Die Erfindung betrifft ein Verfahren zur Herstellung von N-[8-(6-Purinylthio)-valeryl]-dipeptidestem der For mel V

worin n eine ganze Zahl von 0 bis 5, R" eine Alkoxy- gruppe mit gerader oder verzweigter Kohlenstoffkette mit 1 bis 8 Kohlenstoffatomen, Y Wasserstoff, einen geraden oder verzweigten Alkylrest mit 1 bis 5 Kohlenstoffato- men, einen Phenyl- oder einen 3-Indolylrest,
-CHWOH, -CHZSH, -(CH).SCH3 oder -(CHZ)mCORl, worin m eine ganze Zahl von 1 bis 3 und R, eine Alkoxygruppe mit gerader oder verzweigter Kohlenstoffkette mit 1 bis 8 Kohlenstoffatomen sind, bedeuten.
Soweit die Aminosäurereste in der Formel V ein asymmetrisches Kohlenstoffatom haben, gehören sie der L- oder D-Reihe an, oder es sind Reste von inaktiven, razeimischen Säuren.
Die Verbindungen der Formel V wurden im Rahmen der Forschung von Antimetaboliten der Purinkompo- nenten der Nucleinsäuren mit vorausgesetzter antineopla- stischer Wirksamkeit hergestellt.
Die Einführung eines Aminosäure-, Di- oder Tripeptidrestes oder Derivaten derselben, beispielsweise Estern, in das Molekül des cancerostatisch wirksamen 6-(4-Carboxybutyl)-thiopurins [M. Semonsky, A. Cerny, V. Jelinek:
Coll. Czech. Chem. Commun. 25, 1091 (1960)] beeinflusst einige Parameter der physiologischen Disposition des genannten Stoffes erheblich, was vom Gesichtspunkt der Therapie der ma lignen Neoplasien erwünscht ist. Nach Einführung der ge nannten Reste in das 6-(4-Carboxybutyl)-thiopurinmo- lekül bleibt seine geringe Toxicität erhalten, in einigen Fällen wird sie sogar stark vermindert.
Zugleich sind die antineoplastisch wirksamen Dosen der erwähnten 6-(4- -Carboxybutyl)-thiopurinderivate der Formel V bei per oraler Verabreichung an Versuchstiere mit transplantier- baren Tumoren gleich gross oder sogar niedriger als die selben des genannten Grundstoffes.
Im Vergleich mit 6-(4-Carboxybutyl)-thiopurin zeich nen sich insbesondere einige erfindungsgemäss erhältliche Derivate der N-:[8-(6-Purinylthio)-valeryl]-aminosäure- ester durch unterschiedliche Verteilung im Makroorga nismus und durch ausserordentlich hohe Affinität zu einem bestimmten Organ, bzw.
auch zum Gewebe eines bestimmten Organs und durch spezifische antineoplasti- sche Wirksamkeit bei Versuchstieren mit einem bestimm ten transplantierbaren Tumor aus, wodurch Vorausset zungen zur organgezielten Chemotherapie humaner Neo- plasien geschaffen werden.
So z.B. ist der N=[8-(6-Purinylthio)-valeryl]-glycylgly- cinäthylester spezifisch wirksam beim Sarkom SAK, (unwirksam beim Crockersarkom), beim Yoshida Asci- tereticulosarkom der Ratte, beim Adenocarcinom HK bei Mäusen H und beim Tumor S 180 der Maus H und zwar in derselben Gabe und Darreichungsart wie 6-(4- -Carboxybutyl)
-thiopurin. Die chronische Toxicität dieses Glycylglycerinderivats nach peroraler Applikation bei der Maus ist mehr als fünfmal günstiger als die des 6-(4-Carb- oxybutyl)-thiopurins.
Erfindungsgemäss werden die N=[8-(6-Purinylthio)- -valeryl]-dipeptidester der Formel V hergestellt, indem man ein N-:[g-(6-Purinylthio)-valeryl]-aminosäureazid der Formel VI

umsetzt.
Die erfindungsgemäss hergestellten Dipeptidester kön nen in die freien Säuren überführt werden, indem man sie alkalisch verseift und die erhaltene Lösung ansäuert. Die Dipeptide mit einer freien Carboxylgruppe können dann zur Herstellung der entsprechenden Tripeptidester ver wendet werden, indem man sie mit einem Aminosäure ester der Formel Ill umsetzt.
Gemäss einer bevorzugten Ausführungsform der Er findung wird die Umsetzung des Säureazids der Formel VI mit dem Aminosäureester III in einem inerten, orga nischen Lösungsmittel, vorzugsweisve in Dimethylform- amid, durchgeführt.
Die alkalische Verseifung der erfindungsgemäss er hältlichen N-[8-(6-Purinylthio)-valeryl]-dipeptidesterkann vorzugsweise bei Temperaturen zwischen 0 und 25 C durch Einwirkung einer wässrigen Alkalihydroxyd- lösung, vorzugsweise Natrium- oder Kaliumhydroxydlö- sung durchgeführt werden, wobei im Falle eines Amino- monocarbonsäureesters mindestens zwei Äquivalente und im Falle eines Aminodicarbonsäurediesters mindestens drei Äquivalente der Base verwendet werden.
Vorzugsweise führt man die Reaktion der freien N-:[ä- -(6-Purinylthio)-valeryl]-dipeptidsäure mit einem Amino- säureester der Formel III zum entsprechenden N-:[8-(6- -Purinylthio)-valeryl]-tripeptidester in einem inerten orga nischen Lösungsmittel, vorzugsweise einem halogenierten, aliphatischen Kohlenwasserstoff mit 1 oder 2 Kohlenstoff atomen, insbesondere in Methylenchlorid, durch.
Die für die Durchführung des erfindungsgemässen Verfahrens als Ausgangsmaterial erforderlichen N-[8-(6 -Purinylthio)-valeryl]-aminosäureazide der Formel VI kann man aus den entsprechenden Aminosäureestern, de ren Herstellung in der schweizerischen Patentschrift <B>501652</B> beschrieben ist, über die Hydrazide leicht herstel len.
Bei dem erfindungsgemässen Verfahren erfolgt im allgemeinen keine Racemisierung der optisch aktiven Aminosäuren.
Im folgenden werden an Hand von Beispielen bevor zugte Ausführungsformen der Erfindung näher erläutert. Die Schmelzpunkte sind am Koflerblock ermittelt und in Celsiusgraden angegeben. <I>Beispiel 1</I> N-[e-(6-Purinylthio)-valeryl]-glycylglycinäthylester Eine Lösung v.
3,23 g (0,01 Mol) N-[8-(6-Purinylthio)- -valeryl]-glycinhydrazid in 200 ml 0,1 m HCl versetzt man bei 0 bis 5 C unter Rühren tropfenweise mit 10 ml 0,1 m NaNO.= und rührt das Reaktionsgemisch bei derselben Temperatur bis zum Verschwinden der Reaktion auf Jodstärkepapier (etwa 1 Stunde).
Die entstandene Suspen sion des Säureazidhydrochlorids neutralisiert man mit 10 ml 1 m NaHCQ, gibt 100g Ammoniumsulfat zu, und nach 372 Stunde Rühren bei 0 bis 5 C saugt man das ausgeschiedene Azid scharf ab, wäscht es mit 20 ml Was ser, suspendiert es in 25 ml Dimethylformamid, versetzt die Suspension mit<B>5,15</B> g (0,05 Mol) frisch destilliertem Glycinäthylester und lässt das Reaktionsgemisch 2 Tage bei Raumtemperatur stehen.
Nach Abdestillieren des Di- methylformamids im Vakuum bei 50 bis 60 C, versetzt man den Rückstand mit 10 ml Wasser; den pH-Wert des Gemisches stellt man mit Essigsäure auf 6 ein und kri stallisiert den ausgeschiedenen N-:[8-(6-Purinylthio)-vale- ryl]-glycylglycinäthylester aus wässrigem Äthanol um. Man erhält farblose Nädelchen; F. 227 bis 229 ; Ausbeu te 2,80 g<B>(717,).</B>
Das erforderliche N-:[8-(6-Purinylthio)-valeryl]-glycin- hydrazid stellt man auf folgende Weise her: 17,5 ml Hy- drazinhydrat versetzt man unter Rühren bei 20 C mit 3,5 g N-[8-(6-Purinylthio)-valeryl]-glycinäthylester und lässt die Lösung 2 Tage bei Zimmertemperatur stehen.
Nach Abdestillieren des überschüssigen Hydrazinhydrats im Vakuum rührt man den Rückstand mit 7 ml Wasser an, stellt den pH-Wert des Gemisches mit verdünnter Salzsäure auf 7 ein und lässt bei 5 C kristallisieren. Das ausgeschiedene N=[8-(6-Purinylthio)-valeryl]-glycinhydra- zid reinigt man durch Umkristallisieren aus Wasser; Ausbeute 3,05 g (91%); F. 231 - 233 .
<I>Beispiel 2</I> N-:[8-(6-Purinylthio)-valeryl].-glycylglycylglyciniithylester Zu einer Suspension von 3,66 g (0,01 Mol) N=[8-(6 -Purinylthio)-valeryl]-glycylglycin in 36 ml wasserfreiem Methylenchlorid gibt man 1,24 g (0,0l2 Mol) frisch de stillierten Glycinäthylester und danach eine Lösung von 2,27 g (0,011 Mol) N,N'-Dicyclohexylcarbodiimid in 11 ml Methylenchlorid und lässt das Reaktionsgemisch bei 20 C 24 Stunden stehen.
Die entstandene Suspension versetzt man mit 50 ml 90a/oigem wässrigem Äthanol und 1 ml Eisessig, nach 1 Stunde saugt man den ausgeschiedenen N,N'-Dicyclohexylharnstoff ab und dampft das Filtrat zur Trockne ein (bei 40 bis 50 C).
Den Rückstand rührt man zwecks Entfernung der nicht umgesetzten Ausgangssäure 1 Stunde mit 25 ml 1 m NaHC 3, den ungelösten Anteil saugt man ab und chromatographiert ihn an einer Sili- cagelsäure (100 g) unter Verwendung eines Gemisches von Chloroform mit 20% Methanol als Fluierungsmittel, in Fraktionen je 25 ml. In den Ausgangsfraktionen (1. bis 4.) befindet sich überwiegend noch der N,N'-Dicyclohexyl- harnstoff, in weiteren Fraktionen (6.
bis 11.) der praktisch reine N=[8-(6-Purinylthio)-valeryl]-glycylglycylglycinäthyl- ester. Ausbeute 2,15 g, d.i. 48%. Nach Umkristallisieren aus 50%igem wässrigem Äthanol bildet dieser Stoff farb lose Nädelchen; F. 238 bis 240 .
<I>Beispiel 3</I> N=[8-(6-Purinylthio)-valsryl]-glycylglycylgl ycln Eine Lösung von 0,44 g (11 m Mol) Natriumhydroxyd in 20 ml Wasser versetzt man tropfenweise unter Rühren bei 0 bis 5 C mit 5 m Mol N-:[8-(6-Purinylthio)-valeryl]- -glycylglycylglycinäthylester und lässt die Lösung zwei Tage bei derselben Temperatur stehen.
Dann säuert man sie mit verdünnter Salzsäure bis zu einem pH-Wert von 2 bis 3 an und kristallisiert das ausgeschiedene N-:[8-(6- -Purinylthio)-valeryl]-glycylglycylglycin aus Wasser um; der Schmelzpunkt ist F. 231 bis 233 C (Wasser).
Auf dieselbe, in Beispiel 1 beschriebene Weise, wur den in Ausbeuten von 60 bis 80% folgende N-:[8-(6-Puri- nylthio)-valeryl]-dipeptidester hergestellt: N-[8-(6-Purinylthio)-valeryl]-glycyl-L-leucinäthylester, F. 130 - 140 (Aceton-Hexan, fx]DZ = -8,7o (c = 1;
Pyridin); N-:[8-(6-Purinylthio)-valeryl]-glycyl-DL-isoleucinäthyl- ester, F. 141 -- 143 (Äthanol-Hexan). Auf dieselbe, im Beispiel 3 beschriebene Weise, wur- den in Ausbeuten von 95 - 100% folgende N-[8-(6-Pu- rinylthio)-valeryl]-dipeptide hergestellt:
N.#[8-(6-Purinylthio)-valeryl]-glycylglycin, F. 213 - 214 (Wasser); N-[8-(6-Purinylthio)-valeryl]-glycyl-L-leucin, F. 157 - 158 (Wasser); [a]D20 = -9,5o (c = 1, 0,1 m NaOH); N-@[8-(6-Purinylthio)-valeryl]-glycyl-DL-isoleucin, F. 163 -165 (Wasser, Äthanol).

The introduction of an amino acid, di- or tripeptide residue or derivatives thereof, for example esters, into the molecule of the cancerostatically active 6- (4-carboxybutyl) thiopurine [M. Semonsky, A. Cerny, V. Jelinek:
Coll. Czech. Chem. Commun. 25, 1091 (1960)] influences some parameters of the physiological disposition of the substance mentioned considerably, which is desirable from the point of view of the therapy of ma lignen neoplasms. At the same time, the antineoplastically effective doses of the 6- (4- carboxybutyl) -thiopurine derivatives of the formula V when administered orally to test animals with transplantable tumors are the same or even lower than those of the basic substance mentioned.
In comparison with 6- (4-carboxybutyl) -thiopurine, some derivatives of the N-: [8- (6-purinylthio) -valeryl] -amino acid esters which can be obtained according to the invention are particularly distinguished by different distribution in the macroorganism and by extremely high affinity to a certain organ or
also to the tissue of a certain organ and through specific antineoplastic efficacy in test animals with a certain transplantable tumor, which creates the prerequisites for organ-targeted chemotherapy of human neoplasms.
