Verfahren zur Herstellung des neuen Antibiotikums 460
Die Erfindung betrifft ein Verfahren zur Herstellung eines neuen antibiotisch wirksamen Produktes. Insbesondere betrifft die Erfindung ein Verfahren zur fermentativen Herstellung eines antibiotisch wirksamen Produktes, bei welchem eine bisher unbeschriebene Variante des Mikroorganismus Micromonospora chalcea der Ordnung Actinomycetales verwandt wird, die Isolierung eines Antibiotikumgemisches aus diesem Produkt, die Auftrennung dieses Gemisches in die enthaltenen Antibiotika.
Ein erfindungsgemässes Verfahren zur Herstellung eines antibiotisch wirksamen Produktes besteht darin, dass man die vorerwähnte Variante von Micromonospora chalcea oder einen äquivalenten Mikroorganismus bebrütet, bis ein Produkt mit guter antibiotischer Wirksamkeit gebildet ist.
Aus diesem Produkt kann man ein Antibiotikumgemisch isolieren, das im folgenden kurz Antibiotikum 460 genannt wird. Dieses Antibiotikum 460 ist eine Mischung von drei nahe verwandten antibiotischen Substanzen, in welche es aufgetrennt werden kann und die im folgenden Antibiotikum460-Komponenten A, B und C genannt werden. Im folgenden wird die Herstellung dieser Antibiotika beschrieben.
Der Mikroorganismus
Der im erfindungsgemässen Verfahren verwendete Mikroorganismus ist eine Variante von Micromonospora chalcea var. flavida genannt wird. Ein für die Verwendung im erfindungsgemässen Verfahren besonders geeigneter Stamm des Mikroorganismus wurde aus einer von einer bestimmten Stelle im Staate New York (USA) entnommenen Bodenprobe isoliert.
Eine Kultur der erfindungsgemäss vorzugsweise verwendeten Mikroorganismen wurde in der Kulturensammlung des Landwirtschaftsministeriums (Departement of Agriculture) der Vereinigten Staaten von Amerika, Northern Utilization Research and Development Division, Peoria, Illinois, hinterlegt, wo sie unter ihrer NRRL-Nummer 3222 erhältlich ist.
Man erreicht die Isolierung des Mikroorganismus dadurch, dass man etwas von einer getrockneten Bodenprobe in sterilem destilliertem Wasser schüttelt und dann die Suspension nach Verdünnung auf ein geeignetes gesiebtes Agarmedium aufstreicht. Die getrocknete Bodenprobe stammt von der erwähnten Stelle im Staat New York, falls der vorerwähnte Stamm M. chalcea var. flavida im erfindungsgemässen Verfahren verwendet wird.
Die mikroskopische Untersuchung von 21 Tage alten Kulturen von M. chalcea van flavida auf einem Medium, das aus 3 /0 NZ Amin Typ A (ein bei der enzymatischen Verdauung von Casein entstehendes Pepton, das als Stickstoffquelle dient und bei Sheffield Chemical Company, Norwich, New York, erhältlich ist), 1% Dextrose und 1,5 /o Agar besteht, ergibt, dass sich der Mikroorganismus durch ein langes, wenig verzweigtes Mycel mit einem ungefähren Durchmesser von 0,5 lt auszeichnet. Die Sporen sind nicht reichlich und sind Einzelsporen. Sie sind oval bis kugelförmig, haben einen Durchmesser von 1,8 - 1,0 lt und treten gerne bräunlich in Erscheinung.
Diese Kultur zeigt bei makroskopischer Betrachtung gutes Wachstum und kein Luftmycel und hat ein gefaltetes Aussehen.
Kolonien von M. chalcea van flavida kann man ferner durch ihr Aussehen und ihr Wachstum auf verschiedenen Nährmedien charakterisieren. Die folgende Tabelle : enthält die Wachstums- und Farbmerkmale von M. van flavida auf einigen häufig verwendeten Nährböden. Die Beobachtungen wurden an Kolonien gemacht, die durch 21tägige Bebrütung bei 24 bis 26 "C der vorerwähnten Mikroorganismen auf den bezeichneten Medien erhalten wurden. Bei der Beschreibung der Farbmerkmale wurde das folgende System und die folgende Bezeichnungsweise verwandt: Die Farbbezeichnung besteht aus zwei Angaben.
Die jeweils erste Angabe ist der Farbname gemäss Descriptive Color Name Dictionary von Taylor, Knoche und Granville, herausgegeben durch die Container Corporation of America, 1950 (USA), in Verbindung mit einer diesem Namen entsprechenden Nummer; diese Nummer ist dem Color Harmony Manual , vierte Auflage, 1958, herausgegeben ebenfalls von der Container Corporation of America, entnommen. Die zweite Angabe besteht aus einem Farbnamen und einer Nummer entsprechend dem Synonym oder ungefähren Synonym des Circular 553 vom 1. November 1955 des National Bureau of Standards (U.S.A.).

Der Mikroorganismus zeigt gutes Wachstum auf Kohlen hydraten wie Glucose, Arabinose, Galactose, Lactose, Laevu lose, Mannose, Raffinose, Stärke, Saccharose, Xylose und
Melibiose.
Er wächst kümmerlich, wenn überhaupt, auf Me dien, die als einziges Kohlenhydrat Rhamnose, Inosit, Man nit, Sorbit, Ribose oder Glycerin enthalten.

M. chalcea van flavida zeigt ein befriedigendes Wachstum im Temperaturbereich von ungefähr 20 bis 45 "C und bei einem pH-Bereich von ungefähr 6 bis 8, bestes Wachstum jedoch im allgemeinen bei einer Temperatur zwischen 26 und 37 "C und bei einem pH- WµLW /0PH-Wert zwischen 6,5 und 7,8. Wenig oder kein Wachstum findet bei einer Temperatur von 50 "C statt. M. chalcea van flavida ist aerob und reduziert Nitrat zu Nitrit.
Während der mit NRRL 3222 bezeichnete Stamm von M. chalcea van flavida in erster Linie wegen seiner leichten Erhältlichkeit ein besonders geeignetes Ausgangsmaterial für das Verfahren darstellt, versteht es sich, dass andere Stämme der Variante und andere äquivalente Mikroorganismen annehmbare Alternativlösungen sind. Weiterhin können Mutanten von M. chalcea var. flavida, die dadurch erhalten sind, dass der Mikroorganismus, z. B. vom Stamm NRRL 3222, der Einwirkung eines Mutationsmittels, wie Hochfrequenzstrahlung (einschliesslich Röntgenstrahlung und Ultraviolettstrahlung), Actinophagen und/oder Stickstofflost, unterworfen wird, auch als Ausgangsmaterial geeignet sein, die nach Bebrütung unter geeigneten Bedingungen ein Produkt liefern, das gute antibiotische Wirksamkeit hat, aus dem das Antibiotikum 460 isoliert werden kann.
Bebrütung des Mikroorganismus
M. chalcea var. flavida liefert bei kontrollierter Bebrütung ein Produkt mit guter antibiotischer Wirksamkeit, das aus einem komplizierten Gemisch antibiotischer Substanzen besteht, aus dem das Antibiotikum 460 isoliert werden kann.
Zur Herstellung dieses Gemisches kann man erfindungsgemäss M. chalcea var. flavida unter aeroben Bedingungen im Temperaturbereich von 20"-45 "C, vorzugsweise von 25 -40 C, wachsen lassen. Die Bebrütung wird vorzugsweise unter Schütteln oder Rühren unter submersen aeroben Bedingungen bei einem pH-Wert von ungefähr 6,0 bis 8,0 durchgeführt, wobei ein pH-Bereich von 6,5 bis 7,8 häufig besonders vorteilhaft ist. Die Bebrütungszeit beträgt üblicherweise 2 bis 7 Tage, vorzugsweise 4 bis 7 Tage, obwohl kür-zer oder länger bebrütet werden kann.
Die normalerweise bei der Bebrütung verwandten Medien sind wässrig und enthalten von den Mikroorganismen assimilierbare Kohlenstoff- und Stickstoffverbindungen. Geeignete Kohlenstoffverbindungen sind z. B. Kohlenhydrate, wie Stärke, Dextrin und Zucker. Geeignete Stickstoffverbindungen sind sowohl organische als auch anorganische Stickstoffverbindungen, vorzugsweise die ersten, wie z. B. das Handelsprodukt NZ-Amin Typ A.
Zwei Beispiele einer Zusammensetzung eines für den Mikroorganismus geeigneten Bebrütungsmediums sind: Medium 1: Hefeextrakt 0,5%, Fischauszüge 0,1 /o, Cornsteep li- quor 0,1%, Lactose 3% und Calciumcarbonat 0,1 /o; Medium II: Rindfleischextrakt 0,3 /0, Tryptose 0,5 /0, Dextrose 0,1 /o und Hefeextrakt 0,5 /0.
Es ist zweckmässig, wenn die Bebrütungsmedien ein Ingredienz enthalten, das mitwirkt, die Alkalität und Acidität der Medien im Gleichgewicht zu halten. So enthalten Medium 1 und II Calciumcarbonat zu diesem Zweck.
Die so erhaltene Zusammensetzung besteht aus einem Mycel und einer Brühe und zeigt als ganzes eine gute antibiotische Wirksamkeit, die im allgemeinen hauptsächlich in der Brühe konzentriert ist.
Isolierung des Antibiotikums 460
Das Antibiotikum 460 kann aus der Brühe isoliert werden, indem man das ganze bebrütete Nährmedium mit einer starken Säure, wie Schwefelsäure, auf einen pH-Wert von ungefähr 2,0 ansäuert. Dann trennt man das Mycel ab, und zwar normalerweise durch Filtration, wobei man z. B. eine Diatomeenfilterhilfe verwendet. Aus der mycelfreien Brühe trennt man das antibiotische Gemisch nach einer von zahlreichen verschiedenen Methoden ab, z. B. durch Bildung eines unlöslichen Salzes, mittels einer Harzmethode oder mittels Lösungsmittelextraktion.
Eine charakteristische Anwendung der Harzmethode besteht darin, dass die Brühe zuerst mit einem starken Alkali neutralisiert wird, z. B. mit Natrium- oder Kaliumhydroxid.
Die neutralisierte Brühe wird dann mit Ammoniumoxalat behandelt, wobei das im Medium vorhandene Calcium als schwerlösliches Oxalatsalz ausgefällt wird. Man trennt das Calciumoxalat durch Filtration und das Antibiotikum 460 vom Filtrat durch Absorption an einem lonenaustauscherharz, wie ein Harz vom lRC-50-Amberlit-Typ, ab. Beispiele geeigneter Harze vom lRC-50-Amberlit-Typ, sowohl Anionen- als auch Kationenaustauscher, kann man im Handbook of Chemistry and Physics , 42. Auflage, Chemical Rubber Publishing Company, Cleveland, Ohio (USA), finden. Das Antibiotikum wird dann durch Säurebehandlung vom Harz eluiert. Das saure Eluat kann dann beispielsweise mittels eines Anionenaustauscherharzes neutralisiert und die Antibiotikumlösung lyophilisiert werden, bevor sie mittels einer Standard-Agar-Diffusionsmethode geprüft wird.
Auftrennung des Antibiotikums 460 in seine Komponenten
Das Antibiotikum 460 ist, wie vorher erwähnt, ein Gemisch dreier verwandter Antibiotika, in welche es leicht aufgetrennt werden kann. Zahlreiche Methoden sind für diese Auftrennung geeignet. Die Aufteilung erfolgt zweckmässig nach einer Chromatographiemethode. Ein geeignetes Auftrennverfahren ist die Verteilungschromatographie. Dabei wird das Antibiotikumgemisch durch eine Säule geschickt, die Kieselsäure als inertes Trägermaterial enthält, und die Säule wird dann mit einer Lösungsmittelmischung eluiert, die im wesentlichen die folgende Zusammensetzung hat (in Volumenteilen): Äthanol 20, Essigsäure 8, Essigsäureäthylester 8, Wasser 8 und Pyridin 4.
Das von dieser Säule erhaltene Eluat enthält drei unterschiedliche Fraktionen, die in der Reihenfolge ansteigender Polarität, d. h. abnehmender Beweglichkeit unter den gewählten Bedingungen, Fraktionen 1, 2 und 3 genannt werden. Fraktion 1 hat einen bioautographischen RrWert von ungefähr 0,95; für sie benötigt man üblicherweise ungefähr 15 bis 20 /o des Gesamteluatvolumens zur Elution; Fraktion 1 besteht aus ungefähr 75 bis 80 Gewichtsprozent des gesamten erhaltenen wirksamen Materials. Fraktion 2 hat einen bioautographischen RrWert von ungefähr 0,25; man benötigt für sie ungefähr 40 bis 42 /o des gesamten Eluatvolumens zur Elution; Fraktionen 2 besteht aus ungefähr 10 bis 15 Gewichtsprozent des gesamten erhaltenen wirksamen Materials.
Fraktio 3 hat einen bioautographischen RrWert von ungefähr 0,20; man benötigt für sie ungefähr 40 bis 48 /o des gesamten Eluatvolumens zur Elution; Fraktion 3 besteht aus ungefähr 10 bis 15 Gewichtsprozent des gesamten erhaltenen wirksamen Materials.
Jede Fraktion wird dann durch Eindampfen im Vakuum bis zur Trockne eingeengt. Der so erhaltene Rückstand ist das Antibiotikum 460 Komponente A, B oder C, je nachdem von welcher Eluatfraktion er stammt. Der Rückstand wird dann in Wasser gelöst, die Lösung filtrieruunddas Filtrat lyophilisiert.
Das Antibiotikum 460 und seine Komponenten A, B und C können beispielsweise durch Waschen mit einem geeigneten organischen Lösungsmittel und/oder durch Umkristallisation aus solch einem Lösungsmittel gereinigt werden. Sie können auch in Derivate umgewandelt werden, wie z. B. in Säureadditionssalze, in N- und O-Acylverbindungen, in Bisulfit-Additionsprodukte und in Sulfonamide. Das folgende Beispiel dient zur weiteren Veranschaulichung gewisser Züge dieser Erfindung, insbesondere der Bebrütung von M. chalcea van flavida, der Abtrennung der Brühe vom erhaltenen bebrüteten Nährmedium, die Isolierung des Antibiotikums 460 davon und die Auftrennung dieses Antibiotikumgemisches in seine Komponenten.
Beispiel A. Keimung
100 ml steriles Medium II (siehe oben) werden in einem 300-ml-Erlenmeyerkolben mit M. chalcea var. flavida von einer Agarschrägplatte bebrütet. Man setzt den Kolben auf eine rotierende Schüttelmaschine (250-275 Umdrehungen pro Minute) und lässt die Kultur 3 Tage bei 35 "C wachsen.
Diese Kultur ist ausreichend zum Beimpfen von vier 2-Liter Bebrütungen.
B. Bebrütung
Man gibt 25 ml des oben hergestellten Inokulums aseptisch in einen 2-Liter-Erlenmeyerkolben, der 500 ml steriles Medium I enthält. Der Kolben wird auf eine rotierende Schüttelmaschine gesetzt und die Kultur 5 bis 6 Tage bei 28 "C bebrütet, wobei der pH-Wert durch mehrmalige Zugabe von Calciumcarbonat auf ungefähr 7 gehalten wird.
Der Ansatz wird während der 5 bis 6 Tage in gleichen Zeitabständen geprüft, um die maximale Wirksamkeit gegen S.
aureus, S. lutea, B. subtilis und K. pneumoniae zu bestimmen.
C. Abtrennung
Die erhaltene Reaktionsmischung, d. h. das bebrütete
Nährmedium, wird auf einen pH-Wert von 2,0 mit 6N
Schwefelsäure angesäuert und nach Zusatz von Diatomeen erde als Filterhilfe filtriert, um die Brühe abzutrennen. Das
Filtrat wird mit 6N Natriumhydroxid neutralisiert und Am moniumoxalat hinzugefügt, um das im Medium enthaltene
Calcium als sein schwerlösliches Oxalatsalz niederzuschla gen. Die Suspension wird filtriert und das niedergeschlagene
Calciumoxalat verworfen.
D. Isolierung
5 bis 6 g IRC-50-Kationenharz (Natriumsalzform) werden im Filtrat suspendiert und die Suspension eine halbe Stunde gerührt. Die Suspension wird dann filtriert und die Adsorp tionsstufe mit 5 bis 6 g frischem Harz wiederholt. Die Sus pension wird filtriert und das Filtrat verworfen. Das ver einigte Harz wird von Brüheresten frei gewaschen und dann in 50 ml Wasser suspendiert. Zur Suspension fügt man unter
Rühren genügend 3N Schwefelsäure hinzu, um die Suspen sion auf einen pH-Wert von 2,0 zu bringen, auf dem sie 30
Minuten lang gehalten wird. Sie wird dann filtriert, das Harz verworfen, das Filtrat neutralisiert und das Antibiotikumge misch an 20 g neutraler sulfationenfreier Aktivkohle (Sieb nummer 20 bis 40) adsorbiert.
Danach wird die Aktivkohle mit demineralisiertem Wasser gewaschen, bis das Waschwas ser sulfatfrei ist, was mit Bariumchlorid-Lösung geprüft wird.
Das Antibiotikumgemisch wird aus der Aktivkohle extra hiert, indem man sie 1 Stunde in 120 ml einer 1:1 Mischung von 1N Schwefelsäure und Methanol heftig schüttelt oder rührt. Die Aktivkohle wird dann durch Filtration abgetrennt, das Filtrat auf 25 ml durch Erhitzen im Vakuum eingeengt, das Konzentrat lyophilisiert und so ungefähr 50 mg Antibio tikum 460 erhalten.
E. Auftrennung
420 mg des Rohantibiotikums 460 (erhalten durch Verei nigen mehrerer Ansätze) werden in 20 ml Wasser gelöst und zum Absorbieren der Lösung 5,0 g Kieselsäure hinzugefügt.
Die Kieselsäure wird im Vakuum getrocknet und für die wei tere Verarbeitung aufgehoben. Eine Chromatographiersäule (Länge 80 cm, Durchmesser 3,5 cm) wird mit Kieselsäure bis zu einer Länge von 72 cm beschickt und die Antibiotikum Kieselsäure-Mischung von oben in den oberen Teil der Säule gefüllt.
Das Eluierlösungsmittel bestehend aus 20 Teilen Äthanol, 8 Teilen Essigsäure, 8 Teilen Essigsäureäthylester, 8 Teilen Wasser und 4 Teilen Pyridin (jeweils Volumenteile) lässt man mit einer Geschwindigkeit von 1,0 ml pro Stunde durch die Säule laufen und man sammelt das Eluat in 2,0-ml-Fraktionen. Die Säule wird auf antibiotische Wirksamkeit untersucht, indem man Papierscheiben mit von jeder einzelnen 2-ml-Fraktion entnommenen Flüssigkeit befeuchtet, die Scheiben in einem Strom trockener Luft trocknet und dann gegen S. aureus und E. coli in der üblichen Art prüft. Eine Probe von jeder 2-ml-Fraktion wird absteigend papierchromatographiert, wobei man 18 Stunden mit einem Propanol-Pyridin-Essigsäure-Wasser-System (15:10:3:12) arbeitet.
Dann werden die Papiere getrocknet und auf verschiedenen Platten gegen in einem Agar-Medium suspendierte Kulturen von S. aureus und E. coli bioautographiert.
Fraktionen mit übereinstimmenden Bioautographie-Mustern werden vereinigt und die drei so erhaltenen vereinigten Fraktionen jeweils im Vakuum zur Trockene eingeengt. Der von jeder der drei Fraktionen erhaltene Rückstand wird in Wasser gelöst, die Lösung filtriert, das Filtrat lyophilisiert und so die entsprechende Komponente des Antibiotikums 460 erhalten (A, B oder C, je nachdem von welcher Fraktion der Rückstand stammt).
Angaben über gewisse Merkmale der Komponenten A, B und C des beispielsweise nach dem Verfahren des voranstehenden Beispiels erhaltenen Antibiotikums 460 finden sich nachfolgend.

Die Infrarotspektren der Antibiotikum460-Komponenten A, B und C sind in Fig. 1 bis 3 der anhängenden Zeichnungen wiedergegeben, woris-k die Wellenlänge in lt bedeutet, v bedeutet die Frequenz in cm ' und A bedeutet die Absorption. Die Absorptionsmaxima und Bandencharakteristika sind in Tabelle IV unten zusammengefasst, wobei die dort verwandten Abkürzungen die folgende Bedeutung haben: S = stark, M = mittel, M-S = mittel bis stark, W = schwach, M-W = mittel bis schwach, VW = sehr schwach und SH = Schulter.

7. Derivate
Chemisch sind Antibiotikum 460 und seine Komponen ten A, B und C Stickstoffbasen und sie können in die ver schiedenen Arten von Derivaten überführt werden, die mit solchen Basen erhältlich sind. Zum Beispiel können die freien Basen sowohl mit organischen als auch mit anorgani schen Säuren in ihre Säureadditionssalze überführt werden.
Die Derivate, z. B. die Säureadditionssalze, haben die antibio tischen Eigenschaften der Basen mit nur graduellen Unter schieden, sind jedoch manchmal besser löslich und eignen sich manchmal besser als die freien Basen für Zubereitun gen. Beispiele für Säureadditionssalze sind jene, die man mit
Salzsäure, Schwefelsäure, Phosphorsäure, Toluolsulfonsäure,
Methansulfonsäure, Bernsteinsäure, Naphthalin-l-carbon säure, Laurylschwefelsäure, Acetylsalicylsäure und Chol säure erhält. Sie können durch Titrieren einer wässrigen Lö sung der freien Base mit der gewünschten Säure und an schliessende Gefriertrocknung der erhaltenen Lösung herge stellt werden.
Papierchromatographische Eigenschaften
Die Papierchromatogramme können in verschiedenen
Lösungsmittelsystemen gemacht werden und die RrWerte werden nach einer Standard-Bioautographiemethode be stimmt, die darin besteht, dass die Papierchromatogramme entwickelt und getrocknet werden, bevor sie auf eine Staphy lococcus-aureus-Agarplatte gelegt werden; nach einer Kon taktzeit von 15 Minuten wird das Papier von der Platte ent fernt, die dann bei 37 "C 16 bis 20 Stunden bebrütet wird.
Durch Beobachtung der Lage der Hemmzonen kann man die R1rWerte bestimmen. In der folgenden Tabelle Vl sind die mit zwei verschiedenen Lösungsmittelsystemen erhalte nen Rl-Werte der drei Komponenten des Antibiotikums 460 angegeben: 1. Propanol-Pyridin-Essigsäure-Wasser (15:10:3:12), PPEW-System genannt, und 2. Propanol-Essig säure-Wasser (50:40:5), PEW System genannt.

Biologische Eigenschaften In-Vitro-Spektren
Die In-Vitro-Wirksamkeit der Antibiotikum-460-Komponenten gegen eine Reihe von Mikroorganismen wird in der folgenden Tabelle VII dargelegt. In-Vivo-Wirksamkeit
Die In-Vivo-Wirksamkeit der Komponenten A und C des Antibiotikums 460 gegen eine letale Infektion von Staphylococcus aureus (Stamm Gray) und der Komponente C gegen Klebsiella pneumoniae wird in Tabelle VIII angeführt. Die Tests wurden an männlichen Mäusen, Stamm CF Nr. 1, Gewicht 18 bis 22 g, ausgeführt, indem man die Mäuse mit einem Inoculum der entsprechenden Bakterien infizierte und ihnen dann die Antibiotikum-460-Komponente subcutan injizierte. Die Ergebnisse werden in PD50-Werten angegeben (diejenige Konzentration von Antibiotikum, die 50 /O der infizierten Tiere schützt), deren Dimension Milligramm Antibiotikum-Komponente pro Kilogramm Testtier ist.

Die voranstehenden biologischen Daten zeigen, dass die Komponenten des Antibiotikum-460-Gemisches eine wirksame Gruppe von antibiotischen Substanzen sind und einzeln oder als Gemisch zur Bekämpfung von Infektionen verwandt werden können, die von verschiedenen pathogenen Mikroorganismen herrühren, insbesondere von grampositiven Mikroorganismen wie Staphylococcus aureus, Diplococcus pneumoniae und Streptococcus pyogenes.
Diese Antibiotika sind ebenso zur Sterilisation von Laborglasgerät brauchbar, zur Bekämpfung und Ausrottung von gewissen Bakterien-lnfektionen bei Labortieren, zur Sterilisation von Operationsräumen und chirurgischen Instrumenten und in Kombination mit Seifen und Detergentien zur Reinigung von Räumen, die der Zubereitung von Speisen dienen, wie Krankenhausküchen und Speiseräumen in Schulen.
Die einzelnen Komponenten A, B und C des Antibiotikums 460 finden als Standard gegen gewisse Mikroorganismen Anwendung. Die Komponente A kann beispielsweise als Bezugssubstanz dienen, gegen die andere potentiell brauchbare antibiotische Substanzen gemessen werden, wobei Staphylococcus aureus als Testorganismus dient. Die erhaltenen Ergebnisse würden dann ein Mass des Verhältnisses der Aktivität des potentionellen Antibiotikums gegen über derjenigen der Komponente A sein. Ähnlich können die Komponenten B und C als Standard gegen Klebsiella pneumoniae bzw. Escherichia coli Verwendung finden. Sol che Standards stellen eine verhältnismässig einfache Methode zur Bestimmung der relativen Wirksamkeit von antibiotischen Verbindungen dar.
Therapeutische Formen der 460-Antibiotika
Für therapeutische Zwecke kann das Antibiotikum 460 oder jede seiner Komponenten A, B oder C oder ein Derivat des Gemisches oder einer Komponente gegebenenfalls zu einer annehmbaren Form weiter verarbeitet werden.
Zum Beispiel kann das Antibiotikum, beispielsweise durch Mischen, mit einem Träger zu einer therapeutischen Zubereitung vereinigt werden. Als Träger können eine oder mehrere flüssige oder feste, organische oder anorganische Substanzen dienen, die mit dem Antibiotikum verträglich und für die Verabreichungsweise des Antibiotikums geeignet sind. Beispiele für solche Substanzen sind Wasser, Gelatine, Lactose, eine Stärke, Magnesiumstearat, Talk, ein Pflanzenöl, Benzylalkohol, ein Gummi, ein Glycol und Vaseline.
Die Zubereitungen können auch noch ein anderes wirksames Ingrediens enthhalten, z. B. ein Analgetikum, ein anderes Antibiotikum oder ein proteolytisches Enzym.
Method of making the new antibiotic 460
The invention relates to a method for producing a new antibiotic product. In particular, the invention relates to a method for the fermentative production of an antibiotic product in which a previously undescribed variant of the microorganism Micromonospora chalcea of the order Actinomycetales is used, the isolation of an antibiotic mixture from this product, the separation of this mixture into the antibiotics it contains.
A method according to the invention for producing an antibiotic product consists in that the aforementioned variant of Micromonospora chalcea or an equivalent microorganism is incubated until a product with good antibiotic activity is formed.
A mixture of antibiotics can be isolated from this product, which is referred to below as antibiotic 460 for short. This antibiotic 460 is a mixture of three closely related antibiotic substances into which it can be separated and which are referred to below as antibiotic 460 components A, B and C. The production of these antibiotics is described below.
The microorganism
The microorganism used in the process according to the invention is a variant of Micromonospora chalcea var. Flavida. A strain of the microorganism particularly suitable for use in the process according to the invention was isolated from a soil sample taken from a specific location in New York State (USA).
A culture of the microorganisms preferably used according to the invention has been deposited in the culture collection of the Department of Agriculture of the United States, Northern Utilization Research and Development Division, Peoria, Illinois, where it is available under its NRRL number 3222.
The isolation of the microorganism is achieved by shaking some of a dried soil sample in sterile distilled water and then spreading the suspension on a suitable sieved agar medium after dilution. The dried soil sample comes from the location mentioned in New York State, if the aforementioned strain M. chalcea var. Flavida is used in the method according to the invention.
The microscopic examination of 21-day-old cultures of M. chalcea van flavida on a medium made from 3/0 NZ amine type A (a peptone formed during the enzymatic digestion of casein and used as a nitrogen source and obtained from Sheffield Chemical Company, Norwich, New York, available), 1% dextrose and 1.5 / o agar, shows that the microorganism is characterized by a long, poorly branched mycelium with an approximate diameter of 0.5 l. The spores are not abundant and are single spores. They are oval to spherical, have a diameter of 1.8 - 1.0 l and tend to appear brownish.
This culture shows good growth and no aerial mycelium when viewed macroscopically and has a folded appearance.
Colonies of M. chalcea van flavida can also be characterized by their appearance and their growth on various nutrient media. The following table: contains the growth and color characteristics of M. van flavida on some commonly used culture media. The observations were made on colonies obtained by incubating the aforementioned microorganisms at 24 to 26 "C for 21 days on the designated media. M. chalcea van flavida shows satisfactory growth in the temperature range from approximately 20 to 45 "C and at a pH range from approximately 6 to 8, but best growth generally at a temperature between 26 and 37" C and at a pH-WµLW / 0PH value between 6.5 and 7.8. Little or no growth takes place at a temperature of 50 "C. M. chalcea van flavida is aerobic and reduces nitrate to nitrite.
While the M. chalcea van flavida strain designated NRRL 3222 is a particularly suitable starting material for the method primarily because of its ready availability, it will be understood that other strains of the variant and other equivalent microorganisms are acceptable alternative solutions. Furthermore, mutants of M. chalcea var. Flavida, which are obtained in that the microorganism, e.g. B. from strain NRRL 3222, which is subjected to the action of a mutant such as high-frequency radiation (including X-rays and ultraviolet radiation), actinophages and / or nitrogen mustard, may also be suitable as starting material which, after incubation under suitable conditions, provide a product that has good antibiotic activity from which the antibiotic 460 can be isolated.
Incubation of the microorganism
With controlled incubation, M. chalcea var. Flavida provides a product with good antibiotic activity, which consists of a complicated mixture of antibiotic substances from which antibiotic 460 can be isolated.
To produce this mixture, according to the invention, M. chalcea var. Flavida can be grown under aerobic conditions in the temperature range from 20 "-45" C, preferably from 25 -40 C. The incubation is preferably carried out with shaking or stirring under submerged aerobic conditions at a pH of approximately 6.0 to 8.0, a pH range of 6.5 to 7.8 frequently being particularly advantageous. The incubation time is usually 2 to 7 days, preferably 4 to 7 days, although the incubation can be shorter or longer.
The media normally used for incubation are aqueous and contain carbon and nitrogen compounds that can be assimilated by the microorganisms. Suitable carbon compounds are e.g. B. Carbohydrates such as starch, dextrin and sugar. Suitable nitrogen compounds are both organic and inorganic nitrogen compounds, preferably the first, such as. B. the commercial product NZ amine type A.
Two examples of a composition of an incubation medium suitable for the microorganism are: Medium 1: yeast extract 0.5%, fish extracts 0.1 / o, cornsteep liquor 0.1%, lactose 3% and calcium carbonate 0.1 / o; Medium II: beef extract 0.3 / 0, tryptose 0.5 / 0, dextrose 0.1 / 0 and yeast extract 0.5 / 0.
It is useful if the incubation media contain an ingredient that helps to keep the alkalinity and acidity of the media in balance. Medium 1 and II contain calcium carbonate for this purpose.
The composition thus obtained consists of a mycelium and a broth and, as a whole, shows good antibiotic activity, which is generally mainly concentrated in the broth.
Isolation of Antibiotic 460
Antibiotic 460 can be isolated from the broth by acidifying all of the incubated nutrient medium with a strong acid such as sulfuric acid to a pH of about 2.0. The mycelium is then separated off, usually by filtration, e.g. B. used a diatom filter aid. The antibiotic mixture is separated from the mycelium-free broth by one of many different methods, e.g. By forming an insoluble salt, by a resin method, or by solvent extraction.
A characteristic application of the resin method is that the broth is first neutralized with a strong alkali, e.g. B. with sodium or potassium hydroxide.
The neutralized broth is then treated with ammonium oxalate, the calcium present in the medium being precipitated as a poorly soluble oxalate salt. The calcium oxalate is separated by filtration and the antibiotic 460 is separated from the filtrate by absorption on an ion exchange resin such as a resin of the IRC-50 amberlite type. Examples of suitable IRC-50 amberlite type resins, both anion and cation exchangers, can be found in the Handbook of Chemistry and Physics, 42nd Edition, Chemical Rubber Publishing Company, Cleveland, Ohio (USA). The antibiotic is then eluted from the resin by acid treatment. The acidic eluate can then, for example, be neutralized by means of an anion exchange resin and the antibiotic solution lyophilized before it is tested by means of a standard agar diffusion method.
Separation of the antibiotic 460 into its components
Antibiotic 460, as previously mentioned, is a mixture of three related antibiotics into which it can be easily separated. Numerous methods are suitable for this separation. The division is expediently based on a chromatography method. Partition chromatography is a suitable separation process. The antibiotic mixture is passed through a column containing silica as the inert carrier material, and the column is then eluted with a solvent mixture that essentially has the following composition (in parts by volume): ethanol 20, acetic acid 8, ethyl acetate 8, water 8 and Pyridine 4.
The eluate obtained from this column contains three different fractions which are sorted in order of increasing polarity; H. decreasing mobility under the chosen conditions, fractions 1, 2 and 3 are named. Fraction 1 has a bioautographic Rr value of approximately 0.95; they usually require about 15 to 20 / o of the total eluate volume for elution; Fraction 1 consists of approximately 75 to 80 percent by weight of the total active material obtained. Fraction 2 has a bioautographic Rr value of approximately 0.25; they require about 40 to 42 / o of the total eluate volume for elution; Fraction 2 consists of approximately 10 to 15 weight percent of the total active material obtained.
Fraction 3 has a bioautographic Rr value of approximately 0.20; they require about 40 to 48 / o of the total eluate volume for elution; Fraction 3 consists of approximately 10 to 15 weight percent of the total active material obtained.
Each fraction is then concentrated to dryness by evaporation in vacuo. The residue obtained in this way is the antibiotic 460 component A, B or C, depending on which eluate fraction it originates from. The residue is then dissolved in water, the solution filtered and the filtrate lyophilized.
The antibiotic 460 and its components A, B and C can be purified, for example, by washing with a suitable organic solvent and / or by recrystallization from such a solvent. They can also be converted into derivatives such as B. in acid addition salts, in N- and O-acyl compounds, in bisulfite addition products and in sulfonamides. The following example serves to further illustrate certain features of this invention, in particular the incubation of M. chalcea van flavida, the separation of the broth from the resulting incubated nutrient medium, the isolation of the antibiotic 460 therefrom and the separation of this antibiotic mixture into its components.
Example A. Germination
100 ml of sterile medium II (see above) are incubated in a 300 ml Erlenmeyer flask with M. chalcea var. Flavida from an agar inclined plate. The flask is placed on a rotating shaker (250-275 revolutions per minute) and the culture is allowed to grow at 35 ° C. for 3 days.
This culture is sufficient to inoculate four 2 liter incubations.
B. Incubation
25 ml of the inoculum prepared above are aseptically introduced into a 2 liter Erlenmeyer flask containing 500 ml of sterile medium I. The flask is placed on a rotating shaker and the culture is incubated for 5 to 6 days at 28 ° C., the pH being maintained at approximately 7 by adding calcium carbonate several times.
The approach is tested at equal intervals during the 5 to 6 days in order to maximize its effectiveness against S.
aureus, S. lutea, B. subtilis and K. pneumoniae.
C. Separation

The antibiotic mixture is extracted from the activated charcoal by vigorously shaking or stirring it for 1 hour in 120 ml of a 1: 1 mixture of 1N sulfuric acid and methanol. The activated charcoal is then separated off by filtration, the filtrate is concentrated to 25 ml by heating in vacuo, the concentrate is lyophilized and about 50 mg of antibiotic 460 is obtained in this way.
E. Separation
420 mg of the crude antibiotic 460 (obtained by combining several batches) are dissolved in 20 ml of water and 5.0 g of silica are added to absorb the solution.
The silica is dried in vacuo and saved for further processing. A chromatography column (length 80 cm, diameter 3.5 cm) is charged with silica up to a length of 72 cm and the antibiotic silica mixture is poured into the upper part of the column from above.
The eluting solvent consisting of 20 parts of ethanol, 8 parts of acetic acid, 8 parts of ethyl acetate, 8 parts of water and 4 parts of pyridine (each by volume) is allowed to run through the column at a rate of 1.0 ml per hour and the eluate is collected in 2 , 0 ml fractions. The column is tested for antibiotic activity by wetting paper disks with liquid removed from each individual 2 ml fraction, drying the disks in a stream of dry air, and then testing for S. aureus and E. coli in the usual manner. A sample of each 2 ml fraction is paper chromatographed in descending order, working for 18 hours with a propanol-pyridine-acetic acid-water system (15: 10: 3: 12).
The papers are then dried and bioautographed on different plates against cultures of S. aureus and E. coli suspended in an agar medium.
Fractions with matching bioautography patterns are combined and the three combined fractions obtained in this way are each concentrated to dryness in vacuo. The residue obtained from each of the three fractions is dissolved in water, the solution filtered, the filtrate lyophilized and the corresponding component of the antibiotic 460 obtained (A, B or C, depending on which fraction the residue comes from).

4. Melting points
Antibiotic460 components A, B and C have no identifiable specific melting point up to 300 "C.
5. Optical rotation
Antibiotic 460 component A has a specific optical rotation of IcIII, r = + 3.5 (0.62% in water).

7. Derivatives
Chemically, antibiotic 460 and its components A, B and C are nitrogen bases and they can be converted into the various types of derivatives that are available with such bases. For example, the free bases can be converted into their acid addition salts with both organic and inorganic acids.
The derivatives, e.g. B. the acid addition salts have the antibiotic properties of the bases with only gradual differences, but are sometimes more soluble and are sometimes better than the free bases for preparations. Examples of acid addition salts are those that you can use
Hydrochloric acid, sulfuric acid, phosphoric acid, toluene sulfonic acid,
Methanesulfonic acid, succinic acid, naphthalene-l-carboxylic acid, laurylsulfuric acid, acetylsalicylic acid and cholic acid. They can be prepared by titrating an aqueous solution of the free base with the desired acid and then freeze-drying the resulting solution.

Biological properties in vitro spectra
The in vitro efficacy of the antibiotic 460 components against a number of microorganisms is set out in Table VII below. The sensitivity of the microorganisms to the antibiotics was determined using the standard tube dilution method and the growth medium used in each case consisted of 0.8% nutrient broth, 0.3% yeast extract and 0.1% glucose. The results are expressed in minimum inhibitory concentrations, dimension 11g / ml.

In vivo effectiveness
The in vivo efficacy of components A and C of antibiotic 460 against lethal infection of Staphylococcus aureus (strain Gray) and of component C against Klebsiella pneumoniae are reported in Table VIII. The tests were carried out on male mice, strain CF No. 1, weighing 18 to 22 g, by infecting the mice with an inoculum of the appropriate bacteria and then injecting them subcutaneously with the antibiotic 460 component. The results are given in PD50 values (the concentration of antibiotic which protects 50% of the infected animals), the dimension of which is milligrams of antibiotic component per kilogram of test animal.

The above biological data show that the components of the antibiotic-460 mixture are an effective group of antibiotic substances and can be used individually or as a mixture to combat infections originating from various pathogenic microorganisms, especially gram-positive microorganisms such as Staphylococcus aureus, Diplococcus pneumoniae and Streptococcus pyogenes.
These antibiotics can also be used for the sterilization of laboratory glassware, for the control and eradication of certain bacterial infections in laboratory animals, for the sterilization of operating rooms and surgical instruments and, in combination with soaps and detergents, for cleaning rooms that are used for the preparation of meals, such as hospital kitchens and dining rooms in schools.
The individual components A, B and C of antibiotic 460 are used as a standard against certain microorganisms. Component A can, for example, serve as a reference substance against which other potentially useful antibiotic substances are measured, with Staphylococcus aureus serving as the test organism. The results obtained would then be a measure of the ratio of the activity of the potential antibiotic to that of component A. Components B and C can similarly be used as a standard against Klebsiella pneumoniae or Escherichia coli. Such standards are a relatively simple method of determining the relative effectiveness of antibiotic compounds.
Therapeutic forms of the 460 antibiotics
For therapeutic purposes, antibiotic 460, or any of its components A, B, or C, or a derivative of the mixture or component, may optionally be further processed into an acceptable form.
For example, the antibiotic can be combined, for example by mixing, with a carrier to form a therapeutic preparation. One or more liquid or solid, organic or inorganic substances which are compatible with the antibiotic and suitable for the mode of administration of the antibiotic can serve as carriers. Examples of such substances are water, gelatin, lactose, a starch, magnesium stearate, talc, a vegetable oil, benzyl alcohol, a gum, a glycol and petroleum jelly.
The preparations may also contain another active ingredient, e.g. B. an analgesic, another antibiotic or a proteolytic enzyme.
