Diese Erfindung betrifft ein Verfahren zur Herstellung neuer halogenierter Steroide der Formel

worin R1 Fluor, Chlor oder Brom oder Alkyl, R9 Wasserstoff oder Fluor; R3 eine Alkanoyloxygruppe; R4 Wasserstoff oder eine Alkylgruppe; R5 eine Alkanoylgruppe; R6 Wasserstoff, eine Alkylgruppe oder eine Äthinylgruppe; R7 Wasserstoff oder eine Methylgruppe und Y eine Methylen- oder Carbonylgruppe darstellen, wobei die Alkyl und Alkanoyl- bzw. Alkanoyloxygruppen bis zu 8 C-Atomen enthalten, oder entsprechender 1 ,2-Dehydroderivate.
Der Ausdruck niedere Alkylgruppe bezieht sich auf geradkettige und verzweigte Kohlenwasserstoffreste mit bis zu 8 Kohlenstoffatome, wie Methyl, Äthyl, tert. Butyl oder n-Octyl. Eine niedere Alkanoylgruppe ist beispielsweise eine Acetyl-, Butyryl- oder Capronylgruppe.
Das erfindungsgemässe Verfahren ist dadurch gekennzeichnet, dass man eine entsprechende Verbindung, die eine Hydroxygruppe in 17a- bzw. 17ss-Stellung anstelle einer Alkanoyloxygruppe enthält, mit einem entsprechenden Veresterungsmittel behandelt.
Die Ausgangsstoffe für das erfindungsgemässe Verfahren können z. B. wie folgt hergestellt werden (vgl.
auch die belgische Patentschrift Nr. 694 288):
Zu einer auf -80 gekühlten Lösung von 87,0 g 6-Chlor-17a-hydroxypregna-4,6-dien-3,20-dion in einem Gemisch von 870 ml über Calciumhydrid getrocknetem Dimethylformamid und 435 ml Äther gibt man 356 ml einer 0,95m-Lösung von Chlor in Propionsäure. Man lässt zuerst 14 Stunden bei 00, hierauf 6 Stunden bei 250 stehen. Das Reaktionsgemisch wird in 3 Liter Wasser gegossen, hierauf dreimal mit ither-Methylen- chlorid (2,5:1) extrahiert. Man wäscht die organischen Phasen zweimal mit 1,5 Liter 5 %Der Natriumbicarbonatlösung und einmal mit 2 Liter Kochsalzlösung.
Die vereinigten organischen Schichten werden über Natriumsulfat getrocknet und unter vermindertem Druck eingedampft. Das zurückbleibende Öl löst man in Benzol/Hexan (2:1) und chromatographiert an 1100 g synthetischem Magnesia-Silicagel ( Florisil ).
Man eluiert nacheinander mit 2 Liter Benzol/Hexan (Rückstand 0 mg), 1 Liter Benzol (Rückstand 18 mg), 1 Liter Benzol mit 2% Athylacetat (32 mg), 1 Liter Benzol mit 5 % Athylacetat (5,92 g, nicht kristallin), 500 ml Benzol mit 10 % Äthylacetat (19,0 g), 1,5 Liter Benzol mit 20% Äthylacetat (54,1 g) und 1 Liter Benzol mit 40 % Äthylacetat (16 g) ergeben Fraktionen, die nach Verreiben mit Äther kristallisieren.
Die vereinigten Feststoffe werden dreimal aus Methylenchloridliither umkristallisiert, und liefern 4,6-Dichlor-17a-hydroxy-pregna-4,6-dien-
3,20-dion vom Schmelzpunkt 159,5-161 .
Die erfindungsgemäss erhältlichen Verbindungen sind bei einer durch minimale Nebenwirkungen charakterisierten Wirkungsselektivität in hohem Grade endokrin, z. B. progestativ wirksam. Die Verfahrensprodukte können als Heilmittel in Form pharmazeutischer Präparate Verwendung finden, welche sie in Mischung mit einem für die enterale, perkutane oder parenterale Applikation geeigneten pharmazeutischen, organischen oder anorganischen inerten Trägermaterial enthalten.
Die pharmazeutischen Präparate können in fester Form, in halbfester Form oder in flüssiger Form vorliegen.
Gegebenenfalls enthalten sie Hilfsstoffe, wie Konservierungs-, Stabilisierungs-, Netz- oder Emulgiermittel, Salze zur Veränderung des osmotischen Druckes oder Puffer. Sie können auch noch andere therapeutisch wertvolle Stoffe enthalten.
Beispiel 1
Zu einer Suspension von 1,0 g 4,6-Dichlor-17ahydroxy-pregna-4,6-dien-3,20-dion in 15 ml Capronsäure gibt man unter Rühren 4,0 ml Trifluoressigsäureanhydrid und rührt 16 Stunden bei Zimmertemperatur.
Man giesst nun das Reaktionsgemisch in 100 ml einer kalten 5 %igen Natriumbicarbonatlösung und extrahiert das entstandene Gemisch dreimal mit Ather-Methylen- chlorid (2,5:1). Die organischen Schichten werden zweimal mit 0,5n Natronlauge und einmal mit Kochsalzlösung gewaschen, vereinigt, über Natriumsulfat getrocknet und abgedampft.
Der Rückstand wird in Benzol/Hexan (2:1) gelöst und an 15 g synthetischem Magnesia-Silicagel ( Florisil ) chromatographiert. Man eluiert mit Benzol, 30 % Äthylacetat in Benzol und schliesslich mit Äthylacetat.
Die Eluate werden eingedampft, der Rückstand mit Äther verrieben. Umkristallisation aus Methylenchlorid/ Äther gibt 4,6-Dichlor-l 7cr-hydroxy-pregna-4,6-dien- 3,20-dion-capronat vom Schmelzpunkt 118,5-1200; [a]r, + 115,560 (C = 0,98 in CHC13).
Beispiel 2
Man gibt zu einer Suspension von 27,0 g 4,6-Di chlor-17a-hydroxy-pregna-4,6-dien-3 ,20dion in 1350 ml Eisessig und 270 ml Essigsäureanhydrid 27,0 g p-Toluolsulfonsäure, rührt das Gemisch, bis sich der Feststoff völlig gelöst hat, und lässt 6 Stunden bei Zimrnertemperatur stehen. Das Reaktionsgemisch wird in 7 Liter Wasser gegeben, der Niederschlag wird nach 30 Minuten filtriert und gut mit Wasser gewaschen. Das feuchte Produkt wird in ungefähr 1 Liter Methylenchlorid gelöst, über Natriumsulfat getrocknet und die Lösung unter vermindertem Druck auf etwa 200 ml konzentriert. Die heisse Lösung wird unter gleichzeitigem Zusatz von 150 ml Äthylacetat weiter konzentriert, wobei das 4,6- Dichlor - 17a - acetoxy - pregna-4,6-dien- 3,20-dion ausfällt.
Schmelzpunkt nach Umkristallisation aus dem gleichen Lösungsmittel 241,5-243 .

The term lower alkyl group refers to straight-chain and branched hydrocarbon radicals with up to 8 carbon atoms, such as methyl, ethyl, tert. Butyl or n-octyl. A lower alkanoyl group is, for example, an acetyl, butyryl or capronyl group.
The process according to the invention is characterized in that a corresponding compound which contains a hydroxyl group in the 17a or 17ss position instead of an alkanoyloxy group is treated with an appropriate esterifying agent.
The starting materials for the inventive method can, for. B. be produced as follows (cf.

It is eluted successively with 2 liters of benzene / hexane (residue 0 mg), 1 liter of benzene (residue 18 mg), 1 liter of benzene with 2% ethyl acetate (32 mg), 1 liter of benzene with 5% ethyl acetate (5.92 g, not crystalline), 500 ml of benzene with 10% ethyl acetate (19.0 g), 1.5 liters of benzene with 20% ethyl acetate (54.1 g) and 1 liter of benzene with 40% ethyl acetate (16 g) give fractions which, after trituration crystallize with ether.
The combined solids are recrystallized three times from methylene chloride, and give 4,6-dichloro-17a-hydroxy-pregna-4,6-diene-
3,20-dione, melting point 159.5-161.
The compounds obtainable according to the invention are highly endocrine, with a selectivity of action characterized by minimal side effects. B. effective progestationally. The products of the process can be used as medicaments in the form of pharmaceutical preparations which they contain as a mixture with a pharmaceutical, organic or inorganic inert carrier material suitable for enteral, percutaneous or parenteral administration.

To a suspension of 1.0 g of 4,6-dichloro-17ahydroxy-pregna-4,6-diene-3,20-dione in 15 ml of caproic acid, 4.0 ml of trifluoroacetic anhydride are added while stirring and the mixture is stirred for 16 hours at room temperature.
The reaction mixture is then poured into 100 ml of a cold 5% strength sodium bicarbonate solution and the resulting mixture is extracted three times with ether-methylene chloride (2.5: 1). The organic layers are washed twice with 0.5N sodium hydroxide solution and once with brine, combined, dried over sodium sulfate and evaporated.
The residue is dissolved in benzene / hexane (2: 1) and chromatographed on 15 g of synthetic magnesia silica gel (Florisil). It is eluted with benzene, 30% ethyl acetate in benzene and finally with ethyl acetate.
The eluates are evaporated and the residue is triturated with ether. Recrystallization from methylene chloride / ether gives 4,6-dichloro-l-7cr-hydroxy-pregna-4,6-diene-3,20-dione-capronate with a melting point of 118.5-1200; [a] r, + 115.560 (C = 0.98 in CHCl3).
Example 2
27.0 g of p-toluenesulfonic acid are added to a suspension of 27.0 g of 4,6-dichloro-17a-hydroxy-pregna-4,6-diene-3, 20dione in 1350 ml of glacial acetic acid and 270 ml of acetic anhydride, and the mixture is stirred Mix until the solid has completely dissolved and leave to stand for 6 hours at room temperature. The reaction mixture is poured into 7 liters of water, the precipitate is filtered after 30 minutes and washed well with water.
