Die Umsetzung wird bevorzugt bei Temperaturen zwischen 50 und 180 C und vorteilhafterweise in einem organischen Lösungsmittel, z.B. Toluol, Benzol, Xylol, Chlorbenzol, o-Dichlorbenzol, Nitrobenzol, Tetrachlor- äthan, Tetrachloräthylen, Diphenyläther, Diäthyläther oder Essigsäure durchgeführt.
Als saure Kondensationsmittel kommen beispiels weise starke Säuren, insbesondere konzentrierte Schwe felsäure, in Betracht.
Andere geeignete saure Kondensationsmittel sind Phosphorsäure, Polyphosphorsäure, aroma tische oder aliphatische Sulfonsäuren, Bortrifluorid- ätherat in Gegenwart katalytischer Mengen Wasser, Zinckchlorid in Gegenwart von Eisessig und komplexe Halogensäuren, die aus Lewis-Säuren wie z.B. wasser freiem Aluminiumchlorid, Zinntetrachlorid, Antimontri- chlorid, Antimonpentachlorid, Titantetrachlorid, Eisen- -III-chIorid, Zinkchlorid und wasserfreiem Halogenwas- serstoffen in situ erzeugt werden.
Die Cyanpyridine der Formel 2 können entweder in Form der freien Base oder in Form geeigneter Salze, z.B. der Hydrogensulfate, zur Anwendung kommen. Man versetzt sie zweckmässig zunächst mit dem sauren Kondensationsmittel und trägt dann die Verbindung der Formel 3 ein. Es kann jedoch auch ein Gemisch der Formeln 2 und 3 vorgelegt und nachträglich das saure Kondensationsmittel zugegeben werden. Ausserdem be steht die Möglichkeit, das saure Kondensationsmittel auf die Umsetzungstemperatur zu erwärmen und dann in das Gemisch aus den Verbindungen der Formeln 2 und 3 einzutragen.
Bei Verwendung einer komplexen Halogensäure als saures Kondensationsmittel wird zweckmässig zunächst aus einem Cyanpyridin der Formel 2 und einem geeigne ten Metallhalogenid der entsprechende Nitrilmetallhalo genidkomplex hergestellt und dieser ohne weitere Reini gung mit einer Verbindung der Formel 3 bei Tempera turen zwischen 50 und 180 C, vorzugsweise zwischen 100 und 170 C, unter Einleiten von Halogenwasserstoff zu einem Dihydroisochinolin der Formel 1 umgesetzt.
Die Umsetzung des Cyanpyridins der Formel 2 mit der äquimolaren Menge eines Metallhalogenids erfolgt nach an sich üblichen Methoden, z.B. nach der von Kla- ges und Grill, Liebigs. Ann. Chem. 594, 29 (1955), be schriebenen Methode. Es ist dabei von Vorteil, wenn in Anwesenheit .eines inerten Lösungsmittels und bei Tem peraturen zwischen 0 und 70 C gearbeitet wird.
Das 1-(2-Pyridyl)-3,4-dihydro-isochinolin ist literatur bekannt (H. Irving und A. Hampton,]. Chem. Soc. London<I>1955,</I> Seiten 430 - 432). Diese Verbindung konnte bei der Nacharbeitung jedoch nicht rein erhalten wer den; es wurde stets eine starke Verunreinigung an 1-(2- -Pyridyl)-isochinolin und 1-(2-Pyridyl)-1,2,3,4-tetrahydro- -isochinolin festgestellt, die sich mit einfachen Mitteln nicht entfernen liess. Im Gegensatz dazu sind die nach dem erfindungsgemässen Verfahren erhaltenen 1-(2-Py- ridyl)-3,4-dihydro-isochinoline der Formel 1 frei von Disproportionierungsprodukten.
Die als Ausgangsmaterialien verwendeten Allylben- zole der Formel 3 sind grösstenteils bekannt (Christoph Rüchardt, Chem. Ber. 94, 2606, 2622, 2623 (1961); J.F. Bunnett, George T. Davis und Hiroshi Tanida, J. Amer. chem. Soc. 84, 1607 (1962); Percy Warrick, Jr. und Wil- liam H. Saunders, Jr., J. Amer. chem. Soc. 84, 4098 (1962)).
Die erfindungsgemäss hergestellten Verbindungen der Formel 1 können nach an sich üblichen Methoden mit anorganischen oder organischen Säuren in ihre physio logisch verträglichen Salze überführt werden. Als Säu ren kommen z.B. folgende in Betracht: Salzsäure, Brom wasserstoffsäure, Schwefelsäure, Weinsäure, Maleinsäure, Essigsäure, Zitronensäure.
Die Verbindungen der Formel 1 besitzen wertvolle pharmakologische Eigenschaften, insbesondere besitzen sie - bei vergleichsweise schwachen antiphlogistischen und sedativen Eigenschaften - eine beträchtliche akti vierende Wirkung auf die Fermente der Leber, im Rou tinetest gemessen durch die Verkürzung des Barbiturat schlafs von Ratten. Vor allem sind sie jedoch Ausgangs stoffe zur Herstellung von Pharmazeutica.
Zur Messung der aktivierenden Wirkung auf die Fer mente der Leber erhielten Gruppen von je 10 erwachse nen Laboratoriumsratten pro Ratte 75 mg/kg Hexobar- bital intraperitoneal verabreicht. Nach dieser Dosis be trug die Schlafdauer (Aufhebung des Umdrebreflexes) im Mittel von 10 Tieren jeweils eine bestimmte Anzahl von Minuten. Anderen Gruppen von je 10 Ratten wurden die zu prüfenden Substanzen in einer Dosierung von<I>7,5 mg/</I> kg peroral 24 Stunden vor der intraperitonealen Verab reichung von Hexobarbital gegeben.
Dabei wurde die Wirkungsdauer des Hexobarbitals bzw. die Schlafdauer wie folgt verkürzt:

Die .erfindungsgemäss erhältlichen Substanzen sind sehr wenig toxisch, so beträgt die LD50, bestimmt nach peroraler Verabreichung der Wirksubstanzen an Grup pen von je 10 Mäusen (nach einwöchiger Beobachtungs zeit) und berechnet nach der Methode von Litchfield und Wilcoxon, zum Beispiel für 3-Äthyl-3,4-dihydro-3-me- thyl-1-(2-pyridyl)-isochinolin-dihydrochlorid 1350 mg/ kg, für 3,3-Diäthyl-3,4-dihydro-1-(2-pyridyl)-isochinolin- -dihydrochlorid 1150 mg/kg.
Das nachfolgende Beispiel soll die Erfindung näher erläutern: <I>Beispiel</I> 3,4-Dihydro-3,3-dimethyl-1-(2-pyridyl)-isochinolin Die Lösung von 5,2 g 2-Cyanpyridin in 15 ml was serfreiem o-Dichlorbenzol versetzt man tropfenweise mit 13,0 g Zinntetrachlorid und leitet 5 Minuten lang trok- kenes Chlorwasserstoffgas ein. Man gibt 6,6 g (2-Me- thyl-allyl)-benzol zu, wobei die Temperatur der Mischung auf 65 bis 70 C ansteigt und erwärmt anschliessend 10 Stunden auf 130 bis 140 C. Man lässt erkalten, schüttet auf 200 g Eis und arbeitet auf. Man erhält 2,7 g an farb losen Kristallen vom Schmelzpunkt 104 bis 105 C.
Bei Verwendung von 6,65 g Aluminiumchlorid an stelle von 13,0 g Zinntetrachlorid erhält man 2,0 g der gleichen Verbindung.
Die nach dem erfindungsgemässen Verfahren herge stellten Verbindungen können zur pharmazeutischen An wendung in die üblichen pharmazeutischen Präparate eingearbeitet werden. Die Einzeldosis beträgt für Er wachsene 20 bis 300 mg, die bevorzugte Einzeldosis 25 bis 150 mg, für Kinder die Hälfte dieser Dosis.
