This process is illustrated by diagram 1.
It is characterized in that a 3-oxo-methyl-7 α hydroxy-11a R-13 (3-acyloxy-173 gona-4-ene, I, on the action of a dehydrating agent, reacts oxygen in the presence of a tertiary amine with the resulting mixture of dehydrated products, reduces by the action of a reducing agent the hydroperoxide formed to obtain the 3-oxo-7-methyl-7 α
hydroxy-113 R-13 (3-acyloxy-173 gona-4,9-diene, VII, which is subjected to the action of a strong acid, in the presence of a nucleophilic agent, by operating in a solvent aprotic, isolates a 3-oxo-methyl-7 α R-13p acyloxy-173 gona-4,9,11-triene, and saponifies the ester function at 17 of this compound to obtain the 3-oxomethyl-7 α R- 13 (3 hydroxy-17P gona-4,9,11-triene, VIII.
The crude product resulting from the action of the dehydrating agent on oxo-3 methyl-7 α hydroxy-113 R-13 (3-acyloxy-17p gona-4-ene is a mixture comprising various diene compounds whose double bonds are either conjugated or unconjugated.
The process, object of the invention, can be carried out advantageously as follows The dehydrating agent is preferably N-bromoacetamide in the presence of sulfur dioxide, but it is also possible to use phosphorus oxy-chloride or phosphorus or thionyl chloride in the presence of pyridine.
The tertiary amine in the presence of which the oxidation by oxygen of the crude dehydration product is carried out is preferably triethylamine or pyridine. This oxidation is carried out in an organic solvent, in particular in an alcohol such as ethanol or methanol, or in an N, N-dialkoylamide such as dimethylformamide.
The reducing agent used to reduce the crude mixture containing the hydroperoxide from the oxygen oxidation of the mixture of dehydrated dienes is preferably triethyl or trimethyl phosphite. This reduction is carried out in an organic solvent such as an alcohol such as ethanol. The reducing agent can also be an alkali iodide such as potassium iodide.
At the end of the reduction of the crude hydroperoxide, a mixture is obtained mainly containing oxo-3-methyl-7 α R-13P acyloxy-17P gona-4,11, diene and 3-oxo-7 &alpha-methyl; hydroxy-113 R-13 (3-acyloxy-17 gona-4,9-diene, VII, which is conveniently isolated by chromatography.
3-oxo-7 α hydroxy-11P R-13 (3-acyloxy-17P gona-4,9-diene, VII, can be converted into 3-oxomethyl-7 α R 13i acyloxy-17p gona-4,9,11-triene by the action of a mineral acid such as sulfuric acid or perchloric acid, an organic acid such as formic acid or p-toluenesulfonic acid, or a Lewis acid such as boron trifluoride.
The nucleophilic agent is preferably a nitrile of an organic acid such as acetonitrile, malonic dinitrile, dichloroacetonitrile, etc., or an alkali metal cyanide such as sodium cyanide or potassium cyanide.
The aprotic solvent is, for example, methylene chloride, chloroform, cyclohexane, ben zene, ether or tetrahydrofuran.
3-oxo-7 α R-13 (3-acyloxy-173 gona-4,9,11-triene obtained can be saponified, for example, by the action of potassium hydroxide in methanolic medium. 3-oxo-methyl-7 α R-13 (3 hydroxy- 173 resulting gona-4,9,11-triene, VIII, can then be esterified or etherified according to the usual methods. Esterification is carried out in particular by action on the 17-hydroxylated derivative of a functional derivative of an acid suitably chosen, such as a hydride or chloride, in the presence of a tertiary amine.
The etherification is carried out, in particular, by the action of a halide of a suitable hydrocarbon residue on an alkali metal salt of the 17-hydroxylated derivative previously ketalised in position 3.
A method of preparing the starting product (I) is illustrated by scheme 2. According to this method, an acylating agent or a mixture of acylating agents is made to act on an oxo-3 dihydroxy-11p, 17a R-133 gona- 4-ene, II, the acylating agent derived from an organic carboxylic acid comprising from 1 to 10 carbon atoms, to obtain a triacyloxy-3, 11 (3, 17 (3 R-13 (3 gona- 3,5 diene, III, which is selectively brominated in position 6 by the action of a bromination agent, dehydrates 3-oxo-bromo-6 # diacyloxy-113, 17 (3 R-13 (3 gona-4- ene resulting , IV,
