Die Erfindung betrifft ein Verfahren zur Herstellung von 19-Alkylsteroiden der Formel

worin R1 Wasserstoff oder Cl 6-Alkyl, R2 Hydroxy, Acyloxy oder Cl 6-Alkoxy und R3 Wasserstoff C1-Alkyl, C2 6-Alkenyl oder C2-6-Alkinyl darstellen, bzw. von deren hs-Dehydroderivaten.
Eine bevorzugte Gruppe von erfindungsgemäss erhältlichen Verbindungen der Formel I und deren A6-Dehydro- derivaten ist diejenige, in der R9 einen Alkenyl- oder Alkinylrest darstellt.
Das erfindungsgemässe Verfahren ist dadurch gekennzeichnet, dass man ein Steroid der Formel

worin R', R3 und R3 die oben genannten Bedeutungen haben, oder ein A6-Dehydroderivat davon reduziert.
Die vorstehend verwendeten Bezeichnungen Cl 6-AI- kyl, C2 6-Alkenyl und C6-Alkinyl beziehen sich auf solche Kohlenwasserstoffreste, die 1-6 bzw. 2-6 C-Atome enthalten und die geradkettig oder verzweigt sein können.
Beispiele für Cl -Alkyl sind Methyl, Äthyl, Propyl, Isopropyl, Butyl, Isobutyl, tert.-Butyl, Pentyl und Hexyl. Beispiele für C2 ;;-Alkenyl sind Vinyl, Allyl und Methallyl.
Beispiele für C2 6-Alkinylreste sind Äthinyl, Propinyl und Propargyl.
Eine durch das Symbol R dargestellte Acyloxygruppe enthalt vorzugsweise den Rest einer gesättigten oder ungesättigten, aliphatischen oder aromatischen Carbonsäure mit 1-10 Kohlenstoffatomen. Beispiele solcher Säuren sind Essigsäure, Propionsäure, Capronsäure, Pivalinsäure, Buttersäure, Isobuttersäure und Benzoesäure. Beispiele für eine C1-6-Alkoxygruppe sind Methoxy, Äthoxy und tert.Butoxy.
Die erfindungsgemässe Reduktion einer 3-Ketogruppe zu einer 3-Hydroxygruppe kann mit Reduktionsmitteln wie komplexen Metallhydriden, z.B. Lithiumaluminiumhydrid oder Natriumborhydrid in bekannter Weise durchgeführt werden.
Die 19-Alkylsteroide der Formel I sind hormonal wirksam. Sie beeinflussen den Gonadotropinhaushalt des Körpers und hemmen die Wirkung von Progesteron. Sie können als Mittel zur Fertilitätskontrolle Verwendung finden, z.B. in Form phannazeutischer Präparate, welche sie in Mischung mit einem für die enterale oder parenterale Applikation geeigneten pharmazeutischen, organischen oder anorganischen inerten Trägermaterial enthalten. Die pharmazeutischen Präparate können in fester Form oder in flüssiger Form vorliegen. Gegebenenfalls enthalten sie Hilfsstoffe, wie Konservierungs-, Stabilisierungs-, Netzoder Emulgiermittel, Salze zur Veränderung des osmotischen Druckes oder Puffers. Sie können auch noch andere therapeutisch wertvolle Stoffe enthalten.
Beispiel 1
1,3 g 17,5Ç-Äthinyl-17,8-hydroxy-l9-methyl-androsta- -4,6-dien-3-on werden in 40 ml Methanol gelöst und bei 0 unter Argon mit einer Lösung von 840 mg Natriumborhydrid in 4 ml Wasser versetzt. Man rührt während 30 Minuten bei 00, giesst dann auf Eis/Essigsäure, extra hiert mit Methylenchlorid, wäscht die Extrakte mit Natriumbicarbonatlösung und Wasser, trocknet über Pottasche/Natnumsulfat u. dampft das Lösungsmittel ab.
Der Rückstand wird in Aceton gelöst, filtriert und aus AcetonfHexan umkristallisiert. Man erhält 930 mg 17α- -Äthinyl-33,17ss-dihydroxy-19-methyl androsta -4,6- dien.
Schmelzpunkt 148 bis 1490; UV: 23 = 21600 (s) #240 = 23950, #### = 15400 (5); [α]### = 1360 (c - 0,1 in Dioxan).
Beispiel 2
In Analogie zu Beispiel 1 erhält man aus 19-Äthyl- - 17α-äthinyl-17ss -hydroxy - androsta - 4,6-dien - 3- on das 19-Äthyl-17α- äthinyl - 3 - dihydroxy - androsta - 4,6 - dien.
Schmelzpunkt 207 bis 2100. UV: #233 = 21000 (s), ± = 23800, ein = 15100 (s); [α]### = = 1180 (c = 0,1 in Dioxan).
Beispiel 3
5 g 19-Äthyl- 17α-äthinyl-17ss-hydroxy-androst-4-en-3- -on werden mit 3,2 g Natriumborhydrid in Methanol/ Wasser in Analogie zu Beispiel 1 umgesetzt. Man erhält 6 g Gemisch von 3α- und 3,17ss-Dihydroxy-19-äthyl-17α- -äthyl-androst-4-en. Nach Zugabe von Impfkristallen und mehrfacher Kristallisation aus Aceton/Hexan kann das reine 3ss-Isomere abgetrennt werden. Schmelzpunkt 160 bis 163 ; [α]### = 26 (c = 0,1 in Dioxan). Mehrfache Chromatographie der Mutterlaugen an Silicagel mit Hexan/Aceton 9 : 1 liefert schliesslich das nicht kristal line 3-z-Isomere.

A preferred group of compounds of the formula I obtainable according to the invention and their A6 dehydro derivatives is that in which R9 represents an alkenyl or alkynyl radical.
The method according to the invention is characterized in that a steroid of the formula

The terms Cl 6 -alkyl, C2 6-alkenyl and C6-alkynyl used above refer to those hydrocarbon radicals which contain 1-6 or 2-6 C atoms and which can be straight-chain or branched.
Examples of Cl -alkyl are methyl, ethyl, propyl, isopropyl, butyl, isobutyl, tert-butyl, pentyl and hexyl. Examples of C2 ;; - alkenyl are vinyl, allyl and methallyl.
Examples of C2-6-alkynyl radicals are ethynyl, propynyl and propargyl.
An acyloxy group represented by the symbol R preferably contains the residue of a saturated or unsaturated, aliphatic or aromatic carboxylic acid having 1-10 carbon atoms. Examples of such acids are acetic acid, propionic acid, caproic acid, pivalic acid, butyric acid, isobutyric acid and benzoic acid. Examples of a C1-6-alkoxy group are methoxy, ethoxy and tert-butoxy.
The inventive reduction of a 3-keto group to a 3-hydroxy group can be carried out with reducing agents such as complex metal hydrides, e.g. Lithium aluminum hydride or sodium borohydride can be carried out in a known manner.
The 19-alkyl steroids of the formula I are hormonally active. They influence the body's gonadotropin balance and inhibit the effects of progesterone. In analogy to Example 1, 19-ethyl-17α-ethinyl-17ss-hydroxy-androsta-4,6-diene-3-one is obtained from 19-ethyl-17α-ethinyl-3-dihydroxy-androsta-4 , 6 - dien.
Melting point 207 to 2100. UV: # 233 = 21000 (s), ± = 23800, a = 15100 (s); [α] ### = = 1180 (c = 0.1 in dioxane).
Example 3
5 g of 19-ethyl-17α-ethinyl-17ss-hydroxy-androst-4-en-3-one are reacted with 3.2 g of sodium borohydride in methanol / water in analogy to Example 1. 6 g of a mixture of 3α- and 3,17ss-dihydroxy-19-ethyl-17α-ethyl-androst-4-ene are obtained. After adding seed crystals and repeated crystallization from acetone / hexane, the pure 3ss isomer can be separated off. Melting point 160 to 163; [α] ### = 26 (c = 0.1 in dioxane). Multiple chromatography of the mother liquors on silica gel with hexane / acetone 9: 1 finally yields the non-crystalline 3-z isomer.
