in Gegenwart von Cyclisierungsmitteln bei Temperatu ren zwischen 25 und 250 C cyclisiert.
Die Cyclisierung kann in der für Ringschlüsse nach Bischler-Napieralski üblichen Weise durch Behandeln mit den hierfür gebräuchlichen Cyclisierungsmitteln, z.B. Phosphoroxychlorid, Zinkchlorid, Phosphorpentachlorid und Polyphosphorsäure hergestellt werden. Besonders vorteilhaft ist die Verwendung von Phosphorpentoxid oder einem Gemisch aus Phosphoroxychlorid und Phos- phorpentoxid oder einer Mischung aus Phosphoroxy- chlorid und Zinkchlorid oder von Polyphosphorsäure. In manchen Fällen ist es von Vorteil, wenn man das Pyridincarbonsäureamid zuerst mit Phosphorpentachlo- rid und dann mit Aluminiumchlorid behandelt.
Die Cyclisierung erfolgt zweckmässigerweise in Ge genwart wasserfreier, inerter Lösungsmittel, z.B. o-Di- Chlorbenzol, Toluol, Xylol, Tetralin, Nitrobenzol, Chlo roform und bei Temperaturen zwischen 25 und 250 C. Das 1-(2-Pyridyl)-3,4-dihydro-isochinolin ist literatur bekannt (H. Irving und A. Hampton, J. Chem. Soc. Lon don<I>1955,</I> Seiten 430 - 432). Diese Verbindung konnte bei der Nacharbeitung jedoch nicht rein erhalten werden; es wurde stets eine starke Verunreinigung an 1-(2-Pyri- dyl)-isochinolin und 1-(2-Pyridyl)-1,2,3,4-tetrahydro-iso- chinolin festgestellt, die sich mit einfachen Mitteln nicht entfernen liess.
Im Gegensatz dazu sind die nach dem erfindungsgemässen Verfahren erhaltenen 1-(2-Pyridyl)- -3,4-dihydro-isochinoline der Formel I frei von Dispro- portionierungsprodukten.
Die als Ausgangsmaterial verwendeten Pyridincar- bonsäureamide der Formel II lassen sich nach üblichen Methoden erhalten, beispielsweise durch Umsetzung von Pyridincarbonsäurechloriden bzw. -estern mit den ent sprechenden Aminen.
Die erfindungsgemäss hergestellten Verbindungen der Formel I können nach an sich üblichen Methoden mit anorganischen oder organischen Säuren in ihre phy siologisch verträglichen Salze überführt werden. Als Säuren kommen z.B. folgende in Betracht: Salzsäure, Bromwasserstoffsäure, Schwefelsäure, Weinsäure, Mal einsäure, Essigsäure, Zitronensäure.
Die Verbindungen der Formel I besitzen wertvolle pharmakologische Eigenschaften, insbesondere besitzen sie - bei vergleichsweise schwachen antiphlogistischen und sedativen Eigenschaften - eine beträchtliche akti vierende Wirkung auf die Fermente der Leber, im Rou tinetest gemessen durch die Verkürzung des Barbiturat schlafs von Ratten. Vor allem sind sie jedoch Ausgangs stoffe zur Herstellung von Pharmazeutica.
Zur Messung der aktivierenden Wirkung auf die Fer mente der Leber erhielten Gruppen von je 10 erwach senen Laboratoriumsratten pro Ratte 75 mg/kg Hexo- barbital intraperitoneal verabreicht. Nach dieser Dosis betrug die Schlafdauer (Aufhebung des Umdrehreflexes) im Mittel von 10 Tieren jeweils eine bestimmte Anzahl von Minuten.
Anderen Gruppen von je 10 Ratten wurden die zu prüfenden Substanzen in einer Dosierung von 7,5 mg/ kg peroral 24 Stunden vor der intraperitonealen Verab reichung von Hexobarbital gegeben.
Dabei wurde die Wirkungsdauer des Hexobarbitals bzw. die Schlafdauer wie folgt verkürzt:

<tb> -trimethyl-isochinolin-dihydro chlorid Die erfindungsgemäss erhältlichen Substanzen sind sehr wenig toxisch, so beträgt die LD50, bestimmt nach peroraler Verabreichung der Wirksubstanzen an Grup- pen von je 10 Mäusen (nach einwöchiger Beobachtungs zeit)
und berechnet nach der Methode von Litchfield und Wilcoxon, z.B. für 3-Äthyl-3,4-dihydro-3-methyl-1- -(2-pyridyl)-isochinolin-dihydrochlorid 1350 mg/kg, für 3,3 - Diäthyl - 3,4- dihydro-1-(2 - pyridyl)-isochinolin-dihy drochlorid 1150 mg/kg.
Die nachfolgenden Beispiele sollen die Erfindung näher erläutern: <I>Beispiel 1</I> 3,4-Dihydro-3,3-dimethyl-1-(2-pyridyl)-isochinolin 12,7g N-[(1,1-Dimethyl-2-phenyl)-äthyl]-pyridin-2- -carbonsäureamid (Schmp. 76-7811C) in 50m1 Xylol rührt man 8 Stunden bei Raumtemperatur zusammen mit 20g Phosphorpentoxid und 40g Phosphoroxychlo- rid, erhitzt anschliessend 12 Stunden unter Rückfluss, trägt das Reaktionsprodukt in Wasser ein, trennt die Lösungsmittelschicht ab, schüttelt die wässrige Lösung noch mehrmals mit Wasser aus und verwirft die orga nische Phase.
Aus der sauren, wässrigen Lösung scheidet man durch Zugabe von Natronlauge das Dihydroiso- chinolin ab, nimmt es in Chloroform auf, verdampft dieses und destilliert den verbleibenden Rückstand. Man erhält 4,5 g 3,4-Dihydro-3,3-dimethyl-1-(2-pyridyl)-iso- chinolin vom Kp0,03 mm 116 C, Schmp. 104-10511C.
<I>Beispiel 2</I> 3,4-Dihydro-3,3-diäthyl-1-(2-pyridyl)-isochinolin Zu 24,2g N-[(1,1-Diäthyl-2-phenyl)-äthyl]-pyridin-2- -carbonsäureamid (Schmp. 83 - 84 C) in 400 ml o-Di- chlorbenzol gibt man 22,5 g Phosphorpentachlorid, rührt eine Stunde bei Raumtemperatur, fügt dann<I>22,5</I> a ge pulvertes, wasserfreies Aluminiumchlorid hinzu und er hitzt 90 Minuten unter Rühren bei einer Aussentempe ratur von 100 - 120 C.
Nach dem Erkalten zersetzt man durch Zugabe von Eis und Wasser und trennt die wässrige Schicht ab, die man mit Äther ausschüttelt. Die Ätherausschüttelungen werden verworfen. Aus der wässerigen Lösung macht man durch Zugabe von 40%iger Natronlauge die Base frei, nimmt sie in Chloroform auf und destilliert sie nach dem Verjagen des Lösungsmittels i.Vak. Man erhält 8 g eines gelblichen Öles von Kp0.05mm 138 C, dessen Di- hydrochlorid bei 181 C schmilzt.
In derselben Weise und in etwa derselben Ausbeute wie im Beispiel 2 beschrieben erhält man 3,4-Dihydro-3,3-dimethyl-1-(2-pyridyl)-isochinolin vom Schmp. 104 - 105 C aus N-[(1,1-Dimethyl-2-phenyl)-äthyl]-pyridin-2-carbon- säureamid, 3,4-Dihydro-3-äthyl-3-methyl-1-(2-pyridyl)-isochinolin vom Kp0,01 mm 126 - 127 C aus N-[(1-Äthyl-l-methyl-2-phenyl)-äthyl]-pyridin-2- -carbonsäureamid (Kp0,05mm 145 - 146 C), 3,4-Dihydro-3,3,4-trimethyl-1-(2-pyridyl)-isochinolin vom KP"" .
128 C aus N-[(1,1,2-Trimethyl-2-phenyl)-äthyl]-pyridin-2-car- bonsäureamid (Kp0,1mm 157 C), 3,4-Dihydro-3,3,7-trimethyl-1-(2-pyridyl)-isochinolin vom Kp0,05mm 121 C aus N-{[2-(4-Methylphenyl)-1,1-dimethyl]-äthyl}- -pyridin-2-carbonsäureamid (Kp"";""" 155 C). 7-Chlor-3,4-dihydro-3,3-dimethyl-I-(2-pyridyl)-isochino- lin dessen Dihydrobromid unter Schwarzfärbung und Zersetzung bei 240 C schmilzt, aus N-{[2-(4-Chlorphe- nyl) -1,1- dimethyl] - äthyl@ - pyridin - 2 - carbonsäureamid (Schmelzpunkt 58 - 60 C).
Das Dihydrochlorid schmilzt bei 185 C.
Die nach dem erfindungsgemässen Verfahren herge stellten Verbindungen können zur pharmazeutischen An wendung in die üblichen pharmazeutischen Präparate eingearbeitet werden. Die Einzeldosis beträgt für Er wachsene 20 bis 300 mg, die bevorzugte Einzeldosis 25 bis 150 mg, für Kinder die Hälfte dieser Dosis.
