Verfahren zur Herstellung von Oxazaheterocyclen
Die vorliegende Erfindung betrifft die Herstellung von 10-Aminoalkyl-1 1-oxo-10,1 1-dihydro-dibenzo [b,f]- [1,4]oxazepinen, in denen die Aminogruppe monosubstituiert und vom Ringstickstoffatom durch mindestens 2 Kohlenstoffatome getrennt ist, und die in mindestens einem der Benzoringe eine Nitrogruppe enthalten.
Ein die Aminogruppe mit dem Ringstickstoffatom des Dibenzo[b,f][1,4]oxazepinrings verbindender Alkylenrest, welcher die beiden Stickstoffatome durch mindestens 2 Kohlenstoffatome trennt, ist insbesondere eine Niederalkylengruppe mit höchstens 7, vorzugsweise 2 bis 4 Kohlenstoffatomen; geeignete Niederalkylenreste dieser Art sind z.B. 1,2-Äthylen-, 1-Methyl-1,2-äthylen-, 2-Methyl-1,2-äthylen-, i,3-Propylen-, 2-Methyl1,3-propylen-, 1,3- oder 1,4-Butylengruppen, sowie 1,5 Pentylen- oder 1,6-Hexylengruppen.
Substituenten einer monosubstituierten Aminogruppe sind aliphatische, cyclialiphatische, cycloaliphatischaliphatische, aromatische oder araliphatische Reste, welche weitere Substituenten enthalten können. Solche Gruppen sind in erster Linie Alkyl-, besonders Niederalkylgruppen, z. B. Methyl-, Äthyl-, n-Propyl-, Isopropyl-, n-Butyl-, Isobutyl-, n-Pentyl-, Isopentyl-, n-Hexyloder n-Heptylgruppen, sowie Alkenyl-, besonders Niederalkenylgruppen, z. B. Allyl- oder Methallylgruppen, aber auch Cycloalkylgruppen vorzugsweise mit 3-8, insbesondere 5-7 Ringkohlenstoffatomen, z. B. Cyclopentyl-, Cyclohexyl- oder Cycloheptylgruppen, Cycloalkylniederalkylgruppen, vorzugsweise mit 3-8, insbesondere 5-7 Ringkohlenstoffatomen, z. B. Cyclopentylmethyl-, Cyclohexylmethyl-, Cyclohexyläthyl- oder Cycloheptylmethylgruppen, Phenylreste oder Phenylniederalkylreste, z. B.
Benzyl-, 1-Phenyläthyl- oder 2 Phenyläthylreste. Diese Gruppen können zusätzlich Substituenten, wie Niederalkyl-, Hydroxy-, verätherte Hydroxy-, wie Niederalkoxy-, Trifluormethyl-, Nitro-, Amino-, oder veresterte Hydroxygruppen, wie Halogenatome, z. B. Methyl-, Äthyl-, n-Propyl-, Isopropyl-, n Butyl-, Methoxy-, l2ithoxy-, n-Propyloxy-, Isopropyloxy-, Butyloxy-, Methylamino-, Dimethylamino-, Diäthylamino-, Allylamino-, Piperidino-, Morpholino- oder 4 Niederalkyl-piperazinogruppen, oder Fluor-, Chloroder Bromatome enthalten.
Die Verbindungen der vorliegenden Erfindung enthalten in den Benzoringen des tricyclischen 10,1 1-Di- hydrodibenzo[b,f] [1,4]oxazepin-Ringsystems vorzugsweise eine, aberauch zwei oder mehrere Nitrogruppen, die irgend eine Stellung, vorzugsweise die 2- und/oder 7-Stellung, aber auch die 3-Stellung einnehmen können.
Ausser Nitro enthalten die aromatischen Ringe gegebenenfalls einen, zwei oder mehrere weitere, gleiche oder verschiedene Substituenten, wie Niederalkyl-, ver ätherte Hydroxy-, z. B. Niederalkoxy-, Trifluormethyloder veresterte Hydroxygruppen, z. B. Halogenatome, wie die oben erwähnten Substituenten. Sie können aber auch Acyl-, wie Niederalkanoyl-, z. B. Acetyl-, Propionyl- oder Butyrylgruppen, sowie gegebenenfalls substituierte z. B. mono- oder disubstituierte Aminogruppen, enthalten, wobei Substituenten von Aminogruppen in erster Linie aliphatische Gruppen, wie Alkyl-, insbesondere Niederalkylgruppen, sein können.
Die Verbindungen der vorliegenden Erfindung sind demnach 10,1 1-Dihydro-dibenzo [b,f] [1,4] oxazepine der Formel I

worin Am eine monosubstituierte Aminogruppe und A einen die Gruppe Am vom Ringstickstoffatom durch mindestens 2 Kohlenstoffatome trennenden Alkylenrest bedeuten, und worin mindestens eine der 1,2-Phenylengruppen Phi und Ph durch eine Nitrogruppe substituiert ist.
Die'Verbindungen der vorliegenden Erfindung haben wertvolle pharmakologische Eigenschaften und können entsprechend verwendet werden. So zeigen sie ausser Anti-Acetylcholin-, Anti-Serotonin- und Anti Histamin-Wirkungen sowie antiinflammatorischen und lokalanästhetischen Eigenschaften, in erster Linie antidepressive Effekte, die anhand von Tierversuchen, z. B.
an Mäusen und Affen, nachgewiesen werden können, sowie andere psychopharmakologische Eigenschaften.
So zeigen sie z. B. eine reversible Wirkung auf die durch Reserpin hervorgerufene Hypothermie und Ptosis und/ oder potenzieren die Wirkung von 3,4-Dihydroxyphenylalanin in mit einem Monoaminoxydasehemmer behandelten Mäusen. Die Verbindungen können deshalb in erster Linie als antidepressive Mittel zur Behandlung von Depressionszuständen wie auch als Antihistaminika verwendet werden. Ferner können sie als Zwischenprodukte zur Herstellung von anderen, pharmazeutisch verwendbaren Verbindungen verwendet werden.

hat das 10-(3-Methylamino-propyl)-2-nitro-1 1-oxo- 10,1 1-dihydro-dibenzo [b,f] [1,4] oxazepin; diese Verbindung zeigt bei oral oder parenteral verabreichten Tagesdosen von etwa 0,05 g bis etwa 0,2 g hervorragende antidepressive Wirkungen.
Die Verbindungen der vorliegenden Erfindung werden hergestellt, indem man in einem 10-(N-R -N-sub- stituierten Aminoalkyl)-1 1-oxo-1 0,1 1-dihydro-dibenzo- [b,f] [1,4]oxazepin, worin die Aminogruppe durch mindestens 2 Kohlenstoffatome vom Ringstickstoffatom getrennt ist und R" einen Acyl- oder Cyanrest darstellt, und das in mindestens einem der Benzoringe eine Nitrogruppe enthält, die Gruppe RO hydrolytisch abspaltet.
Wenn erwünscht, kann in einer erhaltenen Verbindung eine Aminoalkylgruppe in eine andere Aminoalkylgruppe übergeführt werden.
Ein abspaltbarer Rest R" in einer N-RO-N-substi- tuierten Aminogruppe ist in erster Linie eine hydrolytisch abspaltbare Acylgruppe. Letztere stellt besonders den Acylrest einer monoveresterten Kohlensäure, wie einen Carbo-niederalkoxy-, z. B. Carbomethoxy-, Carb äthoxy- oder Carbo-tert.-butyloxyrest, oder den Acylrest einer Carbonsäure, wie der Ameisensäure, d. h.
den Formylrest, oder einer Niederalkancarbonsäure, z. B. Essig- oder Propionsäure, dar. Die Acylaminobindung in einer N-Acyl-N-substituierten Aminogruppe wird hydrolytisch, insbesondere in Gegenwart von wässrigen Mineralsäuren oder wässrigen Basen gespalten; der Acylrest einer monoveresterten Kohlensäure kann z. B. durch Behandeln mittels Bromwasserstoffsäure in Essigsäure abgespalten werden. Eine Carbo-tert.-butyloxyaminogruppe lässt sich z. B. auch durch Behandeln mit Trifluoressigsäure spalten. Ein weiterer Rest RO ist z. B. eine Cyangruppe, die hydrolytisch, vorzugsweise in Gegenwart von wässriger Mineralsäure, z. B. Salzsäure, oder von Polyphosphorsäure, abgespalten werden kann.
Je nach den Reaktionsbedingungen werden die Verbindungen der vorliegenden Erfindung in freier Form oder in Form ihrer Salze erhalten. Säureadditionssalze sind insbesondere pharmazeutisch verwendbare Säureadditionssalze, z. B. solche mit anorganischen Säuren, wie Salzsäure, Bromwasserstoffsäure, Schwefelsäure oder Phosphorsäure, oder mit organischen Säuren, wie Essig-, Oxal-, Propion-, Pivalin-, Glykol-, Milch-, Malon-, Bernstein-, Malein-, Hydroxymalein-, iipfel-, Wein-, Zitronen-, Benzoe-, Salicyl-, 2-Acetoxybenzoe-, Nicotin-, Isonicotin-, Ascorbin-, Methansulfon-, Äthansulfon-, Athan-1,2-disulfon-, 2-Hydroxy-äthansulfon-, p-Toluolsulfon- oder Naphthalinsulfonsäure.
Andere Säureadditionssalze können als Zwischenprodukte oder zur Identifikation dienen; besonders geeignet zur Charakterisierung sind solche mit organischen Nitroverbindungen, z. B. mit Pikrin-, Pikrolonoder Flaviansäure, mit komplexen Metallsäuren, z. B.
Phosphorwolframsäure, Phosphormolybdänsäure, Chlorplatinsäure oder Reineckesäure, oder mit Perchlorsäure.
Erhaltene Salze können z. B. durch Behandeln mit einer Base oder mit einem geeigneten Anionenaustauscher in die freie Verbindung, oder durch Behandeln mit einem Metall-, z. B. Barium- oder Silbersalz in Gegenwart eines Verdünnungsmittels, in welchem eine gebildete anorganische Verbindung unlöslich ist, oder mit einem geeigneten Anionenaustauscher in andere Salze übergeführt werden; freie Verbindungen werden z. B. durch Behandeln mit geeigneten Säuren oder Ionenausfau- schern in Säureadditionssalze umgewandelt. Infolge der engen Beziehungen der neuen Verbindungen in freier Form und in Form ihrer Salze sind im vorausgegangenen und nachfolgend unter freien Verbindungen sinnund zweckgemäss gegebenenfalls auch die entsprechenden Salze zu verstehen.
Erhaltene Isomerengemische können in an sich bekannter Weise, Racemate z.B. durch Überführen in Gemische von diastereoisomeren Salzen mit geeigneten optisch aktiven Säuren und Trennen des Gemisches auf Grund von Löslichkeitsunterschieden, in die einzelnen Isomeren gespalten werden.
Die Erfindung betrifft auch diejenigen Ausführungsformen des Verfahrens, bei denen man die Ausgangsstoffe in Form eines unter den Reaktionsbedingungen erhältlichen rohen Reaktionsgemisches oder in Form ihrer Salze vorliegen.
Für die erfindungsgemässen Reaktionen werden vor nehmlich solche Ausgangsstoffe verwendet, die die oben erwähnten bevorzugten Verbindungen ergeben.
Die im oben envähnten Verfahren verwendeten Ausgangsstoffe lassen sich nach an sich bekannten Methoden herstellen. So erhält man z. B. die 10-unsubstituier ten 1 1-Oxo-10,1 1-dihydro-dibenzo [b,f] [1,4] oxazepine, indem man a) in einem N (o-Hydroxy-phenyl-O-XOa- benzoesäureamid, worin mindestens einer der aromatischen Reste eine Nitrogruppe enthält und XOa für eine reaktionsfähige veresterte Hydroxylgruppe steht, oder einem Salz davon durch Ringschluss den 4,5-Dihydro 1,4-oxazepinring bildet, oder b) einen o-Amino-o'-XOb-diphenyläther, worin mindestens einer der aromatischen Reste eine Nitrogruppe enthält und Xob einen Substituenten bedeutet,
welcher unter den Bedingungen des Ringschlusses eine Carbonylgruppe bildet, ringschliesst, oder c) einen o-(N-XOa-Amino)-diphenyläther, worin mindestens einer der aromatischen Reste eine Nitrogruppe enthält, und XOG einen Substituenten bedeutet, der unter den Bedingungen des Ringschlusses eine Carbonylgruppe bildet, und in welchem mindestens eine der o' Stellungen unsubstituiert ist, ringschliesst, oder d) in ein 11-Oxo-10,11-dihydro-dibenzo[b,f][1,4]oxa- zepin eine Nitrogruppe einführt oder e) ein 9-Hydroximino-xanthen, worin mindestens eine der beiden carbocyclischen aromatischen Ringe eine Nitrogruppe enthält, der Beckmann-Umlagerung unterwirft.
In ein erhaltenes 10-unsubstituiertes 10-Oxo 10,1 1-dihydro-dibenzo[b,f] [1,4] oxazepin kann dann die N-R -N-substituierte Aminoalkylgruppe eingeführt werden.
Der Ringschluss eines Ausgangsmaterials, in welchem XOa in erster Linie ein Halogen-, z. B. ein Chloroder Bromatom, bedeutet, wird z. B. durch Behandeln mit einer Base, wie einem Alkalimetallhydroxyd, z. B.
Natriumhydroxyd oder Kaliumhydroxyd, besondeis einer Lösung eines solchen, z. B. in Wasser, durchgeführt, wobei man, wenn notwendig, bei erhöhter Temperatur arbeitet, oder durch Erhitzen eines Salzes, z. B. eines Alkalimetall-, wie Natriumsalzes, des Ausgangsmaterials, vorzugsweise in Gegenwart eines geeigneten Lösungsmittels, wie Dimethylformamid.
Eine Gruppe Xob ist in erster Linie eine funktionell abgewandelte Carboxylgruppe, wie eine veresterte Carboxylgruppe, z. B. eine Carbo-niederalkoxy-, wie eine Carbomethoxy- oder Carbäthoxygruppe, oder insbesondere eine Halogencarbonyl-, z. B. eine Chlorcarbonylgruppe. Ringschluss eines Ausgangsstoffes mit einer solchen Gruppe, welche unter den Reaktionsbedingungen in eine Carbonylgruppe umgewandelt wird, kann nach an sich bekannten Methoden durchgeführt werden, z. B. durch Erhitzen, vorzugsweise in Gegenwart eines alkalischen Kondensationsmittels, wie z. B. eines Alkalimetall- oder Erdalkalimetallcarbonates, -hydroxyds oder -niederalkoxyds, vorzugsweise in Gegenwart eines Verdünnungsmittels.
Eine Gruppe X,C in einem Ausgangsmaterial ist eine funktionell abgewandelte Carboxylgruppe, insbesondere eine veresterte Carboxylgruppe, wie eine Carbo-niederalkoxygruppe, kann aber auch eine mit der Aminogruppe einen Isocyanatorest bildende Carbonylgruppe sein. Ringschluss wird z. B. in Gegenwart eines sauren Kondensationsmittels, wie Polyphosphorsäure oder Schwefelsäure, beim Vorhandensein einer Isocyanatgruppe auch von Aluminiumchlorid durchgeführt.
Die Einführung einer Nitrogruppe wird mittels irgendeines geeigneten Nitrierverfahrens durchgeführt, welches sich zur Einführung einer Nitrogruppe in einen aromatischen Kern eignet, z.B. Salpetersäure in Gegenwart von Schwefelsäure.
Die Beckmann-Umlagerung (Variante e) wird in an sich bekannter Weise, in erster Linie durch Behandeln des 9-Hydroximino-xanthens, z. B. mit einem Phosphorhalogenid, wie Phosphorpentachlorid, durchgeführt. Dabei können xwei isomere 11-Oxo-10,11-dihydro-di benzo[b,f][15 oxazepine erhalten werden.
Eine N-R -N-substituierte Aminoalkylgruppe, wie eine N-Acyl-N-substituierte Aminoalkylgruppe, kann durch Behandeln des N-unsubstituierten 11-Oxo-10,11 dihydro-dibenzo[b,f][1,4]oxazepins oder eines Salzes davon mit einem N-Rc-N-substituierten, wie einem N Acyl-N-substituierten Aminoalkylhalogenid, eingeführt werden. Ferner kann man z. B. in einem N,N-disubstituierten 10-Aminoalkyl-1 1-oxo-10,1 1-dihydro-diben zo[b,f][1,4]oxazepin, worin einer der Substituenten der Aminogruppe eine Alkyl- und in erster Linie eine Methylgruppe darstellt, diesen in eine Acyl- oder Cyangruppe umwandeln. So kann man eine Alkyl-, z. B.
Methylgruppe, auf oxydativem Weg durch Behandeln mit Oxydationsmitteln, wie Kaliumpermanganat, Mangandioxyd, Kaliuinferricyanid, Chromsäure, Dibenzoylperoxyd, Gold-III-chlorid oder Quecksilber-II-acetat, in eine Acyl-, insbesondere Formylgruppe, umwandeln. Ferner kann man in einem entsprechenden Ausgangsmaterial eine Alkyl-, insbesondere Methylgruppe, durch Behandeln mit einem Halogencyan, insbesondere Bromcyan, mit einem Halogenameisensäureester, wie Chlorameisensäureäthyl-, -tert.-butyl- oder -benzylester, mit einem Kohlensäuredihalogenid, z. B.
Phosgen, oder mit einer geeigneten Carbonsäure oder einem Anhydrid oder einem Halogenid, z. B. Chlorid davon, einen solchen Substituenten durch eine Cyangruppe, eine funktionell abgewandelte, wie eine veresterte Carboxylgruppe, z. B. eine Carbäthoxy-, Carbotert.-butoxy- oder Carbobenzoxygruppe, bzw. eine Acylgruppe ersetzen.
Die in den obigen Verfahren zur Herstellung von Ausgangsstoffen verwendeten Zwischenprodukte lassen sich in an sich bekannter Weise herstellen. Diejenigen der Variante a) werden z.B. erhalten, indem man ein a-Aminophenol mit einem o-X,"-Benzoesäure- halogenid, besonders einem -chlorid, umsetzt; mindestens eine der beiden Verbindungen enthält eine aromatisch gebundene Nitrogruppe.
Die Ausgangsstoffe der Variante b) können z. B.
durch Reaktion eines o-Aminophenols mit einer o-Halogenbenzoesäure, wobei mindestens einer der beiden Reaktionsteilnehmer eine aromatisch gebundene Nitrogruppe enthält, gebildet werden; im erhaltenen Zwischenprodukt kann die freie Carboxylgruppe in an sich bekannter Weise in die erwünschte Gruppe Xob umgewandelt werden.
Zwischenprodukte, die in der Variante c) verwendet werden können, erhält man z. B. durch Umsetzen eines o-Aminodiphenyläthers, worin mindestens einer der beiden aromatischen Reste eine Nitrogruppe enthält, mit einer geeigneten Halogenkohlensäureverbindung, wie Chlorameisensäureäthylester.
Die 9-Hydroximino-xanthen-Zwischenprodukte der Variante e) erhält man z. B. durch Behandeln eines, in mindestens einem der beiden aromatischen Ringe eine Nitrogruppe enthaltenden Xanthons oder 9-Thionoxanthens mit Hydroxylamin oder einem Salz davon, letzteres in Gegenwart einer Base, wie Pyridin.
Die Verbindungen der vorliegenden Erfindung können z. B. in Form von pharmazeutischen Präparaten Verwendung finden, welche sich zu enteraler, z. B.
oraler, oder parenteraler Verabreichung eignen.
Die Erfindung wird in den folgenden Beispielen näher beschrieben. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel 1
Eine Suspension von 3 g 10-(3-N-Cyan-N-methyl aminopropyl)-2-nitro-1 1-oxo- 10,11 -dihydro-dibenzo- [b,f] [1,4]oxazepin in 75 ml einer etwa 4-n. Salzsäure wird während 4 Stunden am Rückfluss gekocht. Nach dem Abkühlen wird das Reaktionsgemisch mit Äther geschüttelt und die einzelnen Phasen getrennt; die wässrige Lösung wird mit flüssigem Ammoniak basisch gestellt und mit Methylenchlorid extrahiert. Der Methylenchloridextrakt, enthaltend das 1 0-(3-Methylami- no-propyl)-2-nitro-1 oxo-10,11-dihydro-dibenzo[b,f]- [1,4]oxazepin, wird mit Wasser gewaschen, getrocknet und auf ein kleines Volumen eingeengt.
Das Hydrochloridsalz wird durch Sättigen der konzentrierten Methylenchloridlösung mit trockenem Chlorwasserstoffsäuregas erhalten; der Niederschlag wird abfiltriert und aus einem Gemisch von Äthanol und Äther kristallisiert, wobei man das 10-(3-Methylamino-propyl)-2-nitro-1 1- oxo-1 0,1 1-dihydro-dibenzo [b,f] [1,4] oxazepin-hydrochlorid der Formel

erhält, welches bei 215-217" schmilzt.
Das im obigen Verfahren verwendete Ausgangsmaterial wird wie folgt hergestellt:
Ein Gemisch einer Lösung von 5,2 g Natriumbi carbonat in 50 ml Wasser und einer Lösung von 3,3 g o-Aminophenol in 50 ml Äther wird mit Eiswasser ge kühlt und unter Rühren langsam mit einer Lösung von
6,6 g 2-Chlor-5-nitrobenzoylchlorid in 50 ml trockenem Äther, welche über eine Periode einer Stunde zu gegeben wird, versetzt. Das Gemisch wird während meh reren Stunden gerührt und das kristalline Amid abfiltriert. Die Ätherschicht des Filtrates wird abgetrennt und mit verdünnter Salzsäure und dann mit Wasser ge waschen, getrocknet und zur Trockne eingedampft. Man erhält so eine zusätzliche Menge des N-(2-Chlor-5-nitrobenzoyl)-2-hydroxy-anilins, welches aus wässrigem Me thanol umkristallisiert wird und bei 189-192" schmilzt.
Eine Lösung von 4,5 g N-(2-Chlor-5-nitrobenzoyl)
2-hydroxy-anilin in 150 ml Wasser, welche 0,8 g Na triumhydroxyd enthalten, wird unter Rühren auf einem Dampfbad während 16 Stunden erhitzt. Der kri stalline Niederschlag wird abfiltriert, mit Wasser ge waschen und aus einem Gemisch von Aceton und Me thanol umkristallisiert. Man erhält so das 2-Nitro-1 1- oxo-10,1 1-dihydro-dibenzo [b,f] [1,4] oxazepin, das bei 258-260" schmilzt.
Ein Gemisch von 3 g 2-Nitro-11-oxo-10,11-dihydro- dibenzo[b,f] [1,4]oxazepin und 4,7 g 3-Dimethylamino propylchlorid-hydrochlorid in 18 ml Wasser, welche 1,8 g Natriumhydroxyd enthalten, und 30 ml Aceton wird unter vermindertem Druck entfernt und der Rückstand mit Wasser verdünnt. Unter Kühlen scheidet sich ein Ö1 ab, welches mit Äther extrahiert wird. Die Ather- lösung, enthaltend das 10-(3-Dimethylamino-propyl)- 2-nitro-1 1-oxo-10, 1 1-dihydro-dibenzo [b,f] [1,4] oxazepin wird mit Wasser gewaschen, getrocknet und zur Trockne eingedampft. Das Produkt kann als das Hydrochlorid, das bei 223-2260 schmilzt, identifiziert werden.
Eine Lösung von 2,6 g Bromcyan in 15 ml trockenem Äther wird tropfenweise über 15 Minuten und unter Rühren zu einer Lösung von 5,6 g 10-(3-Di methylamino-propyl)-2-nitro-1 1-oxo-10, 1 1-dihydro-di- benzo[b,f] [1,4]oxazepin in 25 ml trockenem Äther gegeben. Das Gemisch wird während 4 Stunden bei Zimmertemperatur gerührt und dann mit 25 ml Wasser versetzt; das feste Material wird abfiltriert, es stellt das kristalline 10-(3 -N-Cyan-N-methyl-aminopropyl)-2-ni- tro-1 1-oxo-10, 1 1-dihydro-dibenzo [b,f] [1,4] oxazepin dar.
Die Ätherlösung des Filtrats wird abgetrennt und mit verdünnter Salzsäure geschüttelt, dann mit Wasser gewaschen, getrocknet und verdampft. Man erhält so eine weitere Menge der erwünschten Verbindung, welche nach Umkristallisieren aus einem Gemisch von Chloroform und n-Hexan bei 135-136" schmilzt.
Beispiel 2
Ein Gemisch von 2 g 10-(3-N-Cyan-N-methyl-ami nopropyl)-2-nitro-1 1-oxo-1 0,1 1-dihydro-dibenzo [b,f]- [1,4]oxazepin und 25 g Polyphosphorsäure wird unter Rühren während 30 Minuten bei 140 (Ölbadtemperatur) erhitzt; die Temperatur wird dann auf 1600 erhöht und während fast 3 Stunden gehalten. Nach dem Abkühlen wird das Gemisch mit eiskaltem Wasser verdünnt und mit 25 ml 3-n. Salzsäure behandelt und das Gemisch mit Methylenchlorid geschüttelt. Die saure wässrige Phase wird mit flüssigem Ammoniak basisch extrahiert; die organische Lösung wird mit Wasser gestellt und das abgeschiedene Öl mit Methylenchlorid gewaschen, über Natriumsulfat getrocknet und dann mit trockenem Chlossvasserstoffsäuregas gesättigt.
Das Hydrochlorid des 10-(3-Methylamino-propyl)-2-nitro-11- oxo-1 0,1 1-dihydro-dibenzo [b,f] [1,4] oxazepins wird abfiltriert, mit einer kleinen Menge eines Gemisches von absolutem Äthanol und Äther gewaschen und dann aus einem Gemisch von Äthanol und Äther umkristallisiert, F. 215-217".
Beispiel 4
Ein Gemisch von 1,4 g 10-(3-N-Carbäthoxy-N-me thyl-aminopropyl)-2-nitro-1 1-oxo-10, 1 1-dihydro-diben- zo[b,f] [1,4]oxazepin und 12 ml einer 480/oigen Lösung von Bromwasserstoffsäure in Essigsäure wird während 3 Tagen unter gelegentlichem Rühren und unter Ausschluss von Feuchtigkeit bei Zimmertemperatur stehen gelassen. Es wird mit 100ml Äther verdünnt, abgekühlt und die Flüssigkeit abdekantiert. Der zähe feste Rückstand wird mit 100 ml Wasser geschüttelt und filtriert; das Filtrat wird mit flüssigem Ammoniak basisch gestellt und mit Methylenchlorid extrahiert. Die organische Lösung wird mit Wasser gewaschen, getrocknet und dann mit trockenem Chlorwasserstoffsäuregas gesättigt.
Das erwünschte 10-(3-Methylamino-propyl)-2 nitro-1 1-oxo-1 0,1 1-dihydro-dibenzo [b,f] [1,4]oxazepin- hydrochlorid scheidet als kristalliner Niederschlag aus und wird aus einem Gemisch von Äthanol und Äther umkristallisiert, F. 215-217".
Das Ausgangsmaterial wird wie folgt hergestellt:
Eine Lösung von 3,4 g 10-(3-Dimethylamino-pro pyl)-2-nitro-1 1-oxo-10, 1 1-dihydro-dibenzo [b,f] [1,4] oxa- zepin in 20 ml trockenem Xylol wird über eine Periode von 10 Minuten und unter Rühren mit einer Lösung von 4,3 g Chlorameisensäureäthylester in 5 ml trockenem Xylol versetzt. Das Gemisch wird während 6 Stunden unter Rühren am Rückfluss gekocht; nach dem Abkühlen wird es mit 50 ml Äther und 25 ml verdünnter Salzsäure geschüttelt und die Phasen getrennt. Die organische Phase wird unter vermindertem Druck eingedampft und der ölige Rückstand, welcher das 10-(3-N Carbäthoxy-N-methyl-aminopropyl)-2-nitro-1 1-oxo 10,1 1-dihydro-dibenzo [b,f] [1,4]oxazepin darstellt, wird ohne weitere Reinigung verwendet.
Beispiel 3
Eine Lösung von 4,6 g 10-(3-N-Formyl-N-methyl aminopropyl)-2-nitro-1 1-oxo-1 0,1 1-dihydro-dibenzo- [b,f][1,4]oxazepin in 70 ml Athanol wird mit 30 ml 6-n.
Salzsäure vermischt und während 6 Stunden am Rückfluss gekocht. Beim Abkühlen kristallisiert das 10-(3 Methylamino-propyl)-2-nitro-11-oxo-10,11-dihydro-di- benzo[b,f] [1,4]oxazepin-hydrochlorid aus, wird abfiltriert und mit einer kleinen Menge eines Gemisches von Äthanol und Ather gewaschen. Die wässrige Phase des Filtrats wird mit flüssigem Ammoniak basisch gestellt und mit Methylenchlorid extrahiert; der organische Extrakt wird über wasserfreiem Natriumsulfat getrocknet und mit trockenem Chlorwasserstoffsäuregas gesättigt; man erhält so eine zusätzliche Menge des erwünschten Produkts, welches nach Umkristallisieren aus einem Gemisch von absolutem Äthanol und Äther bei 215-217" schmilzt.
Das Ausgangsmaterial wird wie folgt erhalten:
Eine Suspension von 1 g pulverförmigem Natriumamid in 25 ml trockenem Dioxan wird mit einer warmen Lösung von 5,1 g 2-Nitro-1 1-oxo-10, 1 1-dihydro- dibenzo[b,f] [1,4]oxazepin in 125 ml trockenem Dioxan versetzt; das Gemisch wird während 1/2 Stunde gerührt und dann mit 3,4 g 3-N-Formyl-N-methyl-aminopropyl- chlorid [hergestellt durch Reaktion von N-Methylformamid und 3-Brom-1-chlor-propan in Gegenwart von Natriumamid nach dem in Chem. Abstr., 62, 13163e (1965) beschriebenen Verfahren] versetzt, welches über eine Periode von 10 Minuten zugegeben wird.
Nach 6-stündigem Kochen unter Rückfluss und Rühren wird das Lösungsmittel durch Destillation unter vermindertem Druck entfernt und der Rückstand mit Wasser und Methylenchlorid ausgeschüttelt; nicht reagiertes Ausgangsmaterial wird durch Filtrieren entfernt. Die Methylenchloridphase des Filtrats wird mit Wasser gewaschen, getrocknet und unter vermindertem Druck zur Trockne eingedampft. Man erhält das 10-(3-N-Formyl N-methyl-aminopropyl)-2-nitro-1 1-oxo-10, 1 1-dihydro- dibenzo[b,f] [1,4]oxazepin als braunes Öl, welches ohne weitere Reinigung verwendet wird.

A suspension of 3 g of 10- (3-N-cyano-N-methyl aminopropyl) -2-nitro-1 1-oxo-10,11-dihydro-dibenzo- [b, f] [1,4] oxazepine in 75 ml of an approximately 4-n. Hydrochloric acid is refluxed for 4 hours. The starting material used in the above procedure is prepared as follows:
A mixture of a solution of 5.2 g of sodium bicarbonate in 50 ml of water and a solution of 3.3 g of o-aminophenol in 50 ml of ether is cooled with ice water and slowly stirred with a solution of
6.6 g of 2-chloro-5-nitrobenzoyl chloride in 50 ml of dry ether, which is added over a period of one hour, are added. The mixture is stirred for several hours and the crystalline amide is filtered off. The ether layer of the filtrate is separated off and washed with dilute hydrochloric acid and then with water, dried and evaporated to dryness. This gives an additional amount of N- (2-chloro-5-nitrobenzoyl) -2-hydroxy-aniline, which is recrystallized from aqueous methanol and melts at 189-192 ".
A solution of 4.5 g of N- (2-chloro-5-nitrobenzoyl)
2-hydroxy-aniline in 150 ml of water, which contain 0.8 g of sodium hydroxide, is heated with stirring on a steam bath for 16 hours. The crystalline precipitate is filtered off, washed with water and recrystallized from a mixture of acetone and methanol. This gives 2-nitro-1 1-oxo-10,1 1-dihydro-dibenzo [b, f] [1,4] oxazepine, which melts at 258-260 ".
A mixture of 3 g of 2-nitro-11-oxo-10,11-dihydrodibenzo [b, f] [1,4] oxazepine and 4.7 g of 3-dimethylamino propyl chloride hydrochloride in 18 ml of water, which 1, Contain 8 g of sodium hydroxide, and 30 ml of acetone is removed under reduced pressure and the residue is diluted with water. An oil separates out under cooling, which is extracted with ether. The ether solution containing the 10- (3-dimethylamino-propyl) -2-nitro-1 1-oxo-10,1 1-dihydro-dibenzo [b, f] [1,4] oxazepine is washed with water, dried and evaporated to dryness. The product can be identified as the hydrochloride melting at 223-2260.
A solution of 2.6 g of cyanogen bromide in 15 ml of dry ether is added dropwise over 15 minutes and with stirring to a solution of 5.6 g of 10- (3-dimethylamino-propyl) -2-nitro-11-oxo-10 , 1 1-dihydro-dibenzo [b, f] [1,4] oxazepine given in 25 ml of dry ether. The mixture is stirred for 4 hours at room temperature and then 25 ml of water are added; the solid material is filtered off, it provides the crystalline 10- (3 -N-cyano-N-methyl-aminopropyl) -2-nitro-1 1-oxo-10, 1 1-dihydro-dibenzo [b, f] [1,4] oxazepine.
The ether solution of the filtrate is separated off and shaken with dilute hydrochloric acid, then washed with water, dried and evaporated. This gives a further amount of the desired compound which, after recrystallization from a mixture of chloroform and n-hexane, melts at 135-136 ".
Example 2
A mixture of 2 g of 10- (3-N-Cyano-N-methyl-aminopropyl) -2-nitro-1 1-oxo-1 0.1 1-dihydro-dibenzo [b, f] - [1.4 ] oxazepine and 25 g of polyphosphoric acid are heated with stirring for 30 minutes at 140 (oil bath temperature); the temperature is then increased to 1600 and held for almost 3 hours. After cooling, the mixture is diluted with ice-cold water and mixed with 25 ml of 3-n. Treated hydrochloric acid and shaken the mixture with methylene chloride. The acidic aqueous phase is extracted basic with liquid ammonia; the organic solution is made with water and the separated oil is washed with methylene chloride, dried over sodium sulfate and then saturated with dry hydrochloric acid gas.
The hydrochloride of 10- (3-methylamino-propyl) -2-nitro-11-oxo-1 0.1 1-dihydro-dibenzo [b, f] [1,4] oxazepine is filtered off with a small amount of a mixture washed by absolute ethanol and ether and then recrystallized from a mixture of ethanol and ether, F. 215-217 ".
Example 4
A mixture of 1.4 g of 10- (3-N-carbethoxy-N-methyl-aminopropyl) -2-nitro-1 1-oxo-10, 1 1-dihydro-dibenzo [b, f] [1 , 4] oxazepine and 12 ml of a 480% solution of hydrobromic acid in acetic acid are left to stand for 3 days with occasional stirring and with exclusion of moisture at room temperature. It is diluted with 100ml ether, cooled and the liquid is decanted off. The viscous solid residue is shaken with 100 ml of water and filtered; the filtrate is made basic with liquid ammonia and extracted with methylene chloride. The organic solution is washed with water, dried and then saturated with dry hydrochloric acid gas.
The desired 10- (3-methylamino-propyl) -2 nitro-1 1-oxo-1 0.1 1-dihydro-dibenzo [b, f] [1,4] oxazepine hydrochloride separates out as a crystalline precipitate and becomes out recrystallized from a mixture of ethanol and ether, F. 215-217 ".
The raw material is produced as follows:
A solution of 3.4 g of 10- (3-dimethylamino-propyl) -2-nitro-1 1-oxo-10,1 1-dihydro-dibenzo [b, f] [1,4] oxazepine in 20 ml of dry xylene is added over a period of 10 minutes and with stirring with a solution of 4.3 g of ethyl chloroformate in 5 ml of dry xylene. The mixture is refluxed with stirring for 6 hours; After cooling, it is shaken with 50 ml of ether and 25 ml of dilute hydrochloric acid and the phases are separated. The organic phase is evaporated under reduced pressure and the oily residue, which contains the 10- (3-N carbethoxy-N-methyl-aminopropyl) -2-nitro-11-oxo 10,1 1-dihydro-dibenzo [b, f ] [1,4] oxazepine is used without further purification.
Example 3
A solution of 4.6 g of 10- (3-N-Formyl-N-methyl aminopropyl) -2-nitro-1 1-oxo-1 0.1 1-dihydro-dibenzo [b, f] [1.4 ] oxazepine in 70 ml of ethanol is mixed with 30 ml of 6-n.
Mixed hydrochloric acid and refluxed for 6 hours. On cooling, the 10- (3-methylamino-propyl) -2-nitro-11-oxo-10,11-dihydro-di-benzo [b, f] [1,4] oxazepine hydrochloride crystallizes out, is filtered off and treated with a small amount of a mixture of ethanol and ether. The aqueous phase of the filtrate is made basic with liquid ammonia and extracted with methylene chloride; the organic extract is dried over anhydrous sodium sulfate and saturated with dry hydrochloric acid gas; this gives an additional amount of the desired product which, after recrystallization from a mixture of absolute ethanol and ether, melts at 215-217 ".
The starting material is obtained as follows:
A suspension of 1 g of powdered sodium amide in 25 ml of dry dioxane is mixed with a warm solution of 5.1 g of 2-nitro-1 1-oxo-10, 1 1-dihydro-dibenzo [b, f] [1,4] oxazepine added in 125 ml of dry dioxane; the mixture is stirred for 1/2 hour and then with 3.4 g of 3-N-formyl-N-methyl-aminopropyl chloride [prepared by reacting N-methylformamide and 3-bromo-1-chloropropane in the presence of Sodium amide according to the method described in Chem. Abstr., 62, 13163e (1965)], which is added over a period of 10 minutes.
After boiling under reflux and stirring for 6 hours, the solvent is removed by distillation under reduced pressure and the residue is extracted by shaking with water and methylene chloride; unreacted starting material is removed by filtration. The methylene chloride phase of the filtrate is washed with water, dried and evaporated to dryness under reduced pressure. The 10- (3-N-formyl N-methyl-aminopropyl) -2-nitro-1 1-oxo-10,1 1-dihydrodibenzo [b, f] [1,4] oxazepine is obtained as a brown oil, which is used without further purification.
