Verfahren zur Substitution von an Stickstoff gebundenen Wasserstoffatomen
Die Erfindung betrifft ein Verfahren zur N-Alkylierung unter Verwendung von hochfluorierten Sulfonsäureestern als Alkylierungsmittel .
Gegenstand der Erfindung ist ein Verfahren zur Herstellung von Verbindungen der Formel R-NR,-CH2Q bzw.
R-NRI-CH2-Q-CH2NR1-R, worin die Reste R organische Reste sind, R1 für einen organischen Rest oder ein Wasserstoffatom steht und Q einen hochfluorierten, gegebenenfalls noch Chloratome aufweisenden Alkyl-, Cycloalkyl-, Oxaalkyl- oder Oxacycloalkylrest, bedeutet, der mindestens 37 Gew.% Fluor enthält, das sich dadurch auszeichnet, dass man ein oder zwei Mol einer Verbindung der Formel R-NHR1 mit einem Sulfonsäureester der Formel (RfSO20CH2)mQ, in der Rf einen hochfluorierten acyclischen oder cyclischen Rest, der 1 bis 8 Kohlenstoffatome sowie mindestens 2 Fluoratome und nicht mehr als 1 Wasserstoffatom aufweist und m 1 oder 2 bedeutet, umsetzt.
In einem Aufsatz von Burdon und McLoughlin, Tetra hydron 21, 1 1 (1965), mit dem Titel Trifluormethan- sulfonate Esters and their Alkylating Properties wird beschrieben, dass Trifluormethansulfonsäure-2,2,2-trifluoräthyl- ester aus dem Alkohol und dem Säurehydrid und weiterhin auch aus dem Alkohol und dem Säurefluorid in Gegenwart von Pyridin hergestellt werden kann. Pyridin quaternisiert jedoch mit diesem Ester sehr rasch, so dass die erzielten Ausbeuten sehr gering waren.
Die beim erfindungsgemässen Verfahren verwendeten Sulfonsäureester der Formel (RfSO20CH2)mQ, können in guten Ausbeuten dadurch hergestellt werden, dass man annähernd stöchiometrische Mengen eines hochfluorierten Alkan- bzw. Cycloalkansulfonylhalogenids der allgemeinen Formel RfSO2X, in der X Cl oder F bedeutet, und eines hochfluorierten Carbinols der allgemeinen Formel Q(CH2OH)rn in Gegenwart eines schwierig quaternisierbaren tertiären Amins, z. B. Triäthylamin (siehe die Schweizer Patentschrift Nr. 496 678) als Säureacceptor umsetzt.
In Gegenwart eines grossen Überschusses an Amin gegenüber der stöchiometrischen Menge kann Quaternisierung eintreten, wobei die Quaternisierungstemperatur um so mehr oberhalb Raumtemperatur (etwa 25 C) liegt, je höher das Molekulargewicht des Esters ist. Insbesondere bei der Herstellung der Ester der Trifluormethansulfonsäure werden daher niedrigere Temperaturen angewendet.
In den beim erfindungsgemässen Verfahren verwendeten Sulfonsäureestern ist der Rest Rf vorzugsweise ein Perfluoralkylrest mit 1 bis 8 Kohlenstoffatomen, ein Perfluorcycloalkylrest mit 4 bis 8 Kohlenstoffatomen oder ein 2-Hydroperfluoralkylrest mit 2 bis 7 Kohlenstoffatomen, wobei der Perfluormethyl-, der Perfluoräthyl- sowie der Tetrafluor äthylrest besonders bevorzugt ist.
Der hochfluorierte Alkylrest Q der Sulfonsäureester, muss mindestens 37 Gew.%, vorzugsweise mindestens 44 Gew.% Fluor enthalten. Bevorzugte Reste Q der beim erfindungsgemässen Verfahren verwendeten Sulfonsäureester sind die folgenden: (a) ein Perfluoralkylrest mit 1-17 Kohlenstoffatomen, (b) ein Perfluorcycloalkylrest mit 4-11 Kohlenstoffatomen, (c) ein -Hydroperfluoralkylrest mit einer geraden Zahl von Kohlenstoffatomen von 2-18, (d) ein 2-Hydroperfluoralkylrest mit 3-11 Kohlenstoffatomen, (e) ein ein oder zwei Chloratome aufweisender Perfluoralkylrest, (f) ein 3-Oxaperfluoralkylrest mit 36 Kohlenstoffatomen und einem Sauerstoffatom, (g) ein Perfluoroxacycloalkylrest mit 4-6 Kohlenstoffatomen, (h) ein Perfluoralkylenrest mit 1-10 Kohlenstoffatomen oder (i) ein Perfluorcycloalkylenrest mit 6-12 Kohlenstoffatomen.
Speziell bevorzugt werden von den unter a bis h genannten Resten Q die folgenden Reste: a) der Perfluormethylrest, der Perfluoräthylrest, ein Perfluorpropylrest, insbesondere der Perfluor-n-propylrest, ein Perfluorheptylrest, insbesondere der Perfluor-n-heptylrest, b) ein Perfluorcyclohexylrest, c) ein Rest der Formel-(CF2)n-CF2H, wobei n 1, 3, 5, 7 oder 9 ist, d) ein Rest der Formel -CF2-CHF-CnF2E+" worin n 5 ist, e) der Chlordifluormethylrest, ein Dichlortrifluoräthylrest, f) der Rest der Formel -(CF2)2OCF3 g) der Rest der Formel cycl-(CsF9O)- oder der Formel

Beispiele für Verbindungen, die NH- oder NH2-Gruppen aufweisen und die nach dem erfindungsgemässen Verfahren umgesetzt werden können, sind die folgenden:
: (a) aliphatische primäre und sekundäre Amine mit
1-20 Kohlenstoffatomen; (b) aromatische Amine und Aralkylamine mit 6-20 Koh lenstoffatomen; (c) heterocyclische Amine mit 2-20 Kohlenstoffatomen; (d) Hydrazinverbindungen, (e) Thioharnstoff.
Die Erfindung sei anhand der folgenden Beispiele näher erläutert, wobei, falls nicht andere Angaben vorliegen, unter den Teilen und Prozenten immer Gewichtsteile und Gewichts prozent zu verstehen sind; Gewichtsteile verhalten sich zu
Volumteilen wie g zu ml.
Beispiel 1
Ein Gemisch aus 3,0 Teilen (0,05 Mol) Äthylendiamin, 30 Volumteilen trockenem Benzol und 33,2 Teilen Trifluormethansulfonsäure-1,1-dihydropcrfluorbutylester wird auf Rückflusstemperatur erhitzt und allmählich innerhalb von 20 Minuten mit 10,1 Teilen (0,10 Mol) Triäthylamin versetzt. Nachdem eine weitere Stunde unter Rückfluss erhitzt worden ist, ist die Lösung homogen geworden. Sie wird dann abgekühlt und zweimal mit Wasser gewaschen. Die Benzolphase wird getrocknet und eingeengt und der Rückstand destilliert, wobeiN,N'-Bis-(1,1-dihydroperfluorbutyl)äthylendiamin vom Kp. 97-100 C/15 mm Hg und nD25=1,3282 erhalten wird.

Beispiel 2
Ein geeignetes Gefäss wird mit 46,4 Teilen (0,20 Mol) Trifluormethansulfonsäure-1,1-dihydroperfluoräthylester und 32,2 Teilen (0,44 Mol) Diäthylamin in 100 Volumenteilen Benzol beschickt. Nach 1stündigem Rühren und Erhitzen auf Rückflusstemperatur scheidet sich ein bräunliches Öl ab, das sich beim Abkühlen auf 0 C verfestigt. Es wird isoliert und mit kaltem Benzol gewaschen. Es handelt sich um Diäthyl ammonium-trifluormethansulfonat.
Die Benzollösung und die Waschflüssigkeit werden zusammengefasst und mit 100 Volumteilen 10 %iger wässriger Salzsäure extrahiert. Der Säureextrakt wird mit Äther gewaschen und mit Natriumhydroxyd neutralisiert. Das ölige Produkt wird mit Äther extrahiert. Diese Extrakte werden über Magnesiumsulfat getrocknet und sorgfältig eingeengt.
Dann wird durch eine Kolonne destilliert, wobei 1,1-Dihydroperfluoräthyldiäthylamin, eine Flüssigkeit vom Kp. 89-91" C/ 740 mm Hg, nD25=1,3508, in einer Ausbeute von 84% erhalten wird. Das Hydrochlorid, Fp. 16X165 C, wird analysiert: Berechnet für CJI13NF3Cl:

Beispiel 3
Dieses Beispiel erläutert die Herstellung von Dihydroperfluoralkylhydrazinen. Ein Gemisch von 26,6 Teilen des nach Beispiel 1 erhaltenen Trifluormethansulfonsäure-1,1-dihydro- perfluoroctylesters, 5 Teilen 95 %igem Hydrazin und 75 Volumteilen Isopropanol wird unter Rühren 3 Stunden auf Rückflusstemperatur erhitzt, wonach das Reaktionsgemisch in Wasser gegossen wird. Die Lösung wird mit Äther extrahiert.
Der Ätherextrakt wird mit Salzlösung gewaschen und getrocknet. Beim Sättigen der ätherischen Lösung mit wasserfreiem Chlorwasserstoff scheidet sich 1, 1-Dihydroperfluor- octylhydrazinhydrochlorid ab, das bei etwa 231-233 C schmilzt. Die freie Base, 1,1-Dihydroperfluoroctylhydrazin, wird durch 10 %ige wässrige Natronlauge freigesetzt. Das Öl hat einen Kp. von 73-75" C/5 mm Hg.

Wenn das obige Verfahren unter Verwendung anderer hochfluorierter Perfluoralkansulfonsäureester wiederholt wird, erhält man andere Hydrazine der Reihe Q(CH2NHNH2)m. Substituierte Hydrazine, wie z.B. Phenylhydrazin und dergleichen, sind bei dem obigen Verfahren ebenfalls einsetzbar und liefern Hydrazine der Reihe Q(CH2NHNHR)m, wobei R einen unsubstituierten oder substituierten Alkyl-, Aryl- oder Aralkylrest bedeutet, wie durch die folgende Tabelle VI erläutert wird.

Diese Hydrazinverbindungen sind brauchbare chemische Zwischenprodukte.
Es sei noch darauf hingewiesen, dass die beim erfindungsgemässen Verfahren eingesetzten Sulfonsäureester der Formel (RfSO20CH2)mQ nicht nur mit NH- oder NH2-Gruppen reagieren, sondern ganz allgemein mit nukleophilen Reaktionspartnern, die eine Nukleophilitätskonstante oberhalb von 1 aufweisen. Beispiele für derartige Verbindungen sind an Kohlenstoff gebundene aktive Wasserstoffatome, SH-Gruppen, OH-Gruppen und Säuren.
In speziellen Fällen kann nicht nur eine Substitution von aktiven, an Schwefel, Sauerstoff oder Kohlenstoff gebundenen Wasserstoffatomen durch die hochfluorierten Reste stattfinden, sondern es kann auch eine Addition eines solchen Restes an eine reaktive Doppelbindung stattfinden, beispielsweise an die C = C-Bindung von Enaminen, wobei dann unter gleichzeitiger Ab spaltung der Stickstoffgruppe als Produkt hochfluorierte Aldehyde und Ketone entstehen. Wenn man daher beim erfindungsgemässen Verfahren als Ausgangsmaterial Verbindungen einsetzt, die neben der NH-Gruppe oder NH2 Gruppe noch eine oder mehrere der oben genannten Gruppierungen mit einer Nukleophilitätskonstante von über 1 aufweisen, dann tritt im allgemeinen zusätzlich zu der N-Alkylierung auch noch eine Reaktion an den anderen genannten Gruppen auf.

The sulfonic acid esters of the formula (RfSO20CH2) mQ used in the process according to the invention can be prepared in good yields by adding approximately stoichiometric amounts of a highly fluorinated alkane or cycloalkanesulfonyl halide of the general formula RfSO2X, in which X is Cl or F, and a highly fluorinated carbinol of the general formula Q (CH2OH) rn in the presence of a tertiary amine which is difficult to quaternize, e.g. B. triethylamine (see Swiss Patent No. 496 678) as an acid acceptor.
In the presence of a large excess of amine compared to the stoichiometric amount, quaternization can occur, the quaternization temperature being more above room temperature (about 25 ° C.) the higher the molecular weight of the ester. Examples of compounds which have NH or NH2 groups and which can be reacted by the process according to the invention are the following:
: (a) aliphatic primary and secondary amines with
1-20 carbon atoms; (b) aromatic amines and aralkylamines with 6-20 carbon atoms; (c) heterocyclic amines having 2-20 carbon atoms; (d) hydrazine compounds, (e) thiourea.

example 1
A mixture of 3.0 parts (0.05 mol) of ethylenediamine, 30 parts by volume of dry benzene and 33.2 parts of 1,1-dihydroprofluorobutyl trifluoromethanesulfonate is heated to reflux temperature and gradually increased over 20 minutes with 10.1 parts (0.10 Mol) triethylamine added. Example 2
A suitable vessel is charged with 46.4 parts (0.20 mol) of 1,1-dihydroperfluoroethyl trifluoromethanesulfonate and 32.2 parts (0.44 mol) of diethylamine in 100 parts by volume of benzene. After stirring for 1 hour and heating to the reflux temperature, a brownish oil separates out, which solidifies on cooling to 0 C. It is isolated and washed with cold benzene. It is diethyl ammonium trifluoromethanesulfonate.
The benzene solution and the washing liquid are combined and extracted with 100 parts by volume of 10% aqueous hydrochloric acid. The acid extract is washed with ether and neutralized with sodium hydroxide. The oily product is extracted with ether. These extracts are dried over magnesium sulfate and carefully concentrated.
It is then distilled through a column, 1,1-dihydroperfluoroethyl diethylamine, a liquid with a boiling point of 89-91 "C / 740 mm Hg, nD25 = 1.3508, being obtained in a yield of 84%. The hydrochloride, melting point 16X165 C, is analyzed: Calculated for CJI13NF3Cl:

Example 3
This example illustrates the preparation of dihydroperfluoroalkyl hydrazines. A mixture of 26.6 parts of the 1,1-dihydroperfluorooctyl trifluoromethanesulfonate obtained according to Example 1, 5 parts of 95% hydrazine and 75 parts by volume of isopropanol is heated to reflux temperature for 3 hours with stirring, after which the reaction mixture is poured into water. The solution is extracted with ether.
The ether extract is washed with salt solution and dried. When the ethereal solution is saturated with anhydrous hydrogen chloride, 1,1-dihydroperfluorooctylhydrazine hydrochloride is deposited, which melts at about 231-233 ° C. The free base, 1,1-dihydroperfluorooctyl hydrazine, is released by 10% aqueous sodium hydroxide solution. The oil has a bp 73-75 "C / 5 mm Hg.

If the above procedure is repeated using other highly fluorinated perfluoroalkanesulfonic acid esters, other hydrazines of the series Q (CH2NHNH2) m are obtained. These hydrazine compounds are useful chemical intermediates.
