Process for the preparation of 3-mono- or 3,17-diesters, substituted in the 17-position by an unsaturated hydrocarbon radical, of 11,13P-dialkylgon-4-ene-3,17p-diols The invention relates to a process for the preparation of in 17- Position by an unsaturated hydrocarbon radical substituted 3-mono- or 3,17-diesters of 11,13 # -dialkylgon-4-ene-3,17 # -diols. The compounds which can be prepared according to the invention have the following formula
EMI0001.0003
in which R and R 'are alkyl radicals, X is an unsaturated aliphatic hydrocarbon radical, Y is hydrogen or an alkanoyl radical, Z is an alkanoyl radical,
the wavy line indicates the alternate stereochemical a- or p-configuration and each of the alkyl, unsaturated aliphatic hydrocarbon or alkanoyl radicals contains fewer than 8 carbon atoms.
Examples of the alkyl radicals symbolized by R and R 'are methyl, ethyl, propyl, butyl, pentyl, hexyl, heptyl and their branched-chain isomers.
Typical of the alkanoyl radicals represented by Y and Z are formyl, acetyl, propionyl, butyryl, valeryl, caproyl, heptanoyl and their branched-chain isomers.
Typical examples of the unsaturated aliphatic hydrocarbon radicals symbolized by X are ethynyl, vinyl, propynyl, allyl, butynyl, propargyl, metallyl, etc. The new compounds of the formula (1) are prepared according to the invention, by reacting the hydroxyl group with a corresponding 17-alkanoyloxy-11,13 # -dialkylgon-4-en-3-ol or 11,13p-dialkylgon-4-en-3,17 # diol by reaction with a reactive alkanoic acid derivative esterified in the 3-position or both hydroxyl groups.
If 3,17P-diols are present, the esterification is preferably carried out by reaction with an alkane carboxylic acid anhydride or halide at room temperature, in particular in the presence of a suitable acid acceptor, e.g. Pyridine, made to the corresponding 3-monoalkanoate of the formula (1), in which Z is an alkanoyl radical and Y is hydrogen. In the esterification of 3,17ss-diols at elevated temperature, the 3,17-bisalkanoate of the formula 1 can be obtained, in which both radicals Y and Z are alkanoyl groups.
On the other hand, the esterification of 17-alkanoyl oxy compounds at room temperature gives 3,17-bisalkanoates of the formula 1.
The compounds which can be prepared according to the invention and in which the 13P-alkyl radical has more than one carbon atom are generally obtained as dl mixtures. The separation of these dl compounds to the individual d- and l-enantiomorphic compounds is expediently carried out by esterification of the hydroxyl groups with the anhydride of a dibasic carboxylic acid, such as succinic or phthalic acid hydride, with the formation of the corresponding half-esters, which with the aid an optically active amine, preferably brucine, morphine, quinine, quinidine or strychnine, splitting the salts obtained into the diastereoisomeric forms and recovering the steroids.
The compounds obtainable according to the invention are more useful because of their valuable pharmacological properties. In particular, they are hormonal agents with strong progestational (the processes that occur immediately before the menses can be influenced) and deeiduogenic (influencing the rejection of the uterine mucous membrane) effectiveness. In the following examples, which explain the process according to the invention, the temperatures are expressed in degrees Celsius and the amounts of material in parts by weight, unless otherwise stated.
The ratio between parts by weight and parts by volume is the same as that between grams and milliliters. Example1 A mixture of 1 part 17α-ethynyl-11p-methylestr--4-en-3p, 17 # -diol, 5 parts acetic anhydride and 10 parts pyridine becomes He warms up on the steam bath for about 18 hours, then cooled and concentrated to dryness under reduced pressure.
