Die Styrylthiazoliumsalze der Formel 1 haben sich als wirksam gegen parasitische Fadenwürmer erwiesen, besonders gegen Hakenwürmer, welche den Magen- Darmtrakt warmblütiger Tiere infizieren. So haben sich die Verbindungen als wirksam gegen Infektionen durch Syphacia obvelata bei Mäusen, durch Stronglyoides ratti bei Ratten, durch Nematospiroldes dubius bei Mäusen und durch Ancylostoma caninum und Uncinaria stenoce phala bei Hunden erwiesen. Ein bevorzugtes Salz der Formel<B>1</B> ist dasjenige mit der folgenden Strukturformel Das Jodid ist wirksam gegen Hakenwürmer bei Hun den und Katzen in Dosen von 5 mg/kg. Seine orale Lä- taldosis LD50 beträgt bei Mäusen 1650 mg/kg.
Beispiele anderer Verbindungen der Formel<B>1</B> sind: 2-(p-Pyrrolidinostyryl)-4-phenylthiazol-methyljodid; 2-(p-Pyrrolidinostyryl)-4-(p-chlorophenyl)-thiazol-methyl- jodid; 2-(p-Dimethylaminostyryl)-4-(p-biphenylyl)-thiazol- -äthyljodid; 2-(p-Dimethylaminostyryl)-4-(p-biphenylyl)-thiazol- -methyljodid; 2-(p-Pyrrolidinostyryl)-4-(p-biphenylyl)-thiazol-methyl- jodid; 2-(p-Pyrrolidinostyryl)-4-(p-biphenylyl)-thiazol-äthyl- jodid;

Geht man bei der Durchführung des erfindungsge- mässen Verfahrens von den Verbindungen der Formeln<B>3</B> und 4 aus, so kann das gebildete Thioimidat der For mel 2 isoliert werden. Man kann jedoch die Umsetzung des Phenacylderivates der Formel 3 mit dem Thiozimt- säureamid der Formel 4 unter solchen Bedingungen durchführen, dass aus dem intermediären Thioimidat eine Verbindung der Formel<B>1</B> entsteht.
Gewöhnlich genügt Erhitzen oder eine Umsetzung der Thioimidate der Formel 2 in Anwesenheit von Säure, um dieses in Verbindungen der Formel<B>1</B> umzuwandeln. Gewöhnlich wendet man Temperaturen zwischen<B>80</B> bis <B>1500C</B> an, um eine rasche Umwandlung zu bewirken. Die Säure kann eine Mineralsäure, wie beispielsweise Salzsäure oder Bromwasserstoffsäure, sein, wobei das Anion vorzugsweise mit der nucleophilen Gruppe oder mit dem Radikal<B>A</B> übereinstimmt. Die Reaktion wird vorzugsweise in Gegenwart einer Flüssigkeit, wie niedere Alkohole, beispielsweise Äthanol oder Butanol, oder einer Mischung Butanol/Wasser, durchgeführt. Die optimalen Reaktionsbedinqungen zur Herstellung von Verbindun gen der Formel<B>1</B> variieren entsprechend der Art des in termediären Thioimidats, der Gruppe A und des ver wendeten Reaktionsmediums.
Weitere Salze der Verbindungen der Formel<B>1</B> lassen sich natürlich gewünschtenfalls durch Umlagerung vor oder nach der Isolierung des Endproduktes herstellen.
Das Infrarot-Spektrum der Säureadditionssalze des Thioimidats der Formel 2 zeigt keine für die Keto-Grup- pe oder seine Enol-Form typische Absorption. Die Mes sung der magnetischen Kernresonanz lässt vermuten, dass eine Resonanzstruktur der Formel 2 a einen wesentlichen Beitrag zum Molekül liefert.

Thioimidate der Formel 2 und ihre Säureadditions salze kann um ihrerseits in Verbindungen der Formel 1 umwandeln, indem<U>man</U> sie mit einer Säure, vorzugsweise einer Mineralsäure, wie Salzsäure oder Bromwasserstoff säure, behandelt, welche mit dem Thioimidatsalz, sofern ein solches verwendet wird, und mit dem gewünschten Styrylthiazoliumsalz der Formel 1 übereinstimmen soll.
Die Säureadditionssalze des Thioimidats der Formel 2 lassen sich ebenfalls durch Erhitzen in die Verbindungen der Formel<B>1</B> überführen. Dabei verwendet man vorzugs weise eine polare Flüssigkeit, wie beispielsweise einen niederen Alkohol wie Butanol, und wendet Temperatu ren von<B>80</B> bis<B>1500C</B> an. Je nach Bedarf kann man das Styrylthiazoliumsalz durch doppelte Umsetzung in an dere Salze überführen. So lassen sich beispielsweise die Chloride und Bromide der Verbindungen der Formel 1 durch Reaktion mit Kaliumjodid, entweder vor oder nach der Isolierung des Endproduktes, in Jodide überführen.
Die Wirkung der Verbindungen der Formel<B>1</B> gegen Fadenwürmer beruht auf dem Kation und die Art des Anions ist ohne Bedeutung, vorausgesetzt, dass das Salz pharmazeutisch anwendbar ist. Beispiele geeigneter Salze sind Chloride, Bromide, Jodide, Sulfate, und Methylsul- fate, wobei die Jodide und Methylsulfate besonders ge- eignet sind. Das Bromid erhält man mittels Methyl- oder Äthylbromid.
Andere Salze erhält man mittels herkömm licher Methoden, z.B. durch Umlagerung des Anions aus einem der obenerwähnten Salze, z.B. aus dem 2-(p-R2- -Aminostyryl)-3-R1-4-Z-phenylthiazolium-Salz. Beispiels weise lässt sich das Chlorid aus dem Jodid in der Weise bilden, dass man letztere mit Silberchlorid in alkanoli- scher Lösung schüttelt.
Zur Behandlung von Fadenwurm-Infektionen bei warmblütigen Tieren verwendet man eine Verbindung der Formel 1 zweckmässig in Form einer pharmazeuti schen Zubereitung, welche die Verbindung und eine ge eignete Trägersubstanz enthält. Es können zu diesem Zweck alle bekannten pharmazeutischen Darreichungs formen angewendet werden, jedoch ist die orale Form bevorzugt. Für eine Verwendung der Verbindungen in trockener fester Form sind vor allem Kapseln, Granulate, Pillen, Pulver, Tabletten, Boli oder in einem Gelatine würfel geeignet, welche die Verbindung in der gewünsch ten Menge in gebräuchlichen Trägersubstanzen enthal ten.<B>Die</B> Zubereitungen werden durch inniges und homo genes Mischen der wirksamen Substanz mit der Träger substanz, welche auch einen oder mehrere Streckmittel, Füllmittel, Sprengmittel und/oder Bindemittel enthal ten kann, hergestellt.
Boli und Tabletten werden mit Hilfe bekannter technischer Einrichtungen geformt, zum Bei spiel durch Pressen mit einer Tablettiermaschine. Die Tabletten sollen so beschaffen sein, dass sie entweder schnell zerfallen, oder eine prothraluierte oder verzögerte Wirkung aufweisen, oder den Wirkstoff in aufeinander folgenden Zeitabschnitten feigeben. Die Tabletten können auch mit einem Überzug versehen sein. Kapseln können einfach durch Mischen der Wirksubstanz mit der Träger substanz und durch Abfüllen in Kapseln bereitet wer den.
Man kann die Verbindungen der Formel<B>1</B> auch in flüssiger Zubereitung oder als Bestandteil von Tierfutter verwenden. Flüssige Zubereitungen können den Wirk stoff entweder in einer Suspension oder Lösung in Wasser oder in pflanzlichem oder Mineralöl oder einer Emulsion davon enthalten. Die flüssige Trägersubstanz selber kann aus einem oder mehreren Stoffen bestehen, wie beispiels weise flüssigen Verdünnungsmitteln, Puffern, Bakterio- statika, Süssungsmitteln, Farbstoffen, Dispergiermitteln, Suspendiermitteln und Emulgatoren.
<I>Beispiel<B>1</B></I> 2-(p-Pyrrolidinostyryl)-3-äthyl-4-(p-biphenyl)-thiazolium- bromid Man stellt p-Pyrrolidinozimtsäure (gelbe Platten, Fp. 2480C unter Aufbrausen) nach der in J. Chem. Soc. 1930, 982 beschriebenen Methode her, wobei man von p-Pyr- rolidinobenzaldehyd (USA-Patentschrift Nr. 3 075 975) ausgeht. Zu einer Suspension von<B>21,7 g</B> dieser Säure in 150 ml Chloroform fügt man unter Rühren 12,5 g Thio- nylchlorid hinzu, dabei wird die Temperatur des Reak tionsgemisches auf<B>00</B> bis 1011C eingestellt.
Nach<B>30</B> Mi nuten dauerndem Rühren gibt man die klare Lösung portionsweise unter Rühren zu 125 ml einer 25%igen wässriggen Lösung von Äthylamin, wobei man die Tem peratur des Gemisches durch Kühlen auf 10 bis 151>C einstellt und diese Temperatur während<B>30</B> Minuten un ter Rühren aufrechterhält.
Der aus der Lösung sich abscheidende Feststoff wird ge sammelt und aus Äthanol umkristallisiert. Man erhält p-Pyrrolidinozimtsäureäthylamid mit einem Fp. bei 2010 bis 2040C.
Ein Gemisch aus 2,3 g des genannten Äthylamids 1,2 g Phosphorpentasulfid in 10 ml Pyridin wird 1 Stunde lang unter Rückfluss erhitzt und in<B>100</B> ml Wasser ge gossen. Der aus der Lösung sich abscheidende Feststoff wird gesammelt und mit Wasser gewaschen. Nach Um kristallisation aus Pyridin erhält man das reine p-Pyrro lidinozimtsäureäthylthioamid, Fp. 246 bis 2480C.
Eine Lösung von 1,3 g dieses Thioamids und 1,38 g p-Phenylphenacylbromid in 25 ml Aceton wird 2 Stun den lang unter Rückfluss. gekocht. Nach dem Abkühlen wird das gebildete p-Phenylphenacyl-N-äthyl-(pyrrolidi- nozimtsäure)-thioimidathydrobromid gesammelt. Es kri stallisiert aus Methanol/Äthylacetat in Form roter Pris men, Fp. 1701 bis 171'C.
1, 15 g des vorstehend genannten Thioimidatsalzes und 3,5 ml 47%ige Bromwasserstoffsäure werden zusammen <B>15</B> Minuten lang auf dem Dampfbad erhitzt. Nach Zu gabe von<B>10</B> ml Wasser und Neutralisation des Gemi sches mit Natriumhydroxydlösung erhält man einen ro ten Feststoff, der gesammelt und mit Wasser gewaschen wird. Nach Umkristallisation aus wässrigem Äthanol er hält man 2-(p-Pyrrolidinostyryl)-3-äthyl-4-(p-biphenyl)- -thiazoliumbromid in Form kleiner tiefroter Plättchen, Fp. 2540 bis 2560C.
Das entsprechende Jodid, welches in Form dunkel roter Platten, Fp. 2640 bis 26511C anfällt, erhält man. durch Behandeln einer methanolischen Lösung des Bro mids mit Kaliumjodidlösung.

</B> hergestellt durch Bro- mierung von 4-p-Methoxyphenylacetophenon in ätheri scher Lösung), 2,9 g p-Pyrrolidinozimtsäuremethylthio amid und 75 ml Aceton wird 1 V2 Stunden lang unter Rückfluss erhitzt. Das gebildete unlösliche 4-p-Methoxy- phenylphenacyl-N-methyl- (p-pyrrolidino-zimtsäure)-thio- imidat-hydrobromid wird gesammelt. Nach dem Umkri stallisieren aus Methanol/Äthylacetat erhält man daraus rote Nadeln, Fp. 1530 bis 1550C.
1,1 g dieses Salzes werden mit 4,5 ml Bromwasser stoffsäure 15 Minuten lang auf dem Dampfbad erhitzt. Danach werden<B>10</B> ml Wasser hinzugefügt und das Ge misch durch Zugabe von Natriumhydroxydlösung neu tralisiert. Der gebildete Feststoff wird gesammelt und mit Wasser gewaschen. Nach dem Umkristallisieren aus Me thanol erhält man kleine tiefrote Plättchen von 2-(p-Pyr- rolidinostyryl)-3-methyl- 4 - (p-methoxybiphenylyl)-thiazo- liumbromid, Fp. 2640 bis 266"C.
Das entsprechende Jodid wird durch Behandeln des Bromids mit Kaliumjodidlösung erhalten und ergibt nach Umkristallisation aus Methanol purpurrote Plättchen, Fp. <B>2791></B> bis<B>2800C.</B> Beispiel 11 2-(p-Pyrrolidinostyryl)-3-methyl-4-(p-phenoxyphenyl)- -thiazoliumchlorid 1. Eine Lösung von 620 mg p-Phenoxyphenacylchlo- rid [Cavallini u. Mitarb., J. Med. Chem. 6 (1963) 573] und 615 mg p-Pyrrolidinozimtsäuremethylthioamid in 10 ml Butanol wird 2V2 Stunden lang in einem Bad auf <B>1200C</B> erhitzt.
Nach dem Abkühlen werden durch Zu gabe von Äther aus der Lösung rote Kristalle ausge fällt, welche nach dem Umkristallisieren aus wässrigem Isopropanol tiefrote Plättchen von 2-(p-Pyrrolidinosty- ryl)-3-methyl-4-(p-phenoxyphenyl)-thiazoliumchlorid, Fp. <B>2260C</B> unter Aufbrausen, ergeben.
11. Zu einer Lösung von 1,24 g p-Phenoxyphenacyl- chlorid in 15 ml trockenem Aceton gibt man eine Lösung von 800 m- Natriumjodid in 10 ml trockenem Aceton. Nach 15 Minuten wird das ausgefallene Natriumchlorid abfiltriert.
Zu der so hergestellten Acetonlösung von p- Phenoxyphenacyljodid fügt man 1,23 g p-Pyrrolidino- zimtsäuremethylthioamid hinzu und erhitzt das Gemisch <B>50</B> Minuten lang unter Rückfluss. Das gebildete unlös liche p-Phenoxyphenacyl-N-methyl-(p-pyrrolidinozimt- säure)-thioimidat-hydrojodid kristallisiert aus Methanol/ Äthylacetat in Form feiner roter Nadeln aus, Fp. 1470 bis 1480C.
Man löst 100 m- des obengenannten Thioimidathy- drojodids in 1,5 ml Methanol, welches 0,1 ml konzen trierte Salzsäure enthält, und lässt die Lösung<B>16</B> Stun den lang bei Raumtemperatur stehen. Die gebildeten tief roten Kristalle werden gesammelt und mit Methanol und Aceton gewaschen. Das so erhaltene 2-(p-Pyrrolidino- styryl) - 3 - methyl - 4 - (p-phenoxyphenyl) -thiazoliumjodid schmilzt bei 2140C unter Zersetzung. Das gleiche Jodid wird durch Behandeln des obenbeschriebenen Chlorids mit Kaliumjodid erhalten.
<I>Beispiel 12</I> 2-(p-Dimethylaminostyryl)-3-methyl-4-(p-biphenyl)- -thiazoliumsalz Zu einer Suspension von 47,75 g p-Dimethylamino- zimtsäure (shoppee, J. Chem. Soc., 1930., 982) in 500 ml Chloroform werden unter Rühren 31,25 g Thionylchlo- rid zugesetzt und das Gemisch durch Kühlen bei 10 C gehalten. Nach 30minütigem Rühren wird die klare Lö sung in Portionen zu 400 ml 25%igem wässrigem Me thylamin unter Rühren zugegeben, wobei die Innentem peratur durch Kühlen bei 011 bis 5 C gehalten wird. Die Mischung wird weitere<B>30</B> Minuten gerührt, dann die Chloroformschicht abgetrennt, mit Wasser gewaschen, getrocknet und eingedampft.
Der zurückbleibende Fest stoff wird nach Waschen mit einer kleinen Menge Ace ton, um farbige Verunreinigungen zu entfernen, aus Ben zol umkristallisiert und liefert fast farblose Nadeln von p-Dimethylaminozimtsäuremethylamid, Fp. 1650 bis <B>1660C.</B>
Ein Gemisch aus 20,4 g des vorstehenden Methyl- amids und 11,1 g Phosphorpentasulfid in 70 ml Pyridin wird<B>15</B> Minuten unter Rückfluss erhitzt und dann in 200 ml Wasser gegossen. Der ausgefallene Feststoff wird gesammelt, gut mit Wasser und dann mit Aceton gewa schen. Nach Umkristallisatiion aus Äthanol erhält man das reine p-Dimethylaminozimtsäuremethylthioamid in Form gelber Nadeln, Fp. 2140C bis 2160C.
Eine Lösung von 8,8 g dieses Thioamids und 11,0 g p-Phenylphenacylbromid in 400 ml Aceton wird unter Rückfluss gekocht. Bald beginnt ein roter Feststoff aus zufallen, welcher gesammelt und mit Aceton gewaschen wird. Das reine p-Phenylphenacyl-N-methyl-(p-dimethyl- aminozimtsäure)-thioimidat-hydrobromid hat einen Smp. bei<B>1670</B> bis<B>1690C.</B>
5.0 g Thioimidat-hydrobromid werden in 50 ml Was ser und 80 ml 48%iger Bromwasserstoffsäure suspendiert und<B>5</B> Minuten auf einem Dampfbad erwärmt, während welcher Zeit die rote Farbe verschwindet und der suspen dierte Feststoff crèmefarbig und kristallin wird. Nach dem Abkühlen wird die Mischung mit Natronlauge neu tralisiert, ein dunkelroter Feststoff gesammelt und gut mit Wasser gewaschen.
Dieses 2-(p-Dimethylaminosty- ryl)-3-methyl-4-(p-biphenylyl)-thiozoliumbromid kristalli siert aus Methanol in Form von grün reflektierenden dunkelroten Plättchen, Schmelzpunkt<B>2530</B> bis 2550C. Das entsprechende Jodid, Fp. 2530 bis 2550C, wird durch Behandlung einer Lösung des Bromids in wässrigem Me thanol mit Kaliumjodidlösung erhalten.
Das Thiazoliumbromid wird ebenfalls durch Erwär men eines Gemisches des obigen Thioimidat-hydrobro- mids mit ungefähr 10 Gewichtsteilen n-Bromwasserstoff- säure auf dem Dampfbad während 1 Stunde und an- schliessendem Verdünnen mit Wasser und Sammeln des gebildeten Thiazoliumsalzes gewonnen, Fp. 25011 bis 2520C.
<I>Beispiel<B>13</B></I> Eine Lösung von 1,15 g p-Phenylphenacylchlorid und 1,1 g p-Dimethylaminozimtsäuremethylthioamid in 50 ml Äthanol wird 2 Stunden unter Rückfluss erhitzt und dann zur Trockne eingedampft. Der Rückstand wird aus einer Mischung von Äthanol und Äthylacetat auskristallisiert und liefert wasserhaltige rote Plättchen von p-Phenyl- phenacyl-N - methyl-(p-dimethylaminozimtsäure)-thioimi- dat-hydrochlorid, Fp. 1450 bis 1500C.
2<B>g</B> dieses Hydrochlorids werden<B>30</B> Minuten auf dem Dampfbad mit 20 ml 0,5n-Salzsäure erwärmt. Darauf werden 20 ml Wasser zugesetzt und die Lösung wird ab gekühlt. Kristallines 2-(p-Dimethylaminostyryl)-3-methyl- -4-(p-biphenyl)-thiazoliumchlorid wird gesammelt und aus einer Mischung aus Äthanol/Äthylacetat umkristalli siert. Man erhält rote Nadeln, welche bei 25111 bis 25311C schmelzen.
<I>Beispiel 14</I> p - Phenylphenacyl - N - methyl-(p-dimethylaminozimt- säure)-thioimidathydrochlorid wird wie in Beispiel 13 her gestellt. 100 mg dieses Salzes werden in 2m1 Butanol 45 Minuten auf 1200C erhitzt. Zu der abgekühlten Lösung wird Äther zugesetzt und das ausgefallene 2-(p-Dimethyl- aminostyryl)-3-methyl-4-(p-biphenylyl)-thiazoliumchlorid <U>gesammelt.</U> Es hat einen Schmelzpunkt bei<B>2510</B> bis <B>2530C</B> nach Kristallisation aus einem Gemisch von Ätha nol und Äthylacetat. Es wird in wässrigem Methanol ge löst und Kaliumjodidlösung zugefügt, worauf man das entsprechende Jodid mit einem Schmelzpunkt bei 2530C bic 2550C erhält.

<I>Beispiel<B>17</B></I> p-Pyrrolidinozimtsäure (gelbe Plättehen, Fq. 248c, un ter Aufbrausen) wird aus p-Pyrrolidinobenzaldehyd (USA-Patent Nr. 3 075 975) nach dem Verfahren von J. Chem. Soc., 1930, 982, hergestellt. Zu einer Suspen sion von<B>21,7 g</B> dieser Säure in<B>150</B> ml Chloroform wer den unter Rühren 12,5 g Thionylchlorid zugesetzt und das Gemisch durch Kühlen zwischen 0 und 10 C gehal ten. Nach Rühren während<B>30</B> Minuten wird die klare Lösung nach undnach zu 125 ml 25%iger wässriger Me thylaminlösung zugegeben, wobei die innere Temperatur durch Kühlen bei<B>10</B> bis<B>150C</B> gehalten wird. Es wird weitere<B>30</B> Minuten gerührt. Der sich absetzende Fest stoff wird gesammelt und aus Äthanol umkristallisiert.
Man erhält p-Pyrrolidinozimtsäuremethylamid als hell gelbe Nadeln, Fp. bei 2370C bis 2390C.
Ein Gemisch von 2,3 g des vorstehend genannten Me- thylamids und 1,2 g Phosphorpentasulfid in 10 ml Pyri- din wird 15 Minuten unter Rückfluss erhitzt und darauf in<B>100</B> ml Wasser gegossen. Der ausgefällte Feststoff wird gesammelt und mit Wasser gewaschen. Nach Umkristal lisation mit Pyridin erhält man das reine p-Pyrrolidino- zimtsäuremethylthioamid, Fp. bei 26211 bis 2641>C.
Eine Lösung von 1,23 g dieses Thioamids, 0,98 g Phenacylbromid und 0,5 ml 47%iger Bromwasserstoff säure in 25 ml Alkohol wird 2 Stunden unter Rückfluss erhitzt. Die sich nach dem Abkühlen ausscheidenden Kristalle von 2-(p-Pyrrolidinostyryl)-3-methyl-4-phenyl- thiazoliumbromid schmelzen bei 2360 bis 2370C unter Aufbrausen. Es wird durch Behandeln mit Kaliumjodid- lösung in das entsprechende Jodid, Fp. 233c> bis 2340C (Aufbrausen) übergeführt.
In ähnlicher Weise erhält man aus der Reaktion von p-Dimethylaminozimtsäuremethylthioamid mit p-Phenyl- phenacylbromid in Gegenwart von Bromwasserstoff säure 2-(p-Dimethylaminostyryl)-3-methyl-4-(p-biphenyl- yl)-thiazoliumbromid, Schmelzpunkt 2530 bis 2550C.

After recrystallization with pyridine, the pure p-pyrrolidinocinnamic acid methylthioamide is obtained, melting point at 26211 to 2641.degree.
A solution of 1.23 g of this thioamide, 0.98 g of phenacyl bromide and 0.5 ml of 47% hydrobromic acid in 25 ml of alcohol is refluxed for 2 hours. The crystals of 2- (p-pyrrolidinostyryl) -3-methyl-4-phenylthiazolium bromide which separate out after cooling melt at 2360 to 2370C with effervescence. It is converted into the corresponding iodide by treatment with potassium iodide solution, melting point 233c> to 2340C (effervescence).
Similarly, from the reaction of p-dimethylaminocinnamic acid methylthioamide with p-phenylphenacyl bromide in the presence of hydrobromic acid 2- (p-dimethylaminostyryl) -3-methyl-4- (p-biphenyl- yl) thiazolium bromide, melting point 2530 bis 2550C.
