Verfahren zur Herstellung von tricyclischen Aminen
5H-Dibenzo[a,d]cycloheptene und 10,11-Dihydro5H-dibenzo[a,d]cycloheptene mit einer basischen Seitenkette in 5-Stellung sind bereits als antidepressiv wirkende Pshchopharmaka bekannt, z.B.
10,11-Dihydro-5-(3'-dimethylaminopropyliden)
5H-dibenzo[a,d]cyclohepten und
10,11-Dihydro-5-(3'-dimethylaminopropyliden)
5H-dibenzo[a,d]cyclohepten.
Gegenstand des Hauptpatentes Nr. 485 648 ist ein Verfahren zur Herstellung von tricyclischen Aminen der Formel

in der R1 ein Wasserstoff oder eine Methylgruppe und X eine Athylengruppe oder eine gegebenenfalls durch ein Halogenatom substituierte Vinylgruppe bedeutet, die im aromatischen Teil durch Chor oder Fluor substi- tuiert sind, wobei ein bzw. der Substituent in 1 -Stellung steht, und von Salzen dieser Verbindungen.
Diese tri acyclischen Amine, die sich von den bekannten Vertretern der betreffenden Substanzgruppe durch die Anwesenheit eines Chlor- oder Fluoratoms in l-Stellung unterscheiden, zeichnen sich durch eine beträchtlich erhöhte antidepressive Wirkung und stark verminderte Toxizität aus. Als besonders vorteilhaft ist die fehlende oder nur geringfügige anticholinergische Wirkung zu er wähnen. Die Verbindungen der Formel Ia zeichnen sich ausserdem durch vielfältige Wirkungen auf das Nervensystem aus. So sind narkosepotenzierende, andrenolytische, sedative, antihistaminartige und lokalanästhetische Wirkungen festgestellt worden.
Es wurde nun gefunden, dass tricyclische Amine der Formel Ia, welche ausser dem Chlor- bzw. Fluoratom in 1-Stellung zusätzlich durch Chlor- bzw. Fluor in den aromatischen Kernen substituiert sind, und zwar Verbindungen der Formel

in der R1 ein Wasserstoffatom oder die Methyligruppe, R ein Chlor- oder Fluoratom, X eine Athylengrilppe oder eine gegebenenfalls durch ein Halogenatom substituierte Vinylengruppe und R2 und R3 je ein Wasserstoff-, Chlor oder Fluoratom bedeuten, wobei mindestens einer der Reste R2 und R3 Chlor oder Fluor darstellt, sowie dieser Verbindungen ebensolche Eigenschaften wie die Verbindungen der oben stehenden Formel In besitzen.
Die Verbindungen der Formel I bilden geometrische Isomere. Auch diese Isomere zeigen die vorstehend erwähnten überragenden Eigenschaften. Sie werden im folgenden als a- bzw. /3-Isomere bezeichnet.
In der obigen Formel I bedeutet R bevorzugt ein Chloratom. Bevorzugte Gruppen von Verbindungen der Formel I sind diejenigen, in der R2 ein Chloratom in 3-Stellung und R3 Wasserstoff bedeuten, sowie auch Verbindugen der Formel I, in der R2 Wasserstoff und R8 ein Chloratom in 9-Steliung darstellen. X ist vorzugsweise eine unsubstituierte Äthylen- oder Vinylengruppe, kann aber auch eine durch ein Halogenatom, insbesondere ein Chlor- oder Bromatom, substituierte Vinylengruppe darstellen. Besonders interessante Vertreter von Verbindungen der Formel I sind z.
1 ,3-Dichlor-1 0,11 -dihydro-5-'(3 -dimethyiamino- propyliden)-5H-dibenzo[a,d]cyclohepten (insbesondere dessen p-Isomeres) und 1 ,9-Dichlor- 10,11 -dihydro-5 -(3 -dimethyiamino- propyliden)-5H-dibenzo[a,d]cyclohepten.
Die Verbindungen der Formel I können in glei- cher Weise wie die oben stehenden Verbindungen der Formel Ia hergestellt werden, z. B. gemäss den Ausführungsformen des obengenannten Hauptpatents.
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von tricyclischen Aminen der Formel I sowie von Salzen dieser Verbindungen gemäss dem Verfahren des Hauptpatentes und ist dadurch gekennzeich- net, dass man eine Verbindung der Formel

in der R1, R R2, Rs und X die oben gegebene Bedeutung haben und eines der Symbole Y1 und Y ein Wasserstoffatom und das andere eine Hydroxylgruppe bedeutet, dehydratisiert, worauf man in beliebiger Reihenfolge erwünschtenfalls die Isomeren aus dem erhaltenen Isomerengemisch isoliert und erwünschtenfalls eine erhaltene Base in ein Säureadditionssalz überführt.
Bevorzugte Ausgangsstoffe sind Verbindungen der Formel II, in denen Y1 eine Hydroxylgruppe und Y Wasserstoff darstellen.
Die erfindungsgemässe Dehydratisierung wird zweckmässig unter Verwendung von Mineralsäuren, wie Chlor- oder Bromwasserstoffsäure, durchgeführt, wobei in wasserfreiem oder wässrigem Medium gearbeitet werden kann. Vorzugsweise wird die Dehydratisierung in äthanolischer Salzsäure bei einer Temperatur zwischen Raumtemperatur und dem Siedepunkt des Reaktionsgemisches ausgeführt. Sie gelingt jedoch auch durch Erwärmen, z.B. bei 50 bis Rückfllusstemperatur, vorzugsweise bei Rückflusstemperatur, mit einem hochsiedenden, wasserfreien Lösungsmittel, wie Dimethylsulfoxid. Andere übliche Wasserabspaltungsmittel können ebenfalls eingesetzt werden, z. B.
Acetylchlorid, Essigsäureanhydrid,
Trifluoressigsäureanhydrid, Schwefelsäure oder
Phosphoroxychlorid, p-Toiuolsulfochlorid,
Zinkchlorid oder Kaliumbisulfat, z.B. in einem inerten organischen Lösungsmittel, beispielsweise in Chloroform oder Methylenchlorid, und bei einer Temperatur zwischen Raumtemperatur und dem Siedepunkt des Reaktionsgemisches.
Das in der obigen Umsetzung eingesetzte Carbinol der Formel II kann z. B. in an sich bekannter Weise durch Umsetzen des entsprechenden tricyclischen 5 Ketons mit geeigneten Grignard-Verbindungen erhalten werden. Die tricyclischen 5-Ketone sind nach an sich bekannten Methoden erhältlich, wobei jedoch zu beachten ist, dass die Einführung eines Halogenatoms in 10 (bzw.11)-Stellung von l-Chlor(oder Fluor)-5H- dibenzo[a,d]cyclohepten-5;on bevorzugt unter Zugabe des entsprechenden Halogens mit gleichzeitiger Belichtung erfolgt.
Das resultierende
1-Chlor(oder Fluor)-10,11-dihalogen-10,11 dihydro-5H-dibenzo[a,d]cyclohepten-5-on spaltet nach Zugabe von Alkali ein Molekül Halogenwasserstoff ab, wobei ein
1-Chlor(oder Fluor)-10(bzw,11)-halogen-5H dibenzo[a,d]cyclohepten-5-on entsteht.
Carbinole der Formel II, in denen Y1 Hydroxyl und Y.2 Wasserstoff darstellen, werden zweckmässig wie folgt erhalten:
Falls ein Carbinol der Formel II, in der R1 Methyl bedeutet, hergestellt werden soll, setzt man ein entsprechendes tricyclisches 5-Keton direkt z.B. mit Dimethylaminopropyl-magnesiumchlorid um und hydrolysiert anschliessend das Reaktionsprodukt. Bedeutet R1 Wasserstoff, so empfiehlt es sich,
Mehylbenzylaminopropyl-magnesiumchlorid als Reagenz für die Umsetzung zu verwenden.
Nach beendigter Umsetzung und anschliessender Hydrolyse setzt man das entstandene Produkt mit Chlorameisensäureäthylester um und hydrolysiert anschliessend die entstandene
5-Hydroxy-5-[3'-(methyl-carbäthoxy-amino) propyl]-Verbindung, wobei eine spontane Decarboxylierung unter Bildung der entsprechenden 5-Hydroxy-5-(3 '-methylaminopropyl)-Verbindung eintritt.
Carbinole der Formel II, in denen umgekehrt Y1 Wasserstoff und Y Hydroxyl bedeuten, erhält man beispielsweise dadurch, dass man ein entsprechendes tricyclisches 5-Keton mit Äthylmagnesiumbromid umsetzt und das Reaktionsprodukt hydrolysiert. Die entstandene 5-Hydroxy-5-Äthyl-Verbindung wird mit Acetylchlorid dehydratisiert und anschliessend mit Ameisensäure und Wasserstoffperoxid behandelt. Es entsteht eine
5-Hydroxy-5-(1'-hydroxyäthyl)-Verbindung, welche mit wässriger Schwefelsäure zu der entsprechenden 5-Acetyl-Verbindung dehydratisiert wird. Durch Behandeln mit Formaldehyd und Methyl- bzw.
Dime thylamin-hydrochiorid erhält man eine 5-(Methyl- bzw. Dimethylaminoprs)pionyl)-
Verbindung, die nach Reduktion mit Natriumborhydrid in das ent sprechende Carbinol übergeht.
Erhaltene, unsymmetrisch substituierte Verbindun gen der Formel I und deren Salze können in ihre geo metrischen Isomeren, d. h. a- bzw. fi-Isomeren, getrennt werden. Die Trennungsmethoden sind an sich bekannt. Bevorzugt trennt man die geometrischen Isomeren durch fraktionierte Kristallisation der Säureadditionssalze aus einem Lösungsmittel, z. B. Aceton, oder aus einem Lösungsmittelgemisch, z. B. MethanoVDi- äthyläther.
Die Erfindung erfasst auch die Herstellung der Säureadditionssalze der tricyolischen Amine der Formel I. Solche Salze sind beispielsweise diejenigen mit organischen Säuren, wie Oxalsäure, Zitronensäure, Essigsäure, Milchsäure, Maleinsäure und Weinsäure, oder mit anorganischen Säuren, wie Chlorwasserstoffsäure, Bromwass erstoffs äure oder Schwefelsäure.
Die Verfahrensprodukte können als Heilmittel in Form pharmazeutischer Präparate Verwendung finden, welche sie oder ihre Salze in Mischung mit einem für die enterale oder parenterale Applikation geeigneten pharmazeutischen, organischen oder aanorganischen inerten Trägermaterial enthalten. Die pharmazeutischen Präparate können in fester Form oder in flüssiger Form vorliegen. Sie können rauch Hilfsstoffe, wie Kon servierungs-, Stabilisierung & , Netz- oder Emulgiermit- tel, Salze zur Veränderung des osmotischen Druckes oder Puffer, sowie auch noch andere therapeutisch wertvolle Stoffe enthalten.
Beispiel 1
25 g 1,3-Dichlor-10,11-dihydro-5-(3'-dimethylamino propyl)-5-hydroxy-5H-dibenzo[a,d]cyclo hepten werden mit 250 ml 30%iger äthanolischer Salzsäure 15 Studen unter Rückflussdedingungen erhitzt. Nach dem Eindampfen unter vermindertem Druck wird das Reaktionsgemisch in Wasser gelöst, mit Äther gewaschen und mit verdünnter Natronlauge alkalisch gestellt. Nach Idem Ausschütteln mit Äther und Eindampfen zur Trockne wird das zurückbleibende ölige Isomerengemisch von
1,3-Dichlor-10,11-dihydro-5-(3'-dimethylamino propyliden)-5H-dibenzo[a,d]cyclohepten durch Behandeln mit methanolischer Salzsäure in das Hydroxychlorid übergeführt. Nach dem Umkristallisieren aus Aceton fällt zuerst das Hydrochlorid des α-Iso- meren aus, Schmelzpunkt 231-233 C.
Aus den Mutterlaugen erhält man Kristallisation aus Methanol/Äther ein Produkt, das aus 94% des Hydrochlorids des ss-Isomeren besteht, Schmelzpunkt 180-183 C.
Das als Ausgangsmaterial eingesetzte
1,3-Dichlor-10,11-dihydro-5-(3'-dimethylamino propyl)-5-hydroxy-5H-dibenzo[a,d]cyclo hepten wird wie folgt hergestellt:
50 g 2-(2',4'-Dichlorphenäthyl)-benzoesäure und 100 ml Thioylchlorid werden 4 Studen unter Rückflussbedingungen erhitzt. Nach dem Einengen unter vermindertem Druck wird das erhaltene Säurechlorid in 500 ml & wefelkohlenstoff gelöst und zu einer siedenden Suspension von 70 g Aluminiumchlorid in 1 Liter Schwefelkohlenstoff eingetropft. Nach 4-stüh- digem Kochen wird unter vermindertem Druck einige dampft, mit Eis hydrolysiert und mit Äther ausgezogen, mit Wasser, 2n Natronlauge und Wasser gewa- schen, getrocknet und eingedampft.
Das rohe
1,3-Dichlor-10,11-dihydro-5H-dibenzo[a,d] cyclohepten-5-on wird an 300 g Silikagel in Benzol chromatographiert und schmilzt dann bei 108-110 C.
Durch Verwendung von 2,9 Gilman Legierung, 13,5 g Dimethyiaminopropylchlorid und 18 g
1,3-Dichlor-10,11-dihydro-5H-dibenzo[a,d] cyclohepten-5-on erhält man nach der im Beispiel 1 angegebenen Arbeizsweise
1,3-Dichlor-10,11-dihydro-5-(3'-dimethylamino propyl)-5-hydroxy-5H-dibenzo[a,d]cyclo hepten, das sofort weiter umgesetzt wird.
Beispiel 2
4,1 g 1,9-Dichlor-10,11-dihydro-5-(3'-dimethylamino propyl)-5-hydroxy-5H-dibenzo[a,d]cyclo hepten werden in 100 ml 30% iger äthanolischer Salzsäure 2 Stunden unter Rückflussbedingungen erhitzt. Nach dem Eindampfen unter vermindertem Druck wird der Rückstand in Wasser gelöst, mit Äther gewaschen, mit Natronlauge alkalisch gestellt und in Ather aufgenommen. Das nach dem Abdampfen des Äthers zurückbleibende, ölige
1,9-Dichlor-10,11-dihydro-5-(3'-dimethylamino propyliden)-5H-dibenzo[a,d]cyclohepten wird in Benzol gelöst, durch Filtration an 50 g Aluminiumoxyd (Aktivitätsstufe II) gereinigt und in das Hydrochlorid übergeführt, das nach dem Umkristallisieren aus Methano/Äther bei 234-2350 C schmilzt.
Das als Ausgangsmaterial eingesetzte
1,9-Dichlor-10,11-dihydro-5-(3'-dimethylamino propyl)-5-hydroxy-5H-dibenzo[a,d]cyclo hepten wird wie folgt hergestellt:
18,2 g 3-Chlor-phthalsäureanhydrid und 20,5 g (2 Chlorphenyl)-essigsäure werden mit 0,5 g Natriumace- tat geschmolzen. Die Schmelze wird unter Rühren in 120 ml Äthanol gegossen, abgekühlt und anschliessend filtriert. Das erhaltene Gemisch der beiden Isomeren
4-Chlor-3-(2'-chlorbenzyliden)-phthalid (A) und
7-Chlor-3-(2'-chlorbenzyliden)-phthalid (B) schmilzt bei 159-190 C. Die Trennung erfolgt durch Umkristallisation aus Aceton. Das gewünschte Isomere A fällt in über 96%iger Reinheit aus und schmilzt bei 204-209 C.
53 g des so erhaltenen Isomeren A werden in 300 ml Jodwasserstoffsäure mit dem spezifischen Gewicht 1,75 zusammen mit 40 g rotem Phosphor 24 Stunden in einer Argonatmosphäre unter Rückflúss- bedingun,gen erhitzt. Das Reaktionsgemisch wird nach dem Abkühlen mit 400 ml Wasser versetzt, filtriert und mit Wasser gewaschen. Der Rückstand wird in heisser 2n Natronlauge aufgelöst und die Lösung zur Entfernung von rückständigem Phosphor nochmals filtriert. Das Filtrat wird mit konzentrierter Salzsäure sauer gestellt und mit Methylenchlorid extrahiert. Der Methylenchlorid-Extrakt wird mit Wasser gewaschen, mit Natriumsulfat getrocknet und eingedampft.
Das erhaltene 4-Chlor-3-(2'-chlorbenzyl)-phthalid schmilzt nach dem Umkristallisieren aus Aceton/tiefsiedendem Petroläther bei 129-130 C.
32,5 g 4-Chlor-3-(2'-chlorbenzyl)-phthalid werden in einer Lösung von 14 g Kaliumhydroxid in 350 rm Wasser gelöst. Nach dem Einengen des Reaktionsge misches unter vermindertem Druck erhält man
Kalium-[3-chlor-2-(2'-chlor-α-hydroxy-phenäthyl) olophen-5-on, als einen festen Schaum.
48,6 g Kalium-[3-chlor-2-(2'-chlor-α-hydroxy-phen- äthyl)-benzoat] werden in 300 mi Dimethylsulfoxid gelöst und in einer Argonatmosphäre 3 1/2 Stunden auf 1700 C erhitzt. Das Reaktionsgemisch wird auf Eiswasser gegossen, das Ganze mit 2n Salzsäure angel säuert und mit Methylenchlorid extrahiert. Der Methylenchloridextrakt wird mit Wasser gewaschen, mit Natriumsulfat getrocknet und eingeengt. Man erhält
3-Chlor-2-(2'-chlorstyryl)-benzoesäure vom Schmelzpunkt 151-155 C.
15 g 3-Chlor-2-(2'-chlorstyryl)-benzoesäure werden in 250 ml Jodwasserstoffsäure vom spezifischen Gewicht 1,75 gelöst und mit 12 g rotem Phosphor 24 Stunden bei 1250 C in einer Argonatmosphäre geführt.
Das Reaktionsgemisch wird darauf mit 200 mi Wasser versetzt und filtriert. Der Rückstand wird in 50%iger Natronlauge unter Erwärmung gelöst. Nach dem Abfiltrieren der Phosphorrückstände wird Lösung mit Äther gewaschen, mit konzentrierter Salzsäure angesäuert und mit Methylenchlorin extrahiert. Der Mc thylenchlorid-Extrakt wird mit Natriumsulfat getrocknet und eingedampft. Man erhält
3-Chlor-2-(2'-chlorphenäthyl)-benzoesäure, die bei 137-139 C schmilzt.
50 g Polyphosphorsäure wird auf 120 C erhizt.
In einer Argonatmosphäre weden nun 7 g 3-Chlor-2 (2'-chlorphenäthyl)-benzoesäure hinzugefügt und das Ganze 5 Stunden auf 1500 C erhitzt. Das Reaktionsge- misch wird auf Eis gegossen und mit Benzol extrahiert.
Der Benzol-Extrakt wird nacheinander mit 2n Natronlauge und Wasser gewaschen, mit Nafriumsulfat getrocknet und eingedampft. Man erhält
1,9-Dichlor-10,11-dihydro-5H-dibenzo[a,d]cy clophen-5-on, das nach dem Umkristallisieren aus Aceton/tiefsieden- dem Petroläther bei 185-187 C schmilzt.
Durch Verwendung von 1,1 g Gilman-Legierung,
5,3 g Dimethylaminopropylchlorid und
3,4 g 1,9-Dichlor-10,11-dihydro-5H-dibenzo [a,d]cyclophen-5-on erhält man mit der im Beispiel 1 angegebenen Arbeitsweise rohes
1,9-Dichlor-10,11-dihydro-5-(3'-dimethylamino propyl)-5-hydroxy-5H-dibenzo[a,d]cyclo hepten als ein gelbes Öl', das sofort weiter umgesetzt wird.

These tri-acyclic amines, which differ from the known representatives of the substance group in question by the presence of a chlorine or fluorine atom in the l-position, are distinguished by a considerably increased antidepressant effect and greatly reduced toxicity. The lack of or only a slight anticholinergic effect should be mentioned as particularly advantageous.
