Verfahren zur Herstellung von Thiazolderivaten Die Erfindung betrifft ein Verfahren zur Herstellung von neuen Thiazolderivaten, die als entzündungs- und fiebermildernde sowie schmerzlindernde Mittel wirksam sind.
Gemäss der Erfindung werden Thiazolderivate der Formel:

und deren Salze hergestellt, wobei Y oder die Gruppe -CR1R2R3 sich in der 2-Stellung des Thiazolkerns befin det, X ein Wasserstoffatom oder einen Alkylrest und Y einen Phenyl- oder Phenylalkylrest, der einen oder zwei Halogensubstituenten oder einen p-Nitro- oder p-Tri- fluormethylsubstituenten im Phenylring trägt, bedeuten und R1 ein Wasserstoffatom oder einen Alkylrest, R2 einen Alkylrest und R3 die Cyangruppe oder die Gruppe -CO2R4 darstellen, wobei R4 einen Alkyl-, Dialkyl-amino- alkyl-, Aralkyl- oder Arylrest bedeutet.
Stellt X einen Alkylrest dar, so kann dieser z.B. ein solcher mit höchstens 3 Kohlenstoffatomen, wie z.B. der Methylrest, sein.
Stellt Y einen substituierten Phenylalkylrest dar, so kann dieser z.B. ein Phenylrest mit höchstens 9 C-Ato- men, beispielsweise der Benzylrest, sein, der in der o.a. Weise substituiert ist. Der Halogensubstituent bzw. die Halogensubstituenten, die eventuell in der Gruppe Y ent halten sind, können aus Fluor-, Chlor- und Bromatomen gewählt werden.
Stellt R1 oder R2 einen Alkylrest dar, so kann dieser z.B. ein solcher mit höchstens 4 C-Atomen, beispielsweise der Methyl-, Äthyl- oder Isopropylrest, sein. Stellt R4 einen Alkylrest dar, so kann er z.B. ein Al kylrest mit höchstens 5 C-Atomen, beispielsweise der Me thyl-, Äthyl- oder n-Butylrest, sein. Stellt R4 einen Dial- kylaminoalkylrest dar, so kann er z.B. ein solcher mit höchstens 8 C-Atomen, beispielsweise der 2-Diäthylami- noäthylrest, sein. Stellt R4 einen Aralkyl- oder Arylrest dar, so kann er z.B. ein Phenylalkylrest mit höchstens 9 C-Atomen, wie z.B. der Benzylrest, oder der Phenylrest sein.
Das erfindungsgemässe Verfahren besteht darin, dass man eine Verbindung der Formel:

worin Y oder die Gruppe -CHR1R3 in 2-Stellung des Thiazolrings haftet und X, Y, R1 und R3 die oben er wähnte Bedeutung besitzen, mono- bzw. dialkyliert.
Die Alkylierung kann z.B, durch Reaktion eines AI- kalimetallderivates, z.B. des Natriumderivats des entspre chenden Thiazolderivats, mit einem Alkylhalogenid, z.B. Methyljodid, durchgeführt werden.
Die Reaktion kann in einem Verdünnungs- oder Lö sungsmittel, wie z.B. wasserfreiem Äther durchgeführt werden.
Als geeignete Säureadditionssalze können beispiels weise die Hydrochloride, Hydrobromide, Sulfate oder Phosphate erwähnt werden.
Die verfahrensgemäss als Ausgangsprodukte verwen deten Thiazolderivate können nach den bekannten all gemeinen Verfahren der heterocyclischen Chemie herge- stellt werden. Die Erfindung wird durch die folgenden Beispiele erläutert, in welchen die Teile Gew.-Teile sind. <I>Beispiel 1</I> Eine Lösung von 28 Teilen Äthyl-2-(4-chlorphenyl)- thiazol-4-ylacetat in 30 Teilen wasserfreiem Äther wird einer Lösung von 3,9 Teilen Natriumamid in 80 Teilen flüssigem Ammoniak zugegeben. Das Gemisch wird während 30 Minuten auf -70 C gerührt, es werden dann 14 Teile Methyljodid zugegeben und das Gemisch bei -70 C während 1 Stunde gerührt. 5 Teile Ammo niumchlorid werden zugegeben und das Ammoniak ver dampfen gelassen.
Der Rückstand wird mit 5%iger wäss- riger Natriumbicarbonatlösung und Äther gerührt, die Ätherschicht vom Gemisch abgetrennt, mit Wasser ge waschen, über wasserfreiem Magnesiumsulfat getrocknet, filtriert und zur Trockne eingedampft. Das verbleibende Öl wird destilliert, wobei man Äthyl-α-[2-(4-chlorphenyl)- -thiazol-4-yl]propionat, Sdp 152-153 C/0,4 mm (132- 134 C/0,12 mm), erhalten wird.
Auf dieselbe Weise erhält man unter Einsatz der ent sprechenden Ausgangsprodukte die folgenden Verbin dungen: Methyl-α-[4-(4-bromphenyl)thiazol-2 -yl]propionat, Smp. 62-63 C; Äthyl-α-[4-(4-bromphenyl)thiazol-2-yl]propionat, Sdp. 150-154 C/0,1 mm; Methyl-α-[4-(4-chlorphenyl)thiazol-2-yl]propionat, Smp. 40-42"C; Äthyl-α-[4-(4-bromphenyl)thiazol-2-yl]isovalerat (die ent sprechende Säure schmilzt bei 110-111"C unter Zers.) Äthyl-α,-[4-(4-bromphenyl)thiazol-2-yl]butyrat (die ent sprechende Säure schmilzt bei 65-67"C unter Zers.) und P-Diäthylaminoäthyl-α-[4-(4-bromphenyl)thiazol-2-yl]- - propionat - dihydrobromid - monohydrat (Zers.p. 127 130"C).
<I>Beispiel 2</I> Eine Lösung von 2,5 Teilen Äthyl-2-(4-chlorphenyl)- -thiazol-4-ylacetat in 15 Teilen wasserfreiem Äther wird einer Lösung von 0,8 Teil Natriumamid in 20 Teilen flüssigem Ammoniak zugegeben. Das Gemisch wird auf -70"C gerührt und es werden insgesamt 2,52 Teile Me- thyljodid in 2 gleichen Anteilen und nach einer Zeitspan ne von 30 Minuten zugegeben. Das Gemisch wird auf -70"C während 1 Stunde gerührt. Es werden 2 Teile Ammoniumchlorid hinzugegeben und das Ammoniak verdampfen gelassen.
Der Äther wird im Vakuum abge dampft und lässt als Rückstand das Äthyl-α-[2-(4-chlor- phenyl)thiazol-4-yl]isobutyrat; die entsprechende Säure schmilzt bei 136-138"C.
Auf dieselbe Weise jedoch unter Einsatz der entspre chenden Ausgangsprodukte werden die folgenden Ver bindungen hergestellt: Äthyl-α-[4-(4-bromphenyl)thiazol-2-yl]isobutyrat, Smp. 53-54"C; und Äthyl-α-[4-(4-chlorphenylthiazol-2-yl]isobutyrat (die ent sprechende Säure zersetzt sich bei 89,5-90,5"C).

The alkylation can, for example, by reaction of an alkali metal derivative, e.g. the sodium derivative of the corresponding thiazole derivative, with an alkyl halide, e.g. Methyl iodide.
The reaction can be carried out in a diluent or solvent, e.g. anhydrous ether.
The hydrochlorides, hydrobromides, sulfates or phosphates, for example, can be mentioned as suitable acid addition salts.
The thiazole derivatives used as starting materials according to the process can be prepared by the known general processes of heterocyclic chemistry.
