Verfahren zur Herstellung von 5-(p-Chlorphenyl)-5-hydroxy-2,3-dihydro- imidazo[2, a]isoindolen
Die Erfindung betrifft ein neues Verfahren zur Herstellung von 5-(p-Chlorphenyl)-5-hydroxy-2,3-dihy- dro-5H-imidazo[2,1-a]isoindolen der Formel I, worin R für Wasserstoff oder Chlor steht, und ihrer Säureadditionssalze.
Ein Verfahren zur Herstellung von Verbindungen der Formel I bildet bereits den Gegenstand unserer älteren Patente Nr. 500 198 und Nr. 516 573. Die Erfindung ist demzufolge gerichtet auf ein neues Verfahren zur Herstellung von Verbindungen der Formel I.
Das erfindungsgemässe neue Verfahren zur Herstellung von Verbindungen der Formel I und ihrer Säureadditionssalze ist dadurch gekennzeichnet, dass man Verbindungen der Formel II bei Temperaturen zwischen 15 und 35 C in ein polares, protonhaltiges Lösungsmittel einträgt und die erhaltenen Verbindungen der Formel I gegebenenfalls in Säureadditionssalze überführt.
Die Herstellung von Verbindungen der Formel I wird vorzugsweise so durchgeführt, dass man Verbindungen der Formel II bei Temperaturen zwischen 15 und 35 C in ein polares, protonhaltiges Lösungsmittel, beispielsweise einen niederen Alkohol, wie Methanol oder Äthanol, einträgt und das Reaktionsgemisch während 6-8 Stunden bei diesen Temperaturen rührt. Die Temperaturgrenze ist für die Reaktion kritisch und soll entsprechend eingehalten werden. Die Verbindungen der Formel I können aus dem Reaktionsgemisch in an sich bekannter Weise isoliert, gereinigt und gegebenenfalls in Säureaddditionssalze übergeführt werden.
Die Verbindungen der Formel I können in tautomeren und/oder mesomeren Formen auftreten. Die zu Verbindungen der Formel I mesomeren Verbindungen sind durch die Formel III versinnbildlicht. Die zu Verbindungen der Formel I tautomeren Verbindungen sind durch die Formel IV, und die zu Verbindungen der Formel III tautomeren Verbindungen durch die Formel V versinnbildlicht.
Die Verbindungen der Formel I besitzen, wie bereits in unseren oben erwähnten älteren Patenten Nur. 500198 und Nur. 516573 beschrieben, günstige pharmazeutische Eigenschaften, die sich insbesondere in einer appetitzügelnden sowie ZNS-stimulierenden Wirkung äussern.
Die Ausgangsprodukte der Formel II sind neu. Zu ihnen kann man gelangen, indem man Verbindungen der Formel VI, worin R obige Bedeutung besitzt, in einem inerten organischen Lösungsmittel oxydiert.
Das Verfahren kann so durchgeführt werden, dass man Verbindungen der Formel VI, gelöst in einem niedrigen Alkohol oder einem Äther, beispielsweise Diäthyläther, oder auch in einem Gemisch entsprechender Lösungsmittel während 10-20 Stunden bei 15-35 C mit Luft oder Sauerstoff in Berührung bringt. Die hierbei gebildeten Verbindungen der Formel II werden aus dem Reaktionsgemisch, das überdies noch bei der Oxydation gebildete Verbindungen der Formel I enthält, auf an sich bekannte Weise abgetrennt. Bei Verwendung von Diäthyläther als Lösungsmittel gelingt die Abtrennung und Isolierung besonders gut, da die gebildeten Verbindungen der Formel II unter bestimmten Bedingungen direkt aus der Lösung auskristallisieren und so durch Filtration isoliert werden können.
Die Verbindungen der Formel II können ebenfalls in ihrer tautomeren Form, die durch die Formel IIa wiedergegeben ist, auftreten. Der Einfachheit halber werden die Verbindungen der Formeln II und IIa unter der Formel II zusammengefassst.
Zu den Ausgangsprodukten der Formel VI und ihren Säureadditionssalzen kann man gelangen, indem man Verbindungen der Formel VII, worin R obige Bedeutung besitzt, in einem inerten organischen Lösungsmittel und in einer inerten Gasatmosphäre bei Temperaturen unter 450 C mit Lithiumaluminiumhydrid behandelt und die Verbindungen der Formel VI gegebenenfalls in Säureadditionssalze überführt.
Als inerte organische Lösungsmittel werden besonders cyclische oder offenkettige Äther, wie Tetrahydrofuran oder Diäthyläther, verwendet, und die Reaktion wird vorzugsweise während 6-96 Stunden bei 15-35 C in einer Stickstoffatmosphäre durchgeführt.
Die Verbindungen der Formel VI sind nur wenig stabil und sollen daher zweckmässigerweise ohne weitere Isolierung zur Herstellung von Verbindungen der Formel II verwendet werden. Hierzu wird das für die obige Reaktion verwendete und hierbei nicht verbrauchte Lithiumaluminiumhydrid beispielsweise durch Zugabe von Natriumhydroxid zersetzt, die Lösung anschliessend filtriert und das Filtrat zur Oxydation verwendet. Andernfalls ist es möglich, durch Behandlung des Filtrats mit einer Säure, beispielsweise gasförmigem Chlorwasserstoff, entsprechende Salze von Verbindungen der Formel VI zu erhalten.
Die Verbindungen der Formel VI können ebenfalls in ihrer tautomeren Form, die durch die Formel VIa wiedergegeben ist, auftreten. Der Einfachheit halber werden die Verbindungen der Formeln VI und VIa unter der Formel VI zusammengefasst.
Gemäss der oben beschriebenen Verfahrensweise kann man somit auch zu Verbindungen der Formel II gelangen, indem man Verbindungen der Formel VII in einem inerten organischen Lösungsmittel und in einer inerten Gasatmosphäre bei Temperaturen unter 45 C mit Lithiumaluminiumhydrid behandelt und die gebildeten Verbindungen der Formel VI in einem inerten organischen Lösungsmittel oxydiert.
Die Ausgangsprodukte der Formel VII bzw. ihre Herstellung sind beispielsweise aus der niederländischen Patentanmeldung Nr. 6 501 647 bekannt.

Beispiel 1
5-(p-Chlorphenyl)-5-hydroxy 2,3-dihydro-5H-imidazo [2, 1-a]isoindol a) 5-(p-Chlorphenyl)-2,3-dihydro-5H-imidazo [2,1-a] - isoindol
In einen mit einem Rührer, Thermometer, Rückflusskühler, Tropftrichter und Gaseinleitungsrohr versehenen und mit Stickstoff ausgespülten Kolben werden 1 000 ml trocknenes Tetrahydrofuran und 10 g (0,263 Mol) Lithiumaluminiumhydrid gegeben. Das hierbei erhaltene Gemisch wird gerührt und anschliessend tropfenweise mit einer Lösung von 30 g (0,105 Mol) 9b-(p Chlorphenyl)-1 ,2,3,9b-tetrahydro-SH-imidazo[2, 1 -a]iso- indol-5-on in 150 ml trockenem Tetrahydrofuran versetzt, wobei darauf geachtet wird, dass die Reaktionstemperatur 30 nicht überschreitet.
Das Gemisch wird während 6 Stunden bei Raumtemperatur gerührt, anschliessend im Eisbad abgekühlt, mit 100 ml Athylacetat, 20 ml einer 2N wässrigen Natriumhydroxid-Lösung und 30 ml Wasser versetzt und schliesslich durch Zusatz von 25 g wasserfreiem Natriumsulfat getrocknet. Danach wird das Gemisch unter Stickstoff abfiltriert und eine Hälfte des Filtrats,das 5-(p-Chlorphenyl)-2,3-dihydro-5H-imidazo [2, 1-a]isoin- dol enthält, mit Chlorwasserstoffgas behandelt, wobei das Hydrochlorid des 5-(p-Chlorphenyl)-2,3-dihydro5H-imidazo[2,1-a]isoindol vom Smp. 280 (Zers.) (aus Chloroform) entsteht.
b) 5-(p-Chlorphenyl)-5-hydroperoxy-2,3-dihydro 5H-imidazo[2, 1-a]isoindol
Die andere Hälfte des im Abschnitt a) beschriebenen, das 5-(p-Chlorphenyl)-2,3-dihydro- 5H-imidazo [2,1-a]isoindol enthaltenden Filtrats wird im Vakuum im Rotationsverdampfer verdampft, der Rückstand in Diäthyläther gelöst und die Lösung während 15 Stunden bei Raumtemperatur mit Luft in Berührung gebracht.
Hierbei erhält man das 5-(p-Chlorphenyl)-5-hydroperoxy-2,3-dihydro5H-imidazo[2,1-a]isoindol vom Smp. 154-157 .
c) 5-(p-Chlorphenyl)-5-hydroxy-2,3-dihydro-5H imidazo [2,1-a]isoindol
Das im Abschnitt b) hergestellte 5-(p-Chlorphenyl)-5-hydroperoxy 2,3-dihydro-5H-imidaw [2, 1-a]isoindol wird bei Raumtemperatur in Methanol eingetragen und die erhaltene Lösung während 8 Std. gerührt. Das hierbei erhaltene 5-(p-Chlorphenyl)-5-hydroxy-2, 3 -dihydro-SH- imidazo [2,1-a]isoindol schmilzt bei 202-2030.
Beispiel 2
5-(3,4-Dihchlorphenyl)-5-hydroxy-
2,3-dihydro-SH-imidazo[2,1-a]isoindol a) 5-(3 ,4-Dichlorphenyl)-2,3-dihydro-5H-imidazo- 2, 1-a]isoindol
In einen mit einem Rührer, Thermometer, Rückflusskühler, Tropfrichter und Gaseinleitungsrohr versehenen und mit Stickstoff ausgespülten Kolben werden 250 ml trockenes Tetrahydrofuran und 2,6 g (0,068 Mol) Lithiumaluminiumhydrid gegeben. Das hierbei erhaltene Gemisch wird gerührt und anschliessend tropfenweise mit einer Lösung von 20 g (0,063 Mol) 9b-(3,4-Dichlorphenyl)-1,2,3,9b tetrahydro-5H-imidazo[2,1-a]isoindol-5-on in 500ml trockenem Tetrahydrofuran versetzt, wobei darauf geachtet wird, dass die Reaktionstemperatur 30 nicht überschreitet.
Das Gemisch wird während 6 Std. bei Raumtemperatur gerührt, anschliessend im Eisbad abgekühlt und mit 5,2 ml einer 2N wässrigen Natriumhydroxidlösung und 7,8 ml Wasser versetzt.
Danach wird filtriert und das Filtrat durch Zusatz von wasserfreiem Natriumsulfat getrocknet. Nach Abfiltrieren des Natriumsulfats wird das Filtrat im Vakuum im Rotationsverdampfer verdampft. Hierbei erhält man das 5-(3,4-Dichlorphenyl)-2,3-dihydro- 5H-imidazo [2, 1-a]isoindol.
b) 5-(3,4-Dichlorphenyl)-5-hydroperoxy-2,3 dihydro-SH4midazo[2,1-a]isoindol
Das im Abschnitt a) erhaltene 5-(3,4-Dichlorphenyl)-2,3-dihydro 5H-imidazo [2,I-a]isoindol wird in Diäthyläther gelöst und die Lösung während 15 Std. bei Raumtemperatur mit Luft in Berührung gebracht. Hierbei erhält man das 5-(3,4-Dichlorphenyl)-5-hydroperoxy- 2,3-dihydro-5H-imidazo [2,1-a] -isoindol.
c) 5-(3,4-Dichlorphenyl)-5-hydroxy-2,3 dihydro-5H-imidazo[2,1-a]isoindol
Das im Abschnitt b) hergestellte 5-(3 ,4-Dichlorphenyl)-5-hydroperoxy-2,3- dihydro-5H-imidazo[2,1-a] -isoindol wird bei Raumtemperatur in Äthanol eingetragen und die erhaltene Lösung während 8 Std. gerührt. Das hierbei erhaltene 5-(3,4-Dichlorphenyl)-5-hydroxy-2,3- dihydro-5H-imidazo-[2s1-alisoindol schmilzt bei 200-201".
Process for the preparation of 5- (p-chlorophenyl) -5-hydroxy-2,3-dihydro-imidazo [2, a] isoindoles
The invention relates to a new process for the preparation of 5- (p-chlorophenyl) -5-hydroxy-2,3-dihydro-5H-imidazo [2,1-a] isoindoles of the formula I, in which R is hydrogen or chlorine stands, and their acid addition salts.

The compounds of the formula II can also occur in their tautomeric form, which is represented by the formula IIa. For the sake of simplicity, the compounds of the formulas II and IIa are combined under the formula II.
The starting materials of the formula VI and their acid addition salts can be obtained by treating compounds of the formula VII, in which R has the above meaning, in an inert organic solvent and in an inert gas atmosphere at temperatures below 450 ° C. with lithium aluminum hydride and the compounds of the formula VI optionally converted into acid addition salts.

250 ml of dry tetrahydrofuran and 2.6 g (0.068 mol) of lithium aluminum hydride are placed in a flask equipped with a stirrer, thermometer, reflux condenser, dropping funnel and gas inlet tube and flushed with nitrogen.
