Das erfindungsgemässe Verfahren ist dadurch gekennzeichnet, dass man in Benzolsulfonylharnstoffe der Formel 2

durch Acylierung den Rest X-CO- einführt oder entsprechend substituierte Benzolsulfonylhalogenide mit R1-substituierten Harnstoffen oder deren Alkalisalzen umsetzt und die Reaktionsprodukte gegebenenfalls zur Salzbildung mit alkalischen Mitteln behandelt.
Die Umsetzung der Benzolsulfohalogenide mit R1-substituierten Harnstoffen erfolgt zweckmässig unter Verwendung von stark basischen Kondensationsmitteln, wie Alkalimetallen, Alkaliamiden oder vorzugsweise -hydriden in indifferenten Lösungsmitteln.
Je nach der Natur der Ausgangsmaterialien insbesondere des Gliedes X wird in einzelnen Fällen das eine oder andere der genannten Verfahren für die Herstellung der unter die allgemeine Formel fallenden individuellen Verbindungen ungeeignet sein oder zumindest Vorkehrungen zum Schutz aktiver Gruppen notwendig machen. Derartige verhältnismässig selten auftretende Fälle können vom Fachmann unschwer erkannt werden, und es bereitet keine Schwierigkeiten, in solchen Fällen die entsprechenden Massnahmen zu ergreifen.
Die Ausführungsformen des Verfahrens gemäss der Erfindung können im allgemeinen hinsichtlich der Reaktionsbedingungen weitgehend variiert und den jeweiligen Verhältnissen angepasst werden. Beispielsweise können die Umsetzungen in Abwesenheit oder Anwesenheit von Lösungsmitteln, bei Zimmertemperatur oder bei erhöhter Temperatur durchgeführt werden.
Die blutzuckersenkende Wirkung der beschriebenen Benzolsulfonylharnstoffderivate konnte dadurch festgestellt werden, dass man sie, z. B. in Form des Natriumsalzes, in Dosen von 10 mg/kg an normal ernährte Kaninchen verfütterte und den Blutzuckerwert nach der bekannten Methode von Hagedorn Jensen oder mit einem Autoanalyzer über eine längere Zeitdauer ermittelte.
So wurde beispielsweise ermittelt, dass 10 mg/kg N- [4-(ss- < 2-Methoxy-5-methyl-benzamido > -äthyl)-benzol- sulfonyl]-N'-(4,7-endomethylen-perhydroindanyl-5-)harnstoff nach 3 Stunden eine Blutzuckersenkung von 24% bewirkt. Gleichermassen bewirken 10 mg N-[4-(ss-(2-Meth- oxy-5-chlorbenzamido ) -äthyl)-benzolsulfonyl]-N'-(4,7- endomethylen-perhydroindanyl-5) -harnstoff nach 6 Stunden eine Blutzuckersenkung von 23 %, während der bekannte N-[4-Methyl-benzolsulfonyl]-N'-butylharnstoff bei einer Dosierung von weniger als 25 mg/kg am Kaninchen keine Senkung des Blutzuckerspiegels mehr hervorruft.
Ferner wurde ermittelt, dass 10 mg N-[4-(ss-(2-Meth- oxy-benzamido > -äthyl)-benzolsulfonyl]-N'-(4,7-endomethy len-perhydroindanyl-5)-harnstoff nach 3 Stunden eine Blutzuckersenkung von 40% bewirken.
Die Verfahrensprodukte besitzen somit eine sehr starke blutzuckersenkende Wirksamkeit bei einer ausserordentlich guten Verträglichkeit.
Die beschriebenen Benzolsulfonylharnstoffe sollen vorzugsweise zur Herstellung von oral verabreichbaren Präparaten mit blutzuckersenkender Wirksamkeit zur Behandlung des Diabetes mellitus dienen und können als solche oder in Form ihrer Salze bzw. in Gegenwart von Stoffen, die zu einer Salzbildung führen, appliziert werden. Zur Salzbildung können beispielsweise alkalische Mittel wie Alkali- oder Erdalkalihydroxyde, -carbonate oder -bicarbonate herangezogen werden.
Als medizinische Präparate kommen vorzugsweise Tabletten in Betracht, die neben den Verfahrenserzeugnissen die üblichen Hilfs- und Trägerstoffe wie Talkum, Stärke, Milchzucker, Tragant oder Magnesiumstearat enthalten.
Ein Präparat, das die beschriebenen Benzolsulfonylharnstoffe als Wirkstoff enthält, z. B. eine Tablette oder ein Pulver mit oder ohne die genannten Zusätze, ist zweckmässig in eine geeignet dosierte Form gebracht. Als Dosis ist dabei eine solche zu wählen, die der Wirksamkeit des verwendeten Benzolsulfonylharnstoffs und dem gewünschten Effekt angepasst ist. Zweckmässig beträgt die Dosierung je Einheit etwa 0,5 bis 100 mg, vorzugsweise 2 bis 10 mg, jedoch können auch erheblich darüber oder darunter liegende Dosierungseinheiten verwendet werden, die gegebenenfalls vor Applikation zu teilen bzw. zu vervielfachen sind.
Beispiel 1
N-[4-(ss- (2-Methoxy-5-chlor-benzamido)-äthyl)- benzolsulfonyl]-N' -(3-methylcyclopentyl)-harnstoff
7,1 g 3-Methylcyclopentylharnstoff werden in 100 ml abs. Benzol suspendiert und mit 2,4 g 50 proz. Natriumhydrid versetzt. Man rührt 3 Stunden bei 50 C, versetzt den Ansatz mit 9,7 g 4-(ss-(2-Methoxy-5-chlor-benzamido)- äthyl)-benzolsulfochlorid in 100 ml abs. Benzol und rührt 3 Stunden bei 80O C nach. Man lässt erkalten, schüttelt das Reaktionsgemisch mehrfach mit Wasser aus und säuert die vereinigten wässrigen Extrakte mit Salzsäure an. Das ausgefällte Reaktionsprodukt wird aus 1 proz. Ammoniak umgefällt und aus Methanol umkristallisiert.
Der N-[4-(ss < 2-Methoxy-5-chlor-benzamido ) -äthyl-benzolsulfonyl] N'-(3-methylcyclopentyl)-harnstoff schmilzt bei 156 bis 158 C.
Beispiel 2
N- [4 -(ss- < 2-Methoxy-5-chlorbenzamido > -äthyl)-benzol sulfonyl]-N' -(4,7-methano-perhydro-indanyl-5-) -harnstoff
8,2 g N-[4-(ss-Acetamidoäthyl)-benzolsulfonyl]- N'-(4,7-methano-perhydro-indanyl-5)-harnstoff werden mit einer Lösung von 1,6 g Natriumhydrqxyd in 30 ml Wasser
2 Stunden unter Rückfluss erhitzt. Man lässt auf Zimmer temperatur abkühlen, versetzt mit 20 ml Aceton und 1,3 g Eisessig und.gibt portionsweise 4,1 g 2-Methoxy-5-chlorbenzoylchlorid zu. Nach zweistündigem Nachrühren bei Zim mertemperatur wird abgesaugt, der Niederschlag mit Bicarbonatlösung behandelt und anschliessend aus verdünnter Ammoniak/Salzsäure umgefällt.
Der erhaltene N-[4-(ss ( 2-Methoxy-5 -chlorbenzamido ) -äthyl) -benzolsulfonyl]- N' -(4,7-endomethylen-perhydro-indanyl-5)-harnstoff schmilzt nach Umkristallisation aus Methanol/Dimethyl formamid bei 196-198'" C.
