worin R31 niedriges Alkyl mit 1 bis 4 Kohlenstoffatomen bedeutet und X für einen reaktionsfähigen Esterrest (beispielsweise Halogen, Methylsulfonyloxy, p-Tolylsulfonyloxy oder Schwefelsäurerest) steht.
Die Reaktion wird gewöhnlich in einem Lösungsmittel durchgeführt, indem man zuerst die Verbindung (11) in ein Alkalimetallsalz mit einem Metallatierungsmittel, wie beispielsweise metallisches Alkali (Li, Na oder K) oder einer Alkalimetallverbindung (Hydrid, Alkoxyd oder Amid eines Alkalimetalls) überführt und sodann das Alkalimetallsalz mit einer Verbindung (III) bei einer Temperatur zwischen Raumtemperatur und Rückflusstemperatur umsetzt. Ein spezifisches Beispiel zur Herstellung der Ausgangsverbindung (II) ist folgendes :
Eine Lösung von 2, 5 g 2-Jodacetamido-3-o-methoxy-benzoyl-5-äthylthiophen in 15 ml Dimethylform- amid wird unter Rühren mit 0, 42 g Natriumazid versetzt und das resultierende Gemisch 30 min lang bei 600C gerührt. Nach der Reaktion werden dem Reaktionsgemisch tropfenweise 20 ml Eiswasser hinzugefügt und wird das ganze auf Raumtemperatur abkühlen gelassen. Die auf diese Weise erhaltene rotbraune gelatinartige Sub- stanz wird mit Ligroin behandelt, wobei man weisses kristallines 2-Azidoacetamido-3-o-methoxybenzoyl-5- - äthyl-thiophen mit einem Schmelzpunkt von 83 bis 840C erhält.
Die Verbindungen gemäss Formel (I) und die für pharmazeutische Zwecke geeigneten Säureadditionssalze derselben haben eine ausgezeichnete pharmakologische Wirksamkeit hinsichtlich Narkosepotenzierung, Unter-

drückung von Aggressivverhalten und als krampfverhindernde Mittel, wie die folgenden Tests zeigen.
I. Narkosepotenzierung.
Der Einfluss einer 30 min langen Vorbehandlung mit der Testverbindung auf die Wirkung von 40 mg/kg (subnarkotische Dosis) von Hexobarbital wurde unter Verwendung von aus 6 männlichen Mäusen bestehenden Gruppen untersucht. Die Wirkung der Narkosepotenzierung durch die Testverbindung wurde durch das mehr als SOhdauerndeVerschwinden vonAggressionsreflexen bestimmt. Der Aggressionsreflex wurde 15 und 30 min nach der Verabreichung von Hexobarbital untersucht. Wenn bei jeder Zeit kein Reflex festgestellt werden konnte, so wurde das Ausmass der Narkosepotenzierung mit 100% bewertet und PD 50 (500/0 Potenziierungsdosis) der Testverbindung graphisch bestimmt.
11. Unterdrückung des Aggressionsverhaltens.
Aggressionshandlungen wurden bei den Mäusen durch die von Tedeschi et al. in Journal of Pharmacology and Experimental Therapeutics, vol. 125 [1959], S. 28ff beschriebene Methode hervorgerufen. Gruppen von 8 weiblichen Mäusen (4 Paare) wurde die Testverbindung oral 60 min vor Erzeugung des elektrischen Fussschocks über 3 min mit 530 V pulsierendem Gleichstrom, 1, 3 mA, 10 Hz/sec, verabreicht. Beim Auftreten von 3 Aggressionsepisoden oder weniger innerhalb 3 min wurde die Unterdrückung durch die Testverbindung angenom- men. Die Vergleichsmäuse von 81 Paaren zeigten Aggressionsepidosen von 8, 7 Mal des Durchschnittes unter denselben Bedingungen. Die ED , d. h. die Dosis die erforderlich ist, 50% der aggressiven Paare zu unterdrükken, wurde graphisch bestimmt.
III. Krampfverhindemde Wirkung
Pentylentetrazol (150 mg/kg) wurde 15 min nach der intraperitonealen VerabreichungderTestverbindung gruppen verabreicht, die jeweils aus 6 Mäusen bestanden. Die Anzahl der toten Mäuse wurde innerhalb 3 h nach der Verabreichung des Pentylentetrazols gezählt und sodann die ED50'die Dosis. die erforderlich war, das letale Ausmass auf 50% herabzusetzen, graphisch bestimmt.

Im Hinblick auf die verschiedenen, oben erwähnten Tests können die erfindungsgemäss herstellbaren Verbindungen der Formel (I) und deren pharmazeutische Säureadditionssalze sicher als kleine Beruhigungsmittel für die Behandlung von Neurosen, Angstgefühlen, Spannungen und Depressionszuständen in Form von pharmazeutischen Präparaten mit einem geeigneten herkömmlichen Träger oder Hilfsstoff, oral ohne Nachteil für den Patienten verabreicht werden.

Die pharmazeutischen Präparate können in jeder konventionellen Form, wie Tabletten, Kapseln oder Pulver vorliegen.
Die orale tägliche Dosis der Verbindung (I) oder eines Salzes hievon für Erwachsene liegt gewöhnlich in einem Bereich von etwa 10 bis 60 mg in Einzel- oder Mehrfachdosen.
Die Erfindung wird durch die folgenden Beispiele näher erläutert.
Beispiel l : Eine Lösung von 4 g 2-Azidoacetamido-3-o-methoxybenzoyl- 5 - äthylthiophen in 60 ml Äthanol wird mit 2 g 5% Palladium auf Holzkohle und sodann mit einer Lösung von 0, 3 g Hydrazinhydrat in 10 ml Äthanol unter Rühren versetzt, wonach das resultierende Gemisch bei Raumtemperatur weitere 2 h (starke Stickstoffgasentwicklung) und sodann weitere 30 min bei 500C gerührt wird. Der unlösliche Katalysator wird sodann abfiltriert und das Filtrat unter vermindertem Druck eingeengt.
Der ölige Rückstand wird aus Hexan umkristallisiert und ergibt blassgelbes kristallines 5-o-Methoxyphe-

Die Kristalle werden abgenutscht und aus einem Gemisch von Hexan und Äthanol umkristallisiert, wobei man weisses kristallines 5-p-Chlorphenyl-1, 6, 7-trimethyl-l, 2-di- hydro-3H-thieno [2, 3-e] [l, 4]-diazepin-2-on mit einem Schmelzpunkt von 182 bis 184 C in 76%iger Ausbeute erhält.
