Der in den obigen Formeln mit Ar bezeichnete Arylrest ist insbesondere der Phenylrest oder ein Halogenphenylrest wie beispielsweise p-Chlorphenyl.
Die durch Verätherung oder Veresterung geschützte und als solche mit R bezeichnete Hydroxylgruppe ist beispielsweise eine solche, in welcher der Wasserstoff der freien Hydroxylgruppe durch eine Alkylgruppe, wie Methyl, Aethyl, n-Propyl, Isopropyl, n-Butyl, Isobutyl, oder durch eine Alkoxyalkylgruppe, wie Methoxyäthyl und Äthoxyäthyl, oder durch einen Aryl- oder Aralkylrest, beispielsweise den Phenylrest oder den Benzyl- oder Phenyläthylrest, oder durch einen Arylsulfonylrest, wie den p-Toluolsulfonsäure- rest, oder aber durch einen Acylrest, beispielsweise durch den Acetylrest, oder durch den Tetrahydropyranylrest ersetzt ist.
Die als Ausgangssubstanzen verwendeten substituierten 4-Amino-penten-(3)-one-(2) der Formel II sind neue Verbindungen, welche beispielsweise durch Umsetzung der entsprechenden 2,4-Pentandione mit Ammoniak hergestellt werden können. So erhält man durch Behandlung von l-Methoxy-3-phenylazo-pentan- dion-(2,4) mit konzentriertem Ammoniak bei Raumtemperatur.
Die Kondensation der substituierten 4-Amino-penten-(3)-one der Formel II mit Cyanacetamid wird vorteilhafterweise in Gegenwart eines alkalischen Kondensationsmittels und bei erhöhter Temperatur, insbesondere bei einer Temperatur von etwa 40-70 C, durchgeführt. Als alkalische Kondensationsmittel eignen sich beispielsweise Alkalihydride wie Natrium- oder Kaliumhydrid, und insbesondere Alkalimetallalkoholate, wie Natriummethylat oder Natrium äthyl at, vorzugsweise in alkoholischer Lösung. Es bildet sich hierbei in nahezu theoretischer Ausbeute ein einheitliches Reaktionsprodukt der Formel I, in welcher R1 das Alkalimetall des als Kondensationsmittel verwendeten Alkalimetallalkoholates, also beispielsweise Natrium oder Kalium, bedeutet.
Die vorliegende Erfindung betrifft ferner die Verwendung der nach dem erfindungsgemässen Verfahren erhaltenen Verbindungen der Formel I zur Herstellung yon Pyridoxin, durch Umsetzung der Verbindung der Formel I mit einem Arylsulfonylhalogenid, Hydrierung der so erhaltenen Verbindung unter reduktiver Spaltung der Azobindung zur entsprechenden 3-Aminoverbindung, Überführung der in 5-Stellung befindlichen Cyanogruppe in die Aminomethylgruppe unter gleich- zeitiger Entfernung der Arylsulfonyloxygruppe, tZber- führung der so erhaltenen Verbindung in die entsprechende 4,5-Di-(brom-methyl)-Verbindung und anschliessende Hydrolyse dieser Verbindung.
Die Umset , zung der Verbindung der Formel I mit einem Arylsulfonylhalogenid, beispielsweise mit p-Toluolsulfochlorid, wird zweckmässig in einem geeigneten Lösungsmittel, wie Dimethylformamid, Aceton oder Wasser, durch führt, unter Bildung einer Verbindung der Formel

Die Hydrierung dieser Verbindung erfolgt zweckmässig mittels Palladium-Kohle in essigsaurer Lösung, wobei die Azobindung reduktiv zur entsprechenden 3-Aminoverbindung gespalten wird. Die Überführung der in 5-Stellung befindlichen Cyanogruppe in die Aminomethylgruppe erfolgt beispielsweise durch katalytische Reduktion mittels Raney-Nickel in alkoholischem Ammoniak, wobei gleichzeitig die Arylsulfonyloxygruppe abgespalten wird, wodurch man zu einer Verbindung der Formel

gelangt.
Die Überführung der in dieser Formel IV in 3- und 5-Stellung befindlichen Aminogruppen in Hydroxylgruppen erfolgt zweckmässig durch Überführung der Verbindung der Formel IV in das Dihydrochlorid mittels Salzsäure und anschliessender Behandlung des Dihydrochlorids mit salpetriger Säure. Die Überfüh- aung der so erhaltenen Verbindung in die entsprechende 4,5 -Di-(brommethyl)-Verbindung erfolgt eweckmässig durch Behandlung mit etwa 480/ger Bromwasserstoffsäure und die hierauf folgende Hydrolyse der 4,5-Di-(brommethyl)-Verbindung zu Pyrido xinhydrochlorid zweckmässig durch Erhitzen in Wasser in Gegenwart von Silberchlorid.
Beispiel
Es werden 4,6 g Natrium in 300 ml absolutem Äthanol gelöst, worauf man die so erhaltene Lösung unter Rühren mit 46,5 g 1-Methoxy-3-phenylazo-4-ami- no-penten-(3)-on-(2) versetzt. Anschliessend werden 17 g Cyanacetamid zugegeben und das Reaktionsgemisch wird langsam unter Rühren bis zum Siedepunkt erhitzt. Hierbei klärt sich die ursprüngliche Suspension unter starker Ammoniakentwicklung. Bereits nach 15 Minuten beginnt die Ausscheidung des Natriumsalzes von 2-Methyl-3 -phenylazo-4-methoxy-methyl-5-cyan- 6-hydroxy-pyridin. Es wird nun noch 2 Stunden am Rückfluss erhitzt, worauf dann das Reaktionsgemisch auf 00 abgekühlt und vom Rückstand abgenutscht wird.
Nach dem Trocknen erhält man 64 g des Natriumsalzes von 2-Methyl-3-phenylazo-4-methoxymethyl-5-cyan- 6-hydroxy-pyridin, welches in Wasser leicht löslich ist.
30, 4 g des so erhaltenen Natriumsalzes, welches vorher gesiebt wurde, werden in einem mit einer Rührvorrichtung, einem Thermometer, einem Tropftrichter und einem Rückflusskühler versehenen 250 ml 4-Halskolben in 60 ml Dimethylformamid gelöst, worauf die so erhaltene Lösung während 30 Minuten mit einer Lösung von 28, 5 g p-Toluolsulfochlorid in 30ml Dimethylformamid tropfenweise versetzt wird, wobei die Temperatur auf 450 C steigt. Man rührt nun weitere 3 Stunden bis die Temperatur wieder auf Raumtemperatur gesunken ist und destilliert hierauf das Dimethylformamid am Vakuumverdampfer ab.
Der verbleibende Rückstand wird mit 40 ml Wasser, 40 ml Äther und 40 ml 3 n Natronlauge geschüttelt, wobei sich 2-Methyl-3 -phenylazo-4-methoxymethyl-5-cyan-
6-tosyloxy-pyridin kristallin abscheidet. Das so erhaltene tiefrot gefärbte Produkt schmilzt bei 1500, kristallisiert dann nach einiger Zeit in farblosem Zustande und schmilzt zuletzt wiederum bei 1930, Ausbeute
30,5 g.
27 g des so erhaltenen 2-Methyl-3-phenylazo 4-methoxymethyl-5-cyan-6-tosyloxy-p yridin werden in
500 ml 800/oiger Essigsäure suspendiert und hierauf in Gegenwart von 3 g Palladium-Kohle bei Raumtemperatur und unter Normaldruck hydriert. Die Hydrierung kommt nach Aufnahme von 2 Mol Wasserstoff zum Stillstand, was etwa 2 Stunden erfordert. Hierauf wird die Essigsäure abgedampft und der Rückstand nach Abkühlen abfiltriert. Man erhält auf diese Weise
17,5 g von 2-Methyl-3-amino-4-methoxymethyl5-cyan-6-tosyloxy-pyridin vom Schmelzpunkt 125-126 . Beim Umkristallisieren dieses Produktes tritt keine Schmelzpunktsveränderung ein.
Es werden nun 17,5 g des so erhaltenen 2-Methyl 3-amino-4-methoxymethyl-5-cyan-6-tosyloxy-pyridin in 300 mol alkoholischem Ammoniak (etwa 80/oig durch Sättigung von Alkohol mit gasförmigen Ammoniak bei Raumtemperatur erhalten) in Gegenwart von 20 g Raney-Nickel bei Raumtemperatur und unter Normaldruck hydriert. Die Wasserstoffaufnahme erfolgt hierbei sehr regelmässig und kommt nach Aufnahme von 3,25 Liter Wasserstoff zum Stillstand. Hierauf wird vom Katalysator abgetrennt und das Filtrat im Vakuum eingedampft. Um den restlichen Ammoniak zu entfernen, wird der Rückstand in 40 mol Alkohol gelöst und die so erhaltene Lösung im Vakuum zur Trockene eingedampft.
Das zurückbleibende viskose Öl wird nun nochmals in 20 ml Alkohol gelöst und mit alkoholischer Salzsäure stark kongosauer gestellt.
Die hierbei ausfallenden Kristalle werden abfiltriert und mit Aceton gewaschen. Man erhält ohne Aufarbeitung der Mutterlaugen 9 g 2-Methyl-3-amino-4-metho- xymethyl-5-aminomethyl-pyridin-dihydrochlorid mit einem Schmelzpunkt von 228". Eine weitere Menge dieser Substanz kann noch aus den Mutterlaugen gewonnen werden.
Die Weiterverarbeitung dieses Produktes zum Pyridoxin kann gemäss den Angaben in J. Am. chem.
Soc. 61 (1939), 1245-1247 erfolgen.
Das als Ausgangsmaterial verwendete l-Methoxy3-phenylazo-4-amino-5-penten-(3)-on-(2) kann wie folgt hergestellt werden: In einen 5-Liter-Rundkolben wird ein Gemisch von 145,6 g Anilin (roh) und 1500 g Eis eingebracht und es werden diesem Gemisch unter kräftigem Rühren und bei Aufrechterhaltung einer Temperatur von etwa 00, 470 mol konzentrierte Salzsäure langsam zugetropft. Anschliessend wird bei weiterer Aufrechterhaltung einer Temperatur von 0 eine Lösung von 114 g Natriumnitrit in 220 ml Wasser zugetropft und hierauf werden noch 180 ml konzentrierte Salzsäure zugegeben. Auf diese Weise erhält man die für die im folgenden beschriebene Umsetzung mit dem Natriumsalz von l-Methoxy-2-hydroxy-penten- (2)-on-(4) benötigte Diazoverbindung.
In einem mit Aussenkühlung, einer Rührvorrichtung und einem Thermometer versehenen Emailgefäss wurden 237 g des Natriumsalzes von l-Methoxy-2-hydroxy-penten-(2)-on-(4) in 2000 mol Methanol gelöst, worauf diese Lösung mit einer Lösung von 468 g Natriumacetat in 1000 ml Wasser versetzt wird. Hier auf wird tropfenweise das durch Diazotierung von Anilin gemäss den obigen Angaben erhaltene Reaktionsgemisch unter Aufrechterhaltung einer Temperatur von 0 zugesetzt. Nach beendeter Zugabe wird das Reaktionsgemisch noch 30 Minuten bei 0 gerührt und anschliessend abgenutscht. Der auf der Nutsche verbleibende Rückstand wird einige Male mit Wasser gewaschen und zuletzt getrocknet. Man erhält auf diese Weise 337 g 1 -Methoxy-3 -phenylazo-pentandion-(2,4).
SChmelzpunkt: 69-700 C.
200 g der so erhaltenen Verbindung werden in 3000 ml konzentriertem Ammoniak während 96 Stunden bei Raumtemperatur gerührt, wobei gasförmiger Ammoniak sehr langsam in die Suspension eingeleitet wird. Hierauf wird das Reaktionsgemisch abgenutscht, mit Wasser gewaschen und im Vakuum bei Raumtemperatur getrocknet. Man erhält 175 g eines Rohproduktes, welches bei 106 sintert und bei 120-1230 schmilzt. 175 g dieses Rohproduktes werden eine Stunde lang mit 500 ml Methanol bei 500 C verrührt.
Nach Abkühlen auf Raumtemperatur wird abgenutscht, wobei man 138 g 1-Methoxy-3-phenylazo-4- amino-penten-(3)-on-(2) mit einem Schmelzpunkt von 135 erhält. Dieses Produkt wird der eingangs beschriebenen Reaktionsfolge unterworfen.

A uniform reaction product of the formula I, in which R1 is the alkali metal of the alkali metal alcoholate used as the condensing agent, for example sodium or potassium, is formed here in almost theoretical yield.
The present invention also relates to the use of the compounds of the formula I obtained by the process according to the invention for the preparation of pyridoxine by reacting the compound of the formula I with an arylsulfonyl halide, hydrogenation of the compound thus obtained with reductive cleavage of the azo bond to the corresponding 3-amino compound, conversion the cyano group in the 5-position into the aminomethyl group with simultaneous removal of the arylsulfonyloxy group, conversion of the compound thus obtained into the corresponding 4,5-di- (bromomethyl) compound and subsequent hydrolysis of this compound.

30.4 g of the sodium salt obtained in this way, which has been sieved beforehand, are dissolved in a 250 ml 4-necked flask equipped with a stirrer, a thermometer, a dropping funnel and a reflux condenser in 60 ml of dimethylformamide, whereupon the resulting solution for 30 minutes with a solution of 28.5 g of p-toluenesulfonyl chloride in 30 ml of dimethylformamide is added dropwise, the temperature rising to 450.degree. The mixture is then stirred for a further 3 hours until the temperature has dropped back to room temperature and the dimethylformamide is then distilled off on a vacuum evaporator.
The remaining residue is shaken with 40 ml of water, 40 ml of ether and 40 ml of 3N sodium hydroxide solution, 2-methyl-3-phenylazo-4-methoxymethyl-5-cyano-
6-tosyloxy-pyridine separates in crystalline form. The deep red colored product thus obtained melts at 1500, then crystallizes after some time in a colorless state and finally melts again at 1930, yield

27 g of the 2-methyl-3-phenylazo 4-methoxymethyl-5-cyano-6-tosyloxy-pyridine thus obtained are in
500 ml of 800% acetic acid suspended and then hydrogenated in the presence of 3 g of palladium-carbon at room temperature and under normal pressure. 17.5 g of the 2-methyl 3-amino-4-methoxymethyl-5-cyano-6-tosyloxypyridine thus obtained are now obtained in 300 mol of alcoholic ammonia (about 80% by saturating alcohol with gaseous ammonia at room temperature ) hydrogenated in the presence of 20 g of Raney nickel at room temperature and under normal pressure. The hydrogen uptake takes place very regularly and comes to a standstill after the uptake of 3.25 liters of hydrogen. The catalyst is then separated off and the filtrate is evaporated in vacuo. In order to remove the remaining ammonia, the residue is dissolved in 40 mol of alcohol and the resulting solution is evaporated to dryness in vacuo.
The remaining viscous oil is now dissolved again in 20 ml of alcohol and made strongly Congo acidic with alcoholic hydrochloric acid.
The crystals which precipitate out are filtered off and washed with acetone. Without working up the mother liquors, 9 g of 2-methyl-3-amino-4-methoxymethyl-5-aminomethylpyridine dihydrochloride with a melting point of 228 "are obtained. A further amount of this substance can still be obtained from the mother liquors.

The 1-methoxy-3-phenylazo-4-amino-5-penten- (3) -one- (2) used as starting material can be prepared as follows: A mixture of 145.6 g of aniline is placed in a 5 liter round bottom flask ( raw) and 1500 g of ice are introduced and concentrated hydrochloric acid is slowly added dropwise to this mixture, while stirring vigorously and while maintaining a temperature of about 0.00470 mol. Then, while maintaining a temperature of 0, a solution of 114 g of sodium nitrite in 220 ml of water is added dropwise and 180 ml of concentrated hydrochloric acid are then added. In this way, the diazo compound required for the reaction described below with the sodium salt of l-methoxy-2-hydroxypentene- (2) -one- (4) is obtained.

200 g of the compound obtained in this way are stirred in 3000 ml of concentrated ammonia for 96 hours at room temperature, gaseous ammonia being introduced very slowly into the suspension. The reaction mixture is then filtered off with suction, washed with water and dried in vacuo at room temperature. 175 g of a crude product are obtained which sinters at 106 and melts at 120-1230. 175 g of this crude product are stirred with 500 ml of methanol at 500 ° C. for one hour.
After cooling to room temperature, it is suction filtered, 138 g of 1-methoxy-3-phenylazo-4-aminopenten- (3) -one- (2) having a melting point of 135 being obtained. This product is subjected to the reaction sequence described at the outset.
