dans laquelle R représente un radical alcoyle comportant de 1 à 4 atomes de carbone.
Le substituant R des composés I représente, de pré férence, un radical méthyle, éthyle, propyle, ou butyle. A partir des composés de formule I, on peut pré parer les esters correspondants ; entre autres et de préférence, ceux qui dérivent d'acides organiques car boxyliques comportant de 1 à 18 atomes de carbone, ap partenant à la série aliphatique, alicyclique, aromatique ou hétérocyclique, tels que les acides formique, acétique, propionique, butyrique, valérianique, caprique, undécy- lénique, oléique, cyclopropane carboxylique, cyclohexane carbooxylique, cyclohexyl acétique, phényl acétique, ben zoïque, phénoxy acétique,
furanne 2-carboxylique, nico- tinique ou isonicotinique.
De même, on peut préparer aussi des éthers des composés de formule I, de préférence ceux qui pro viennent d'alcools aliphatiques inférieurs comme le mé thanol, l'éthanol, le propanol, l'isopropanol, les butanols, les pentanols, l'alcool allylique, d'alcools arylaliphati- ques comme l'alcool benzylique, ou d'alcools hétéro- cycliques comme le tétrahydropyranol.
Les nouveaux composés de formule I ainsi que leurs esters et éthers sont dotés de propriétés pharmacologi ques intéressantes. Ils possèdent des propriétés anaboli santes et androgènes qui les rendent aptes à être utilisés pour le traitement des troubles de l'anabolisme protidi que, de l'asthénie, des maigreurs, ostéosporose, andro pause, sénescence, retards de consolidation des fractures, troubles métaboliques des corticothérapies prolongées, syndrome adiposo génital, méno-métrorragies fonction nelles, fibromes, endométriose, comme traitement com plémentaire du cancer du sein, et comme agent de cica trisation dans le traitement des ulcères variqueux.

Les composés de formule I, ainsi que leurs esters et éthers peuvent être employés par voie buccale, per- linguale, transcutanée, par voie locale en application topique sur la peau et les muqueuses ou par voie rectale.
Ils peuvent se présenter sous forme de solutions ou de suspensions buvables ou injectables, de comprimés, de comprimés enrobés, de comprimés sublinguaux, de capsules, de suppositoires, de pommades, de crèmes et de poudres topiques.
La posologie utile de ces composés s'échelonne entre 250 y et 5 mg par jour, chez l'adulte, en fonction de la voie d'administration.
Les formes pharmaceutiques sous lesquelles peuvent être utilisés ces composés, telles que: solutions ou sus pensions buvables ou injectables, comprimés, comprimés enrobés, comprimés sublinguaux, capsules, suppositoires, pommades, crèmes et poudres topiques, peuvent être préparées selon les méthodes usuelles.
Le procédé de préparation, objet de l'invention, des composés I, est illustré par le schéma annexé; il est caractérisé en ce que l'on soumet un oxo-3 méthyl-7α hydroxy-113 R-133 XO-173 gona-4-ène, II, où X dé signe un radical acyle, dérivé d'un acide organique carboxylique comportant de 1 à 18 atomes de carbone, à l'action d'un agent déshydratant, puis fait agir sur le mélange de produits déshydratés résultant, un acide fort en milieu protique, isole l'oxo-3 méthyl-7α
R-133 XO- 17a gona-4,11-diène correspondant, saponifie ce dernier par action d'un agent basique et obtient l'oxo-3 méthyl- 7α R-133 hydroxy-17 gona-4,11-diène, I. On peut sou mettre le composé obtenu, le cas échéant, à l'action d'un agent d'estérification ou à l'action d'un agent d'éthérifi- cation.
Le produit brut provenant de l'action de l'agent dés hydratant est un mélange comportant divers composés diéniques, caractérisables par leurs spectres infrarouge et ultraviolet et dont les doubles liaisons sont soit conjuguées [5(10),9(11) ou 4,9] soit non conjuguées [4,9(11) ou 4,1l].
Le procédé de l'invention peut être exécuté avanta geusement comme suit L'agent déshydratant est, de préférence, le N-bromo- acétamide en présence d'anhydride sulfureux; mais il est également possible d'employer l'oxychlorure de phos phore ou le chlorure de thionyle en présence de pyri- dine.
L'acide fort est choisi, de préférence, dans le groupe constitué par l'acide perchlorique, l'acide sulfurique, l'acide p-toluènesulfonique et une résine sulfonique échangeuse de cations, le milieu protique pouvant être formé par l'acide acétique ou un alcool aliphatique inférieur.
L'action de l'acide fort en milieu protique sur le mélange de produits déshydratés, conduit à l'obtention d'un mélange d'oxo-3 méthyl-7α R-133 XO-17B gona- 4,9-diène et d'oxo-3 méthyl-7α R-13(3 XO-173 gona-4,11- diène, ce dernier composé pouvant comme précédem ment être isolé par les méthodes classiques de sépara tion telles que cristallisation ou chromatographie.
La saponification de l'oxo-3 méthyl-7α R-13(3 X0- 17(3 gona-4,11-diène par un agent basique tel que la potasse méthanolique conduit à l'obtention de l'oxo-3 méthyl-7α R-13P hydroxy-173 gona-4,11-diène que l'on peut estérifier ou éthérifier selon les méthodes usuelles, si on le désire.
Pour estérifier la fonction alcool en position 17, on peut, par exemple, utiliser un dérivé fonctionnel d'un acide convenablement choisi, tel qu'un chlorure ou un anhydride d'acide, en opérant, de préférence, en présence d'une amine tertiaire.
Pour effectuer l'éthérification de la fonction alcool en position 17, on peut, par exemple, faire réagir un halogénure d'un reste hydrocarboné convenablement choisi, sur un dérivé alcalin de l'alcool en position 17.
Les oxo-3 méthyl-7α hydroxy-113 R-13(3 XO-17P gona-4-ènes, II, utilisés au départ du procédé, peuvent être préparés comme suit On fait agir un agent d'acylation ou un mélange acylant convenable sur. un oxo-3 dihydroxy-113, 17(3 R-17(3 gona-4-ène pour obtenir un triacyloxy-3, l1(3, 17(3 R-13[3 gona-3,5-diène que l'on brome sélectivement en position 6 par action d'un agent de bromuration convenable, débromhydrate l'oxo-3 bromo-6% diacyloxy- 113, 17(3 R-13(3 gona-4-ène résultant par action d'un halogénure de lithium en présence d'un agent basique pour obtenir un oxo-3 diacyloxy-113, 17P R-13(3 gona- 4,
6-diène sur lequel on fait agir un dérivé organo-métal- lique comportant un groupement méthyle, en présence d'un catalyseur tel que le chlorure cuivreux, puis sapo nifie par action d'une base les fonctions esters incom plètement hydrolysées pour obtenir l'oxo-3 méthyl-7α dihydroxy-11p, 17(3 R-13(3 gona-4-ène que l'on trans forme par les méthodes usuelles, en dérivé 17-acyloxylé correspondant.
Un exemple de préparation d'oxo-3 méthyl-7α hy- droxy-113, éthyl-133 acétoxy-173 gona-4-ène est donné à titre indicatif dans la partie expérimentale.
Les oxo-3 dihydroxy-113, 17(3 R-13p gona-4-ènes utilisés au départ de ce procédé sont eux-mêmes obtenus selon le brevet français N 1574693.
L'exemple suivant illustre l'invention. Préparation Oxo-3 méthyl-7α hydroxy-113 éthyl-13p acétoxy-173 gona-4-ène (II avec R = C2H5 et X = CH3CO) Stade A : triacétoxy-3, 11(3, 17(3 éthyl-133 gona-3,5-diène.
Dans un mélange de 250 cm3 d'anhydride acétique, 8,75 cm3 de chlorure d'acétyle et 8,75 crus de pyridine, on introduit sous atmosphère d'azote, 25 g d'oxo-3 dihy- droxy-113, 17(3 éthyl-133 gona-4-ène (produit obtenu selon le brevet français N <B>1574693)</B> porte le mélange réactionnel au reflux, l'y maintient pendant cinq heures et trente minutes, le refroidit, le concentre à sec sous pression réduite, dissout le résidu dans 150 cm3 d'un mélange bouillant d'acide acétique et de méthanol (8-2), ajoute du noir, filtre à chaud, concentre le filtrat à 100 cm3, refroidit, laisse cristalliser, filtre et obtient 25,5 g de triacétoxy-3,113, 17(3 éthyl-133 gona-3,5-diène, F = 1810 C.

Par concentration des liqueurs mères puis cristallisa tion dans un mélange acide acétique-méthanol, on ob- tion un 2e jet de 4,4 g, F = 1800 C.

Dans un mélange de 450 cm3 de diméthylformamide et de 7 cm3 d'eau distillée, on introduit, sous atmosphère d'azote, 133,4 g de triacétoxy-3, 11(3, 17(3 éthyl-133 gona- 4-ène, refroidit 18 mélange à 0 C et y ajoute en une heure 62 g de N-bromosuccinimide (test positif au papier amido-ioduré) on obtient ainsi l'oxo-3 bromo-6b diacétoxy-113, 17(3 éthyl-133 gona-4-ène, que l'on utilise, tel quel, en solution sans iolement intermédiaire, pour le stade suivant.
Ce composé peut être isolé, si on le désire, par con centration du mélange réactionnel, dilution à l'eau, extraction par un solvant organique puis concentration à sec.

A la solution obtenue au stade B, on ajoute 33,2 g de bromure de lithium et 66,4 g de carbonate de lithium. On porte le mélange réactionnel à 951, C et l'agite à cette température pendant vingt-sept heures. On refroidit, verse le mélange réactionnel dans de l'eau acidulée par de l'acide acétique, agite, isole par essorage le précipité formé, le sèche et obtient 120 g d'oxo-3 diacétoxy-11p, 17(3 éthyl-133 gona-4-diène brut à l'état amorphe.

Dans 545 cm3 d'une solution 0,92 M de bromure de méthyl magnésium dans le tétrahydrofuran on intro duit 2,8 g de chlorure cuivreux en poudre, agite pendant quinze minutes à température ambiante, refroidit à -100 C, introduit progressivement une solution de 44 g d'oxo-3 diacétoxy-113, 17(3 éthyl-133 gona-4,6-diène dans 290 cm3 de tétrahydrofuran, laisse la température remonter à 200 C, agite pendant vingt minutes, verse le mélange réactionnel dans un mélange d'eau et d'acide chlorhydrique, extrait la phase aqueuse au chlorure de méthylène,

On amène le mélange au reflux, l'y maintient pendant une heure, le refroidit, le verse dans l'eau, acidifie à pH 3,5 par une solution aqueuse d'acide chlorhydrique, extrait la phase aqueuse au chlorure de méthylène, lave les extraits chlorométhyléniques à l'eau, les sèche, les con centre à sec, dissout le résidu dans le chlorure de méthy lène, fait passer la solution obtenue sur une colonne de Florisil (silicate de magnésium activé), concentre à sec la solution résultante, cristallise l'extrait sec dans un mélange de méthanol et d'éther isopropylique et ob tient 13,9 d'oxo-3 méthyl-7α
dihydroxy-113, 17(3 éthyl- 133 gona-4-ène brut, F = 2460 C qui est utilisé tel quel pour le stade suivant.
Un échantillon de ce produit est purifié par cristal lisation dans le méthanol, F = 248o C.

Un échantillon d'oxo-3 méthyl-7α éthyl-133 acétoxy- 17p gona-4,11-diène est purifié par empâtage dans l'éther de pétrole (Eb. = 35-700 C) puis cristallisation dans l'éther isopropylique, F = 1440 C la-ID = -I- 650 (c = 0,55%, chloroforme).
