufgenommen wird, wobei Verbindungen der Formel IV erhalten werden. Hierauf werden die Verbindungen der Formel IV dehydratisiert"wodurch man die Verbindungen der Formel I erhält.
Die Erfindung sei durch die folgenden Beispiele erläutert.
Beispiel I a) Herstellung des Ausgangsproduktes
Zu einer Lösung von 1,68 g Diphenylmethan in 5 cm3 Tetrahydrofuran gibt man tropfenweise 11 cm3 einer 15% eigen Hexanlösung von Butyllithium innerhalb von 10 Minuten in einer Stickstoffgasatmosphäre bei einer Temperatur im Bereiche von -20 bis -25"C hinzu. Nach einstündigem Rühren versetzt man das Gemisch bei niedriger Temperatur mit 1,27 g 1-Äthyl-2-methyl-3-pyrrolidinon, worauf man das Reaktionsgemisch während 2 Stunden rührt. Dann wird das Reaktionsgemisch mit weiteren 15 cm3 Äther versetzt und die gesamte Lösung in kaltes Wasser gegossen, wodurch Hydrolyse eintritt.
Die Ätherschicht wird mit Wasser gewaschen und getrocknet und das Lösungsmittel durch Destillation entfernt, worauf man 1,5 g l-Äthyl-2-methyl-3-diphenylmethyl -3-pyrrolidinol in Form von Kristallen vom Smp. 135 bis 137"C erhält. 0,51 g nicht umgesetztes Diphenylmethan und 0,3 g der 3-Pyrrolidinonverbindung, welche als Ausgangsmaterial verwendet werden, werden aus der Reaktionsflüssigkeit zurückgewonnen.
b) 100 mg Kristalle der 3-Pyrrolidinolverbindung, wie sie oben erhalten worden ist, wird zu 2 cm3 60%iger Schwefelsäure hinzugegeben, worauf man das Gemisch auf dem siedenden Wasserbade während 1 Stunde erwärmt. Nach dem Kühlen wird das Reaktionsgemisch mittels Natriumhydroxyd neutralisiert und hierauf mit Äther extrahiert. Der Äther wird aus dem Extrakt durch Destillation entfernt, wobei man 80 mg 1 -Äthyl-2-methyl-3 -diphenyl-methylenpyrrolidin in Form einer öligen Substanz in einer Ausbeute von 85% erhält.
Diese Substanz ist völlig identisch mit dem in üblicher Weise durch Reduktion von l-Äthyl-2-methyl-3- diphenyl- methylen-l-pyrroliniumjodid erhaltenen l-Äthyl-2-methyl-3 -diphenylmethylenpyrrolidin, wie dies durch das Infrarotabsorptionsspektrum und durch Dünnschichtchromatographie feststellbar ist.
Beispiel 2 a) Herstellung des Ausgangsproduktes
Zu einer Lösung von 9,7 g 10,11-Dihydro-5H-dibenzo -[a,d]-cyclohepten in 50 cm3 Tetrahydrofuran -gibt man tropfenweise 50 cm3 einer 15%igen Hexanlösung von Butyllithium in einer Stickstoffgasatmosphäre bei einer Temperatur von -200C hinzu. Nach dem Rühren versetzt man das Gemisch mit 6,4 g 1-Äthyl-2-methyl-3-pyrrolidinon, worauf man das Reaktionsgemisch während 2 Stunden bei Zimmertemperatur rührt. Dann wird das Reaktionsgemisch mit 100 cm3 Äther versetzt und die gesamte Lösung in kaltes Wasser gegossen, um die Hydrolyse durchzuführen. Die Ätherschicht wird mit Wasser gewaschen und getrocknet und das Lösungsmittel durch Destillation entfernt, wobei man l-Äthyl-2-methyl-3-(10,1 l-dihydro-SH-dibenzo-[a,d]-cyclo- hepten-5-yl)-3-pyrrolidinol in Form von Kristallen vom Smp.
124 bis 1260C erhält. 3,2 g nicht umgesetztes l0,ll-Dihydro- -SH-dibenzo-[a,d]-cyclohepten und 1 g 3-Pyrrolidinonverbindung, welche als Ausgangsmaterialien verwendet worden sind, werden aus der Reaktionsflüssigkeit zurückgewonnen.
b) 100 mg der erhaltenen Kristalle, bestehend aus der 3-Pyrrolidinolverbindung, werden einer Mischung von 200 mg p-Toluolsulfonsäure und 3 cm3 absolutem Toluol hinzugegeben, worauf man das Gemisch während 17 Std. unter Rückfluss erhitzt. Nach dem Kühlen versetzt man das Reaktionsgemisch mit.l0 cm3 Äther, worauf man die Lösung mit einer 10%igen Natriumhydroxydlösung 2mal und hierauf mit Wasser wäscht und anschliessend über Magnesiumsulfat trocknet. Das Lösungsmittel wird abdestilliert, wobei man 90 mg l-Äthyl-2-methyl-3-(10,11-dl'hydro-5H-dibenzo-[a,dl- -cyclohepten-5-yliden)-pyrrolidin in Form einer öligen Substanz in einer Ausbeute von 95% erhält.
Diese Substanz ist identisch mit dem üblicherweise erhaltenen l-Äthyl-2-methyl-3-(10,1 1 -dihydro-SH-dibenzo-[a,d]- -cyclohepten-5-yliden)-pyrrolidin, das man durch Reduktion von 1 -Äthyl-2-methyl-3-(10,11 -dihydro- 5H-dibenzo [a,d]- -cyclohepten-5-yliden)- 1 -pyrroliniumjodid erhalten hat, wie dies sowohl durch das Infrarotabsorptionsspektrum als auch durch Dünnschichtchromatographie feststellbar ist.

At first, the starting product IV is obtained by reacting a methane derivative of the formula II with a 3-pyrrolidinone of the formula III in the presence of an alkaline condensing agent or, if X in the compound II represents a halogen atom, in the presence of magnesium. As the alkaline condensing agent, for example an alkali metal, e.g. Lithium, sodium, potassium, etc., or an alkali metal compound of the formula R4-X3, wherein R4 is an alkali metal and X3 is hydrogen, an amine, an alkyl radical or an aryl radical, for example a phenyl, tolyl, xylyl, naphthyl radical, etc., or an aralkyl radical, for example a benzyl, tolylmethyl, xylylmethyl, phenylethyl, 3-phenylpropyl radical, etc., use.
These alkaline condensing agents are selected depending on the methane compound of the formula II to be used. A solvent such as e.g. Tetrahydrofuran, ether, n-hexane, dioxane, or any other solvent which is inert during the reaction. The reaction is generally carried out at temperatures which are between a low temperature and about 1000 ° C., but the temperature will be as low as possible.
In this reaction, the alkali metal will preferably be used in excess if the compound of the formula II, in which X is a halogen atom, is reacted with the alkali metal, the reaction also being carried out at a low temperature in order to avoid side reactions, e.g. a replacement of the halogen atom by the radical X3, which would be an alkyl or arykyl radical. This happens because of a possible self-condensation.
The reaction product thus obtained is then hydrolyzed. The hydrolysis occurs easily by adding water and can be accelerated by the presence of an acidic or basic substance. The compounds of the formula IV obtained in this way, namely 1,2-di-lower-alkyl-3-pyrrolidinols, are new compounds and, if desired, can be isolated from the reaction mixture in crystal form. However, they can also be used for the subsequent process, namely the dehydration stage, without isolation.
The dehydration reaction is preferably carried out by heating and / or by treating compounds of formula IV with a catalyst, e.g. Aluminum oxide, aluminum salts, e.g. Aluminum chloride, etc., or with a dehydrating agent such as e.g. Hydrochloric acid, sulfuric acid, an aromatic sulfonic acid, phosphorus pentoxide, phosphoric acid, metaphosphoric acid, oxalic acid, formic acid, an anhydride of an organic acid, an inorganic or organic acid chloride, phosphorus oxychloride, an anhydrous inorganic salt, etc. The reaction temperature depends on the agent to be used.
The reaction mechanism can be described as follows. It is assumed that the group X represents hydrogen or halogen in the methane derivative of the formula II. In this case, this group is replaced by the alkali metal in the alkaline condensing agent, the product obtained reacting with the 3-pyrrolidinone compounds of the formula III, the metal introduced in the compound II entering into a combination with the oxygen atom in the compounds of the formula III, whereby a metal-O bond is formed on the one hand and on the other hand the carbon atom in the methane portion of the compounds of the formula II is combined with the carbon atom which is in the 3-position of the pyrrolidine ring of the compounds III to form a CC bond,
whereupon the metal in the product obtained is set free and replaced by a hydrogen atom by hydrolysis, compounds of the formula IV being obtained. The compounds of the formula IV are then dehydrated, whereupon the desired compounds of the formula I are obtained. In the case of a reaction of a compound of the formula II, in which X represents a halogen atom, with magnesium, the reaction takes place according to a reaction mechanism similar to that described above.
In this case, a halogenated magnesium group is first generated by bonding magnesium as the methane derivative of the formula II, whereupon this group, in connection with the oxygen atom in the 3-pyrrolidinone compounds of the formula III, changes into a halogenated magnesium-O group, while a CC -Bond is formed between the carbon atom in the methane portion of the compound II and the carbon atom in the 3-position of the pyrrolidine ring of the compounds of the formula III, whereupon the halogenated magnesium part in the halogenated magnesium-O group is set free and a hydrogen atom is added by hydrolysis , whereby compounds of the formula IV are obtained. The compounds of the formula IV are then "dehydrated", whereby the compounds of the formula I are obtained.
The invention is illustrated by the following examples.
Example I a) Production of the starting product
To a solution of 1.68 g of diphenylmethane in 5 cm3 of tetrahydrofuran is added dropwise 11 cm3 of a 15% own hexane solution of butyllithium within 10 minutes in a nitrogen gas atmosphere at a temperature in the range from -20 to -25 ° C. After stirring for one hour 1.27 g of 1-ethyl-2-methyl-3-pyrrolidinone are added to the mixture at low temperature, whereupon the reaction mixture is stirred for 2 hours, a further 15 cm3 of ether are added to the reaction mixture and the entire solution is poured into cold water poured, causing hydrolysis to occur.
The ether layer is washed with water and dried and the solvent is removed by distillation, whereupon 1.5 g of 1-ethyl-2-methyl-3-diphenylmethyl -3-pyrrolidinol are obtained in the form of crystals with a melting point of 135 to 137 ° C. 0.51 g of unreacted diphenylmethane and 0.3 g of the 3-pyrrolidinone compound, which are used as the starting material, are recovered from the reaction liquid.
b) 100 mg of crystals of the 3-pyrrolidinol compound as obtained above are added to 2 cm3 of 60% strength sulfuric acid, whereupon the mixture is heated on a boiling water bath for 1 hour. After cooling, the reaction mixture is neutralized with sodium hydroxide and then extracted with ether. The ether is removed from the extract by distillation, 80 mg of 1-ethyl-2-methyl-3-diphenyl-methylenepyrrolidine being obtained in the form of an oily substance in a yield of 85%.
This substance is completely identical to the 1-ethyl-2-methyl-3-diphenylmethylene pyrrolidine obtained in the usual way by reducing l-ethyl-2-methyl-3-diphenyl-methylene-l-pyrrolinium iodide, as indicated by the infrared absorption spectrum and by Thin layer chromatography can be determined.
