Verfahren zur Herstellung von Polymethinen
Die Erfindung betrifft ein Verfahren zur Herstellung von Polymethinen der Formel

wobei R1 Wasserstoff oder Alkyl R Alkyl oder Aryl X Arylamino oder Dialkylamino bedeuten und n eine ganze Zahl von 2 bis 4 darstellt.
Polymethine werden als optische Sensibilisatoren in der Fotoindustrie verwendet oder dienen zu ihrer Herstellung. Es ist bekannt, dass man Pentamethine (I, n=2) durch Spaltung quartärer Pyridiniumverbindungen mit Aminen herstellen kann. So führt die Umsetzung von N-2,4-Dinitrophenylpyridiniumchlorid mit Methylanilin zur Bildung von Bismethylanilinopentamethin. Heptamethine (I,n=3) werden durch Kondensation von l-Arylalkyl-aminopropen-(l)-al-(3) mit Malonsäure oder ihren Derivaten hergestellt.
Hepta-(I,n=3) und Nonamethine (I,n4) können durch Kettenverlängerung der Pentamethine mit Hilfe von Grignardverbindungen gewonnen werden. Zum Bei spiel führt die Reaktion von 5-Methylanilinopentadien (2,4)-al-( 1) mit Äthinylmagnesiumbromid zu Heptadien (2,4)-in-(6)-al-(1), das mit sekundären Aminen Bis aminoheptamethine ergibt. In analoger Weise führt die Umsetzung von 5-Methylanilinopentadien-(2,4)-al-(1) mit 1 -Methoxybuten-( 1 )-in-(3)-ylmagnesiumbromid zu einem Aldehyd, der nach katalytischer Hydrierung und Zugabe von Dimethylamin Bisdimethylaminononamethin ergibt.
Weiterhin sind Verfahren bekannt, nach denen ungesättigte Trialkoxyverbindungen oder ungesättigte Acetale durch Formylierung in Hepta-(I,n=3) und Nonamethine (In= 4) überführt werden. So ergibt die Umsetzung von 1.1 .3-Triäthoxyhexen-4 mit Chlormethenyldimethylammoniumchlorid und anschliessender Zugabe von Dimethylamin Bisdimethylaminohetpamethin. In analoger Weise führt die Reaktion von 1,1,3 Triäthoxyoctadien-(4,6) bzw. vom Acetal des Octatrien (2,4,6)-al-(l) mit Chlormetenyldimethylammoniumchlorid zu Nonamethinen.
Die angeführten Verfahren zur Herstellung von Polymethinen weisen Mängel auf. Die in der Literatur angegebenen Ausbeuten beziehen sich auf ungereinigte Produkte und lassen sich technisch nicht reproduzieren.
Es treten schwer abzutrennende Nebenprodukte auf. In einigen Fällen sind die Synthesewege kompliziert und führen erst über viele Reaktionsstufen zu den gewünschten Produkten. Das Verfahren zur Herstellung von Pentamethin aus quartären Pyridiniumverbindungen hat den Nachteil, dass die Ausgangsverbindungen teuer sind.
Der Erfindung liegt die Aufgabe zugrunde, die Herstellung substituierter Polymethine der Formel I, ausgehend von einfachen Ausgangsverbindungen mit guter Ausbeute an reinen Produkten zu ermöglichen.

Die Umsetzungen können bei einer Temperatur von -30 bis +350, vorzugsweise bei Zimmertemperatur, gegebenenfalls in Gegenwart eines Lösungsmittels, vorzugsweise von Tetrahydrofuran, durchgeführt werden.

In ähnlicher Weise gewinnt man ausgehend vom 5 Methylanilinopentadien-(2,4)-al-(1) und Acetaldehyd bzw. aus Sorbinaldehyd und Dimethylformamid Heptamethine, aus Crotonaldehyd und Dimethylformamid Pentamethine.
Wie bereits erwähnt, kann statt des Aldehyds der Formel III auch das entsprechende Acetal oder die Schiff'sche Base umgesetzt werden.
Ein Beispiel für die Umsetzung einer Verbindung der Formel II mit einer Schiff'schen Base (Immoniumsalz) der Formel III ist die Reaktion von 5-Methylanilinopentadien-(2, 4)-al-(1) mit Bis (1-äthylidenpiperidinium)hexachlorostannat unter Zusatz katalytischer Mengen von Pyridin zur Bildung von Heptamethin

Die Erfindung soll nachstehend an einigen Ausführungsbeispielen näher erläutert werden.
Beispiel I Heptamethin
1 Mol 5-Methylanilinopentadien-(2, 4)-al-(1) (Zinkkealdehyd) wird in 1 1 Tetrahydrofuran gelöst und bei -30 "C unter Rühren mit 1,2 Mol Phosphoroxychlorid versetzt. Bei gleicher Temperatur wird 1 Mol Acetaldehyd zugegeben, dann lässt man die Temperatur innerhalb einer Stunde auf + 20 0C ansteigen. Das Reaktionsgemisch wird über Nacht bei Zimmertemperatur stehengelassen und dann auf Eis gegeben. Es werden 3 Mol Dimethylaminhydrochlorid zugefügt, darauf mit 10 0/oiger NaOH alkalisch gemacht und mit 1,5 Mol Natriumperchlorat in wenig Wasser versetzt. Das ausgeschiedene Bisdimethylaminoheptamethinperchlorat wird mit wenig Wasser gewaschen und getrocknet.
Ausbeute: 13 O/o der Theorie.
Beispiel 2 Nonamethin
2 Mol Reaktionsprodukt aus Phosphoroxychlorid und Dimethylformamid werden unter Rühren bei 25-30 OC mit einem Mol Octatrien-(2,4,6)-al-(1) (Fraktion 45-70 OC 0,1 mm, dargestellt nach bekannten Vorschriften durch Selbstkondensation von Crotonaldehyd) versetzt. Nach Stehen über Nacht bei Zimmertemperatur wird die Reaktionsmischung auf Eis gegeben und mit Soda alkalisch gemacht. Das ausgefallene Produkt wird abgesaugt und in Alkohol gelöst. Zu dieser Lösung wird eine alkoholische Lösung von 2 Mol Methylanilin und 1 Mol Perchlorsäure gegeben. Nach einer Stunde Stehen bei Zimmertemperatur wird das Bismethylanilinononamethinperchlorat abgesaugt.
Ausbeute: 29 01o der Theorie (sehr reines Produkt)
Beispiel 3 Nonamethin
2 Mol Reaktionsprodukt aus Phosphoroxychlorid und Dimethylformamid werden unter Rühren bei 25-30 0C mit einem Mol Octatrien-(2,4,6)-al-(1) (Fraktion 45-70 OC 0,1 mm dargestellt nach bekannten Vorschriften durch Selbstkondensation von Crotonaldehyd) versetzt. Nach Stehen über Nacht bei Zimmertemperatur wird die Reaktionsmischung auf Eis gegeben. Dann werden 2 Mol Natriumperchlorat in wenig Wasser zugesetzt. Das ausgefallene o-Chlor-o'- dimethylammoniumnonamethinperchlorat wird abgesaugt.
Ausbeute: 30 o/o der Theorie.
Beispiel 4 Heptamethin
2 Mol Reaktionsprodukt aus Phosphoroxychlorid und Dimethylformamid werden unter Rühren bis 25-30" mit einem Mol Sorbinalaldehyd versetzt. Nach Stehen über Nacht bei Zimmertemperatur wird die Reaktionsmischung, wie unter Beispiel 1 angegeben, aufgearbeitet. Die schliesslich erhaltenen Kristalle werden in Athanol aufgenommen und mit 1,5 Mol Dimethylamin und 1,5 Mol Dimethylaminperchlorat 10 Minuten unter Rückfluss erhitzt. Nach dem Abkühlen wird abgesaugt.
Ausbeute an Bisdimethyl aminoheptamethinper- chlorat: 36 O/o der Theorie
Beispiel 5 Heptamethin
Zum Gemisch von 40 cm:3 Dimethylformamid und 3,2 g (0,1 Mol) Methanol werden bei 10 bis 15 OC 31 g (0,2 Mol) Phosphoroxychlorid getropft. Dazu gibt man bei 65 bis 70 OC innerhalb von 5 Minuten 9,6 g (0,1 Mol) Sorbinaldehyd, rührt 3 Stunden bei 80 C und giesst anschliessend auf ca. 300 g Eis. Nach Zugabe von gesättigter wässriger Natriumperchloratlösung scheidet sich das [7-Chlor-heptatrien-(2,4,6)-yliden] -dimethyl- ammoniumperchlorat als zähes öl aus.
Das Reaktionsprodukt wird in 50cm:3 heissem Aceton gelöst und zusammen mit 20 g Anilin in 100 cm3 Äthanol 5 Minuten zum Sieden erhitzt.
Das ausgefallene Heptamethinsalz wird abgesaugt und mit heissem Äthanol gewaschen.
Ausbeute: 12 g (32 0/o) Schmp.: 168 bis 171 "C
Beispiel 6 Nonamethin
46 g (0,3 Mol) Phosphoroxychlorid werden bei 0 "C in 80 cm Dimethylformamid getropft. Zu dieser Mischung gibt man innerhalb von 30 Minuten bei 45 OC 12,2 g (0,1 Mol) kristallines Octatrien-(2,4,6)-al-(1) in 20 cm3 Dimethylformamid. Bei Verwendung von rohem Octatrienal, Fraktion 52 bis 65 C/0,1 Torr, wird eine geringere Ausbeute (10 bis 15 O/o) erhalten, ohne dass störende Nebenprodukte auftreten. Es wird 2 Stunden bei 50 OC gerührt und auf ca. 500 g Eis gegossen. Nach Alkalisieren mit Natriumcarbonat fällt das 9-Chlor-nonatetraen-(2,4,6,8)-al-(1) aus.
Das Reaktionsprodukt wird in 50 cm3 Äthanol gelöst und mit 20 g Anilin und 5 cml kons. Salzsäure 2 Stunden bei Raumtemperatur stehengelassen. Anschliessend wird das Nonamethinsalz rnit Äthanol oder Aceton mehrmals ausgekocht.
Ausbeute: 14 g (42 O/o) Schmp.: 186 bis 188 "C
Beispiel 7
1 Mol 5-Methylanilinopentadien-(2,4)-al-(1) wird in Acetanhydrid gelöst und mit einem Überschuss an Immoniumsalz (Bis(1-aethylidenpiperidinium) hexachlorestannat) versetzt. Nach Zugabe katalytischer Mengen Pyridin wird 3 Stunden bei Zimmertemperatur stehengelassen. Der ausfallende Farbstoff wird abgesaugt.
Ausbeute an Bisdimethylaminoheptamethinsalz: 32 O/o der Theorie.
Beispiel 8 Nonamethin
Eine Lösung von 1 Mol 5-Methylanilinopentadien (2, 4)-al-(1) in Acetanhydrid mit einem Überschuss an Immoniumsalz (Bis-( 1 -buten-(2)-ylidenpiperidinium) hexachlorostannat) wird mit katalytischen Mengen Pyridin versetzt. Nach 3 Stunden Stehen bei Zimmertemperatur wird nach Beispiel 8 aufgearbeitet.
Ausbeute an Bisdimethylaminononamethinperchlorat: 23 O/o der Theorie.
