Die neuen Verbindungen werden erfindungsgemäss erhalten, indem man in 1-Stellung halogenierte Phthalazine der Formel

mit f-Dimethylaminoäthylalkohol in Form seiner Alkaliverbindung umsetzt. In dieser Formel steht Hal für ein Halogenatom.
Als Ausgangsstoffe für das Verfahren kommen beispielsweise folgende Phthalazinderivate in Be tracht: 1-Chlor-4-benzyl-phthalazin, 1-Chlor-4-p-chlorbenzyl-phthalazin, 1-Brom-4 p-methoxybenzyl phthalazin.
Zur Durchführung der Kondensation, die nach an sich bekannten Methoden erfolgt, lässt man z. B. auf die Lösung eines Alkalimetalls in überschüssigem ;B-Dimethylaminoäthanol das halogenierte Phthalazin in äquimolekularen Mengen (bezogen auf Alkali- metall) zunächst unter Rühren und Kühlen einwirken und vervollständigt dann die Reaktion durch mehr stündiges Erhitzen bei der Siedetemperatur des Aminoalkohols. Man kann auch so verfahren,
dass man den Aminoalkohol in Gegenwart eines mit Wasser nicht mischbaren organischen Lösungsmittels, z. B. Benzol, Toluol oder Xylol, mit der äquimoleku= Laren Menge eines Alkaliamids, vorzugsweise Na- triumamid, wie üblich behandelt und nach Beendi gung der Ammoniakentwicklung bei Siedetemperatur des Lösungsmittels mit dem halogenierten Phthalazin umsetzt.
Die Aufarbeitung der Reaktionsprodukte erfolgt in üblicher Weise, beispielsweise durch Auswaschen der Reaktionslösung mit Wasser oder durch Aus ziehen derselben mit einem mit Wasser nicht misch baren Lösungsmittel, z. B. Äther oder Benzol, und Waschen der Lösung mit Wasser. Die Gewinnung der Endprodukte kann entweder durch Vakuumdestil lation oder Überführung in geeignete Salze vor genommen werden. Die freien Basen lassen sich auch in üblicher Weise quaternisieren.
Die als Ausgangsstoffe verwendeten Phthalazin- derivate sind nach bekannten Verfahren erhältlich, z. B. durch Behandlung von in 4-Stellung aralkyl substituierten Phthalazonen mit Phosphoroxychlorid. Die in 4-Stellung substituierten Phthalazone selbst können nach gleichfalls bekannten Methoden gewon nen werden, z.
B. durch Kondensation von Hydrazin mit Benzalphthalid, p-Chlorbenzalphthalid, p-Meth- oxybenzalphthalid.
Die nach dem beanspruchten Verfahren erhält lichen Verbindungen stellen wertvolle Arzneimittel dar, die in der Literatur bisher nicht beschrieben sind. Bekannt sind lediglich basisch substituierte Bis-alkyläther des Pyridazins (Helv. Chim. Acta 37 [1954], 121 ff.) sowie basisch substituierte Mono äther des Phthalazons (1-Oxo-1,2-dihydrophthalazin),
die zusätzlich am Amidstickstoff basisch substituiert sind (DAS <B>1005</B> 072). Die bekannten Verbin dungen, von denen die letztgenannten analgetische, antipyretische und antiphlogistische Wirkung auf weisen, unterscheiden sich wesentlich von den nach der vorliegenden Erfindung erhältlichen Stoffen, und zwar sowohl bezüglich ihrer chemischen Konstitu- tion als auch ihrer pharmakologischen Eigenschaften.
Es wurde nämlich gefunden, dass sich die neuen Ver bindungen im Gegensatz zu den erwähnten bekann ten Stoffen vor allem durch histaminolytische, spasmolytische und lokalanästhetische Wirksamkeit auszeichnen, wobei insbesondere die starke, spezifi sche und langanhaltende Antihistaminwirkung der neuen Verbindungen hervorzuheben ist. Von bereits bekannten, ähnlich wirkenden Stoffen weichen sie in ihrer Konstitution weitgehend ab und stellen somit einen neuen Typ hochwirksamer Antihistaminica dar.
Sie lassen sich auch zur Herstellung von Penicillin salzen mit Depotwirkungen verwenden.
<I>Beispiel 1</I> 8,9 Gewichtsteile ss-Dimethylaminoäthylalkohol, gelöst in 75 Volumteilen Xylol, werden mit 4 Ge wichtsteilen Natriumamid unter Rühren so lange auf dem Wasserbad erhitzt, bis kein Ammoniak mehr entweicht.
Nach dem Abkühlen auf etwa 1011C werden hierzu allmählich 25,5 Gewichtsteile 1-Chlor- 4-benzyl-phthalazin, F.<B>1520</B> C [hergestellt durch Er hitzen von 4-Benzyl-phthalazon-(1) mit Phosphoroxy- chorid] zugegeben und das Gemisch einige Stunden auf etwa 1400 C erhitzt. Anschliessend wird vom Kochsalz abgenutscht und die Xylollösung mit Wasser gewaschen.
Nach dem Abdestillieren des Lö sungsmittels hinterbleiben 25,8 Gewichtsteile 1-(ss Dimethylaminoäthoxy)-4-benzyl-phthalazin (Kp o,5 201 bis<B>2080</B> C), dessen Hydrochlorid bei 178a C schmilzt. Das Jodmethylat der Base hat einen Schmelzpunkt von 186a C.
<I>Beispiel 2</I> 2,3 Gewichtsteile metallisches Natrium werden in einem Gemisch, bestehend aus 20 Gewichtsteilen ,B-Dimethylaminoäthylalkohol und 100 Volumteilen Toluol, zur Auflösung gebracht und unter Kühlen auf -f- <B>100</B> C und Rühren allmählich mit 30 Ge wichtsteilen 1- Chlor-4-p-methoxybenzyl - phthalazin [hergestellt durch Erhitzen von 4-p-Methoxybenzyl- phthalazon-(1), F. 196 C, mit Phosphoroxychlorid] versetzt. Anschliessend wird einige Stunden zum Sie den erhitzt.
Nach dem Abkühlen wird mit Wasser gewaschen und die Toluolschicht getrocknet. Nach Abdestillieren des Toluols werden 21,3 Gewichtsteile 1- (ss - Dimethylaminoäthoxy) - 4 - p - methoxybenzyl- phthalazin (Kpo,l.5 219-226 C) erhalten.
Analog hierzu werden aus 50 Gewichtsteilen - Dimethylaminoäthylalkohol, 1,2 Gewichtsteilen metallischem Natrium und 18 Gewichtsteilen 1-Brom- 4 - p - chlorbenzyl - phthalazin [hergestellt durch Er hitzen von 4-p-Chlorbenzylphthalazon-(1), F. 218 C, und Phosphorpentabromid], 14,5 Gewichtsteile 1-(ss Dimethylaminoäthoxy) - 4 - p-chlorbenzyl-phthalazin (Kp,a,s 218-222a C) erhalten, dessen Hydrochlorid bei 239-240a C schmilzt.

with f-dimethylaminoethyl alcohol in the form of its alkali compound. In this formula, Hal stands for a halogen atom.
The following phthalazine derivatives, for example, are suitable starting materials for the process: 1-chloro-4-benzyl-phthalazine, 1-chloro-4-p-chlorobenzyl-phthalazine, 1-bromo-4-p-methoxybenzyl phthalazine.
To carry out the condensation, which takes place according to methods known per se, z. B. on the solution of an alkali metal in excess; B-dimethylaminoethanol, the halogenated phthalazine in equimolecular amounts (based on alkali metal) initially act with stirring and cooling and then complete the reaction by heating for more hours at the boiling point of the amino alcohol. The free bases can also be quaternized in the usual way.
The phthalazine derivatives used as starting materials can be obtained by known processes, e.g. B. by treating phthalazones substituted in the 4-position aralkyl with phosphorus oxychloride. The phthalazones substituted in the 4-position themselves can be won by likewise known methods, for.
B. by condensation of hydrazine with benzalphthalide, p-chlorobenzalphthalide, p-methoxybenzalphthalid.
The compounds obtained by the claimed process are valuable drugs that have not yet been described in the literature. Only basic-substituted bis-alkyl ethers of pyridazine are known (Helv. Chim. Acta 37 [1954], 121 ff.) And basic-substituted monoethers of phthalazone (1-oxo-1,2-dihydrophthalazine),
which are also basic substituted on the amide nitrogen (DAS <B> 1005 </B> 072). The known compounds, the latter of which have analgesic, antipyretic and antiphlogistic effects, differ significantly from the substances obtainable according to the present invention, both with regard to their chemical constitution and their pharmacological properties.
It has been found that the new compounds, in contrast to the known substances mentioned, are primarily characterized by histaminolytic, spasmolytic and local anesthetic effectiveness, with particular emphasis on the strong, specific and long-lasting antihistamine action of the new compounds. They largely differ in their constitution from already known, similarly acting substances and thus represent a new type of highly effective antihistamines.
They can also be used for the production of penicillin salts with depot effects.
<I> Example 1 </I> 8.9 parts by weight of β-dimethylaminoethyl alcohol, dissolved in 75 parts by volume of xylene, are heated with 4 parts by weight of sodium amide on a water bath with stirring until no more ammonia escapes.
After cooling to about 1011C, 25.5 parts by weight of 1-chloro-4-benzyl-phthalazine, F. 1520 C [prepared by heating 4-benzyl-phthalazone- (1) with Phosphorus oxychloride] was added and the mixture was heated to about 1400 ° C. for a few hours. The sodium chloride is then filtered off with suction and the xylene solution is washed with water.
After the solvent has been distilled off, 25.8 parts by weight of 1- (ss dimethylaminoethoxy) -4-benzyl-phthalazine (bp 0.5201 to 2080C), the hydrochloride of which melts at 178.degree. The iodine methylate of the base has a melting point of 186a C.
<I> Example 2 </I> 2.3 parts by weight of metallic sodium are dissolved in a mixture consisting of 20 parts by weight of B-dimethylaminoethyl alcohol and 100 parts by volume of toluene and, with cooling, to -f- <B> 100 </ B > C and stirring gradually add 30 parts by weight of 1-chloro-4-p-methoxybenzyl-phthalazine [prepared by heating 4-p-methoxybenzyl-phthalazone- (1), melting point 196 C, with phosphorus oxychloride]. Then it is heated for a few hours.
After cooling, it is washed with water and the toluene layer is dried. After the toluene has been distilled off, 21.3 parts by weight of 1- (ss-dimethylaminoethoxy) -4-p-methoxybenzyl-phthalazine (Kpo, 1.5 219-226 C) are obtained.
Analogously to this, from 50 parts by weight of dimethylaminoethyl alcohol, 1.2 parts by weight of metallic sodium and 18 parts by weight of 1-bromo-4-p-chlorobenzyl-phthalazine [prepared by heating 4-p-chlorobenzylphthalazone- (1), F. 218 C, and phosphorus pentabromide], 14.5 parts by weight 1- (ss dimethylaminoethoxy) -4-p-chlorobenzyl-phthalazine (bp, a, s 218-222a C), the hydrochloride of which melts at 239-240a C.
