Mittel zum qualitativen Nachweis bzw. zur quantitativen Bestimmung von Hydroperoxyden bzw.
peroxydatisch wirksamen Substanzen und seine Verwendung Gegenstand der vorliegenden Erfindung ist ein Mit tel zum qualitativen Nachweis bzw. zur quantitativen Bestimmung von a) Hydroperoxyden oder von Substan aen, die unter Freisetzung von Hydroperoxyden reagie ren oder b) Peroxydase oder peroxydatisch wirkenden
Substanzen unter Ausnützung der Farbreaktion, welche a) und b) gegebenenfalls unter Einwirkung anderer Sub stanzen zusammen mit einem Chromogen eingehen. Von besonderem Interesse ist der Nachweis von Glucose in
Harn, Blut und Serum bei Diabetes, sowie der Nach weis peroxydatisch wirkender Substanzen, wie Hämo globin im Harn und Blut und der Naohweis von Hydro peroxiden z.
B. in der Milchwirtschaft, Kosmetik und Polymerchemie.
Es ist eine Reihe von Verbindungen bekannt, die von Wasserstoffperoxyd mit Peroxydase als Katalysator zu einem Farbstoff oxydiert werden. Die Substanzen, die hierfür verwendet wenden, sind z. B.: Nadi-Reagenzien, p-Phenylendiaminderivate, Carboazine, Guajakharz, Aldazine etc.
Bevorzugt eingesetzt werden bisher Benzidin, o-Dia- nisidin und o-Toiidin. Diese Verbindungen sind aber teilweise nicht sehr stabil und können nach den neue sten Erkenntnissen gesundheitsschädlich sein, so dass ihre Handhabung nicht ungefährlich erscheint.

absolut beständig sind und sich besonders gut dazu eig nen, Wasserstoffperoxyd und andere Hydroperoxyde so wohl im optischen Tiefst hals auch mit Hilfe von Reagenz papieren und Reagenzfilmen zu bestimmen. Es ist zwar bekannt, dass sich einige dieser Substanzen mittels star ker Oxydationsmittel wie Bleitetraacetat, Cer-IV-Sulfat und Kaijumpersulfat oxydieren lassen und dabei gefärb te Oxydationsprodukte liefern; vgl. S. Hünig et al., Lie bigs Ann. Chem. 676, (1964), S. 3265 sowie H. Lang et al. Z. analyt. Chem. 201, (1964), S. 321.
Da jedoch mit Wasserstoffperoxyd alleine keine Reaktion erfolgt, war es nicht vonauszusehen, Idass sich in Anwesenheit von Peroxydase oder peroxydatisch wirksamen Substan zen ebenfalls reproduzierbare deutliche Verfärbungen erzielen lassen.
Das erfindungsgemässe Mittel, das a) ein Hydroper oxyd oder eine unter Freisetzung von Hydroperoxyd reagierende Substanz oder b) Peroxydase oder eine pe roxydatisch wirksame Substanz und ein Chromogen ent hält, ist dadurch gekennzeichnet, dass es als Chromo gen mindestens eine Verbindung der Formel I enthält.
Das erfindungsgemässe Mittel ist vorzugsweise für gekoppelte und ungekoppelte Enzymreaktionen geeignet, wie z. B. zur Bestimmung von Glucose, Galactose, Aminosäuren, Harnsäure, Peroxyden, Hämoglobin, Peroxydase oder anderen peroxydatisch wirksamen Substanzen sowie Enzymaktivitäten. Die routinemässige Bestimmung von derartigen Substraten ist wegen ihrer ele mentaren Wichtigkeit aus wider klinischen Chemie und aus der Lebensmittelchemie nicht mehr wegzudenken.
Bei der Bestimmung von Glucose beispielsweise wird diese von Glucoseoxydase zu Gluconsäure oxydiert, wobei der Luftsauerstoff zu Wasserstoffperoxyd reduziert wird. Das Wasserstoffperoxyd oxydiert dann mittels Peroxydase oder einer peroxydatisch wirkenden Substanz das erfindungsgemäss als Indikator eingesetzte Chromogen zum entsprechenden - Farbstoff. Weitere Beispiele für derartige analytisch brauchbare Enzymsysteme, die unter Freisetzung von Wasserstoffperoxyd reagieren, sind L-Aminosäureoxydase + L-Aminosäuren, D-Aminosäureoxydase + D-Aminosäuren, Uricase + Harnsäure, Xanthinoxydase + Hypoxanthin bzw.
Xanthin, Olycinoxydase + Glycin, Monoaminooxydasfe + Monoamin (wie Adrenalin, Mescalin etc.), Diaminooxydase + Diamine (wie Hisbamin), Luciferase + Luciferin, D-Asparaginsäureoxydase + D-Asparaginsäure, Leber-Aldehydoxydase + Aldehyd, Galactoseoxydase + galaktose, Edson's Flavinenzym + Milchsäure.
Der Nachweis bzw. die quantitative Bestimmung mit Hilfe der erfindungsgemässen Mittel erfolgt durch Auswertung der bei der Reaktion der Hydroperoxyde mit Peroxydase bzw. peroxydatisch wirksamen Substanzen auftretenden Verfärbungen, die auf den Übergang des Chromogens in den entsprechenden Farbstoff zurückzuführen sind. Die Auswertung erfolgt beispielsweise durch optische Ausmessung im Spektrophotometer oder bei Verwendung von Testpapierstreifen und Testfilmen durch Vergleich der Farbstärke mit Blindproben bekannter Zusammensetzung bzw. Farbtafeln.
Besonders gute Ergebnisse und vor allem besonders gut abgestufte Farbändlerungen erhält man überraschen derveise, wenn das diagnostische Mittel ausser den Verbindungen der Formel 1 noch ein Reduktionsmittel der Formel

In der Formel II bedeuten: Ar einen Benzol- oder Naphthalinkern, V eine Hydroxyl- oder Aminogruppe, V' Wasserstoff oder eine gegebenenfalls alkylierte oder arylierte Hydroxyl- oder Aminogruppe, W eine Amino-, Alkoxy-, Alkyl-, Aryl-, Carboxy oder Sulfogruppe und W' Wasserstoff oder eine Amino-, Alkoxy-, Alkyl-,
Aryl-, Carboxy- oder Sulfogruppe, sowie gegebenenfalls deren Alkalisalze.
Die Verbindungen der Formel II, die als Modifier wirken, geben für sich alleine keine oder nur sehr unspezifische Verfärbungen und sind des'alb als Indi- katoren im allgemeinen nicht geeignet. Zusammen mit den Verbindungen der Formel 1 erhält man aber besonders deutliche und gut abgestufte Farbumschläge.
Bevorzugte Ausführungsformen der vorliegenden Erfindung enthalten daher eine Kombination der Verbindungen der Formel 1 und II als Indikator.
Selbstverständlich lässt sich umgekehrt das erfindungsgemässe Mittel auch zur Bestimmung der Chromogene 1 sowie der Modifier II mit Hilfe von Hydroperoxyden und einer peroxydatisch wirksamen Substanz benutzen, was bei der Produktionsüberwachung dieser Diagnostica sehr nützlich ist.
Die erfindungsgemässen Mittel sowie ihre Verwendung werden anhand der folgenden Beispiele näher er ,läutern.
Beispiel 1 Reagenzfilm zum Nachweis von Glukose
45 g Propiofan (BASF), 35 g einer Lösung von 37 g Algipon in 2 Liter eines 0,5 molaren Phosphat- oder Citratpuffers vom pH 5,7, 1 g Texapon P (Fa. Henkel Düsseldorf), 10 ml Wasser, 0,075 g Peroxydase und 0,15 g Glucoseoxydase werden zu einem homogenen Brei verrührt. Zu dieser Mischung gibt man 6 ml einer 5 /0-igen wässrigen Lösung von 2,2'-Azino-di-[3-äthylbenzthia- zolon-sulfonsäure (6)]. Mit den so hergestellten Mischungen, in denen jeweils 0,2-0,3 io Indikator enthalten sind, werden Folien in einer Schichtdicke von 300 u beschichtet und getrocknet. Mit wässrigen Lösungen verschiedener Olukosekonzentrationen erhält man schwach abgestufte Grünfärbungen.
Setzt man den obigen Mischungen anstelle der wässrigen Lösungen von 2,2'-Azino-di-[3-äthyl-benzthiazolon-sulfonsäure (6)] 6 ml einer 5 /o-igen Lösung von p-Amino-salicylsäure zu, so erhält man Teststreifen, die sich beim Benetzen mit wässrigen Glukoselösungen nur schwach gelb verfärben und keinerlei Abstufungen der Verfärbung aufweisen. Verwendet Iman jedoch 6 ml einer wässrig-methanolischen Lösung des Gemischs von 5 /0 2,2'-Azino di-[3-äthyl-benzthiazolon-sulfonsäure (6)] und 5 /o p-Aminosalicylsäure, so erhält man Teststreifen, die sich je nach der Glukosekonzentration in Abstufungen von grün nach violett verfärben.
Beispiel 2 Nachweis von Glukose in Blut, Harn oder Serum
In analoger Weise wie im Beispiel 1 beschrieben, werden Reagenzfilme mit folgenden Substanzen 1 hergestellt: A = 2,2'-Azino-di-[3-äthyl-benzthiazolon sulfonsäure(6)] B = 2,2'-Azino-di-[1-äthyl-chinolin-sulfonsäure(6)] C = 2,2-Azino-di-[1,3-diäthyl-benzimidazolon- sulfonsäure(6)] D = 1-[3-Methylbenzthiazolon(2)]-2 [1 -äthylpyridon(2)] -azin E = 1-[3 -Äthylbenzthiazolon(2)] -2-[1-phenyl-3- methyl-4-äthyl-1,2,4-triazolon(5)]-azin F = 1-[3-Äthylbenzthiazolon(2)]-2 [1-äthylchinolinon(2)]-azin G = 1-[3-Methylbenzthiazolon(2)]-2-[1-phenyl-3-
methyl-4-äthyl-1,2,4-triazolon(5)]-azin H = 1 -[1 -Äthylchinolinon(2)]-2-[ 1-äthylpyridon(2)]- azin J = 1-[3-Äthylbenzthiazolon(2)]-2 [1-äthylpyridon(2)j-azin K = 1 -[3 -Methylbenzthiazolon(2)] -2- [1-äthylchinolinon(2)J-azin L = 1-[3-Methylbenzthiazolon(2)]-2-[1-äthyl-6- chlorpyridon(2)]-azin M = 1-[1-Athylchinolinon(2)]-2-[1-äthyl-6- chlorpyridon(2)] -azin N = 2,2'-Azino-di-(3-äthylbenzthiazolon) wobei diese mit einer Reihe von Substanzen II kombiniert werden. Die Farbumschläge mit glucosehaltigen Lösungen (z. B.
Blut, Harn oder Serum) werden nach kurzer Zeit abgelesen und sind in der nachfolgenden Tabelle 1 zusammengestellt:

Beispiel 3 Nachweis von Blut im Harn
Ein Filterpapier (2316 der Firma Schleicher & Schüll) wird mit einem 0,1 molaren Phosphatpuffer vom pH 7,0 in dem 1 o/o Texapon P (Fa. Henkel, Düsseldorf) gelöst ist, imprägniert und getrocknet. Danach wird das imprägnierte Papier mit einer wässrigen Lösung des Indikatorgemisches (0,03 O/o 2,2'-Azino-di-[1-äthylchino linsulfonsäure (6)] und 0,1 /o o-Aminophenol-p-sulfonsäure) nochmals imprägniert und getrocknet. Bringt man auf ein so hergestelltes Papier einen Tropfen eines bluthaltigen Urins und einen Tropfen 30/obiges Wasserstoffperoxyd, so färbt sich das Testpapier noch bei einer Verdünnung von 1:50 000 rotviolett.
Nimmt man statt o-Aminophenol-p-sulfonsäure, 1-Amino-8-naphthol-2,4- disulfonsäure, so färbt sich das Papier noch bei einer Verdünnung von 1: 50 000 blauviolett.
Beispiel 4 Nachweis von Hydrnpernxyden in Flüssigkeiten
Ein Filterpapier (2316 der Firma Schleicher & BR< Schüll) wird mit 50 ml eines 0,1 molaren Phosphatpuffers vom pH 6,0, in dem 25 mg Peroxydase und 0,25 ml Texapon P (Fa. Henkel, Düsseldorf) gelöst sind, ijmpräg- niert und getrocknet. Danach wird das Papier mit einer Lösung von 0,01 g 2,2'-Azino-di-[1-äthylchinolinsul- fonsäure (6)] und 0,1 g o-Aminophenol-p-sulfonsäure in 100 ml Wasser imprägniert und getrocknet. Bringt man auf ein so hergestelltes Testpapier einen Tropfen einer wässrigen Hydroperoxydlösung, so zeigt das Papier noch bei einer Konzentration von 0,2 y/ml eine rotviolette Färbung.
Beispiel 5 Nachweis von Glucose in Flüssigkeiten
Jeweils 0,01 ml von Glucoselösungen mit Konzentrationen von 0-300 mg0/ werden zu 2 ml einer wässrig-methanolischen 0,00125 molaren Indikatorlösung von äquimolaren Mengen 2,2'-Azino-di-[3 -äthylb enz- thi azolonsulfonsäure (6)] und p-Aminosalicylsäure pipettiert. Hierzu gibt man 2 ml einer Lösung von 10 mg Peroxydase und 20 mg Glucoseoxydase in 100 ml eines 0,5 molaren Phosphatpuffers vom pH 5,7, schüttelt um, misst bei einer Wellenlänge von 490 nm und berechnet mit Hilfe einer Eichkurve die entsprechenden Glucose- werte.
Verwendet man als Indikatorgemisch 2,2'-Azinowdi- [1-äthylchinolinsulfonsäure (6)] und m-Aminophenol, so liegt das Absorptionsmaximum bei 510 nm.
Beispiel 6
Ein gemäss Beispiel 1 hergestellter Reagenzfilm der 0,02 0/0 2,2'-Azino-di-[l-äthylchinolinsulfonsäure (6)] und 0,3 0/0 2,2-Azinodi-[3-äthylbenzthiazolon-sulfon- säure (6)] als Indikatorgemisch enthält, verfärbt sich mit steigender Glucosekonzentration von violett nach grün; dieser Farbumschlag lässt sich gut beobachten, da das menschliche Auge auf Änderungen in diesem Farbbereich empfindlich reagiert.
Beispiel 7 Nachweis von Glucose im Blut
0,1 ml Blut werden mit 1 ml 0,3 molarer Perchlorsäure enteiweisst. Von dem Überstand werden 0,2 mi in 5 ml einer Lösung folgender Zusammensetzung einpipettiert:
0,1 molarer Phosphatpuffer vom pH 7,0, welcher pro ml 40 zug Peroxydase, 250 itg Olucoseoxydase und 500 ,ug des Diammoniumsalzes der 2,2'-Azinodi-[3- äthyl-benzthiazolon-sulfonsäure (6)] enthält. Man misst die Extinktion bei einer Wellenlänge von A= 420 nIn und entnimmt die Glucosewerte einer Eichkurve.
Beispiel 8 Blutglucose-Teststreifen
45 g einer wässrigen Dispersion von Polyvinylpropionat (Propiofan, BASF), 35 g einer Lösung von 37 g Natriumalginat (Algipon) in 2 Liter eines 0,5 molaren Phosphat- oder Citratpuffers von pH 5,7, 1 g Texapon P (Fa. Henkel, Düsseldorf), 5 ml Wasser, 0,07 g Peroxydase, 0,16 g Glueoseoxydase werden zu einem homogenen Brei verrührt. Dazu kommen 0,3 g Ammoniumsalz der 2,2'-Azinodi-[3 -äthylbenzth,iazolon-sulfonsäure- (6)] und 0,04 g des Ammoniumsalzes der o-Aminophenol-p-sulfonsäure in 7 ml Wasser gelöst, sowie 0,3 g N, N,N,N'Tetramethyl-4,4'-diamino-diphenyl-methan in 4 ml Aceton gelöst. Die so hergestellte Mischung wird in einer Schichtdicke von 300 u auf eine Folie aufgetragen und getrocknet.
Bringt man auf einen so hergestellten Reagenzfilm Blut mit verschiedenen Glucosekonzentrationen, lässt ca. 1 Minute einwirken und wisoht wieder ab, so findet man im physiologischen Bereich Abstufungen von gelb über grün nach blau. (Abstufungen: bei 60 mg0/o gelb, 120 mgo/o grün, 180 mg0lo blaugrün, 240 mg0/o blau, 300 mg0/o blauviolett).
Texapon P ist ein waschaktives Produkt der Dehydag, Düsseldorf, dessen Zusammensetzung geheim gehalten wird.. Man kann stattdessen auch Natriumlau rylsulfat benutzen.

However, since there is no reaction with hydrogen peroxide alone, it was not to be expected that in the presence of peroxidase or peroxidatically active substances, reproducible, clear discolorations could also be achieved.
The agent according to the invention, which contains a) a hydroperoxide or a substance which reacts to release hydroperoxide or b) peroxidase or a peroxidatically active substance and a chromogen, is characterized in that it contains at least one compound of the formula I as chromogen .
The agent according to the invention is preferably suitable for coupled and uncoupled enzyme reactions, such as. B. for the determination of glucose, galactose, amino acids, uric acid, peroxides, hemoglobin, peroxidase or other peroxidatically active substances and enzyme activities. The routine determination of such substrates has become indispensable for clinical chemistry and food chemistry because of their elementary importance.
When determining glucose, for example, it is oxidized by glucose oxidase to gluconic acid, the oxygen in the air being reduced to hydrogen peroxide. The hydrogen peroxide then oxidizes the chromogen used according to the invention as an indicator to the corresponding dye by means of peroxidase or a substance with a peroxidic effect. Example 1 reagent film for the detection of glucose
45 g Propiofan (BASF), 35 g of a solution of 37 g Algipon in 2 liters of a 0.5 molar phosphate or citrate buffer with a pH of 5.7, 1 g Texapon P (Henkel Düsseldorf), 10 ml water, 0.075 g Peroxidase and 0.15 g of glucose oxidase are stirred to a homogeneous paste. 6 ml of a 5/0 strength aqueous solution of 2,2'-azino-di- [3-ethylbenzthiazolone-sulfonic acid (6)] are added to this mixture. The mixtures produced in this way, which each contain 0.2-0.3 io indicator, are used to coat films in a layer thickness of 300 μm and dry them. With aqueous solutions of different olucose concentrations, weakly graduated green colors are obtained.
If 6 ml of a 5% solution of p-aminosalicylic acid are added to the above mixtures instead of the aqueous solutions of 2,2'-azino-di- [3-ethyl-benzthiazolone-sulfonic acid (6)] test strips that only turn slightly yellow when wetted with aqueous glucose solutions and do not show any gradations of color. Example 3 Detection of blood in urine
A filter paper (2316 from Schleicher & Schüll) is impregnated with a 0.1 molar phosphate buffer of pH 7.0 in which 1 o / o Texapon P (from Henkel, Düsseldorf) is dissolved, and dried. The impregnated paper is then treated with an aqueous solution of the indicator mixture (0.03 O / o 2,2'-azino-di- [1-ethylquinoline sulfonic acid (6)] and 0.1 / o-aminophenol-p-sulfonic acid) again impregnated and dried. Example 4 Detection of Hydrogen Oxides in Liquids
A filter paper (2316 from Schleicher & BR <Schüll) is filled with 50 ml of a 0.1 molar phosphate buffer of pH 6.0 in which 25 mg peroxidase and 0.25 ml Texapon P (from Henkel, Düsseldorf) are dissolved, Impregnated and dried. The paper is then impregnated with a solution of 0.01 g of 2,2'-azino-di [1-ethylquinolinesulfonic acid (6)] and 0.1 g of o-aminophenol-p-sulfonic acid in 100 ml of water and dried . Example 5 Detection of Glucose in Liquids

Example 6
A reagent film produced according to Example 1 of 0.02 0/0 2,2'-azino-di- [1-ethylquinolinesulfonic acid (6)] and 0.3 0/0 2,2-azinodi- [3-ethylbenzthiazolone-sulfone- acid (6)] as an indicator mixture, changes in color from purple to green with increasing glucose concentration; this color change can be easily observed because the human eye is sensitive to changes in this color range.
Example 7 Detection of Glucose in Blood
0.1 ml of blood are deproteinized with 1 ml of 0.3 molar perchloric acid. 0.2 ml of the supernatant are pipetted into 5 ml of a solution of the following composition:
0.1 molar phosphate buffer of pH 7.0, which per ml contains 40 pounds peroxidase, 250 μg olucose oxidase and 500 μg of the diammonium salt of 2,2'-azinodi- [3-ethyl-benzthiazolone-sulfonic acid (6)]. If blood with different glucose concentrations is applied to a reagent film produced in this way, allowed to act for about 1 minute and then removed again, gradations from yellow to green to blue are found in the physiological range. (Gradations: at 60 mg0 / o yellow, 120 mg0 / o green, 180 mg0 / o blue-green, 240 mg0 / o blue, 300 mg0 / o blue-violet).
