Die Erfindung betrifft ein Verfahren zur Herstellung von neuen 4H-m-Dithiino[5,4-d]pyrimidinen der Formel

Die neuen Verbindungen werden erfindungsgemäss erhalten durch Umsetzung eines 4H-m-Dithiino[5,4-d]pyrimidins der Formel

in der einer der Reste A1 und A2 ein Halogenatom oder eine freie oder durch einen niederen Alkylrest substituierte Mercaptogruppe bedeutet,
und der andere der Reste A1 und A2 entweder ein Halogenatom oder eine freie oder durch einen niederen Alkylrest substituierte Mercaptogruppe bedeutet oder eine der anderen oben für R3 und R4 angegebenen Bedeutungen besitzt, mit Verbindungen der Formeln Rs'H und R4H, wobei R's eine Gruppe der Formel -NR,R, darstellt.
Sollen nach diesem Verfahren Verbindungen hergestellt werden, in denen R3 und R4 die gleichen Reste bedeuten, so geht man von einer Verbindung der Formel II aus, in der beide Reste A1 und A2 ein Halogenatom oder eine freie oder alkylsubstituierte Mercaptogruppe bedeuten, und setzt diese Verbindung mit der zweifachen molaren Menge oder einem Überschuss der Verbindung R5,H = R4H um.
Sollen Verbindungen mit verschiedenen Resten R3, und R4 hergestellt werden, so kann entweder erst der Rest A2 gegen den Rest R4 und dann der Rest A1 gegen den Rest R3 ausgetauscht werden, oder man verwendet als Ausgangsstoff eine Verbindung, in der A1 oder A2 bereits eine andere Bedeutung besitzt als ein Halogenatom oder eine freie oder alkylsubstituierte Mercaptogruppe.
Die Umsetzung erfolgt vorteilhaft in Gegenwart eines inerten organischen Lösungsmittels bei Temperaturen zwischen 0 und 200 C; falls A1 und/oder A2 ein Halogenatom bedeuten, ist die Anwesenheit eines halogenwasserstoffbindenden Mittels erforderlich. Als solches kann eine anorganische oder tertiäre organische Base verwendet werden; man kann auch das umzusetzende Amin, wenn man es in einem mindestens molaren Überschuss zugibt, als säurebindendes Mittel verwenden. Ein weiterer Überschuss dieses Ambis kann auch als Lösungsmittel verwendet werden.
Die Reaktionstemperatur hängt von der Reaktionsfähigkeit der Reaktionspartner ab. Im allgemeinen verläuft der Austausch eines Halogentoms gegen die einer der angegebenen Gruppen in Gegenwart eines halogenwasserstoffbindenden Mittels bereits bei Zimmertemperatur oder mässig erhöhter Temperatur, während der Austausch einer Mercaptogruppe gegen einen Rest der Formel

erst bei Temperaturen zwischen 100 und 2000 C vonstatten geht. Bei Anwendung eines niedrig siedenden Lösungsmittels oder einer Verbindung der Formeln R5H oder R4H mit einem niederen Siedepunkt wird die Reaktion zweckmässig in geschlossenen Gefäss durch geführt.
Soll eine Verbindung der Formel I erhalten werden, in der R3 die oben angegebenen Bedeutungen, mit Ausnahme der einer Gruppe -NR5R; innehat, so muss von solchen Verbindungen der Formel II ausgegangen werden, in der A1 diese Bedeutung bereits besitzt.
Die als Ausgangsstoffe verwendeten Verbindungen der Formel II können nach dem Verfahren des Schweizer Patentes Nr. 469 737 erhalten werden. Sollen die Reste A1 und/oder A2 freie oder alkylsubstituierte Mercaptogruppen bedeuten, so können diese Verbindungen direkt durch Ringschluss erhalten werden, bei5pielsweise durch Umsetzung eines 5-IminemZi- thian-4-carbonsäurethioamids mit einem Thioharnstoff. Verbindungen, in denen A, und A, Halogenatome bedeuten, können aus dem aus den Ann. - Chim. 49,1844-1849 (1959) bekannten 6,8-Dihydroxy-4H-m-dithüno[5,4-d]pyrimidin durch Umsetzung mit einem Phosphoroxyhalogenid hergestellt werden.
Verbindungen, in denen nur A2 ein Halogenatom bedeuten, können aus den entsprechenden gemäss dem Verfahren des Schweizer Patentes Nr. 469 737 herstellbaren 8-Hydroxyverbindungen durch Umsetzung mit einem Phosphoroxyhalogenid erhalten werden. Im Beispiel 1 wird die Herstellung einer Reihe solcher Ausgangsstoffe für das vorliegende Verfahren beschrieben.
Werden Verbindungen erhalten, in denen die Reste R3 und/oder R4 eine freie Aminogruppe bedeuten, so kann diese Gruppe gegebenenfalls nachträglich auf bekannte Weise in eine substituierte Aminogruppe übergeführt werden, beispielsweise durch Umsetzung mit reaktionsfähigen Estern der entsprechenden Alkohole, insbesondere mit Halogenwasserstoffsäure- oder Sulfonsäureestern. Aryl- und Pyridylreste können auf diese Weise nicht eingeführt werden.
Die Verbindungen der Formel I können gegebenenfalls nachträglich in ihre Säureadditionssalze mit physiologisch verträglichen anorganischen oder organischen Säuren übergeführt werden. Als solche kommen beispielsweise Salzsäure, Schwefelsäure, Phosphorsäure, Essigsäure, Zitronensäure, Weinsäure oder Maleinsäure in Frage.
Die neuen Verbindungen besitzen wertvolle pharmazeutische Eigenschaften, insbesondere wirken sie sedativ, analgetisch, antiphlogistisch, antipyretisch und cardiovaskulär; sie können ausserdem als Zwischenprodukte zur Herstellung von Schädlingsbekämpfungsmitteln verwendet werden.
Nachstehend wird die Herstellung einiger Ausgangsstoffe der Formel II beschrieben.
A: 6-Chlor-8-morpholino-4-H-m dithiino [5,4-d] pyrimidin
Zu einer kräftig gerührten Suspension von 1 g (0,0043 Mol) 6,8-Dichlor-4-H-m-dithüno[5,4-d]pyrimidin in 10 ml absolutem Äthanol werden bei Raumtemperatur 0,8 ml (0,0092 Mol) Morpholin getropft. Dabei ist eine schwach positive Wärmetönung zu beobachten; es tritt jedoch keine Lösung ein. Die Reaktionsmischung wird noch 1,5 Stunden bei Raumtemperatur gerührt.
Man saugt ab, wäscht die Kristalle mit Äthanol und Wasser und kristallisiert aus Äthanol um. Die Verbindung hat den F = 181-1820 C. Die Ausbeute beträgt 0,7 g (58 /o der Theorie).

B: 6-Morpholino-8-chlor-4-H-m dithüno[5,4-d]pyrimidin
6,75 g (0,025 Mol) 6-Morpholino-8-oxy-4-H-mdithiino[5,4-d]pyrimidin und 30 ml Phosphoroxychlorid werden 2 Stunden zum Rückfluss erhitzt. Es tritt völlige Lösung ein. Das überschüssige Phosphoroxychlorid wird im Vakuum abgezogen, der zähe Rückstand mit Eiswasser zersetzt und mehrmals mit Chloroform extrahiert.
Nach dem Abziehen des Chloroforms erhält man gelbbraune Kristalle, die aus Äthanol umkristallisiert werden.
Die Verbindung hat den F = 129-131 C. Die
Ausbeute beträgt 6,0 g (83 /o der Theorie)

5 g (0,02 Mol) 6-Äthylmercapto-8-hydroxy-4-H-m- dithiino [5,4-d] -pyrimidin und 50 ml Morpholin werden 8 Stunden unter Rühren unter Rückfluss erhitzt (Badtemperatur 1400 C). Die klare Reaktionsmischung wird nach Abkühlen in 250 ml Wasser gegossen, das ausgefallene kristalline Produkt abgesaugt, mit Wasser gewaschen und aus Dimethylformamid umkristallisiert. Die Verbindung hat den F = 285 C (unter Zersetzung). Die Ausbeute beträgt 4,8 g (89 /o der Theorie).

6-Isopropyl-8-methylmercapto-4H-m dithüno[5,4-d]pyrimidin
Eine kräftig gerührte Lösung von 1,0 g (0,004 Mol) 6-Isopropyl-8-mercapto-4-H-mdithiino[5,4-d]pyrimidin (hergestellt aus 6-Isopropyl-8-chlor-4H-m dithiino[5,4-d]pyrimidin und Natriumhydrogensulfid) in 15 ml 2n-NaOH versetzt man bei 50 C tropfenweise mit 2,8 g (0,02 Mol) Methyljodid. Nach einer Reaktionszeit von 15 Minuten wird kurz auf 400 C erwärmt, wobei sich eine kristalline Substanz abscheidet. Diese wird abgenutscht, mit Wasser gewaschen und aus Athanol umkristallisiert.
Schmelzpunkt 753 C, Ausbeute 0,6 g (58 /o der Theorie).

Beispiel 1
6-Morpholino-8-pyrrolidino-4-H-m dithüno[5,4-d]pyrimidin
3,0 g (0,01 Mol)6-Athylmercapto-8- pyrrolidino-4-H-m-dithüno[5,4-d]pyrimidin und 50 ml Morpholin werden 30 Stunden in der Bombe auf 1750 C erhitzt. Nach dem Abkühlen wird die Reaktionsmischung in Wasser gegossen, wobei sich ein kristallines Produkt abscheidet. Es wird abgesaugt, mit Wasser nachgewaschen und aus Äthanol umkristallisiert. Die Verbindung hat den F = 145-146 C. Die Ausbeute beträgt 2,1 g (65 /o der Theorie).

Beispiel 2
6-Pyrrolidino-8-morpholino-4-H-m dithiino [5,4-d]pyrimidin.
1 g (0,0034 Mol) 6-Chlor-8-morpholino-4-H-mdithiino-[5,4-d]pyrimidin und 15 ml Pyrrolidin werden 4 Stunden unter Rückfluss erhitzt. Nach dem Abkühlen giesst man die klare Lösung in 100 ml Wasser. Die sich abscheidende Kristalle werden abgesaugt, mit Wasser und Äthanol umkristallisiert. Die Verbindung hat den F = 133-134 C.
Die Ausbeute beträgt 0,8 g (71% der Theorie).

Beispiel 3
6-Äthylmercapto-8-pyrrolidino-4-H-m dithiino [5,4-d]pyrimidin.
1 g (0,0038 Mol ) 6-Sithylmercapto-8-chlor-4-H-m- dithüno-[5,4-d]pyrimidin und 2 ml (0,024 Mol) Pyrrolidin werden zusammengegeben, wobei unter heftiger Reaktion Lösung und Sieden unter Rückfluss eintritt. Man lässt ohne zusätzliches Erhitzen 1 Stunde stehen, saugt die ausgefallenen Kristalle ab und kristallisiert aus Äthanol um. Die Verbindung hat den F = 104-105 C. Die Ausbeute beträgt 0,8 g (70 /o der Theorie).

Beispiel 4 6,8-Dimorpholino-444-m- dithiino [5,4-d]pyrimidin.
5,8g (0,02) 6-Morpholino-8-chlor-4-H-mdithüno[5,4-d]-pyrimidin und 15 ml Morpholin werden 3 Stunden zum Rückfluss erhitzt (Badtemperatur 150 C). Nach dem Abkühlen wird die reaktionsmischung mit Äther versetzt, das ausgefallene Produkt abgenutscht, mit Wasser nachgewaschen und aus Äthanol umkristallisiert. Die Verbindung hat den F = 1260 C. Die Ausbeute beträgt 5,4 g (790/o der Theorie).

Beispiel 5
6-Morpholino-8-amino-4-H-m dithüno['%-D)pyrimklin.
5,8 g (0,02 Mol) 6-Morpholino-8-chlor-4-H-m dithiino [5,4-d]-pyrimidin und 50 ml kondensiertes Ammoniak werden in einer Bombe 5 Stunden auf 100 C (Badtemperatur) erhitzt.

6-Morpholino-8-äthanolamino-4-H-m dithiino [)5,4-d]pyrimidin.
5,8 g (0,02 Mol) 6-Morpholino-8-chlor-4-H-mdithiino[5,4-d]-pyrimidin und 15 ml Äthanolamin werden 2 Stunden auf 1500 C (Bad-temperatur) erhitzt. Das Reaktionsprodukt kristallisiert beim Abkühlen der Lösung aus. Es wird abgenutscht, mit Äther und Wasser nachgewaschen und aus Äthanol umkristallisiert. Die Verbindung hat den F=145-146 C. Die Ausbeute beträgt 3,5 g (58 % der Theorie).

Beispiel 7
6-Phenyl-8-morpholino-4-H-m dithüno[5,4-d]pyrimidin.
3,7 g (0,013 Mol) 6-Phenyl-8-chlor-4-H-mdithiino [5,4-d]-pyrimidin werden mit 10 ml Morpholin 24 Stunden am Rückfluss erhitzt. Nach dem Abkühlen wird das überschüssige Morpholin unter vermindertem Druck abdestilliert und der Rückstand in 30ml 2-normaler Salzsäure aufgenommen. Man schüttelt zweimal mit je 25 ml Äther aus und stellt die wässrige Phase dann alkalisch. Das abgeschiedene Öl wird in Chloroform aufgenommen, mit Wasser gewaschen und über Natriumsulfat getrocknet.
Nach Abdestillation des Lösungsmittels bleibt ein zäher Rückstand, der beim Anreiben durchkristallisiert. Das Produkt wird abgenutscht und zweimal aus Essigester umkristallisert. Die Verbindung hat den F = 1030 C. Die Ausbeute beträgt 1,8 g (41 /o der Theorie).

Beispiel 8
2-Methyl-6-propyl-8-morpholino-4-H-m dithüno[5,4-d]pyrimidin-maleinat
8,4 g (0,032 Mol) 2-Methyl-6-propyl-8-chlor-4-H-mdithiino-[5,4-d]pyrimidin und 20 ml Morpholin werden 24 Stunden unter Rückfluss erhitzt. Nach dem Abkühlen wird das überschüssige Morpholin unter vermindertem Druck abdestilliert und der halbfeste Rückstand mit 2-normaler Salzsäure angesäuert. Man schüttelt zweimal mit 20 ml Ather aus und stellt dann die wässrige Phase alkalisch. Das ausgefallene Oel wird in Äther aufgenommen, mit Wasse ausgewaschen und über Natriumsulfat getrocknet.
Nach Eindampfen des Lösungsmittels bleiben 6,85 g öliger Rückstand. Dieser wird in etwas absolutem Atha- nol gelöst und mit 22 ml einer molaren, äthanolischen Maleinsäurelösung versetzt. Nach Abdampfen des Äthanols bleibt ein kristalliner Rückstand, der mehrmals aus absolutem Essigester umkristallisiert wird.
Die Verbindung hat den F = 94-950 C. Die Ausbeute beträgt 5 g (37 /o der Theorie).

Example 6
6-Morpholino-8-ethanolamino-4-H-m dithiino [) 5,4-d] pyrimidine.
5.8 g (0.02 mol) of 6-morpholino-8-chloro-4-H-mdithiino [5,4-d] pyrimidine and 15 ml of ethanolamine are heated to 1500 ° C. (bath temperature) for 2 hours. The reaction product crystallizes out on cooling the solution. It is suction filtered, washed with ether and water and recrystallized from ethanol. The compound has the F = 145-146 C. The yield is 3.5 g (58% of theory).

Example 7
6-phenyl-8-morpholino-4-H-m dithuno [5,4-d] pyrimidine.
3.7 g (0.013 mol) of 6-phenyl-8-chloro-4-H-mdithiino [5,4-d] -pyrimidine are refluxed with 10 ml of morpholine for 24 hours. After cooling, the excess morpholine is distilled off under reduced pressure and the residue is taken up in 30 ml of 2N hydrochloric acid. It is extracted twice with 25 ml of ether each time and the aqueous phase is then made alkaline. The separated oil is taken up in chloroform, washed with water and dried over sodium sulfate.
After the solvent has been distilled off, a viscous residue remains, which crystallizes on trituration. The product is suction filtered and recrystallized twice from ethyl acetate. The compound has the F = 1030 C. The yield is 1.8 g (41 / o of theory).

Example 8
2-methyl-6-propyl-8-morpholino-4-H-m dithüno [5,4-d] pyrimidine maleate
8.4 g (0.032 mol) of 2-methyl-6-propyl-8-chloro-4-H-mdithiino- [5,4-d] pyrimidine and 20 ml of morpholine are refluxed for 24 hours. After cooling, the excess morpholine is distilled off under reduced pressure and the semi-solid residue is acidified with 2N hydrochloric acid. It is extracted twice with 20 ml of ether and the aqueous phase is then made alkaline. The precipitated oil is taken up in ether, washed out with water and dried over sodium sulfate.
After evaporation of the solvent, 6.85 g of an oily residue remain. This is dissolved in a little absolute ethanol and mixed with 22 ml of a molar, ethanolic maleic acid solution. After evaporation of the ethanol, a crystalline residue remains, which is recrystallized several times from absolute ethyl acetate.
The compound has the F = 94-950 C. The yield is 5 g (37 / o of theory).

3.1 g (0.01 mol) of 6- (2-methylmorpholino) -8-äthylamino4-Hm-dithüno [5,4-d] pyrimidine, dissolved in 100 ml of absolute ethyl acetate, are treated with ethereal hydrochloric acid until the solution is Congo acidic.

6-Morpholino-8-benzylamino-4H-m dithiino [5,4-d] pyrimidine.
0.5 g (0.0018 mol) 6-morpholino-8-amino-4-H-m dithiino- [5,4-d] pyrimidine [melting point 137-139 C (ethanol); prepared from 4-carbamidino-5-imino-m-dithiane and N-guanylmorpholine and 0.35 g (0.0018 mol) of benzyl bromide are dissolved in 15 ml of absolute benzyl alcohol and heated to 150 ° C. for 24 hours.
