Verfahren zur Herstellung von 2-(4'-Thiazolyl)-benzimidazol Die Erfindung betrifft ein verbessertes Verfahren zur Herstellung von 2-(4'-Thiazolyl)-benzimidazol.
Es ist bekannt, dass 2-(4'-Thiazolyl)-benzimidazol bzw.dessen Alkylderivate in der Pharmazie für die Be handlung oder Prevention der Helminthis erfolgreich verwendet werden können. (Belgische Patentschrift Nr. <B>599</B> 143). In dieser Patentschrift sind verschiedene Verfahren zur Herstellung des 2-(4'-Thiazolyl)-benzi- midazols beschrieben. So kann man Thiazol-4-carbon- säure, oder Thiazol-4-aldehyd mit o-Phenylendiamin umsetzen.
Nach einem anderen Verfahren wird Thia- zol-4-carbonsäurechlorid mit o-Nitranilin umgesetzt und das erhaltene Produkt mit Wasserstoff reduziert und bei saurem pH-Wert einem Ringschluss unterwor fen. Die bekannten Verfahren weisen den Nachteil auf, dass Thiazolylbenzimidazol-Derivate nur mit niedriger Ausbeute hergestellt werden können.
Das verbesserte Verfahren der vorliegenden Erfin dung zur Herstellung von 2-(4'-Thiazolyl)-benzimida- zol ist dadurch gekennzeichnet, dass man N-(o-Nitro-phenyl)-2-chlor- 4-thiazol-carboxamid in Gegenwart von Palladium mit Wasserstoff unter Er wärmen behandelt.
Das als Ausgangsstoff verwendete N-(o-Nitro-phenyl)-2-chlor- 4-thiazol-carboxamid kann durch Umsetzung von 2-Chlor-4-thiazol-carbon- säurechlorid und o-Nitranilin hergestellt werden.
Bei der Behandlung in Gegenwart von Palladium wird in einem Verfahrensschritt die Reduktion der Nitrogruppe, die Ausbildung des Benzimidazol-Ringes und die Entfernung des Chloratoms ausgeführt. Mit Hilfe dieses Verfahrens kann die Ausbeute, berechnet auf das Carboxamid, auf ungefähr 80-90 % erhöht werden. Die Wirtschaftlichkeit des erfindungsgemässen Verfahrens ist besonders günstig.
Die Reaktion wird vorzugsweise in Gegenwart eines mit Wasser mischbaren organischen Lösungsmit tels, wie z. B. Alkohol bei 3-5 atü Druck ausgeführt. Die Reaktion erfolgt bei erhöhter Temperatur, vorteil haft bei etwa 80-100 C. Im Laufe des Verfahrens wird die Reaktion am be sten nächst in Gegenwart von Ammoniak und dann in Schwefälsäuremedium ausgeführt. Bei dem erfindungsgemässen Verfahren wird vor zugsweise der Katalysator in mehreren Portionen dem Reaktionsgemisch zugeführt.
Weitere Einzelheiten des erfindungsgemässen Ver fahrens gehen aus dem folgenden Beispiel hervor. <I>Beispiel</I> 0,5 g 9 o%ige wässrige Palladiumlösung wird in einem Gemisch von 20 ml Wasser, 5 ml konzentrier tem Ammoniumhydroxyd und 100 ml Methanol bei 4 atü Druck und 20 C während einer Stunde mit Schüt teln vorreduziert, sodann wird 5 g N-(o-Nitrophenyl)-2- chlor-4-thiazolcarboxamid zugegeben und weiter hydriert (4 atü Druck; 20 C, 2 Stunden Schütteln), nachher werden dem Reaktionsge misch 0,5 g 9 %ige wässrige Palladiumlösung und 53 ml 5n Schwefelsäure zugegeben und die Hydrierung fortgesetzt (6 atü Druck, 80-100 C, 4 Stunden);
der Katalysator wird dann filtriert und das Methanol de stilliert. (Das Destillieren wird unter vermindertem Druck beendet). Der Destillations-Rückstand wird ab gekühlt, worauf sich 7,3 g feuchtes Thiazolyl-benzimi- dazol-sulfat ausscheiden. Dieses Produkt wird in 100 ml heissem Wasser gelöst, mit 0,5 g Tierkohle be handelt und der pH-Wert mittels wässrigem Ammoni umhydroxyd auf 7 eingestellt. Das ausgeschiedene Thiazolyl-benzimidazol wird abgesaugt, mit Wasser ge waschen und getrocknet. Die Ausbeute beträgt 3,1 g (91 C/Fp) (in geschlossenem Schmelzpunktrohr) 301- 303 C.
Das als Ausgangsstoff verwendete N-(o-Nitro-phenyl)-2-chlor-4-thiazol-carboxamid kann wie folgt hergestellt werden: 1,6 g 2-Chlor-4-thiazol-carbonsäure wird mit 5,6 ml Thionylchlorid 2 Stunden gekocht und im Vakuum zur Trockne eingedampft, wobei 2-Chlor- 4-thiazol-carbonsäurechlorid (Fp.: 114-116 C) erhal ten wird. Das Produkt wird in 15 ml Benzol gelöst, worauf 0,84g o-Nitralin in 10 ml Benzol zugesetzt werden.
Nach 5-stündigem Kochen wird zur Trockne eingedampft und der Rückstand in 100 ml Methanol heiss gelöst, mit Tierkohle geklärt und filtriert. Die ausgeschiedenen Kristalle ergeben 1,25 g des gelben kristallinen N-(o-nitro-phenyl)-2-chlor- 4-thiazol-carboxamids. Fp: 160-161 C. Analyse: N % = 14,84; Cl % = 12,96; S % = 11,44 (ber. 14,84; 12,52, 11,30). Aus der Mut terlauge werden noch weitere Mengen des Produktes erhalten.

The known processes have the disadvantage that thiazolylbenzimidazole derivatives can only be prepared with low yield.
The improved process of the present invention for the preparation of 2- (4'-thiazolyl) -benzimidazole is characterized in that N- (o-nitro-phenyl) -2-chloro-4-thiazole-carboxamide in the presence of Palladium treated with hydrogen under heat.
The N- (o-nitro-phenyl) -2-chloro-4-thiazole-carboxamide used as starting material can be produced by reacting 2-chloro-4-thiazole-carboxamide and o-nitroaniline.
In the treatment in the presence of palladium, the reduction of the nitro group, the formation of the benzimidazole ring and the removal of the chlorine atom are carried out in one process step. With the help of this process the yield, calculated on the carboxamide, can be increased to approximately 80-90%. The economy of the process according to the invention is particularly favorable.
The reaction is preferably carried out in the presence of a water-miscible organic solvent such as. B. running alcohol at 3-5 atmospheric pressure. The reaction takes place at an elevated temperature, advantageously at about 80-100 C. In the course of the process, the reaction is best carried out next in the presence of ammonia and then in sulfuric acid medium. In the process according to the invention, the catalyst is preferably added to the reaction mixture in several portions.
