In dieser Formel bedeuten: R1 und R2, die gleich oder verschieden sein können, Wasserstoff- oder Halogenatome, Niederalkyl- oder Niederalkoxygruppen, die Trifluormethyl-, Nitril- oder Hydroxylgruppe oder zusammen mit dem aromatischen Ring einen Naphthyl-, Indanyl-, Tetrahydronaphthyl-, Methylendioxyphenyl- oder Benzodioxanylrest, R3 ein Wasserstoffatom, die Methyl- oder Äthylgruppe oder eine gegebenenfalls mit einer Niederalkylgruppe ein- oder mehrfach substituierte Phenylgruppe, R4 ein Was serstoffatom, die Methyl- oder Äthylgruppe oder eine Phenylgruppe, die gegebenenfalls durch ein Halogen atom oder eine Niederalkyl- oder Niederalkoxygruppe substituiert ist, und R5 ein Wasserstoffatom, eine Nie deralkylgruppe oder eine Benzylgruppe,
die gegebenen falls durch ein Halogenatom und/oder eine Niederalkyl gruppe substituiert ist, und X ein Sauerstoffatom oder die Methylengruppe. Unter niederen Alkylresten bzw. Alkoxygruppen sind vorzugsweise solche mit 1-4 Koh lenstoffatomen zu verstehen.
Die erfindungsgemäss erhältlichen Verbindungen enthalten mindestens zwei Asymmetriezentren, näm lich die Kohlenstoffatome 4 und 5 der Formel I. Sie können dementsprechend in vier stereoisomeren For men existieren, von denen je zwei ein Paar optisch ak tiver Antipoden darstellen. In dem einen Paar befinden sich die 5ständige Hydroxylgruppe und die 4ständige Aminogruppe in trans-, in dem anderen in cis-Stellung. Wenn R4 eine andere Bedeutung als Wasserstoff besitzt, so besitzen die erfindungsgemäss erhältlichen Verbindungen ein weiteres Asymmetriezentrum. Die Zahl der stereoisomeren Formern erhöht sich dann auf B.
Die jeweiligen optisch aktiven Antipoden bilden in üblicher Weise Racemate.

worin R1 eine hydrolytisch oder hydrogenolytisch ab spaltbare Schutzgruppe bedeutet, wobei, falls R;, eine gegebenenfalls substituierte Benzylgruppe bezeichnet, R' eine hydrolytisch abspaltbare Schutzgruppe ist, die Schutzgruppe abspaltet.
Als Schutzgruppe kommen beispielsweise ein Acyl- oder der Benzylrest in Betracht. Zur Herstellung von Verbindungen der Formel I, in denen R, einen Benzyl rest bezeichnet, sind nur Ausgangsverbindungen mit einer hydrolytisch abspaltbaren Schutzgruppe geeignet.
Nach diesem Verfahren entstehen nebeneinander die Racemate der cis- und der trans-Form. Eine Tren nung der beiden Racemate kann durch fraktionierte Kristallisation oder Chromatographie an Kieselgel er folgen. Die Verbindungen, bei denen einer der Reste R;, und R,, ein Wasserstoffatom bedeutet (sekundäre Aminoverbindungen), können auch durch Behandlung des Isomerengemisches mit Aldehyden oder Ketonen, vorzugsweise mit Benzaldehyd oder p-Nitrobenzaldehyd aufgetrennt werden, da die trans-Verbindungen der Formel I mit diesen Substanzen zu Oxazolidinen rea gieren, während die cis-Verbindungen dabei unverändert bleiben.
Das gebildete Oxazolidin lässt sich aufgrund seiner stark veränderten Lösungseigenschaften leicht von der unveränderten cis-Verbindung abtrennen und anschliessend wieder durch Behandeln mit verdünnter Mineralsäure in die Ausgangs-Komponenten zerlegen.
Die Racemate der cis- bzw. trans-Form können ihrerseits nach üblichen Verfahren, beispielsweise durch Salzbildung mit optisch aktiven Hilfssäuren wie Di- benzoyl-D-weinsäure oder (+)3-Bromcampher-8-sul- fonsäure, anschliessende fraktionierte Kristallisation der diasteromeren Salze und Freisetzung der Basen, in ihre optisch aktiven Antipoden übergeführt werden.
Die vorstehend beschriebenen Abspaltungsreaktio nen lassen sich sowohl in der cis- als auch in der trans- Reihe ausführen. Dabei können sowohl racemische als auch optisch aktive Ausgangsmaterialien eingesetzt werden.
Die Substanzen der Formel 1 sind infolge der An wesenheit einer Aminogruppe im Molekül Basen und werden daher für eine therapeutische Anwendung zweckmässig in üblicher Weise in ihre physiologisch verträglichen Säureadditionssalze überführt. Dies kann beispielsweise durch Umsetzung mit geeigneten Säuren geschehen. Als geeignete Säuren kommen z. B. in Frage: Salzsäure, Bromwasserstoffsäure, Schwefelsäure, Bern steinsäure, Maleinsäure, Weinsäure, Milchsäure, Me- thansulfonsäure oder saure Harze vom vernetzten Polystyrol-Typ, die Sulfonsäuregruppen enthalten, wie Zeo-Karb 225 (eingetragenes Warenzeichen).
Die Verbindungen der Formel 1 haben interessante pharmakologische, insbesondere antidepressive und antikonvulsive Eigenschaften bei geringer Toxizität. Als besonders wertvoll haben sich dabei die Verbindungen der trans-Reihe erwiesen, und zwar insbesondere solche, bei denen R,, R_, und R7, eine niedere Alkylgruppe (vor zugsweise Methyl) und R " Wasserstoff bedeuten, wäh rend R., und X die oben angegebene Bedeutung haben (dabei kann auch lediglich einer der Reste R, und R2 niederes Alkyl sein, während der andere eine der übri gen für R, und R2 möglichen Bedeutungen hat).
Eine besonders ausgeprägte antidepressive Wirkung haben beispielsweise die optischen Antipoden und das Race- mat der trans-Form des 4-Methyl-amino-5-hydroxy-7,8-dimethyl-2,3,4,5- tetrahydro-1-benzoxepin-hydrochlorids, des 4-Methyl-amino-5-hydroxy-7,9-dimethyl-2,3,4,5- tetrahydro-l-benzoxepinhydrochlorids, des 4-Methyl-amino-5-hydroxy-7,8-dimethyl-2,3,4,5- tetrahydro-benzocycloheptens und des 4-Dimethylamino-5-hydroxy-7,8-dimethyl-2,3,4,5- tetrahydro-1-benzoxepinhydrochlorids gezeigt;
während sich die optischen Antipoden und das Racemat der trans-Form des 4-Methylamino-3,7,8-tri- methyl-5-hydroxy-2,3,4,5-tetrahydro-1-benzoxepins als gute Antikonvulsiva herausgestellt haben. Als Dosis für die orale Anwendung der trans-Ver- bindungen der Formel 1 werden 1 bis 250 mg als Ein zeldosis und 30 bis 500 mg als Tagesdosis vorgeschla gen; für die drei formelmässig bezeichneten Racemate und ihre optisch aktiven Antipoden kommt beispiels weise eine Einzeldosis von 2 bis 75 mg und eine Tages dosis von 30 bis 150 mg in Frage.
Die erfindungsgemäss erhältlichen Verbindungen können allein oder in Kombination mit anderen erfin dungsgemässen Wirkstoffen, gegebenenfalls auch in Kombination mit weiteren pharmakologisch aktiven Wirkstoffen wie Sympathicomimetika oder Psychophar maka, zur Anwendung gelangen. Geeignete Anwen dungsformen sind beispielsweise Tabletten, Kapseln, Zäpfchen, Lösungen, Säfte, Emulsionen oder dispersible Pulver.
Entsprechende Tabletten können beispielsweise durch Mischen des oder der Wirkstoffe mit bekannten Hilfsstoffen, beispielsweise inerten Verdünnungsmitteln, wie Calciumcarbonat, Calciumphosphat oder Milch zucker, Sprengmittel, wie Maisstärke oder Alginsäure, Bindemitteln, wie Stärke oder Gelatine, Schmiermitteln, wie Magnesiumstearat oder Talk, und/oder Mitteln zur Erzielung eines Depoteffekts, wie Carboxypolymethy- len, Carboxymethylcellulose, Celluloseacetatphthalat, oder Polyvinylacetat erhalten werden.
Die Tabletten können auch aus mehreren Schichten bestehen. Entsprechend können Dragees durch über ziehen von analog den Tabletten hergestellten Kernen mit üblicherweise in Dragéeüberzügen verwendeten Mitteln, beispielsweise Kollidon oder Schellack, Gummi arabicum, Talk, Titandioxid oder Zucker, hergestellt werden. Zur Erzielung eines Depoteffektes oder zur Vermeidung von Inkompatibilitäten kann der Kern auch aus mehreren Schichten bestehen. Desgleichen kann auch die Dragéehülle zur Erzielung eines Depot effekts aus mehreren Schichten bestehen, wobei die oben bei den Tabletten erwähnten Hilfsstoffe verwendet werden können.
Säfte der erfindungsgemässen Wirkstoffe bzw. Wirk stoffkombinationen können zusätzlich noch ein Sü- ssungsmittel, wie Saccharin, Cyclamat, Glycerin oder Zucker, sowie ein geschmacksverbesserndes Mittel, z. B. Aromastoffe, wie Vanillin oder Orangenextrakt, enthalten. Sie können ausserdem Suspendierhilfsstoffe oder Dickungsmittel, wie Natriumcarboxymethylcellu- lose, Netzmittel, beispielsweise Kondensationsprodukte von Fettalkoholen mit Äthylenoxid, oder Schutzstoffe, wie p-Hydroxybenzoate, enthalten.
Injektionslösungen werden in üblicher Weise, z. B. unter Zusatz von Konservierungsmitteln, wie p-Hy- droxybenzoaten, oder Stabilisatoren, wie Alkalisalzen der Äthylendiamintetraessigsäure, hergestellt und in In jektionsflaschen oder Ampullen abgefüllt.
Den oder die Wirkstoffe bzw. Wirkstoffkombina tionen enthaltende Kapseln können beispielsweise her gestellt werden, indem man die Wirkstoffe mit inerten Trägern, wie Milchzucker oder Sorbit, mischt und in Gelatinekapseln einkapselt.
Geeignete Zäpfchen lassen sich beispielsweise durch Vermischen der dafür vorgesehenen Wirkstoffe bzw. Wirkstoffkombinationen mit üblichen Trägermitteln, wie Neutralfetten oder Polyäthylenglykol bzw. dessen Derivaten, herstellen.
Die folgenden Beispiele dienen zur Erläuterung der Erfindung: <I>Beispiel 1</I> 4-Methylamino-5-hydroxy-7,8-dimethyl-2,3,4,5- tetrahydro-1-benzoxepin 30 g racemisches trans-4-N-Benzyl-N-methylamino-7,8-dimethyl-2,3,4,5- tetrahydro-5-hydroxy-1-benzoxepin vom Schmp. 111-112 C, welches wie untenstehend be schrieben erhalten wurde, werden in 200-250 cm3 Eisessig mittels Pd-Kohle hydriert, nach Filtration, Ein dampfen, Lösen in Wasser und Fällen mit Ammoniak erhält man 70-80 % d. Th. an farblosen Kristallen vom Schmp. 117-118 C.
Mit alkoholisch-ätherischer Salzsäure wird das farb lose Hydrochlorid der racemischen trans-Titelverbin- dung vom Schmp. 210 C erhalten.
Herstellung des Ausgangsmaterials: 0,2 Mol - 61,8 g racemisches 4-N-Benzyl-methylamino-7,8-di-methyl-2,3,4,5- tetrahydro-1-benzoxepin-5-on (Fp. 85-87 C), gelöst in 1 1 Methanol, werden ohne Küh lung unter Rühren während 10-15 Minuten mit 15 g Natriumboranat versetzt. Unter Aufschäumen erwärmt sich das Reaktionsgemisch allmählich. Anschliessend wird noch 1 Stunde unter Rückfluss gekocht, im Va kuum eingedampft, mit Wasser verdünnt und mit Me- thylenchlorid ausgeschüttelt.
Nach Abdampfen des Lö sungsmittels hinterbeibt 60-62 g des kristallinen Iso- merengemisches. Durch wiederholtes Kristallisieren aus Methanol kann man die racemischen trans-Komponente erhalten. Es ist aber günstig, das Gemisch über eine Säule von 400-500 g thermisch aktiviertes Kieselgel zu chromatographieren, wobei sich eine Mischung Iso- propyläther/Diäthylamin 50 : 1 als Fliessmittel bewährt hat. Ausbeute 25 bis 30 g.
In analoger Weise wird das cis-Isomere hydriert und die erhaltene racemische cis-Base der Titelverbindung, F. 150-151' C in das Hydrochlorid, F. 175-176 C, überführt.
<I>Beispiel 2</I> 4-Methylamino-5-hydroxy-7,8-dimethyl-2,3,4,5- tetrahydro-l-benzoxepin Ein durch Borhydridreduktion erhältliches Gemisch der isomeren Racemate des 4-N-Benzyl-N-methyl-amino-5-hydroxy-7,8- dimethyl-2,3,4,5-tetrahydro-1-benzoxepins (60-62 g) wird direkt in Eisessig hydriert und das Reaktionspro dukt mit Ammoniak gefällt (30-35 g). Das Gemisch aus cis- und trans-Racemat der Titelverbindung wird mit 30 g p-Nitrobenzaldehyd in 400 cm-" Xylol 2 Stunden am Wasserabscheider gekocht. Man dampft im Vakuum ein und gibt über eine kurze Säule aus Kieselgel (Fliess mittel Isopropyläther). Als erste Fraktion werden 15 bis 20 g des korrespondierenden Oxazolidins vom Schmp. 101-102 C erhalten.
Das Oxazolidin wird mit 1n Salzsäure 15 Minuten auf dem Wasserbad erwärmt. Aus dem Filtrat erhält man mit Ammoniak die ge wünschte trans-Base vom Schmp. 1l4-116 C.
<I>Beispiel 3</I> 8-Chlor-4-methylamino-5-hydroxy-2,3,4,5- tetrahydro-1-Benzoxepin 11 g racemisches trans-8-Chlor-4-benzylmethyl amino-5-hydroxy-2,3,4,5-tetrahydro-1-benzoxepin-hy- drochlorid werden in 100 cm3 Wasser und 50 cm3 Methanol nach Zugabe von 1,5 g Kohle und 15 cm3 2 % iger Palladiumchlorürlösung bei 60 C und 5 atm. hydriert. Nach 3 bis 5 Minuten sind 90 % des zur Ab- spaltung der Benzylgruppe nötigen Wasserstoffs auf genommen und die Hydrierung wird wesentlich lang samer. Man bricht ab und arbeitet auf, indem man mehrfach umkristallisiert und chromatographiert. Aus beute: 75 % d. Th. farblose Kristalle aus Isopropyläther; F. 121-122 C (trans-Base).
<I>Beispiel 4</I> 4-Methylamino-5-hydroxy-2,3,4,5- tetrahydro-1-benzoxepin Die Herstellung erfolgt wie in Beispiel 1 beschrie ben, ausgehend von 4-N-Benzyl-N-methylamino-5-hy- droxy-2,3,4,5-tetrahydro-1-Benzoxepin. Man erhält hierbei: Racemische trans-Titel-Verbindung (Base): F. 99 bis 100 C. (Hydrochlorid: F. 196-197 C).
Racemische cis-Titel-Verbindung (Base): F.125 bis 126 C. (Hydrochlorid: F. 150-152 C).
<I>Beispiel 5</I> 4-Methylamino-5-hydroxy-7,9-dimethyl-2,3,4,5- tetrahydro-1-benzoxepin Die Herstellung erfolgt wie in Beispiel 1 beschrie ben, ausgehend von 4-N-Benzyl-N-methylamino-5-hydroxy-7,9- dimethyl-2,3,4,5-tetrahydro-1-benzoxepin. Man erhält hierbei: Racemische trans - Titel - Verbindung (Base): F. 112-113 C. (Hydrochlorid 185-186 C).
Racemische cis-Titel-Verbindung (Base): F. 114 bis 115 C (Hydrochlorid: F. 215-216 C).
<I>Beispiel 6</I> trans-4-Methylamino-3,7,8-trimethyl-5-hydroxy- 2,3,4,5-tetrahydro-1-benzoxepin trans-4-N-Benzyl-N-methylamino-3,7,8-trimethyl-5- hydroxy-2,3,4,5-tetrahydro-1-benzoxepin, erhalten aus 3,7,8 - Trimethyl - 2,3,4,5 - tetrahydro-1-benzoepin-5-on vom Kp0,7 110-1l2 C durch Bromierung, Umsetzung mit N-Benzyl-N-methylamin, Reduktion mit Natrium- boranat und anschliessender säulenchromatographischer Trennung wird analog Beispiel 1 entbenzyliert. Schmp. der Verbindung: 111-113 C.
<I>Beispiel 7</I> 4-Methylamino-8-methyl-5-hydroxy-2,3,4,5- tetrahydro-1-benzoxepin Durch Entbenzylierung von 4-N-Benzyl-N-methylamino-8-methyl-5-hydroxy- 2,3,4,5-tetrahydrobenzoxepin entsprechend Beispiel 1 erhält man die Titelverbindung. Trans-Base: F. 81-82 C (Hydrochlorid: F. 231 bis 232 C) Cis-Base: F. 146-148 C (Hydrochlorid: F. 182 bis <B>183-</B> C).
<I>Beispiel 8</I> trans-4-Methylamino-7-chlor-9-methyl-5-hydroxy- 2,3,4,5-tetrahydro-1-benzoxepin Schonende katalytische Entbenzylierung von trans-4-N-Benzyl-N-methylamino-7-chlor-9-methyl-5- hydroxy-2,3,4,5-tetrahydro-1-benzoxepin; F. 140-141 C; in. Wasser/Methanol analog Beispiel 3 ergibt die Titel verbindung; F. 141-142 C: Die enthalogenierte Verbindung ist durch reduktive Enthalogenierung der Titelverbindung zugänglich. F. 79-80 C.
<I>Beispiel 9</I> trans-4-Methylamino-5-hydroxy-8,9-dimethyl-2,3,4,5- tetrahydro-1-benzoxepin Ausgehend von 2,3-Dimethylphenol und Butyro- lacton wird die ölige Benzylverbindung erhal ten, indem man 2,3-Dimethylphenol nach O. Dann und W. D. Arndt, Liebigs Annalen 587, S. 50 oder besser als Natriumsalz mit Butyrolacton nach B. Dotzauer, Diss. Marburg (1959) S.55 in 3,4-Dimethylphenoxy- buttersäure überführt und mit Polyphosphorsäure cy- clisiert.
0,5 Mol = 95 g 7,8-Dimethyl-2,3,4,5-tetrahydro-1- benzoxepin-5-on werden in 500 cm3 Chloroform mit 80 g Brom bromiert. Die sofort einsetzende Reaktion ist nach 15 Minuten beendet. Man dampft i. V. zur Trockne. Der feste Rückstand wird mit 121g N-Ben- zyl-N-methylamin und 400 cm-' Xylol oder Toluol 30 Min. unter Rückfluss gekocht; das N-Benzyl-N- methylaminhydrobromid mit Wasser ausgeschüttelt und das Reaktionsprodukt mit Zn Salzsäure extrahiert.
Alkalisieren der salzsauren Extrakte, Ausschütteln mit Methylenchlorid und Umkristallisieren aus Metha nol ergibt 78-85 % = 50--55 % d. Th. an racemischem 4-N-Benzyl-N-methylamino-8,9-dimethyl-2,3,4,5- tetrahydro-1-benzoxepin-5-on. Katalytisch-reduktive Entbenzylierung in Eisessig oder Methanol analog Beispiel 3 ergibt die Titelverbin dung.
trans-Base F. 103-104 C. trans-Hydrochlorid F. 218-219 C.
Die nach den vorstehenden Beispielen hergestellten Verbindungen erwiesen sich bei NMR-spektroskopi- scher Untersuchung als sterisch einheitlich.
