Verfahren zur Herstellung von Phthalimidinderivaten Die vorliegende Erfindung bezieht sich auf ein Ver fahren zur Herstellung von neuen Phthalimidinderivaten mit wertvollen biologischen Eigenschaften. Die neuen Verbindungen können daher als Arzneimittel und als Zwischenprodukte für die Herstellung von Arzneimitteln eingesetzt werden.
Die neuen Produkte entsprechen der folgenden allgemeinen Formel:

worin R1 für ein Wasserstoffatom, für gegebenen falls durch freie, veresterte oder verätherte Oxy- oder Mercaptogruppen, durch Aminogruppen, Carboxylgrup- pen oder Halogenatome substituierte Alkyl- oder Alke- nylreste mit 1 bis 6 Kohlenstoffatomen oder für einen Aralkyl-, Cycloalkyl- oder Arylrest steht und m und n die Zahlen 0, 1 oder 2 bedeuten, wobei die Summe von m und n 1 oder 2 beträgt.
Bedeutet R, einen Alkylrest, der durch eine Amino- gruppe substituiert ist, so kann es sich dabei um eine pri märe, sekundäre oder tertiäre Aminogruppe oder auch um eine Aminogruppe handeln, deren Stickstoffatom Bestandteil eines 5- oder 6gliedrigen Restes ist, wie z.B. eine Pyrrolidino-, Piperidino-, Morpholino- oder N-Me- thylpiperazinogruppe. R, kann also beispielsweise für eine Aminoäthylgruppe, eine Methylaminoäthylgruppe, eine Diäthylaminoäthylgruppe oder z.B. für eine Morpholino- methylgruppe stehen.
Bevorzugt werden solche Verbin dungen der allgemeinen Formel I hergestellt, in denen die Summe von m und n 2 ist und in denen R, für Wasser stoff oder einen unsubstituierten oder durch nichtreaktive Gruppen substituierten Kohlenwasserstoffrest aus den oben genannten Gruppen steht.
Die Verbindungen der allgemeinen Formel I können ein asymmetrisch substituiertes Kohlenstoffatom enthal ten und dementsprechend in isomeren Formen auftreten. Zum Gegenstand der Erfindung gehört sowohl die Her stellung von racemischen Verbindungen der Formel I bzw. deren Isomerengemischen als auch die Herstellung der reinen Isomeren, wie z.B. der optisch aktiven Formen der Verbindungen der Formel I.
Die erfindungsgemäss erhältlichen neuen Phthalimi- dinderivate haben eine Anzahl äusserst wertvoller phar mazeutisch verwertbarer Wirkungen. Insbesondere zeigen sie ausgeprägte neuroendokrine Effekte und sind unter anderem in der Lage, das Tumorwachstum günstig zu beeinflussen, wie beispielsweise mit folgender Versuchs anordnung gezeigt werden kann.
Gibt man weiblichen Sprague-Dawley (SD)-Ratten im Alter von 50 - 65 Tagen und im Gewicht von durchschnitt lich 160 g eine einmalige Dosis von 20 mg Dimethylbenz- anthracen in 1 ml Sesamöl mit der Schlundsonde, so ent wickeln sich vorwiegend im Bereich der Milchleiste Tu moren, deren Anzahl man auszählen und deren Grösse man durch Messung feststellen kann. Behandelt man Tie re, welchen auf die beschriebene Art Mammatumoren in duziert wurden, mit den Verbindungen der allgemeinen Formel I, so verringern sich Tumoranzahl und Tumor grösse im Vergleich zu den unbehandelten Kontrolltieren.
Bezeichnet man das Produkt aus dem grössten und dem kleinsten Durchmesser des percutan tastbaren Tumors als Tumorfläche in mm2, so steigt z.B. bei einem Ausgangs wert von 509,6 mm Tumorfläche als Mittel von 10 Tieren die Tumorfläche bei unbehandelten Tieren in 6 Wochen auf einen Wert von 1 180,5 mm-' an und die Tumoranzahl wächst von 2,3 auf 4,5 ebenfalls als Mittelwert von 10 Tie- ren.
Gibt man jedoch den Tieren für die Dauer von 6 Wochen ein Futter, das 0,47 /0 3-Phthalimidino-piperidin- dion-2,6 bzw. 0,47% 4-Phthalimidino-piperidindion-2,6 enthält, so verändert sich in dieser Zeit die Tumorfläche von 473,5 mm= auf 490,2 mm= bzw. von 555,5 mm= auf 364,2 mm= und die Tumoranzahl geht von 2,0 auf 2,2 bzw. von 2,8 auf 2,5, ebenfalls als Mittelwert von 10 Tie ren.
Durch ihre immunosuppressive Wirkung begünstigen die Verbindungen der allgemeinen Formel I das Anwach sen von Transplanten und haben einen vorteilhaften Einfluss auf den Verlauf von zum Beispiel Lepra-Erkran kungen, Strahlenschäden und Autoimmunkrankheiten.
Bei vielen der Verbindungen der allgemeinen Formel I konnte weiterhin eine starke sedierende Wirkung festge stellt werden.
Zur Herstellung der Verbindungen der allgemeinen Formel I geht man von einem Mono- oder Diamid eines Amins der allgemeinen Formel H2N-R1 II worin R1 die vorgenannte Bedeutung hat, mit einer Säure der allgemeinen Formel

Der Formaldehyd wird vorzugsweise in Form von wässrigen oder alkoholischen Lösungen verwendet. Ver bindungen, die Formaldehyd unter den Reaktionsbedin gungen zu bilden vermögen, sind beispielsweise Paraform- aldehyd oder Chlormethylmethyläther. Die Umsetzung wird zweckmässig in Gegenwart von Lösungs- oder Ver dünnungsmitteln durchgeführt. Vorzugsweise werden or ganische Lösungsmittel, wie Methanol, Äthanol oder Äthylenglykolmonoäthyläther, oder Gemische aus Wasser und organischen Lösungsmitteln verwendet. Die Reaktion wird zweckmässig bei erhöhter Temperatur, z.B. zwischen 50 und 80 , durchgeführt, sie kann jedoch auch bei Raumtemperatur oder unter Kühlung durchgeführt wer den.
Man kann stöchiometrische Mengen der Verbindun gen der allgemeinen Formeln IV und V sowie Formalde hyd verwenden, man kann jedoch auch eine oder zwei der drei Komponenten im Überschuss einsetzen. Die Iso lierung der entstandenen Verbindungen der allgemeinen Formel I kann erfolgen, indem man-gegebenenfalls nach Einengen des Reaktionsgemisches - durch Kühlen die Reaktionsprodukte auskristallisieren lässt oder das Reak tionsgemisch mit einem flüssigen Mittel, in dem die Ver fahrensprodukte nicht oder nur wenig löslich sind, ver setzt. Beispielsweise kann man zur Isolierung der Produk te aus einem Alkohol-Wasser-Gemisch Alkohol zufügen.
Man kann auch das verwendete Lösungs- oder Verdün nungsmittel durch Destillation entfernen u. den Rückstand aus geeigneten Lösungsmitteln oder Lösungsmittelgemi schen umkristallisieren.
Die Verbindungen der Formel II können erhalten werden, indem man beispielsweise in einer Säure der all gemeinen Formel

worin m und n die vorgenannte Bedeutung haben, eine der Carbonylgruppen des Phthalimidrestes zu einer CH2-Gruppe reduziert. Man kann auch z.B. von einem Ester einer Säure der Formel VIII ausgehen. Wird in einem solchen Fall die Reduktion mit Hilfe der Clemmen- sen-Methode durchgeführt, so kann unter den Reduk tionsbedingungen gleichzeitig eine Verseifung des Esters erfolgen, so dass man als Produkt der Reduktion die Säu re der Formel 1I erhält.
Zur Reduktion kann man nicht nur das Verfahren nach Clemmensen verwenden, sondern auch elektrolytisch oder unter Verwendung von z.B. Kobalt- oder Nickelka talysatoren mit elementarem Wasserstoff reduzieren. Die x-Phthalimidinoglutarsäure ist beispielsweise folgender- massen erhältlich: 133 g α-Phthalimido-glutarsäure werden unter Erwär men in 1,2 1 Essigsäure gelöst.
Bei einer Temperatur von 60 C setzt man potrionsweise 168g Zinkstaub hinzu und erhitzt das Reaktionsgemisch 4 Stunden auf Rückfluss- temperatur. Man filtriert in der Wärme, wäscht den Rück stand mit Essigsäure und erhält aus den vereinigten Fil traten beim Abkühlen die α-Phthalimidino-glutarsäure in einer Ausbeute von 70% der Theorie in Form weisser Kristalle vom Schmelzpunkt 236-237 C, nach Umkristalli sation aus Wasser.
Analog erhält man unter Verwendung von ss-Phthal- imidoglutarsäure die ss-Phthalimidino-glutarsäure vom Schmelzpunkt 280 - 281 C in einer Ausbeute von 910/a der Theorie.
Aus Phthalimido-bernsteinsäure als Ausgangsprodukt entsteht unter den genannten Bedingungen die Phthalimi- dino-bernsteinsäure vom Schmelzpunkt 229 - 235 C un ter Zersetzung.
In den folgenden Beispielen sind die Temperaturan gaben unkorrigiert.
<I>Beispiel 1</I> 108 g α-Phthalimidino-glutarsäure werden mit einer Mischung von 833 ml Essigsäureanhydrid und 29,7 ml Thionylchlorid bis zur Beendigung der Salzsäureentwick lung auf Rückflusstemperatur erhitzt. Beim Abkühlen er hält man das α-Phthalimidino-glutarsäureanhydrid in einer Ausbeute von 90% der Theorie, Schmelzpunkt 254 - 257 C unter Zersetzung.
187 g α-Phthalimidino-glutarsäureanhydrid werden in 1870 ml absolutem Dioxan suspendiert. Man leitet zu nächst bei Raumtemperatur, später bei 80 C, trockenes Ammoniakgas ein. Der Niederschlag wird abgetrennt, mit Äther gewaschen und im Vakuum bei 60 C getrocknet. 208 g des so erhaltenen Ammoniumsalzes werden in 420 ml Wasser gelöst. Zu der gegebenenfalls nach Filtration klaren Lösung lässt man unter Rühren 63 ml konzentrier ter Salzsäure zutropfen. Nach einiger Zeit scheiden sich 192 g des Monoamides der α-Phthalimidino-glutarsäure ab. Die Substanz schmilzt nach Umkristallisation- aus Äthanol bei 191 - 192 C.
132 g der vorstehenden Verbindung werden mit einer Mischung von 518 ml Essigsäureanhydrid und 57,8 ml Thionylchlorid langsam auf 90 - 95 C erwärmt und bis zum Abklingen der Salzsäureentwicklung bei dieser Tem peratur gehalten. Durch Abkühlen, gegebenenfalls nach Einengen, erhält man das 3-Phthalimidino-piperidindion- -2,6 in einer Ausbeute von 76% der Theorie.
<I>Beispiel 2</I> Analog Beispiel 1 erhält man aus α-Phthalimidino- glutarsäureanhydrid und Methylamingas das 1-Methyl-3- -phthalimidino-piperidindion-2,6 vom Schmelzpunkt 139 141 C nach Umkristallisation aus Äthanol.
<I>Beispiel 3</I> 40 g α-Phthalimidino-glutarsäureanhydrid werden in 400 ml absolutem Dioxan suspendiert. Unter Rühren lässt man bei Raumtemperatur 18,5g Äthylamin zutropfen und erwärmt das Reaktionsgemisch anschliessend 2 Stun den auf 80 - 85 C. Man entfernt das Lösungsmittel durch Destillation im Vakuum und löst den Rückstand in 80 ml Wasser. Durch Zugabe von 13,5 ml konzentrierter Salz säure erhält man das α-Phthalimidino-glutarsäure-N- -äthyl-amid in Form weisser Kristalle vom Schmelzpunkt 141 - 144 C nach Umkristallisation aus Aceton. 23,5 g der vorstehenden Verbindung werden mit einer Mischung von 97 ml Essigsäureanhydrid und 9,8 ml Thionylchlorid versetzt und bis zum Nachlassen der Salz säureentwicklung auf 90 - 95 C erwärmt.
Nach Abkühlen, gegebenenfalls nach Einengen, er hält man durch Zugabe von 200 ml Äther das 1-Äthyl-3- -phthalimidinopiperidindion-2,6 in Form weisser Kri stalle vom Schmelzpunkt 177 - 180 C nach Umkristalli sation aus Äthanol.
<I>Beispiel 4</I> Analog Beispiel 3 erhält man durch Variation der Aminkomponente, gegebenenfalls ohne Isolierung der Zwischenprodukte, folgende Verbindungen:

<tb> Äthanol. <I>Beispiel 5</I> 302 g ss-Phthalimidino-glutarsäure werden mit einer Mischung von 1725 ml Essigsäureanhydrid und 84,5 ml Thionylchlorid bis zur Beendigung der Gasentwicklung auf Rückflusstemperatur erhitzt. Beim Abkühlen erhält man das p-Phthalimidino-glutarsäureanhydrid in einer Ausbeute von 86,5% der Theorie, Schmelzpunkt 230 - 235 C. 254 g der vorstehenden Verbindung werden in 2,5 1 absolutem Dioxan suspendiert.
Man leitet, zunächst bei Raumtemperatur, später bei 80-85 C, so lange trockenes Ammoniakgas ein, bis sich eine Probe des Niederschla ges in Wasser klar löst. Der Niederschlag wird abgetrennt und in 582 ml Wasser gelöst. Zu der gegebenenfalls nach Filtration klaren Lösung lässt man unter Rühren 87 ml konzentrierter Salzsäure zutropfen. Beim Abkühlen schei den sich 270 g des Monoamides der ss-Phthalimidino-glu- tarsäure ab. Schmelzpunkt 228 - 230 C.
270 g der vorstehenden Verbindung werden mit einer Mischung von 1,140 ml Essigsäureanhydrid und 118 ml Thionylchlorid langsam auf 90 - 95 C erwärmt und bis zum Abklingen der Salzsäureentwicklung bei dieser Tem peratur gehalten. Beim Abkühlen erhält man das 4- -Phthalimidino-piperidindion-2,6 in einer Ausbeute von 74% der Theorie, Schmelzpunkt 244 - 249 C nach Um kristallisation aus Essigsäure.
<I>Beispiel 6</I> 30 g p-Phthalimidino-glutarsäureanhydrid werden in 300 ml absolutem Dioxan suspendiert. Unter Rühren tropft man 14,5 g Äthylamin zu und erwärmt auf 80 C. Nach Abkühlen wird der Niederschlag abgesaugt, mit Äther gewaschen und in 150 ml Wasser gelöst. Zu der filtrierten Lösung gibt man 10 ml konz. Salzsäure. Man lässt stehen, saugt dann das ausgefallene ss-Phthalimidino- glutarsäure-N-äthylamid ab und wäscht mit Wasser. Ausbeute 31 g, Schmelzpunkt 215 C - 217 C.
Die ge- trocknete Amidsäure wird nun 7 Stunden mit einer Mi schung aus 130 ml Essigsäureanhydrid und 130 ml Acetyl- chlorid unter Rückfluss gekocht. Durch Abkühlen, gege benenfalls nach Einengen und Versetzen mit Äther er hält man das 1-Äthyl-4-phthalimidino-piperidindion-2,6 in einer Ausbeute von 24 g, Schmelzpunkt 191 - 195 C nach Umkristallisation aus n-Butanol.
<I>Beispiel 7</I> Analog Beispiel 6 erhält man durch Variation der Aminokomponente folgende Verbindungen:

Man lässt 150 ml 30%iger wässriger Formaldehydlösung zutropfen und ver setzt anschliessend mit 60 ml Pyrrolidin. Man erhitzt eini ge Zeit zum Rückfluss.
Durch Stehen unter Kühlung er hält man das 1-Pyrrolidinomethyl-3-phthalimidino-piperi- dindion-2,6 in einer Ausbeute von 48% der Theorie. Nach Umkristallisation aus Dioxan/Äther schmilzt das Pro dukt bei 164 - 165 C.
<I>Beispiel 9</I> 12 g 4-Phthalimidino-piperidindion-2,6 werden in 150 ml Äthanol aufgeschlämmt und unter Erwärmen mit 25 ml 35%iger wässriger Formaldehydlösung versetzt. Man erhitzt bis zur klaren Lösung und erhält nach Ab kühlen das 1-Methylol-4-phthalimidino-piperidindion-2,6 in weissen Kristallen vom Schmelzpunkt 152 - 154 C unter Zersetzung.
5 g 1-Methylol-4-phthalimidino-piperidindion-2,6 wer den unter Erwärmen in 50 ml Äthanol gelöst und mit 2 g Piperidin versetzt. Man erwärmt 10 Minuten und erhält nach Abkühlen das 1-Piperidinomethyl-4-phthalimidino- piperidindion vom Schmelzpunkt 162 - 165 C nach Um kristallisation aus Dioxan/Äther.
<I>Beispiel 10</I> Man verfährt wie in Beispiel 8 und erhält aus 4- -Phthalimidino-piperidindion-2,6, Formaldehydlösung u. Pyrrolidin das 1-Pyrrolidinomethyl-4-phthalimidino-pi- peridindion-2,6 in einer Ausbeute von 53% der Theorie. Schmelzpunkt 154 bis 159 C nach Umkristallisation aus Dioxan/Äther.

where R1 stands for a hydrogen atom, optionally with free, esterified or etherified oxy or mercapto groups, alkyl or alkenyl radicals substituted by amino groups, carboxyl groups or halogen atoms and having 1 to 6 carbon atoms or for an aralkyl, cycloalkyl or aryl radical and m and n are the numbers 0, 1 or 2, the sum of m and n being 1 or 2.
If R is an alkyl radical which is substituted by an amino group, it can be a primary, secondary or tertiary amino group or an amino group whose nitrogen atom is part of a 5- or 6-membered radical, e.g. a pyrrolidino, piperidino, morpholino or N-methylpiperazino group. R, can for example represent an aminoethyl group, a methylaminoethyl group, a diethylaminoethyl group or e.g. stand for a morpholino methyl group.
Those compounds of the general formula I are preferably prepared in which the sum of m and n is 2 and in which R 1 is hydrogen or an unsubstituted or substituted by nonreactive hydrocarbon radical from the groups mentioned above.
The compounds of the general formula I can contain an asymmetrically substituted carbon atom and accordingly occur in isomeric forms. The subject of the invention includes both the preparation of racemic compounds of the formula I or their isomer mixtures and the preparation of the pure isomers, such as e.g. the optically active forms of the compounds of formula I.
The novel phthalimidine derivatives obtainable according to the invention have a number of extremely valuable pharmaceutically utilizable effects. In particular, they show pronounced neuroendocrine effects and are able, among other things, to favorably influence tumor growth, as can be shown, for example, with the following experimental set-up.
