dans laquelle R3 représente un radical alcoyle compor tant de 1 à 4 atomes de carbone ou un reste hydro- carboné non saturé comportant de 2 à 4 atomes de carbone.
Les produits de formule générale I sont dotés de propriétés pharmacologiques intéressantes. Ils possèdent notamment une action antiandrogène importante.
Ils se distinguent donc très nettement par leurs pro priétés pharmacologiques des 3-oxo estra-4,9,11-triènes déjà décrits. On connaissait, en particulier par la publi cation Th. Feyel Cabannes [Annales d'Endocrinologie, 26, 95 (1965)] et par la publication Velluz et Coll., C.R. Ac. Sci., 264 (1967) 1396, la haute activité androgène et anabolisante de ce type de composés. En particulier, le dérivé 17a-méthylé s'est révélé être le composé andro gène et anabolisant le plus actif connu.
En outre, les composés à chaîne latérale non saturée comme le com posé 17a-éthynylé ou le composé 17a-chloroéthynylé se sont révélés être progestomimétiques ou inhibiteurs hy pophysaires sans manifester d'effet estrogène alors que le dérivé 17a-vinylé possède une activité androgène et anabolisante (voir le brevet américain N 3257278).
Les 3-oxo estra-4,9,11-triènes forment donc une fa mille de composés doués de fortes propriétés androgènes ou progestomimétiques. Leur effet estrogène est inexis tant aux doses usuelles.
L'introduction d'un radical méthyle en position 12 dans une telle molécule a donc entraîné un changement profond des propriétés pharmacologiques de la molé cule.
Ce substituant a provoqué, d'une part, 1a disparition des propriétés androgènes ou progestomimétiques de la molécule, et, d'autre part, l'apparition de propriétés estrogènes marquées mises en évidence soit par des tests d'estrogénie, soit par la manifestation sur l'animal mâle de propriétés antiandrogènes.
Cette apparition de propriétés estrogènes par simple insertion d'un méthyle en 12, est, en outre, inattendue, car l'insertion d'un méthyle en d'autres positions (1 ou 6 ou 7 par exemple) entraîne soit l'exaltation, soit une atténuation relative, des propriétés androgènes.

Le procédé de préparation des composés de formule 1 est caractérisé en ce que l'on soumet le 3-oxo 12-mé- thyl 17ss-hydroxy estra-4,9,11-triène à l'action d'un agent de blocage de la fonction cétone en 3, fait réagir le dé rivé 12-méthyl 17ss-hydroxy estra-4,9,11-triénique résul tant avec un agent d'oxydation, soumet le dérivé 12-mé- thyl 17-oxo estra-4,9,11-triénique formé, dont la fonction cétone en 3 est bloquée, à l'action d'un dérivé organo métallique où le radical organe, est représenté par R.., ce qui conduit au 12-méthyl 17α

Le produit de départ, le 3-oxo 12-méthyl 17(ss-hy- droxy estra-4,9,11-triène, peut être préparé en faisant réagir un halogénure de méthylmagnésium sur le 3-oxo 17ss-hydroxy estra-4,9,11-triène (décrit dans le brevet français No 1380414), en présence d'un halogénure cui vreux pour former le 3-oxo 12-α-méthyl 17ss-hydroxy estra-5(10),9(11)-diène que l'on déshydrogène au moyen d'une dihalogéno dicyanobenzoquinone au sein d'un sol vant inerte.
On décrira ci-dessous plus en détail la préparation du produit de départ (stades A et B).
Stade A 3-oxo 12α-méthyl 17ss-hydroxy estra-S(10),9(11)- diène On introduit 0,94 g de chlorure cuivreux dans 300 cm3 d'une solution tétrahydrofuranique de bromure de méthylmagnésium 0,8 N et 300 cm3 de tétrahydro- furan, amène la température à -20 C, sous atmosphère d'azote, et ajoute 15 g de 3-oxo 17ss-hydroxy estra-4,9,11- triène (décrit dans le brevet français No 1380414) ; on agite pendant deux heures à 00 C environ et ajoute un litre d'acide chlorhydrique N ;
on extrait au chlorure de méthylène, lave les phases organiques à l'eau jusqu'à neutralité des eaux de lavage, sèche sur sulfate de sodium et évapore à sec, on recueille 18 g de produit brut que l'on purifie par chromatographie sur gel de silice, élution au mélange benzène-acétate d'éthyle (1-1), évaporation à sec de l'éluat, puis cristallisation dans l'éther, on ob tient 8,28 g de 3-oxo 12α-méthyl 17ss-hydroxy estra-5 (10),9(l 1)-diène.
Le produit se présente sous forme d'aiguilles incolo res, solubles dans la plupart des solvants organiques et insolubles dans l'eau, F = 1550 C.

Stade B 3-oxo 12-méthyl 17ss-hydroxy estra-4,9,11-triène On dissout 7,8g de 3-oxo 12α-méthyl 17ss-hydroxy estra-5(10),9(11)-diène dans 390 cm3 de benzène sous agitation, ajoute 10g de dichloro dicyanobenzoquinone et agite à température ambiante, sous azote, pendant trois heures trente minutes, on filtre, lave le filtrat avec une solution aqueuse de thiosulfate de sodium 0,1 N, puis à l'eau, sèche sur sulfate de sodium et évapore à sec sous vide, on recueille 3,721 g de composé 12-méthyl triénique brut.
Par traitement du précipité par le mélange de 0,5 litre de soude 5 N et de 0,5 litre de solution de thiosulfate de sodium 0,1 N et extraction au benzène, on recueille en outre un 2e jet de 0,407 g de dérivé 12-méthylé.
On purifie le produit par chromatographie sur gel de silice, élution au mélange benzène-acétate d'éthyle (1-1), évaporation à sec puis empâtage du résidu sec dans l'éther isopropylique ; on obtient le 3-oxo 12-méthyl 17ss-hydroxy estra-4,9,11-triène fondant à 145e C.
Ce composé se présente sous forme d'un produit incolore, soluble dans l'acétone, le benzène et le chloro forme; il est peu soluble dans l'alcool et l'éther; il est insoluble dans l'eau.

Ils peuvent être utilisés pour le traitement de l'adé nome prostatique, du cancer de la prostate, de l'hyper- androgénie, de l'acné, de l'hirsutisme.
Ils peuvent être utilisés par voie buccale, perlinguale. transcutanée, rectale et topique.
Ils peuvent se présenter sous forme de solutions ou de suspensions injectables, conditionnées en ampoules, en flacons à prises multiples, de comprimés, de compri més enrobés, de comprimés sublinguaux, de suppositoi res, de crèmes et de pommades.
La posologie utile s'échelonne généralement entre 10 mg et 500 mg par jour chez l'adulte, en fonction de la voie d'administration.
